Passiflora foetida is a horticultural plant and vital traditional Chinese herbal medicine. In our previous study, the characterization and immuno-enhancing effect of fruits polysaccharide 1 (PFP1), a water-eluted hetero-mannan from wild Passiflora foetida fruits, were investigated. Herein, another three salt-eluted novel polysaccharides, namely PFP2, PFP3, and PFP4, were obtained and structurally characterized. The results showed that PFP2, PFP3, and PFP4 were three structurally similar hetero-galacturonans with different molecular weights of 6.11 × 104, 4.37 × 104, and 3.48 × 105 g/mol, respectively. All three of these hetero-galacturonans are mainly composed of galacturonic acid, galactose, arabinose (75.69%, 80.39%, and 74.30%, respectively), and other monosaccharides including mannose, fucose, glucose, ribose, xylose, and glucuronic acid (24.31%, 19.61, and 25.70%, respectively), although differences in their backbone structure exist. Additionally, immunomodulatory assay indicated that the three hetero-galacturonans possess the ability to promote the production of nitric oxide (NO), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) in RAW264.7 macrophages in a concentration-dependent manner (p < 0.05). Especially, PFP3 displayed a stronger enhancing effect than PFP2 and PFP4 at the minimum effective concentration. Therefore, the results suggested that the obtained three salt-eluted hetero-galacturonans, especially PFP3, could be utilized as immunomodulatory effectivity ingredients in nutritional/pharmaceutical industries.

ABSTRACT Hypermutator Pseudomonas aeruginosa strains, characterized by an increased spontaneous-mutation rate, are found at high frequencies in chronic lung infections. Hypermutability is associated with the loss of antimutator genes related to DNA repair or damage avoidance systems. Only a few antimutator genes have been described in P. aeruginosa, although there is some evidence that additional genes may be involved in naturally occurring hypermutability. In order to find new P. aeruginosa antimutator genes, we constructed and screened a library of random insertions in the PA14 strain. Some previously described P. aeruginosa and/or Escherichia coli antimutator genes, such as mutS, mutL, uvrD, mutT, ung, and mutY, were detected, indicating a good coverage of our insertional library. One additional mutant contained an insertion in the P. aeruginosa PA14-04650 (pfpI) gene, putatively encoding a member of the DJ-1/ThiJ/PfpI superfamily, which includes chaperones, peptidases, and the Parkinson's disease protein DJ-1a. The pfpI-defective mutants in both PAO1 and PA14 showed higher spontaneous mutation rates than the wild-type strains, suggesting that PfpI plays a key role in DNA protection under nonstress conditions. Moreover, the inactivation of pfpI resulted in a dramatic increase in the H2O2-induced mutant frequency. Global transcription studies showed the induction of bacteriophage Pf1 genes and the repression of genes related to iron metabolism, suggesting that the increased spontaneous-mutant frequency may be due to reduced protection against the basal level of reactive oxygen species. Finally, pfpI mutants are more sensitive to different types of stress and are affected in biofilm formation.

The aim of this study was to propose a new index called Postural Force Production Index (PFPI) for evaluating the force production during handcycling. For a given posture, it assesses the force generation capacity in all Cartesian directions by linking the joint configuration to the effective force applied on the handgrips. Its purpose is to give insight into the force pattern of handcycling users, and could be used as ergonomic index. The PFPI is based on the force ellipsoid, which belongs to the class of manipulability indices and represents the overall force production capabilities at the hand in all Cartesian directions from unit joint torques. The kinematics and kinetics of the arm were recorded during a 1-min exercise test on a handcycle at 70 revolutions per minute performed by one paraplegic expert in handcycling. The PFPI values were compared with the Fraction Effective Force (FEF), which is classically associated with the effectiveness of force application. The results showed a correspondence in the propulsion cycle between FEF peaks and the most favorable postures to produce a force tangential to the crank rotation (PFPI). This preliminary study opens a promising way to study patterns of force production in the framework of handcycling movement analysis.

