Artificial neural networks have been recently used in different fields of science in applications ranging from pattern recognition to semi-quantitative analysis. In this work, two types of neural networks were applied to the problems of spectral interferences, matrix effects, and the measurement drift in ICP-AES. Their performance was compared to that of the more conventional technique of multiple linear regressions (MLR). The two types of neural networks examined were “traditional” multilayer perceptron neural networks and generalized regression neural networks (GRNNs). The GRNN is comparable to, or better than, MLR for modeling spectral interferences and matrix effects covering several orders of magnitude. In the case of an Fe spectral interference on Zn, the GRNN reduced the error from 81% to 24%, while MLR reduced the average error to only 49%. For matrix effects caused by large backgrounds of Mg (0–10,000 ppm) on Zn, average error was reduced to 55% from 67%. In the case of combinations of spectral overlaps and matrix effects, the GRNN reduced average error by approximately 10%. MLR performed poorly on systems involving matrix effects. GRNN is also a very promising tool for the correction of drift caused by fluctuations in power levels, reducing drift over a two-hour period from 2.3% to 0.6%. GRNNs, both by themselves and in multinetwork combinations, seem to be highly promising for the correction of nonlinear matrix effects and long-term signal drift in ICP-AES.

This paper deals with the problem of robust and perceptual logo watermarking for colour images. In particular, we investigate trade-off factors in designing efficient watermarking techniques to maximize the quality of watermarked images and the robustness of watermark. With the fixed size of a logo watermark, there is a conflict between these two objectives, thus a multiobjective optimization problem is introduced. We propose to use a hybrid between general regression neural networks (GRNNs) and multiobjective memetic algorithms (MOMA) to solve this challenging problem. Specifically, a GRNN is used for efficient watermark embedding and extraction in the wavelet domain. Optimal watermark embedding factors and the smooth parameter of the GRNN are searched by a MOMA for optimally embedding watermark bits into wavelet coefficients. The experimental results show that the proposed approach achieves robustness and imperceptibility in watermarking.

Three different artificial neural network (ANN) methods, namely, feed-forward back-propagation (FFBP), radial basis function (RBF), and generalized regression neural networks (GRNNs) were applied to predict peak ground acceleration (PGA). Ninety five three-component records from 15 ground motions that occurred in Northwestern Turkey between 1999 and 2001 were used during the applications. The earthquake moment magnitude, hypocentral distance, focal depth, and site conditions were used as inputs to estimate PGA for vertical (U-D), east-west (E-W), and north-south (N-S) directions. The direction of the maximum PGA of the three components was also added to the input layer to obtain the maximum PGA. Testing stage results of three ANN methods indicated that the FFBPs were superior to the GRNN and the RBF for all directions. The PGA values obtained from the FFBP were modified by linear regression analysis. The results showed that these modifications increased the prediction performances.

Accurately estimating crop evapotranspiration (ET) is essential for agricultural water management in arid and semiarid croplands. This study developed extreme learning machine (ELM) and generalized regression neural network (GRNN) models for maize ET estimation on the China Loess Plateau. Maize ET, meteorological variables, leaf area index (LAI), and plant height (hc) were continuously measured during maize growing seasons of 2011–2013. The meteorological data and crop data including LAI and hc from 2011 to 2012 were used to train the ELM and GRNN using two different input combinations. The performances of ELM and GRNN were compared with the modified dual crop coefficient (Kc) approach in 2013. Results indicated that ELM1 and GRNN1 using meteorological and crop data as inputs estimated maize ET accurately, with root mean square error (RMSE) of 0.221 mm/d, mean absolute error (MAE) of 0.203 mm/d, and NS of 0.981 for ELM1, RMSE of 0.225 mm/d, MAE of 0.211 mm/d, and NS of 0.981 for GRNN1, respectively, which confirmed better performances than the modified dual Kc model. Performances of ELM2 and GRNN2 using only meteorological data as input were poorer than those of ELM1, GRNN1, and modified dual Kc approach, but its estimation of maize ET was acceptable when only meteorological data were available.

