Wheat powdery mildew, caused by Blumeria graminis f. sp. tritici (an obligate biotrophic pathogen) is a worldwide threat to wheat production that occurs over a wide geographic area in China. For monitoring genetic variation and virulence structure of Blumeria graminis f. sp. tritici in Liaoning, Heilongjiang, and Sichuan in 2015, 31 wheat lines with known Powdery mildew resistance genes and 2 EST-SSR markers were used to characterize the virulence and genetic diversity. Results indicated that 90% of all isolates were virulent on Pm3c, Pm3e, Pm3f, Pm4a, Pm5, Pm6 (Timgalen), Pm7, Pm16, Pm19, and Pm1 + 2 + 9 and 62.6% to 89.9% of isolates were virulent on Pm3a, Pm3b, Pm3d, Pm4b, Pm6 (Coker747), Pm8, Pm17, Pm20, Pm23, Pm30, Pm4 + 8, Pm5 + 6, Pm4b + mli, Pm2 + mld, Pm4 + 2X, Pm2 + 6. The Pm13 and PmXBD genes were effective against most collected isolates from Liaoning and Heilongjiang Provinces. Only Pm21 exhibited an immune infection response to all isolates. Furthermore, closely related isolates within each region were distinguished by cluster analyses using EST-SSR representing some gene exchanges and genetic relationships between the flora in Northeast China (Liaoning, Heilongjiang) and Sichuan. Only 45% of the isolates tested show a clear correlation between EST-SSR genetic polymorphisms and the frequency of virulence gene data. However, the EST-SSR polymorphism of isolated genes did not correspond to the virulence diversity of isolates in the single-gene lineage identification of hosts.

Calcium wave pacemakers in fertilized eggs of ascidians and mouse are associated with accumulations of cortical endoplasmic reticulum in the vegetal hemisphere. In ascidians, two distinct pacemakers (PM1 and PM2) generate two series of calcium waves necessary to drive meiosis I and II. Pacemaker PM2 is stably localized in a cortical ER accumulation situated in the vegetal contraction pole. We now find that pacemaker PM1 is situated in a cortical ER-rich domain that forms around the sperm aster and moves with it during the calcium-dependant cortical contraction triggered by the fertilizing sperm. Global elevations of inositol (1,4,5)-trisphosphate (Ins(1,4,5)P3) levels produced by caged Ins(1,4,5)P3 or caged glycero-myo-PtdIns(4,5)P2 photolysis reveal that the cortex of the animal hemisphere, also rich in ER-clusters, is the cellular region most sensitive to Ins(1,4,5)P3 and acts as a third type of pacemaker (PM3). Surprisingly, the artificial pacemaker PM3 predominates over the natural pacemaker PM2, located at the opposite pole. Microtubule depolymerization does not alter the activity nor the location of the three pacemakers. By contrast, blocking the acto-myosin driven cortical contraction with cytochalasin B prevents PM1 migration and inhibits PM2 activity. PM3, however, is insensitive to cytochalasin B. Our experiments suggest that the three distinct calcium wave pacemakers are probably regulated by different spatiotemporal variations in Ins(1,4,5)P3 concentration. In particular, the activity of the natural calcium wave pacemakers PM1 and PM2 depends on the apposition of a cortical ER-rich domain to a source of Ins(1,4,5)P3 production in the cortex. Background: Percutaneous intramyocardial (PIM) septal radiofrequency ablation (SRA) is a novel treatment approach for hypertrophic obstructive cardiomyopathy patients, but there has been lack of a large animal model to study PIM-SRA. We aimed to validate the long-term safety and efficacy of PIM-SRA and to observe pathological changes of the ablated interventricular septum (IVS) in a healthy sheep model. Methods and Results: Twelve sheep were randomized to the PIM-SRA group (n = 6) and the sham group (n = 6). In the PIM-SRA group, a radiofrequency (RF) electrode was inserted into the IVS with a maximum power of 80 W for 5 min. In the sham group, the RF electrode tip was positioned in the IVS segment but without RF power delivery. Septal hypokinesis was seen in all PIM-SRA group animals immediately after the procedure; the systolic wall thickening rate and motion amplitude of the ablated region decreased (p < 0.01), and the diastolic IVS thickness also decreased significantly over time (p < 0.01). ECG showed that all the sheep had normal sinus rhythm during the follow-up. Pathological examinations revealed scar tissue in the ablated region as expected. Conclusions: PIM-SRA produced precisely ablated myocardial tissue, reduced the IVS thickness significantly, preserved the global LV function, and avoided the incidence of conduction system damage in the long term. PIM-SRA was found to be a safe and effective minimally invasive septal reduction therapy.

Objective: To identify the best method among the radiologic, laparoscopic and laparotomic scoring assessment to predict the outcomes of cytoreductive surgery in patients with advanced ovarian cancer (AOC). Methods: Patients with AOC who underwent pre-operative computed tomography (CT) scan, laparoscopic evaluation, and cytoreductive surgery between August 2016 and February 2021 were retrospectively reviewed. Predictive Index (PI) score and Peritoneal Cancer Index (PCI) scores were used to estimate the tumor load and predict the residual disease in the primary debulking surgery (PDS) and interval debulking surgery (IDS) after neoadjuvant chemotherapy (NACT) groups. Concordance percentages were calculated between the two scores. Results: Among 100 eligible patients, 69 underwent PDS, and 31 underwent NACT and IDS. Complete cytoreduction was achieved in 72.5% of patients in the PDS group and 77.4% in the IDS. In patients undergoing PDS, the laparoscopic PI and the laparotomic PCI had the best accuracies for complete cytoreduction (R0) [area under the curve (AUC) = 0.78 and AUC = 0.83, respectively]. In the IDS group, the laparotomic PI (AUC = 0.75) and the laparoscopic PCI (AUC= 0.87) were associated with the best accuracy in R0 prediction. Furthermore, radiological assessment, through PI and PCI, was associated with the worst accuracy in either PDS or IDS group (PI in PDS: AUC = 0.64; PCI in PDS: AUC = 0.64; PI in IDS: AUC = 0.46; PCI in IDS: AUC = 0.47). Conclusion: The laparoscopic score assessment had high accuracy for optimal cytoreduction in AOC patients undergoing PDS or IDS. Integrating diagnostic laparoscopy in the decision-making algorithm to accurately triage AOC patients to different treatment strategies seems necessary.

