Introduction: Zika virus (ZIKAV) disease is a public health problem of international concern. Recent evidence has documented imported ZIKAV cases into the Middle East and the existence of ZIKAV-transmitting mosquitoes in Jordan. However, limited data exist on the role of physicians in public awareness in this regard. This study aimed to assess ZIKAV knowledge, attitudes and counseling practices (KAP) of general physicians and gynecologists in Amman, Jordan. Methodology: In this cross-sectional study, a structured paper-based questionnaire was completed by 119 participants during 2016-2017. Results: Only 4.2% of the physicians correctly addressed ZIKAV-complication questions. A misconception of considering direct contact between individuals and breastfeeding as modes of ZIKAV transmission was observed. Only one participant correctly recognized that isolation of infected or exposed persons is not recommended. Having at least five years of experience in medical practice was the only factor that was significantly associated with a high knowledge score (P-value=0.011). Although prevention measures are the sole method to control ZIKAV spread, only 50% of participants believed in the efficacy of such measures. Despite a quarter of participants perceiving ZIKAV as a threat to their patients, none of them have counseled a patient in this regard before. The presence of an evidence of ZIKAV in Jordan and health authorities' recommendations were the most important predictors for adoption of counseling practice. Conclusions: General physicians and gynecologists in Jordan had several gaps in knowledge of key aspects of ZIKAV disease, and there is a need for specific training programs of physicians and gynecologists.

Introdução: O virús Zika (ZIKAV) no Brasil foi identificado no início de 2015, concomitantemente houve um aumento de casos de microcefalia, sendo 1.248 novos casos, representando um salto de vinte vezes em relação aos últimos anos. Objetivo: Avaliar o desenvolvimento neuropsicomotor (DNPM) em portadores de microcefalia pelo ZIKAV. MATERIAIS e Métodos: Pesquisa observacional, de corte transversal, com análise descritiva, através da revisão de dados em prontuários com a amostra composta por 22 lactentes com microcefalia associada ao ZIKAV. Resultados: A idade média na consulta de 8,9 ± 2,13 meses, idade gestacional de 38,13 ± 2,38 semanas ao nascimento, peso de 2603.9 gramas ± 579,5 e perímetro cefálico de 28,86±1.74 centímetros. 10 (45,45%) pacientes apresentaram histórico de convulsões, 11 (50%) alterações visuais, 2 (9,09%) auditivas e 4 (18,18%) articulares. Observou-se atrasos no DNPM. O tônus de membros superiores e membros inferiores apresentaram-se aumentados, em que os membros superiores e membros inferiores apresentou-se aumentado, sendo que os membros superiores apresentaram o valor médio superior aos membros inferiores, porém sem significância estatística (p=0,1). Conclusão: Os portadores de microcefalia associada ao ZIKAV apresentam atraso no desenvolvimento neuropsicomotor,alterações visuais, auditivas e sensoriais, impactando na independência funcional e na inserção social dessa população.

Objectives: Zika virus (ZikaV) is currently one of the most important emerging viruses in the world which has caused outbreaks and epidemics and has also been associated with severe clinical manifestations and congenital malformations. Traditional approaches to combat the ZikaV outbreak are not effective for detection and control. The aim of this study is to propose a cloud-based system to prevent and control the spread of Zika virus disease using integration of mobile phones and Internet of Things (IoT).Methods: A Naive Bayesian Network (NBN) is used to diagnose the possibly infected users, and Google Maps Web service is used to provide the geographic positioning system (GPS)-based risk assessment to prevent the outbreak. It is used to represent each ZikaV infected user, mosquito-dense sites, and breeding sites on the Google map that helps the government healthcare authorities to control such risk-prone areas effectively and efficiently.Results: The performance and accuracy of the proposed system are evaluated using dataset for 2 million users. Our system provides high accuracy for initial diagnosis of different users according to their symptoms and appropriate GPS-based risk assessment.Conclusions: The cloud-based proposed system contributed to the accurate NBN-based classification of infected users and accurate identification of risk-prone areas using Google Maps.

