With the development of mass spectrometer technology, recent studies revealed the critical roles of cancer-specific metabolism for tumor propagation in several types of cancers. In leukemia, many studies have been conducted to elucidate a leukemia-specific metabolism, and several effective treatments such as IDH1/2 inhibitors targeting acute myeloid leukemia (AML) with IDH1/2 mutation have been developed. To identify the new metabolic pathways on which acute leukemia cells depend, we purified water-soluble metabolites from CD34+ hematopoietic stem and progenitor cells (HSPCs) of healthy donors, AML and acute lymphoblastic leukemia (ALL) patients, and we comprehensively measured 116 metabolites using mass spectrometer analysis. From this experiment, we found that the cellular content of glycerol 3-phosphate (G3P) in CD34+ AML and ALL cells was lower than that of normal CD34+ HSPCs. G3P is an intermediate metabolite in the glycolysis metabolic pathway and is utilized as a substrate for phospholipids synthesis. The initial and rate-limiting step of phospholipids synthesis is the synthesis of lysophosphatidic acid (LPA) from G3P and acyl-CoA mediated by glycerol 3-phosphate acyltransferases (GPATs). Since CD34+ acute leukemia cells contained significantly lower level of G3P, we hypothesized that leukemia cells actively consumed G3P and synthesized LPA by GPATs. GPATs are classified into four isoforms based on intracellular localization and substrate preference. GPAT1 and GPAT2 are mitochondrial GPATs that are localized to the mitochondrial outer membrane, but on the other hand, GPAT3 and GPAT4 are microsomal GPATs that are localized to the endoplasmic reticulum membrane, each encoded by independent genes. GPAT1 is identified as an essential gene for the growth of leukemia cells by RNAi screen analysis in the public database (DepMap). We found that CD34+ immature AML cells exhibited higher GPAT1 expression as compared to CD34- more differentiated AML cells and normal T cells. GPAT1 knockdown inhibited the proliferation of several acute leukemia cell lines including THP-1 and Kasumi-1 in vitro and in vivo. Moreover, a mitochondrial GPATs specific inhibitor (FSG67), which was originally developed as a drug to treat obesity and diabetes, suppressed the growth of the leukemia cell lines through the induction of G1 cell cycle arrest. Growth inhibition was rescued by exogenous administration of LPA, suggesting that the synthetic activity mediated by mitochondrial GPATs should be required for acute leukemia growth. Furthermore, FSG67 induced the apoptosis of leukemia cells derived from AML and ALL patients without affecting normal CD34+ HSPCs at least in vitro. We also confirmed that the injection of FSG67 resulted in the suppression of AML and ALL propagation in vivo using patient-derived xenograft models (see figure). GPAT1 regulates the mitochondrial function by producing LPA which is an essential metabolite for maintaining mitochondrial fusion. Actually, the amount of LPA was decreased in GPAT1 knockdown acute leukemia cells. We next examined mitochondrial energy production by extracellular flux assay, and found that GPAT1 knockdown as well as FSG67 significantly suppressed oxygen consumption rate of acute leukemia cells. Consistent with the impaired mitochondrial function, FSG67 suppressed the mitochondrial membrane potential, indicating that GPAT1 should play a pivotal role in maintaining leukemia-specific mitochondrial function. These results collectively suggest that the synthesis of LPA from G3P catalyzed by GPAT1 has a critical role in propagation of acute leukemia cells irrespective of their lineage origin. Thus, GPAT1 is a novel and common therapeutic target for human acute leukemia through suppressing leukemia-specific mitochondrial function. Glycerol-3-phosphate acyltransferases (GPATs) catalyze the first step in the synthesis of glycerolipids and glycerophospholipids. Microsomal GPAT, the major GPAT activity, is encoded by at least two closely related genes, GPAT3 and GPAT4. To investigate the in vivo functions of GPAT3, we generated Gpat3-deficient ( Gpat3 −/−) mice. Total GPAT activity in white adipose tissue of Gpat3 −/− mice was reduced by 80%, suggesting that GPAT3 is the predominant GPAT in this tissue. In liver, GPAT3 deletion had no impact on total GPAT activity but resulted in a 30% reduction in N-ethylmaleimide-sensitive GPAT activity. The Gpat3 −/− mice were viable and fertile and exhibited no obvious metabolic abnormalities on standard laboratory chow. However, when fed a high-fat diet, female Gpat3 −/− mice showed decreased body weight gain and adiposity and increased energy expenditure. Increased energy expenditure was also observed in male Gpat3 −/− mice, although it was not accompanied by a significant change in body weight. GPAT3 deficiency lowered fed, but not fasted, glucose levels and tended to improve glucose tolerance in diet-induced obese male and female mice. On a high-fat diet, Gpat3 −/− mice had enlarged livers and displayed a dysregulation in cholesterol metabolism. These data establish GPAT3 as the primary GPAT in white adipose tissue and reveal an important role of the enzyme in regulating energy, glucose, and lipid homeostasis.

