Анотація:

Efficient delivery of functional factors into target cells remains challenging. Although extracellular vesicles (EVs) are considered to be potential therapeutic delivery vehicles, a variety of efficient therapeutic delivery tools are still needed for cancer cells. Herein, we demonstrated a promising method to deliver EVs to refractory cancer cells via a small molecule-induced trafficking system. We generated an inducible interaction system between the FKBP12-rapamycin-binding protein (FRB) domain and FK506 binding protein (FKBP) to deliver specific cargo to EVs. CD9, an abundant protein in EVs, was fused to the FRB domain, and the specific cargo to be delivered was linked to FKBP. Rapamycin recruited validated cargo to EVs through protein-protein interactions (PPIs), such as the FKBP-FRB interaction system. The released EVs were functionally delivered to refractory cancer cells, triple negative breast cancer cells, non-small cell lung cancer cells, and pancreatic cancer cells. Therefore, the functional delivery system driven by reversible PPIs may provide new possibilities for a therapeutic cure against refractory cancers.

Protein delivery to cells in vivo has great potential for the functional analysis of proteins in nonmodel organisms. In this study, using the butterfly wing system, we investigated a method of protein delivery to insect epithelial cells that allows for easy access, treatment, and observation in real time in vivo. Topical and systemic applications (called the sandwich and injection methods, respectively) were tested. In both methods, green/orange fluorescent proteins (GFP/OFP) were naturally incorporated into intracellular vesicles and occasionally into the cytosol from the apical surface without any delivery reagent. However, the antibodies were not delivered by the sandwich method at all, and were delivered only into vesicles by the injection method. A membrane-lytic peptide, L17E, appeared to slightly improve the delivery of GFP/OFP and antibodies. A novel peptide reagent, ProteoCarry, successfully promoted the delivery of both GFP/OFP and antibodies into the cytosol via both the sandwich and injection methods. These protein delivery results will provide opportunities for the functional molecular analysis of proteins in butterfly wing development, and may offer a new way to deliver proteins into target cells in vivo in nonmodel organisms.

Extracellular vesicles (EVs), specifically exosomes and microvesicles (MVs), are presumed to play key roles in cell–cell communication via transfer of biomolecules between cells. The biogenesis of these two types of EVs differs as they originate from either the endosomal (exosomes) or plasma (MVs) membranes. To elucidate the primary means through which EVs mediate intercellular communication, we characterized their ability to encapsulate and deliver different types of macromolecules from transiently transfected cells. Both EV types encapsulated reporter proteins and mRNA but only MVs transferred the reporter function to recipient cells. De novo reporter protein expression in recipient cells resulted only from plasmid DNA (pDNA) after delivery via MVs. Reporter mRNA was delivered to recipient cells by both EV types, but was rapidly degraded without being translated. MVs also mediated delivery of functional pDNA encoding Cre recombinase in vivo to tissues in transgenic Cre-lox reporter mice. Within the parameters of this study, MVs delivered functional pDNA, but not RNA, whereas exosomes from the same source did not deliver functional nucleic acids. These results have significant implications for understanding the role of EVs in cellular communication and for development of EVs as delivery tools. Moreover, studies using EVs from transiently transfected cells may be confounded by a predominance of pDNA transfer.

AbstractThe design of safe and efficient gene transfer vectors remains one of the key challenges in gene therapy. Despite their remarkable transfection efficiency, viral vectors suffer from known safety issues. Consequently, significant research activity has been undertaken to develop nonviral approaches to gene transfer during the last decade. Numerous academic and industrial research groups are investigating synthetic cationic vectors, such as cationic amphiphiles, with the objective of increasing the gene transfection activity. Within this area, the development of functional synthetic vectors that respond to local environmental effects have met with success. These synthetic vectors are based on mechanistic principles and represent a significant departure from earlier systems. Many of these systems for gene delivery in vitro and in vivo are discussed in this article.

