First steps toward a classification of irreducible symplectic 4-folds whose integral 2-cohomology with 4-tuple cup product is isomorphic to that of (K3)[2]. We prove that any such 4-fold deforms to an irreducible symplectic 4-fold of Type A or Type B. A 4-fold of Type A is a double cover of a (singular) sextic hypersurface and a 4-fold of Type B is birational to a hypersurface of degree at most 12. We conjecture that Type B 4-folds do not exist.

This study aimed to clarify the characteristics of rat vocal fold scarring by examining the alteration of key components in the extracellular matrix: hyaluronic acid, collagen, and fibronectin. Under monitoring with a 1.9-mm-diameter telescope, unilateral vocal fold stripping was performed, and larynges were harvested at 2, 4, 8, and 12 weeks after operation. The vocal folds were histologically analyzed with Alcian blue stain, trichrome stain, and immunofluorescence of collagen type I, collagen type III, and fibronectin. The scarred vocal folds showed less hyaluronic acid and more collagen types I and III than did the controls at all time points. Type III was stable for 12 weeks, while type I declined until 8 weeks and thereafter remained unchanged. Fibronectin increased for 4 weeks and then decreased; it was close to the control level at 8 and 12 weeks. These results suggest that the tissue remodeling process in scarred vocal folds slows down around 2 months after wounding.

This study investigated the prethyroplasty and postthyroplasty voices of patients with glottic incompetence of mobile vocal folds related to vocal fold bowing and scarring. Seventeen patients underwent vocal function evaluation preoperatively and 1 month postoperatively with videostrobolaryngoscopic examination, acoustic and aerodynamic analysis, and perceptual judgment of voice characteristics. The postoperative voice outcome in this group of patients was compared to that of a group of patients with unilateral vocal fold paralysis. Patients with vocal fold bowing showed significant improvement in glottic gap size and hoarseness after the surgery. There was minimal improvement on other test measures. Patients with vocal fold scarring exhibited worse preoperative and postoperative vocal functions, with little voice improvement after surgery. The outcome of thyroplasty type I in cases of vocal fold bowing or scarring is not as good as that in unilateral vocal fold paralysis.

This study reviewed the current literature on technical aspects regarding controlled vocal fold injuries in the rat model. Data from PubMed, Embase, and Scopus database for English language literature was collected to identify methodological steps leading to a controlled surgical injury of the rat vocal fold. Inclusion criteria: full disclosure of anesthesia protocol, positioning of the rat for surgery, vocal fold visualization method, instrumentation for vocal fold injury, vocal fold injury type. Articles with partial contribution were evaluated and separately included due to the limited number of original methodologies. 724 articles were screened, and eleven articles were included in the analysis. Anesthesia: ketamine hydrochloride and xylazine hydrochloride varied in dose from 45 mg/kg and 4.5 mg/kg to 100 mg/kg and 10 mg/kg. Visualization: The preferred method was the 1.9 mm, 25–30 degree endoscopes. The widest diameter endoscope used was 2.7 mm with a 0 or 30 degree angle of view. Instruments for lesion induction range from 18 to 31G needles, microscissors, micro forceps to potassium titanyl phosphate, and blue light lasers. Injury types: vocal fold stripping was the main injury type, followed by vocal fold scarring and charring. One article describes scaffold implantation with injury to the superior aspect of the vocal fold. Rats are good candidates for in vivo larynx and vocal folds research. A more standardized approach should be considered regarding the type of vocal fold injury to ease data comparison.

Kanjamalai one of the fascinating location in Southern Granulite Terrain (SGT) for studying Archaean geology and structures as the entire hill is made up of variety of rock types like two pyroxene granulite, amphibolites, quartzo - feldsapthic gneisses, banded iron formation, and intrusive rocks like dunite, peridotite and pegmatite and beautifully carved structures. The entire hill resembles a canoe shape with doubly plunging fold structure with E-W elongation. The entire hillock seems to sit pretty on mylonitised hornblende biotite gneisses which also have a common N70-95 degree trend and sub vertical dip with NE plunge which is in contradiction to centrally plunging lineations of the hill. The SW part of Kanjamalai near Chinasrirangapadi was displaying beautiful fold structures, with interference pattern out of which six domains were selected for detailed study and analysis. The multiple generation folded structure will have a clue in reconstructing the deformation history of this Kanjamalai. The observed f1, f2 and f3 folds show significant Type III interference pattern as that of Ramsay and 01 and 03 type folds of Bernhard Grasemann. Wavelength –amplitude analysis was made to generalize and regroup the observed folds in to high amplitude, high wavelength or open folds, low wavelength and Mesoscopic folds. And visual harmonic analysis was made to analyse the symmetry of the folds and analyze the geometry, symmetry and harmony and genesis of the fold in terms of relative timing of the events.

CPA/AT transporters are made up of scaffold and a core domain. The core domain contains two non-canonical helices (broken or reentrant) that mediate the transport of ions, amino acids or other charged compounds. During evolution, these transporters have undergone substantial changes in structure, topology and function. To shed light on these structural transitions, we create models for all families using an integrated topology annotation method. We find that the CPA/AT transporters can be classified into four fold-types based on their structure; (1) the CPA-broken fold-type, (2) the CPA-reentrant fold-type, (3) the BART fold-type, and (4) a previously not described fold-type, the Reentrant-Helix-Reentrant fold-type. Several topological transitions are identified, including the transition between a broken and reentrant helix, one transition between a loop and a reentrant helix, complete changes of orientation, and changes in the number of scaffold helices. These transitions are mainly caused by gene duplication and shuffling events. Structural models, topology information and other details are presented in a searchable database, CPAfold (cpafold.bioinfo.se).