Acyl-CoA:glycerol-sn-3-phosphate acyltransferase (GPAT) is an enzyme responsible for the rate-limiting step in the synthesis of glycerophospholipids and triacylglycerol (TAG). The enzymes of mammalian species are classified into four isoforms; GPAT1 and GPAT2 are localized in the mitochondrial outer membrane, whereas GPAT3 and GPAT4 are localized in the endoplasmic reticulum membrane. The activity of each enzyme expressed is associated with physiological and pathological functions. The transcriptional regulation is well known, particularly in GPAT1. GPAT1 mRNA expression is mainly regulated by the binding of the transcriptional factor SREBP-1c to the specific element (the sterol regulatory element) flanking the GPAT1 promoter. The TAG level is controlled by the insulin-induced transcriptional expression of GPAT1, which occupies most of the GPAT activity in the liver. The transcriptional regulation of the other three GPAT isoforms remains undetermined in detail. It is predicted that retinoic acid serves as a transcription factor in the GPAT2 promoter. PPARγ (peroxisome proliferator-activated receptor γ) increases the mRNA expression of GPAT3, which is associated with TAG synthesis in adipose tissues. Although GPAT has been considered to be a key enzyme in the production of TAG, unexpected functions have recently been reported, particularly in GPAT2. It is likely that GPAT2 is associated with tumorigenesis and normal spermatogenesis. In this review, the physiological and pathophysiological roles of the four GPAT isoforms are described, alongside the transcriptional regulation of these enzymes.

Abstract Berardinelli-Seip congenital lipodystrophy type 2 (BSCL2) is the most severe form of human lipodystrophy and is caused by loss-of-function mutations in the BSCL2/seipin gene. Exactly how seipin may regulate adipogenesis remains unclear. A recent study in vitro suggested that seipin may function to inhibit the activity of glycerol-3-phosphate acyltransferases (GPATs), and increased GPAT activity may be responsible for the defective adipogenesis under seipin deficiency. Here we generated Seipin−/−Gpat3−/− mice, which had mild but significant recovery of white adipose tissue mass over Seipin−/− mice. The mass of brown adipose tissue (BAT) of the Seipin−/−Gpat3−/− mice was almost completely restored to normal level. Importantly, the Seipin−/−Gpat3−/− mice showed significant improvement in liver steatosis and insulin sensitivity over Seipin−/− mice, which is attributable to the increased BAT mass and to the enhanced browning of the subcutaneous fat of the Seipin−/−Gpat3−/− mice. Together, our results establish a functional link between seipin and GPAT3 in vivo and suggest that GPAT inhibitors may have beneficial effects on BSCL2 patients.

ABSTRACT Glycerol-3-phosphate acyltransferase (GPAT) catalyzes the initial step in the synthesis of all glycerolipids. It is the committed and rate-limiting step and is redundant in Saccharomyces cerevisiae, mammals, and plants. GPAT controls the formation of lipid intermediates that serve not only as precursors of more-complex lipids but also as intracellular signaling molecules. Saccharomyces cerevisiae possesses two GPATs, encoded by the GAT1 and GAT2 genes. Metabolic analysis of yeast lacking either GAT1 or GAT2 indicated partitioning of the two main branches of phospholipid synthesis at the initial and rate-limiting GPAT step. We are particularly interested in identifying molecular determinants mediating lipid metabolic pathway partitioning; therefore, as a starting point, we have performed a detailed study of Gat1p and Gat2p cellular localization. We have compared Gat1p and Gat2p localization by fluorescence microscopy and subcellular fractionation using equilibrium density gradients. Our results indicate Gat1p and Gat2p overlap mostly in their localization and are in fact microsomal GPATs, localized to both perinuclear and cortical endoplasmic reticula in actively proliferating cells. A more detailed analysis suggests a differential enrichment of Gat1p and Gat2p in distinct ER fractions. Furthermore, overexpression of these enzymes in the absence of endogenous GPATs induces proliferation of distinct ER arrays, differentially affecting cortical ER morphology and polarized cell growth. In addition, our studies also uncovered a dynamic posttranslational regulation of Gat1p and Gat2p and a compensation mechanism through phosphorylation that responds to a cellular GPAT imbalance.

