Background: The PRESENILIN genes (PSEN1, PSEN2) encoding for their respective proteins have critical roles in many aspects of Alzheimer’s disease (AD) pathogenesis. The PS2V transcript of PSEN2 encodes a truncated protein and is upregulated in AD brains; however, its relevance to AD and disease progression remains to be determined. Objective: Assess transcript levels in postmortem AD and non-AD brain tissue and in lymphocytes collected under the Australian Imaging Biomarker and Lifestyle (AIBL) study. Methods: Full length PSEN2 and PS2V transcript levels were assessed by quantitative digital PCR in postmortem brain tissue (frontal cortex and hippocampus) from control, AD, frontotemporal dementia (FTD), and Lewy body dementia (LBD). Transcript levels were also assessed in lymphocytes obtained from the Perth subset of the AIBL study (n = 160). Linear regression analysis was used to assess correlations between transcript copy number and brain volume and neocortical amyloid load. Results: PS2V levels increased in AD postmortem brain but PS2V was also present at significant levels in FTD and LBD brains. PS2V transcript was detected in lymphocytes and PS2V/PSEN2 ratios were increased in mild cognitive impairment (p = 0.024) and AD (p = 0.019) groups compared to control group. Increased ratios were significantly correlated with hippocampal volumes only (n = 62, β= –0.269, p = 0.03). Conclusion: Taken together, these results suggest that PS2V may be a marker of overall neurodegeneration.

ABSTRACT The virulence plasmid of Salmonella enterica serovar Gallinarum-Pullorum (pSPV) but not those of Salmonella enterica serovars Enteritidis (pSEV) and Typhimurium (pSTV) can be readily mobilized by an F or F-like conjugative plasmid. To investigate the reason for the difference, the oriT-traM-traJ-traY-traA-traL regions of the three salmonella virulence plasmids (pSVs) were cloned and their nucleotide and deduced amino acid sequences were examined. The cloned fragments were generally mobilized more readily than the corresponding full-length pSVs, but the recombinant plasmid containing the oriT of pSPV was, as expected, more readily mobilized, with up to 100-fold higher frequency than the recombinant plasmids containing the oriT of the other two pSVs. The nucleotide sequences of the oriT-traM-traJ-traY-traA-traL region of pSEV and pSTV were almost identical (only 4 bp differences), but differed from that of pSPV. Major nucleotide sequence variations were found in traJ, traY, and the Tra protein binding sites sby and sbm. sby of pSPV showed higher similarity than that of pSEV or pSTV to that of the F plasmid. The reverse was true for sbm: similarity was higher with pSEV and pSTV than with pSPV. In the deduced amino acid sequences of the five Tra proteins, major differences were found in TraY: pSEV's TraY was 75 amino acids, pSTV's was 106 amino acids, and pSPV's was 133 amino acids; and there were duplicate consensus βαα fragments in the TraY of pSPV and F plasmid, whereas there was only a single βαα fragment in that of pSEV and pSTV.

Abstract Leaf samples collected from 1,883 peanut plants in 158 fields in 14 counties during July-August, 1990 and 1991, were tested for peanut mottle (PMV), peanut stripe (PStV), peanut stunt (PSV), and tomato spotted wilt (TSWV) viruses by sap inoculations onto indicator plants and/or by enzyme-linked immunosorbent assay (ELISA). Of 889 plants showing virus-like symptoms, 58% were infected with TSWV alone or mixed with PMV or PSV, 36% with PMV alone or mixed with TSWV or PSV, and 5% with PSV alone or mixed with TSWV or PMV. Double infections of PMV with TSWV, PSV with TSWV, and PMV with PSV were detected in 24%, 3%, and 1% of the symptomatic plants. Of 994 apparent asymptomatic plants, 16% were infected with PMV alone or mixed with TSWV, 8% with TSWV alone or mixed with PMV or PSV, and 5% with PSV alone or mixed with TSWV. Double infections of PMV with TSWV and PSV with TSWV were detected in 2% and 1% of the asymptomatic plants. PMV and TSWV were found in at least one field in every county sometime during the two seasons; PSV was found in 13 counties. The most notable difference in virus incidence between the two years was that TSWV was found in 20% of the asymptomatic plants in 1990 as compared to 6% in 1991. PStV was not detected by ELISA in 1990 or 1991, and no reactions suggestive of PStV or any virus other than PMV, PSV, or TSWV were observed on indicator plants used to assay the 1990 collections.