The effects of maturation on the interrelationship between skeletal muscle fiber area and capillarization was investigated in specific fiber types (I, IIa, IIb, IIc) of male Wistar rats at seven developmental periods ranging from 8 to 85 days postnatal. Fiber type specific developmental properties were compared in three different muscles, the diaphragm (DIA), extensor digitorum longus (EDL), and soleus (SOL), which are known to differ widely in function. All fiber types in each of the three muscles examined exhibited large increases in area (FA), the magnitude and time course of the increase being related to both the type of fiber and the muscle in which the fiber was located. For type I fibers, areas increased from 3- to 18-fold (SOL greater than EDL greater than DIA), whereas in type IIa fibers, area increased ranged between 5- to 11-fold (SOL greater than EDL greater than DIA). Growth rates in IIb fibers were more homogeneous between muscles ranging from 11- to 14-fold. Capillarization, as indicated by the capillary contacts per fiber (CC), increased in all fiber types regardless of muscle origin. These increases ranged between 1.7- and 2.2-fold for type I fibers, between 2.4- and 2.5-fold for type IIa fibers, and between 2.0- and 3.0-fold for type IIb fibers. In general, capillary density expressed as the ratio of the number of capillary contacts divided by the fiber area (CC/FA) progressively declined in all fiber types with age. The rate of the decline in CC/FA was mediated in large part by the changes in fiber area.(ABSTRACT TRUNCATED AT 250 WORDS)

Injection laryngoplasty is a widely used therapeutic option for drug delivery into vocal folds (VFs). Efficient injectable materials are urgently needed for treating intractable VF disease. Liquid-type non-thermal atmospheric plasma (LTP) has been found to be useful for various biological applications, including in regenerative medicine. We evaluated the effects of LTP on VF regeneration. Migration and matrix metalloproteinase-2 expression of lipopolysaccharide (LPS)-treated human vocal fold-derived mesenchymal stem cells (VF-MSCs) were enhanced by LTP treatment. LTP treatment not only ameliorated nuclear factor-κB and interleukin-6 activation, induced by LPS treatment, but also the increased manifestation of α-smooth muscle actin and fibronectin, induced by transforming growth factor-ß. In a rabbit VF scarring animal model, histological analyses showed increased hyaluronic acid deposition and decreased collagen accumulation after LTP injection. Videokymographic analysis showed more improved vibrations in LTP-treated VF mucosa compared to those in non-treated group. In conclusion, LTP treatment enhanced the recruitment and activation of VF-MSCs. Regulated extracellular matrix (ECM) synthesis and eventual functional improvement of scarred VFs were observed upon LTP treatment. The results of this study suggest that LTP injection can enhance wound healing and improve functional remodeling following VF injury. Impact statement Voice disorder has a significant impact on life quality, and one of the major causes of this voice disorder is vocal fold scarring. Therefore, various approaches have been tried to treat for voice disorder. However, no method has satisfied all requirements until now. Plasma medicine, which involves the medical application of plasma, is a rapidly developing field. We have confirmed that liquid-type plasma improved vocal fold scarring by mobilizing and activating vocal fold fibroblast. In conclusion, liquid-type plasma is a potential therapeutic agent for promoting vocal fold scarring through simple injection and it may be an alternative therapeutic agent for the current situation to treat voice disorder.

Abstract Multifunctional enzyme type 2 (MFE-2) is a peroxisomal enzyme participating in the breakdown of fatty acids in eukaryotes. Depending on the organism, MFE-2 is composed of two to four functional units, out of which the two enzymatic ones, 2-enoyl-coenzyme A (CoA) hydratase 2 and (3R)-hydroxyacyl-CoA dehydrogenase, are found in the all MFE-2s. These units are responsible for the catalysis of the second and third steps of the peroxisomal β-oxidation of various CoA thioesters of fatty acids and fatty acyl derivatives. Their (R)-stereospecificity and ability to accept a broad range of fatty acid CoA esters as substrates, in addition to the fact that they do not share any sequence similarity with the classical mitochondrial counterparts, make the enzymatic units of MFE-2 structurally very interesting. In this study, the three-dimensional structures of the (3R)-hydroxyacyl-CoA dehydrogenase and 2-enoyl-CoA hydratase 2 units were solved by crystallographic methods. The crystal structure of the (3R)-hydroxyacyl-CoA dehydrogenase unit of rat MFE-2 reveals a dimeric enzyme with an α/β short-chain alcohol dehydrogenase/reductase (SDR) fold. A unique feature of (3R)-hydroxyacyl-CoA dehydrogenase, however, is the separate C-terminal domain, which completes the active site cavity of the adjacent monomer and extends the dimeric interactions. The 2-enoyl-CoA hydratase 2 unit is a dimer with a unique two-domain structure proposed to evolve via gene duplication. The fold consists of two side-by-side arranged repeats of the hot-dog fold motifs, thus being highly reminiscent of the tertiary structures of the (R)-specific 2-enoyl-CoA hydratase of the polyhydroxyalkanoate synthesis pathway and the β-hydroxydecanoyl thiol ester dehydrase of fatty acid synthesis type II, both from prokaryotic sources. The importance of the N-domain in the binding of bulky substrates was shown by the enzyme-product complex structure, which also indicates the active site. For the first time, it was shown that the eukaryotic hydratase 2 uses an Asp/His catalytic dyad in catalysis. Moreover, a novel catalytic mechanism was proposed for (R)-specific hydration/dehydration. The solved structures also provide a molecular basis for understanding the effects of the patient mutations of MFE-2. They also allow disussion of the possible organisation of the three units in full-length MFE-2 of mammals.