Glycerol-3-phosphate acyltransferases (GPATs) play an important role in glycerolipid biosynthesis, and are mainly involved in oil production, flower development, and stress response. However, their roles in regulating plant height remain unreported. Here, we report that Arabidopsis GPAT1 is involved in the regulation of plant height. GUS assay and qRT-PCR analysis in Arabidopsis showed that GPAT1 is highly expressed in flowers, siliques, and seeds. A loss of function mutation in GPAT1 was shown to decrease seed yield but increase plant height through enhanced cell length. Transcriptomic and qRT-PCR data revealed that the expression levels of genes related to gibberellin (GA) biosynthesis and signaling, as well as those of cell wall organization and biogenesis, were significantly upregulated. These led to cell length elongation, and thus, an increase in plant height. Together, our data suggest that knockout of GPAT1 impairs glycerolipid metabolism in Arabidopsis, leading to reduced seed yield, but promotes the biosynthesis of GA, which ultimately enhances plant height. This study provides new evidence on the interplay between lipid and hormone metabolism in the regulation of plant height.

Glycerol-3-phosphate acyltransferase (GPAT) enzymes catalyze the first step in triacylglycerol (TAG) synthesis. Genes that belong to the GPAT family are potential genetic markers for intramuscular fat content (IMF) content and thus meat quality. The objective of this study was to analyze the expression of GPAT1 and GPAT2 genes in musculus longissimus lumborum, liver and subcutaneous fat of various breeds of pigs. Furthermore, correlations between the genes’ expression abundance and utility traits, meat quality and meat texture parameters of pork were determined. The results obtained showed significant differences in the mRNA level of GPAT1 between analyzed tissues and breeds. The highest expression of GPAT1 gene was observed in liver tissue (p ≤ 0.01). Furthermore, significantly higher GPAT1 transcript level in the m. longissimus lumborum was observed for duroc in comparison to other analyzed breeds (p ≤ 0.05). Expression of the GPAT2 gene was shown only in the liver tissues, however statistically significant differences between the analyzed breeds were not observed. Correlation analysis confirmed the highest association between GPAT2 gene expression level in liver and cohesiveness and resilience traits of m. longissimus lumborum (p ≤ 0.01).

Glycerol-3-phosphate acyltransferases (GPATs) catalyze the first and rate-limiting step in the de novo glycerolipid synthesis. The GPAT2 isoform differs from the other isoforms because its expression is restricted to male germ cells and cancer cells. It has been recently reported that GPAT2 expression in mouse testis fluctuates during sexual maturation and that it is regulated by epigenetic mechanisms in combination with vitamin A derivatives. Despite progress made in this field, information about GPAT2 role in the developing male germ cells remains unclear. The aim of the present study was to confirm the hypothesis that GPAT2 is required for the normal physiology of testes and male germ cell maturation. The gene was silenced in vivo by inoculating lentiviral particles carrying the sequence of a short-hairpin RNA targeting Gpat2 mRNA into mouse testis. Histological and gene expression analysis showed impaired spermatogenesis and arrest at the pachytene stage. Defects in reproductive fitness were also observed, and the analysis of apoptosis-related gene expression demonstrated the activation of apoptosis in Gpat2-silenced germ cells. These findings indicate that GPAT2 protein is necessary for the normal development of male gonocytes, and that its absence triggers apoptotic mechanisms, thereby decreasing the number of dividing germ cells.

