Resumo: Todos os dias milhares de cirurgias cardíacas envolvendo circulação extracorpórea (CEC) são realizadas ao redor do mundo, mas a técnica ainda apresenta problemas relacionados à segurança. Um dos principais tipos de acidentes que podem ocorrer em circulação extracorpórea envolve os oxigenadores e consiste no esvaziamento rápido do reservatório de sangue sem que o perfusionista (operador de CEC) perceba, que provoca a entrada maciça de ar no paciente (embolia aérea maciça), podendo resultar em seqüelas graves ou até mesmo em sua morte. Este acidente é mais comum quando utilizada a bomba de roletes, sendo raros os relatos de casos de embolia aérea maciça com bomba centrífuga. Procurando aumentar a segurança do procedimento de CEC em relação à prevenção deste tipo de acidente, foi desenvolvido o SACEC (Sistema de Apoio à Circulação Extracorpórea), que opera juntamente com uma bomba de roletes e oxigenadores de bolha ou membrana para utilização em adultos. o SACEC é um equipamento microcontrolado que, a partir da medição discreta do nível de sangue no reservatório do oxigenador (com resolução de 100 ml) utilizando um transdutor ultra-sônico de 40 KHz (também desenvolvido neste trabalho) colocado no topo do reservatório, procura manter o nível de sangue acima do nível mínimo de segurança (400 ml nos oxigenadores para adultos) por meio do controle automático da rotação da bomba sistêmica (desligando a bomba se o nível cai abaixo de 500 ml), ou apenas realiza a monitoração do nível no reservatório e da rotação da bomba... Abstract:Thousands of cardiac surgeries involving extracorporeal circulation are perfonned everyday around the world, but the technique still presents safety problems. Oxygenator accidents are among the major accidents that can occur in extracorporeal circulation, being the most serious the rapid and inadvertent emptying of the blood reservoir, that causes massive infusion of air into the patient (massive air embolism) and can result in serious consequences and even patient death. This accident is most common when roller pump is used, being rare reports of massive air embolism with centrifugal pump. Aiming at increasing the safety related to the prevention of this accident, it has been developed the SACEC (Extracorporeal Circulation Support System), that works together with a roller pump and bubble or membrane oxygenators for adults. SACEC is a microcontroller based equipment that, based on the discrete blood leveI measurement in the oxygenator reservoir (with 100 ml resolution) using a 40 KHz ultrasonic transducer (also developed in this work) placed on the oxygenator top, tries to keep the blood leveI above the minimum safety leveI (400 ml in oxygenators for adults) by means of the automatic control of the systemic pump rotation (turning off the pump if the leveI falls below 500 ml), or only performs the reservoir blood leveI and pump rotation monitoring... Повний текст джерелаThe yolk sac plays an important role in embryonic development of all mammals. Its function is being studied, proving to be one of the initials of haematopoiesis. In the period when the placenta is not real yet formed during embryonic development, the yolk sac is the main source of nutrition of the embryo, providing a place for the transfer and synthesis of proteins. Proteins are molecules that govern all cellular functions and are from a chemical structurally the most complex biological molecules. The proteomics is the large-scale study of proteins in a complex biological sample. The aim of this work was make the analisys of present proteins in the yolk sac of bovine embryos by 2D-PAGE two-dimensional electrophoresis. The gels obtained from 2D-PAGE of protein samples contained in the yolk sac of 37 days of pregnancy had about 1230 proteins, and gels of samples from 23 days of pregnancy had about 970 proteins. The yolk sac of 23 days showed essential proteins differentially expressed in stages of development, as Hemogen, protein expressed in stage, responsible for controlling the proliferation and differentiation of hematopoietic cells, the protein acetylgalactosamine glycoprotein-N-3-beta-galactosyltransferase-1, involved in the process of angiogenesis, trombopoiese homeostasis and development of the kidney, the protein factor chorion-specific transcription (GCMa) transcription factor necessary for the development of the placenta. The yolk sac of 37 days showed essential proteins differentially expressed in stages of development , as apolipoprotein E (APOE), which mediates the binding, intersignaling and catabolism of lipoprotein particles, and can serve as a link to the receptor to LDL (apo B/E) and specific receptors for apo E in the liver tissue embryogenesisThese proteins are of vital importance to the development of the embryo until the formation of the placenta.

Повний текст джерелаNesta dissertação, são apresentados alguns dos algoritmos exactos existentes que permitem resolver o Problema do Saco-mochila com Estrutura em Árvore (TKP) e, em particular, são apresentadas de forma uniforme e completa as técnicas de resolução do TKP recorrendo a algoritmos de programação dinâmica. Posteriormente, o TKP é reformulado e são desenvolvidas duas formulações (SP1 e SP2) deste problema como um Problema de Caminho Mais Curto com Restrições de Capacidade. Em seguida, são desenvolvidas duas formulações estendidas (SP3 e SP4) do TKP como um Problema de Caminho Mais Curto. Para comparar a eficiência destas duas últimas formulações, foi feita uma análise da sua implementação computacional considerando diferentes instâncias do problema. Os resultados mostram que a sua eficiência depende da topologia da árvore. Finalmente, é apresentada uma aplicação do TKP ao Desenho de Redes de Acesso Local de Telecomunicações (LANTD). ABSTRACT: In this dissertation, we survey exact algorithms to solve the Tree Knapsack Problem (TKP) and, in particular, we provide a complete and unified presentation of dynamic programs. Then we present two distinct reformulations of the TKP (SP1 and SP2) as a Capacitated Shortest Path Problem. After that, we extend these formulations and develop another two models (SP3 and SP4) of TKP, as a Shortest Path Problem. In order to show the efficiency of the last two models we conduct a computational analysis that concern different problem instances. Our results show that the efficiency of these formulations depends from the tree topology. Finally, we present an application of the TKP to Local Access Network Telecommunications Design (LANTD).

Повний текст джерелаThe yolk sac is a unique structure of vertebrates that evolved a great diversity of developmental, structural and functional features. It is regarded to have a fundamental role for early embryo survival. Although placentation has been intensely studied in the guinea pig, there are important gaps as in regard to the yolk sac. This research will undergo a detailed morphological and developmental characterization of the yolk sac in Cavia porcellus. Seventeen specimens have been collected at gestational ages from day 12 to 55 and investigated by conventional histology, histochemistry, immunohistochemistry including proliferation marker and transmission electron microscopy. Historical material on earlier stages was used from the Embryological Collection of the Museum of Natural History Berlin, Germany. A temporary parietal yolk sac and yolk sac cavity occurred during the blastocyst implantation. The yolk sac covering the chorioallantoic placenta showed great structural differentiation between day 12 and 18 such as a transition from a pseudostratified epithelium to projections that resemble a tree. A Reicherts membrane was not developed on day 12, but a space filled with extracellular material and vacuoles was present. The membrane was found from day 16 onward and became thick and composed of amorphous and acellular material. The morphological data suggest that a transfer of nutrients from the yolk sac to the fetus was most advanced between 22th and 40th day, especially on the 40th day. In the last third of gestation a decrease in both, placenta and visceral, yolk sacs tissues was observed. In conclusion, the development of the yolk sac in the guinea pig showed similarities to other caviomorphs. Data suggest that the yolk sac in caviomorphs is most important for mid gestation, but not so much for early pregnancy as it is for other vertebrates.
