Due to its direct relevance to post-disaster operations, meter reading and civil refuse collection, the Uncertain Capacitated Arc Routing Problem (UCARP) is an important optimisation problem. Stochastic models are critical to study as they more accurately represent the real world than their deterministic counterparts. Although there have been extensive studies in solving routing problems under uncertainty, very few have considered UCARP, and none consider collaboration between vehicles to handle the negative effects of uncertainty. This article proposes a novel Solution Construction Procedure (SCP) that generates solutions to UCARP within a collaborative, multi-vehicle framework. It consists of two types of collaborative activities: one when a vehicle unexpectedly expends capacity ( route failure), and the other during the refill process. Then, we propose a Genetic Programming Hyper-Heuristic (GPHH) algorithm to evolve the routing policy used within the collaborative framework. The experimental studies show that the new heuristic with vehicle collaboration and GP-evolved routing policy significantly outperforms the compared state-of-the-art algorithms on commonly studied test problems. This is shown to be especially true on instances with larger numbers of tasks and vehicles. This clearly shows the advantage of vehicle collaboration in handling the uncertain environment, and the effectiveness of the newly proposed algorithm.

Abstract Based on compelling evidence from a vast number of in vitro and in vivostudies, Tregs have become an attractive cell population to treat or even prevent auto- and alloimmunity including Graft-versus-Host disease (GvHD). However, several safety concerns still exist as for example the risk of global immunosuppression using polyclonal Tregs. In fact, experiments in mice showed that adoptive transfer or induction of antigen-specific Tregs is more potent regarding suppression of pathogenic immune responses when compared to polyclonal Treg populations. Unfortunately, the isolation and expansion of naturally occurring antigen-specific Tregs is technically difficult, labour-intensive, and time-consuming. An attractive way to overcome these limitations and to endow polyclonal Treg populations with a desired antigen-specificity is their engraftment with chimeric antigen receptors (CARs). In this context, CAR-modification represents a promising approach to redirect polyclonal Tregs in an antigen-specific manner to suppress ongoing self-destructive immune responses at the site of inflammation. Nevertheless, until now redirection of CAR-engineered T cells is limited to a single target antigen, restricting this approach to an unflexible monospecific therapy. Therefore, we developed a more flexible universal CAR (UCAR) platform that allows redirection of T cells to an in principal unrestricted number of surface antigens. T cells are engrafted with UCARs that bind to a small peptide epitope derived from a human nuclear protein. Cross-linkage to target cells is mediated by independent target modules that provide antigen-specificity and comprise the peptide epitope recognized by the UCAR. In order to target different tissue antigens, the target modules can easily be exchanged. Thereby, once established, the treatment strategy can easily be applied to various auto- and alloimmune diseases. At present, the CD45RA+ population is the Treg subset of choice for a clinical application as these cells have the highest capacity to maintain phenotypic and functional Treg properties upon prolonged ex vivo expansion. Here we show that highly pure, sorted CD4+CD25+CD127lowCD45RA+ Tregs can be genetically manipulated using lentiviral gene transfer, resulting in approximately 70 % of UCAR-expressing Treg cells. The transduction procedure itself did not affect the phenotype of UCAR-engineered Tregs as it was similar to non-transduced wildtype cells. Both Treg populations presevered FOXP3 expression even after prolonged in vitro cultivation (> 95 % FOXP3+). Upon incubation with antigen-positive target cells and a respective target module UCAR-engineered Tregs upregulate the activation markers CD69 and LAP demonstrating that the cells can be restimulated antigen-specifically. Most importantly, UCAR-engrafted Tregs were functionally activated upon antigen encounter, demonstrated by suppression of proliferation and expansion of cocultured autologous T effector cells. Taken together, our results pave the way towards an application of UCAR technology for a site-specific recruitment of CAR-modified Tregs into inflamed tissues aiming at re-establishing immune homeostasis. Due to its high flexibility UCAR-engrafted Tregs can easily and universally be used for treatment of various autoimmune diseases or GvHD just by exchanging the tissue-specific target modules. Abstract The reliability of spot urinary Ca to P ratio (uCa:P) to assess P intake adequacy in sows was evaluated. Thirty-six multiparous sows were fed one of six concentrations of dietary total P (0.40, 0.48, 0.56, 0.64, 0.72, and 0.80%), with a constant Ca to total P ratio (1.25:1), from day 7.5+1 after breeding until the end of lactation (day 26.6+1). Total 24-hour urine samples were collected in mid (day 77.1+2) and late gestation (day 112.4+1), and early (day 4.5+1 and late (day 18.2+1) lactation. In parallel to 24-hour collections, spot urine samples were collected at three times (early morning, late morning, and late afternoon) in late gestation and late lactation. Urine Ca and P concentrations were measured and uCa:P was calculated. Sows were classified as P-adequate or P-deficient according to dietary P intake. Sows fed P-deficient diets had greater uCa:P than sows fed P-adequate diets (P < 0.001). Receiver operator characteristic (ROC) curves were used to determine cut-off values for uCa:P to predict P intake adequacy. The area under the ROC for uCa:P was 0.88 (95% CI 0.81–0.95). Best cut-off value of uCa:P was 1.5 (sensitivity 94% and specificity 68%) to identify sows fed P-deficient diets, and 0.5 for P-excessive diets (sensitivity 82% and specificity 82%). A strong relationship between uCa:P in 24-hour and spot urine samples was determined (r = 0.93, P < 0.01), independent of physiological state and collection time of spot samples (adjusted-R2 = 0.86, P < 0.01). The degree of agreement between spot and 24-hour urine for P intake adequacy, assessed by Cohen’s weighted kappa analysis, was substantial (0.78, 95% CI 0.69–0.88). Measurements of uCa:P in spot urine samples provide a reliable prediction of the adequacy of P intake in reproducing sows. Values of uCa:P > 1.5 were associated with P-deficient diets, whereas uCa:P < 0.5 reflected excessive P intake.