Four glycerol-3-phosphate acyltransferase (GPAT) isoforms, each encoded by a separate gene, catalyze the initial step in glycerolipid synthesis; in liver, the major isoforms are GPAT1 and GPAT4. To determine whether each of these hepatic isoforms performs a unique function in the metabolism of fatty acid, we measured the incorporation of de novo synthesized fatty acid or exogenous fatty acid into complex lipids in primary mouse hepatocytes from control, Gpat1−/−, and Gpat4−/− mice. Although hepatocytes from each genotype incorporated a similar amount of exogenous fatty acid into triacylglycerol (TAG), only control and Gpat4−/− hepatocytes were able to incorporate de novo synthesized fatty acid into TAG. When compared with controls, Gpat1−/− hepatocytes oxidized twice as much exogenous fatty acid. To confirm these findings and to assess hepatic β-oxidation metabolites, we measured acylcarnitines in liver from mice after a 24-h fast and after a 24-h fast followed by 48 h of refeeding with a high sucrose diet to promote lipogenesis. Confirming the in vitro findings, the hepatic content of long-chain acylcarnitine in fasted Gpat1−/− mice was 3-fold higher than in controls. When compared with control and Gpat4−/− mice, after the fasting-refeeding protocol, Gpat1−/− hepatic TAG was depleted, and long-chain acylcarnitine content was 3.5-fold higher. Taken together, these data demonstrate that GPAT1, but not GPAT4, is required to incorporate de novo synthesized fatty acids into TAG and to divert them away from oxidation.

La colonisation réussie et durable des terres par les plantes a nécessité l'apparition de certains traits afin de survivre aux nouvelles contraintes telles que l'accès limité aux nutriments et à l'eau, la gravité, l'environnement oxydatif et les radiations UV. Chez les plantes terrestres existantes, les polymères hydrophobes déposés à la surface de différents tissus, tels que la sporopollénine, la cuticule, la subérine et la lignine, sont connus pour contribuer à la limitation de la perte d'eau et à la résistance à d'autres stress abiotiques, ce qui en fait des "innovations de terrestrialisation" potentielles. Comment et quand ces traits ont évolué chez les plantes restent insaisissables. Les données fossiles et l'analyse phylogénétique des gènes impliqués dans la biosynthèse de ces polymères chez les plantes ont proposé la cutine, la partie hydrophobe et polymérisée de la cuticule, comme le meilleur candidat pour la terrestrialisation des plantes. Cette thèse décrit des recherches menées pour déterminer la distribution de la cutine dans les plantes, l'évolution des gènes nécessaires à sa biosynthèse et la fonction de deux d'entre eux chez la Bryophyte modèle, Marchantia polymorpha. La présence de la cutine dans les espèces de Zygnematophyceae, qui sont les algues les plus proches des plantes terrestres, et dans diverses plantes terrestres a été surveillée par des observations microscopiques et des analyses biochimiques. La cutine n'a été détectée que dans les plantes terrestres, de même qu'une enzyme terminale de la biosynthèse des monomères de cutine, les glycérol-phosphates acyle transférases (GPATs). The successful and long-lasting colonization of lands by plants required the evolution of innovations in order to survive to the novel constraints such as limited nutrient and water access, gravity, oxidative environment and UV radiations. In extant land plants, hydrophobic polymers deposited on the surface of different tissues, such as sporopollenin, cuticle, suberin and lignin, are known to contribute to the limitation of water loss, and resistance to other abiotic stresses, making them potential "terrestrialization innovations". How and when these traits evolved in plants remains elusive. Fossils data and phylogenetical analysis of genes involved in cutin biosynthesis in plants proposed cutin as the best candidate for plant terrestrialization. However, works testing this hypothesis are actually scares and the evidence limited. This thesis describes conducted investigations to determine the distribution of cutin in plants, the evolution of the genes required for its biosynthesis and the function of two of them in the model bryophyte, Marchantia polymorpha. The presence of cutin in species from the zygnematophyceae, which are the closest algal relatives to land plants, and in diverse land plants was monitored through microscopic observations and biochemical analyses. Cutin was only detected in land plants, as was one terminal enzyme in the biosynthesis of the cutin monomer, the Glycerol-Phosphate Acyl Transferases (GPATs). To confirm the origin of cutin in Embryophytes we have studied GPAT in the Bryophyte M. polymorpha. First, the expression pattern of M. polymorpha GPAT were studied using promoter:GUS fusion. The M. polymorpha GPATs promoter activities were mainly detected where cutin is formed. Then a mutant line of M. polymorpha GPAT was isolated. Macroscopic analysis of this mutant have shown that such as in Embryophytes, Marchantia cutin-related genes are involved in development of aerial organs. Biochemical analysis, reverse-genetics and phylogenetics were combined to reach the conclusion that cutin evolved in the first land plants together with one class of cutin-biosynthesis enzymes, the GPATs, and likely play a function in development in Bryophytes. This works indicates that cutin evolution in Embryophytes played a role in the terrestrialization event