Trong các vi rút truyền qua muỗi Aedes, bên cạnh vi rút DENV vi rút ZIKAV, CHIKV đang có xu hướng lan rộng ra nhiều quốc gia trên thế giới. Nghiên cứu này được tiến hành nhằm xác định sự lưu hành của muỗi Aedes nhiễm DENV, CHIKV và ZIKAV tại một số địa phương có số ca mắc sốt xuất huyết hàng năm cao ở miền Bắc Việt Nam trong thời gian 2016-2019. Trong năm 2016, nghiên cứu được thực hiện tại 3 xã phường của tỉnh Nam Định và 6 xã phường của Hà Nội. Muỗi Ae. aegypti và Ae. albopictus phát hiện được hầu hết ở các xã phường nghiên cứu. Muỗi Ae. aegypti chiếm tỷ trọng lớn ở các phường nội thành, nơi luôn có các ổ dịch sốt xuất huyết được phát hiện hàng năm. Phân tích tổng số 622 muỗi Ae. aegypti và Ae. albopictus từ các điểm nghiên cứu bằng kỹ thuật real-time RT PCR đa mồi phát hiện DENV/CHIKV/ ZIKAV chưa phát hiện thấy muỗi Aedes nhiễm DENV/CHIKV/ZIKAV. Áp dụng thêm kỹ thuật Pan RT-PCR chi Flaviviruses, Alphaviruses và Phelbovirus phân tích 478 muỗi Ae. aegypti thu thập tại Hà Nội năm 2019 cũng không phát hiện muỗi nhiễm các vi rút này. Vẫn có sự lưu hành của muỗi Aedes tại Nam Định và Hà Nội, chưa phát hiện thấy muỗi Aedes nhiễm DENV, CHIKV và ZIKAV. Các giám sát lưu hành Arbovirus trên muỗi cần tiếp tục duy trì, áp dụng thêm các kỹ thuật sinh học phân tử như Pan RT-PCR sàng lọc các chi/nhóm vi rút sẽ giúp tăng hiệu quả giám sát.

Analisamos 23 artigos, publicados entre 2016 e 2017, selecionados em periódicos brasileiros, no campo das Ciências Sociais e Humanas em Saúde. Há um discurso mestiço de reivindicação por direitos – grandes narrativas de apelo social e explicação pelas forças sociais de um Estado negligente – associado a pequenas narrativas de cuidado de um filho com deficiência. Provisoriamente, concluímos que a zona de contato se dá pelo movimento de resgate das pequenas narrativas na tradução das iniquidades produzidas pelo Estado na vida das pessoas comuns.

Zika, an emerging Aedes-mosquito-borne virus are currently being identified with alarming outbreak is spreading throughout the America. Health expert warn that anytime virus could enter Bangladesh due to worldwide easy communication of the people. Concerns have grown even stronger in Bangladesh after news media in Thailand and Taiwan reported cases of the viral infection among locals. Both places are popular destinations for Bangladeshi travellers, increasing the risk of the virus also spreading here. Aedes aegypti, the carrier of the virus, is also responsible for spreading dengue fever throughout the Indian sub-continent region, especially in Bangladesh and India. Pregnant women are at increased risk of neonatal complication like microcephaly if infected with ZIKA virus. Has the objective of identifying social vulnerabilities on the ZIKAV epidemic that occurred in Brazil in 2015. Search realize the main focuses of social vulnerability and possible impacts of the disease on policies for the promotion of women's reproductive health. Develops by means of the analysis of the official reports of the Ministry of the health of Brazil and of the data presented by the Brazilian Institute of geography and statistics (IBGE). Concludes by pointing to poverty and the lack of basic infrastructure services suitable for the main factor that caused the Brazilian outbreak. Describes how the disease affected mostly northeastern poor women. And weaves considerations of environmental racism and the report by the action of unconstitutionality (ADIN 5.581/2016), notably on the protection and defense of reproductive rights, especially the right to abortion.

Abstract Background Proyecto Vigilancia de Embarazadas con Zika (VEZ) was an intensified surveillance system built upon existing national surveillance of pregnant women with symptoms of Zika virus (ZIKV) disease and conducted in three Colombian cities with a high prevalence of Zika. This analysis of data from VEZ estimates the risk of Zika-associated birth defects among pregnant women with symptoms of ZIKV disease, and among a subset with laboratory evidence of possible ZIKV infection during pregnancy. Methods During April–November 2016, pregnant women were enrolled if they were reported to the surveillance system (Sivigila) or visited participating clinics with symptoms of ZIKV disease. Maternal and pediatric data were abstracted from prenatal care, ultrasound, and delivery records, as well as from pediatric or specialist visit records. Available maternal and infant specimens were tested for the presence of ZIKV RNA and/or anti-ZIKV immunoglobulin (IgM) antibodies. Results Of 1,223 women enrolled, 47.8% and 34.3% reported first or second trimester symptom onset, respectively. Of 381 pregnancies with maternal and/or infant specimens tested, 108 (29%) had laboratory evidence of possible ZIKV infection during pregnancy; half of these (53.3%) were positive for ZIKV RNA only, 37.4% for IgM antibodies only, and 9.3% for both. Of 1,190 of pregnancies with known outcome, 63 (5%) had Zika-associated brain or eye defects; among the subset with any laboratory evidence, 12 (11%) had Zika-associated brain or eye defects. The prevalence of Zika-associated brain or eye defects was 5.9% (35/593) and 4.5% (19/423) among pregnancies with symptom onset in the first and second trimester, respectively. Conclusion Among pregnant women with symptoms of ZIKV disease enrolled during the height of the ZIKV epidemic in Colombia, prevalence of any Zika-associated brain or eye defect was 5%, with a higher prevalence among those with laboratory evidence of possible ZIKV infection. Rapid enhancements to Colombia’s national surveillance enabled the estimation of the risk of birth defects associated with ZIKV disease in pregnancy. Disclosures All Authors: No reported Disclosures.