ObjectivesLower limb posterior chain injury (PCI) is common among athletic populations, with multifactorial risk factors including age, previous injury, strength measurements, range of motion and training load. Biomechanics are commonly considered in the prevention and rehabilitation of PCI by performance staff. However, there is no documented testing method to assess for associations between biomechanics and PCI. The aim of this study was to investigate whether there is an association between an easily applicable, novel biomechanical assessment tool and PCI.MethodsFifty male elite-level rugby union athletes (age 22.83±5.08) participating in the highest tier of England were tested at the start of the 2019 preseason period and PCIs (N=48) were recorded over the 2019/2020 playing season. Participants’ biomechanics were analysed using two-dimensional video analysis against an injury risk score (IRS) system in the performance of the combined movement—prone hip extension and knee flexion. Participants’ biomechanics in carrying out this movement were scored against the 10-point IRS, where the more compensatory movement recorded sees an increase in an individual’s IRS. Participants’ IRS was then compared against the number of PCIs sustained and Spearman’s correlation coefficient was used for statistical analysis.ResultsThere is a significant association between IRS and PCI (R=0.542, p<0.001). Linear regression demonstrated that an increase in 1 in IRS was associated with a 35% increase in PCI incidence (R²=0.346).ConclusionA significance between the IRS and PCI provides preliminary support for its use as an injury risk assessment tool.

This study evaluated the role of H-K-adenosinetriphosphatase (H-K-ATPase) with chronic metabolic acidosis (CMA) in intercalated cells (ICs) of rabbit cortical collecting duct (CCD). CMA was induced by replacing drinking water with 75 mM NH4Cl in 5% sucrose for 10-14 days. CCDs isolated from CMA and control rabbits were split open and exposed to the intracellular pH (pHi) indicator 2',7'-bis(carboxyethyl)-5(6)-carboxyfluorescein. In N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid-buffered solutions, the resting pHi in ICs was similar for both groups. K-dependent pHi recovery (5 mM K, 140 mM N-methyl-D-glucamine) was monitored in response to a pulse of NH4Cl (10 mM). The K-dependent pHi recovery rate was threefold higher in CMAICs compared with controls and was abolished with the gastric H-K-ATPase inhibitor, Sch-28080 (10(-5) M). Polarity of the H-K-ATPase was studied in microperfused CMA and control CCDs. Luminal K-dependent pHi recovery was monitored in response to an acute pulse of NH4Cl in individual peanut lectin agglutinin (PNA)-binding ICs. The apical Sch-28080-inhibitable K-dependent pHi recovery rate was significantly greater in CMA ICs than control ICs. In summary, CMA enhances functional activity of an apical H-K-ATPase in PNA-binding ICs of rabbit CCD.

Secondary injuries are associated with bad outcomes in the case of severe traumatic brain injury (sTBI). Patients with a Glasgow Coma Scale (GCS) < 9 should undergo pre-hospital intubation (PHI). There is controversy about whether PHI is beneficial. The aim of this study was to estimate the effect of PHI in patients after sTBI. A multicenter, prospective cohort study was performed in Switzerland, including 832 adults with sTBI. Outcomes were death and impaired consciousness at 14 days. Associations between risk factors and outcomes were assessed with univariate and multivariate Cox models for survival, and univariate and multivariate regression models for impaired consciousness. Potential risk factors were age, GCS on scene, pupil reaction, Injury Severity Score (ISS), PHI, oxygen administration, and type of admission to trauma center. Age, GCS on scene < 9, abnormal pupil reaction and ISS ≥ 25 were associated with mortality. GCS < 9 and ISS ≥ 25 were correlated with impaired consciousness. PHI was overall not associated with short-term mortality and consciousness. However, there was a significative interaction with PHI and major trauma. PHI improves outcome from patients with sTBI and an ISS ≥ 25.

Abstract Paediatric brain tumour survivors (PBTS) experience slower information processing speed (IPS) that contributes to difficulty performing tasks of minimal (MC) and greater complexity (GC), and is related to aberrant neural communication. It is still unknown whether deficient IPS exists during increasing complexity. We aim to determine if PBTS experience deficient IPS and neural communication relative to typically developing children (TDC) during an increasingly complex visual-motor reaction time (RT) task. During magnetoencephalography recording, participants (n=58, 12.69 ±3.24 years) pressed a button with their left or right thumb after an arrow pointing in the corresponding direction appeared on a screen. During two MC conditions, the arrow pointed in a single direction. During a GC condition, the arrow alternated direction randomly. Mean RT &gt;3SD and signal artifacts were removed prior to analyses. The phase lag index (PLI) estimated neural communication between 90 cortical sources. Linear regression and Network Based Statistics assessed group differences in mean RT and the PLI. PBTS demonstrated increased RT relative to TDC during the GC condition (p=0.04, MPBTS=354.00s, MTDC=326.00s). Group differences in mean RT during MC conditions and the PLI during all conditions were not detected (p&gt;0.05). These results suggest PBTS experience slower IPS during GC. Reduced IPS is thought to contribute to difficulty recruiting cognitive resources needed to perform more complex tasks. Subtle deficits in neural communication may underlie slower IPS. The weighted PLI is superior to the PLI when estimating small differences in neural communication. We will now use the weighted PLI to assess task-related neural communication.