Abstract The peroxisomal multifunctional enzyme type-1 (perMFE-1) is a monomeric protein of β-oxidation possessing 2-enoyl-CoA hydratase-1, Δ3-Δ 2-enoyl-CoA isomerase, and (3S)-hydroxyacyl-CoA dehydrogenase activities. The amino-terminal part of perMFE-1 shows sequence similarity to mitochondrial 2-enoyl-CoA hydratases (ECH-1) and Δ3-Δ 2-enoyl-CoA isomerases, and belongs to the hydratase/isomerase superfamily. Family members with known structures are either homotrimers or homohexamers. The purpose of this work was to elucidate the structure-function relationship of the rat perMFE-1 with special reference to the hydratase/isomerase fold. The structural adaptations required for binding of a long chain fatty acyl-CoA were studied with rat ECH-1 via co-crystallization with octanoyl-CoA. The crystal structure revealed that the long chain fatty acyl-CoA is bound in an extended conformation. This is possible because, a flexible loop moves aside and opens a tunnel, which traverses the subunit from the solvent space to the intertrimer space. Structural and enzymological studies have shown the importance of Glu144 and Glu164 for the catalysis by ECH-1. In the present work the enzymological properties of Glu144Ala and Glu164Ala variants of ECH-1 were studied. The catalytic activity of hydration was reduced about 2000-fold. It was also demonstrated that rat ECH-1 is capable of catalyzing isomerization. The replacement of Glu164 with alanine reduced the isomerase activity 1000-fold, confirming the role of Glu164 in both the hydratase and isomerase reactions. The structural factors favoring the hydratase over the isomerase reaction were addressed studying the enzymological properties of the Gln162Ala, Gln162Met, and Gln162Leu variants. These mutants had similar enzymatic properties to wild type, thus the catalytic function of the Glu164 side chain in the hydratase and isomerase reaction does not depend on interaction with the Gln162 side chain. The perMFE-1 was divided into five functional domains based on amino acid sequence comparisons with the homologous proteins with known structures. Deletion variants of perMFE-1 showed that the folding of an enzymatically active amino-terminal hydratase/isomerase domain requires stabilizing interactions from the two carboxy-terminal domains of perMFE-1. The last carboxy-terminal domain is also required for the folding of the dehydrogenase part of perMFE-1. The dehydrogenase part of perMFE-1 was crystallized.

La determination de la premiere structure d'une molecule d'immunoglobuline (ig) a montre une chaine polypeptidique organisee en domaines ou les brins sont disposes en deux feuillets antiparalleles. La disposition et la connectivite des brins est typique et caracterise le repliement de type ig (ig fold). Ce mode de repliement a ete retrouve dans d'autres proteines distinctes des igs. Ces proteines constituent un groupe heterogene, communement appele la superfamille des igs. Nous nous sommes pose la question suivante : l'apparition de ce type de repliement dans des proteines non apparentees est-elle commandee par un ancetre commun, ou ce repliement constitue-t-il un etat particulierement stable et robuste sur le plan thermodynamique. Pour tenter d'y repondre, nous proposons dans un premier temps une banque de donnees regroupant les membres de la superfamille, et nous montrons l'heterogeneite de cette superfamille en terme de fonctions biologiques, de distribution tissulaire et d'apparition dans diverses especes. Dans un deuxieme temps, nous comparons les sequences et l'architecture des membres de la superfamille dont on connait la structure 3d. Finalement, nous modelisons la structure 3d de la prolactine de lapin en complexe avec le domaine extracellulaire de son recepteur, lui-meme membre de la superfamille des igs. L'hypothese la plus plausible concernant le cheminement evolutif de cette superfamille serait qu'evolutions convergente et divergente aient opere de concert, l'une au plan global de la superfamille, l'autre au sein des sous-familles la composant.

The purpose of this research was to examine food security for people with prediabetes participating in the Supplemental Nutrition Assistance Program (SNAP), focusing on their food choice decisions and coping strategies over a 30-day benefit cycle that potentially increases the risk of Type 2 diabetes. A cross-sectional, quantitative design based on food choice process model constructs was used. SNAP participants (n = 36) with prediabetes, aged 21â??70 years, were recruited as outpatients from Bronx Lebanon Hospital and completed self-reported questionnaires on demographics and health, food security, and food frequency. Descriptive statistics, Pearson chi square tests, and regression analysis were performed using SPSS. Also, independent t test, and Levene's test were used for ad hoc analysis to assess variation of food choice decisions over 30 days. Of the sample, 5% had low and 95% very low food secure status. Food security status did not predict coping strategies (p = .724); however, food security status and type of coping strategy had a moderate relationship (p < 0.01; r =.60). Food choices of 11 food categories changed over a 30-day cycle with greatest variation for Week 1, compared to Weeks 2â??4 (p < .005). Use of coping strategies to minimize hunger was limited. Very low food security associated with certain coping strategies disrupted eating patterns. Disrupted eating patterns affect food variation over time, increasing the intake of non-nutrient-dense foods and the risk of obesity and Type 2 diabetes. The implications for positive social change include the potential to change SNAP's benefit allotments, make nutrition education mandatory, and create a nutrition package, thereby lowering food insecurity and the risk of Type 2 diabetes.

