In their post-authoritarian period, civilian governments in transitional democracies have often been battered by unelected power centers. Where do these unelected forces derive their power? This article addresses this question through a case study of Pakistan. Since the 2018 elections, a decade after leaving formal political office, the Pakistani military has asserted greater control over civilian government. Using the concept of informal institutions of political participation as an analytical framework, we argue that when formal forms of control become untenable due to legitimacy and/or functional constraints, the military turns into a Janus-faced institution, visibly acting as a formal state organ while invisibly protecting its institutional interests through what we call “informal mechanisms.” The article explains how Pakistan’s pre-2018 political situation dictated a quasi-military regime more suited to the military’s interests than direct military rule.

Abstract Endogenous thymic regeneration is a crucial function that allows for renewal of immune competence following immunodepletion caused by common cancer therapies such as cytoreductive chemotherapy or radiation; however, the mechanisms governing this regeneration remain poorly understood. Moreover, despite this capacity, prolonged T cell deficiency is a major clinical hurdle in recipients of hematopoietic stem cell transplantation (HSCT) and can precipitate high morbidity and mortality from opportunistic infections, and may even facilitate malignant relapse. We have recently described a central role for group 3 innate lymphoid cells (ILC) in a complex cellular and molecular network that drives endogenous thymic regeneration (Dudakov 2012 Science 336:91). Although IL-22 contributes considerably towards thymic regeneration and mice deficient for IL-22 lag behind WT controls in their recovery of thymic function, there is still some tissue regeneration in these mice, suggesting that other regeneration pathways also contribute to thymic repair. Unbiased transcriptome analysis on the damage-resistant non-hematopoietic compartemtn of the thymus revealed significant upregulation of Bmp4 and its downstream signalling targets (Fig. 1a). Further interrogation revealed that while thymic expression of BMP4 was restricted to fibroblasts and endothelial cells (ECs), only ECs increase their expression of Bmp4 after damage; and specific and inducible deletion of BMP4 in ECs led to significantly worse regeneration (Fig. 1b). Thymopoiesis is dependent on the close interaction between developing thymocytes and the non-hematopoietic stromal microenvironment, which includes highly specialized thymic epithelial cells (TECs) and ECs. While the role of TECs has been well studied, the contribution of ECs to thymopoiesis and thymic regeneration has thus far remained largely unclear. Careful interrogation of ECs after damage revealed that, much like ILCs, ECs are extremely resistant to multiple clinically relevant models of acute tissue injury including corticosteroids, chemotherapy and TBI. However, whole organ imaging analysis using light sheet field microscopy suggested that even though the number of ECs remain unchanged after damage, there is considerable structural changes to the vasculature including shortening of the vessels and reduced branching. Although BMP4 receptors are widely expressed in the thymus, there was enriched expression for BMP4 receptor subunits on TECs, which is consistent with the role of BMP4 in thymus ontogeny by promoting TEC development, at least partially due to its ability to induce expression of Foxn1 (Fig. 1c), a key transcription factor for the development and maintenance of TECs. Consistent with these findings, after thymic damage we observed a significant increase in the expression of Foxn1 after damage as well as GSEA enrichment for downstream FOXN1 target genes (Fig. 1d); including Dll4, the Notch ligand critical for T cell development and whose concentration we have previously shown can directly regulate thymic size (Velardi 2014 J Exp Med 211:2341). Finally, using a technique whereby ECs are transduced with the adenoviral gene E4ORF1 - ECs could be expanded ex vivo (exEC) and, when administered to mice after SL-TBI, significantly boost recovery of thymic function; but only when the exEC were derived from the thymus but not from heart or kidney (Fig. 1e). Consistent with endogenous regeneration, in vivo administration of exEC(Thy) induced the expression by TECs of Foxn1 and Dll4 . Here we demonstrate that rather than just being passive conduits that deliver oxygen and nutrients, ECs are active participants in organ function producing distinct paracrine factors that orchestrate thymic renewal. These studies thus not only detail a novel pathway promoting endogenous thymic regeneration, but also offer an innovative clinical approach to enhance T cell immunity in recipients of allo-HSCT and for individuals with T cell deficiencies due to aging, infectious disease, and common cancer treatments such as chemo- and radiation-therapy. This thesis considers modelling one of the mechanisms, which governs the formation of wool fibres. The problem is investigated by constructing a mathematical model that is based on the geometry of the ultrastructure of the wool fibres. The deformed shapes of the wool fibres are obtained by minimising the strain energy calculated from the mathematical model. The objective of this study is to explore the relationship between the ultrastructure and the crimpy shape of the wool fibres. The first chapter gives the background information of wool fibre, the outline of the thesis and defines the problem. The second chapter examines the structures inside the wool fibres and the mechanisms, which govern the shapes of the wools fibres. The variables used to describe the fibre shapes are introduced here. Then, some simple examples are given to demonstrate the mathematical model. In the third chapter, the structures of the wool fibres are simplified with several assumptions and used to construct the basic model. Then, some results are given with various values of different parameters to demonstrate the properties of the model. Some modifications of the model are made, which include the addition of the cuticle shell and an approximate analytical model by Taylor's expansion. The fourth chapter develops another modification of the mathematical model with the addition of a 'macrofibril' structure. The fifth chapter gives the last modification of the model uses integration to calculate the total energy of the whole fibre. The sixth and the seventh chapters show the practical methods to measure the natural shape of a single fibre and the cross-section pattern of the fibre respectively. The measured natural shape of the wool fibre can be used to verify the local estimated shape of the wool fibre, which is calculated by the mathematical model with the measured cross-section pattern.