Penelitian ini merupakan sebuah Penelitian Tindakan Kelas (PTK) yang bertujuan: 1) meningkatkan aktivitas siswa kelas XI IPS dengan menerapkan model Problem Based Instruction (PBI), 2) respon, kendala, dan cara mengatasi model Problem Based Instruction (PBI) di Kelas XI IPS, dan 3) meningkatkan hasil belajar siswa Kelas XI IPS dengan penerapan model Problem Based Instruction (PBI). Penelitian dilaksanakan sebanyak tiga siklus dengan subyek penelitian siswa kelas XI IPS 3 SMA N 3 Kota Bengkulu. Setiap siklus melalui empat tahap PTK yang meliputi, perencanaan, pelaksanaan, observasi, dan refleksi. Instrumen penelitian ini adalah lembar observasi aktivitas, pedoman wawancara berbasis tugas, dan lembar tes tertulis. Berdasarkan hasil penelitian dapat disimpulkan: 1) Penerapan model PBI di kelas XI IPS dapat meningkatkan aktivitas belajar siswa dengan cara, menjelaskan masalah, mengorganisasikan siswa untuk berdiskusi dengan bantuan LKS, membimbing kegiatan diskusi, memberi kesempatan siswa untuk mempresentasikan hasil dan berpendapat, dan mengevaluasi hasil kegiatan dengan tanya jawab, 2) Respon dalam memecahkan masalah baik dan respon siswa dengan penerapan PBI positif. Selain itu ditemukan kendala untuk kegiatan presentasi karena siswa kurang percaya diri untuk ikut aktif dalam kegiatan ini. Pembiasaan kegiatan ini dan motivasi merupakan cara yang baik untuk meningkatkan keaktifan siswa, 3) Penerapan model PBI dapat meningkatkan hasil belajar siswa. Nilai rata-rata hasil belajar siswa di siklus I adalah 70,63 dan ketuntasan belajar klasikal 74,07%. Di siklus II mengalami peningkatan menjadi 77,78 dan ketuntasan belajar klasikal menjadi 81,48%. Peningkatan terjadi lagi di siklus III yakni sebesar 82,34 dan ketuntasan belajar klasikal 89,29%.

Membranes with high CO2 solubility are essential for developing a separation technology with low carbon footprint. To this end, physical blend membranes of [BMIM][Ac] and [BMIM][Succ] as Ionic Liquids (ILs) and PIM-1 as the polymer were prepared trying to combine the high permeability properties of PIM-1 with the high CO2 solubility of the chosen ILs. Membranes with a PIM-1/[BMIM][Ac] 4/1 ratio nearly double their CO2 solubility at 0.8 bar (0.86 cm3 (STP)/cm3 cmHg), while other ratios still maintain similar solubilities to PIM-1 (0.47 cm3 (STP)/cm3 cmHg). Moreover, CO2 permeability of PIM-1 /[BMIM][Ac] blended membranes were between 1050 and 2090 Barrer for 2/1 and 10/1 ratio, lower than PIM-1 membrane, but still highly permeable. The here presented self-standing and mechanically resistant blend membranes have yet a lower permeability compared to PIM-1 yet an improved CO2 solubility, which eventually will translate in higher CO2/N2 selectivity. These promising preliminary results will allow us to select and optimize the best performing PIM-1 /ILs blends to develop outstanding membranes for an improved gas separation technology.

La Capacité d'Absorption (CA) de l'information et la perspective « Knowledge Based View » (KBV) ont fait l'objet d'un intérêt académique significatif et croissant à travers un grand nombre de disciplines pendant la dernière décennie. En dépit de cet important intérêt académique, la littérature développée sur la CA et la perspective KBV a privilégié l'étude des entreprises à forte composante technologique, visant à modéliser principalement l'innovation sur la base de l'absorption de l'information technologique. En conséquence, la connaissance et l'information concernant l'environnement du marché international d'une organisation ont été principalement ignorées, malgré leur importance accrue pour le développement des activités d'exportation des entreprises, laquelle a été fréquemment soulignée dans la littérature portant sur l'internationalisation. Particulièrement dans le cas des PME, il est unanimement accepté qu'afin de mieux faire face à la turbulence de marchés internationaux, les PME doivent mieux gérer l'information et développer des niveaux supérieurs de connaissances et compétences de gestion des activités internationales (Kuivalainen et Bell 2004). Ce travail de thèse vise à répondre au moins en partie à ce manque dans la littérature et fournit deux principales contributions. En premier lieu, cette thèse propose une conceptualisation de la CA de l'information à l'export, adaptée dans le cas des PME, basée sur la réitération du concept de CA par Zahra et George (2002) en tant que capacité dynamique. En second lieu, des éléments de la littérature sur la CA, la KBV, l'orientation de marché (OM), le marketing et le management international ont été combinés pour développer un cadre conceptuel qui décrit l'évolution de l'état d'information à l'export des PME et son impact sur leur niveau de performance internationale. La nature causale de la problématique de recherche nous a mené à retenir une méthodologie quantitative. Néanmoins, tenant compte de la nature qualitative et multidimensionnelle de l'information, une première recherche préliminaire qualitative a été entreprise afin d'explorer si l'utilisation des éléments des perspectives KBV, AC et OM est appropriée pour décrire l'impact de l'évolution de l'état d'information à l'export dans les PME sur leur niveau de performance internationale. De plus, il nous a semblé important de vérifier si la mobilisation de ces éléments de la littérature est congruente avec les perceptions managériales. Ainsi, 24 entretiens semi-directifs ont été réalisés avec les dirigeants des entreprises exportatrices. Par la suite, sur la base de la revue de la littérature et de la recherche qualitative préliminaire, un questionnaire a été développé. Le questionnaire a été pré-testé et envoyé aux dirigeants des PME de l'industrie des métaux. 107 questionnaires exploitables ont été recueillis (taux de réponse de 19,1%). L'évaluation des relations causales de l'influence qui composent le modèle de recherche selon l'approche PLS a mené à la validation de neuf des dix propositions de recherche énoncées. Ainsi, les résultats suggèrent que la perspective de la CA de Zahra et de George (2002) constitue un cadre attrayant et efficace pour explorer l'impact de la dynamique liée au traitement de l'information à l'export sur le niveau de la performance internationale des PME. Les résultats suggèrent que la qualité de l'exposition aux sources d'information à l'export des PME (évaluée à travers la richesse des sources d'information à l'export) et la richesse des expériences internationales des employés export (évaluée en termes de variété, complémentarité et durée), ainsi que la réelle expérience internationale de PME apparaissent comme des causes déterminantes de la capacité des PME à acquérir efficacement de l'information à l'export
The Absorptive Capacity (AC) and the Knowledge-Based View (KBV) perspectives have generated significant scholarly interest across a large number of disciplines over the last decade. Despite this important scholarly interest, both the AC and KBV literature streams have focused primarily on technology-intensive and dynamic industries, aiming to model innovation on the basis of technological and R&D knowledge absorption. As a result, knowledge and information relating to an organization's international market environment were mainly ignored, even though their utmost importance for foreign business development is overall emphasized within the international marketing literature. Especially in the case of SMEs, it is oftenly suggested that in order to handle the international market place turbulence, they shall better manage information and develop superior knowledge and capabilities relative to the management of foreign operations (Kuivalainen and Bell 2004). This dissertation aims to address this gap in the literature and provides two major contributions. First, a conceptualization of the AC of export information adapted in the case of SMEs is proposed, based on Zahra and George's (2002) re-lecture of the AC concept. Second, insights from the AC, marketing, international business, market orientation (MO) and knowledge management literature were combined to develop and test a research model for describing the export information state evolution in SMEs and its impact on their international performance. The causal nature of the research question led us to the retaining of a quantitative methodology. Nonetheless, taking into account the preponderant qualitative and multi-dimensional nature of information, a first qualitative preliminary research has been undertaken aiming to explore whether the use of insights from the KBV, MO and AC literature for describing the impact of the evolution of export information state in SMEs on their levels of international performance was consistent with the managerial perceptions. To this end, 24 semi-structured interviews were realized with French and Romanian SMEs' managers. Furthermore, based on the literature review and the preliminary qualitative investigation, a questionnaire was developed. The questionnaire was pre-tested and then sent to the managers of exporting SMEs from the steel industry. 107 usable questionnaires were gathered, yielding for a response rate of 19,1%. The estimation of the causal relationships of influence composing the research model with PLS path modeling (XLSTAT software) led to the validation of nine out of the ten research propositions composing the research model. The results suggest that the AC perspective of Zahra and George (2002) is an appealing and efficient framework for exploring the dynamics associated with export information processing in small firms and their impact upon SMEs' international performance levels. The results suggest that the richness of export information sources, the real export experience of SMEs, and the richness of the international experiences of export employees positively and significantly influence the efficiency of export information acquisition in small firms. Furthermore, the efficiency of export information acquisition and assimilation practices, namely knowledge transfer and integration and coordination efficiency, determine the export responsiveness capacity of SMEs. In turn, the export responsiveness capacity has a strong impact upon the SMEs' positional advantage in terms of international marketing competences. The results suggest that by means of responding and taking corrective action in order to better manage foreign market shifts, firms appear to build superior levels of positional advantage in terms of foreign market-related competences compared to their competitors