Here, we screen a panel of 13 mutants defective in intracellular proteases and demonstrate that, in addition to the known alterations in Lon and AsrA mutants, mutation of three protease-related proteins PfpI, ClpS, and ClpP differentially affected antibiotic resistance, swarming motility, and biofilm formation.

The DJ-1/ThiJ/PfpI superfamily is a large protein group over diverse organisms, under this superfamily, there are multi-types of proteins such as protease, chaperones, heat shock protein, human parkinson's disease protein. The conserved protein from Staphylococcus aureus SAV1875 is a member of DJ-1 superfamily, but its function is unknown. We have determined the crystal structure of SAV1875 to a resolution of 1.8Å . As expected, the overall fold of the core domain of SAV1875 is similar to that of DJ-1. SAV1875 appears to be a dimer both in solution and the crystal, displaying an oligomerization interface similar to that observed for DJ-1. SAV 1875 contains a possible catalytic triad (Cys105-Glu17-His106) analogous to PfpI, YhbO, and DR1199. The cysteine in this triad (Cys-105) is oxidized in this crystal structure, similar to modifications seen in the cysteine of the DJ-1. This Cys-sulfenic acid is stabilized by hydrogen bonding with Glu17, Gly72, His106. We also have determined the crystal structure of mutated form of reactive Cys, SAV1875 C105D to a resolution of 2.1 Å. Aspartate mutation mimics the the Cys-sulfinic acid, more oxidized form. The aspartate stabilization by hydrogen bonding with neighboring residues are maintained. On the basis of these results, we suggest that SAV1875 might work as a general stress protein involved in the detoxification of the cell from oxygen reactive species.

Pullulan conjugates (i.e. PFP1, PFP4) formed via DCC/DMAP coupling of porphyrin derivatives possessing one (P1) or four (P4) carboxylic acid moieties with folate-appended pullulans (PF) were prepared for application as photosensitizers (PS) for photodynamic therapy. To maximize the desired PS characteristics, the assembly size of PS should be less than 200 nm in water medium. In a PBS buffer solution, PFP4 forms smaller assemblies (~100 nm) than PFP1 (~320 nm) due to its enhanced intramolecular hydrophilicity.

Floating offshore wind turbine is a complex rigid-flexible coupling nonlinear system, and the accurate dynamic model is difficultly established. Therefore, the wind-wave interference cannot be improved by adopting the conventional control strategy. In order to solve this problem, an adaptive fuzzy controller (AFC) is used to suppress the dynamic response of floating wind turbine. Two correction factors are introduced to optimize the fuzzy rule, and the traditional fuzzy controller (FC) is firstly obtained. Since the balance positions change and structural parameter perturbation of the wind turbine, an AFC is designed and validated. Finally, the suppression vibration responses ability of floating offshore wind turbine by using the different control strategies is studied under the random wind-wave disturbance and blade pitch control system coupling effect. The simulation results show that the tracking ability of the AFC to the target value is obviously higher than that of the FC; Comparing with the passive control strategy, the suppression vibration effect on the power spectral density (PSD) of the platform pitch (PFPI) motion peak can increase by 39.06% by adopting the AFC.

