Recentlyμopioid receptor (MOR) has been shown to be closely associated with depression. Here we investigated the action of Shuyu, a Chinese herbal prescription, on repeated restraint stress induced depression-like rats, with specific attention to the role of MOR and the related signal cascade. Our results showed that repeated restraint stress caused significant depressive-like behaviors, as evidenced by reduced body weight gain, prolonged duration of immobility in forced swimming test, and decreased number of square-crossings and rearings in open field test. The stress-induced depression-like behaviors were relieved by Shuyu, which was accompanied by decreased expression of MOR in hippocampus. Furthermore, Shuyu upregulated BDNF protein expression, restored the activity of CREB, and stimulated MEK and ERK phosphorylation in hippocampus of stressed rats. More importantly, MOR is involved in the effects of Shuyu on these depression-related signals, as they can be strengthened by MOR antagonist CTAP. Collectively, these data indicated that the antidepressant-like properties of Shuyu are associated with MOR and the corresponding CREB, BDNF, MEK, and ERK signal pathway. Our study supports clinical use of Shuyu as an effective treatment of depression and also suggests that MOR might be a target for treatment of depression and developing novel antidepressants.

Shuni virus (SHUV) is a neglected teratogenic and neurotropic orthobunyavirus that was discovered in the 1960s in Nigeria and was subsequently detected in South Africa, Zimbabwe, and Israel. The virus was isolated from field-collected biting midges and mosquitoes and shown to disseminate efficiently in laboratory-reared biting midges, suggesting that members of the families Culicidae and Ceratopogonidae may function as vectors. SHUV infections have been associated with severe neurological disease in horses, a variety of wildlife species, and domesticated ruminants. SHUV infection of ruminants is additionally associated with abortion, stillbirth, and congenital malformations. The detection of antibodies in human sera also suggests that the virus may have zoonotic potential. To understand how SHUV crosses the ruminant placenta, we here infected pregnant ewes and subsequently performed detailed clinical- and histopathological examination of placental tissue. We found that SHUV targets both maternal epithelial cells and fetal trophoblasts, that together form the maternal-fetal interface of the ovine placenta. Experiments with human placental explants, furthermore, revealed replication of SHUV in syncytiotrophoblasts, which are generally highly resistant to virus infections. Our findings provide novel insights into vertical transmission of SHUV in sheep and call for research on the potential risk of SHUV infection during human pregnancies.

The effects of the Shuyu capsule on5-HT3ARand5-HT3BRexpression in a rat model of premenstrual syndrome (PMS) depression and on5-HT3ARand5-HT3BRexpression and hippocampal neuron 5-HT3channel current were investigated, to elucidate its mechanism of action against PMS depression. PMS depression model rats were divided into depression and Shuyu- and fluoxetine-treated groups, which were compared to control rats for frontal lobe and hippocampal5-HT3ARand5-HT3BRexpression and behavior. The depressed model rats displayed symptoms of depression, which were reduced in treated and normal control rats. Frontal lobe and hippocampal5-HT3ARand5-HT3BRlevels were significantly higher in the model versus the control group and were significantly lower in the Shuyu group. As compared to control rats, the 5-HT3R channel current in the model group was significantly higher; the 5-HT3R channel current in hippocampal neurons treated with serum from Shuyu group rats was significantly lower than that in those treated with model group serum. Thus, PMS depression may be related to5-HT3ARand5-HT3BRexpression and increased 5-HT3channel current. Shuyu capsules rectified abnormal5-HT3ARand5-HT3BRexpression and 5-HT3channel current changes in a rat model; this finding may provide insight into treating PMS depression.

The genus Orthobunyavirus (family Peribunyaviridae, order Bunyavirales) comprises over 170 named mosquito- and midge-borne viruses, several of which cause severe disease in animals or humans. Their three-segmented genomes enable reassortment with related viruses, which may result in novel viruses with altered host or tissue tropism and virulence. One such reassortant, Schmallenberg virus (SBV), emerged in north-western Europe in 2011. Shuni virus (SHUV) is an orthobunyavirus related to SBV that is associated with neurological disease in horses in southern Africa and recently caused an outbreak manifesting with neurological disease and birth defects among ruminants in Israel. The zoonotic potential of SHUV was recently underscored by its association with neurological disease in humans. We here report a reverse genetics system for SHUV and provide first evidence that the non-structural (NSs) protein of SHUV functions as an antagonist of host innate immune responses. We furthermore report the rescue of a reassortant containing the L and S segments of SBV and the M segment of SHUV. This novel reverse genetics system can now be used to study SHUV virulence and tropism, and to elucidate the molecular mechanisms that drive reassortment events.