455 Background: Whether pT3 urothelial carcinoma of the renal pelvis (UCRP) and urothelial carcinoma of the ureter (UCU) show the same prognosis or not remains controversial.This study aims to reveal whether the prognosis of pT3 UCRP shows similar to those of pT3 UCU. Methods: We retrospectively evaluated 954 patients who underwent nephroureterectomy between 1983 and 2017 at our institutions.None of the patients received neoadjuvant therapy.All HE slides for each patient were reviewed by a single genitourinary pathologist (TT).pT3 UCRP was subdivided pT3a (urothelial carcinomas that extend only to the renal medulla) and pT3b (urothelial carcinomas that extend to the renal cortex and/or peripelvic adipose tissue) according to the criteria previously we proposed.Fine and Gray model was developed to predict the reccurence-free survival (RFS) and cancer-specific survival (CSS). Results: Overall, 954 patients were eligible for this study, 493 of whom were pT3 with only renal pelvis and 461 of whom were pT3 with only ureter. A total of 202 cases were pT3 UCRP and 146 were pT3 UCU. The pT3 subclassification of UCRP resulted in 79 pT3a and 120 pT3b cases.No significant difference was found in the 5-yr RFS and 5-yr CSS between pT3a UCRP and pT2 UCRP and pT2 UCU. (RFS; pT3a UCRP vs pT2 UCRP: HR = 1.19 p = 0.74; pT3a UCRP vs pT2 UCU HR = 0.88, p = 0.72, CSS;pT3a UCRP vs. pT2 UCRP: HR = 0.69; p = 0.56; pT3a UCRP vs. pT2 UCU: HR = 0.66, p = 0.31) The RFS rate was significantly higher in the pT3a UCRP than that of pT3b, (pT3a vs pT3b: HR = 2.59, p = 0.0038) and so was the CSS rate. (pT3a vs. pT3b: HR = 3.1 p = 0.001) In fine and gray model analysis for RFS, pT stage, lymph node metastasis, WHO/ISUP grade, lymphovascular invasion were independently associated with tumor recurrence, whereas pT stage, lymph node metastasis, operation method, histologic type, lymphovascular invasion, gross type, adjuvant chemotherapy were independently associated with cancer mortality. Conclusions: Subclassifying pT3 UCRP into pT3a and pT3b was useful in determining the prognosis of different patients.The pT3b UCRP showed similar outcome compared to those of UCU.