The superfamily Sauropodomorpha comprised some of the largest terrestrial herbivorous vertebrates that have ever walked on this planet. The study of these fascinating beings has been at the forefront of palaeontological research throughout the years. Especially in the last 20 years, due to the advent of modern techniques and technologies, significant steps have been made towards our understanding of their physiology. One of the sauropodomorphs’ central morphological characteristics is the expression of postcranial skeletal pneumaticity (PSP), a condition that is also expressed in the other superfamily of the order Saurischia, the Theropoda, as well as in their extant relatives, the Avians. This condition remodels the vertebrae and, occasionally the girdles and appendicular elements, producing perforations, depressions, excavations, cavities and internal chambers. This is a result of bone invasion and resorption from the development of the lung-air sac diverticula of the respiratory system. The various forms of this expression have been addressed across all subfamilies of sauropodomorphs mostly from an evolutionary aspect and, recently, from a developmental scope too. The resulting hypotheses state that PSP may have acted as a mechanism for weight reduction, allowing for sauropods to attain large sizes without having to suffer analogous gravitational constraints from equally attained masses. Any possible associations, though, between the expression of pneumaticity and body size have not been put under test. Through this study, a method of quantification and categorisation of PSP, and therefore, classification of the sauropodomorphs which express it, is created from the data retrieved from 61 taxa across all subfamilies, permitting us not only to trace any correlation between PSP and metric size data (body mass and body length) but also to visualise the evolution of PSP throughout Sauropodomorpha. This classification scheme from highest to lowest expression of PSP, ‘Alpha’, ‘Beta’, ‘Gamma’, ‘Delta’, and ‘Epsilon’ stems from the numerical estimate of pneumaticity in terms of a percentage, called the Pneumaticity Degree Index (PDI%). The revised scheme, Pneumaticity Degree Index (PDI%), takes account of the number of vertebral elements that are pneumatized in a single vertebra, the nature of pneumaticity traits, as well as the intensity of pneumatization in different body regions of interest (e.g. vertebral column), resulting in an integral and comprehensive measure of PSP. The proposed method ranks each pneumaticity trait with a value from 1 to 5 with 1 (100%) representing the most invasive unambiguous trait (e.g. camellae) and 5 (~ 10%) the least invasive and most ambiguous trait (i.e. fossa). By adding and dividing by their number all of the observed traits of every available vertebra of a region we retrieve a decimal numeric outcome and this outcome is translated to a percentage. The total average pneumatization of any taxon is calculated by retrieving the total average of the pneumaticity from all available body regions. In this study, only vertebral and pelvic elements were used for the retrieval of pneumaticity data since they are the most frequently exhibiting pneumatic traits in comparison to pectoral and appendicular elements. Results show that the total average PDI% range of basal Sauropodomorpha is 0%-59%, of the non-neosauropod Eusauropoda is 23%-90%, of Diplodocoidea is 45%-73%, of Macronaria is 45%-92% and of Somphospondyli is 46%-94%. The most pneumatized vertebral landmarks are the centrum, neural arch and neural spine. The vertebral regions most commonly pneumatized are the cervicals and dorsals. No genus or subfamily of sauropodomorphs ever reaches 100% pneumatization in all vertebral regions. Furthermore, PSP is not always positively correlated with mass, rather its expression is mostly correlated with length. Taxa with low or high masses may exhibit either low or high PDI’s. In addition, increasing progression of the extent and expression of PSP occurs mostly on a subfamilial level and less throughout the entire lineage. Finally, modelling calculations result in an Ornstein-Ulenbeck with an early burst progress of the expression of pneumatisation in sauropodomorphs. After that, the expression proceeds relatively steadily throughout the entire superfamily. The biotic causes of PSP are still unclear, as it could be an artefact of inheritance and genetic drift throughout speciation events. The pneumatization degree index (PDI%) is a means of quantifying and categorising pneumatization in any archosaurian taxon that is faster and cheaper, though less accurate, than CT scanning. It is more precise than the Pneumaticity Index since it integrates a wider selection of the qualities and osteological characteristics of pneumaticity we want to measure, provided that the bone under study is at least 50% free of matrix.

