Antimicrobial resistance poses a significant global health challenge, with the World Health Organization ranking it among the top 10 global threats to public health worldwide. Hence, the urgent need for new anti-infectives targeting novel pathways.In this context, the Methylerythritol phosphate (MEP) pathway becomes particularly significant. The MEP pathway is composed of seven enzymes and is responsible for the biosynthesis of the two isoprenoids precursors: isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP). This pathway is completely absent in humans but is essential to pathogenic microorganisms such as Mycobacterium tuberculosis (Mt), Plasmodium falciparum (Pf), Pseudomonas aeruginosa (Pa).The aims of this project were to elucidate the crystallographic structures of the last two enzymes of the MEP pathway (IspG and IspH) from three different pathogens (P. aeruginosa, M. tuberculosis and P. falciparum) and by combining X-ray fragment screening and in silico virtual screening of chemical libraries, to identify or design new specific inhibitors of these enzymes.Due to the presence of a 4Fe-4S cluster in their active sites, all these targets are oxygen-sensitive enzymes that must be handled under anaerobic conditions using a glove-box. The first part of this work consisted in establishing a protocol for the production and purification of each of the six proteins in quantity suitable for crystallization studies. Pure proteins were obtained for all the IspH orthologues and for Pa and MtIspG.The reactions catalyzed by IspG and IspH are dependent on an electron donating system. For Plasmodium, this system is composed of two proteins: the ferredoxin (a 2Fe-2S cluster, PfFd) and the ferredoxin-reductase (a FAD dependent protein, PfFNR). Obtaining these two active proteins in sufficient quantities was necessary for using the natural reduction system for measuring the enzymatic activities of PfIspG and PfIspH. That’s why the optimization of the expression and purification of PfFd and PfFNR was also achieved during the first part of this project. The activity tests were performed successfully with PfIspH by the group of Dr. Myriam Seemann, our close collaborators at the University of Strasbourg. They also used the Plasmodium system to screen different series of chemical compounds in vitro.Crystallization screening under anaerobic conditions were performed for all the IspG and IspH proteins obtained. Several crystals were obtained and analyzed at the European Synchrotron Radiation Facility (ESRF). The collected data led to the elucidation of the first crystallographic structures of PaIspH and MtIspH at 1.5 Å and 2.8 Å resolution, respectively. Both enzymes show a three-leaf clover folding typical of the IspH family.PaIspH contains an incomplete 3Fe-4S cluster and a molecule of bicine coming from the crystallization buffer in its active site. The bicine is interacting with key residues involved in the substrate binding, but has no inhibitory activity. However, this has provided us structural information on possible interactions within the active site, which may help in the design of new inhibitors.For MtIspH, a shift in the third domain leads to a previously unseen opening of the active site. Surprisingly, despite this opening, MtIspH displays a 4Fe-4S cluster. This can be explained by the fact that the displacement of the third domain has brought the cluster closer to the threonine 189, which can bind and stabilize the apical iron.Some of the compounds identified in silico as well as new inhibitors selected in vitro by our collaborators in Strasbourg have been used for co-crystallization assays. The new structural information combined with the in silico docking analysis and the enzymatic activity assays, opens the door for new biophysical and structural studies of these fascinating enzymes that are targeted for the development of new anti-infectives

A leishmaniose visceral, causada pelos parasitas das espécies Leishmania infantum e Leishmania donovani, do Velho Mundo, e pela Leishmania infantum chagasi, do Novo Mundo, é uma doença infecciosa e letal se não for tratada. O número de casos desta doença está aumentando no Estado de São Paulo. Uma vez diagnosticada, o tratamento para a leishmaniose visceral tem efeitos colaterais importantes, além da ocorrência de parasitas resistentes aos medicamentos disponíveis. Sabe-se que algumas peptidases desempenham um papel importante na fisiologia e doença causada por parasitas do género Leishmania. A inibição específica destas enzimas pode constituir uma importante estratégia para a produção de potentes agentes antiparasitários. Em bactérias E. coli, inibidores de serino peptidases (ISPs) denominados ecotinas, têm sido descritos, sendo também identificados em espécies de Leishmania. A caracterização funcional da ISP1 e ISP2 de L. major mostrou o seu papel na formação de flagelo de promastigotas e na inibição da elastase de neutrófilos de hospedeiro vertebrado, respectivamente. Portanto, os objetivos deste projeto são a expressão e a purificação das proteínas ISP1 e ISP2 de L. i. chagasi (LcISP1 e LcISP2) recombinantes. A amplificação das sequências que codificam as LcISP1 e LcISP2 foi realizada por PCR a partir do DNA genômico, de L. i. chagasi extraído de uma estirpe de referência cedida pelo Laboratório Nacional de Referência de Espécies de Leishmania da Fiocruz, Rio de Janeiro. Os fragmentos de PCR foram clonados no vetor de expressão bacteriano pET28a e as proteínas recombinantes LcISP2 e LcISP1 foram obtidas na fração solúvel do extrato proteico bacteriano. Após expressão das proteínas recombinantes, elas foram purificadas por cromatografia de afinidade em níquel.
Visceral leishmaniasis, caused by the Old World parasites species Leishmania infantum and Leishmania donovani, and by the New World Leishmania infantum chagasi, is an infectious and lethal disease, if untreated. The number of cases of this disease is increasing in the State of São Paulo. Once diagnosed, treatment for visceral leishmaniasis has important side effects, besides occurrence of parasites resistant to the available drugs. It is known that some peptidases play an important role in physiology and disease caused by parasites of the genus Leishmania. The specific inhibition of these enzymes may constitute an important strategy for producing potent antiparasitic agents. In bacteria E. coli, inhibitors of serine peptidases (ISPs) called ecotins, have been described, being also identified in Leishmania species. Characterization of genes encoding ISP1 and ISP2 of L. major, showed its role in the promastigote flagellum formation and in the inhibition of vertebrate host neutrophil elastase, respectively. Therefore, the aims of this project are to produce L. i. chagasi ISP1 (LcISP1) and ISP2 (LcISP2) recombinant forms and to evaluate the its biochemical activity. Amplification of the LcISP1 and LcISP2 encoding sequences were performed by PCR from L. i. chagasi DNA extracted from a reference strain obtained from the Leishmania collection of the Leishmania National Reference Laboratory of the Instituto Oswaldo Cruz. PCR fragments were cloned into the pET28a bacterial expression vector and the recombinant LcISP1 and LcISP2 proteins were present in soluble bacterial extract fraction. After purification by niquel affinity chromatography, recombinant LcISP1 and LcISP2 proteins e were tested for biochemical activity.

