Abstract Poly (ADP-ribose) polymerase (PARP) is a critical enzyme for DNA repair and PARP inhibitors have been found to be efficacious against cancers with defects in DNA repair pathways such as with BRCA mutations. In addition, in vitro and in vivo studies have found that for PARP inhibitors with good trapping activity, combination with temozolomide (TMZ) produced the best synergetic effects among different types of chemotherapies. Several clinical combination studies of PARP inhibitors with TMZ have been reported, such as the combination of olaparib with TMZ in small cell lung cancer (SCLC). In this presentation, we will report in vitro and in vivo studies of PARP inhibitor senaparib (IMP4297) in combination with TMZ. Synergetic effects were observed when senaparib was combined with TMZ in vitro against human SCLC NCI-H209 cells. In vivo studies of high-dose senaparib in combination with low-dose TMZ once a day continuously in human SCLC NCI-H209 xenograft model was found to be highly synergetic and well tolerated. A clinical study of senaparib in combination with TMZ in patients with advanced solid tumors and SCLC is ongoing (ClinicalTrials.gov Identifier: NCT04434482). Abstract Poly (ADP-ribose) polymerase (PARP) is a key enzyme for DNA repair. PARP inhibitors have been shown to be effective against cancers with DNA repair defects, such as with BRCA1 or BRCA2 mutations. Several PARP inhibitors, including olaparib, rucaparib, niraparib, talazoparib, fluzoparib and pamiparib have been approved for the treatment of several types of cancer. In this presentation, we will report our discovery and development of senaparib (IMP4297) as a potent PARP inhibitor. We will present studies led to the identification of senaparib as a clinical candidate, as well as IND enabling in vitro and in vivo characterization of senaparib. Preclinical data indicated that senaparib had a wide therapeutic window (single agent effective dose of 2.5-21 mg/kg QD, in a BRCA1 mutated MDA-MB-436 human breast cancer xenograft model), which was confirmed in clinical studies (effective dose of 20-120 mg QD, and no DLT was found up to 150 mg QD). Senaparib is currently in several phase I/II, phase II and phase III clinical studies for the treatment of multiple tumor types either as single agent or in combination with temozolomide (TMZ). The most advanced study of senaparib is a phase III study as maintenance therapy in platinum-sensitive advanced ovarian cancer patients following first-line chemotherapy (ClinicalTrials.gov Identifier: NCT04169997). The BP artificial neural network model in type 7-5-5 was constructed with the surface water quality standard (GB3838-2002) and the surface water quality items such as BOD5 (5 day biochemical oxygen demand), COD (chemical oxygen demand), permanganate index, fluoride, NH3-N, TP (total phosphorus) and TN (total nitrogen), and the water environmental quality evaluation was conducted using the trained BP artificial neural network with the water contamination concentration data in 6 sections of Weihe river Baoji segment in year 2009. Results showed that the water quality were GradeIand GradeII in Lin Jia Cun section and Sheng Li Qiao section, and Grade III in the rest section (Wo Long Si Bridge, Guo Zhen Bridge, Cai Jia Po Bridge and Chang Xing Bridge).