Whole sorghum kernels were directly processed into whole grain snacks with acceptable texture. Extrudates made from whole sorghum had a harder gritty texture than those made from decorticated materials. Extrusion of whole sorghum provides significant savings in processing: there are no dry matter losses; no equipment or energy is required for decortication or milling; the extruder consumes less power and processes more material per time unit. In addition, the extruder utilized is a simple adiabiatic, friction extruder of relatively low cost. Grinding whole sorghum and removing fines did not improve product expansion during extrusion but altered the gritty pieces in the extrudates. The best products were obtained when whole sorghum (ground or un-ground) was extruded at 14% moisture. The whole sorghum extrudates had larger bubbles with thick cell walls, which made extrudates more crunchy. Decortication of sorghum improved extrusion performance and products by allowing adequate formation and retention of air cells. Decortication to remove 20% of the original sorghum weight was enough to produce extrudates with characteristics comparable to those made from commercial yellow corn meal. Sorghum milled fractions with composition and particle size distribution similar to corn meal produced extrudates with higher expansion, lower bulk density and similar texture. In addition, sorghum extrudates were rated equal to corn meal extrudates by a taste panel for appearance, flavor, texture and overall characteristics. Extrudates made from polished rice were less expanded and whiter than extrudates made from sorghum. When processed under similar conditions, sorghum extrusion required more energy than corn meal extrusion. However, whole sorghum extrusion required less energy than corn meal extrusion. Unground sorghum samples (decorticated or non-decorticated) produced harder extrudates compared to those made from ground raw material. White sorghum is a feasible option for snack extrusion because of its versatility, product characteristics, cost and processing properties.

AbstractThis paper considers parametricity and its resulting free theorems for nested data types. Rather than representing nested types via their Church encodings in a higher-kinded or dependently typed extension of System F, we adopt a functional programming perspective and design a Hindley-Milner-style calculus with primitives for constructing nested types directly as fixpoints. Our calculus can express all nested types appearing in the literature, including truly nested types. At the term level, it supports primitive pattern matching, map functions, and fold combinators for nested types. Our main contribution is the construction of a parametric model for our calculus. This is both delicate and challenging: to ensure the existence of semantic fixpoints interpreting nested types, and thus to establish a suitable Identity Extension Lemma for our calculus, our type system must explicitly track functoriality of types, and cocontinuity conditions on the functors interpreting them must be appropriately threaded throughout the model construction. We prove that our model satisfies an appropriate Abstraction Theorem and verifies all standard consequences of parametricity for primitive nested types.

AbstractLarge solar energetic-particle (SEP) events are clearly associated in time with eruptive phenomena on the Sun, but how? When large SEP events were first observed, flares were the only visible candidate, and diffusion theory was stretched to explain how the particles could spread through space, as widely as observed. The observation of coronal mass ejections (CMEs), and the wide, fast shock waves they can drive, provided better candidates later. Then small events were found with 1000-fold enhancements in 3He/4He that required a different kind of source—should we reconsider flares, or their open-field cousins, solar jets? The 3He-rich events were soon associated with the electron beams that produce type III radio bursts. It seems the radio astronomers knew of both SEP sources all along. Sometimes the distinction between the sources is blurred when shocks reaccelerate residual 3He-rich impulsive suprathermal ions. Eventually, however, we would even begin to measure the source-plasma temperature that helps to better distinguish the SEP sources.

AbstractDynamic processes have always been of profound interest for scientists and engineers alike. Often, the mathematical models used to describe and predict time-variant phenomena are uncertain in the sense that governing relations between model parameters, state variables and the time domain are incomplete. In this paper we adopt a recently proposed algorithm for the detection of model uncertainty and apply it to dynamic models. This algorithm combines parameter estimation, optimum experimental design and classical hypothesis testing within a probabilistic frequentist framework. The best setup of an experiment is defined by optimal sensor positions and optimal input configurations which both are the solution of a PDE-constrained optimization problem. The data collected by this optimized experiment then leads to variance-minimal parameter estimates. We develop efficient adjoint-based methods to solve this optimization problem with SQP-type solvers. The crucial test which a model has to pass is conducted over the claimed true values of the model parameters which are estimated from pairwise distinct data sets. For this hypothesis test, we divide the data into k equally-sized parts and follow a k-fold cross-validation procedure. We demonstrate the usefulness of our approach in simulated experiments with a vibrating linear-elastic truss.

The human vocal folds are subjected to complex dynamic biomechanical stimulation during phonation. The aim of the present study was to develop and evaluate an airflow-induced self-oscillating mechanical model, i.e., a bioreactor, which mimics the geometry and the mechanical microenvironment of the human vocal folds. The bioreactor consisted of two composite synthetic vocal fold replicas loaded into a custom-built airflow supplied tube. A cell-scaffold mixture was injected into cavities within the replicas. The folds were phonated using a variable speed centrifugal blower for two hours a day over a period of seven days. The static and dynamic subglottal pressures and the dynamic supraglottal pressure were monitored. A similar bioreactor without mechanical excitation was used as positive control. The cell-scaffold mixture was harvested for cell viability and collagen type I immunohistochemistry tests seven days after injection. The flow-induced self-oscillations of the vocal fold replicas were shown to produce mechanical excitations that are typical of those in the human vocal fold lamina propria during phonation. The results confirmed that human vocal fold fibroblasts survived inside the present bioreactor, and maintained cellular functions of protein production.