The majority of viral pathogens that cause emerging and re-emerging infectious diseases are membrane-enveloped viruses. In this thesis work a new series of antiviral compounds, with 1H-pyrrol-methylene thioxodihydropyrimidine structure, were designed and synthesized. They were endowed with antiviral activity on enveloped viruses, likely ZIKAV, DENV-2, and five influenza virus strains, including the pandemic H7N9 model. The intriguing mechanism of action was pursued and confirmed the hypothesis of compounds able to intercalate in the viral lipid envelope membrane and to oxidize phospholipids, impairing the physical and chemical structure of the viral membrane and impacting the fusion of viral and cell membranes. With the aim of investigating in vitro and in vivo pre-clinical features, compounds were assayed for their chemical-physical properties. Finally, a preliminary in vivo PK was assessed.

No Brasil a confirmação do primeiro caso de febre zika, resultante da infecção pelo vírus zika (ZIKV), foi no primeiro semestre de 2015, na região nordeste. Os achados característicos eram de uma doença que apresentava sintomas semelhantes aos sintomas causados pelo vírus dengue (DENV), porém mais amenos, sendo inicialmente denominada de \"Síndrome Dengue-Like\". Além desta semelhança no quadro clínico, ZIKV e os DENV são arboviroses endêmicas no Brasil e pertencem à mesma família viral, sendo muito próximos filogeneticamente, resultando em uma forte reação cruzada entre os anticorpos induzidos por estas infecções. O diagnóstico específico para o ZIKV requer cuidados uma vez que o DENV circula há muito mais tempo no Brasil e muitas pessoas já são imunes para, no mínimo, um sorotipo da doença. Desta forma, anticorpos contra os DENV nos testes sorológicos podem reagir inespecificamente contra o ZIKV, originando um resultado falso-positivo. As maneiras para diferenciar as duas doenças são o isolamento e a detecção do RNA viral durante a fase aguda da doença e dos anticorpos neutralizantes durante a fase de convalescença. Atualmente com a escassez de testes registrados pela ANVISA e a dificuldade em diferenciar as duas viroses usando testes sorológicos, amostras bem caracterizadas e confirmadas quanto à infecção pelo ZIKV foram utilizadas para o teste de comparação com vários kits comerciais aprovados ou não pela ANVISA, para detecção de anticorpos. Estas amostras também foram testadas por um ensaio soro-molecular padronizado e desenvolvido no Laboratório de Virologia Molecular.
In Brazil, the first case of zika fever, resulting from zika virus (ZIKV) infection, was confirmed in the first half of 2015, in the northeast region. The characteristic findings were of a disease that presented symptoms similar to the symptoms caused by dengue viruses (DENV), but milder, being initially called \"Dengue-Like Syndrome\". Although this similarity in the clinical presentation, ZIKV and (DENV) are arboviruses endemic in Brazil and belonging to the same viral family, being very close phylogenetically, and resulting in a strong cross-reaction between the antibodies induced by these infections. The specific diagnosis for ZIKV requires careful evaluation since DENV has been circulating much longer in Brazil and many people are already immune to at least one serotype of the disease. Thus, the antibodies against DENV may react non-specifically against ZIKV in serological tests, resulting in a false-positive result. The ways to differentiate the two diseases are the isolation and detection of viral RNA during the acute phase of the disease and the neutralizing antibodies during the convalescent phase. Actually, with the scarcity of tests registered by ANVISA and the difficulty in differentiating the two viruses using serological tests, well-characterized and confirmed samples for ZIKV infection were used for the comparison of several commercial kits for detection of antibodies approved or not by ANVISA. These samples were tested by a standardized serummolecular assay developed at the Molecular Virology Laboratory.

