3018 Background: Vascular endothelial growth factor (VEGF) plays a central role in neoangiogenesis and thus represents an important therapeutic target in antiangiogenic cancer therapy strategies. We have developed an anti-VEGF vaccine VEGF kinoid that elicits high titres of neutralizing anti-VEGF antibodies in mice. In this study we have evaluated the immunogenicity, safety and efficacy of VEGF kinoid in preventing tumor growth and metastases in mice. Methods: The efficacy of VEGF kinoid was evaluated in a model of active immunization (CT26 colon carcicoma lung metastases model) and in two models of passive immunization. On the passive immunization setting two VEGF secreting xenograft tumor models, A673 rhabdomyosarcoma and HT29 colon carcinoma, were used to evaluate the ability of IgGs derived from kinoid immunized mice to inhibit tumor growth in comparison with bevacizumab. Results: Active immunization with autologous VEGF Kinoid (mVEGF kinoid) triggered a strong anti-VEGF antibody but no cellular immune response in mice. The anti-VEGF Abs exhibited high neutralizing capacity as assessed via inhibition of human umbilical vein endothelial cell proliferation in the presence of VEGF and binding to the Flk-1 receptor. In mVEGF kinoid immunized BALB/c mice challenged with syngeneic CT26 colorectal tumor cells the lung metastases were inhibited. In human VEGF (hVEGF) kinoid immunized Balb/c mice neutralizing anti-hVEGF Abs were elicited. Purified IgG from these mice inhibited tumor growth of human A673 rhabdomyosarcoma and HT29 colon carcinoma cells xenografted in Swiss nude and NOD/SCID mice respectively as assessed by T/C% measurements. Tumor cell growth inhibition was similar to that in mice receiving therapeutic doses of bevacizumab. Active immunization against VEGF showed an excellent safety profile in mice and no impairment of would healing processes was observed after surgical interventions in immunized mice. Conclusions: These experiments show that active immunization with VEGF kinoid elicits polyclonal neutralizing anti-VEGF Abs that inhibit metastases and tumor growth in mice. It may thus represent an alternative strategy to safely combat VEGF-dependent neovascularization and metastases occurring in malignant tumors. No significant financial relationships to disclose.

The paper reports on research at two well-preserved Iron Age settlement sites in north-east Scotland, occupied between the 2nd century BC and 2nd century AD. At Old Kinord, trenches first excavated in 1903 were reopened, shedding new light on the chronology and structural history of a pair of stone roundhouses and two souterrains. The project extended to new surveys of this site and its neighbour at New Kinord. It investigated the character of the unusually large stone structures found there and the ways in which they were built and used. This report also considers the character of the original excavation, which was conducted by the future Lord Abercromby, a significant figure in the history of Scottish archaeology. Kinolin çekirdeği, geniş bir spektrumda biyolojik aktivite sergileyen birçok doğal ve farmakolojik açıdan aktif bileşiğin ana iskeletini oluşturur. Sübstitüe edilmiş siyano kinolin bileşikleri, büyüme faktörü reseptörü protein tirozin kinaz aktivitesini inhibe edebilmektedir. Farklı gruplara sahip ajanlar ayrıca biyolojik sistemler arasında bağlanan küçük molekül inhibitörleri olarak da işlev görür. Son zamanlarda, sübstitüe edilmiş veya sübstitüe edilmemiş 1,2,3,4-tetrahidrokinolinin bromlama reaksiyonu temelli 6-bromo-, 6,8-dibromo- ve 3,6,8-tribromokinolinlerin sentezi için yeni bir sentetik stratejiler keşfettik. Metal-brom değişimi reaksiyonlarıyla bromlu kinolinlerden 6,8-disübstitüe edilmiş kinolin türevlerinin uygun sentez yollarını ve bromlu ürünlerin disübstitüe kinolinler için öncü bileşikler olduğu rapor edildi. Bu çalışmaların akabinde, 6,8-dibromokinolin, kenetleme ve yer değiştirme reaksiyonları ile fenil, siyano, metoksi ve hidroksi türevlerine dönüştürüldü. Bu strateji ile sentezlenen kinolin türevleri, spesifik antikanser aktiviteleri gösterdi. AbstractClimbing beans play a central role in food security of rural households in the densely populated highlands of East and Central Africa. Soil fertility degradation and the lack of nutrient inputs are major limitations to yield of beans and other crops. We conducted field trials in Northern Rwanda in Kinoni and Muko villages to evaluate the effect of mineral N, P, and K fertilizers (both alone and in combination) and farmyard manure on nitrogen fixation and grain yields of climbing bean in smallholder farmers’ fields. The trials were laid down in a randomized complete block design with seven replicate blocks in each village. Manure and fertilizer application led to greater yields in all fields, and the largest yields were achieved when manure was combined with NPK. Large variability in yield between fields was observed. Application of fertilizer together with manure increased the grain yield from 1.5 to 3.9 t ha−1 in Kinoni and from 2.6 to 5.4 t ha−1 in Muko. Fertilizer and/or manure increased stover yield from 0.8 to 2.3 t ha−1 in Kinoni and from 1.5 to 3.4 t ha−1 in Muko. Application of 30 kg P ha−1 and 5 t manure ha−1 led to increased N and P uptake (from 49 to 106 kg N ha−1 and from 6.1 to 12.4 kg P ha−1 in Kinoni and from 46 to 128 kg N ha−1 and from 5.3 to 17.9 kg P ha−1 in Muko). There was no clear relationship between soil fertility characteristics and the response of climbing bean to applied inputs at Muko site. However, at Kinoni site, limited response to manure and NPK application was observed in plots where soil available P and soil exchangeable K were relatively low. Our results show the benefits of using manure along with mineral fertilizers for increased climbing bean yields and nutrient uptake in smallholder farming systems.