The current study looks at the impact of compliance to Shari’ah principles on the capital structure for Malaysian firms. Examination of impact of compliance is based on the classification by the Securities Commission of Malaysia. Given that the literature on adjustment tends to ignore non-linear models, the current study utilises Generalised Regression Neural Network (GRNNs). Results are compared to conventional panel data regression models via performing a hold-out sample. Initial results confirm stability of the data allowing predictive ability. The results indicate that compliant firms tend to finance a greater portion of their financing imbalance via equities relative to non-compliant firms. This provides a strong indication towards compliant firms reducing overall risk taking where the financing pattern incorporates a greater aspect of risk sharing which is in-line with Shari’ah principles. In addition, two more factors are ranked as important in deciding compliant firms issue choice to resolve financial imbalance: profitability and size. The rest of the determinants have low impact on explaining net debt issues. Diagnostics for results provide evidence of lower RMSE and MSE for GRNNs for the training, testing and overall datasets. The potential benefit of this research allows managers and investors of Islamic capital markets to understand potential risk exposure and financing costs of compliant firms. Findings also provide a roadmap for development of a sustainable capital market model which has wider implications on a global scale.

Unlike feedforward neural networks (FFNN) which can act as universal function approximators, recursive, or recurrent, neural networks can act as universal approximators for multi-valued functions. In this paper, a real time recursive backpropagation (RTRBP) algorithm in a vector matrix form is developed for a two-layer globally recursive neural network that has multiple delays in its feedback path. This algorithm has been evaluated on two GRNNs that approximate both an analytic and nonanalytic periodic multi-valued function that a feedforward neural network is not capable of approximating.

Agriculture is the principal basis of livelihood that acts as a mainstay of any country. There are several changes faced by the farmers due to various factors such as water shortage, undefined price owing to demand–supply, weather uncertainties, and inaccurate crop prediction. The prediction of crop yield, notably paddy yield, is an intricate assignment owing to its dependency on several factors such as crop genotype, environmental factors, management practices, and their interactions. Researchers are used to predicting the paddy yield using statistical approaches, but they failed to attain higher accuracy due to several factors. Therefore, machine learning methods such as support vector regression (SVR), general regression neural networks (GRNNs), radial basis functional neural networks (RBFNNs), and back-propagation neural networks (BPNNs) are demonstrated to predict the paddy yield accurately for the Cauvery Delta Zone (CDZ), which lies in the eastern part of Tamil Nadu, South India. The performance of each developed model is examined using assessment metrics such as coefficient of determination (R2), root mean square error (RMSE), mean absolute error (MAE), mean squared error (MSE), mean absolute percentage error (MAPE), coefficient of variance (CV), and normalized mean squared error (NMSE). The observed results show that the GRNN algorithm delivers superior evaluation metrics such as R2, RMSE, MAE, MSE, MAPE, CV, and NSME values about 0.9863, 0.2295 and 0.1290, 0.0526, 1.3439, 0.0255, and 0.0136, respectively, which ensures accurate crop yield prediction compared with other methods. Finally, the performance of the GRNN model is compared with other available models from several studies in the literature, and it is found to be high while comparing the prediction accuracy using evaluation metrics.

The purpose of this paper is to improve the accuracy of solving prediction tasks of the missing IoT data recovery. To achieve this, the authors have developed a new ensemble of neural network tools. It consists of two successive General Regression Neural Network (GRNN) networks and one neural-like structure of the Successive Geometric Transformation Model (SGTM). The principle of ensemble topology construction on two successively connected general regression neural networks, supplemented with an SGTM neural-like structure, is mathematically substantiated, which improves the accuracy of prediction results. The effectiveness of the method is based on the replacement of the summation of the results of the two GRNNs with a weighted summation, which improves the accuracy of the ensemble operation in general. A detailed algorithmic implementation of the ensemble method as well as a flowchart of its operation is presented. The parameters of the ensemble operation are determined by optimization using the brute-force method. Based on the developed ensemble method, the solution of the task of completing the partially missing values in the real monitoring dataset of the air environment collected by the IoT device is presented. By comparing the performance of the developed ensemble with the existing methods, the highest accuracy of its performance (by the parameters of Mean Absolute Percentage Error (MAPE) and Root Mean Squared Error (RMSE) accuracy) among the most similar in this class has been proved.