Accumulation of triglyceride (TG) in enterocytes of the small intestine follows two specific absorption pathways: the monoacylglycerol acyltransferase pathway (MGAT) and glycerol acyltransferase pathway (GPAT), the latter (GPAT) is usually found in the small intestine. In this study, we investigated the effects of Lactobacillus rhamnosus MRS6AN, an isolate from “amabere amaruranu” a Kenyan traditional cultured milk, on triglyceride accumulation and expression of GPAT3, I-FABP, MTP and NPC1L1 in Caco-2 cell enterocyte model. Intracellular triglyceride level (TG) of Caco2 cells was significantly reduced by live L. rhamnosus (LB) compared with other bacterial products. MTP expression in Caco2 cells was minimally reduced in live L. rhamnosus (LB) treated Caco2 cells. However, the expression level GPAT3, I-FABP and NPC1L1 was reduced in Caco2 cells treated with live bacteria. Data from this study suggests that Lactobacillus rhamnosus MRS6AN may reduce lipid uptake and accumulation perhaps via modulation of GPAT3 pathway.

La symbiose mycorhizienne à arbuscules (AM) est une association entre la plupart des embryophytes et des champignons ubiquitaires du sol qui appartiennent à la famille des Glomeromycotina. La symbiose mycorhizienne a été proposé comme l'une des innovations clés qui ont permis la colonisation de la terre par des plantes il y a 450 millions d'années. Les champignons mycorhiziens colonisent leur hôte de façon interne et produisent des structures intracellulaires, appelées arbuscules, où ils fournissent de l'eau et des nutriments minéraux à leur hôte en échange de carbone sous forme de sucres et de lipides. Chez les angiospermes, des études récentes ont permis de mieux comprendre les mécanismes moléculaires à l'origine de l'établissement de la symbiose mycorhizienne et des échanges trophiques. Les lipides ont été identifiés comme étant une des principales sources de carbone transférée au partenaire fongique qui est auxotrophe pour les lipides. Une voie de biosynthèse des lipides associée à la symbiose limité dans son expression dans les cellules colonisées par les champignons AM a été caractérisé chez des plantes modèles. Cependant, les études génétiques ont jusqu'à présent été limitées à quelques espèces d'angiospermes. Nous avons utilisé ici le modèle d'hépatique Marchantia paleacea, récemment mis au point, pour vérifier si les mécanismes permettant le transfert des lipides de la plante au champignon ont une origine profonde dans les plantes ou ont évolué plus récemment chez les Angiospermes. Premièrement, nous avons observé une accumulation de lipides dans les cellules de M. paleacea contenant des arbuscules. Ensuite, une stratégie originale basée sur l'ingénierie enzymatique nous a permis de suivre des composés lipidiques spécifiques supposés être transférés au champignon. De cette manière, nous avons démontré la production de lipides et leur transfert de M. paleacea au champignon mycorhizien Rhizophagus irregularis. La conservation de ce transfert, qui avait été initialement démontrée dans les angiospermes, souligne son importance dans la symbiose mycorhizienne. The Arbuscular Mycorrhiza (AM) symbiosis is an association between most embryophytes and ubiquitous soil-born fungi that belong to the Glomeromycotina. It has been proposed as one of the key innovation that allowed the colonization of land by plants 450 million years ago. AM fungi colonize internally their host and produce intracellular structures, called arbuscules, where they provide water and mineral nutrients to their host in exchange for carbon delivered as sugars and lipids. In angiosperms, recent studies have provided a deeper understanding of the molecular mechanisms behind AM establishment and trophic exchanges. Lipids have been identified as the main carbon source transferred to the fungal partner which is lipid auxotroph. The associated symbiotic lipid-biosynthesis pathway expressed in cells colonized by AM fungi have been deciphered in model plants. However, genetic studies have so far been limited to a few angiosperm species. Here, we used the model liverwort Marchantia paleacea, recently developed, to test whether the mechanisms that allow the transfer of lipids from the plant to the fungus have a deep origin in plants or evolved more recently in Angiosperms. First, we observed lipid accumulation in the arbuscule-containing cells of M. paleacea. Then, an original strategy based on enzymatic engineering allowed us to follow the fate of specific lipid compounds believed to be transferred to the fungus. By this way, we demonstrated the production of lipids and their transfer from M. paleacea to the AM fungus Rhizophagus irregularis. The conservation of this transfer, that had been originally demonstrated in angiosperms, outlines its importance in the AM symbiosis. Thanks to the recently sequenced genome of M. paleacea, we identified in this organism the genes known in angiosperms as being involved in the symbiotic lipid biosynthesis and transfer pathway. I focused on one essential gene encoding for the Glycerol-3 Phosphate Acyl Transferase RAM2 which is thought to act downstream of this pathway. High fat feeding increases hepatic fat accumulation and is associated with hepatic insulin resistance. AMP Activated Protein Kinase (AMPK) is thought to inhibit lipid synthesis by the acute inhibition of glycerol-3-phosphate acyltransferase (GPAT) activity and transcriptional regulation via SREBP-1c. The purpose of this study was to determine if chronic activation of AMPK prevented an increase in GPAT1 activity in rats fed a high fat diet. Rats were fed a control (C), or a high fat (HF) diet (60% fat) for 6 weeks and injected with saline or a daily AICAR dose of 0.5 mg/g body weight. Chronic AMPK activation by AICAR injections resulted in a significant reduction in hepatic triglyceride accumulation in both the C and HF fed animals (C, 5.5±0.7; C+AICAR, 2.7 ±0.3; HF, 21.8±3.3; and HF+AICAR, 8.0±1.8 mg/g liver). HF feeding caused an increase in total GPAT and GPAT1 activity which was not affected by chronic AMPK activation (GPAT1 activity vs. C, C+AICAR, 92±19%; HF, 186±43%; HF+AICAR, 234±62%). Markers of oxidative capacity, including citrate synthase activity and cytochrome c abundance, were not affected by chronic AICAR treatment. Interestingly, HF feeding caused a significant increase in LCAD (up 66% from C), a marker of fatty acid oxidation capacity. These results suggest that chronic AMPK activation limits hepatic triglyceride accumulation independent of a reduction in total GPAT1 activity.

