As doenças cardiovasculares representam a principal causa de morte nos países ocidentais. Dentre essas doenças, a aterosclerose é que mais se destaca, sendo caracterizada pelo acúmulo de células musculares lisas vasculares (CMLV). O efeito patológico das CMLV em resposta a diferentes estímulos pode acarretar em disfunções nestas células. É notável que a aterosclerose ocorra principalmente em vasos sinuosos onde ocorre um forte turbilhonamento do fluxo sanguíneo, que pode acarretar em hemólise e, consequentemente, acúmulo de heme livre. Além disso, no processo de aterogênese as moléculas de adesão, principalmente integrinas, são de crucial importância durante a resposta de CMLV. Nesse trabalho nosso objetivo inicial foi avaliar o efeito do heme livre nas funções de CMLV, bem como os mecanismos moleculares por trás desses efeitos. Em uma segunda parte, investigamos o envolvimento da integrina α1ß1 no efeito da Angiotensina II (Ang II) em CMLV. Nós observamos que o heme livre é capaz de induzir a proliferação e migração de CMLV via espécies reativas de oxigênio (ERO) provenientes da NADPHoxidase (NADPHox). Adicionalmente vimos que o heme ativa vias de sinalização redox-sensíveis relacionadas à proliferação celular, como MAPKinases e o fator de transcrição NFκB. Também observamos que há uma ligação entre a NADPHox e o sistema heme oxigenase (HO), uma vez que o heme induz a expressão de HO-1 e o pré-tratamento das CMLV com inibidores de HO levam ao aumento tanto o efeito proliferação quanto a indução de ERO promovidas pelo heme. Além disso, vimos que o efeito contra-regulatório promovido pela HO ocorre devido as metabolites do heme: biliverdina, bilirrubina e monóxido de carbono. Por último, quando bloqueamos tanto a NADPHox quanto o sistema HO o heme não teve efeito algum na proliferação de CMLV. Em um segundo estudo, observamos que o efeito da Ang II sobre a migração de CMLV foi inibido quando as células foram pré-tratadas com o ligante da integrina α1ß1, a desintegrina Obtustatina. A seguir observamos que o efeito da Ang II na ativação de FAK e na colocalização actina-ILK é dependente da integrina α1ß1, que possivelmente ativa PKCα, uma vez que vimos que a produção de ERO induzida por Ang II foi inibida pela Obtustatina. Vimos que a indução da expressão de ILK por Ang II em CMLV é dependente da integrina α1ß1 e também observamos que a Obtustatina inibibiu o desacoplamento de ILK da FAK, uma vez que a Obtustatina bloqueou a fosforilação de FAK induzida por Ang II (processo crucial para o desacoplamento da ILK). Nós também observamos que a Ang II induz, via integrina α1ß1, a fosforilação de AKT e a diminuição da expressão de p21, provavelmente via ILK. Corroborando estes dados, nós mostramos que o pré-tratamento com Obtustatina induziu um estacionamento na fase G0 e diminuição da proliferação de CMLV tratadas com Ang II. Portanto, mostramos nesse trabalho que o heme livre induz a ativação de CML via NADPHox, que é elegantemente contra-regulado pelo sistema HO. Além disso, sugerimos que a integrina α1ß1 pode ser um importante alvo molecular para o desenvolvimento de intervenções mais efetivas para a aterosclerose.