Several global leaf area index (LAI) products were generated using neural networks, but the training dataset for the neural networks was sensor specific, and the construction of the training dataset was time consuming. In this paper, an unsupervised domain adaptation-based method was proposed to estimate LAI from the Visible Infrared Imaging Radiometer Suite (VIIRS) surface reflectance dataset based on a training dataset constructed from the Moderate Resolution Imaging Spectroradiometer (MODIS) surface reflectance dataset. A transfer component analysis (TCA) algorithm was first utilized to map the MODIS and VIIRS surface reflectance into the same subspace to reduce the distribution discrepancies between the MODIS and VIIRS surface reflectance. Then, the embedded data obtained from MODIS surface reflectance dataset, along with the LAI values produced by fusing the MODIS and the Carbon cYcle and Change in Land Observational Products from an Ensemble of Satellites (CYCLOPES) products, were employed to train general regression neural networks (GRNNs). Finally, for retrieving the LAI values, the embedded data acquired from the VIIRS surface reflectance dataset was input into the trained GRNNs. For multiple field sites with different biome types, we used this developed method to retrieve LAI values based on the VIIRS surface reflectance dataset. The results indicate that, based on the training dataset built from MODIS surface reflectance dataset, the domain adaptation-based retrieval method can effectively estimate LAI values from VIIRS surface reflectance dataset. By comparison with the VIIRS and MODIS LAI products, the retrieved LAI values with TCA are more consistent with the reference LAI values acquired from high-resolution remote sensing images. The coefficient of determination (R2) and root mean square error (RMSE) of the retrieved LAI values with TCA at all selected sites are 0.88 and 0.68, respectively. Furthermore, the accuracy of the retrieved LAI values with TCA is higher than the retrieved LAI values without TCA with the R2 0.81 and the RMSE 0.79.

The purpose of this survey is to analyze how civics teachers relate to freedom of speech in their classrooms. To examine this I want to study following questions a) What arguments for and against freedom of speech in the classroom do the teachers use? b) What strategies do the teachers use in class discussions to not risk result into conflicts or violating expressions? To analyze this, I have used a qualitative method in terms of interviews with six different teachers from both primary schools and secondary high schools. This was made on different locations in Sweden. The main theoretical points for this survey have been from Ulf Petäjäs freedom of speech theories how the western countries of democracy and its populations today doesn´t consider its value, but instead mostly talk about its boundaries. Also, Carsten Ljunggren and Ulrika Unemar Östs strategies amongst teachers when it comes to class discussions, what roles they choose to assume. For example, a moderator who let the students discuss freely, or an educator to maintain control over the classroom. And finally, Tomas Englunds deliberative discourse and Olga Dysthes dialogical dialogue regarding strategies how teachers engage in class discussions. The conclusions that I find are that the freedom of speech, teachers strategies and the teachers view towards the freedom of speech is never static but conditional to the climate of the discourse, depending on which subject they teach and how well they know their students. Which leads to my most definitive conclusion, that the freedom of speech among these teachers in this suvery is situation contingent.

Vetandets gräns är en epistemologisk fråga om vilken kunskap människan kan få och ha om allt runt omkring oss. Alla dessa ting som finns omkring oss, men även se på självet1 som objekt. Immanuel Kant skriver om kunskapens gräns i sin bok Kritik av det rena förnuftet och den 1400-tals Juris doktorn Nicolaus Cusanus2 skriver även han om vår vetandets gräns i sin bok ”Gudsseendet”.Det uppsatsen vill söka efter hos Kant och Cusanus är om människans tänkande har någon gräns. Det studiet söker är en större kunskap om vårt egna tänkande, vilket visar på en större kunskap om vår inre värld. ”Den primära världen” som påverkar allt handlande som människan gör i ”den sekundära världen”. Relevansen med detta studium är att människan kan få en mer detaljerad och fördjupad syn på människans tänkandes gränser.Under hela mitt vuxna liv har jag burit på frågan ”varför gör jag det jag gör?”. Under mitt första år som teologistuderande fick jag höra att Immanuel Kant anser att människan inte kan tänka utan tid och rum. Min naturliga reaktion blev att fråga, ”varför inte det?” och fick till svar att ”Det går men det är svårt”. Sedan den dagen har denna tanke varit ämne för reflektion.Detta studium vill se om det kan finna den bortre gräns som människans tänkande har, sin egna förståelse om sig själv och i slutändan kanske få en större kunskap om vad det är att vara en människa i denna tid och rum.

