ABSTRACT A putative ABC transporter, fit, with significant homology to several bacterial iron transporters was identified in Escherichia coli. The E. coli fit system consists of six genes designated fitA, -B, -C, -D, -E, and -R. Based on DNA sequence analysis, fit encodes an outer membrane protein (FitA), a periplasmic binding protein (FitE), two permease proteins (FitC and -D), an ATPase (FitB), and a hypothetical protein (FitR). Introduction of the E. coli fit system into E. coli strain K-12 increased intracellular iron content and transformed bacteria were more sensitive to streptonigrin, which suggested that fit transports iron in E. coli. Expression of fit was studied using a lacZ reporter assay. A functional, bidirectional promoter was identified in the intergenic region between genes fitA and fitB. The expression of the E. coli fit system was found to be induced by iron limitation and repressed when Fe2+ was added to minimal medium. Several fit mutants were created in E. coli using an in vitro transposon mutagenesis strategy. Mutations in fit did not affect bacterial growth in iron-restricted media. Using a growth promotion test, it was found that fit was not able to transport enterobactin, ferrichrome, transferrin, and lactoferrin in E. coli.

The suffused, noneverted cheek pouch of ovalbumin (OA)-immunized hamsters was employed to study changes in vascular permeability utilizing intravital light microscopy and direct measurements of plasma to suffusate tracer efflux. Suffusion of the cheek pouches of immunized animals with OA for 10 min stimulated the formation of focal venular fluorescein isothiocyanate dextran (FITC-D, 70,000 Da) leakage sites and produced increases in the plasma to suffusate FITC-D efflux resulting in marked increases in concentration of FITC-D in suffusate ([FITC-D]s). The intravenous injection of the FITC-D 15 to 60 min after the start of the OA suffusion failed to reveal the formation of vascular FITC-D leakage sites or increases in [FITC-D]s. In contrast, the intravenous injection of the tracer at the start or 10 min after the start of the OA suffusion revealed the formation of venular FITC-D leakage sites and marked increases in [FITC-D]s. Treatment with diphenhydramine or isoproterenol completely inhibited the antigen-stimulated formation of venular FITC-D leakage sites and increases in [FITC-D]s. It is concluded that antigen-antibody reactions resulting in the production of immune complexes trigger immediate increases in the plasma to suffusate FITC-D clearance via the formation of venular FITC-D leakage sites subsequent to the release of endogenous histamine. The increase in venular permeability is marked but transient, lasting approximately 10 min, and is subject to inhibition by either diphenhydramine or isoproterenol.

The purpose of this study was to determine the pathology of cigarette smoke-increased permeability at the bronchioalveolar junction of the guinea pig. After exposure to either smoke or room air, guinea pigs were anesthetized and fluorescein isothiocyanate-dextran (FITC-D, mol wt 10,000) was aerosolized into their lungs. Blood samples taken through a carotid arterial cannula were analyzed by gel chromatography and spectrofluorometry for the presence of FITC-D. The results confirmed that, after smoke exposure, increased amounts of intact FITC-D molecules with a reported Einstein-Stokes radius of 22.2 A crossed the respiratory epithelium into the vascular space. Transmission electron-microscopic studies showed that the FITC-D diffused across damaged type I pneumocyte membranes and cytoplasm to reach the basal lamina and entered the alveolar capillaries through endothelial tight junctions. Damage to the alveolar epithelium was more frequent for the smoke-exposed animals than the room air-exposed animals (P less than 0.05). We conclude that smoke exposure damages type I cells and that inhaled FITC-D crosses the epithelial barrier at damaged type I cells of the bronchioloalveolar junctions.