Abstract Cyclin E1 amplification is prevalent in cancers with high unmet medical need such as high grade serous ovarian cancer, stomach cancer and esophageal cancer. Cancer cell lines with high expression level of cyclin E1 gene exhibited profound sensitivity to CDK2 gene depletion, suggesting CDK2 selective inhibitors have the potential to treat patients harboring Cyclin E1 alterations. Here we report the development and preclinical characterization of ARTS-021, an orally bioavailable small molecule CDK2 selective inhibitor. In enzymatic assay against a panel of different CDKs, ARTS-021 potently inhibits the activity of CDK2/CyclinE1 complex with 50% inhibitory concentration (IC50) in the sub-nanomolar range, displaying superior selectivity against other CDKs (CDK1,4,6,7,9). Very low hit rate (S(10)=0.022, at 1uM) is observed when ARTS-021 is assessed in whole Kinome profiling, further revealing the selectivity of this compound. ARTS-021 also demonstrates potent CDK2 inhibition and selectivity against other CDK family members (CDK1,4,6,9) in a series of cellular assays. Consistent with genetic CDK2 dependence data, cell lines with CCNE1 amplification are particularly sensitive to ARTS-021. Double nanomolar ARTS-021 inhibits Rb phosphorylation and blocks G1/S transition, leading to cell growth arrest specifically in CCNE1 amplified cell lines. This CDK2 dependent anti-tumor activity is further validated in vivo, where daily ARTS-021 administration leads to tumor stasis in CCNE1 amplified but not wild type xenograft models. Taken together, these data demonstrate ARTS-021 as a promising CDK2 selective inhibitor with strong potential towards the development of effective therapies for CCNE1 altered cancer. Recent research on Daoism has distinguished various models of self-cultivation present in the tradition, in particular those which aim at returning humanity to a natural, spontaneous form of existence (often associated with early pre-Qin “philosophical” Daoism), and those which aim at transcending human nature through technical practices (often associated with later “religious” Daoism). During the Wei-Jin period, organized Daoist religion was still in its early stages, yet the difference between the two models was very much an issue in the Dark Learning (xuanxue) thought of the intellectual elite. In this paper, I trace this debate as expressed in Wei-Jin thinker Guo Xiang’s Commentary to the Zhuangzi, in particular in Guo’s criticisms of the desire or attempt to exceed the limits (ji) of one’s inherent nature and his reinterpretation of the Zhuangzi’s criticisms of technical practices. While Guo follows Xiang Xiu in rejecting many of the claims of radical transcendence through self-cultivation, I argue that this does not imply that he lacks any positive conception of self-cultivation, but rather that he sees such cultivation as only possible through an immanent historical process in which both natural spontaneity and artificial techniques have a role to play.

Abstract CD73 expressed in Treg surface facilities the hydrolysis of AMP into adenosine which inhibits the activation, expansion and homing of T effector cells.Therapeutic efficiency of CD73 mAb had been studied in C57BL/6 and BALB/C mice xenografted with several murine cancer cell lines. However, for human CD73 mAb with no binding to murine CD73, CD73 humanized mouse models are essential for medicinal development.We established CD73 humanized mouse model (hCD73) on C57BL/6 and BALB/c background in which the mCD73 was replaced with hCD73. Similar expression level of hCD73 in peripheral blood was observed compared with mCD73 in wildtype mice. Besides, the proportion of T, B and NK cells in spleen of hCD73 mice were also analogy to that in wildtype mice. After treating with anti-hCD73 antibody, tumor growth was inhibited in our CT-26-hCD73 tumor model.Furthermore, hPD1/hPDL1/hCD73 mouse models were also established on C57BL/6 and BALB/c backgrounds based on hCD73 mice. However, a better anti-tumor effect of anti-CD73 antibody was found on BALB/c-hPD1/hPDL1/hCD73 bearing CT26 than that on B6-hPD1/hPDL1/hCD73 bearing MC38. Besides, Combination of anti-CD73 and anti-PD1 significant elevated anti-tumor effect comparing with anti-CD73 and anti-PD1 mono therapy in BALB/c-hPD1/hPDL1/hCD73 mice bearing CT26-hPDL1hCD73.In conclusion, BALB/c-hPD1/hPDL1/hCD73 is a well efficient mouse model for therapeutic effect study of anti-CD73 and relevant combination therapy. BackgroundRheumatoid arthritis (RA) is a common autoimmune disease with inflammation[1]. Total saponins of Panax japonicus (TSPJs) are effective components extracted from Panax japonicus[2]. They are known to exhibit anti-inflammatory and immunosuppressoive properties, but their effect of anti-inflammation in collagen-induced arthritis (CIA) remains unclear.ObjectivesTo investigate the