För att ett gemenskapsvarumärke ska kunna registreras krävs att märket uppfyller kravet på särskiljningsförmåga och därmed tillåts inte märken som endast är beskrivande att registreras. Deskriptiva varumärken anses inte kunna särskilja varor och kan därför inte uppfylla varumärkets grundläggande funktion, förmågan att ange ett kommersiellt ursprung. Vidare ska inte beskrivande termer kunna förbehållas enskilda näringsidkare eftersom det finns ett frihållningsintresse beträffande sådana beteckningar. Det är emellertid inte alltid helt enkelt att avgöra om ett figurmärke innehållande deskriptiv text bör anses uteslutande beskrivande eller ej. Logiskt sett ligger det nära till hands att utgå från att sådana märken bör tillåtas registrering i större mån än motsvarande ordmärken. Uppsatsens primära syfte är därför att utreda i vilken utsträckning det är möjligt att registrera gemenskapsfigurmärken delvis bestående av deskriptiva komponenter samt om det är möjligt att dra några slutsatser om särskiljningsförmågans nedre gräns i detta avseende. Utifrån en granskning av drygt 300 OHIM-fall kan jag konstatera att det under vissa omständigheter är möjligt att registrera beskrivande figurmärken, trots att korresponderande ordmärken säkerligen skulle ha nekats registrering. Deskriptiva figurmärken tycks dock endast undantagsvis beviljas registrering och i praktiken framstår det som att OHIM bedömer om ett figurmärke till störst del kommer att uppfattas som beskrivande, snarare än om det endast kommer att uppfattas på så vis. I regel verkar det krävas ”något extra” för att ett deskriptivt figurmärke ska tillåtas registrering, som ett visst mått av fantasifullhet, kreativitet eller åtminstone godtycklighet. Beträffande särskiljningsförmågans nedre gräns förefaller det sig inte möjligt att ange mer specifikt vad som krävs för att en deskriptiv text ska kunna varumärkesregistreras som en del av ett figurmärke, vilket bl.a. beror på att registreringsärenden oundvikligen präglas av ett visst mått av subjektivitet. Med det sagt går det ändå att skönja vissa egenskaper och faktorer som tillmäts stor vikt i registreringssammanhang.

Empathy is the ability to understand another person's subjective condition, including emotions. Depending on culture, emotions are expressed differently, hence there might occur difficulties for one person to understand emotions in another if they belong to different cultures. This study explores how depression is described in western psychiatry, compared to alternative cultural forms of similar conditions. Building on how the understanding of the client's emotions is shaped in the psychiatric meeting, the limit of empathy is discussed as well as what might influence the understanding of another person's emotions. Two limits of empathy are revealed, both depending on differences in the individuals relation to the surrounding world. The first limit is found in differences between emotions, the second limit is related to differences in experience due to when they are based in either emotional or cognitive conceptions. The conclusion of this study is that the scientific thinking and the cognitively based understanding of emotions changes the comprehension of them – from being interpreted as a way to relate to the world to being interpreted more as characteristics within the individual. Consequently, the possibility to empathetically understand the other's emotions diminishes.

La tesi Cases grans. Interiors nobles a Barcelona 1739-1761 té com eix fonamental estudiar el que podem convenir en denominar "l'art de la casa", durant el període de Ferran VI, a la ciutat de Barcelona. Intenta dibuixar la configuració interna de les arquitectures particulars, percebudes aquestes sempre com espais ocupats en constant evolució, com a reflex de personalitats, gustos i modes d'una època determinada, com escenaris polièdrics d'accions, deures, desitjos i funcions, en definitiva com "obra composada". Per tant, Cases grans. Interiors nobles a Barcelona no ha pretès ser mai un estudi acabat i tancat sinó la confecció d'una sòlida trama on sustentar futures línies de recerca.

Metodològicament el corpus teòric es fonamenta amb l'anàlisi d'uns vuitanta, concretament 85 inventaris post-mortem de personatges nobles per tal d'accedir a la descripció dels espais domèstics que havien ocupat. Molta rellevància han tingut en la confecció del discurs i les conclusions les notícies que ofereixen els fons familiars i patrimonials, on cal destacar les despeses domèstiques, o la relectura de documentació sobre afers sanitaris, o gremials, entre altres. Informacions, que en darrera instància van ser posades en relació a un gruix considerable d'assumptes consignats davant notari, on podem destacar la revisió de les voluntats testamentàries, els contractes matrimonials, àpoques o acords comercials