This is the first survey to determine the occurrence, prevalence, and distribution of peanut (Arachis hypogaea) viral diseases in Argentina. It was conducted in the province of Córdoba, which has 92% of the country's peanut production. It included the main peanut viruses Peanut mottle virus (PeMoV), Peanut stripe virus (PStV), Cucumber mosaic virus (CMV), Peanut stunt virus (PSV), Tomato spotted wilt virus (TSWV), and Groundnut ringspot virus (GRSV). Leaf samples from 1,028 individual peanut plants with virus-like symptoms and 986 samples from asymptomatic plants were collected in six counties of Córdoba over 3 years and serologically tested for the presence of viruses. PeMoV was the most frequently detected virus, found in 58.8, 34.2, and 23.4% of samples from the 2003–04, 2004–05, and 2005–06 growing seasons, respectively, and it was found in all sampled counties. Also, it was the only virus detected in asymptomatic plants. Less than 4% of symptomatic plants were infected with CMV or GRSV; 0.5, 3.6, and 2% of samples were positive for CMV; and 0.5, 3.1, and 1.6% were positive for GRSV in the 2003–04, 2004–05 and 2005–06 seasons, respectively. Some mixed infections were found: CMV-PeMoV and GRSV-PeMoV. During this survey, PSV, PStV, and TSWV were not detected in any peanut samples.

Dipositronium, Ps2, was recently prepared [1]. This is significant because: • It is the first laboratory observation of a molecule that contains more than one positron; • It is the most symmetrical molecule possible; • It is the most non-rigid (floppiest) molecule possible; • The interval between the theoretical establishment of its existence [2] and its laboratory observation [1] is inordinately long – 60 years; and • An extension of the technology developed for the Ps2 observation may soon lead to an observation of the Bose-Einstein condensation of positronium and the development of a gamma ray laser. We briefly discuss the symmetry of Ps2 and how an understanding of it will underlie its characterization in the future. Ps2O and CPs2 might be the next two-positron compounds to be prepared and characterized in the laboratory. A discussion of the contrasting eigenstates of these two molecules is given. An understanding of these states is required in order to identify them.

A study of the genetic structure and differentiation of eight populations of Pinus sylvestris L. on the Russian Plain showed that the populations of PsI and PsII (D=0.066) are located at the smallest genetic distance, and between the populations of PsI and PsIV (D=0.308) at the greatest genetic distance. On the dendrogram, the studied populations formed four clusters: PsI and PsII; PsIII and PsIV; PsV and PsVI; PsVII and PsVIII. Analysis of the genetic structure of eight populations of P. sylvestris showed that the expected proportion of heterozygous genotypes (HT) for the total sample was 0.320, the expected proportion of heterozygous genotypes in a single population for all loci (HS) was 0.170, therefore, the population subdivision (GST) was high and amounted to 0.468. The studied populations are highly differentiated, since the interpopulation component accounts for 46.8% of the genetic diversity. In all studied populations, the indicator h has values less than 0.3. An analysis of the fraction of rare alleles showed that the genetic structure is less balanced in the populations PsIII (h=0.254) and PsIV (h=0.273). The most balanced genetic structure in the populations of PsVII (h=0.112) and PsVIII (h=0.127). Data on the genetic structure and differentiation of the common pine populations should be taken into account when developing recommendations for preserving their gene pools.

In prostate cancer (PCa), stromal tissues co-evolve through reciprocal interactions with the tumour to support tumour growth and suppress the immune response. WFDC1/ps20 is a secreted whey acid protein (WAP) four-disulphide core (WFDC) family member highly expressed within the prostate stroma. Ps20 expression has been frequently observed to be down-regulated or lost in cancers including in prostate cancer, and numerous lines of evidence suggest that ps20 has intrinsic growth suppressive function in numerous tumour model systems. However, ps20 remains biochemically uncharacterised and the mechanisms by which it functions are unknown. WFDC1/ps20 is expressed in numerous mRNA and protein isoforms some of which result from proteolytic cleavage. These post-translational modifications affect the ability of the protein to inhibit the proliferation of PCa cells. We demonstrated that ps20 undergoes oligomerisation by transglutaminase, and crosslinking to fibronectin. We show that ps20 is cleaved by cathepsin L, which while failing to abrogate function of the protein, does liberate cross-linked ps20 from solid phase fibronectin. We show that ps20 binds to glycosaminoglycans (GAG) and interacts with cell surfaces in a GAG dependent manner. To further investigate the cellular effects of ps20 we overexpressed ps20 in PCa cell lines. Transgenic overexpression of WFDC1/ps20 reduced proliferation in WPMY-1 cells, and conditioned media (CM) from these cells had potent pro-apoptotic effects on a range of PCa cell lines. Whole genome differential transcriptome analysis of WPMY-1-EV v WPMY-1-ps20FL/WPMY-1-ps20TR cells identified numerous factors, including IL-8, IL-32, COX2, and SerpinF1 to be upregulated in WPMY-1 cells. Addition of a COX2 inhibitor reversed the suppressive effect of WPMY-1-ps20 CM, suggesting that expression of ps20 in the prostate stroma can regulate growth of epithelial and other tissues through the prostaglandin synthase pathway. Lastly, using ps20 overexpression in WPMY-1 prostate stromal cells, we demonstrated that CM from ps20 expressing cells inhibits proliferation of CD4 and CD8 T cells. Using depletion of ps20 from CM we show that suppression is indirectly mediated. We show that ps20 expression in WPMY-1 cell CM inhibits both anti-CD3/28 and IL-7/15 dependent T cell. However no effect on the secretion of IFNγ or expression of common T cell activation markers is observed. We demonstrate that the suppression of T cell proliferation is partly due to enhanced COX-2 activity.