ABSTRACTMycobacterium aviumsubsp.paratuberculosisis the causative agent of Johne's disease (JD) in cattle.M. aviumsubsp.paratuberculosisinfects the gastrointestinal tract of calves, localizing and persisting primarily in the distal ileum. A high percentage of cattle exposed toM. aviumsubsp.paratuberculosisdo not develop JD, but the mechanisms by which they resist infection are not understood. Here, we merge an establishedin vivobovine intestinal segment model forM. aviumsubsp.paratuberculosisinfection with bovine-specific peptide kinome arrays as a first step to understanding how infection influences host kinomic responses at the site of infection. Application of peptide arrays toin vivotissue samples represents a critical and ambitious step in using this technology to understand host-pathogen interactions. Kinome analysis was performed on intestinal samples from 4 ileal segments subdivided into 10 separate compartments (6M. aviumsubsp.paratuberculosis-infected compartments and 4 intra-animal controls) using bovine-specific peptide arrays. Kinome data sets clustered into two groups, suggesting unique binary responses toM. aviumsubsp.paratuberculosis. Similarly, twoM. aviumsubsp.paratuberculosis-specific immune responses, characterized by different antibody, T cell proliferation, and gamma interferon (IFN-γ) responses, were also observed. Interestingly, the kinomic groupings segregated with the immune response groupings. Pathway and gene ontology analyses revealed that differences in innate immune and interleukin signaling and particular differences in the Wnt/β-catenin pathway distinguished the kinomic groupings. Collectively, kinome analysis of tissue samples offers insight into the complex cellular responses induced byM. aviumsubsp.paratuberculosisin the ileum and provides a novel method to understand mechanisms that alter the balance between cell-mediated and antibody responses toM. aviumsubsp.paratuberculosisinfection.

In the era of precision medicine, research of biomarkers for identification of responders to nivolumab therapy is a major challenge. Peripheral blood mononuclear cells (PBMC) could be an interesting surrogate tissue for identifying pharmacodynamic biomarkers. The aim of this exploratory study was to investigate the global serine/threonine kinase (STK) activity in PBMC from non-small-cell lung cancer (NSCLC) patients using a high throughput kinomic profiling method. PamChip® microarrays were used to explore the STK kinomic profile in PBMC from 28 NSCLC patients before nivolumab initiation (D0) and on day 14 (D14) of the first administration. Two clusters of patients (A and B) were identified at D0, median overall survival (OS) tended to be longer in cluster A than in B (402 vs. 112.5 days, respectively; p = 0.15). Interestingly, the PD-L1 tumor cell score (p = 0.045), the count of CD8+ cells (p = 0.023) and the total body weight (p = 0.038) were statistically different between the clusters. On D14, clusters C and D were identified. Greater activity of most STK, especially those of the PI3K/Akt signaling pathway, was noticed among cluster C. No significant difference between C and D was observed regarding OS. Considering the small number of patients, results from this preliminary study are not conclusive. However, the 4-fold longer median OS in cluster A paves the way to further investigate, in a larger cohort of NSCLC patients, the benefit of basal STK kinomic profile in PBMC to identify responders to nivolumab therapy.