Compostos butílicos de estanho (BTs) estão na lista dos contaminantes impactantes relacionados ao ambiente marinho. Sua ampla distribuição associada à elevada toxicidade faz com que o monitoramento desses compostos no ambiente costeiro seja de fundamental importância. A ocorrência de BTs foi verificada em sedimentos superficiais, material particulado em suspensão e nos tecidos (fígado e brânquias) de três espécies de bagres estuarinos: Cathorops spixii, Aspistor luniscutis e Genidens genidens; amostrados em 8 estuários da região sul e sudeste brasileira. As análises foram realizadas através de GC-PFPD, seguindo padrões de controle de qualidade analítico e onde foi constatada a ocorrência de efeito matriz sobre o sistema analítico a partir da quantificação das amostras de tecido. Dentre os estuários amostrados, verificou-se um maior impacto dos BTs sobre os sistemas estuarinos de Paranaguá e Itajaí, em decorrência da presença destes compostos em todos os compartimentos ambientais analisados. As três espécies investigadas puderam ser consideradas como biomonitoras da presença de BTs no ambiente, uma vez que são passíveis de assimilação destes compostos, mesmo em organismos amostrados em regiões distantes de potencias fontes de entrada. Os efeitos sobre a fisiologia de C. spixii amostrado em Paranaguá também foi investigada a partir do uso de biomarcadores no fígado desses organismos, constatando um impacto associado à presença destes contaminantes no sistema.
Butyltin compounds (BTs) are in the list of related contaminants striking marine environment. Their widespread distribution associated with high toxicity causes the monitoring of fundamental importance. The occurrence of BTs were checked through superficial sediments, suspended particulate matter and tissues (liver and gill) analysis on three estuarine catfishes: Cathorops spixii, Aspistor luniscutis e Genidens genidens; sampled in eight estuaries from Brazilian south and southeast coast. Analysis were carried by GC-PFPD, following quality control procedures, where was verified the presence of matrix effect over the system from tissues quantification. Among estuaries, the major impact was found in Paranaguá-PR and Itajaí-SC, due the presence of these compounds in all investigated compartments. These three catfish species have shown good biomonitors of BTs presence once were liable of uptake, even in groups sampled far from potential BTs sources. The effects over C. spixii physiology were also checked in Paranaguá from the use of biomarkers on liver of these organisms, noting the impact related with the presence of these compounds on the system.

Few studies have been undertaken which examine the correlation between design of the receptionl/waiting areas of the outpatients' departments and the implications for Private Finance Initiative (PFI) and non-PFI hospitals, in particular the interior environment with reference to user satisfaction. This study investigates to what degree user satisfaction has been achieved in the design of the receptionlwaiting areas in PFI and non-PFI hospitals. The aim of the investigation is to determine whether user satisfaction can be achieved in PFI or non-PFI hospital environments, particular in the outpatients' department. To ascertain whether hospital environments facilitate user friendly and therapeutic characteristics/attributes conducive to user satisfaction, two strands of investigation were undertaken; a) investigation and analysis of PFI and non-PFI hospital design; b) the study of users (PFI and non-PFI) via questionnaire surveys and analysis of their perceptions. The research methods utilised combinations of qualitative information from interviews, discussions with hospital end users, architects/designers and Consortium executives. The surveys undertaken with patients, hospital staff and NHS Trust Managers provided quantitative data to measure the degree to which user satisfaction had been achieved. The main findings of the design analysis identify the strengths and weaknesses in the design of the 'main' and 'sub' reception/waiting areas respectively. The results of the patient surveys, discussions and interviews revealed more positive perceptions of the hospital facilities for PFI hospitals and a general acceptance of the hospital facilities in the non-PFI hospitals. However, the other comments section of the questionnaires reveals some psychological needs of the user were not being met. The hospital staff surveys, discussions and interviews revealed the spatial planning was not ideal for their functional needs. The survey of NHS Trust Managers, Architects/Designers and Building Contractors revealed the difficulties associated with the collaborative process and the implications for the design development process, when reflecting upon 'cost effectiveness' and 'value for money' issues. The conclusions drawn from the study suggest that there is a case for the standardisation of therapeutic environments in the development of 'new build' hospital projects via the design development and collaborative process. The recommendation (see p. 313) provides a design protoeo/that enhance and aids the design development process via selective expertise, which addresses the functional and psychological needs of the hospital end user.