På uppdrag av innovationsföretaget EmpirBus AB skulle en produkt utvecklas. Projektet innefattade utvecklingsprocessen ända ifrån ide till slutprodukt. Syftet var att utveckla ett gränssnitt för kommunikation mellan CAN-bus och USB/PS2 eller RS232. Gränssnittet sammankopplas med ett CAN-bussbaserat elsystem för marina installationer. Produktens uppgift var att utföra bildskärmsväxlingar på informationsskärmar ombord på fartyg. Genom att sända ett kommando till en bildskärmsväxel innehållande information om till vilken bild som skall växlas fram. Projektet gick enligt plan och en färdig produkt blev slutresultatet, benämnd UPS-B Ver. 1.0.

Resumo: Introdução: Durante o desenvolvimento e a vida adulta de um organismo, células troncoindiferenciadas, capazes de dar origem a um ou mais tipos celulares, proliferam e se diferenciam para, tanto produzir os tecidos de um organismo em desenvolvimento, quanto repor células perdidas pelo envelhecimento, doença ou injúria. Essas células residem em um nicho especializado contendo células diferenciadas e elementos responsáveis por manter o estado predominantemente quiescente. Acredita-se que as células tronco da próstata contribuam para o desenvolvimento de câncer através da desregulação dos elementos do nicho. ps20 é um elemento secretado por células musculares lisas da próstata, o qual demonstra capacidade de inibir o crescimento da linhagem de células prostáticas cancerígenas PC3 em cultura. Evidências sugerem uma possível relação entre ps20 com a adesão celular. No entanto, seu funcionamento é pouco conhecido. Objetivos: Analisar o efeito do silenciamento do gene WFDC1/ps20 sobre o comportamento das células p63+ e arquitetura tecidual durante o desenvolvimento prostático pós-natal. Materiais e métodos: Próstatas ventrais de ratos Wistar recém nascidos foram cultivadas sobre membranas flutuantes permeáveis, na presença de RNA de interferência para reduzir a expressão de ps20. Os órgãos foram cultivados por sete dias e em seguida analisados através da contagem da formação de estruturas epiteliais, colorações histológicas, imunohistoquímica e PCR em Tempo Real. Resultados: O silenciamento de WFDC1/ps20 levou a uma redução de 36% na quantidade de mRNA de WFDC1/ps20. Foi observada redução no número de pontas de ductos epiteliais entre o segundo e quarto dia cultura e entre o quarto e sétimo dia. A análise histológica revelou ductos com ramificação reduzida e com as pontas mais dilatadas, em resposta a redução de WFDC1/ps20. Foi observada polarização celular prejudicada e cavitação reduzida. A imunohistoquímica para p63 revelou que os níveis reduzidos de ps20 promoveram a concentração das células p63+ nas pontas dilatadas dos ductos epiteliais. Real Time PCR revelou que o silenciamento do gene WFDC1/ps20 promoveu um aumento de nove vezes na quantidade do mRNA de MMP9.Discussão: O silenciamento de WFDC1/ps20afetou o desenvolvimento prostático com um efeito sem ambiguidade no comportamento das células basais p63+, como representado pelo seu acumulo nas pontas dos ductos e diminuição ao longo dos ductos estendidos. O tratamento também prejudicou a polarização de células luminais e comprometeu a cavitação.O conteúdo elevado de mRNA de MMP9 pode estar relacionado com o acumulo da proteína e a degradação descontrolada da matriz extracelular ao redor pontas dos ductos, levando ao fenótipo dilatado. Como a degradação correta da matriz em locais específicos é requerida para a formação do ponto de ramificação, é sugerido que a ps20 regula o balanço da atividade de MMP9 necessária durante o desenvolvimento prostático. Essa ideia coloca o sistema uPA/uPAR acima da regulação e ativação de MMP9.Conclusões:A redução deWFDC1/ps20comprometeu a morfogênese em ramos, polarização das células epiteliais e cavitação. Esses aspectos estão associados com a desestabilização do comportamento das células p63+, como sugerido pelo seu acumulo nas pontas dilatadas e diminuição do compartimento de células basais ao longo dos ductos estendidos