The purpose of this study was to examine the role of circulating neutrophils in endotoxin-induced increase in microvascular permeability in vivo. Fifteen hamsters were anesthetized, and a plastic chamber was placed in each cheek pouch to observe the microvasculature. Fluorescein-labeled dextran (FITC-D, 150 kDa) was injected intravenously, and changes in leaky sites and FITC-D clearance were measured in three groups: control (saline, n = 4), endotoxin suffusion (n = 6), and endotoxin suffusion after neutropenia induction (n = 5). We found a significant increase in leaky sites and FITC-D clearance with endotoxin (45 +/- 18/cm2 and 20 +/- 6 x 10(-6) ml/min, respectively; mean +/- SD, P less than 0.05) in comparison to control (7 +/- 6/cm2 and 7 +/- 5 x 10(-6) ml/min) and endotoxin suffusion in neutropenic animals (19 +/- 11/cm2 and 12 +/- 4 x 10(-6) ml/min). There was a significant correlation between the number of leaky sites and FITC-D clearance (r = 0.91, P less than 0.01) and between the number of circulating neutrophils and FITC-D clearance (r = 0.87, P less than 0.01). We conclude that endotoxin-mediated increase in microvascular permeability in the peripheral circulation is dependent in part on circulating neutrophils.

Fluorescence recovery after photobleaching (FRAP) was used to quantify the translational diffusion of microinjected FITC-dextrans and Ficolls in the cytoplasm and nucleus of MDCK epithelial cells and Swiss 3T3 fibroblasts. Absolute diffusion coefficients (D) were measured using a microsecond-resolution FRAP apparatus and solution standards. In aqueous media (viscosity 1 cP), D for the FITC-dextrans decreased from 75 to 8.4 × 10−7 cm2/s with increasing dextran size (4–2,000 kD). D in cytoplasm relative to that in water (D/Do) was 0.26 ± 0.01 (MDCK) and 0.27 ± 0.01 (fibroblasts), and independent of FITC-dextran and Ficoll size (gyration radii [RG] 40–300 Å). The fraction of mobile FITC-dextran molecules (fmob), determined by the extent of fluorescence recovery after spot photobleaching, was >0.75 for RG < 200 Å, but decreased to <0.5 for RG > 300 Å. The independence of D/Do on FITC-dextran and Ficoll size does not support the concept of solute “sieving” (size-dependent diffusion) in cytoplasm. Photobleaching measurements using different spot diameters (1.5–4 μm) gave similar D/Do, indicating that microcompartments, if present, are of submicron size. Measurements of D/Do and fmob in concentrated dextran solutions, as well as in swollen and shrunken cells, suggested that the low fmob for very large macromolecules might be related to restrictions imposed by immobile obstacles (such as microcompartments) or to anomalous diffusion (such as percolation). In nucleus, D/Do was 0.25 ± 0.02 (MDCK) and 0.27 ± 0.03 (fibroblasts), and independent of solute size (RG 40–300 Å). Our results indicate relatively free and rapid diffusion of macromolecule-sized solutes up to approximately 500 kD in cytoplasm and nucleus.

We studied the dynamics of macromolecular transport across the microvascular wall in the hamster cheek pouch using intravital microscopy and digital video-image analysis. We used fluorescein isothiocyanate-dextrans of 70,000 and 150,000 Da (FITC-Dextran 70 and 150, respectively) as tracers. We applied our mathematical model and our in vivo calibration to determine the diffusion coefficient (D) and the average fluid velocity (V) in the microvascular wall and in the interstitium from the experimental data. The value of D for FITC-Dextran 70 was 0.90 +/- 0.04 x 10(-11) cm2/s in the wall and 1.29 +/- 0.05 x 10(-8) cm2/s in the interstitium. In both regions, V was 2.05 +/- 0.05 x 10(-8) cm/s. The transport parameters for FITC-Dextran 150 were 0.27 +/- 0.02 x 10(-11) cm2/s, 0.55 +/- 0.05 x 10(-8) cm2/s, and 1.71 +/- 0.48 x 10(-8) cm/s for D in the wall and interstitium and V, respectively. The topical application of either calcium ionophore A23187 (7 x 10(-7) M) or bradykinin (5 x 10(-7) M) increased D for FITC-Dextran 70 and 150 2-fold and V 10-fold relative to their control values. We used these values to quantify the relative importance of the diffusive and convective mechanisms in the total solute flux. Molecular diffusion dominates convective transport in both the microvascular wall and the interstitial space.