La tesi s'estructura en deu capítol amb un seguit d'annexos documentals i gràfics. A banda de l'introducció que correspon al primer capítol, en el segon, s'analitzen i valoren les diferents fons documentals emprades. En el tercer, titulat Barcelona entre dos tractats de pau, hi ha una aproximació als aspectes que defineixen l'elit barcelonina i la pròpia ciutat. Perfilar la configuració dels seus interiors domèstics amb correcció sols era factible construint la imatge de la família en les múltiples vessants que la conformen, - ocupacions, llinatge, patrimoni- perquè no oblidem que la casa i el seu agençament esdevé un univers simbòlic únic i particular del món clos de la família. En el següent, Figurar, construir i habitar, la mirada es centra en dibuixar les possibles imatges de la ciutat. Após a reestruturação da indústria de eletricidade, a energia elétrica tornou-se uma mercadoria que pode ser comprada e vendida nos mercados de eletricidade. Dentro deste contexto, novas ferramentas para previsão de cargas e preços de eletricidade ainda são objetos de estudo. Redes Neurais Artificiais são bastante usadas para previsão de preços juntamente com outras técnicas estatísticas. Nesta pesquisa apresenta-se um método para a previsão de preço do dia seguinte, usando uma Rede Neural de Regressão Generalizada (GRNN) e Perceptron Multicamadas (MLP) com algoritmo de treinamento Levenberg – Marquardt. Os preços nodais são determinados resolvendo um DC-OPF (fluxo de potência ótimo), gerando os preços para todas as barras do sistema elétrico, denominado Locational Marginal Prices (LMP). Para treinar a rede neural, foram gerados vários cenários carga-preço e variando aleatoriamente a carga em cada barra do sistema elétrico. Os resultados são analisados através do erro percentual médio absoluto, uma medida muito comum encontrada na literatura. Os resultados foram comparados com literatura especializada com o caso base bem como com o Erro Máximo que varia de 3% a 8 %. Os resultados obtidos foram satisfatórios, dentro das margens encontradas na literatura, conforme apresentado na seção de resultados. A presente proposta foi aplicada no sistema teste IEEE 24- RTS.

This work presents a method for day-ahead price forecasting using a generalized regression neural network where nodal prices are determined by solving a DC optimal power flow. In order to train the neural network several load-price scenarios are generated by randomly varying the loads in each bus of the electric energy system. The results are validated by analyzing the mean absolute percentage error, a common measure adopted in the technical literature as well as the base case and the maximum error which are within those found in the literature. The proposed methodology has been successfully applied to the IEEE 24-RTS system.

Resumo: Após a reestruturação da indústria de eletricidade, a energia elétrica tornou-se uma mercadoria que pode ser comprada e vendida nos mercados de eletricidade. Dentro deste contexto, novas ferramentas para previsão de cargas e preços de eletricidade ainda são objetos de estudo. Redes Neurais Artificiais são bastante usadas para previsão de preços juntamente com outras técnicas estatísticas. There were many ways of bordering manliness during the historical period covered by my research. Borders have been metaphorically understood as those invisible, often non-enunciated limits that have safeguarded manliness. There were borders separating masculinity from femininity and from childishnes, but there is also a more distant border, separating masculinity from the bestial. The term un-manliness is a useful concept for this analysis, for it can be used to illuminate the different ways in which masculinity has been interrogated. The concept can also be used in comparative analyses of how tolerance towards men deviating from ideas of ideal masculinity has differed according to situation and culture. It has been my ambition to elucidate the particular attitudes, values, customs, knowledge and requirements that influenced the view of masculinity at both individual and the group level. The clearest-cut aspects of manliness and un-manliness expressed in court proceedings were those having to do with sexuality, relations within the household, and the subordinate and dominant masculinities displayed by different court-room actors. The latter, in turn, reflected contemporary social structures, including the social gap that divided the Livonian peasantry’s serfs or former serfs from the ruling Baltic-German elite. Records from the court proceedings have shown the subordinate masculinity of the defendants, subordinate not only to that of the officers of the court but to that of the witnesses. This subordination was an inevitable consequence of the nature of the accusations, regardless of whether they were deemed well-founded or false.  The defendants were placed in a situation where they were forced constantly to be on the alert, ready to defend themselves and show their best sides. As a result, they would often give extremely clear expression to their views of proper masculinity. Such actors stressed, consciously or unconsciously, certain manly traits and behaviour patterns that characterised themselves and others. Their arguments provide insights into what they thought of each other and how they conceived a man should generally be, behave and act in different situations. By the same token, they clearly showed what kinds of behaviours were considered undesirable or outright unmanly. The positioning of the borders of manliness was linked both to time and to space. Deviations have helped different societies set the borders for what they considered acceptable behaviour. There was a clear cultural and geographical border between Sweden as such, and the Swedish province Livonia. This emerges clearly when one compares Livonian results with earlier studies on bestiality in Sweden. This shows that the view of manliness and the tolerance towards certain kinds of behaviour changed as one moved East.