Public key infrastructure (PKI) was created to provide the basic services of confidentiality, authenticity, integrity and non-repudiation for sensitive information that may traverse public (un-trusted) networks. This thesis provides a brief description of the background and functional components of a PKI, and then "builds" a PKI to be used for research at the Naval Postgraduate School (NPS). Deficiencies of this PKI with respect to DoD PKI policy are delineated. The thesis addresses details of software selection, installation, configuration and operation; using Netscape's Certificate Management System as its Certificate Authority application of choice. The functionality of this PKI was validated by testing all major certificate lifecycle events (creation, archival, revocation, validation, etc.) All but two of these tests were successful-key escrow and revocation checking-and thus these two remain to be addressed by further work to make the NPS PKI fully functional.

This thesis describes the synthesis of soluble Polymer of Intrinsic Microporosity (PIM-1), fluoro-endcapped PIM-1 (F-PIM-1) and copolymers of F-PIM-1 with poly(ethylene glycol) monomethyl ether (MeOPEG). The main aim of the project was to alter the porosity of microporous PIM-1 in three ways: (a) synthesis of copolymers of F-PIM-1 with MeOPEG (b) blending of PIM-1 with MeOPEG in various proportions; and (c) adsorption of MeOPEG from aqueous solution byPIM-1. PIM-1 and F-PIM-1 were synthesized by step growth polymerization of tetrafluoroterephthalonitrile (TFTPN) with 5,5',6,6'-tetrahydroxy-3,3,3',3'-tetramethyl-1,1'-spirobisindane (THSB), using the conventional method and a newly reported high shear mixing method. F-PIM-1 oligomers were then coupled to poly(ethylene glycol) monomethyl ether (MeOPEG). The products were analyzed by NMR, IR, MALDI ToF MSS, TGA and polystyrene based GPC as well as multidetector GPC techniques. The high shear technique generally produced high molar mass products and yields. This method was also more successful for copolymerization.Blending of PIM-1 and MeOPEG in different proportions resulted in macrophase separation. Copolymer products were used to facilitate mixing of blends (as compatibilizers), however only 5% of MeOPEG could be solubilised into a PIM-1 phase. The effect of compatibilizer was found to be affected by interaction between PIM-1 and copolymer. However, N2 adsorption studies showed that after thermal removal of MeOPEG, PIM-1 regained stable porosity with significant BET surface area.Fluorescence studies were aimed at applications of PIM-1 and copolymers in sensors. PIM-1 and copolymers, spin-coated on the polyester-based substrate Melinex, were studied with and without methanol treatment in an environment of different solvent vapours. The effect of time and volume on wavelength shift and change in intensity was studied. Polar solvents tended to cause a red shift with decrease in intensity while less polar solvents behaved otherwise. Based on fluorescence experiments, solvent profiles for PIM-1 and copolymers were established.

L'investigation des savoirs-faire et sentiments mobilisés dans les deux types d'interventions au moyen de 122 observations non participantes et de 44 entretiens de recherche, a permis de sélectionner et de se concentrer plus spécifiquement ici sur l'éthique construite pour ces situations de travail au travers de la double analyse - Cognitivo-Discursive sous Tropes® et Clinique - des entretiens de recherche. Les résultats mettent en visibilité l'autonomie morale subjective et la responsabilité morale expansée des soignants, la métamorphose de la portée de l'agir dès lors qu'il s'agit de (dé)procréation humaine, ainsi que la nécessité de changer de paradigme pour les sciences humaines qui travaillent ces sujets. This thesis in psychology emerge from a larger qualitative research conjunctively approaching all the nursing staff activity in 2 infertility clinics and 2 abortion clinics. The investigation of the abilities and feelings mobilized in the operations types by the way of 122 non participant observations and 44 research interviews, has allowed to select and put the focus more specifically on the ethic built up within work situations throughout the research interviews double analysis Cognitivo-Discursive under Tropes® and Clinical -. Abstract The correct three dimensional structures of proteins are essential for their ability to function properly. Proteins start to fold as soon as they are synthesized in the ribosomes from activated amino acids. Many secreted, cell-surface, secretory pathway and endoplasmic reticulum (ER) lumenal proteins have in their amino acid sequence cysteine residues which form intra- and intermolecular disulfide bridges that stabilize the overall fold of the proteins and protein complexes. The formation of correct disulfide bonds is a complex process which takes place within the ER. Protein disulfide isomerase (PDI) is the key enzyme in the formation and rearrangement of correct disulfide bonds in the ER. It is an archetypal and the best studied member of the PDI family, i.e. a group of ER proteins that resemble thioredoxin (TRX), a protein reductase, in their structure. PDI has a four domain a-b-b'-a' structure the a and a' domains having the catalytic activity and amino acid sequence similarity to TRX. In addition to its function as a thiol-disulfide oxidoreductase, PDI acts as the β subunit in two protein complexes: collagen prolyl 4-hydroxylase (C-P4H) and microsomal triglyceride transfer protein (MTP). The closest homologue of PDI is the multifunctional enzyme and chaperone ERp57 that functions in concert with two lectins, calnexin (CNX) and calreticulin (CRT) specifically in the folding of proteins that have sugar moieties linked to them. ERp57 is 56% similar to PDI in its amino acid sequence and has also the four-domain architecture. Despite the high similarity in their structures ERp57 cannot substitute for PDI as the β subunit of C-P4H. The minimum requirement for the C-P4H tetramer assembly is fulfilled by domains b' and a' of PDI, while domains a and b enhance this function and can be substituted in part by those of ERp57. Until very recently the structural information of any of the PDI family members, which contains the TRX active site was limited to solution structures of human PDI domains a and b. In this research the domain boundaries of the full length ERp57 were defined and the individual domains characterized. Furthermore the solution structures of the catalytically active domains a and a' of ERp57 were studied by nuclear magnetic resonance (NMR).