The endothelial cell type plasminogen activator inhibitor (PAI-1) may exist in an active, latent form that can be converted into an active form upon exposure to denaturants such as sodium dodecyl sulphate (SDS), guanidine-HCl or urea. Here we show that latent PAI-1 can be activated with lipid vesicles, consisting of the negatively charged phospholipids phosphatidylserine (PS) or phosphatidylinositol (PI). The presence of a net negative charge on the phospholipid headgroup is essential for activation. Incubation with lipid vesicles, consisting of the zwitterionic phospholipids phosphatidylcholine (PC) and phosphatidylethanol-amine (PE), does not result in activation of the inhibitor. In the presence of PS vesicles, the capacity of PAI-1 to inhibit tissue type plasminogen activator (t-PA) is 50-fold higher than that of the untreated protein. For comparison, the activity of PAI-1 was enhanced 25-fold by treatment of the protein with SDS. PS induces activation of the inhibitor at much lower concentrations than SDS. For example, to achieve 50% inhibition of t-PA with a more than 100-fold excess of PAI-1, 0.25 nmoles of PS are required, whereas L.60 nmoles of SDS are necessary to reach half maximal inactivation of t-PA. Activation of PAI-1 by PS can be reversed by the addition of Ca2+-ions, suggesting that Ca2+-ions interfere with the interaction of PAI-1 with the negatively charged lipid surface, thus preventing its activation. The lipid-induced activation of PAI-1 points to a possibly important role of phospholipids in fibrinolysis; regulation of the fibrinolytic activity in blood plasma may ultimately be determined by the extent to which these phospholipids activate the inhibitor of t-PA.This study was supported by the Netherlands Thrombosis Foundation.

Human plasminogen activator inhibitor type-1 is an Mr∼54,000 protein which specifically inhibits urokinase-type (u-PA) and tissue-type (t-PA) plasminogen activators. During inhibition, u-PA and t-PA convert PAI-1 to an inactive form with Mr∼50,000. We have determined the amino-terminal amino acid sequence of native and converted PAI-1, and isolated and partly sequenced PAI-1 cDNA. The data show that the conversion of PAI-1 consists of cleavage of an Arg-Met bond 33 residues from the carboxy-terminus, thus localizing the reactive center of the inhibitor to that position, and identifying PAI-1 as an "arg-serpin". PAI-1 activity is known to be influenced by a number of agents; we have studied the mechanisms of the stimulation of PAI-1 activity by transforming growth factor-β (TGF-β) and the synthetic glucocorticoid dexame-thasone in human WI-38 lung fibroblasts and HT-1080 fibrosarcoma cells. Bytheuse of PAI-1 cDNA, TGF-β was found to course a rapid increase in PAI-1 mRNA level in WI-38 cells, reaching a maximal 50-fold enhancement after 8 hours. Dexamethasone caused a 10-fold increase in PAI-1 mRNA in HT-1080 cells, which was detectable after 4 hours and became maximal after 16 hours. In both cases, the 3.4 as well as the 2.4 Kb-PAI-1-mRNA species were increased. Quantitative studies on the effect of these agents on PAI-1 protein levels in cell extracts and culture media by ELISA gave results consistent with the effects on PAI-1 mRNA. These studies suggest that TGF-β and glucocorticoids may exert important controls over plasminogen activation-mediated extracellular proteolysis through an enhancement of PAI-1 gene transcription.

Site-specific mutagenesis was used to produce a mutant form of tissue-type plasminogen activator (t-PA) which was missing the first 44 amino acids. This domain has sequence homology with the type 1 regions in proteins such as fibronectin and is commonly called the finger domain. The mutant protein (des 1-44 t-PA) was expressed in Chinese Hampster Ovary cells, and was purified using chromatography on Zn-chelate sepharose and lysine-sepharose. Sequence analysis indicated that the resulting protein was homogeneous and started at amino acid 45 in the sequence of the normal protein. The two-chain forms of both des 1-44 t-PA and normal sequence t-PA exhibited similar kinetic constants with a small synthetic substrate (H-D-Isoleucyl-L- prolyl-L-arginyl-p-nitroani 1 ide). The ability of des 1-44 t-PA to activate plasminogen was decreased to 70% of the rate of normal t-PA. The rate of plasminogen activation by normal t-PA was stimulated 51-fold in the presence of fibrin, whereas with des 1-44 t-PA it was stimulated only 40-fold. Although des 1-44 t-PA bound to lysine-agarose, little (if any) binding was observed to either intact or degraded fibrin indicating that fibrin stimulation is due in part to the ability of t-PA to recognize plasminogen bound to fibrin as a preferable substrate. The mutant t-PA was capable of forming complexes in vitro with all of the inhibitors in blood which react with normal sequence t-PA. The rate of reaction with α2-macroglobulin, however, was slower with des 1-44 t-PA than with normal sequence t-PA. The similar resistance of des 1-44 t-PA and normal sequence t-PA to proteolysis and the ability to react with a battery of monoclonal antibodies suggests that the deletion did not cause perturbed folding, but rather that alterations in function of des 1-44 t-PA were due to the lack of the finger domain.