Purpose: To evaluate long-term biocompatibility, optical clarity and the efficacy of Perfluoropolyether (PFPE) polymer as a corneal inlay in humans. Materials and Methods: Corneas of 5 unsighted eyes of 5 patients received inlays: one patient selected as control. All were examined at baseline, immediately post-operative, 7 days and monthly thereafter for 12 months and at 3-monthly intervals for the next 12 months. Clinical evaluation includes detailed slit-lamp biomicroscopy, refraction, keratometry, corneal topography, and other routine ocular examinations. Results: Surgery was uneventful. None of the patients developed corneal oedema or increased vascularisation during study period. All corneas were clear except for minimal haze around the edge of the implant and mild interface reflectivity. Two patients developed epithelial erosion with minimal stromal involvement at 4 and 24 months post surgery resulted in removal of inlay. Inlays remained stable in position in all patients, however clarity has reduced (4% - 26%) gradually with time. We believe mechanical stress from inlay to the surrounding tissue triggered keratocyte activation and extra cellular matrix material deposits around inlay may be responsible for epithelial erosions as well as the reduction in inlay clarity. All test eyes showed acute myopic shift by 8.47D ?? 1.97 (mean ?? SD) following implantation. The epithelial pigment deposits were detected in all test patients following surgery, related to acute corneal curvature changes. The removal data from two patients showed the reversal of the refractive effect to preoperative levels. One patient followed up for an extensive period of 19 months post inlay removal demonstrated stable results. Conclusion: The PFPE inlays have demonstrated excellent biocompatibility within the human corneal tissue for 2 years. The epithelial erosion and inlay clarity reduction were the two major problems encountered in this study. We believe these problems were related to the mechanical stress induced tissue response from inlay. Critical patient selection and the modification of inlay design should improve the success of the procedure. Inlays are effective in altering the refractive status of the eye by altering the anterior corneal curvature. Also, inlay implantation and removal procedures are found to be easy, safe and quick without any complications.

Patellofemoral pain syndrome (PFPS) is one of the most common musculoskeletal disorders affecting adults. Patellar taping provides an effective treatment in alleviating the symptoms of a high proportion of subjects who present with PFPS. However, the mechanisms of pain reduction have not been established. The aim of this work was to evaluate the effects of patellar taping on the biomechanics of the patellofemoral joint and level of pain in PFPS subjects, during the eccentric phase of single leg squatting at 30° of knee flexion. These were determined with a combined kinetic and kinematic approach using data from video analysis, force plate and EMG. A series of pilot tests was performed, involving a group of asymptomatic volunteers performing a number of knee flexion procedures. These tests demonstrated a high reliability in both the estimated patellofemoral joint reaction force (PFJRF) and relative amplitude of vasti, but less impressive reliability for the parameters associated with muscle onset. The main case-controlled study involved 18 PFPS subjects, recruited from a local Physiotherapy Department, and a similar number of healthy controls. The test procedure was performed on the knees of both subject groups and, where appropriate, during and after patellar taping. The results revealed that the mean value of PFJRF, which was 2025 N prior to taping, had decreased significantly to 1720 N (p< 0.05) following a period of taping and exercises which lasted between 2-5 weeks. Equivalent changes were also recorded with the pain levels, although the ratio of the amplitude of the vasti did not change during this period. The only significant differences between the two subject groups were with the relative timing of muscle onset, for which VMO preceded VL in the healthy controls. The reduction of biomechanical characteristics following the application of patellar tape and exercises in PFPS subjects, may explain the mechanism of pain reduction during the early phase of single leg squatting.

Private Finance Initiative (PFI) has successfully transformed the management of public sector construction projects by minimising construction risks and delivering projects with higher certainty in the time, cost, quality, and customer satisfaction than is with the counterpart traditional approach. The success rests in the principles of better Value for Money (VfM) demonstrated through the focus on whole-life costs and quality as the measure for economic, effective, and efficient procurement of the private sector and the delivery of the project and services thereafter. However, PFI has not addressed in full the expectation to provide better VfM due to uncertainties in the estimated project costs and persistent decrease in the specified client requirements as the project advances in the procurement process; hence, attracting distrust and resistance to the initiative. This highlights the need for a new approach to ensure that VfM is not only demonstrated at a point of procurement, but also monitored and sustained over the whole PFI contract period. This research explored the exposure of PFI projects to VfM uncertainty and developed a VfM Optimisation and Sustainability (VaS) model as it main scientific contribution. The developed model works abreast a Process Map, Taxonomy of uncertainty factors, and VfM Simulation and Monitoring Templates to define and develop VfM processes, create VfM, assess and test for its robustness, and sustain it through the entire project life. The model follows from a systematic analysis of the current practice and the uncertainties it creates. The analysis based on an empirical research that adopted a mixed-methods approach involving healthcare and transport PFI projects. The model was evaluated by experts from PFI projects and academics and by using simulation case studies. Adopting the model has the potential to improve the delivery of VfM through optimization of the benefits and costs during the project appraisal process, monitor and sustain VfM during the operating phase, and utilise lessons learnt for improvements in the future.