У розділа особливості реалізаціх програмного додатку для мобільної платформи показані теоретичні відомості про алгоритми, які були використанні. У розділі аналіз додатку розгялнуті методи тестування додатку, аналіз подальшого вдоскналення.
Structure and scope of work. The explanatory note of the diploma project consists of six sections, contains 8 figures, 5 tables, 5 appendices, 45 sources. Thesis project is dedicated to the development of an application based on Android "Kinogid". In the section analysis of existing solutions for systematization of information about film screenings will consider the existing services, their functions and shortcomings that need to be improved. The section analysis of programming languages and technologies of Android application development considers theoretical aspects of execution methods causing all possible complications and shortcomings of different systems in Android application development. Theoretical information on the topic of the diploma. The methods of application development, algorithms of actions at the set task, and also the decision of a problem which could arise are shown. The section features of the implementation of the software application for the mobile platform shows theoretical information about the algorithms that were used. In the section analysis of the application the methods of testing the application, analysis of further improvement are explained.

Cette these decrit l'evolution des ecoles de la "kinotis" (communaute) grecque d'alexandrie, en essayant de reperer comment s'inscrivent les influences du contexte egyptien et de la metropole sur cette organisation decentralisee, financierement et administrativement independante de l'egypte et de la grece. Il est montre que la politique educative est le coeur de l'activite de la "kinotis" et vise premierement, a dispenser une education de haut niveau, adaptee aux besoins du lieu d'installation et a la fois "nationale", et deuxiemement, a faire que l'ensemble de la population grecque de la ville puisse en jouir. Les effets de ladite politique expliquent les motifs qui incitent les bourgeois grecs alexandrins a se charger du cout de l'education de leurs compatriotes co-citoyens : les ecoles incarnent une strategie collective de la bourgeoisie qui les gere, dont le but est de consolider (ou proteger) sa place dans le milieu pluri-ethnique alexandrin. Car, ne pouvant compter sur l'aide effective de l'etat hellenique pauvre et impuissant, elle necessite l'appui d'une population grecque solidaire, ayant un sentiment national fort. De meme, pour faire face a la concurrence internationale, elle a besoin des employes competents, possedant le savoir-faire necessaire
This thesis sets out to describe the evolution of the schools of the greek "kinotis"* in alexandria. It does so by attempting to show the impact of the egyptian context and of the metropolis on this financially and administratively decentralised organisation, independent both from egypt and greece. It is shown that the educational policy primarily aims at providing a high level education, which is adapted to the needs of the context whilst simultaneously cultivating a national allegiance ; secondly, it aims at rendering this education accessible to the totality of the alexandrian greek population. The effects of this policy come to explain the motives of the bourgeois greeks who undertake the financing of the education of their compatriotes. Specifically this bourgeois strategy compensates for the lack of effective support from the poor greek state. Thus the attempted construction of an ideologically homogenised and professionally competent greek population is supposed to respond to the conditions of international antagonism in that historical context. *community