The Shuni virus (SHUV) causes an endemic viral infection in Israel and South Africa. It belongs to the Simbu serogroup within the order Bunyavirales, family Peribunyaviridae, genus Orthobunyavirus. Recently, it has been identified in aborted cases of domestic ruminants, young cattle and horses manifesting neural signs and acute death, symptomatic cows, and in carcasses of wild animals. Moreover, SHUV was isolated and identified in humans. In this study, we describe clinical cases of SHUV infection in Israeli domestic ruminants in 2020–2021, which represented clinical manifestations of simbuviral infection including abortions, a neural lethal case in a fattening calf, and an acute symptomatic case in a beef cow. In all cases, SHUV was confirmed by complete or partial viral genome sequencing. There is a significant difference of M and L segments of the novel strains compared with those of all known SHUV strains, while the S segments have more than 99% nucleotide (nt) identity with Israeli and African “Israeli-like” strains previously circulated in 2014–2019. This indicates a reassortment origin of the strain. At the same time, M and S segment nt sequences showed about 98–99% nt identity with some South African strains collected in 2016–2018. Nevertheless, the viral origin and the geographical place of the reassortment stayed unknown.

The Orthobunyavirus genus, family Peribunyaviridae, contains several important emerging and re-emerging arboviruses of veterinary and medical importance. These viruses may cause mild febrile illness, to severe encephalitis, fetal deformity, abortion, hemorrhagic fever and death in humans and/or animals. Shuni virus (SHUV) is a zoonotic arbovirus thought to be transmitted by hematophagous arthropods. It was previously reported in a child in Nigeria in 1966 and horses in Southern Africa in the 1970s and again in 2009, and in humans with neurological signs in 2017. Here we investigated the epidemiology and phylogenetic relationship of SHUV strains detected in horses presenting with febrile and neurological signs in South Africa. In total, 24/1820 (1.3%) horses submitted to the zoonotic arbovirus surveillance program tested positive by real-time reverse transcription (RTPCR) between 2009 and 2019. Cases were detected in all provinces with most occurring in Gauteng (9/24, 37.5%). Neurological signs occurred in 21/24 (87.5%) with a fatality rate of 45.8%. Partial sequencing of the nucleocapsid gene clustered the identified strains with SHUV strains previously identified in South Africa (SA). Full genome sequencing of a neurological case detected in 2016 showed 97.8% similarity to the SHUV SA strain (SAE18/09) and 97.5% with the Nigerian strain and 97.1% to the 2014 Israeli strain. Our findings suggest that SHUV is circulating annually in SA and despite it being relatively rare, it causes severe neurological disease and death in horses.

The Simbu serogroup of orthobunyaviruses includes several pathogens of veterinary importance, among them Schmallenberg virus (SBV), Akabane virus (AKAV) and Shuni virus (SHUV). They infect predominantly ruminants and induce severe congenital malformation. In adult animals, the intra vitam diagnostics by direct virus detection is limited to only a few days due to a short-lived viremia. For surveillance purposes the testing for specific antibodies is a superior approach. However, the serological differentiation is hampered by a considerable extent of cross-reactivity, as viruses were assigned into this serogroup based on antigenic relatedness. Here, we established a glycoprotein Gc-based triplex enzyme-linked immunosorbent assay (ELISA) for the detection and differentiation of antibodies against SBV, AKAV, and SHUV. A total of 477 negative samples of various ruminant species, 238 samples positive for SBV-antibodies, 36 positive for AKAV-antibodies and 53 SHUV antibody-positive samples were tested in comparison to neutralization tests. For the newly developed ELISA, overall diagnostic specificities of 84.56%, 94.68% and 89.39% and sensitivities of 89.08%, 69.44% and 84.91% were calculated for SBV, AKAV and SHUV, respectively, with only slight effects of serological cross-reactivity on the diagnostic specificity. Thus, this test system could be used for serological screening in suspected populations or as additional tool during outbreak investigations.