Dysfunctional endothelium causes more disease than any other cell type. Systemically administered RNA delivery to nonliver tissues remains challenging, in large part because there is no high-throughput method to identify nanoparticles that deliver functional mRNA to cells in vivo. Here we report a system capable of simultaneously quantifying how >100 lipid nanoparticles (LNPs) deliver mRNA that is translated into functional protein. Using this system (named FIND), we measured how >250 LNPs delivered mRNA to multiple cell types in vivo and identified 7C2 and 7C3, two LNPs that efficiently deliver siRNA, single-guide RNA (sgRNA), and mRNA to endothelial cells. The 7C3 delivered Cas9 mRNA and sgRNA to splenic endothelial cells as efficiently as hepatocytes, distinguishing it from LNPs that deliver Cas9 mRNA and sgRNA to hepatocytes more than other cell types. These data demonstrate that FIND can identify nanoparticles with novel tropisms in vivo.

The ability of cell penetrating peptides (CPPs) to deliver biologically relevant cargos into cells is becoming more important as targets in the intracellular space continue to be explored. We have developed two assays based on CPP-dependent, intracellular delivery of TEM-1 β-lactamase enzyme, a functional biological molecule comparable in size to many protein therapeutics. The first assay focuses on the delivery of full-length β-lactamase to evaluate the internalization potential of a CPP sequence. The second assay uses a split-protein system where one component of β-lactamase is constitutively expressed in the cytoplasm of a stable cell line and the other component is delivered by a CPP. The delivery of a split β-lactamase component evaluates the cytosolic delivery capacity of a CPP. We demonstrate that these assays are rapid, flexible and have potential for use with any cell type and CPP sequence. Both assays are validated using canonical and novel CPPs, with limits of detection from <500 nM to 1 µM. Together, the β-lactamase assays provide compatible tools for functional characterization of CPP activity and the delivery of biological cargos into cells.

We compared the performance and levofloxacin (Quinsair) lung deposition of three nebulisers commonly used in CF (I-Neb Advance, eFlow rapid, and LC Plus) with the approved nebuliser Zirela. The delivered dose, delivery rate, and aerosol particle size distribution (APSD) for each device were determined using the methods described in the Pharmacopeia. High-resolution computed tomography scans obtained from seven adult patients with mild CF were used to generate computer-aided, three-dimensional models of their airway tree to assess lung deposition using functional respiratory imaging (FRI). The eFlow rapid and the LC Plus showed poor delivery efficiencies due to their high residual volumes. The I-Neb, which only delivers aerosols during the inspiratory phase, achieved the highest aerosol delivery efficiency. However, the I-Neb showed the largest particle size and lowest delivery rate (2.9 mg/min), which were respectively associated with a high extrathoracic deposition and extremely long nebulisation times (>20 min). Zirela showed the best performance considering delivery efficiency (159.6 mg out of a nominal dose of 240 mg), delivery rate (43.5 mg/min), and lung deposition (20% of the nominal dose), requiring less than 5 min to deliver a full dose of levofloxacin. The present study supports the use of drug-specific nebulisers and discourages the off-label use of general-purpose devices with the present levofloxacin formulation since subtherapeutic lung doses and long nebulisation times may compromise treatment efficacy and adherence.

Анотація:

Toxins efficiently deliver cargo to cells by binding to cell surface ligands, initiating endocytosis, and escaping the endolysosomal pathway into the cytoplasm. We took advantage of this delivery pathway by conjugating an attenuated diphtheria toxin to siRNA, thereby achieving gene downregulation in patient-derived glioblastoma cells. We delivered siRNA against integrin-β1 (ITGB1)—a gene that promotes invasion and metastasis—and siRNA against eukaryotic translation initiation factor 3 subunit b (eIF-3b)—a survival gene. We demonstrated mRNA downregulation of both genes and the corresponding functional outcomes: knockdown of ITGB1 led to a significant inhibition of invasion, shown with an innovative 3D hydrogel model; and knockdown of eIF-3b resulted in significant cell death. This is the first example of diphtheria toxin being used to deliver siRNAs, and the first time a toxin-based siRNA delivery strategy has been shown to induce relevant genotypic and phenotypic effects in cancer cells.