Cardiovascular diseases represent the major mortality reason in western countries. Among these diseases, atherosclerosis is the most prominent one, which is characterized by vascular smooth muscle cell (VSMC) accumulation. The pathological effect of VSMC in response to different stimuli is able to induce VSMC dysfunctions. Notably, this cardiovascular disease occurs mainly in sinuous vessels with turbulent blood flow, which may lead to hemolysis and consequent free heme accumulation. Furthermore, in atherogenesis the adhesion molecule, mainly integrins, were of crucial importance during the VSMC response. In this work our aim was to elucidate the effect of free heme in VSMC, as well the molecular mechanisms underlying this process. In a second part, we investigated the role of α1ß1 integrin in Angiotensin II (Ang II) effect on VSMC. We observed that free heme is able to induce VSMC proliferation in a Reactive Oxygen Species (ROS) derived from NADPHoxidase (NADPHox) dependent manner. Additionally, heme activates proliferation-relationed redox-sensitive signaling routes, such as MAPKinases and the transcription factor NFκB. It was also observed a critical crosstalk between NADPHox and heme oxygenase (HO) system, once heme induces HO-1 expression and VSMC pretreatment with HO inhibitors increased heme proliferative effect and ROS production. Accordingly, we observed that the counter-regulatory effect promoted by HO occurs due heme metabolites: biliverdin, bilirubin and carbon monoxide. Finally, when both NADPHox and HO system were blocked, heme had no effect on VSMC proliferation. In a second part, we observed that the chemotactic effect of Ang II on VSMC was abolished when the cells were pretreated with the α1ß1 integrin ligand, the disintegrin Obtustatin. Then, we observed that the Ang II effect on FAK activation and actin-ILK colocalization is integrin α1ß1 dependent, which possibly activates PKCα, once we observed that the ROS production induced by Ang II was inhibited by Obtustatin. We demonstrated by western blotting that ILK induction by Ang II is dependent of α1ß1 integrin and we also observed that Obtustatin inhibited the uncoupling of ILK to FAK, once Obtustatin blocked the FAK phosphorylation induced by Ang II (crucial process to ILK uncoupling). We also observed that Ang II induced, via α1ß1 integrin, AKT phosphorylation, and p21 expression reducement, probably via ILK. Corroborating these data we demonstrated that the pretreatment with Obt induced G1 phase arrest and diminishment of VSMC proliferation treated with AngII. Thus we showed that free heme induces VSMC activation via NADPHox, which is elegantly counter-regulated by HO-1. Furthermore, we suggest that α1ß1 integrin may be an important target molecule to the development of more effective therapeutic interventions in atherosclerosis.

Повний текст джерелаObjectives: To determine if the use of RxSkills effectively improves student scores on the North American Pharmacist Licensure Examination (NAPLEX)-based questions among third and fourth-year pharmacy students at the University of Arizona (UA) College of Pharmacy. Methods: This is a pre-post interventional study using a survey of volunteer students from the classes of 2016 and 2017 at the UA College of Pharmacy. An email calling for volunteers was sent using the listserv to students providing information regarding the study. Once the pre-test was completed, instructions for downloading the mobile application were sent to the students. After six weeks of RxSkills use, a link to the post-test was sent to the students for completion. Student scores on the NAPLEX-based questions, use of the application (frequency and duration) and helpfulness, and demographic data were collected. Results: The number of participants in the study from each class was similar with 15 students from the Class of 2016 (52%) and 14 from the Class of 2017 (48%). There was no correlation between how often RxSkills was used and improvement in scores. Overall, student score on the NAPLEX-based questions were significantly improved post-RxSkills use (mean 10.48, SD 2.49) compared to pre-RxSkills use (mean 9.68, SD 2.24) with a p-value of 0.03. Conclusions: The use of RxSkills resulted in an improvement in student scores on the NAPLEX-based questions, indicating its usefulness in studying for the NAPLEX. Students would benefit from using the application when studying for this exam.

Повний текст джерелаReplace of missing teeth by osseointegrated implants has become an accepted treatment modality in clinical dentistry. The clinical efficacy of implant therapy has been demonstrated through a myriad of long-term studies following an established criteria for success and showing high survival rates. In those studies most of the failures were concentrated at the second stage surgery and in the first year following the insertion of the prostheses. In the present study , the success of 371 implants of the NAPIO (Center of Supporting Research in Dental Implants) implant system placed in function for 2 years were evaluated. These implants were used to support 135 single units and a variety of fixed and removable prothesis placed in 109 patients presenting partial and complete edentulism. The gender distribution encompassed 43% of males and 66% of females. The age range was between 17 and 82 years old. The implants were made of titanium grade I and acid etched. Clinical and radiografic evaluation were done every 6 months in the first year and then once a year for 2 years. The clinical evaluation was performed using several known periodontal parameters such as, probing depth, attachment level, presence of plaque, suppuration, edema and color alteration. For radiographic assessment, a standardized technique using an individually customized film holder with acrylic resin was used. The implants were evaluated using the success criteria described by SMITH; ZARB186. The success index for the mandible was 95.90% and for the maxilla 90.21% reflecting a clinical efficacy similar to others osseointegrated implant systems. Of the failing implants, 77% of those placed in the maxilla and 45.5% of the ones placed in the mandible were detected at second surgery, and were all of short length. The radiographs were digitized and measured using the software Sigma Scan Pro 4 (Jandel Scientifics) and the bone loss calculated for every time point. Results demonstrated an overall bone loss of 0.99 mm during 2 years. There were very little changes in probing depth and attachment level during the first 2 years and, almost no changes were found using parameters to access inflammation of the peri-implant tissue, presence of plaque, edema and bleeding on probing. The most frequent complication found in the prostheses assessment was loosening and fracturing of the retaining screw. The grades of confort, esthetics and function of the prostheses were assessed by the patients and the results were superior to 9.4 attesting the efficacy of the treatment from the patients point of view. It was concluded that the results found for clinical parameters, implants success, bone loss measured radiografically and complications found in the prosthesis during the first two years of follow-up were in agreement with results presented in the literature for other osseointegrated implant systems.