AbstractUnsafe care results in over 2 million deaths per year and is considered one of the world’s leading causes of death. In 2019, the 72nd World Health Assembly issued a call to action, The Global Action on Patient Safety, that called for Member States to democratize healthcare by engaging with the very users of the healthcare system—patients, families, and community members—along with other partners—in the “co-production” of safer healthcare.The WHO’s Patients for Patient Safety (PFPS) Programme, guided by the London Declaration, addresses this global concern by advancing co-production efforts that demonstrate the powerful and important role that civil society, patients, families, and communities play in building harm reduction strategies that result in safer care in developing and developed countries. The real-world examples from the PFPS Programme and Member States illustrate how civil society as well as patients, families, and communities who have experienced harm from unsafe care have harnessed their wisdom and courageously partnered with passionate and forward-thinking leaders in healthcare including clinicians, researchers, policy makers, medical educators, and quality improvement experts to co-produce sustainable patient safety initiatives. Although each example is different in scope, structure, and purpose and engage different stakeholders at different levels, each highlights the necessary building blocks to transform our healthcare systems into learning environments through co-production of patient safety initiatives, and each responds to the call made in the London Declaration, the WHO PFPS Programme, and the World Health Assembly to place patients, families, communities, and civil society at the center of efforts to improve patient safety.

In order to clarify the degradation mechanisms of the PFPE fluid under a head/disk sliding contact, decomposition reactions of z-dol fluids were investigated under triboplasma generation and in the atmospheres of oxygen, nitrogen and air in gas-discharge-plasma. The results showed that the PFPF degradation proceeds through intermolecular reactions with ambient air molecules by the tribomicroplasma.

The need for precise positioning technique draws significant attention for optical engineering, semi-conductor industry, biotechnology, and nanotechnology, etc. This paper presents a novel piezoelectric (PZT) actuated precision fast positioning system (PFPS), which consists of control module, PZT driving module, precision fast positioning mechanism (PFPM), and micro-displacement measure module. It provides fast and precise control in tilt and piston movements. The driving, mechanism and measure integrative model of the PPS is built. Due to the inherent hysteresis nonlinearity, the PZT actuator always causes positioning error in the open-loop operation and instability in the closed-loop operation. In order to improve the positioning accuracy and response speed, a control method for PZT actuator based on a PID feedback controller with a feedforward compensation is proposed. Finally, the tracking control experiments of piezoelectric actuators for a desired sinusoidal trajectory are performed according to the proposed control method. The experimental results demonstrate that the positioning precision is noticeably improved. The maximum error in tracking a sinusoidal signal is lowered about one order of magnitude in comparison with only using PID controller.