An experimental study of the vibratory deformation of the human vocal folds was conducted. Experiments were performed using model vocal folds [1, 2], Fig. 1, made of silicone rubber implemented into an air supply system, Fig. 2. The material used to cast the model is an isotropic homogeneous material, [3] with a tangent modulus E=5 kPa at ε = 0, i.e. elastic properties similar to those of the human vocal fold cover [4]. The advantages of the use of model larynx systems over the use of excised larynges include easy accessibility to fundamental studies of the vocal fold vibration without invasive testing. Acoustic analysis of voice or electroglottography provide certain insight into voice production processes but optical techniques for the study of vocal fold vibrations have drawn considerable attention. Videoendoscopy, stroboscopy, high-speed photography, and kymography have shown to provide a visual impression of vocal fold dynamics but are limited in providing insight into the fundamental deformation processes of the vocal folds. Quantitative measures of deformation have been conducted through micro-suture techniques but are invasive and allows for measurements of only view image points. Laser triangulation is non-invasive but is limited to only one local measurement point. Here, digital image correlation technique with the software VIC 3D [5] is applied. For the experimental set-up see Fig. 2. The analysis consists of (1) stereo correlation to obtain in-plane displacements and (2) stereo triangulation step to obtain out-of-plane deformation. For the stereo correlation images of the object at two different stages of deformation are compared. A point in the image of the undeformed object is matched with the corresponding point in the deformed stage. “Subsets” of digital images are traced via their gray value distribution from the undeformed reference image to the deformed image. The uniqueness of the matching is enabled by the creation of a speckle pattern on the object’s surface. Here, a white pigment is mixed into the silicone rubber and subsequently black enamel paint is sprayed onto the superior surface of the vocal folds. The stereo triangulation requires two images of the object at each stage of deformation. These are obtained in a single CCD frame by placing a beam splitter in the optical axis between camera and object. These images provide a “left” and “right” view of the model larynx. Thus, the deformed shape of the vocal folds can be obtained. The method allows for noninvasive measurement of the full-field displacement fields. Images of the superior surface of the model larynx are obtained by the use of a high speed digital camera with a frame rate of 3000 frames per second allowing for more than 30 image frames for each vibration cycle. For the 3D digital image correlation analysis two images of the object are obtained for each time instance as a beam splitter is placed in the optical axis between the camera and the model larynx. Phonation frequencies and onset pressure are given in Fig. 3, showing that the model larynx behavior is close to actual physiological data. Figs 4(a) and (b) provide superior views of the model larynx at maximum glottal opening and at glottal closure, respectively. As one example of measured strain fields, Figs 5(a) and (b) depict the distributions of the transverse strain component, on the glottal surface in a contour plot on the deformed superior surface. The knowledge of the distribution of this strain component is relevant to the assessment of the impact of vocal fold collision on potential tissue damage. In the position of maximum opening the vocal folds are deformed by a combination of a bulging-type deformation and the opening movement. At this time instance, the transverse strains at the medial surface are found to be negative, an indication of Poisson’s deformation. During the closing stage, vocal folds collide and simultaneously a mode 3 vibration pattern emerges. Closure of the glottal opening is not complete and two incomplete closure areas are formed during the closure stage. These open areas are located at the anterior and posterior ends of the model larynx, see Fig. 4(b). The finding of this type of incomplete closure is agreement with both actual glottal measurements [6] and 3D finite element simulations of [7]. Transverse strains during that stage are now positive and considerably larger that during the opening stage. Finally, Fig. 6 depicts the time evolution of the out of plane displacements along the medial surface for the closing phase and Fig. 7 depicts the maximum values of the longitudinal strain (at the coronal section of the medial surface) in dependence of the flow rate. These examples of measurements indicate that the DIC method is promising for studies of vocal fold dynamics.

Review question / Objective: To assess the beneficial effect and safety of finerenone for patients with chronic kidney disease associated with type 2 diabetes. Condition being studied: Chronic kidney disease (CKD) is a major contributor to morbidity and mortality from non-communicable diseases, affecting almost 700 million people worldwide. Approximately 40% of patients with diabetes have CKD, which exposes them to a 3-fold higher risk of cardiovascular death versus those with T2D alone. Strategies to protect the kidneys of patients with CKD and T2D may reduce their risk of cardiovascular events. Finerenone, a nonsteroidal, selective mineralocorticoid receptor antagonist, reduced composite kidney and cardiovascular outcome in trials involving patients with chronic kidney disease. Recently, quite a few clinical studies have been conducted to compare finerenone and placebo. Original objectives The general goal of the project was to utilize the bactericidal potential of curcumin- functionalizednanostructures (CFN) for reinforcement of food safety by developing active antimicrobial food-contact surfaces. In order to reach the goal, the following secondary tasks were pursued: (a) further enhancement of the CFN activity based on understanding their mode of action; (b) preparing efficient antimicrobial surfaces, investigating and optimizing their performance; (c) testing the efficacy of the antimicrobial surfaces in real food trials. Background to the topic The project dealt with reducing microbial food spoilage and safety hazards. Cross-contamination through food-contact surfaces is one of the major safety concerns, aggravated by bacterial biofilm formation. The project implemented nanotech methods to develop novel antimicrobial food-contact materials based on natural compounds. Food-grade phenylpropanoidcurcumin was chosen as the most promising active principle for this research. Major conclusions, solutions, achievements In agreement with the original plan, the following research tasks were performed. Optimization of particles structure and composition. Three types of curcumin-functionalizednanostructures were developed and tested: liposome-type polydiacetylenenanovesicles, surface- stabilized nanoparticles and methyl-β-cyclodextrin inclusion complexes (MBCD). The three types had similar minimal inhibitory concentration but different mode of action. Nanovesicles and inclusion complexes were bactericidal while the nanoparticlesbacteriostatic. The difference might be due to different paths of curcumin penetration into bacterial cell. Enhancing the antimicrobial efficacy of CFN by photosensitization. Light exposure strengthened the bactericidal efficacy of curcumin-MBCD inclusion complexes approximately three-fold and enhanced the bacterial death on curcumin-coated plastic surfaces. Investigating the mode of action of CFN. Toxicoproteomic study revealed oxidative stress in curcumin-treated cells of E. coli. In the dark, this effect was alleviated by cellular adaptive responses. Under light, the enhanced ROS burst overrode the cellular adaptive mechanisms, disrupted the iron metabolism and synthesis of Fe-S clusters, eventually leading to cell death. Developing industrially-feasible methods of binding CFN to food-contact surfaces. CFN binding methods were developed for various substrates: covalent binding (binding nanovesicles to glass, plastic and metal), sonochemical impregnation (binding nanoparticles to plastics) and electrostatic layer-by-layer coating (binding inclusion complexes to glass and plastics). Investigating the performance of CFN-coated surfaces. Flexible and rigid plastic materials and glass coated with CFN demonstrated bactericidal activity towards Gram-negative (E. coli) and Gram-positive (Bac. cereus) bacteria. In addition, CFN-impregnated plastic material inhibited bacterial attachment and biofilm development. Testing the efficacy of CFN in food preservation trials. Efficient cold pasteurization of tender coconut water inoculated with E. coli and Listeriamonocytogeneswas performed by circulation through a column filled with CFN-coated glass beads. Combination of curcumin coating with blue light prevented bacterial cross contamination of fresh-cut melons through plastic surfaces contaminated with E. coli or Bac. licheniformis. Furthermore, coating of strawberries with CFN reduced fruit spoilage during simulated transportation extending the shelf life by 2-3 days. Understanding mode of antimicrobial action of curcumin will allow enhancing its dark efficacy, e.g. by targeting the microbial cellular adaptation mechanisms.