Повний текст джерелаThe main goal of this work has been the synthesis of tungstate phosphate glasses based on sodium polyphosphate NaPO3 and tungsten oxide WO3 with increasing amounts of lead fluoride PbF2 and doped with rare earth ions in order of obtaining new glass compositions for optical applications. The investigated ternary system has been NaPO3-WO3-PbF2 with WO3 being used as a glass intermediary for higher chemical and termal stabilities. Firstly, glass compositions were investigated in the binary system (100-x)NaPO3-xWO3 with x=20, 30 and 40% in order to define the best NaPO3/WO3 ratio for further PbF2 incorporation. The molar ratio 60NaPO3-40WO3 has been identified as the best precursor glass for lead fluoride incorporation. Glass samples were obtained by the melt-quenching method in the ternary system NaPO3-WO3-PbF2 with PbF2 concentration varying from 0 to 60 mol% and a fixed NaPO3/WO3 molar ratio of 3/2. Synthesis conditions such as melting time and melting temperature were improved for the formation of transparent, homogeneous and stable glasses with low fluorine loss during the melting process. Two series of rare earth doped glasses were then prepared: the first series has been synthesized with 0,2% Er3+, 1% Yb3+ and increasing amounts of PbF2 in order to investigate the luminescent behavior of Er3+ in function of PbF2 concentration. The second series has been prepared with 0,2% Eu3+ and increasing amounts of PbF2 with the aim to use the Eu3+ luminescence as a structural probe. These glasses were characterized by DSC in order to determine the termal behavior in function of composition. Structural investigations were also performed by Raman scattering and allowed to identify the modifier behavior of PbF2 in the covalent network with a progressive break of the covalent bridging bonds. A crystallization study performed on the most PbF2 concentrated glass samples (40%, 50% and 60%) identified a preferential precipitation of lead fluorophosphate Pb5F(PO4)3 for samples containing 40% and 50% of PbF2 with a dominant volume crystallization mechanism while sample containing 60% of PbF2 exhibited precipitation of cubic ß-PbF2 with a dominant volume crystallization. UV-Vis absorption spectra obtained on Er3+/Yb3+ codoped glass samples allowed to identify absorption bands due to f-f electronic transitions and the absorption bands related with the dark color of less PbF2 concentrated glass samples. Emission spectra of Eu3+ doped glasses in the visible range pointed out changes in the intensity ratio between electronic transitions 5D0?7F2 and 5D0?7F1, related with structural changes in the Eu3+ neighborhood with a symmetry increase with increasing PbF2 content, probably related with progressive coordination of Eu3+ with F-. Finally, excitation and emission spectra of Er3+/Yb3+ codoped glass samples also pointed out important changes in the luminescent properties in function of PbF2 concentration related with structural changes around rare earth ions as well as a higher energy transfer efficiency between Er3+ and Yb3+ ions with increasing PbF2 contents

O recozimento foi realizado nesta temperatura por 1 h para minimizar as tensões mecânicas. As propriedades dielétricas e a dependência da condutividade com a composição química, temperatura e freqüência foram avaliadas pela espectroscopia de impedância (IS). Um mecanismo de condução iônico foi observado para composições de WO3 < 30 %mol. Uma mistura entre os mecanismos de condução iônica e eletrônica foi observada para WO3 > 30 %mol. Aglomerados das unidades de WO6 foram observados nas amostras contendo grandes quantidades de WO3 sugerindo uma possível conexão entre estes aglomerados e os mecanismos de condução. As técnicas espectroscopia no infravermelho com transformada de Fourier (FT-IR) e difração de raios-X foram usadas para a caracterização no presente trabalho.