Les récentes avancées technologiques dans le domaine de l'acquisition et du calcul ont permis une croissance rapide d'une branche de production de média: la capture volumétrique. En particulier, la montée en puissance de la réalité virtuelle et augmentée engendre un besoin accru de contenus adaptés à ces nouveaux médias, notamment des contenus 3D obtenus à partir de scènes réelles. En effet, la possibilité d'enregistrer une performance et de la rejouer sous n'importe quel point de vue permet de créer une expérience dans un environnement réaliste et immersif pour l'utilisateur.Ce manuscrit présente le problème de la reconstruction de forme 4D à partir d'images RVB multi-vues, qui est une des stratégies permettant de créer un tel contenu. Nous nous intéressons particulièrement à la capture de performances dynamiques en situations réelles, contenant des détails de surface complexes. Les défis typiques de ces situations de capture incluent une plus faible densité d'observation des objets d'intérêt en raison des champs de vision plus larges nécessaires pour capturer le mouvement; des occultations et auto-occultations de plusieurs sujets en interaction; un manque de texture typique de l'apparence et des vêtements du sujet réel; ou du flou de bougé avec des sujets en mouvement rapide tels que des scènes d'action sportive. Un aspect essentiel et qui peut encore être amélioré à cet égard est la fidélité et la qualité des formes récupérées, notre objectif dans ce travail.Nous pr'esentons un pipeline complet de reconstruction adapt'e `a ce sc'enario, auquel nous avons contribu'e de nombreuses mani`eres. En premier lieu, on peut noter que les m'ethodes bas'ees sur la technologie st'er'eo multi-vues (MVS) ont atteint un bon niveau de qualit'e avec des pipelines qui comprennent g'en'eralement l'extraction de descripteurs caract'eristiques, une 'etape de mise en correspondance et l'inf'erence de forme 3D. Mais il est surtout int'eressant de noter que des travaux tr`es r'ecents ont r'eexamin'e le probl`eme de st'er'eo et st'er'eo multi-vues en introduisant des fonctions de similarit'e automatiquement inf'er'ees `a l'aide d'apprentissage profond. La principale promesse de ce type de m'ethode 'etant d'inclure un meilleur a-priori, appris sur les donn'ees r'eelles. Dans une premi`ere contribution, nous examinons dans quelle mesure ces am'eliorations sont transf'er'ees au cas plus g'en'eral et complexe de la capture de performances dynamiques, o`u diverses difficult'es suppl'ementaires se pr'esentent. Nous expliquons ensuite comment utiliser cette strat'egie d'apprentissage pour construire de mani`ere robuste une repr'esentation de forme `a chaque instant, `a partir desquelles une s'equence de mod`eles 3D peut ^etre extraite. Une fois que nous obtenons cette repr'esentation `a chaque instant de la s'equence captur'ee, nous expliquons comment il est possible d'exploiter la redondance temporelle pour affiner la pr'ecision des mod`eles en propageant les d'etails des formes observ'ees aux instants pr'ec'edents et suivants. En plus d'^etre b'en'efique pour de nombreux sc'enarios dynamiques `a vues multiples, cela permet 'egalement de capturer des sc`enes plus grandes o`u une pr'ecision accrue peut compenser la r'esolution spatiale r'eduite. Recent advances in acquisition and processing technologies lead to the fast growth of a major branch in media production: volumetric video. In particular, the rise of virtual and augmented reality fuels an increased need for content suitable to these new media including 3D contents obtained from real scenes, as the ability to record a live performance and replay it from any given point of view allows the user to experience a realistic and immersive environment.This manuscript aims at presenting the problem of 4D shape reconstruction from multi-view RGB images, which is one way to create such content. We especially target real life performance capture, containing complex surface details. Typical challenges for these capture situations include smaller visual projection areas of objects of interest due to wider necessary fields of view for capturing motion; occlusion and self-occlusion of several subjects interacting together; lack of texture content typical of real-life subject appearance and clothing; or motion blur with fast moving subjects such as sport action scenes. An essential and still improvable aspect in this matter is the fidelity and quality of the recovered shapes, our goal in this work.We present a full reconstruction pipeline suited for this scenario, to which we contributed in many aspects. First, Multi-view stereo (MVS) based methods have attained a good level of quality with pipelines that typically comprise feature extraction, matching stages and 3D shape inference. Interestingly, very recent works have re-examined stereo and MVS by introducing features and similarity functions automatically inferred using deep learning, the main promise of this type of method being to include better data-driven priors. We examine in a first contribution whether these improvements transfer to the more general and complex case of live performance capture, where a diverse set of additional difficulties arise. We then explain how to use this learning strategy to robustly build a shape representation, from which can be extracted a 3D model. Once we obtain this representation at every frame of the captured sequence, we discuss how to exploit temporal redundancy for precision refinement by propagating shape details through adjacent frames. In addition to being beneficial to many dynamic multi-view scenarios this also enables larger scenes where such increased precision can compensate for the reduced spatial resolution per image frame. The source code implementing the different reconstruction methods is released to the public as open source software

The concern of the pipeline industry and general population for a safe and green environment is higher than ever. This highlights the need for efficient leak detection to prevent environmental catastrophes and operational disruption. Therefore, accurate techniques to detect and locate very small leaks that could develop into larger leaks are a valuable asset for the construction of key pipelines. External pipeline leak detection systems based on distributed fiber optic sensing emerge as the most appropriate solution for automatic detection and localization of very small leaks. In the case of the Kinosis pipeline system, two 11km Electrically Heat Traced Pipe-In-Pipe (EHTPIP) pipelines have been built between the Nexen Long lake upgrader and Nexen Kinosis SAGD facilities. The fiber optic sensing cable is directly in contact with the EHTPIP external surface. These pipelines carry Produced Emulsion and Boiler Feed Water at temperatures as high as 120°C and 150°C respectively. The fiber optic distributed sensing system provides temperature feedback information to the operator, not only in operation and in case of a leak but also when the Electrical Heat Trace system is turned on; in this case, the monitoring system can detect and locate overheating problems and/or signs of heating failures. In the case of a leak, the outer temperature of the pipeline will increase; this will automatically be detected and monitored by the DITEST temperature monitoring system and will trigger an alarm to the Nexen LONG LAKE upgrader SCADA system for that specific location. Furthermore, the combination of fiber optic distributed monitoring with the PIP technology enables to detect and locate a leak in the inner pipeline at a very early stage, therefore avoiding any environmental damage (the leak is still contained by the outer PIP tube) and giving time to the pipeline operator to plan a sectional replacement.