anti-inflammatory targets of TSPJ predicted by bioinformatics and the verification in CIA mice.MethodsThe targets of RA are obtained in the GeneCards database. we used Cytoscape 3.7.2 software to construct a protein-protein interactions (PPI) network and obtain the hub genes. There are four effective components of TSPJ: Araloside A, chikusetsusaponin IVa, ginsenoside Rg2, and ginsenoside Ro. Through molecular docking between the screened hub genes and the four effective components of TSPJ, the possibility of TSPJ treating CIA mice can be predicted. The collagen II (CII) and complete Freund’s adjuvant (CFA) were used to induce the CIA model. After establishing the model, 32 DBA1/J mice were divided into C group (n=8), M group (n=8), L group(n=8), and H group(n=8). The L and H groups were gavaged with TSPJ at 30 mg/kg or 150 mg/kg, and the C and M groups were gavaged with normal saline. The thickness of the hind paw, number of swollen joints, and arthritis index were evaluated. After 11 days of treatment, all the mice were sacrificed after anesthesia. Sera were collected to centrifuge tubes and the levels of inflammatory factor were determined by the ELISA kit following the instruction.ResultsA gene list that enriches 263 genes was obtained by searching RA from the GeneCards database. The hub genes of the top 3 obtained from Cytoscape 3.7.2 software were tumor necrosis factor (TNF), interleukin-1β (IL-1β), and interleukin-6 (IL-6). In addition, interleukin-17A (IL-17A), a classical inflammatory index in the top 10, was selected and included in the predicted target. The results of molecular docking between the predicted target and the components of TSPJ showed that the combined pose has good stability. The numerical value of hind paw thickness, swollen joint counts, and arthritis index in the intervention groups were lower than those in the M group, suggesting TSPJ played a critical role in improving pathological changes. Compared to those of the C group, the serum levels of TNF-α, IL-1β, IL-6, and IL-17A were increased in the M group. Compared to those of the M group, the levels of TNF-α, IL-1β, IL-6, and IL-17A in the L and H groups were decreased. Compared to those of the L group, the levels of TNF-α, IL-1β, IL-6, and IL-17A in the H group were decreased. The results suggested that TSPJ may decrease the levels of TNF-α, IL-1β, IL-6, and IL-17A in CIA mice. These results suggest that TSPJ may inhibit the inflammatory effects of CIA mice.ConclusionCurrent study demonstrated a novel inhibitory effect of TSPJ on inflammation in CIA mice, and TSPJ can act on the targets predicted by bioinformatics of CIA mice, suggesting the potential of TSPJ as a therapeutic agent for RA and providing new ideas for the clinical treatment of RA.References[1]Scherer HU, Haupl T, Burmester GR. Background:Rheumatoid arthritis (RA) is a chronic, inflammatory synovitis based systemic disease of unknown etiology1. The genes and pathways in the inflamed synovium of RA patients are poorly understood.Objectives:This study aims to identify differentially expressed genes (DEGs) associated with the progression of synovitis in RA using bioinformatics analysis and explore its pathogenesis2.Methods:RA expression profile microarray data GSE89408 were acquired from the public gene chip database (GEO), including 152 synovial tissue samples from RA and 28 healthy synovial tissue samples. The DEGs of RA synovial tissues were screened by adopting the R software. The Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis were performed. Protein-protein interaction (PPI) networks were assembled with Cytoscape software.Results:A total of 654 DEGs (268 up-regulated genes and 386 down-regulated genes) were obtained by the differential analysis. The GO enrichment results showed that the up-regulated genes were significantly enriched in the biological processes of myeloid leukocyte activation, cellular response to interferon-gamma and immune response-regulating signaling pathway, and the down-regulated genes were significantly enriched in the biological processes of extracellular matrix, retinoid metabolic process and regulation of lipid metabolic process. The KEGG annotation showed the up-regulated genes mainly participated in the staphylococcus aureus infection, chemokine signaling pathway, lysosome signaling pathway and the down-regulated genes mainly participated in the PPAR signaling pathway, AMPK signaling pathway, ECM-receptor interaction and so on. The 9 hub genes (PTPRC, TLR2, tyrobp, CTSS, CCL2, CCR5, B2M, fcgr1a and PPBP) were obtained based on the String database model by using the Cytoscape software and cytoHubba plugin3.Conclusion:The findings identified the molecular mechanisms and the key hub genes of pathogenesis and progression of RA.References:[1]Xiong Y, Mi BB, Liu MF, et al. The Sixteen Kingdoms was a period of political disintegration