In the framework of the Terrafirma project, Persistent Scatterers Interferometry (PSI) has be used for mapping land subsidence at basin scale in Gioia Tauro plain (Italy). The investigated area is built over unconsolidated fine-grained sediments, where the increasing groundwater demands for irrigation have caused the natural sediment consolidation to progressively accelerate. Both historical (1992-2001; ERS1/2 images) and recent (2002-2006; ENVISAT images) scenarios are analyzed to solve the spatial variability and temporal evolution of ground displacements affecting the plain. The results show deformation rates as high as 10-12 mm/yr in 1992-2007, with highest velocities occurred between 1992 and 2000 within the central part of the basin, in the area of Rizziconi (5 km ESE of Gioia Tauro). The outcomes of this PSI study could support the future improvement of groundwater management and the implementation of best strategies for land use planning and sustainable use of groundwater resources.

Calcium pyrophosphate (CPP) dihydrate crystals form extracellularly. Their formation requires sufficient extracellular inorganic pyrophosphate (ePPi), calcium, and pro-nucleating factors. As inorganic pyrophosphate (PPi) cannot cross cell membranes passively due to its large size, ePPi results either from hydrolysis of extracellular ATP by the enzyme ectonucleotide pyrophosphatase/phosphodiesterase 1 (also known as plasma cell membrane glycoprotein 1) or from the transcellular transport of PPi by ANKH. ePPi is hydrolyzed to phosphate (Pi) by tissue non-specific alkaline phosphatase. The level of extracellular PPi and Pi is tightly regulated by several interlinked feedback mechanisms and growth factors. The relative concentration of Pi and PPi determines whether CPP or hydroxyapatite crystal is formed, with low Pi/PPi ratio resulting in CPP crystal formation, while a high Pi/PPi ratio promotes basic calcium phosphate crystal formation. CPP crystals are deposited in the cartilage matrix (preferentially in the middle layer) or in areas of chondroid metaplasia. Hypertrophic chondrocytes and specific cartilage matrix changes (e.g. high levels of dermatan sulfate and S-100 protein) are related to CPP crystal deposition and growth. CPP crystals cause inflammation by engaging with the NALP3 inflammasome, and with other components of the innate immune system, and is marked with a prolonged neutrophilic inflitrate. The pathogenesis of resolution of CPP crystal-induced inflammation is not well understood.

AbstractPomegranates are known for being rich in polyphenols and are considered to have immense therapeutic potential. The present study investigates the hypothesis that the Methanolic Extract of Pomegranate (PME), a rich source of antioxidants, may reverse the adverse effects of TNF-α in endothelial cells. This was done by pre-treating the endothelial cells EA.hy926 with PME (80 µg/ml) before subjecting them to apoptotic stimuli, which was TNF-α in combination with cyclohexamide. PME was found to rescue a population of cells from apoptosis induced by TNF-α modulating the levels of BCL2 and BAX involved in intrinsic apoptotic pathway. PME was found to increase the BCL-2/BAX ratio and reverse the elevated levels of effector caspase and thus assist cells to escape from apoptotic stimuli. Also, the extract was found to attenuate oxidative stress by reducing the levels of Reactive Oxygen Species (ROS). Supplementing its anti-atherosclerotic potential, PME pre-treatment diminished the elevated levels of adhesion molecules like VCAM upon TNF-α treatment. PME may therefore have therapeutic implications in protecting the endothelium from TNF-α triggered atherosclerosis.

Reliability-based robust design optimization deals with two objectives of structural design methodologies subject to various uncertainties: reliability-based design and robust design. A reliability-based design optimization deals with the probability of failure, while a robust design optimization minimizes the product quality loss. In general, the product quality loss is described by using the first two statistical moments: mean and standard deviation. In this paper, a performance moment integration (PMI) method is proposed by using numerical integration scheme for output response to estimate the product quality loss. For the reliability part of the reliability-based robust design optimization, the performance measure approach (PMA) and its numerical method, hybrid-mean value (HMV) method, are used. New formulations of reliability-based robust design optimization are presented for three different types of robust objectives, such as smaller-the-better, larger-the-better, and nominal-the-better types. Examples are used to demonstrate the effectiveness of reliability-based robust design optimization using the proposed PMI method for different types of robust objective.

Various industries related to daily life are related to the precision machinery industry (PMI). With the pace of Industry 4.0, PMI is one of the industries with great development potential. Renewal progress has not only caused changes in life but also revealed a significant impact on occupations. In recent years, the rise of Industry 4.0 has increased the output value of various industries. The launch of Industry 4.0 has led to global manufacturing initiatives to improve production efficiency and to reduce its burden. In order to increase its industrial benefit as the goal, it is important to discuss the problems and challenges that people face. Therefore, this study uses the Analytic Hierarchy Process (AHP) method to focus on the objectives of the research on the critical success factors of intelligentization in PMI. This study not only expects to provide the existing machinery manufacturing industry and the management policies for the future investment in PMI manufacturer reference value but also expects to serve as an academic reference for government policy considerations and scholars in the machinery manufacturing industry.

The United States represents the world’s largest market for pharmaceutical drugs. It is also the only advanced economy in the world that does not regulate drug prices. There is no upper threshold for the prices of medicines in the United States. List prices are instead set by manufacturers in negotiation with supply-chain intermediaries, though some federal programs have degrees of discretion in price determinations. In practice, this deregulated system means that drug prices in the United States are generally far higher than in other advanced economies, adversely affecting patient accessibility and system affordability. In this paper, we draw on the “theory of innovative enterprise” to develop a framework that provides both a critique of the existing pricing system in the United States and a foundation for developing a new model of pricing regulation to support safety and effectiveness through drug development as well as accessibility and affordability in the distribution of approved medicines to patients. We introduce a regulatory approach we term “Pricing for Medicine Innovation” (PMI), which departs dramatically from the market-equilibrium assumptions of conventional (neoclassical) economics. The PMI approach recognizes the centrality of collective investments by government agencies and business firms in the productive capabilities that underpin the drug development process. PMI specifies the conditions under which, at the firm level, drug pricing can support both sustained investment in these capabilities and improved patient access. PMI can advance both of these objectives simultaneously by regulating not just the level of corporate profit but also its allocation to reinvestment in the drug development process. PMI suggests that although price caps are likely to improve drug affordability, there remain two potential issues with this pricing approach. Firstly, in an innovation system where a company’s sales revenue is the source of its finance for further drug development, price caps may deprive a firm of the means to invest in innovation. Secondly, even with adequate profits available for investment in innovation, a firm that is run to maximize shareholder value will tend to use those profits to fund distributions to shareholders rather than for investment in drug innovation. We argue that, if implemented properly, PMI could both improve the affordability of medicines and enhance the innovative performance of pharmaceutical companies.