The mechanism(s) of the waning of the vasodilation and increase in vascular permeability during prolonged local intraarterial infusions of bradykinin (BK) was investigated in this study. Treatment with phentolamine or saralasin failed to prevent the waning of the vasodilation during the prolonged infusion of BK into forelimbs perfused at constant flow. In contrast, BK produced a sustained vasodilator response after treatment with captopril. Forelimb weight and lymph analysis were used to quantitate edema formation and to determine the duration of the increase in vascular permeability during prolonged local intra-arterial infusions of BK into forelimbs perfused at constant flow. The lymph-to-plasma ratios (L/P) for protein and FITC-Dextrans (fluorescein isothiocyanate dextrans, 70,000 Da) were determined, and clearances for protein and FITC-D were calculated. BK markedly increased fluid filtration, the protein L/P, and protein clearance resulting in edema formation. The protein L/P remained markedly elevated throughout the experimental period. The FITC-D L/P was markedly increased in the groups of animals in which the tracer was injected intravenously at the start or 8 min after the start of the prolonged BK infusion. In the groups of animals in which the tracer was injected intravenously 15-60 min after the start of the prolonged BK infusion, the FITC-D L/P failed to exceed the FITC-D L/P in control animals, although the protein L/P remained elevated. Pretreatment with both captopril and propranolol dramatically potentiated the magnitude of the increase in protein clearance, the filtration rate, and edema formation produced by BK but failed to affect the duration of the transient increase in vascular permeability.

The purpose of this study was to examine whether antioxidants attenuate endotoxin-induced microvascular hyper-permeability for macromolecules in the hamster cheek pouch. Twenty-two adult male Syrian hamsters were anesthetized, and a removable plastic chamber was placed in the cheek pouch to observe and collect suffusate from the microvasculature. Fluorescent-labeled dextran (FITC-D; mol wt 150,000) was injected intravenously, and changes in the number of microvascular leaky sites and microvascular clearance of FITC-D were measured in five groups: saline control (group 1, n = 4), endotoxin (0.1 mg/ml) suffusion for 120 min (group 2, n = 6), endotoxin plus dimethyl sulfoxide (1.0 g/kg iv; group 3, n = 4), endotoxin plus allopurinol (30 mg/kg ip; group 4, n = 4), and endotoxin plus dimethyl sulfoxide and allopurinol (group 5, n = 4). The number of leaky sites and the FITC-D clearance were significantly higher in group 2 [45 +/- 18 (SD) sites/cm2 and 20 +/- 6 X 10(-6) ml/min, respectively; P less than 0.01] than in group 1 (7 +/- 6 sites/cm2 and 7 +/- 5 X 10(-6) ml/min), group 3 (9 +/- 5 sites/cm2 and 8 +/- 2 X 10(-6) ml/min), group 4 (11 +/- 7 sites/cm2 and 9 +/- 4 X 10(-6) ml/min), and group 5 (11 +/- 6 sites/cm2 and 7 +/- 1 x 10(-6) ml/min). The leaky sites appeared predominantly in postcapillary venules. Antigen transport from the airway mucosa to the thoracic lymph nodes (TLNs) was studied in vivo by intratracheal instillation of fluorescein isothiocyanate (FITC)-conjugated macromolecules. After instillation, FITC+ cells with stellate morphology were found deep in the TLN T cell area. Using flow cytometry, an FITC signal was exclusively detected in CD11cmed-hi/major histocompatibility complex class II (MHCII)hi cells, representing migratory airway-derived lymph node dendritic cells (AW-LNDCs). No FITC signal accumulated in lymphocytes and in a CD11chiMHCIImed DC group containing a CD8αhi subset (non–airway-derived [NAW]-LNDCs). Sorted AW-LNDCs showed long MHCIIbright cytoplasmic processes and intracytoplasmatic FITC+ granules. The fraction of FITC+ AW-LNDCs peaked after 24 h and had reached baseline by day 7. AW-LNDCs were depleted by 7 d of ganciclovir treatment in thymidine kinase transgenic mice, resulting in a strong reduction of FITC-macromolecule transport into the TLNs. Compared with intrapulmonary DCs, AW-LNDCs had a mature phenotype and upregulated levels of MHCII, B7-2, CD40, and intracellular adhesion molecule (ICAM)-1. In addition, sorted AW-LNDCs from FITC-ovalbumin (OVA)–instilled animals strongly presented OVA to OVA-TCR transgenic T cells. These results validate the unique sentinel role of airway DCs, picking up antigen in the airways and delivering it in an immunogenic form to the T cells in the TLNs.