O Zika virus (ZIKV) é um arbovírus do gênero Flavivirus pertencente à família Flaviviridae. Duas principais linhagens são reconhecidas, Asiática e Africana, sendo a linhagem Asiática encontrada no Brasil. Os sintomas da infecção de ZIKV geralmente são brandos, mas existem relatos de casos de óbitos e manifestações neurológicas. Variações genéticas virais estão relacionadas com as diferenças nas manifestações clínicas da doença, portanto pesquisas sobre o genoma, filogenia e distribuição geográfica do vírus podem esclarecer como essas variações afetam a manifestação clínica do ZIKV, além de servir para o desenvolvimento de produtos biotecnológicos. Apesar da importância, poucos estudos avaliam as variações genéticas virais. Este estudo realizou uma abordagem epidemiológica e filogenética a nível populacional que analisou a diversidade genética do ZIKV no Espírito Santo (ES), e as relações filogenéticas das cepas de ZIKV identificadas com sequências virais disponíveis no GenBank. Constatou-se que os municípios de Vitória, Cariacica e Cachoeiro de Itapemirim foram os mais afetados, sendo a linhagem Asiática a circulante no ES, assim como no restante do país. Foram encontradas 4 variações em 2 genes estudados, no gene E A1023G e C1050T e no gene NS5 C1891T e T1945C. As variações A1023T e C1891T foram descritas pela primeira vez em linhagens Asiáticas neste trabalho, e as variações C1050T e T1945C foram descritas anteriormente em linhagem Asiática apenas uma vez em uma sequência do Rio de Janeiro e três sequências na China, respectivamente. Por fim, apesar das variações genéticas encontradas nas sequências do ES não estarem associadas à mudança de aminoácidos, não se pode descartar que outras mudanças, seja em ligações entre enzimas e o RNA, seja na estrutura das proteínas finais, podem ser causadas por essas variações. Os resultados apresentados podem ser úteis a futuros trabalhos permitindo uma expansão da base de dados apresentando uma visão mais clara da epidemiologia do ZIKV, o que poderá ajudar a determinar a origem geográfica de um novo surto e, além disso, monitorar a eficácia das medidas de controle.