Gla-rich protein (GRP) was described in sturgeon as a new vitamin-K-dependent protein (VKDP) with a high density of Gla residues and associated with ectopic calcifications in humans. Although VKDPs function has been related withγ-carboxylation, the Gla status of GRP in humans is still unknown. Here, we investigated the expression of recently identified GRP spliced transcripts, theγ-carboxylation status, and its association with ectopic calcifications, in skin basal cell and breast carcinomas. GRP-F1 was identified as the predominant splice variant expressed in healthy and cancer tissues. Patterns ofγ-carboxylated GRP (cGRP)/undercarboxylated GRP (ucGRP) accumulation in healthy and cancer tissues were determined by immunohistochemistry, using newly developed conformation-specific antibodies. Both GRP protein forms were found colocalized in healthy tissues, while ucGRP was the predominant form associated with tumor cells. Both cGRP and ucGRP found at sites of microcalcifications were shown to havein vitrocalcium mineral-binding capacity. The decreased levels of cGRP and predominance of ucGRP in tumor cells suggest that GRP may represent a new target for the anticancer potential of vitamin K. Also, the direct interaction of cGRP and ucGRP with BCP crystals provides a possible mechanism explaining GRP association with pathological mineralization.

Ubiquitin cross-reactive protein (UCRP), a 15-kDa interferon-induced protein, is a sequence homolog of ubiquitin that is covalently ligated to intracellular proteins in a parallel enzymatic reaction and is found at low levels within cultured cell lines and human tissues not exposed to interferon. Ubiquitin and UCRP ligation reactions apparently target distinct subsets of intracellular proteins, as judged from differences in the distributions of the respective adducts revealed on immunoblots. In this study, successive passages of the human lung carcinoma line A549 in the presence of neutralizing antibodies against alpha and beta interferons had no effect on the levels of either free or conjugated UCRP, indicating that these UCRP pools are constitutively present within uninduced cells and are thus not a consequence of autoinduction by low levels of secreted alpha/beta interferon. In an effort to identify potential targets for UCRP conjugation, the immunocytochemical distribution of UCRP was examined by using affinity-purified polyclonal antibodies against recombinant polypeptide. UCRP distributes in a punctate cytoskeletal pattern that is resistant to extraction by nonionic detergents (e.g., Triton X-100) in both uninduced and interferon-treated A549 cells. The cytoskeletal pattern colocalizes with the intermediate filament network of epithelial and mesothelial cell lines. Immunoblots of parallel Triton X-100-insoluble cell extracts suggest that the cytoskeletal association largely results from the noncovalent association of UCRP conjugates with the intermediate filaments rather than direct ligation of the polypeptide to structural components of the filaments. A significant increase in the sequestration of UCRP adducts on intermediate filaments accompanies interferon induction. These results suggest that UCRP may serve as a trans-acting binding factor directing the association of ligated target proteins to intermediate filaments.

Saving the ovarian function of premature ovarian failure (POF) patients undergoing chemotherapy is an important problem in the field of reproductive medicine. At present, human umbilical cord mesenchymal stem cells (HucMSCs) have been used in the treatment of POF, but the effect is still not optimal. The purpose of this study was to determine whether human umbilical cord blood platelet-rich plasma (ucPRP) enhances the beneficial effects of HucMSCs in the treatment of POF. First, we observed the effects of changes in the biological activity of ucPRP on HucMSCs in vitro. Subsequently, we tracked the distribution and function of the HucMSCs in POF rats, and the rats’ estrus cycle and serum sex hormones, follicular development, ovarian angiogenesis, ovarian granulosa cell proliferation, and apoptosis were assessed. The results of the study showed that the addition of ucPRP in vitro accelerates proliferation and reduces apoptosis of the HucMSCs while upregulating the stemness gene of the HucMSCs. The combined transplantation of HucMSCs and ucPRP resulted in more stem cells being retained in the ovaries of POF rats, the estrus cycle of the POF rats being restored, the levels of serum E2, AMH, and FSH improving, and damaged follicles beginning to grow. Finally, we confirmed that the potential mechanism of the combination of HucMSCs and ucPRP to rescue the ovarian function of POF rats is to promote ovarian angiogenesis and to promote the proliferation and reduce the apoptosis of ovarian granulosa cells. The upregulation of AMH and FHSR expression and the downregulation of caspase-3 expression in granulosa cells are potential mechanisms for the recovery of ovarian function. Our research results suggest that the combined application of HucMSCs and ucPRP is a safe and efficient transplantation program for the treatment of POF, thus providing a reliable experimental basis for the clinical application of stem cell therapy in POF.