The aim of this study was to explore the mechanism underlying the antidepression activity of paeoniflorin, the main active ingredient of paeony extract and Shuyu capsules, and determine its effect on the calmodulin/calmodulin-dependent protein kinase II (CaM/CaMKII) signalling pathway and on the possible target, the voltage-gated calcium channel (Cav). Rats at the nonacceptance stage were selected for premenstrual syndrome (PMS) depression modelling. Behavioural assays were used for model testing. Rats were given Shuyu capsules, paeony extract, and bupleurum. Western blot analysis was used to assess the expression levels of calcium voltage-gated channel subunit alpha 1 C (CACNA1C), brain-derived neurotrophic factor, and CaM/CaMKII signalling pathway proteins. Intracellular Ca2+ concentration in CHO cell line was measured using Fluo-4-AM and whole-cell patch clamps. The PMS depression model was successfully established and demonstrated that Shuyu can mitigate depressive behaviour in a rat PMS model. Paeony extract did not affect CACNA1C protein expression in rat hippocampi but did affect Cav1.2-mediated CaM/CaMKII signalling pathways. Paeoniflorin significantly inhibited KCl-induced increases in intracellular Ca2+ concentration and Cav1.2 current density. Further, it may function via the CaM/CaMKII pathway and its downstream signalling molecules by regulating Cav1.2, thus playing an important role in the treatment and alleviation of affective disorders.

A lack of well-distributed, high-resolution records of Late Quaternary Asian palaeomonsoon variability remains an outstanding issue for palaeoclimatologists, and is especially marked in remote regions such as the mountains of southwestern China (Wang et al., 2005). Characterising the nature, timing and magnitude of climate variability in southwestern China is essential for understanding the regional climate as a whole, and the potential social, economic and environmental impacts that may result from Asian monsoon system changes. The NERC-funded research presented in this thesis focuses on a high altitude lake sediment record obtained from Lake Shudu, Yunnan Province, China. The lake is located on the southeastern edge of the Tibetan Plateau. The primary aims of this research were to identify and examine key environmental and climatic shifts which occurred in southwestern China during the Late Pleistocene (Dali) - Early Holocene Period; to examine the possible drivers of these changes; and to compare the findings with other regional proxy records in order to better understand climate dynamics in southwestern China. These aims were chosen in order to test the hypothesis that Late Quaternary millennial to centennial scale climatic and environmental changes in southwestern China were driven by changes in solar insolation and / or glacial climate boundary conditions, characterised by stepwise increases in palaeomonsoon intensity. AMS 14C radiocarbon dates obtained from bulk sediment samples and pollen concentrations indicated that the seven metre core (06SD) that forms the focus of this research spans the last c. 22.6 ka cal. yr BP, making it one of the longest high-resolution Late Quaternary records available for southwestern China. 06SD was examined using a multi-proxy approach incorporating physical, organic and palaeoecological analyses. The record captures the shift from colder, drier Pleistocene (Dali) conditions to warmer, wetter Holocene conditions and is punctuated by two events. The first event, centred at c. 17.3 ka cal. yr BP, possibly represents a phase of warmer and / or wetter conditions in response to rising solar insolation during the deglacial period. The second event, commencing at c. 11.7 ka cal. yr BP, possibly denotes the Pleistocene - Holocene Boundary. Overall, the findings of this research support the view that during the Late Pleistocene, Asian summer monsoon strengthening was non-linear and driven by changes in glacial dynamics and / or solar insolation.

Polyketides are belong to secondary metabolites obtained from bacteria, fungi, plants, mollusks, algae, insects, sponges etc. and contain diverse chemical structure and extensive bioactivity. They have been applied in the field of agriculture, industry and pharmaceuticals such as antibiotics erythromycin and tetracycline used in the human, veterinary antibiotic tylosine, anticancer drug doxorubicin, immunosuppressant rapamycin, or cholesterol-lowering drug lovastatin. Therefore, the discovery and producing of novel polyketides may have a high contribution in the development of new drugs. In order to establish a strategy for heterologous expression of polyketide synthases (PKS) that underline the biosynthesis of polyketide, we choice the PKS genes, pfaA, pfaB, pfaC, pfaD and pfaE, involving EPA biosynthesis in Shewanella oneidensis MR-1. These genes was analyzed by bioinformation to design cloning primers. For cloning EPA PKSs, we extracted genomic DNA and performed long-range PCR to obtain full-length EPA PKS genes. These genes were cloned into pENTER/D-TOPO cloning vector and then used gateway cloning system to construct yeast expression vectors in which harbor GAL1 promoter in their 5’ end and in-frame GFP in their 3’-end and contain ura3, leu, his or trp auxotrophic selection marker, respectively. These EPA PKS expression vectors were transformed into INVSc1 and BJ2168 Saccharomyces cerevisiae by lithium chloride method. After induction, the protein expression of EPA PKSs were analyzed by western blot using GFP antibody, and EPA production was detected by gas chromatography. A Leptosphaeria sp fungus, NTOU806, contains bioactive compounds that inhibited the production of nitric oxide and lipopolysaccharide-induced inflammation and showed low cytotoxicity in RAW264.7 cells. For identification of PKS genes and the biosynthesis of polyketides, we performed whole genome sequencing of NTOU806, finding that NTOU806 contains 10 genes of PKS and 3 genes of nonribosomal peptide synthetase. The PKS genes, not containing intron, of PKS2271, PKS2675, PKS3034A and PKS3034B were performed cloning from genomic DNA of NTOU806 and transformed into S. cerevisiae for expression. Moreover, PKS64 obtained from NTOU2362 were performed large scale culture to purify its polyketide. These yeast transformants were collected culture medium, extracted by ethyl acetate, evaporated to concentrate their products and dissolved by methanol. These collected material were analyzed and purified by high performance liquid chromatography, estimated molecular weight by mass spectrometry, and determined chemical structure by nuclear magnetic resonance. Accroding to analysis of HPLC which shows that gene PKS2271 and PKS2675 can produce new chemical compound. NTOU 2362(PKS 64 gene) can express the new protein by induction of S. cerevisiae BJ2168. We use HPLC analysis to ensure the production of the new compund. after the HPLC analysis,we use MS analysis to analyze the new compound. we found out that the new chemical compoud has the molecular weight 144 now we use the final analysis,NMR,and we found that the formula of the chemical compound is tryptophol.Our results show that PKS genes are successfully expressed in S. cerevisiae, and we establish a reliable and feasible strategy to purify and identify polyketides from heterologous expression.