Carnivorous plants adsorb prey-derived nutrients partly by endocytosis. This study quantifies endocytosis in Drosophyllum lusitanicum, Drosera capensis, Drosera roseana, Dionaea muscipula and Nepenthes × ventrata. Traps were exposed to 1% fluorescent-labeled albumin (FITC-BSA), and uptake was quantified repeatedly for 64 h. Formation of vesicles started after ≤1 h in adhesive traps, but only after 16 h in species with temporary stomach (D. muscipula and N. × ventrata). In general, there are similarities in the observed species, especially in the beginning stages of endocytosis. Nonetheless, further intracellular processing of endocytotic vesicles seems to be widely different between species. Endocytotic vesicle size increased significantly over time in all species except in D. capensis. Fluorescence intensity of the endocytotic vesicles increased in all species except D. muscipula. After 64 h, estimates for FITC-BSA absorption per gland ranged from 5.9 ± 6.3 ng in D. roseana to 47.8 ± 44.3 ng in N. × ventrata, demonstrating that endocytosis substantially contributes to the adsorption of prey-derived nutrients.

The purpose of mixture experiments is to explore the optimum blends of mixture components, which will provide desirable response characteristics in finished products. D-Optimal minimal designs have been considered for a variety of mixture models, including Scheffe's linear, quadratic, and cubic models. Usually, these D-Optimal designs are minimally supported since they have just as many design points as the number of parameters. Thus, they lack the degrees of freedom to perform the Lack of Fit tests. Also, the majority of the design points in D-Optimal minimal designs are on the boundary: vertices, edges, or faces of the design simplex. In this dissertation, extensions of the D-Optimal minimal designs are developed to allow additional interior points in the design space to enable prediction of the entire response surface. First, the extensions of the D-Optimal minimal designs for two commonly used second-degree mixture models are considered. Second, the methodology for adding interior points to general mixture models is generalized. Also a new strategy for adding multiple interior points for symmetric mixture models is proposed. When compared with the standard mixture designs, the proposed extended D-Optimal minimal design provides higher power for the Lack of Fit tests with comparable D-efficiency.

Sigmoid curves have found broad applicability in biological sciences and biopharmaceutical research during the last decades. A well planned experiment design is essential to accurately estimate the parameters of the model. In contrast to a large literature and extensive results on optimal designs for linear models, research on the design for nonlinear, including sigmoid curve, models has not kept pace. Furthermore, most of the work in the optimal design literature for nonlinear models concerns the characterization of minimally supported designs. These minimal, optimal designs are frequently criticized for their inability to check goodness of fit, as there are no additional degrees of freedom for the testing. This design issue can be a serious problem, since checking the model adequacy is of particular importance when the model is selected without complete certainty. To assess for lack of fit, we must add at least one extra distinct design point to the experiment. The goal of this dissertation is to identify optimal or highly efficient designs capable of checking the fit for sigmoid curve models. In this dissertation, we consider some commonly used sigmoid curves, including logistic, probit and Gompertz models with two, three, or four parameters. We use D-optimality as our design criterion. We first consider adding one extra point to the design, and consider five alternative designs and discuss their suitability to test for lack of fit. Then we extend the results to include one more additional point to better understand the compromise among the need of detecting lack of fit, maintaining high efficiency and the practical convenience for the practitioners. We then focus on the two-parameter Gompertz model, which is widely used in fitting growth curves yet less studied in literature, and explore three-point designs for testing lack of fit under various error variance structures. One reason that nonlinear design problems are so challenging is that, with nonlinear models, information matrices and optimal designs depend on the unknown model parameters. We propose a strategy to bypass the obstacle of parameter dependence for the theoretical derivation. This dissertation also successfully characterizes many commonly studied sigmoid curves in a generalized way by imposing unified parameterization conditions, which can be generalized and applied in the studies of other sigmoid curves. We also discuss Gompertz model with different error structures in finding an extra point for testing lack of fit.