Neste trabalho foram investigadas as propriedades elétricas de materiais híbridos de Sílica/orgânico à base de GPTS (3-glicidoxipropiltrimetoxisilano) e TEOS (tertraetilortosilicato), preparadas pelo processo sol-gel, a partir da hidrólise ácida. Os Materiais híbridos de Sílica/orgânico foram preparados por deposição do sol de GPTS/TEOS por dip-coating, em substratos de vidro. Eletrodos de ouro foram depositados paralelamente às faces da amostra para a realização de medidas de espectroscopia de impedância. O estudo foi realizado em amostras com diferentes composições de GPTS/TEOS e diferentes temperaturas de tratamentos térmicos de densificação, destas amostras em diferentes composições, foram realizadas medidas em diferentes temperaturas. Foi determinado que a condutividade aumenta com a concentração de GPTS na matriz híbrida, os valores foram de 8,04.10-9 S/cm e 8,74.10-10 S/cm, para os filmes GT(7:1) e GT(1:1), respectivamente. O mecanismo de transporte envolve íons H+ que transferem carga protônica ao oxigênio do epóxi proveniente do GPTS, esse mecanismo de transporte é descrito pelo mecanismo de Grotthuss. A energia de ativação diminuiu com a aumento da razão de GPTS, sendo 0,54 eV e 0,83 eV, para os filmes GT(7:1) e GT(3:1), respectivamente. Organic/silica hybrid materials also known as Organically Modified Silicates (ORMOSIL) have been object of study due to the diversity of organically functionalized silicon alcoxides with organic groups covalently bonded to the silicon (epoxy, vinyl, amino, phenyl, etc). This work aims the study of the electrical properties of the organic/silica hybrid materials prepared by sol-gel technique derived from the acid hydrolysis of 3- glycidoxypropyltrimethoxysilane (GPTS) and tetraethylorthosilicate (TEOS). Organic/silica hybrid films were deposited on glass substrates by dip-coating technique and sandwiched between gold electrodes to allow the impedance spectroscopy measurements. The study was carried out on film samples with different GPTS/TEOS molar ratio and films submitted to different thermal treatments for densification. Also impedance spectroscopy measurements were performed at different temperatures for samples with different GPTS/TEOS molar ratio. It was determined regarding the increase of the GPTS concentration in the hybrid matrix that the conductivity increases from 8.74x10- 10 S/cm to 8.04x10-9 S/cm for films GT(1:1) and GT(7:1), respectively. The transport mechanism involves H+ ions transfer protonic charge to the oxygen of epoxy groups, and this transport mechanism is described by Grotthuss model. The activation energy decreases with the increase of the increase of the GPTS concentration, being 0.83 eV and 0.54 eV and for the GT (3: 1) and GT (7: 1), respectively. Concerned educators have been implementing ninth-grade transition programs to help freshmen adjust to the demands in high school and to reduce ninth-grade failure rates. The purpose of this quasi-experimental quantitative study was to investigate the impact of a ninth-grade transition program. The research questions addressed impact on cumulative GPAs and credits, ninth-grade dropout rates, and student satisfaction with ninth grade as measured by a survey of 120 ninth-grade transition program participants and 102 nonparticipants. Theoretical foundations were provided by the construction of new knowledge based on different background knowledge, different social conditions, and different life experiences. Another theoretical foundation was social control theory predicting less student delinquency if students have strong bonds with society such as school, family, and peers. After using a posttest-only with nonequivalent control-group design and independent-measures t tests for all hypotheses, results showed that the transition program did not have any effect on dropout rates, student satisfaction, and cumulative credits earned. The program had a negative effect on GPAs, possibly because of an increased margin of error (low return rate of parental consent for participation) and a new math curriculum for the experimental group. Recommendations for future study include conducting a longitudinal study with more participants, adding qualitative aspects, and teaching the same curricula to all study participants. Findings are significant for positive social change because they challenge educators to reexamine transition procedures, implement improved strategies, and review their teaching methods to provide students with the skills required by employers for a successful workforce that will contribute to America's well being and strong economic status in the 21st century.