In the midst of Nativist literary movement in 1970s, the Notes of Daimashuzu series, written by essayist and poet Zhang Tuo-Wu (1928-), has gained its popularity in literary market that was reaching its popularity. This presenting thesis would investigate this literary phenomenon in terms of literary field, genre aesthetics and literary history, and tries to explicate what such phenomenon brings to literary history of Taiwan. Zhang retired from military service in 1973, and after the attack of stroke, Zhang started to write biographical essays under the encouragement of his writer friends. Those essays were published on Zhonghua Wenyi and related magazines, and he collected his such writings, spanning 6 years, into Daimashuzu series. In the first section of this presenting thesis, I would focus on Zhang’s position in the literary field in 1970s, and firstly indicate that the genre shift in Zhang’s writing, from poetry to essay, is not only the effect of his change in aesthetic ideology, but as well an action based on his identity as “a military writer not in military service,” in which he tries to extend and transform his cultural capital he gained in his military life. What’s more, his essay writing can be further analyzed under the negotiating history of post-war literary essay and the geopolitical dilemma of Taiwan, and this section would conclude that the 1970s is in effect an little-studied decade of essay-ology, when a new paradigm of essay writing started shaping, effecting military writers changing their topics from concerns of the nation to reflection of the self. Zhang’s essays, as I would argue in the second section, count as a genre of multiplicity. This characteristic relates to the problem of its medium, which is situated between magazine columns and private notes, its genre aesthetics, oscillating from poetry to essay, and how the generation of Zhang’s writing style is affected by his experiences in broadcast industry. Such multilayered characteristics of Daimashuzu series, as this section argues, is a response the modernist aesthetic heritage from the 1960s. Also, by investigating into his essays in light of recent studies on representation of body in literature, this section also indicates that in his essays, the reinforced verb forms act as resistance against the disciplinary power imposing on individuals and genres, and that his essays are unconventional “motivational” narratives in light of the presence of his identity of disability and the characteristics of his essays mentioned above. In the final chapter, I would like to extend my discussion to arguments over military writings in literary history. The debate around Dabing Wenxue (military writings) in the 1970s, as this chapter would argue, indicates the authority’s attempt to reform the literary order in military system, but the attempt faltered owing to lacking followers and theoretical bases, and the discussion then turned toward writings by old solders. However, Zhang’s writings, as the argument shows, stands between the two literary category, of the military literary order and the nativist literature, and Zhang’s style actually shows a strand connecting the early anti-communist nostalgia with more recent trend of “returning to reality,” which resonates to the paradigm set by the nativist literary movements in the 1970s. Zhang’s Notes of Daimashuzu series precede its age. The Dabing Wenxue debate his writings bring out actually counts as the momentary recovery of the military literary order; his writings also inherit the literary tradition (anti-communist nostalgic writings) and anticipate literary phenomena (publications of biographies and memoirs of old soldiers). Investigating Zhang Tuo-wu and his writings in terms of literary fields, genre aesthetics and literary history helps not only to analyze Zhang’s own works, but as well to understand the meaning of similar Wai-sheng old soldier writers in literary history.