in medieval China. Foreign rulers of Wu Hu (“Five Barbarian Tribes”) captured the northern China during the 3rd – 5th centuries and established independent states through continuous military actions. This thesis aims at studying the relationship between the emperors and Buddhist monks in the Northwest empires in the Sixteen Kingdoms Period, including the Former Liang (320-376), the Western Qin (385-400; 409-431), the Later Liang (386-403), the Northern Liang (397-439) and the Xia (407-431), by developing a theoretical model called “Hourglass Model”. The model provides a holistic framework for investigating not only the emperors’ changing attitudes towards Buddhism but also the dissemination of the Dharma by monks. In addition, the concept of “data visualization” is implemented to re-interpret various historical sources. The complex interaction among imperial clans and Buddhist practitioners is further analyzed with computer-aided historical research methods. This thesis hopes to reveal the inspiring nature of the political religion in China’s early medieval age and expand the scope of Chinese Studies by rethinking about foreign rulers and Buddhist monks’ roles in reshaping and vitalizing the Chinese civilization. Moreover, this study develops alternative approaches to the research on the history of Chinese Buddhism, and attempts to shed new lights on theories and research methodology.

One of the ten objectives of the 2004 Australian National Water Initiative is to manage surface and groundwater as a single resource. In order to do that it is necessary to understand the interactions between surface and groundwater, as well as the impacts of water abstraction, land use change and climate variability. In Australia, not just the quantity of water, but also its quality and particularly its salinity are critically important. Some of the difficulties facing agencies in managing surface and groundwater as a single resource are the extreme variability of climate in Australia, the lack of long-term streamflow and groundwater level data sets and the very limited temporal records on water quality. This thesis presents a study of surface and groundwater interaction and salinity in a selected catchment in the Hunter Valley in mid New South Wales, eastern Australia, where data records are limited and incomplete. The hypotheses tested in this work are that (1) salinity discharge in the Hunter is largely determined by mineral weathering and deep groundwater inflows and (2) a simple parameter-efficient coupled surface and groundwater model can accurately predict groundwater and streamflow behaviour over monthly time scale and is useful in determining surface-groundwater interactions.<....>

ABSTRACT: The key to develop the geothermal energy economically and effectively is forming a high quality enhanced geothermal system (EGS) with complex fracture network. Clarifying the interaction between hydraulic fracture (HF) and natural fracture (NF) is of positive significance to study how to establish an efficient EGS. In this paper, based on meso damage mechanics, elastic thermodynamics and Biot’s classical seepage mechanics, a thermo-hydro-mechanical-damage (THMD) coupling model of hydraulic fracturing in geothermal reservoir with randomly distributed NFs is established. Then the evolution of thermal field (T), seepage field (H), solid field (M) and damage (D) during hydraulic fracturing is simulated, and the effects of injection flow rates, rock thermal expansion coefficients and the NFs parameters (including length, dip angle and distribution density) on HF propagation are analyzed. The results indicate that under the action of water pressure and thermal stress, the normal stress on the NF surface will increase, resulting in the fracture damage. High injection flow rate would create longer fracture and significantly increase the fracture width, which is conducive to the development of deep thermal reservoirs. And high thermal expansion coefficient of rock is more conducive to form complex fractures. NFs will guide the HF to turn along the dip angle of NFs ,and the longer the NFs are, the stronger their guiding effect on HF. When the angle between the dip angle of NFs and the expansion direction of HF is large, the longitudinal expansion of HF is hindered, but the HF could communicate and activate more NFs. The NFs density directly determines the degree of hydraulic fracturing damage, and the damage area of HF increases with the increase of NFs density. 