The rapid and vast expansion of Zika virus in the Americas has heightened interest in members of the health sciences community, including public health agencies and officials. Concern of a future Zika outbreak in the United States is imminent. Prior studies, in combination with surveys administered by the author, suggest that mosquito avoidance behaviors could use improvement, especially in areas of high susceptibility of mosquito-borne illness dissemination. Border communities often face more health-related complications due to lack of appropriate resources, indicating the need for preventive education in these areas. The purpose of this thesis is to bring awareness to this outbreak potential, as well as providing a comprehensive health educational unit plan for high school students on the Tohono O’odham reservation in Southern Arizona. Mosquito-borne diseases are responsible for significant human morbidity and mortality throughout the world. Current vector control strategies have been impeded by mosquitoes acquiring resistance to insecticide. Therefore, development of new vector control strategies is urgently needed to complement current strategies. In this thesis, efforts have focused on characterizing the glycan-lectin interactions of Ross River virus (RRV; Togaviridae: Alphavirus) with their mosquito vectors. RRV is the most common arbovirus in Australia that causes clinical manifestations including arthralgia and myalgia. Many studies have shown the importance of viral surface glycans in mediating viral entry into host cells. Moreover, the viral surface glycans varies depending on the cells that they replicate in and this variation can affect the infectivity of virus. However, gaps remain in the role of viral glycans in virus host cell recognition. In Chapter 2, the surface glycans of RRV derived from two different cell lines, C6/36 (Ae. albopictus) and Vero (African green monkey kidney) were characterized using lectin array. Lectin array data revealed that RRV derived from two different cell lines exhibited similar glycan profiles. The glycan structures present on the surface of RRV are mannose, galactose, N-acetylglucosamine (GlcNAc) and N-acetylgalactosamine (GalNAc). To investigate the importance of these viral surface glycans in mediating viral entry into host cells, six lectins targeting these glycan structures were assessed for their ability to bind and block RRV entry into host cells. Of these lectins, two mannose-binding lectins, GNA and ConA, showed inhibition of RRV entry into C6/36 and Vero cells. These results suggest the potential use of mannose-binding lectins to block RRV transmission by mosquitoes. It is well known that cell surface glycans or lectins play important role in viral entry. Therefore, comprehensive surface glycan profiles and carbohydrate-binding characteristics of lectins from five mosquito cell lines were established in Chapter 3. Using lectin and glycan arrays, our results showed differences between the glycan structures and carbohydrate-binding characteristics of mosquito cell lines. In particular, complex-type glycans were detected on the cell surface of Ae. albopictus and An. gambiae. The presence of complex-type glycans as authentic constituents of insect glycans is still controversial and this is an important finding as complex-type glycans play diverse roles in regulation of biological functions. Zika virus (ZIKV) is primarily transmitted by the Aedes (Ae) mosquito, with Ae. aegypti and Ae. albopictus being the primary vectors. However, controversial findings on the potential of other mosquito species belonging to the genera of Anopheles (An) and Culex (Cx) have been reported. To date, the rationale underlying the specificity of ZIKV in infecting Aedes mosquitoes remain to be an unaddressed issue. In Chapter 4, we seek to characterize the susceptibility of seven cell lines derived from Ae, An and Cx mosquitoes towards ZIKV infection. Indeed, Ae cell lines were permissive to ZIKV infection and supported viral replication up to seven days post infection, while cells lines from An and Cx mosquitoes were unable to support replication. To specifically address if non-susceptible cell lines were due to the incompetence of ZIKV in establishing viral entry, a pseudoZIKV replicon system was utilized. Interestingly, while all Ae cell lines were highly susceptible to pseudoZIKV infection, the non-susceptible An. gambiae cell line (4a-3B) was also highly permissive to pseudoZIKV entry, in contrast to other An and Cx cell lines tested. Therefore, to identify the host factors involved in ZIKV replication in mosquito cells, RNA sequencing (RNAseq) analysis was performed on ZIKV-susceptible Ae and non-susceptible An cells after infection. Through comparative transcriptomics approach, we observed a differential regulation of attacin, an antimicrobial peptide (AMP) that may potentially play a critical role in modulating ZIKV replication in mosquito cells. Further investigations on the expression profile of different classes of AMPs including attacin, cecropin, defensin, diptericin and gambicin in Aedes; and attacin, cecropin, defensin and gambicin in Anopheles cells by qRT-PCR demonstrated that these AMPs were differentially regulated in ZIKV-susceptible and -resistant mosquito cells. These results suggest that the innate immunity may have a role to influence mosquito vector competence. Species specificity often relies on a specific interaction between a virus ligand and its host cell receptor. Therefore, expression levels of the receptor largely determine the tropisms of viruses. In Chapter 5, we constructed a representative cDNA library from the ZIKV-susceptible Ae. aegypti Aag-2 cell line using the SMART (Switching Mechanism At 5’ end of RNA Template) cDNA synthesis technology. This library will be a useful tool to provide genetic resources for many applications using the proposed strategy including identification of the cellular surface receptors for ZIKV viral entry and host factors required for sustained ZIKV replication in Aedes cells. Overall, this thesis provides in depth investigations into the glycan-lectin interactions between RRV and their mosquito vectors that may affect vector competence. Our study also identified the potential antiviral lectins that can block virus transmission. Finally, our study shed insights into the host factors involved in modulating ZIKV replication, providing a molecular platform for the future development of effective vector control and to evaluate the risk of emergence of a new vector for mosquito-transmitted viruses.

As the current growing threat to maternal-fetal health, the most recent and largest outbreak of the Zika virus has introduced the devastating fetal effects of microcephaly and other central nervous system deficits. Therefore, the need for appropriate recognition, treatment, management, and prevention of the Zika virus prompts the necessity for further education and high quality level research to be conducted and utilized. A search of the literature using the databases PubMed, UptoDate, and CINAHL was conducted for articles published between 2009 and 2016. In addition, key informant interviews from various specialties including clinical genetics and public health were conducted. The proposed best practice recommendations for education regarding the Zika virus and appropriate prevention and treatment methods are outlined in an electronic education module that would be delivered to patients and their families prior to visiting their healthcare providers. As the Zika virus continues to spread and further research is conducted regarding its teratogenic effects, the need for concise and effective education is critical in order to raise awareness and conversely decrease the potential for maternal exposure and adverse fetal outcomes.