本文採取道教文獻研究方法,以蔣叔輿(1162-1223)所撰的《無上黃籙大齋立成儀》為例子,探討南宋道教黃籙齋儀的發展與變革。
黃籙齋儀是具有久遠歷史發展之道教科儀,其儀式傳統可接源六朝道教科範。蔣叔輿承其師留用光(?-1206)的啟導,從六朝靈寶經典中編修齋儀,其中《金籙簡文》和《黃籙簡文》的科範更是蔣氏整理「古法」黃籙齋儀時最重要的參考;而內容則以懺謝為中心。
另一方面,在一個新科儀、新法術傳統出現之變革時代,南宋黃籙齋也不可避免地出現了一些新變革,其中最明顯是文書系統和薦拔度亡儀式的加入。
黃籙齋之文書系統,是一個涉及「文書」、「天上官署」、「印篆」、「法」、「籙」的龐大系統;這是吸收了世俗行政文書之應用,亦是道教科儀能區別於一般民間祭祀的重要特點。另各法派之文書系統、印篆運用、對行儀者法籙要求也各成一系,這亦是讓我們判別不同法派齋儀的重要依據。
此外,南宋黃籙齋晚上新增的度亡儀式,包括了破獄、召魂、沐浴、設斛、鍊度、傳戒環節,並會分正薦亡魂、普薦幽魂兩線並行,而且兩種薦度的重點也不一樣。對比「古法」黃籙齋儀,這些新儀式更著重採用行符、存想、唸呪的法術來薦拔度亡。然而通過對這些法術行持的分析,又會發展其源與六朝道教經科有不少聯繫。
南宋黃籙齋儀一方面延續了六朝靈寶黃籙齋儀懺謝的傳統,另一方面又能確保齋儀能從新法術中吸取養份,以更有效而全面地拔度亡魂。《無上黃籙大齋立成儀》的黃籙齋儀同時存在著新、古兩類儀式;這種試圖於新法術和舊傳統中以爭取平衡,只怕亦是南宋黃籙齋在儀式變革的一種體現。
This thesis adopts the study of Daoist liturgical texts, based on Standardized Rituals of the Supreme Yellow Register Retreat (WushangHuanglu dazhai licheng yi 無上黃籙大齋立成儀) written by Jiang Shuyu 蔣叔輿 (1162-1223), to investigate the development and transformation of rituals of the Yellow Register Retreat (Huanglu zhai 黃籙齋).
The thesis attempts to demonstrate that rituals of the Huanglu zhai had a long historical Daoist ritual background. The rituals could be tracked back to the liturgies of Six Dynasties. Jiang Shuyu was inspired by his teacher Liu Yongguang 留用光 (?-1206) to compile the rituals of Retreat (zhai 齋) from Lingbao 靈寶 scriptures. Among the scriptures, Jinlu jinwen 金籙簡文 and Huanglu jinwen 黃籙簡文 were the most important references for Jiang to compile "old ways"(gufa 古法). The central idea of them is confession and apology (chanxie 懺謝).
It was an era with the transformation of new rites, the Huanglu zhai in Southern Song unavoidably brought in some reforms. The most obvious two examples were the application of the system of written-documents (wenshu 文書) and the ritual of salvation of souls (duwang 度亡).
The system of written-documents in the Huanglu zhai is enormous and it involved written-documents, supernal offices (guanshu 官署), stamps (yin 印), rites (fa 法) and register (lu 籙). It absorbed the administrative documents system. And it was a crucial differentiation between Daoist ritual and folk sacrifice. Different rites adopted different systems of written-documents, different use of seals and different requirements on the ritual master. These are important evidences for us to differentiate various traditions of the rituals of zhai.
Besides that, the Huanglu zhai of Southern Song introduced the ritual of salvation of souls at night, including breaking the hell, summoning the souls, bathing, feeding, refining and teaching the precepts. They were divided into two parallel lines, the salvation of the ancestors and the universal salvation of orphan souls, and their focus were different. Comparing to Lingbao "old ways", these new salvation of souls rituals valued more on sending talismans (fu 符), visualization (cunxiang 存想), and chanting incantations (nianzhou 唸呪). And through analyzing these practices, we discovered that the origin has many connections with the scriptures and liturgies of Six Dynasties.
The ritual of Huanglu zhai, on the one hand continued the practice of confession and apology of LingbaoHuanglu zhai ritual in Six Dynasties, on the other hand it ensured the ritual could absorb new elements from the new rites in order to save the souls effectively and thoroughly. The ritual of Huanglu zhai in WushangHuanglu dazhai licheng yi included both old and new rituals. It tried to strike a balance between the new practice and the old tradition. That also demonstrated the transformation of Huanglu zhai in the Southern Song Dynasties.