Background to the topic: For food safety and security reasons, hundreds of pesticides, veterinary drugs, and environmental pollutants should be monitored in the food supply, but current methods are too time-consuming, laborious, and expensive. As a result, only a tiny fraction of the food is tested for a limited number of contaminants. Original proposal objectives: Our main original goal was to develop fast, practical, and effective new approaches for the analysis of hazardous chemicals in the food supply. We proposed to extend the QuEChERS approach to more pesticides, veterinary drugs and pollutants, further develop GC-MS and LC-MS with SMB and combine QuEChERS with GC-SMB-MS and LC-SMB-EI-MS to provide the “ultimate” approach for the analysis of hazardous chemicals in food. Major conclusions, solutions and achievements: The original QuEChERS method was validated for more than 200 pesticide residues in a variety of food crops. For the few basic pesticides for which the method gave lower recoveries, an extensive solvent suitability study was conducted, and a buffering modification was made to improve results for difficult analytes. Furthermore, evaluation of the QuEChERS approach for fatty matrices, including olives and its oil, was performed. The QuEChERS concept was also extended to acrylamide analysis in foods. Other advanced techniques to improve speed, ease, and effectiveness of chemical residue analysis were also successfully developed and/or evaluated, which include: a simple and inexpensive solvent-in-silicone-tube extraction approach for highly sensitive detection of nonpolar pesticides in GC; ruggedness testing of low-pressure GC-MS for 3-fold faster separations; optimization and extensive evaluation of analyte protectants in GC-MS; and use of prototypical commercial automated direct sample introduction devices for GC-MS. GC-MS with SMB was further developed and combined with the Varian 1200 GCMS/ MS system, resulting in a new type of GC-MS with advanced capabilities. Careful attention was given to the subject of GC-MS sensitivity and its LOD for difficult to analyze samples such as thermally labile pesticides or those with weak or no molecular ions, and record low LOD were demonstrated and discussed. The new approach of electron ionization LC-MS with SMB was developed, its key components of sample vaporization nozzle and flythrough ion source were improved and was evaluated with a range of samples, including carbamate pesticides. A new method and software based on IAA were developed and tested on a range of pesticides in agricultural matrices. This IAA method and software in combination with GC-MS and SMB provide extremely high confidence in sample identification. A new type of comprehensive GCxGC (based on flow modulation) was uniquely combined with GC-MS with SMB, and we demonstrated improved pesticide separation and identification in complex agricultural matrices using this novel approach. An improved device for aroma sample collection and introduction (SnifProbe) was further developed and favorably compared with SPME for coffee aroma sampling. Implications, both scientific and agricultural: We succeeded in achieving significant improvements in the analysis of hazardous chemicals in the food supply, from easy sample preparation approaches, through sample analysis by advanced new types of GC-MS and LCMS techniques, all the way to improved data analysis by lowering LOD and providing greater confidence in chemical identification. As a result, the combination of the QuEChERS approach, new and superior instrumentation, and the novel monitoring methods that were developed will enable vastly reduced time and cost of analysis, increased analytical scope, and a higher monitoring rate. This provides better enforcement, an added impetus for farmers to use good agricultural practices, improved food safety and security, increased trade, and greater consumer confidence in the food supply.