Background: Infection by Zika virus (ZIKV) is associated with damage to the Central Nervous System, such as Congenital Zika Syndrome1 . Due to its transplacental transmission, ZIKV induces neuroinflammation and microglial activation, resulting in lesions that can compromise neurodevelopment2-4. The fetus protection can be provided by maternal antibodies. However, this protection is still controversial5 . In this context, it is necessary to elucidate the effects of ZIKV and the mechanisms involved. Objectives: The present work aim to evaluate the role of the ZIKV+IgG+ complex in murine microglia cells (BV2). Design and setting: BV2 were exposed for 24 or 72 hours, to ZIKV, ZIKA-IgG+ or ZIKV+IgG+ complex. Methods: Effects of exposure to treatments were evaluated by MTT, oxidation of DCFHDA (ROS production)6 and mitochondrial membrane potential (Δ∴ϑ), measured by JC-17 assay. Results: It was observed that ZIKV-IgG+ and the ZIKV+IgG+ complex are cytotoxic to microglia, impairing the viability of these cells, altering Δ∴ϑ and inducing the production of ROS, especially in long-term exposure8,9. Negative action mediated by these antibodies may be a result of oxidative stress and a intervention in the Δ∴ϑ. Conclusion: ZIKV-IgG+ antibodies are harmful to microglia and these mechanisms may be related to the potential for ZIKV neuroinflammation.

Introduction: The Zika virus (ZIKV) is an arbovirus of RNA, whose transmission is mainly vector - by mosquitoes of the genus Aedes - but it also occurs through sexual, blood and transplacental transmission, with the last mentioned it was possible to verify serious neurological effects in the epidemic in South America, especially in Brazil, between 2015 and 2016. Objectives: To analyze the relationship between Zika virus infection and microcephaly in recent scientific literature. Methodology: Refers to a bibliographic review in the databases SciELO, LILACS and MEDLINE / Pubmed, with the terms “zika virus”, “infection” and “microcephaly” correlated in Portuguese and in English; 78 articles were found, but only 7 followed for analysis. Articles published more than 5 years ago and out of the proposed theme were disregarded. Results: The Zika virus, although similar to the dengue and chikungunya virus, it has a tendency to cause damage to the central nervous system such as Guillain-Barré Syndrome. However, the association between microcephaly and ZIKV started to be more observed through the increase of the disease among fetuses and newborns of mothers who had been infected during the gestational phase in the epidemic that happened in Brazil. It is known that the development of the nervous system is the product of processes of high proliferation and cellular differentiation, in which even small errors generate dangerous impacts, and it is during this period that ZIKV affects the CNS of the fetus. The disease is characterized by the reduction of the brain perimeter, in this context, is a consequence of abnormalities influenced by the virus. Conclusions: Microcephaly is a complex disease; therefore, it is necessary to emphasize the importance of primary care and other spheres for monitoring Zika virus infections, prenatal care and constant psychosocial monitoring. Furthermore, it is necessary to understand the relevance of studies about ZIKV and microcephaly, and to encourage scientific production in this area.

Introdução: Por Zika vírus (ZIKV) entende-se o patógeno arboviral pertencente ao gênero Flavivirus, notório por ser transmitido vetorialmente por artrópodes do gênero Aedes - dentre eles o Aedes aegypti - e hábil a desencadear a infecção sistêmica Zika. Foi diagnosticado documentalmente, no Brasil, em maio de 2015, por métodos biomoleculares sistematizados por pesquisadores da Universidade Federal da Bahia. O progressivo aumento das notificações relacionadas ao nascimento de crianças com microcefalia, no Nordeste, exigiu atenção do Ministério da Saúde, a fim de investigar correlações fisiopatológicas reportadas, tais como acometimentos oculares e deficiências cognitivas, com alterações morfológicas em recém-nascidos com ZIKV. Objetivo: Revisar estudos sobre impactos congênitos neuronais em crianças infectadas verticalmente pelo ZIKV para compreender eventuais alterações morfológicas. Material e Métodos: Trata-se de uma Revisão Integrativa de Literatura, realizada a partir da leitura íntegra de 23 publicações científicas, escritas em Português e em Inglês entre 2016 e 2021, acerca das disfunções morfológicas embrionárias desencadeadas pela infecção por ZIKV. Tal elegibilidade deu-se pelo levantamento em Bases de Dados (Scopus, PubMed, Scielo e Portal CAPES) com cruzamento dos Descritores Zika virus, Embryology e Congenital Abnormalities. Resultados: A infecção fetal pelo ZIKV ainda não é totalmente esclarecida e, apesar de estudos sugerirem que o vírus seja capaz de driblar a imunoproteção placentária, ainda há um hiato quanto à influência viral em anomalias congênitas. Entretanto, já foi comprovado que o ZIKV induz efeitos citopáticos por desencadear lesões histológicas no citotrofoblasto ao subverter o sistema imunológico placentário, com tropismo por Macrófagos, via expressão do sfRNA viral. Alguns estudos identificaram antígenos virais em Células Progenitoras de Sinais Neuronais (hNPCs), sendo tal conteúdo viral hábil a modular negativamente a neurogênese por induzir a apoptose a partir da regulação, via mecanismos ainda obscuros, da expressão gênica. Particularmente, as hNPCs superexpressam Receptores AXL, capazes de viabilizarem o egresso intracelular do ZIKV e de mediarem vias de sinalização supressoras da resposta imunitária inata, magnificando a neurovirulência. Conclusão: A infecção congênita por ZIKV induz alterações morfológicas que lesam o Sistema Nervoso Fetal pela depleção das hNPCs corticais, resultando em malformações neurológicas.