Metastatic dissemination of ovarian tumors involves the invasion of multi-cellular tumor cell clusters into the mesothelial cell lining of organs in the peritoneal cavity. We developed an in vitro assay that models this initial step of ovarian cancer metastasis to investigate the mechanisms of invasion. Pre-clustered ovarian cancer multicellular spheroids are incubated with GFP-expressing mesothelial monolayers and the extent of mesothelial invasion is monitored by time-lapse video microscopy.

The processes involved in mammary gland development are intimately linked with those that drive breast oncogenesis. Regulation of growth, tissue polarity and genome stability are a few of the factors the maintain homeostasis in breast epithelium and prevent malignant progression. In this work, a series of clonal 184-hTERT cell lines were generated that modeled the in vitro growth characteristics of bi-potent mammary progenitor cells; they were dependent upon fibroblasts for low-density growth and formed dual-lineage acini in 3D culture. These lines were subsequently used in a genome-wide siRNA screen to identify the factors that regulate fibroblast-driven epithelial cell growth. Fibroblasts constitute the majority of cells within stroma, which plays a major role in supporting mammary progenitor cell growth. From this screen, 49 surface and secreted factors were identified that putatively transduce the signals emanating from the fibroblasts that are required for epithelial cell growth. These factors were more potent than any of the previously described growth factor receptors. When assessed in primary tissue, Gpr39, Scarb2, Ntn1, Efna4, Nptx1, and Ctnna1 were found to have the greatest effect on overall progenitor cell growth, while SerpinH1 differentially suppressed luminal progenitor cells, and Nkain4 and Kcnj5 differentially suppressed bi-potent progenitor cells. Further profiling of these lines identified the planar cell polarity protein Celsr1 as differentially regulated under fibroblast-dependent conditions. Silencing of Celsr1 increased the number of bi-potent progenitor cells detected in the colonyforming assay. Furthermore, it induced branching morphogenesis within normally spherical acini and disrupted the apical polarity of these structures in 3D culture. Within this system, Celsr1 is suspected of signaling through Shisa4. This is the first description of a noncanonical Celsr1 interactor. Finally, a curious variant line was identified amongst the collection of 184-hTERT cells generated for this work. This line harbours mitotic spindle and cell cycle checkpoint defects, and rapidly gains chromosome 20 during passaging. Through an elimination process, de novo promoter hypomethylation and subsequent overexpression of CENPI was identified as likely being responsible for this phenotype. This is the first description of CENPI deregulation and one of a few descriptions of gene promoter hypomethylation resulting in genome instability.