Hydrostatic pressure testing is the most widely accepted approach to verify the integrity of assets used for the transportation of natural gas. It is required by Federal Regulations 49 CFR §192 to substantiate the intended maximum allowable operating pressure (MAOP) of new gas transmission pipelines. The Pipeline and Hazardous Materials Safety Administration (PHMSA) Notice of Proposed Rulemaking (NPRM) with Docket No. PHMSA-2011-0023 [1], proposes an additional requirement for MAOP verification of existing pipelines that: i) do not have reliable, traceable, verifiable, or complete records of a pressure test; or ii) were grandfathered into present service via 49 CFR §192.619(c). To meet this requirement, the NPRM proposes that an Engineering Critical Assessment (ECA) can be considered as an alternative to pressure testing if the operator establishes and develops an inline inspection (ILI) program. The ECA must analyze cracks or crack-like defects remaining or that could remain in the pipe, and must perform both predicted failure pressure (PFP) and crack growth calculations using established fracture mechanics techniques. For assets that cannot be assessed by ILI, however, the implementation of an ECA is hindered by the lack of defect size information. This work documents a statistical approach to determine the most probable PFP and remaining life for assets that cannot be assessed by ILI. The first step is to infer a distribution of initial defect size accumulated through multiple ILI and in-ditch programs. The initial defect size distribution is established according to the as-identified seam type, e.g. low-frequency electric resistance weld (LF-ERW), high-frequency electric resistance weld (HF-ERW), flash weld (FW), single submerged arc weld (SSAW), or seamless (SMLS). The second step is to perform fracture mechanics assessment to generate a probabilistic distribution of PFPs for the asset. In conjunction with the defect size distribution, inputs into the calculation also include the variations of mechanical strength and toughness properties informed by the operator’s materials verification program. Corresponding to a target reliability level, a nominal PFP is selected through its statistical distribution. Subsequently applying the appropriate class location factor to the nominal PFP gives the operator a basis to verify their current MAOP. The last step is to perform probabilistic fatigue life calculations to derive the remaining life distribution, which drives reassessment intervals and integrity management decisions for the asset. This paper will present some case studies as a demonstration of the methodology developed and details of calculation and establishment of database.

Thin-film lubricants play a key role in high-density data storage capabilities via increased reliability and performance of hard disk drive systems. The commercially-used perfluoropolyether (PFPE) lubricants are random copolymers with a linear, polymeric backbone chain structure: X–[(OCF2CF2)p–(OCF2)q]–O–X (p/q ≅ 2/3). Here, X (representing the endgroups) is CF3 in PFPE Z (non-reactive) and CF2CH2OH in PFPE Zdol (reactive), where the hydroxyl groups in PFPE Zdol exhibits strong polar interactions with solid surfaces and with other endgroups. Figure 1 illustrates the representative bead-spring models [1,2] for PFPE Zdol examined in this paper.

Perfluoropolyether (PFPE) lubricants were chemically modified to maximize their thermal stability for application in heat assisted magnetic recording (HAMR). Benzophenone (BP) was introduced to react with the hydroxyl end group in PFPE to act as a free radical stabilizer. Modification of the PFPE by benzophenone was confirmed by evaluation of the chemical shift and the area change of the hydroxyl peak in the 1H NMR spectra. The thermal stability of modified PFPE lubricants was studied using thermo-gravimetry. The improvement in the thermal stability was found to be a function of the amount of substituted hydroxyl groups in PFPE.

The static and dynamic properties of lubricant PFPE are important for the service durability and reliability of the computer head-disk device. Thus molecular dynamic simulations based on a coarse-grained, bead-spring model are adopted to study those properties. On the one hand, we investigate the static properties and infer the structure of both nonpolar and polar PFPE films. For a nonpolar PFPE film, there is a layering structure in the surface layer. And for a polar PFPE film, besides layering structure, there is a bi-polymer structure in the bulk layer. On the other hand, we investigate the dynamic properties and find that for nonpolar PFPE film, a precursor film around one atomic diameter thickness develops according to layering structure; while for polar PFPE film, besides a precursor film, a much steeper and slower spreading shape appears according to bi-polymer structure.

Effect of thermal treatment on the friction and wear of monomolecular layers of SAMs (Self-assembled monolayers) overcoated with PFPE was studied using ball-on-disk tribometer. Two SAMs, Octadecyltrichlorosilane (OTS) and 3-aminopropyltrimethoxysilane (APTMS) were formed on the Si substrate by self-assembly followed by dip-coating with perfluoropolyether (PFPE). Modified Si samples are thermally treated at 150 °C under vacuum for 2h. The physical and chemical properties of modified samples, before and after thermal treatment, are analyzed using contact angle measurements, X-ray photoelectron spectroscopy (XPS). Tribological properties are evaluated using Universal Micro Tribometer (UMT). A Si3N4 ball of 4mm diameter is used as the counterface. Thermal treatment of the PFPE coated SAMs and Si surface reduced the coefficient of friction to slightly lower values in all cases. Thermal treatment has shown slight increase in wear life in the case of PFPE coated APTMS SAM surfaces whereas it decreased the wear life in the case of PFPE coated OTS SAM and Si surfaces. The exact reasons for this behavior are not clear at present, but it is being speculated that thermal treatment effects the extent of bonding between the surface groups of PFPE and SAMs and mobile characteristics of the PFPE, which finally influence the wear durability characteristics. Therefore, the differences in the wear durability characteristics are analyzed using changes in the surface energies and the amount of strongly adsorbed PFPE, as a result of thermal treatment.