Tato práce si klade za cíl obeznámit čtenáře s 2D CNC obráběním a rozličnými metodami popisu a manipulace s dvojrozměrnými objekty. Poté je představen tzv. Nesting problém a jsou popsány různé metody přístupu k jeho vyřešení. Praktická část pokrývá implementaci vlastního nesting softwaru, jenž je schopný efektivně rozmístit nepravidelné tvary do nepravidelné ohraničené plochy. Na závěr je provedeno zhodnocení řečeného softwaru a také je zmíněno jeho možné využití v praxi.

Ce travail traite de la simulation numérique des équations de Navier Stokes et Euler pour un fluide non homogène avec des conditions aux limites sur la pression et divergence non nulle. Dans un premier temps, on linéarise en partie le problème en découplant la densité du reste du système. La difficulté principale pour le traitement numérique des équations provient de l'équation sur la divergence non nulle (fluide échangeant de la chaleur avec le milieu extérieur). La discrétisation en espace, est opérée par la méthode des moindres carrés en formalisme éléments finis. Une des caractéristiques histopathologiques de la maladie d'Alzheimer (AD) est la présence de plaques amyloïdes formées par les peptides amyloïdes β (Aβ) de 40 et 42 résidus, qui sont les produits de clivage par des protéases de l'APP. Afin de comprendre le rôle des variations structurelles du TM dans le traitement de l'APP, les peptides APP_TM4K ont été étudiés dans la bicouche lipidique en utilisant l’ATR-FTIR et ssNMR. Tandis que la structure secondaire globale du peptide APP_TM4K est hélicoidale, hétérogénéité de conformation et d'orientation a été observée pour le site de clivage γ et , que peuvent avoir des implications dans le mécanisme de clivage et donc dans la production d’Aβ. Les peptides Aβ s'agrègent pour produire des fibrilles et aussi de manière transitoire d'oligomères neurotoxiques. Nous avons constaté qu'en présence de Ca2+, l’Aβ (1-40) forme de préférence des oligomères, tandis qu'en absence de Ca2+ l'Aβ (1-40) s’agrège sous forme de fibrilles. Dans les échantillons sans Ca2+, l’ATR-FTIR révèle la conversion des oligomères en feuillets β antiparallèles en la conformation caractéristique des fibrilles en feuillets β parallèles. Ces résultats nous ont amené à conclure que les Ca2+ stimulent la formation d'oligomères d'Aβ (1-40), qui sont impliqués dans l’AD. Les positions et une orientation précise de deux nouveaux médicaments puissants modulateurs de la γ-sécrétase - le benzyl-carprofen et le sulfonyl-carprofen  dans la bicouche lipidique, ont été obtenus à partir des expériences des ssNMR. Ces résultats indiquent que le mécanisme probable de modulation du clivage par la y-sécrétase est une interaction directe avec le domaine TM de l’APP
A histopathological characteristic of Alzheimer’s disease (AD) is the presence of amyloid plaques formed by amyloid β(A) peptides of 40 and 42 residues-long, which are the cleavage products of APP by proteases. To understand the role of structural changes in the TM domain of APP, APP_TM4K peptides were studied in the lipid bilayer using ATR-FTIR and ssNMR. While the overall secondary structure of the APP_TM4K peptide is helical, conformational and orientational heterogeneity was observed for the y- and for the -cleavage sites, which may have implications for the cleavage mechanism and therefore the production of Aβ. Starting from its monomeric form, Aβ peptides aggregate into fibrils and / or oligomers, the latter being the most neurotoxic. We found that in the presence of Ca2 +, Aβ (1-40) preferably forms oligomers, whereas in the absence of a2 + Aβ (1-40) aggregates into fibrils. In samples without Ca2 +, ATR-FTIR shows conversion from antiparallel β sheet conformation of oligomers into parallel β sheets, characteristic of fibrils. These results led us to conclude that Ca2 +stimulates the formation of oligomers of Aβ (1-40), that have been implicated in the pathogenesis of AD. Position and precise orientation of two new drugs  powerful modulators of γ-secretase  benzyl-carprofen and carprofen sulfonyl  in the lipid bilayer were obtained from neutron scattering and ssNMR experiments. These results indicate that carprofen-derivatives can directly interact with APP. Such interaction would interfere with proper APP-dimer formation, which is necessary for the sequential cleavage by β -secretase, diminishing or greatly reducing Aβ42 production