We produced self-assembled quantum dot (QD) samples of InAs on GaAs by molecular beam epitaxy (MBE). With these, we explored growth effects as a function of InAs coverage for three arsenic pressures, and as a function of arsenic pressure at a specific InAs coverage. During growth, the samples were studied using reflection high energy electron diffraction (RHEED). These RHEED measurements were compared to low energy electron diffraction (LEED) measurements. To perform this ex-situ LEED characterisation, some samples were covered with an amorphous arsenic cap. This cap was thermally evaporated producing a clean, non-oxidised surface that was studied using LEED. We obtained non-ambiguous identification of the GaAs (001) surface reconstructions as well as timing information for the 2D to 3D transition during the growth of InAs on GaAs. Post growth characterisation of two sets of self-assembled QD samples, twelve samples in all, revealed the following: As a function of increasing the arsenic pressure used in QD growth, the photoluminescence (PL) of capped QDs is first redshifted at low arsenic pressures, and then blueshifted at high arsenic pressures. Scanning electron microscopy and atomic force microscopy of uncapped QDs show that as the arsenic pressure increases, the QD density increases while the average QD width and height decrease monotonically; these trends are consistent with the shift in PL for the high arsenic pressure samples, but are inconsistent with the shift in PL for the low pressure samples. This leads us to proposing a mechanism by which QDs may be modified as they are overgrown with capping material. We discuss the effects of adjusting the arsenic pressure on the formation of QDs and the mechanism by which QDs may be modified during capping.

Breakdown of electrical equipment is unwanted and the use of reliable methods of insulation are therefore necessary. Liquid insulation is one preferred material for electrical devices used in subsea installations due to outstanding qualities related to insulation, heat transfer, safety and incompressibility properties. To ensure that the selected insulating liquid is suitable for the specific equipment it will be necessary to test its insulating quality. This is verified by stressing the liquid with voltages higher than the partial discharge inception voltage (PDIV). While there are existing tests for these properties, such as use of partial discharge measurements, this thesis examines an experimental approach to testing the properties of different liquids in a point-plane gap through the use of high AC voltage.Charges, like ions, in an electrical field creates a current flow. In a liquid, dissociative ionization under applied voltage is the mechanism that generates the most charges. Field emission originating from the negative polarity contributes in some liquids. Moving charges in the liquids results in measurable currents of the order 10 - 80 nA under applied AC voltage of 20 kV$_{peak}$ in a 20 mm point-plane gap in different liquids. Space charges are also found to affect the partial discharge (PD) behaviour in liquids. Different liquids have different chemical and physical properties and therefore different PD behaviour. It has been observed that PDs are stochastic, and in some sense correlated to earlier stress due to residual ions from previous half periods and permanently change of the chemical structure of some liquid molecules after a PD. The PD rate increases exponentially with increasing voltage, while the maximum charge per half period tends to increase linearly. This observed behaviour is in agreement with what is found in earlier reports.The presence of free electrons is important. A significant increase in the rate of PD at low magnitudes in both polarities occurred when the test cell was exposed to X-rays. PDs start under the influence of a strong field or an electron avalanche. It is essential to be able to test different liquids in laboratory conditions, for economical reasons, in order to identify the most suitable insulating media. Power electronics utilizing quality insulation is more reliable with a lower probability of downtime. The test setup and methods used in this thesis may easily be adapted for test of electrical components and equipment instead of the simple point-plane gap method.Different liquids have different PD phase patterns. This thesis intends to describe the reasons to explain this difference. It is found to be the result of different properties in the liquids for charge creation and differences in their electrical field threshold for PD initiation. PDs of a certain size are rarely occurring events and are strongly dependent on the prehistory of the stressed liquid gap. We do not know whether PDs in itself is harmful to insulating properties or not. It is therefore uncertain whether the IEC 61294 test method, based on PDs, provides any useful information or not. More research is therefore needed in order to fully understand the PD phenomenon.