Ultraviolet (UV) light exposure can lead to photo degradation of perfluoropolyether (PFPE) lubricants, resulting in a change of their physical properties. In this paper, 2,3,4,5,6-pentafluorobenzophenone was used as ultraviolet stabilizer (UVS) to slow down the photo degradation of PFPE. PFPE/UVS mixtures were exposed to UV light and the mechanism of photo degradation was studied with nuclear magnetic resonance (NMR). Small amounts of UVS (less than 0.3 wt%) were found to effectively stabilize both UV photo degradation at the main chain and the end chain of PFPE.

The overall theme of this project was to increase the speed of analysis for monitoring pesticide residues in food. Traditionally, analytical methods for multiple pesticides are time-consuming, expensive, laborious, wasteful, and ineffective to meet critical needs related to food safety. Faster and better methods were needed to provide more cost-effective detection of chemical contaminants, and thus provide a variety of benefits to agriculture. This overarching goal to speed and improve pesticide analysis was successfully accomplished even beyond what was originally proposed by the investigators in 1998. At that time, the main objectives of this project were: 1) to further develop a direct sample introduction (DSI) device that enables fast sampling and introduction of blended-only agricultural products for analysis by gas chromatography (GC); 2) to evaluate, establish, and further develop the method of simultaneous pulsed flame photometric detector (PFPD) and mass spectrometry (MS) detection for enhanced pesticide identification capabilities; and 3) to develop a new and novel MS pesticide analysis method, based on the use of supersonic molecular beams (SMB) for sampling and ionization. The first and third objectives were successfully accomplished as proposed, and the feasibility of the second objective was already demonstrated. The capabilities of the GC/SMB-MS approach alone were so useful for pesticide analysis that the simultaneous use of a PFPD was considered superfluous. Instead, the PFPD was investigated in combination with an electron-capture detector for low-cost, simultaneous analysis of organophosphorus and organochlorine pesticides in fatty foods. Three important, novel research projects not originally described in the proposal were also accomplished: 1) development of the quick, easy, cheap, effective, rugged, and safe (QuEChERS) method for pesticides in foods; 2) development and optimization of a method using low-pressure (LP) GC/MS to speed pesticide residue analysis; and 3) innovative application of analyte protectants to improve the GC analysis of important problematic pesticides. All of the accomplishments from this project are expected to have strong impact to the analytical community and implications to agriculture and food safety. For one, an automated DSI approach has become commercially available in combination with GC/MS for the analysis of pesticide residues. Meanwhile, the PFPD has become the selective detector of choice for the analysis of organophosphorus pesticides. Great strides were made in SMB-MS through the manufacture of a prototype "Supersonic GC/MS" instrument, which displayed many advantages over commercial GC/MS instruments. Most notably, the QuEChERS method is already being disseminated to routine monitoring labs and has shown great promise to improve pesticide analytical capabilities and increase lab productivity. The implications of these developments to agriculture will be to increase the percentage of food monitored and the scope of residues detected in the food, which will serve to improve food safety. Developed and developing countries alike will be able to use these methods to lower costs and improve results, thus imported/exported food products will have better quality without affecting price or availability. This will help increase trade between nations and mitigate certain disputes over residue levels in imported foods. The improved enforcement of permissible residue levels provided by these methods will have the effect to promote good agricultural practices among previously obstinate farmers who felt no repercussions from illegal or harmful practices. Furthermore, the methods developed can be used in the field to analyze samples quickly and effectively, or to screen for high levels of dangerous chemicals that may intentionally or accidentally appear in the food supply.