1. INTRODUCTION Geothermal energy is a widely distributed, low-carbon, environment-friendly and sustainable clean energy (Potten and Thuro 2017; Zhao et al. 2020). Hot Dry Rock (HDR) refers to the high-temperature rock mass with general temperature greater than 180 °C, buried depth of several kilometers, no fluid or only a small amount of underground fluid (Hofmann et al. 2014; Xu et al. 2012). As a new geothermal resource, it has advantages in geothermal energy development and application, which has attracted more and more attention. However, the HDR found at present is mainly composed of granite, showing low porosity and permeability (Lu and Wang 2015). Hydraulic fracturing technology is a mature and effective method to reconstruct low quality reservoir in oil and gas field development projects (Qu et al. 2021). This technology can be applied to HDR geothermal reservoir to realize efficient development of geothermal energy, too. Through the circulation of cold water in high-temperature rock formations with fractures, the geothermal energy is efficiently exploited. And then the high-temperature fluid will be used for step applications such as power generation or heating, which is the so-called enhanced geothermal system (EGS) (Bujakowski et al. 2015; Guo et al. 2019; Olasolo et al. 2016). Studies have shown that in the production process of EGS, the number of fractures or the complexity of fracture network will significantly affect the exploitation efficiency of geothermal energy (Guo et al. 2020b; Zhang et al. 2021; Jin et al. 2020). In order to form EGS with complex fracture networks for efficient development of geothermal energy, it is necessary to clarify the interaction between artificial fractures and natural fractures, and find out the evolution law of fracture during hydraulic fracturing.

Abstract Pressure transverse in foam drilling operation is sensitive and difficult to predict particular at the start of flow that follows the unavoidable shut in due to inevitable procedure of stop and proceed arising from re-connection of additional drilling pipe to further drill depth. The practice in drilling may not enable the flow to attain steadiness flow region before running in the length of drill pipe. Most existing models in the literature for predicting pressure transverse in foam drilling operation only captured the steadiness flow region of the foam drilling operation by keeping out restriction terms induced by accumulation and kinetic for simplicity sake, hence unsteadiness flow region experienced during foam drilling operation was rarely modelled. It is highly expedient to derive a model that evident the unsteadiness region in order to accurately predict pressure transverse, hence sufficiently analyses the well stability during foam drilling operation. In this study, a model for forecasting pressure transverse in foam drilling operation was established considering restriction term caused by accumulation and kinetic that constitute for accurate formulation of hydraulic model that govern flow of foam during underbalanced drilling. By applying the proposed model to a case study reported in literature, pressure transverse at unsteadiness flow region for foam drilling operation can be quantitatively estimated and analyzed. The result obtained in a case study carried out indicates high variance in pressure as function time at the beginning of flow in foam drilling where unsteadiness is promoted before matching up closely with the results obtained from the existing Guo et al 2003 model at the steadiness flow region. The new model has a better accuracy with a percentage error of 0.74% and 6.4% as compared to previous models by Guo et al 2003. The proposed model make possible for drilling engineer to take decision with larger precision during hydraulic design of foam drilling operation and guaranteeing well stability in complex drilling system.

In a previous publication (Guo & Chen et al., 2007), the authors solved the unsteady, 3-D Navier-Stokes equations with the k-ε turbulence model using CFX software to show that there is a volute stall coincided with the stage stall of a turbocharger centrifugal compressor operated at 423m/s tip speed and the stage stall frequency is dictated by a volute standing wave. This paper presents the flow condition at the vaneless diffuser and volute from the same simulation at various mass flow rates from stage peak efficiency to deep stage stall. Time averaged flow conditions show that (1) the influence of exducer blade passing at the volute inlet rapidly diminishes at the compressor peak pressure ratio point and the influence vanishes when the stage is in stall; (2) only at the peak pressure ratio point, circumferentially averaged, spanwise distribution of radial velocity at the volute inlet has an inflection point and the distribution meets the requirement of the Fjo̸rtoft instability theorem; (3) in the volute discharge section, the flow stalls after the stage stalls and the vortex core at the cross sectional center of the section breaks down; (4) impeller total pressure rise curve has a flat region in the middle before the stage stalls and (5) diffuser stall triggers the stage stall and drives the volute into stall.