A naturally occurring sorbent, kaolinite, is injected into a down-flow pilot scale combustor burning natural gas, doped with a surrogate metal containing waste (usually lead acetate). When sorbent is not introduced, lead vaporized in the flame ultimately forms a unimodal particle size distribution centered at approximately 0.1 μm., often referred to as fume. The injection of sorbent prevents the formation of this fine particulate, by reactively scavenging the lead as vapor above its dewpoint temperature. The approximately 1 μm lead alumina-silicate particles formed capture nearly 100% of the lead, yet yield very low water soluble lead mass fractions. Thus, lead is chemically fixed in particles that are approximately an order of magnitude larger in size than the lead aerosol fume. Consequently, air pollution control systems can capture these particles relatively efficiently, and the metal's isolation from the environment is ensured when subsequently disposed of in a landfill. Chlorine is a commonly occurring halogen in many incinerable waste streams. With increasing Cl/Pb ratio, the mass fraction of lead captured decreases. At a chlorine gas injection rate resulting in a Cl/Pb ratio of 2/1, 10/1, and 17/1, the obtained mass fraction of lead captured was 79.7%, 44.0%, and 37.8%. The uncaptured lead remained in the gas phase as the relatively volatile PbCl₂, which formed a water soluble fume upon temperature quench in the sampling probe. These results indicate that sorbent injection could be less effective at metal capture for incinerable waste streams with both chloride and metal fractions. A multi-component aerosol simulations package (MAEROS) is utilized to model the temporal evolution of a lead aerosol particle size distribution in the combustor. Base case simulations model the Pb-O₂ system, and accounts for homogeneous nucleation, physical condensation, and coagulation. These simulations match the data fairly well under conditions which promote self nucleation (i.e. steep temperature profiles). The MAEROS model is also utilized to simulate the Pb-O₂-sorbent and Pb-O₂-Cl₂-sorbent systems in an attempt to elucidate the lead capture mechanisms. Also, the model-fitted intrinsic reaction rates are much greater than the rates obtained from the lead/sorbent bench scale experiments from the literature.

A key issue is how public values can be forced upon the ecosystem’s architecture—an architecture whose core is overwhelmingly controlled by (US) tech giants pushing economic values and corporate interests, often at the expense of a (European) focus on social values and collective interests. The mechanisms of datafication, commodification, and selection seem to afford tech companies unprecedented infrastructural, sectoral, and intersectoral powers. However, the ecosystem’s architecture is adaptable to changing societal norms and awareness about potential harms. This book’s search for underlying patterns and systemic mechanisms prompts a final reflection on the “what,” “how,” and “who” of governance: what kind of public values do we want to incorporate into the design of the platform society, how do we do that, and who is responsible for doing so?

This chapter examines key legal instruments and mechanisms relevant to international renewable energy regulation. These play an important role in governing unified action and enhancing collaboration and information-sharing on effective policies and investment frameworks aimed at reducing barriers and risks to investments in renewable energy. The mechanisms that are analysed are the International Renewable Energy Agency (IRENA) Statute, the United Nations Framework Convention on Climate Change (UNFCCC), the Kyoto Protocol and related international climate change negotiations and declarations, the Energy Charter Treaty (ECT), and various sector-specific treaties. The chapter also turns its attention to the primary international organizations that influence present and future directions in international renewable energy policy, such as the Nairobi Programme of Action for the Development and Utilization of New and Renewable Sources of Energy, International Energy Agency, Development Banks, and the Renewable Energy and Energy Efficiency Partnership.

On the received view, the resolution of context-sensitivity is at least partly determined by non-linguistic features of utterance situation. If I say ‘He’s happy’, what ‘he’ picks out is underspecified by its linguistic meaning, and is only fixed through extra-linguistic supplementation: the speaker’s intention, and/or some objective, non-linguistic feature of the utterance situation. This underspecification is exhibited by most context-sensitive expressions, with the exception of pure indexicals, like ‘I.’ While this received view is prima facie appealing, I argue it is deeply mistaken. I defend an account according to which context-sensitivity resolution is governed by linguistic mechanisms determining prominence of candidate resolutions of context-sensitive items. Thus, on this account, the linguistic meaning of a context-sensitive expression fully specifies its resolution in a context, automatically selecting the resolution antecedently set by the prominence-governing linguistic mechanisms.

The Roman Empire systematically recognised alien polities in provincial landscapes and allowed them local powers of jurisdiction and legislation. It also created conditions of heightened human mobility and regularly endowed aliens with Roman citizenship. This chapter explores the procedural and theoretical resources developed by Roman authorities to deal with the conflicts of law that resulted from these processes. It explores principles governing choice of law, to wit, those of personality and territoriality. It also investigates procedural mechanisms that temporarily bracketed distinctions that rendered actions non-justiciable, for example, the legal fiction that assimilated aliens to citizens for the purposes of dispute resolution.