Resumo: Este trabalho analisa a experiência em pesquisa e desenvolvimento da Cooperativa de Produtores de Cana, Açúcar e Álcool do' Estado de São Paulo (COPERSUCAR). São apontandos os fatores que levaram à internalização da pesquisa por parte desta cooperativa no contexto das relações de produção predominantes na agroindústria canavieira. Por outro lado, são analisados os diversos programas de pesquisa desenvolvidos pelo Centro de Tecnologia da COPERSUCAR desde 1969, na área de melhoramento vegetal, no processamento industrial e na área de máquinas agrícolas. Por último, é avaliada a contribuição desta experiência para a capacitação tecnol6gica no setor sucro-alcooleiro no país
Abstract: This text analyses the R&D experience of the COPERSUCAR (Alcohol and Sugar Cane São Paulo State Producers Cooperative). The factors that have induced "in house" research are discussed, on the context of the prevalescent production relations of the sugar cane agro-industry. The distinct research programs developed by the CTC (COPERSUCAR Technology Center) since 1969 are also analysed, with special emphasis on the vegetable improvement, industrial process and agricultural machinery areas. As a last point, it is analysed the CTC contribution to the technological capacitation of the brazilian sugar-alcohol productive sector

El objetivo principal de la presente tesis es la elaboración de un perfil de plan estratégico que contribuya al fortalecimiento de la “Unión de Cerveceros Artesanales del Perú” (UCAP), tanto en temas de representatividad como en la mejora de los servicios que ofrece. La asociación, foco de la tesis, se creó en el año 2013 con Diego Rodríguez Mondragón a la cabeza. La idea surge como iniciativa de un pequeño grupo de amigos, los cuales estaban convencidos de que la promoción y el crecimiento del segmento tendría mejores resultados si se llevaban a cabo de manera conjunta. Poco a poco, la asociación fue creciendo y en la actualidad cuentan con más de 27 cervecerías y 5 “homebrewers” (cerveceros caseros) asociados en todo el Perú, según Silvia De Tomás, persona encargada de las gestiones de la UCAP. Según Diego Rodríguez (presidente de la UCAP), el segmento de cerveza artesanal local posee una participación de mercado menor al 1%; sin embargo, los índices de crecimiento muestran una tendencia positiva. La necesidad de elaborar la presente tesis nace de la oportunidad que brinda este nuevo modelo de negocio (cervecerías artesanales). Estamos frente a un segmento que ofrece un bien con una propuesta de valor que se basa en la experiencia de compra y consumo; apelando a la calidad por encima de la cantidad. La generación del conocimiento y las experiencias compartidas, son las principales fortalezas del segmento, brindando la oportunidad de aplicar los conocimientos adquiridos durante la carrera de gestión; con el fin de lograr encontrar una ruta que contribuya al crecimiento del segmento y al fortalecimiento de este tipo de empresas. La presente tesis se ha desarrollado de acuerdo con las pautas de la investigación acción. La principal fuente de información primaria son las entrevistas. Estas fueron aplicadas a la asociación, actores relacionados, expertos en investigación y analistas de mercado. Además, se utilizó el método de observación; principalmente, para conocer la forma de pensar y trabajar de los cerveceros artesanales y del comportamiento del consumidor en los puntos de venta. La estructura de la tesis se desarrolla en seis capítulos, los cuales se detallan a continuación. El primer capítulo versará sobre el planteamiento de los problemas de investigación, antecedentes, objetivos del proyecto y la justificación. El segundo capítulo presenta el marco metodológico, donde se detalla el tipo de estudio realizado, así como las herramientas de recopilación de información utilizadas durante el desarrollo de la tesis. Finalmente se delimita el alcance de la propuesta a elaborar. En el tercer capítulo, se expone el marco contextual. En primer lugar, se describe la cerveza, su historia y los tipos de cerveza. Luego se realiza una aproximación a la situación actual del segmento cervecero artesanal a nivel mundial. Del mismo modo, se realizó un Benchmarking sobre asociaciones de cerveza artesanal en el mundo, lo que permitirá conocer qué actividades y estrategias se están llevando a cabo en otros países. Finalmente, se detalla la situación actual del segmento de cerveza artesanal en el Perú. El cuarto capítulo describe el marco teórico sobre el cual se desarrollaron los conceptos de asociatividad y planeamiento estratégico. En el quinto capítulo se desarrolla el Planeamiento Estratégico para la UCAP. Finalmente, en el sexto capítulo, se presentan las conclusiones finales de la tesis. Del mismo modo, se proponen ciertas recomendaciones que la asociación debería implementar para lograr mejores resultados en el sector al cual pertenecen.