Review question / Objective: A meta-analysis of cross-sectional, case-control and cohort studies (S) was conducted to explore whether women with POI/POF (P) could increase risk of depression, anxiety, and poor quality of life (O). Intervention studies (I) are eligible only if pre-intervention data were available(I) and the comparator(C) of this meta-analysis was people who do not suffer from POI (age-matched). Condition being studied: Several studies have been conducted to show that POI affects the quality of life of patients, of which psychological and psychosocial factors cannot be ignored as a part. Most studies have shown that POI patients are more prone to negative emotions such as depression and anxiety than normal people, but some studies have also shown that normal POI patients are psychologically indistinguishable from normal people. Therefore, meta-analysis is needed to determine the effect of POI on psychological factors and to better guide the clinical treatment plan.

Review question / Objective: Perineural invasion (PNI) is a possible route for metastatic spread in various cancer types, including colorectal cancer (CRC). PNI is linked to poor prognosis. For patients with lymph node positive colorectal cancer, a number of large-scale RCT studies have confirmed that they can benefit from chemotherapy, but there are still many controversies about whether colorectal patients with negative lymph nodes need adjuvant chemotherapy. At present, there is a general consensus that patients with stage II colorectal cancer who have risk factors such as PNI+ need chemotherapy. However, there are many recent literatures that show that patients with stage II colorectal cancer with nerve invasion risk factors can not prolong the OS and DFS of patients. At the same time, chemotherapy increases the toxicity, economic and mental burden of patients. Therefore, we hope to write this review to summarize the current research findings and provide some clinical guidance on whether patients with lymph node negative colon cancer who have perineural invasion should receive chemotherapy. Condition being studied: Patients with high-risk such as PNI+ stage II colon cancer (CC) are recommended to undergo adjuvant chemotherapy (ACT). However, whether such patients can benefit from ACT remains unclear. And recently studies shown that, ACT had no significant benefit among patients with PNI.

This report describes the early success of the PLI on students’ learning outcomes, evidence on what elements of the implementation are working, and the process and principles of the analytics partnership. The report aims to share with other education systems the lessons learned from this journey.

The principal accomplishment of the research supported by BARD was progress toward a comprehensive view of cell-surface electrical effects (both in cell walls [CWs] and at plasma membrane [PM] surfaces) upon ion uptake, intoxication, and amelioration. The research confirmed that electrostatic models (e.g., Gouy-Chapman-Stern [G-C-S]), with parameter values contributed by us, successfully predict ion behavior at cell surfaces. Specific research objectives 1. To characterize the sorption of selected heavy metals (Cu, Zn, Pb, Cd) to the root PM in the presence of other cations and organic ligands (citric and humic acids). 2. To compute the parameters of a G-C-S model for heavy-metal sorption to the root PM. 3. To characterize the accumulation of selected heavy metals in various plant parts. 4. To determine whether model-computed ion binding or ion activities at root PM surfaces predict heavy-metal accumulation in whole roots, root tips, or plant shoots. 5. To determine whether measured ion binding by protoplast-free roots (i.e., root CWs) predicts heavy-metal accumulation in whole roots, root tips, or plant shoots. 6. To correlate growth inhibition, and other toxic responses, with the measured and computed factors mentioned above. 7. To determine whether genotypic differences in heavy-metal accumulation and toxic responses correlate with genotypic differences in parameters of the G-C-S model. Of the original objectives, all except for objective 7 were met. Work performed to meet the other objectives, and necessitated on the basis of experimental findings, took the time that would have been required to meet objective 7. In addition, work with Pb was unsuccessful due to experimental complications and work on Cd is still in progress. On the other hand, the uptake and toxicity of the anion, selenate was characterized with respect to electrostatic effects and the influences of metal cations. In addition, the project included more theoretical work, supported by experimentation, than was originally planned. This included transmembrane ion fluxes considered in terms of PM-surface electrical potentials and the influence of CWs upon ion concentrations at PM surfaces. A important feature of the biogeochemistry of trace elements in the rhizosphere is the interaction between plant-root surfaces and the ions present in the soil solution. The ions, especially the cations, of the soil solution may be accumulated in the aqueous phases of cell surfaces external to the PMs, sometimes referred to as the "water free space" and the "Donnan free space". In addition, ions may bind to the CW components or to the PM surface with variable binding strength. Accumulation at the cell surface often leads to accumulation in other plant parts with implications for the safety and quality of foods. A G-C-S model for PMs and a Donnan-plus-binding model for CWs were used successfully to compute electrical potentials, ion binding, and ion concentration at root-cell surfaces. With these electrical potentials, corresponding values for ion activities may be computed that are at least proportional to actual values also. The computed cell-surface ion activities predict and explain ion uptake, intoxication, and amelioration of intoxication much more accurately than ion activities in the bulk-phase rooting medium.