Abstract: Introduction: During the development and adult life of an organism, undifferentiated stem cells, capable of giving rise to one or more cell types in an adult organism, proliferate and differentiate to both produce the tissues of an developing organism and replace cells lost by aging, disease or injury. These cells reside in a specialized niche containing differentiated cells and elements responsible for maintaining their predominantly quiescent state. Prostate stem cells are believed to contribute to cancer development via deregulation of niche elements. ps20 is a protein secreted by prostate smooth muscle cells which was shown to inhibit the growth of prostatic PC3 cancer cell line in culture. Evidences exist favoring a possible relationship between ps20 and cell adhesion. However its functioning is still poorly known. Objectives: Analyze the effect of the silencing of the WFDC1/ps20 gene in the behavior of p63-positive cells and tissue architecture during the post-natal prostatic development. Materials and methods: Ventral prostate of newborn Wistar rats were cultured on floating permeable membranes, in the presence of small interference RNA to reduce ps20 expression. The organs were cultured for seven days, and then analyzed by counting of epithelial structures formation, histological and immunohistochemical staining and Real Time PCR. Results: The silencing of WFDC1/ps20 led to a 36% reduction in WFDC1/ps20 mRNA amount. It was observed reduction in the number of epithelial duct tips between the second and fourth day of culture, and between the fourth and seventh day. Histological analysis revealed reduced branching and dilated tips in response toWFDC1/ps20knocking down. It was observed hindered polarization and reduced cavitation. Immunohistochemistry for p63 revealed that reduced levels of ps20 promoted the concentration of p63+ cells at the enlarged ductal tips. Real Time PCR revealed that the knocking down the WFDC1/ps20 gene promoted a 9-fold increase in the amount of MMP9 mRNA. Discussion: The silencing of WFDC1/ps20affected prostate development with an unambiguous effect on p63+ cells behavior, as represented by their accumulation at the ductal tips and depletion along the extended ducts. The treatment also hampered the differentiation of luminal cells and compromised cavitation. The increased MMP9 mRNA content might be correlated with the accumulation of the protein and uncontrolled degradation of the extracellular matrix around the duct tips, leading to the dilated phenotype. Since the correct degradation of the matrix in specific sites is required for the branch point formation we suggest that ps20 regulates the balance for MMP9 activity required during prostate development. This idea places the uPA/uPAR system upstream of MMP-9 regulation and activation. Conclusions: The reduction in the WFDC1/ps20expressioncompromised branching morphogenesis, epithelial cell polarization and cavitation. These aspects are associated with a destabilization of p63+cell behavior, as suggested by their accumulation at the dilated tips and depletion from the basal cell compartment along the extended ducts