Introdução: O vírus Zika (ZIKV) é uma doença infecciosa emergente transmitida por meio da picada do mosquito Aedes aegypti. Infecções tornaram-se cada vez mais comuns, e no ano de 2015 o primeiro caso foi registrado em território brasileiro, no nordeste, que concentrou 84% dos casos de microcefalia em recém nascidos e após o ano de 2016, registrou a maior quantidade de casos no país. Nesse contexto, alguns estados nordestinos apresentam variáveis favoráveis para a dinâmica vetorial de arboviroses, como o Zika. Logo, torna-se imprescindível o conhecimento acerca das formas de distribuição e desfechos da doença nesses estados. Objetivos: Analisar a prevalência e apresentação do Zika vírus no Ceará e Maranhão, ressaltando o perfil epidemiológico e o desfecho das infecções, a partir dos casos notificados no período de 2016 a 2021. Metodologia: Tratou-se de um estudo epidemiológico, retrospectivo, descritivo, quantitativo, que usou dados secundários acerca do Zika vírus nos estados Ceará e Maranhão, de 2016 a 2021, disponibilizados pelo Sistema de Informação de Agravos de Notificação (SINAN). Realizou-se a coleta de dados na plataforma do DATASUS, com base em indicadores e dados básicos sobre Zika vírus. Resultados: No intervalo de 2016 a 2021 foram notificados 15.947 casos de Zika vírus no Ceará e 6.657 no Maranhão. Nesse espaço de tempo é possível observar que a raça mais atingida foi a parda, correspondendo a 73,73% e 80% dos casos no Ceará e Maranhão, respectivamente. Em relação ao sexo, o mais afetado pelo ZIKV foi o feminino, no Ceará 70,22% eram mulheres e no Maranhão cerca de 65,73%, sendo que 16% estavam grávidas. De acordo com os dados obtidos no DATASUS, a quantidade de óbitos nos dois estados não foi significativa, no Ceará não houveram óbitos. A incidência média no Ceará totalizou 172,2 casos/100.000 habitantes, já no Maranhão 93,1 casos/100.000 habitantes. Conclusão: Dessa forma, os dados obtidos demonstram que os impactos causados pelo ZIKV, em ambos os estados foram expressivos em mulheres, e que o quesito raça/cor foi importante. Nesse contexto, ações conjuntas de combate e prevenção são necessárias para evitar a expansão do Zika vírus em regiões que favorecem a dinâmica vetorial.

However, diagnostics platforms at the point-of-care (POC) are highly desirable for understanding and preventing ZIKV transmission. To address this need, we have developed a testing platform that (1) can be operated in the field for pathogen detection, (2) is rapid, cost-effective, and reliable, and (3) does not require a power supply. To realize the platform, we have developed (1) a series of ball-based valves for the storage and sequential delivery of reagents and (2) microheater-enabled RNA amplification, both of which are integral components of this POC device. The multiple reagents are needed for virus lysis, RNA enrichment and purification. These ball-based are employed for fluid-control and they are actuated manually by sliding the unit and a pole under it, which can lift the balls. Nucleic acid amplification is then performed by a smart coffee mug that provides a constant temperature for reverse transcription loop mediated isothermal amplification (RT-LAMP), followed by colorimetric detection. We have demonstrated the detection of Zika virus in human urine and saliva samples using this testing platform.

Due to evidence that Zika virus (ZIKV) infection during pregnancy caused congenital brain anomalies, including microcephaly, in 2016 the WHO declared this disease a worldwide public health problem. The objective of this work is to identify the most important characteristics for the diagnosis of children with congenital syndrome due to ZIKV virus infection. We applied machine learning algorithms to RESP-Microcephaly, a database from the Brazilian Ministry of Health that records suspected cases of congenital abnormalities. At the end of the process, the most relevant characteristics were: weight, age of the pregnant woman, length, head circumference and region where the mother lives. This information is very significant as it is in agreement with the literature that associates these attributes with critical factors for the occurrence of congenital infection.