Amphiphilic block and graft copolymers have been studied because of the possibility of tailoring their complex and fascinating chemical properties. Potential applications include wetting agents, foaming agents, plastic modifiers as well as biomedical applications in drug delivery, owing to their biocompatible and low toxic nature. This thesis describes the study of a series of amphiphilic block copolymers, known as Pluronics, and their aqueous interaction with a hydrophobic drug, flurbiprofen. Synthesis and characterisation of novel graft copolymers with interesting associative behaviour that is less influenced by concentration is also another major aspect of the work described in this thesis. Pulsed-field gradient stimulated-echo nuclear magnetic resonance (pFGSE- NMR) and surface tension measurements have been used to show that the addition of flurbiprofen promotes micellisation of Pluronic triblock copolymers (PI03, P123, and L43). Structural changes in the micelles ofPluronics PI03 and P123, as a function of temperature, eo-solvent (ethanol, 10 wt/vol %), and the addition of the hydrophobic drug flurbiprofen were also investigated by small- angle neutron scattering (SANS). Flurbiprofen was shown to be released from micelles by increasing the solution pH. At higher pH, the drug is ionised and the fraction of polymer in micelles reduces. Synthesis of novel graft copolymers was carried out using a "grafting onto" method. In this approach, the functional group of a hydrophobic backbone will react with the functional group of the copolymer chains. The graft copolymers composed of either hydrophilic Pluronics or sulfonated poly(ethylene oxide) chains and hydrophobic backbones, displayed formation of core-shell micelles in selective solvents. The Pluronic graft copolymers showed the potential for solubilising hydrophobic drug molecules in aqueous solution even at low polymer concentrations. These graft copolymers and their interactions with flurbiprofen were characterised by PFGSE-NMR and SANS.

Анотація:

Funtional εCL monomers have been successfully synthesised utilising a primary amine (protected as a CBZ or BOC carbamate) and also a protected ketone (using an acetal protecting group). Amine derivatives were synthesised from the commercially available trans-4-aminocyclohexanol. The acetal protected ketone was synthesised from the commercially available 1,4-cyclohexanedione monoethylene acetal. The subsequent exposure of these monomers to ring opening polymerisation conditions (using stannous octanoate) in the presence of εCL, yielded random copolymers with a functionalised polyester backbone. The acetal copolymer was successfully deproteceted and reduced to yield an alcohol pendant. A polymer-drug composite was successfully synthesised through the use of this alcohol polymer reacting with ciprofloxacin. A polymer-drug conjugate was also successfully synthesised using surface chemistry on PCL and PLA films. PCL and PLA films were cast as films and subsequently aminolysed using 1,6-hexamethylene diamine in aqueous media at pH = 11.5. These aminolysed films were then treated with both ciprofloxacin and penicillin G, together with amide coupling chemistry to synthesise biologically active surfaces. Ciprofloxacin and penicillin G were successfully attached to the PCL surface with ciprofloxacin being attached to the PLA variant. A functional PCL monomer was also successfully synthesised from the commercially available PCL diol. Both methacrylate and acrylate varients were synthesised through acid chloride chemistry. The acrylate and methacrylate analogue were UV cured using both type I (Irgacure 2959) and II (DETX/EDB) photoinitiators. The relative ratios of these initiators were changed along with the composition of the curing material to investigate curing parameters, to aid in 3D printing conditions. To achieve viscocity criteria for 3D printing a ratio of 70:30 (PCL DMA:PEG DA) was used, utilising PEG DA as a reactive diluent. A ratio of 3 wt% DETX and 3 wt% EDB was found to give the best UV curing results and were successfully applied on a jetting machine to yield 3D printed constructs.

The objective of this research is to explore the use of ionic liquds in crystal engineering and drug delivery. Ionic liquids have a wide range of applications in pharmaceutical field due to their unique physicochemical propertie ssuch as chemical, thermal stability, low melting point, nonvolatility, nonflamability, low toxicity and recyclability which offer unique and interesting potential for pharmaceuitcal applications. Currently, many research groups are working on the development of ionic liquids to use in this field but there is need to develop systematic understanding about new techniques for synthesis and applications of ionic liquids to obtain new crystal form and potential of drug ionic salts. The synthesis of fifteen phosphonium ionic liquids under microwave irradiation and their physicochemical properties was investigated. The reaction time was significantly reduced compared to conventional methods, and higher yields were reported. The crystallisation of pharmaceutical drugs such as sulfathiazole, chlorpropamide, phenobarbital and nifedipine were investigated using imidazolium ionic liquids. The supramolecular complex of sulfathiazole and phenobarbital with imidazolium ionic liquids and polymorphic change in chlorpropamide was achieved. The ionic liquids provides unique environment for the crystallisation. The imidazolium salts of ibuprofen and diclofenac were synthesised and evaluated for physicochemical properties and their pharmaceutical performances especially transdermal absorption. The investigation of physicochemcal properties and pharmaceutical performance of imidazolium drug salts indicated opportunity to optimise lipophilicity and other physicochemical properties such as molecular size, osmolality, viscosity to achieve desired skin deposition and permeation. This study will provide a new approach to design of new drug salts develop using the interdisciplinary knowledge of chemical synthesis and drug delivery.