Use of Lqh2 mutants (produced at TAU) and rNav1.2a mutants (produced at the US side) for identifying receptor site-3: Based on the fact that binding of scorpion alpha-toxins is voltage-dependent, which suggests toxin binding at the mobile voltage-sensing region, we analyzed which of the toxin bioactive domains (Core-domain or NC-domain) interacts with the DIV Gating-module of rNav1.2a. This analysis was based on the assumption that the dissociation of toxin mutants upon depolarization would vary from that of the unmodified toxin should the substitutions affect a site of interaction with the channel Gating-module. Using a series of toxin mutants (mutations at both domains) and two channel mutants that were shown to reduce the sensitivity to scorpion alpha-toxins, and by comparison of depolarization-driven dissociation of Lqh2 derivatives off their binding site at rNav1.2a mutant channels we found that the toxin Core-domain interacts with the Gating-module of DIV. Details of the experiments and results appear in Guret al (2011). Mapping receptor site 3 at Nav1.2a by extensive channel mutagenesis (Seattle): Since previous studies with photoaffinity labeling and antibody mapping implicated domains I and IV in scorpion alpha-toxin binding, Nav1.2 channel mutants containing substitutions at these extracellular regions were expressed and tested for receptor function by whole-cell voltage clamp. Of a large number of channel mutants, T1560A, F1610A, and E1613A in domain IV had ~5.9-, ~10.7-, and ~3.9-fold lower affinities for the scorpion toxin Lqh2, respectively, and mutant E1613R had 73-fold lower affinity. Toxin dissociation was accelerated by depolarization for both wild-type and mutants, and the rates of dissociation were also increased by mutations T1560A, F1610A and E1613A. In contrast, association rates for these three mutant channels at negative membrane potentials were not significantly changed and were not voltage-dependent. These results indicated that Thr1560 in the S1-S2 loop, Phe1610 in the S3 segment, and Glu1613 in the S3-S4 loop in domain IV participate in toxin binding. T393A in the SS2-S6 loop in domain I also showed a ~3.4-fold lower affinity for Lqh2, indicating that this extracellular loop may form a secondary component of the toxin binding site. Analysis with the Rosetta-Membrane algorithm revealed a three-dimensional model of Lqh2 binding to the voltage sensor in a resting state. In this model, amino acid residues in an extracellular cleft formed by the S1-S2 and S3-S4 loops in domain IV that are important for toxin binding interact with amino acid residues on two faces of the wedge-shaped Lqh2 molecule that are important for toxin action. The conserved gating charges in the S4 transmembrane segment are in an inward position and likely form ion pairs with negatively charged amino acid residues in the S2 and S3 segments (Wang et al 2011; Gurevitz 2012; Gurevitzet al 2013).

The extensive use or non-specific, hazardous, chemical insecticides demands the development of "healthier" alternative means for pest control. Insect-specific, baculoviruses expressing anti-insect toxin genes (from mites or scorpions) demonstrated in laboratory assays and field trials enhanced insecticidal activity and provided some protection from lepidopterous larvae to agricultural plantations. To utilize recombinant baculoviruses as commercial biopesticides in row crop agriculture, further increase in their speed of kill should be achieved and the reduction in crop damage should be comparable to the levels obtained with organic insecticides (the problem). In this project we developed strategies to improve further the efficacy of recombinant baculoviruses which included: I) Synergism among baculoviruses expressing different anti-insect toxins: a) Synergism among two complementary anti-insect scorpion neurotoxins each expressed by a separate recombinant baculovirus, both regulated by the same or a different viral promoter. b) Synergism among two complementary anti-insect scorpion neurotoxins expressed by the same recombinant virus, both regulated by the same or a different viral promoter respectively. The above included two classes of pharmacologically complementary toxins: i) toxins with strictly anti-insect selectivity (excitatory and depressant); ii) toxins with preferential anti-insect activity (anti-insect alpha toxins). c) Synergism among wild type viruses, recombinant baculoviruses and chemicals (insecticides and phytochemicals) II) Identification of more potent toxins against lepidopterous pests for their expression by baculoviruses. Our approach was based on the synergistic effect displayed by the combined application of pairs of anti-insect toxins to blowfly and lepidopterous larvae that resulted in 5 fold increase in their insecticidal activity without apparent increase in their anti-mammal toxicity (toxins LqhIT2 and LqhaIT, LqhIT2 and AaIT, and LqhaIT and AaIT (1). Thus, we developed new concepts and produced a "second generation" of recombinant baculoviruses with enhanced potencies and speeds of kill comparable to classical insecticides. These achievements contribute to make these biopesticides a viable alternative to minimize the use of hazardous chemicals in pest control. Also, our project contributed new tools and model systems to advance the study of insect sodium channels.

Increasing food self-sufficiency means rethinking global supply chains, (re-)adapting to local contexts, and ensuring optimal conditions for selling and buying locally produced food. Increased self-sufficiency and improved local food systems can have positive environmental, social, and economic consequences. However, whether increased self-sufficiency adds to more sustainable food systems depends on myriad factors, including production methods, the type of food in question, and the availability of local food on the local market. Previous research shows that local food production does not automatically equate to sustainable food production. This project sought to increase knowledge of how greater food self-sufficiency can contribute to increased sustainability and resilience in the food systems of five Nordic island societies: Bornholm, the Faroe Islands, Greenland, Iceland, and Åland.

Based on the generalization and systematization of scientific and accounting data, the article considers agriculture as one of the leading sectors of the Ukrainian economy, providing 50% of foreign exchange earnings from exports of all goods from the country. The structure and geography of agricultural exports from Ukraine are analyzed. The existing export potential of the studied sector of the national economy and the level of self-sufficiency of Ukraine in agri-food products are revealed. Four specialization types of agricultural exports from Ukrainian regions are established: areas with dominance in export of animal origin products; areas with a dominance in the export of plant products; areas with a predominance in the export of animal or vegetable origin fats and oils; area, with mixed export specialization. Recommendations for the re-profiling of agricultural production in a number of the country’s regions in order to increase exports of this product type.