The relationship between Objective Metrics (OM) and Subjective Assessments (SA) has been analyzed by people using different methods. This paper continues Gaspar Gil Gómez’s research over test method development by use of SOM-GRNN, aiming to find correlations between OM and SA. In this paper, CAE simulation is performed to analyze the relationship between OM and vehicle parameters. First impression test is refined and one more dataset has been added in order to populate SOM-GRNN map. This paper also conducts analysis over SOM and GRNN algorithms, and explores several possible applications using SOM-GRNN map. Finally the whole SOM-GRNN system is integrated and a User Interface is built in GUI for future research and application. The system can still be improved by populating database, refining SA evaluation method and optimizing SOM-GRNN training algorithms.
Relationen mellan målmetri (OM) och subjektiv bedömning (SA) har analyserats av personer som använder olika metoder. I detta dokument fortsätter Gaspar Gil Gómezs forskning kring testmetodutveckling med hjälp av SOM-GRNN, som syftar till att finna korrelationer mellan OM och SA. I detta papper utförs CAE-simulering för att analysera förhållandet mellan OM och fordonsparametrar. Första intryckstestet är raffinerat och ytterligare en dataset har lagts till för att fylla i SOM-GRNN-kartan. I detta dokument analyseras även SOM- och GRNN-algoritmer, och undersöker flera möjliga tillämpningar med hjälp av SOM-GRNN kartan. Slutligen är hela SOM-GRNN-systemet integrerat och ett användargränssnitt är byggt i GUI för framtida forskning och tillämpning. Systemet kan fortfarande förbättras genom att fylla i databasen, förädla SA utvärderingsmetod och optimera SOM-GRNN träningsalgoritmer.

A inferência de Redes de Regulação Gênica (GRNs) é importante em Biologia Sistêmica, pois permite o entendimento de padrões de interações entre genes. Essas descobertas sãoúteis para fornecer compreensão sobre doenças e ajudar no desenvolvimento de fármacos. Técnicas de computação evolutiva, como a Programação Genética Cartesiana (CGP), têm sido utilizadas para inferir GRNs com resultados promissores. Entretanto, a CGP tem problemas de escalabilidade. Aqui, GRNs são inferidas de forma eficiente usando abordagens de computação de alto desempenho. Experimentos computacionais mostram que o método desenvolvido nesta iniciação científica é capaz de inferir GRNs mais rapidamente do que outros da literatura com soluções simbólicas. O ganho em tempo de processamento da técnica paralela apresentada em relação ao formato sequencial é de até 104%.

The problem of graph similarity measurement is fundamental in both complex networks and bioinformatics researches. Gene regulatory networks (GRNs) describe the interactions between the molecules in organisms, and are widely studied in the fields of medical AI. By measuring the similarity between GRNs, significant information can be obtained to assist the applications like gene functions prediction, drug development and medical diagnosis. Most of the existing similarity measurements have been focusing on the graph isomorphisms and are usually NP-hard problems. Thus, they are not suitable for applications in biology and clinical research due to the complexity and large-scale features of real-world GRNs. In this paper, a fast similarity measurement method called FSM for GRNs is proposed. Unlike the conventional measurements, it pays more attention to the differences between those influential genes. For the convenience and reliability, a new index defined as influence power is adopted to describe the influential genes which have greater position in a GRN. FSM was applied in nine datasets of various scales and is compared with state-of-art methods. The results demonstrated that it ran significantly faster than other methods without sacrificing measurement performance.

The use of CRISPR/Cas9 has been successfully applied in various plant species to induce targeted genome editing, including soybean. Soybean is recalcitrant to transformation and thus, plants with stable CRISPR gene edits are costly and take a long time to produce. Moreover, soybean is allotetraploid and editing paralogous genes are often necessary to obtain observable phenotype(s). For each gene target, we designed two gRNAs to increase editing efficiency and allow rapid genotyping by PCR. We also tested the CRISPR reagents in transient hairy root transformation to determine if the Cas9 and gRNAs would perform properly in transgenic soybean plants. Our results showed that we can indeed obtain highly efficient, cost-effective CRISPR/Cas editing in soybean to generate novel genotypes for gene discovery and downstream field propagation and breeding efforts. Examples of CRISPR-edited genes and their associated seed traits will be discussed.