O ISEG é relativamente pouco conhecido, no geral, devido às seguintes principais vertentes: a comunicação no mercado chinês e os programas de cooperação com as IES na China. Na fase inicial de exploração do mercado chinês, é fundamental compreendê-lo, assim como conhecer o perfil dos alunos chineses, o que inclui a sua família. O presente projeto tem como principal objetivo explorar o perfil dos alunos chineses e a perceção dos mesmos do contexto português, através de dados secundários e do questionário destinado a alunos que já frequentam cursos de licenciatura, mestrado e doutoramento em Portugal. De modo a perceber os principais pontos fortes e fracos, realiza-se uma análise de concorrência externa e interna. A partir desta análise, identificam-se as partes que se devem melhorar, tais como o aumento de oferta de cursos lecionados em inglês, e a comunicação e a cooperação com as instituições de ensino superior na China. Assim, o plano de marketing desenvolvido apresenta diversas ações e estratégias, que visam melhorar a imagem do ISEG no mercado chinês, aumentar a sua notoriedade, incrementar o número das inscrições de alunos chineses, etc.
For chineses students, the general knowledge for ISEG's is relatively low, mainly due to the two aspects: communication in the Chinese market and cooperation programs with HEIs in China. In the initial phase of exploring Chinese market, it is crucial for ISEG to understand the Chinese market and the profile of Chinese students, even their parents. The main objetivo for the present project is to explore the profile of Chinese students and their perception in terms of Portugal, through secondary data and the questionnaire for chinese students who are already attending undergraduate, masters and doctorate courses in Portugal. In order to understand the main strengths and weaknesses of ISEG compared with its competitors external and internal, a competition analysis is carried out. Based on that, we find out the main parts to be improved, such as: increase the number of courses taught in English, the communication and cooperation with higher education institutions in China. As a result, the Marketing Plan developed presents several actions and strategies, aimed at improving the image of ISEG in the Chinese market, increase its notoriety, and the enrollment of Chinese students, etc.

Med anledning av ISPS- kodens inträdande 2004 och personliga erfarenheter från fartyg och skyddsvaktsarbete har vi valt att fördjupa oss i ämnet portvaktens arbetsuppgifter. Vad vi har observerat sedan tidigare är att den allmänna uppfattningen bland ombordanställda är att portvakten är i vägen och det tar lång tid att komma till sin arbetsplats på fartyget.
Detta är en deskriptiv uppsats, där vi gjort en kvalitativ studie som bygger på intervju med vaktpersonal. Intervjun är sedan jämförd med de regler som finns för vakten i porten. Vi har avgränsat oss till att undersöka Göteborgs hamn, då denna var först i Europa att införa ISPS. Vi har intervjuat Magnus Rosenqvist, gruppchef på G4S i Göteborgs hamn. Detta har vi sedan jämfört med vad sjöbefälsstudenter på Högskolan i Kalmar har för erfarenheter och uppfattningar.
Vakten ansvarar för tillträdet till hamnområdet. Anledningen varför kontrollerna är så höga är för att upprätthålla hamnskyddet, inte fartygsskyddet. Vakten har samma ISPS- utbildning som resterande hamnpersonal, den utbildning en väktare måste inneha, men i Göteborgs hamn måste man dessutom vara skyddsvakt.
In connection with the ISPS- code's arising in 2004 and personal experiences from ships and security guarding, we have selected to deepen ourselves in the matter the gatekeeper’s duties. We have observed already that the general view among the vessel’s apployes, that the gatekeeper is in the way and it takes a long time to get to the workplace onboard the ship.
This is a descriptive essay, where we have done a qualitative study that builds on interview with local security personnel. The interview is then compared with the rules that are for the security guard in the gate. We have delimited ourselves to examining Gothenburg's port, because this is the first port in Europe to introduce the ISPS- code. We have interviewed Magnus Rosenqvist, squad commander at G4S in Gothenburg’s port. This result is then compared with the experiences and thoughts of students at Kalmar Maritime Academy.
The security guards are responsible for the access to the port area. The reason why the controls are so high is in order to maintain the port’s protection, not the ship’s protection. The security guard has the same ISPS education that all port personnel have, the education a general security guard must hold, but in Gothenburg’s port a security guard also must have a “skyddsvakt’s” education.