AbstractAgainst the backdrop of an energy system moving from vertically integrated monopolies towards a decentral system with a multitude of actors in ever-changing roles, we observe a gradual strengthening of central governance mechanisms on the nation-state and on the European level. Such a top-down approach to the governance of the energy system might have been necessary to open up energy markets to competitive processes and innovation. With social goals shifting and security of supply and environmental concerns gaining importance, the governance of the energy system has to be reshaped anew, enabling, e.g., the optimization of regional energy systems by local actors. In particular, strict unbundling rules may hinder or preclude system-serving behavior, to the detriment of all market participants and consumers. Lawmakers and regulators should provide some leeway to cooperative approaches, such as the empowerment of local actors to devise their own energy regimes.

Compliant mechanisms are defined as mechanisms that gain some, or all of their mobility from the flexibility of their members. Suitable use of pseudo-rigid-body models for compliant segments, and relying on the state-of-the-art knowledge of rigid-body mechanism synthesis types, greatly simplifies the design of compliant mechanisms. Assuming a pseudo-rigid-body four-bar mechanism, with one to four torsional springs located at the revolute joints to represent mechanism compliance, a simple, heuristic approach is provided to develop various compliant mechanism types. The synthesis with compliance method is used for three, four and five precision positions, with consideration of one to four torsional springs, to systematically develop design tables for standard mechanism synthesis types. These tables appropriately reflect the mechanism compliance by specification of either energy or torque. Examples are presented to demonstrate the use of weakly or strongly coupled sets of kinematic and energy/torque equations, as well as different compliant mechanism types in obtaining solutions.

Original objectives: The long-term goal of the research is to achieve higher protein content in the milk of ruminants by modulating the translational apparatus of the mammary gland genetically, nutritionally, or pharmacologically. The short-term objectives are to obtain a better understanding of 1) the role of amino acids (AA) as regulators of translation in bovine and mouse mammary epithelial cells and 2) the mechanism responsible for the synergistic enhancement of milk-protein mRNA polyadenylation by insulin and prolactin. Background of the topic: In many cell types and tissues, individual AA affect a signaling pathway which parallels the insulin pathway to modulate rates and levels of protein synthesis. Diverse nutritional and hormonal conditions are funneled to mTOR, a multidomain serine/threonine kinase that regulates a number of components in the initiation and elongation stages of translation. The mechanism by which AA signal mTOR is largely unknown. During the current grant period, we have studied the effect of essential AA on mechanisms involved in protein synthesis in differentiated mammary epithelial cells cultured under lactogenic conditions. We also studied lactogenic hormone regulation of milk protein synthesis in differentiated mammary epithelial cells. In the first BARD grant (2000-03), we discovered a novel mechanism for mRNA-specific hormone-regulated translation, namely, that the combination of insulin plus prolactin causes cytoplasmic polyadenylation of milk protein mRNAs, which leads to their efficient translation. In the current BARD grant, we have pursued the signaling pathways of this novel hormone action. Major conclusions/solutions/achievements: The positive and negative signaling from AA to the mTOR pathway, combined with modulation of insulin sensitization, mediates the synthesis rates of total and specific milk proteins in mammary epithelial cells. The current in vitro study revealed cryptic negative effects of Lys, His, and Thr on cellular mechanisms regulating translation initiation and protein synthesis in mammary epithelial cells that could not be detected by conventional in vivo analyses. We also showed that a signaling pathway involving Jak2 and Stat5, previously shown to lead from the prolactin receptor to transcription of milk protein genes, is also used for cytoplasmic polyadenylation of milk protein mRNAs, thereby stabilizing these mRNAs and activating them for translation. Implications: In vivo, plasma AA levels are affected by nutritional and hormonal effects as well as by conditions of exercise and stress. The amplitude in plasma AA levels resembles that applied in the current in vitro study. Thus, by changing plasma AA levels in the epithelial cell microenvironment or by sensitizing the mTOR pathway to their presence, it should be possible to modulate the rate of milk protein synthesis. Furthermore, knowledge that phosphorylation of Stat5 is required for enhanced milk protein synthesis in response to lactogenic opens the possibility for pharmacologic approaches to increase the phosphorylation of Stat5 and, thereby, milk protein production.