Sugarcane has an importance in Brazil due to sugar and biofuel production. Considering this aspect, there is basic research being done in order to understand its physiology to improve production. The aim of this research is the Base Excision Repair pathway, in special the enzyme MUTM DNA-glycosylase (formamidopyrimidine) which recognizes oxidized guanine in DNA. The sugarcane scMUTM genes were analyzed using four BACs (Bacterial Artificial Chromosome) from a sugarcane genomic library from R570 cultivar. The resulted showed the presence in the region that had homology to scMUTM the presence of transposable elements. Comparing the similarity, it was observed a highest similarity to Sorghum bicolor sequence, both nucleotide and peptide sequences. Furthermore, promoter regions from MUTM genes in some grass showed different cis-regulatory elements, among which, most were related to oxidative stress, suggesting a gene regulation by oxidative stress
A cana-de-a??car ? uma das principais culturas brasileiras e importante, principalmente, pela produ??o de a??car e biocombust?vel. Por isso, manter a qualidade das cultivares desta esp?cie tornou-se alvo das pesquisas envolvendo gen?tica e bioqu?mica moleculares. Um dos objetivos destas pesquisas ? descobrir informa??es ?teis sobre o material gen?tico que as cultivares da cana-de-a??car possuem, para utiliz?-las como ferramentas no melhoramento contra intemp?ries que afetam sua produ??o, muitas vezes, de forma dr?stica. O foco deste trabalho ? a via de reparo de DNA conhecida por Reparo por Excis?o de Base, mais precisamente, a enzima DNA-glicosilase MUTM (formamidopirimidina-DNA-glicosilase), a qual reconhece e repara guaninas oxidadas no DNA. A caracteriza??o dos genes MUTM da cana-de-a??car foi realizada a partir das an?lises de quatro BACs (Bacterial Artificial Chromosome) de uma biblioteca gen?mica da cultivar R570. Os resultados obtidos dos alinhamentos mostraram a presen?a marcante de elementos de transposi??o. Al?m disso, foi verificado que os genes MUTM foram altamente similares aos de Sorghum bicolor, tanto em sequ?ncias nucleot?dicas e pept?dicas, como na estrutura g?nica. Esta dissertação tem por objetivo analisar a implantação de um modelo de gestão em operações baseado na teoria das restrições na UCAR Produtos de Carbono S/A. Trata-se de um estudo de caso, onde, após uma síntese teórica dos princípios gerais da teoria das restrições, faz-se uma avaliação dos resultados obtidos e das dificuldades encontradas na área de operações. Através de aplicação de questionário e suportado por análises de indicadores obtidos nos quatro anos em que vem sendo implementada, conclui-se que a teoria das restrições facilita a gestão, tendo ajudado à obtenção de resultados crescentes nas operações da UCAR, principalmente nos últimos dois anos da implantação. Sendo um processo ainda em andamento, é visualizado pelos gestores como de grande potencial, capaz de trazer resultados significativos. Apesar desta conclusão positiva, alguns desvios foram detectados. Desta forma, por se tratar de uma dissertação de mestrado profissional, também contribui para apontar deficiências encontradas durante a implementação, direcionando algumas possíveis soluções para as etapas seguintes.

ABSTRACT The techniques and approaches within geoinformatics and data science rely on the effective coupling of supporting infrastructures and institutions. Without underlying infrastructures for data discovery, analysis, management, distribution, and preservation, new computational techniques wither on the vine for lack of input or remain isolated as niche tools that miss broader potential audiences. Likewise, without supporting institutions that enable governance of policies and finances, coordination of stakeholders, and validation of new knowledge and tools, technological advances become detached from the people and organizations that operate and use them. This paper centers on a case study of work within the National Center for Atmospheric Research (NCAR) and University Corporation for Atmospheric Research (UCAR) to develop effective systems and processes for research data curation, access, discovery, and preservation. By emphasizing iterative alignment of institutional work (policies, intermediaries, governance processes, routines, and financial instruments) and infrastructural work (data storage systems, repositories, tools, and interfaces), balanced progress has been made toward developing solutions to gaps in organizational data services.