Introdução: O Aedes aegypti é uma espécie de mosquito pantropical responsável pela doença Zika. Sabe-se, que o vírus Zika (ZIKV) é transmitido por artrópodes (arbovírus) e atualmente, traz um grande problema de saúde pública para os brasileiros. Estudos filogenéticos sugerem, que o vírus Zika foi provavelmente importado do Pacífico para o Brasil, em 2015, e foi declarado uma emergência nacional de saúde pública. Objetivos: Quantificar os casos de Zika vírus, no Distrito Federal, registrados no Sistema de informações de Agravos de notificação, em 2018 a 2020. Metodologia: Estudo descritivo, quantitativo e qualitativo, que buscou coletar dados, através do Sistema de informações de Agravos de notificação (SINAN), que é um programa alimentado, principalmente, pela notificação e investigação de casos de doenças e agravos, que constam da lista nacional de doenças de notificação compulsória (Portaria de Consolidação nº 4/2017). Foram solicitados relatórios de casos dos anos 2018 a 2020, delimitando-se apenas ao Distrito Federal. Resultados: No Distrito Federal, em 2018, foram registrados 135 casos de Zika Vírus; em 2019, foram 420 casos, e em 2020, período pandêmico COVID-19, 2.293 casos. Observa-se um aumento considerável durante a Pandemia de Covid-19. A Zika provoca febre baixa, dor nas articulações, dor muscular, dor de cabeça, dor atrás dos olhos, conjuntivite, entre outros. Se fazem necessárias medidas sanitárias imediatas do Estado, campanhas educativas em nível nacional e visitas de agentes de saúde, além do uso de produtos químicos e biológicos, no Distrito Federal e no Brasil. Há estudos, que buscam descobrir a relação entre o vírus Zika e o surto de microcefalia na região Nordeste e o impacto do vírus na gestante. Urgem-se por maiores pesquisas a respeito do vírus. Conclusão: Conclui-se, que há necessidade de maiores estudos a respeito do Zika vírus. O saneamento básico é um problema de saúde pública, que colabora com a presença da doença Zika. Comprovou-se, que o Aedes aegypti é o mosquito mais fácil de criar e manipular em laboratório, sendo assim, são necessários investimentos em pesquisas com o objetivo de se obterem modificações genéticas, a fim de impedir sua descendência.

Background: Autoimmune encephalitis (AIE) is the main cause of non-infectious encephalitis and results from the peripheral immune response against cell surface antigens in the central nervous system. The clinical presentations are varied and known triggers are tumors and herpetic infections. Arboviruses Zika (ZIKV), Dengue (DENGV) and Chikungunya (CHIKV) are neurotropic infections that present neurological manifestations whose mechanism is unknown. Objective: Verify the frequency of arboviruses antibodies in patients with autoimmune encephalitis in a Brazilian cohort Design and setting: It is a transversal study performed by Hospital Israelita Albert Einstein in Brazil Methods: Patients who met the criteria for probable AIE (Graus 2016) evaluated at the 18 centers of the BrAIN network were included. Clinical, epidemiological and laboratory data were compiled. Antineuronal antibodies were detected using TBA, CBA and immunoblot in serum and CSF; antibodies against ZIKV, DENGV and CHIKV were detected by ELISA. The cohort was divided into two groups: seropositive encephalitic (SPE) and non-encephalitic (NE) and the frequencies of viral serologies were compared. Results: Among 619 patients included in the BrAIN cohort, serology for arboviruses was performed in 482 patients, being 79 SPE and 99 NE. The SPEgroup showed the following frequency of antibodies: 58.2% anti-NMDA, 7.6% antiLGI1, 6.3% anti-Caspr2, 2.5% anti-GABA B, 1.3% anti-GABA A, 3.8% anti-AMPAr, 1.3% anti-AQ4, 8.9% anti-MOG, 1.3% anti-IgLON5, 7.6% anti-GlyR and 5.1% others. The frequency of serology was IgG DENG (SPE 42.3% X NP 43.4%, p = 0.82); IgG CHIK (SPE 16.5% X 3.1% NP, p = 0.001); IgG ZIKV (SPE31.6% X NP 28.3%, p = 0.62). The frequency of triple positive serology (IgG DENG, ZIKV, CHIK) was 11.39% (SPE X 2.02% NP, p = 0.009). Conclusions: Patients with SPE have IgG CHIKV antibodies most commonly. In addition, they present a higher frequency of positivity for IgG CHIKV, ZIKV, DENGV simultaneously. Future studies should assess the association between arboviruses as a trigger for AIE or as a marker of susceptibility to immunological alteration.