The present work proposes a methodology to define the electric propulsion system for PSVs. This methodology was applied to a case study: the conceptual design of a PSV for operation at the pre-salt fields at Santos basin. First, four different alternatives of electric propulsion systems for the PSV are presented and sized. The first one has induction motors as main propulsion motors, the second one has synchronous motors as main propulsion motors; the third and fourth alternatives are the same as the first and second, respectively, with a batteries bank connected to the main switchboard. Each of the four arrangements was contemplated with two options for Diesel generators: high speed and medium speed generator sets. The mass, volume, fuel consumption, exhaust gases emissions and reference capital costs for each arrangement are estimated and analyzed. Moreover, an economic analysis through the Net Present Value (NPV) is performed. The methodology ends with the Analytic Hierarchy Process (AHP) to support the decision making procedure. Some of the parameters obtained for each arrangement (mass, volume, fuel consumption, exhaust gases and NPV) are used as criteria and sub-criteria for selection. Two scenarios are evaluated, the first scenario gives more importance to the financial component of the design; the second scenario sets the exhaust gases emissions as the more significant parameter. The results were different, the arrangements 1 and 2 with medium speed Diesel generator sets appear as the most suitable option from the economical point of view; whereas the arrangements with batteries and high speed Diesel gensets are the best options to reduce the exhaust emissions.
O presente trabalho propôs uma metodologia para definir o sistema de propulsão elétrica para PSVs. A metodologia foi aplicada para um caso estudo: o projeto conceitual de um PSV para operar nos campos do pré-sal na Bacia de Santos. Primeiramente, as quatro diferentes alternativas de sistemas de propulsão elétrica para PSV são apresentadas e dimensionadas. A primeira alternativa tem motores de indução como motores de propulsão principal, a segunda alternativa tem motores síncronos como motores de propulsão principal; a terceira e quarta alternativas são as mesmas que a primeira e a segunda, respectivamente, com um banco de baterias conectado ao quadro principal. Cada um dos quatro arranjos foi considerado com duas opções para Diesel geradores: Diesel geradores de alta e média rotação. A massa, volumem, consumo de combustível, emissão de gases e os custos capitais de referência para cada arranjo são estimados e analisados. Adicionalmente, uma análise económica usando o Valor Presente Líquido (VPL) é feita. A metodologia finaliza com o Analytic Hierarchy Process (AHP) para apoiar o processo de escolha de alternativa. Alguns dos parâmetros obtidos para cada arranjo (massa, volumem, consumo de combustível, gases poluentes e o VPL) são utilizados como critérios de seleção. Dois cenários são avaliados, o primeiro cenário dá maior importância à parte financeira do projeto, o segundo cenário estabelece as emissões de gases poluentes como o parâmetro mais significativo. Os resultados foram diferentes, os arranjos 1 e 2 com Diesel geradores de média rotação se apresentam como a opção mais adequada desde o ponto de vista econômico; enquanto os arranjos com baterias e Diesel geradores de alta rotação são a melhor opção para reduzir as emissões de gases poluentes.

Abstract Prior research into Ultra Compact Combustors (UCC) showed an axial length savings compared to traditional gas-turbine combustors. This savings is achieved by swirling the reactants circumferentially in a recessed cavity around the outside diameter of the engine. A similar circumferential combustor is envisioned for a new engine configuration that positions the combustor outboard of a radial compressor and an inflow turbine. This configuration will offer an axial length savings for the entire engine, not just the combustor. The new engine configuration will not utilize a core flow path and thus requires all engine air from the compressor to pass through, or around, the combustor cavity. This report characterizes the cavity flow behavior as the core flow quantity was reduced from 80% of the total engine mass flow rate down to zero, representing the new engine configuration, while maintaining constant cavity mass flow rates. Velocity profiles were obtained with particle-shadow image velocimetry (PSV) in cold flow and with particle streak emission velocimetry (PSEV) in reacting flow experiments. The cold flow results showed that the core flow produced a suction and removed fluid from the circumferential cavity resulting in a lower cavity mass flow rate. This behavior resulted in lower circumferential velocities at higher core flow percentages and the fastest cavity velocity with zero core flow. Reacting flows produced a similar result with the fastest cavity velocities achieved at reduced, but non-zero core flows, and the slowest velocities at the highest and zero core flows. Overall, it was found that there was no negative impact on performance from the removal of the core flow that would prohibit development of the new engine.

We describe the dynamic analysis of a spring-loaded pressure safety valve (PSV) using a moving mesh technique and transient analysis in computational fluid dynamics (CFD). Multiple domains containing pure structural meshes are generated to ensure that the correlative mesh could change properly without negative volumes. With a geometrically accurate CFD model including the PSV and vessel rather than only the PSV, the entire process from valve opening to valve re-closure is presented. A detailed picture of the compressible fluid flowing through the PSV is obtained, including flow features in the very small seat region. In addition, the forces on the disc and its motion are monitored. Results from the model were very useful in investigating the dynamic and fluid characteristics of the PSV. Our practical CFD model has the potential to reduce the costs and risks associated with the development of new pressure safety valve designs. Future work will focus on improving the spring stiffness and seat region to eliminate or reduce vibration during the re-closure process.

As the ultimate protection of a pressure system, pressure safety valves (PSV) can respond in an instable manner such as flutter and chatter which will affect service life, reliability and performance. In order to study the dynamic instability caused by multi-source forces including the flow force, the spring compressing force and the pressure wave force, a more realistic CFD model containing a PSV and different connected pipes as well as the pressure vessel is developed, in which advanced techniques in Fluent such as User Defined Function (UDF) and Dynamic Layering method are combined to allow the PSV to operate. Based on this model, the process of the valve’s opening and reclosing is monitored to examine the influence of design parameters on the dynamic instability of the PSV. Specifically, the propagation of pressure waves along the connecting pipes is successfully captured, which is of great help to explain the instability mechanism and optimize the design and setup of pressure relief systems.