The overall objective was to understand the cloud flocculation of citrus juice by characterization of the interactions between proteins and pectins, and to determine the role of PE isozymes in catalyzing this phenomenon. Specific objectives were to: 1. identify/characterize cloud-proteins in relation to their coagulable properties and affinity to pectins; 2. to determine structural changes of PME and other proteins induced by cation/pectin interactions; 3. localize cloud proteins, PME and bound protein/pectates in unheated and pasteurized juices; 4. to create "sensitized" pectins and determine their effect on clarification. The original objectives were not changed but the methods and approach were modified due to specific research requirements. Two i postulates were: 1. there is a specific interaction of cloud proteins with de-esterified regions of ! pectin and this contributes to cloud loss; 2. isozymes of pectin-methyl-esterase (PME) vary in efficiency to create sensitized pectins. The appearance of citrus fruit juice is an important quality factor and is determined by the color and turbidity that .are conferred by the suspended particles, i.e., by the cloud and its homogeneity. Under some circumstances the cloud tend to flocculate and the juice clarifies. The accepted approach to explain the clarification is based on pectin demethoxylation by PME that promotes formation of Ca-pectate. Therefore, the juice includes immediate heat-inactivation upon ~ squeezing. Protein coagulation also promotes cloud instability of citrus fruit extracts. However, the clarification mechanism is not fully understood. Information accumulated from several laboratories indicates that clarification is a more complex process than can be explained by a single mechanism. The increasing trend to consume natural-fresh juice emphasizing the importance of the knowledge to assure homogeneity of fresh juice. The research included complementary directions: Conditions that induce cloud-instability of natural- juice [IL]. Evaluate purification schemes of protein [USA]. Identifications of proteins, pectin and neutral sugars ([IL]; Structure of the cloud components using light and electron microscopy and immuno-labeling of PME, high-methoxyl-pectin (HMP) and low-methoxyl-pectin (LMP); Molecular weight of calcium sensitized pectins [US]; Evaluation of the products of PME activity [US]. Fractions and size distribution and cloud components [IL-US]. The optimal pH activity of PME is 7 and the flocculation pH of the cloud is 3-4. Thus, the c roles of PME, proteins and pectins in the cloud instability, were studied in pH ranges of 2- 7. The experiments led to establish firstly repeatable simulate conditions for cloud instability [IL]. Thermostable PME (TS-PE) known to induce cloud instability, but also thermolabile forms of PME (TL-PE) caused clarification, most likely due to the formation and dissolution of inactive :. PE-pectin complexes and displacement of a protective colloid from the cloud surface [US]. Furthermore, elimination of non-PME protein increases TS-PE activity, indicating that non-PME proteins moderate PME activity [US]. Invasive exotic plant (IEP) species are a significant threat to natural ecosystem integrity and biodiversity, and controlling them is a high priority for the National Park Service. The North-ern Colorado Plateau Network (NCPN) selected the early detection of IEPs as one of 11 monitoring protocols to be implemented as part of its long-term monitoring program. This report represents work completed at Colorado National Monument during 2019. During monitoring conducted June 12–19, a total of 20 IEP species were detected on monitoring routes and transects. Of these, 12 were priority species that accounted for 791 separate IEP patches. IEPs were most prevalent along riparian areas. Yellow sweetclover (Melilotis officinale) and yellow salsify (Tragopogon dubius) were the most commonly detected priority IEPs along monitoring routes, representing 73% of all priority patches. Patches of less than 40 m2 were typical of nearly all priority IEP species except yellow sweetclover. A patch management index (PMI) was created by combining patch size class and percent cover for each patch. In 2019, a large majority of priority IEP patches were assigned a PMI score of low (46%) or very low (50%), indicating small and/or sparse patches where control is generally still feasible. This is similar to the numbers for 2017, when 99% of patches scored low or very low in PMI. Seventy-eight percent of tree patches were classified as seedlings or saplings, which require less effort to control than mature trees. Cheatgrass (Anisantha tectorum) was the most common IEP recorded in transects, found in 30–77% of transects across the different routes. It was the only species found in transects on all monitoring routes. When treated and untreated extra areas near the West Entrance were compared, the treated area had comparable or higher lev-els of IEPs than the untreated area. When segments of monitoring routes conducted between 2003 and 2019 were compared, results were mixed, due to the different species monitored in different time periods. But in general, the number of IEPs per 100 meters is increasing or remaining constant over time. There were notable increases in IEP patches per 100 meters on several routes in 2019: field bindweed (Convolvulus arvensis) along East Glade Park Road; Siberian elm (Ulmus pumila) in Red Canyon; yellow salsify along East Glade Park Road, No Thoroughfare Canyon, No Thoroughfare Trail, and Red Canyon; and yellow sweetclover in No Thoroughfare Canyon and Red Canyon. Network staff will return to re-sample monitoring routes in 2021.

The study has been focused on proline-rich proteins from the HyPRP family. Three proline-rich proteins have been characterized with the CWLP as the main objective. We showed that this unique protein is assembled in the plasma membrane (PM) and forms a continuum between the cell wall (CW) and cytosol via the PM. While spanning the PM, it is arranged in lipid rafts as CWLP-aquaporin complexes that recruit PP2A-β”, as a part of PP2A enzyme, close to the aquaporin moiety where it dephosphorylates two crucial Ser residues and induces closure of the aquaporin water channels. The closure of water channels renders cells more tolerant to plasmolysis and plants to dehydration. This unique effect was observed not only in Arabidopsis, but also in potato plants over expressing the CWLP, suggesting a possible usage in crop plants as a valve that reduces loss of water or/and elevates cold resistance. The CWLP is a member of the HyPRP protein family that all possess structurally similar 8CM domain, predicted to localize to PM lipid rafts. In this study, two additional highly homologous HyPRP proteins were also studied. The GPRP showed the same localization and it’s over expression increased tolerance to lack of water. However, the third one, PRP940, despite sharing high homology in the 8CM domain, is completely different and is assembled in parallel to cortical microtubules in the cell. Moreover, our data suggest that this protein is not involved in rendering plants resistant to lack of water. We suggest implying CWLP as a tool for better regulation of water maintenance in crop plants.

Review question / Objective: Can Baduanjin exercise improve the cardiac rehabilitation of patients with coronary artery disease after percutaneous coronary artery surgery? Condition being studied: Coronary heart disease (CHD), also known as coronary artery disease (CAD), is the single most common cause of death globally, with 7.4 million deaths in 2013, accounting for one-third of all deaths (WHO 2014). PCI has been shown to be effective in reducing mortality in patients with CHD. During follow-up, it has been shown that the benefits of PCI can be offset by the significant risks of coronary spasm, endothelial cell injury, recurrent ischemia, and even restenosis or thrombus. Numerous guidelines endorse the necessity for cardiac rehabilitation (CR), which is recommended for patients with chronic stable angina, acute coronary syndrome and for patients following PCI. Baduanjin have been widely practised in China for centuries, and as they are considered to be low risk interventions, their use for the prevention of cardiovascular disease is now becoming more widespread. The ability of Baduanjin to promote clinically meaningful influences in patients with CHD after PCI, however, still remains unclear.