The purpose of the current study was to investigate the relationships between natural mentoring and youth academic achievement, attitudes towards school, and psychological well-being in a cross cultural sample of adolescents. It also sought to investigate whether it is the quality of the mentoring relationship and not simply the categorical presence of a natural mentor that is associated with positive youth outcomes. This study also examined the reliability and validity of the RHI-Y-M. Participants (n = 62) were recruited from two schools in rural Southern Illinois. Youth were asked to complete a survey packet of self-report questionnaires that included measures of attitudes towards school, life satisfaction, stress, and depression. The primary hypotheses were not supported. However, the RHI-Y-M demonstrated good reliability and concurrent validity. Limitations with regards to data collection and statistical power are discussed. The majority of youth identified natural mentors, and there was a significant association between the mentoring relationship and life satisfaction. These findings have implications for community and intervention programs involving youth.

Анотація:
Argonne National Laboratory is further developing fault diagnosis algorithms for use by the operator of a nuclear plant to aid in improved monitoring of overall plant condition and performance. The objective is better management of plant upsets through more timely, informed decisions on control actions with the ultimate goal of improved plant safety, production, and cost management. Integration of these algorithms with visual aids for operators is taking place through a collaboration under the concept of an operator advisory system. This is a software entity whose purpose is to manage and distill the enormous amount of information an operator must process to understand the plant state, particularly in off-normal situations, and how the state trajectory will unfold in time. The fault diagnosis algorithms were exhaustively tested using computer simulations of twenty different faults introduced into the chemical and volume control system (CVCS) of a pressurized water reactor (PWR). The algorithms are unique in that each new application to a facility requires providing only the piping and instrumentation diagram (PID) and no other plant-specific information; a subject-matter expert is not needed to install and maintain each instance of an application. The testing approach followed accepted procedures for verifying and validating software. It was shown that the code satisfies its functional requirement which is to accept sensor information, identify process variable trends based on this sensor information, and then to return an accurate diagnosis based on chains of rules related to these trends. The validation and verification exercise made use of GPASS, a one-dimensional systems code, for simulating CVCS operation. Plant components were failed and the code generated the resulting plant response. Parametric studies with respect to the severity of the fault, the richness of the plant sensor set, and the accuracy of sensors were performed as part of the validation exercise. The background and overview of the software will be presented to give an overview of the approach. Following, the verification and validation effort using the GPASS code for simulation of plant transients including a sensitivity study on important parameters will be presented. Finally, the planned future path will be highlighted.