In this paper, the investigation on the instability in parallel channel system is summarized systematically. This phenomenon in parallel channel system is very typical, interesting and challengeable. The experiment data of a twin-channel system is used as the validation. Two typical methods are adopted to simulate this phenomenon for deciding the instability boundary. One is the integral method, which is based on the model of Clausse and Lahey and developed by Lee and Pan and GUO; the other is the classical system analysis code: Relap5/MOD3.4. In the experiment the influences of inlet resistance, system pressure and nonuniform heating are obtained. The influences of system pressure and inlet resistance can be simulated by both methods. However, there are some differences between the results of two methods. And for the effects of nonuniform heating and asymmetric inlet resistances, which are very popular in the nuclear power system, the results of numerical methods cannot get a good numerical agreement with those of experiment. It should be noticed in the practical engineering design. Finally, the typical “Ledinegg” instability phenomenon may occur in the parallel channel system according to the numerical results. Sometimes it will induce the burnout before the parallel channel instability. Both methods predict the same tendency. And a detailed explanation is given. The slope of the pressure drop-mass flux curve is the key to avoid the flow excursion phenomenon in parallel channel system.

MEMS accelerometers have found applications in harsh environments with pressure, temperatures above ambient conditions, high g shock and vibrations. The complex structure of these MEMS devices has made it difficult to understand the failure modes and failure mechanisms of present day MEMS accelerometers. Little work has been done by the researchers in investigating the high g reliability of the MEMS accelerometers by continuous high g drops and quantifying the failure modes. There is little literature addressing the multiphysics finite element modelling of MEMS accelerometers subjected to high g shocks. In defense applications, where these devices are integrated with several other compactly assembled subsystems, lack of knowledge on the physics of failure for the MEMS sensor in harsh environment operation, can be detrimental to the success of the system on the whole. Being able to successfully model inside of an accelerometer, enables the user to better understand the change in parameters like time delay induced in response of successive drops, change in pulse width that indicate failure, reduction in sensed g levels. Some researchers have subjected various accelerometers to repeated drops at their maximum sensing g(not high g) level, and used optical microscopy to detect damaged sensing elements [Beliveau, 1999]. Few researchers have modeled the internal structure of the MEMS device, along with the device packaging under the stresses of operation [Fang 2004, Ghisi 2008, Xiong 2008]. In this paper, a multiphysics model of capacitive and the moving elements of the accelerometer has been developed to model the change in capacitance with respect to stroke and understand the correlation with g-levels, in addition to the transient dynamic response of the accelerometer under high-g shock. This has not been much explored in the past. The accelerometer studied in the paper is the ADXL193, and subjected to repeated drops of 3000g in each 3 axes as per 2002.4 of MIL-STD-883 without preconditioning. A characteristic graph of capacitance vs accelerometer stroke has been obtained from a series of electrostatic simulations and is then used to relate g levels, capacitance, stroke deflection and voltage change using electromechanical transducer elements. The drift in the performance characteristics of the accelerometer have been measured versus the number of shock events. In addition, an attempt has been made to investigate the failure mode in the accelerometer.

Review question / Objective: In this study, we conducted a Bayesian network meta-analysis to evaluate the efficacy and safety of interleukin (IL) inhibitors, tumor necrosis factor-alpha (TNF-α) inhibitors, and Janus kinase (JAK) inhibitors on ankylosing spondylitis (AS).The purpose of this study is to compare the effectiveness and safety of different interventions for treating AS to provide insights into the decision-making in clinicalpractice. Condition being studied: Ankylosing spondylitis. Based on the Bayesian hierarchical model, we conducted a network meta-analysis using the gemtc package in R software (version 4.1.3) and Stata software (version 15.1). Cong Tian and Jianlong Shu contributed to the conception and design of the study and supervised the tweet classification. All authors drafted the manuscript.