Biomaterials serve as interventional tools in medicine to treat, improve or replace diseased tissues, organs or function of the body. Although several polymeric biomaterials already exist, they often present challenges, at material level, such as non-biodegradability, degradation into acidic by-products or tissue incompatibility, or at functional level such as failure to sustain prolonged release of therapeutic payload for a desired period. Research has been focused on investigating new polymeric candidates to address these problems of current systems. The use of renewable resources to generate smart polymers for biomedical and pharmaceutical purposes presents a new and exciting avenue for biomaterials. As part of these efforts, a new set of biomaterials were developed from plant-derived high molecular weight (~3.0 kDa) compounds. The advantages of biobased materials include availability for large-scale synthesis, facile post-synthetic modification, biocompatibility, improvement of functional properties and affordability. In this project we used sucrose soyates, i.e. octa-esters obtained from conjugation of sucrose molecules and multiple soybean oil fatty acid chains, to prepare three groups of functional biomaterials namely: a). self-assembled soy-based nano-constructs, b). blended soy-based free-standing films and c). three-dimensional cross-linked soy-based soft matrices. Here, we will discuss the fabrication and physical, chemical and mechanical characterization of these biomaterials prepared from soy-based compounds, as well as, the assessment of their functional performance in biological environment.

Анотація:

Thermoresponsive hydrogels exhibit the unique property of volume change as a function of change in temperature as they transition between hydrophilic and hydrophobic states. These hydrogels can be loaded with drug/protein and serve as reservoirs for drug/protein delivery applications. A hydrogel based device for controlled drug delivery is designed with a number of subsystems that are interfaced with LabVIEW for development of a functional device. The device was designed using analytical and finite element analysis procedures and fabricated. In this manuscript, the device design will be reviewed and discussed. A parametric study was performed to examine the device operation and performance as function of hydrogel heating/cooling temperature profiles. Subsequently, the device was employed in a series of experiments to examine the delivery of a protein as a function of thermal stimuli. The matrix used in this study was poly(ethylene glycol) diacrylate (PEGDA) and the drug delivery nanoparticles carriers were poly(N-isopropylacrylamide-co-acrylamide (PNIPAM) with a lower critical solution temperature (LCST) around 40°C. The protein of choice was bovine serum albumin (BSA). The results of this study illustrate that the development of a multi-drug or therapeutic delivery device is possible and that individual drugs can be delivered on demand using a closed loop control system.

Анотація:

The Philippines embraced a devolution agenda in 1991, leading to the enactment of the Local Government Code (LGC). Despite over three decades of implementation, devolution has remained incomplete, resulting in blurred accountability between the national government and local government units (LGUs) and inefficient delivery of basic devolved services. In March 2023, President Ferdinand Marcos Jr. ordered a review of the functions, services, and facilities that will be fully devolved to local governments. As part of this review, the study assesses LGU functional assignments across three critical sectors: health, infrastructure, and agriculture. The study performs an “unbundling” of the broad functions identified in the LGC and examines them in light of the principles of functional assignment and the binding constraints that have hindered devolution in the country. An expenditure analysis is employed to assess whether the increase in available financing in the first year of implementing the Mandanas ruling has translated into an increase in service delivery of devolved functions, as reflected in LGU spending. The study finds that the emphasis on subsidiarity and local autonomy, coupled with the country’s binding constraints, has resulted in significant gaps between de jure and de facto functional assignments, as well as disparities in assumed devolved functions across LGUs. The data further indicates uneven uptake or utilization of the increased funding across different levels of LGUs. The discussion highlights the need for a reevaluation of functional assignments, as well as the government's strategy to address the binding constraints that continue to hinder devolution: fiscal imbalances, institutional capacity, enforcement, and political economy factors. Specifically, the study recommends a re-assignment of the devolved functions—between national and local, and among different levels of local government—that not only accounts for institutional constraints but also aligns benefits and costs and considers inter-jurisdictional spillovers.

Grapevine is perhaps the most widely grown fruit crop. To understand the genetic make-up so as to improve the yield and quality of grapes and grape products, researchers in Europe have recently sequenced the genomes of Pinot noir and its inbred. As expected, function of many grape genes is unknown. Functional genomics studies have become the major focus of grape researchers and breeders. Current genetic approaches for gene function studies include mutagenesis, crossing and genetic transformation. However, these approaches are difficult to apply to grapes and takes long periods of time to accomplish. It is thus imperative to seek new ways for grape functional genomics studies. Virus-induced gene silencing (VIGS) offers an attractive alternative for this purpose and has proven highly effective in several herbaceous plant species including tomato, tobacco and barley. VIGS offers several advantages over existing functional genomics approaches. First, it does not require transformation to silence a plant gene target. Instead, it induces silencing of a plant gene through infection with a virus that contains the target gene sequence, which can be accomplished within a few weeks. Second, different plant genes can be readily inserted into the viral genome via molecular cloning and functions of a large number of genes can be identified within a short period of time. Our long-term goal of this research is to develop VIGS-based tools for grapevine functional genomics, made of the genomes of Grapevine virus A (GVA) from Israel and Grapevine rupestris stem pitting-associated virus (GRSPaV) from Canada. GVA and GRSPaV are members of the Flexiviridae. Both viruses have single-stranded, positive sense RNA genomes, which makes them easy to manipulate genetically and excellent candidates as VIGS vectors. In our three years research, several major breakthroughs have been made by the research groups involved in this project. We have engineered a cDNA clone of GVA into a binary vector that is infectious upon delivery into plantlets of micropropagated Vitis viniferacv. Prime. We further developed the GVA into an expression vector that successfully capable to silence endogenous genes. We also were able to assemble an infectious full-length cDNA clones of GRSPaV. In the following sections Achievements and Detailed description of the research activities, we are presenting the outcome and results of this research in details.

Review question / Objective: To summarise measures or aspects targeted at reducing the incidence of adverse events during the delivery of exercise interventions through telerehabilitation in patients after stroke. Background: The sequelae in people with stroke are diverse. Regarding physical function post-stroke, functional impairment of the upper and lower extremities is common, which may be due to weakness or paralysis, sensory loss, spasticity, and abnormal motor synergies. In addition, a near 15% prevalence of sarcopenia has been found in people with stroke. Gait impairment has been observed in a high percentage of people with stroke, a dysfunction that may persist despite rehabilitation. More than 50% of people with stroke may experience limitations in activities such as shopping, housework, and difficulty reintegrating into community life within 6 months. These restrictions can result in a diminished health-related quality of life.

In recent years, Indonesia has introduced reforms to its bureaucracy in response to critiques of the quality of government policy design and delivery. The Grand Design of Bureaucratic Reform strategy seeks to reduce the number of civil servants employed in administrative or managerial positions (structural appointments) in favour of skills-based recruitment into ‘functional’ positions. Specifically, the introduction of the ‘policy analyst’ position as a functional position in the civil service has sought to improve evidence-based policy making and the quality of policy outcomes, by incorporating merit-based recruitment, appointment and promotion. The role of functional policy analysts (Jabatan Fungsional Analis Kebijakan or JFAKs) is to assist policy makers in identifying policy issues, analyse evidence available on these issues, and ultimately make policy recommendations. This report overviews the recent experiences of different policy analyst cohorts since the role’s creation in 2015. It investigates these experiences to better understand the extent to which policy analysts are playing the role intended for them, and the factors enabling or inhibiting this.

This paper reviews the literature and documented evidence on capacity and coordination issues in crisis situations, where social protection and humanitarian assistance intersect. The paper finds that while there is a burgeoning literature that mentions capacity and coordination, very little of this focuses on crisis situations. Although both terms are mentioned frequently, they are rarely defined or robustly and systematically assessed. The little literature that does exist points to a substantial knowledge gap on both the ways in which capacity and coordination deficits undermine the delivery of social assistance in crisis situations and what can be done to overcome these deficits. Frameworks that could be useful in exploring these questions in crisis situations are identified including those that differentiate between technical and functional elements of capacity, and between technical, political, social and behavioural aspects of coordination.