Enterocytes comprising the single layer of epithelial cells in the intestine are linked together by a series of proteins known as tight junction proteins (TJP). TJP absorb nutrients through selective permeability and act as a barrier between intestinal contents and blood. When the barrier function is compromised this is known as increased intestinal permeability (IP). The aim of the thesis was to investigate biomarkers of increased IP in chickens. Increased IP has been studied extensively in rats, pigs and humans. In contrast, this is a relatively a new area in chickens (Chapter 2). Based on published studies in rats and pigs, lipopolysaccharide (LPS), an endotoxin produced by Gram-negative bacteria, was selected as a method for increasing IP in chickens. Three experiments were conducted utilizing LPS to increase IP in chickens (Chapter 3). These studies concluded that LPS failed to increase IP as measured by permeability of lactulose, rhamnose and mannitol sugars (LMR) and fluorescein isothiocyanate dextran (FITC-d) across the epithelial barrier. However, this work generated useful data and indicated for the first time that 90 minutes post-oral gavage was the optimal time-point for blood sampling. Since fasting for 19.5 hours was applied in the protocol of LPS administration (Chapter 3), a second experiment was conducted with fasting for 19.5 hours and dextran sodium sulphate ingestion along with the LMR sugars and FITC-d (Chapter 4). Additional biomarkers such as intestinal fatty acid binding protein, fecal antitrypsin inhibitor, diamine oxidase and d-lactate were also utilized. This paper concluded that 19.5 hours fasting increased IP that could be evaluated by LMR sugar and FITC-d methods. However, DSS ingestion did not show any IP increase in chickens. Since fasting is routinely applied in the chicken meat industry, fasting and its effects on IP were further investigated (Chapter 5). This study revealed that fasting for as little as 4.5 and 9 hours also increased IP in chickens. The study also confirmed that both methods (LMR sugars and FITC-d) could be utilized to evaluate increased IP. Chapter 6 focused on delayed feeding at the time of chicken placement at the farm. However, this study revealed that delayed feeding did not alter IP in very young chickens. Finally, as short-term fasting also increased IP in older chickens, a further study was conducted to investigate whether mRNA expression of gut barrier associated genes was altered (Chapter 7). This study showed that mRNA expression of claudin-3 only was significantly different from control for 9 and 19.5 hours fasting. This thesis has identified several biomarkers that could be utilized to evaluate increased IP in chickens. Some of these biomarkers such as fecal antitrypsin inhibitor, fatty acid binding protein, diamine oxidase and d-lactate could not detect IP changes induced by fasting in the current studies, they could potentially eliminate the need for oral gavage which is required in the FITC-d and LMR sugar tests. Nonetheless, this research project has shown that permeation of LMR sugars and FITC-d across the intestinal epithelium was able to demonstrate IP changes in chickens.

This study investigate the influence of the fitness of functional strategy with new product performance based on the concept of dynamic industry typology of thecnology trajectory taking industries of food, machinery, automobile and electronics as research object. The conclusions are as follow: 1.By cluster analysis, we divide the R&D strategy into offensive R&D and defensive R&D, they are the leader of technology able to evaluate the technology competences of their competitors, and to predict formal technology, they are usually aggressive, challenging, acquiring opportunity, so there are high performances resulted. On the other side, opposite effects are founded on factories with defensive R&D. 2.We divide manufacturing strategy into strategy of flexible-respone and strategy of stable-low cost. Factories with flexible respone strategy are able to proactive to extenal changes, closed to their customer, high volume buffer and internally reactive to higher rescheduling, rerouting, closed job relvance, high quality authorization, high process change. They emphasize quickly external response, customer service, and more flexible operation, while stable-low cost strtegy resulted oppositely. These two groups have no significant difference on performance, it seems we need to include the influence of other strategy fitness for the explanation. 3.On the total performance of strategy fitness, the best is the fiting of offensive R&D with flexible-respone strategy, the worst is the fiting of defensive R&D with flexible- respone strategy. This is due to to the reason that R&D investment and involvement are the critical factor of new product performance. 4.For the relationship of technology enviroments, strategy fitnesss and new product performance.Factories under technology enviroments of low uncertainty have high performance than that under enviroments of high uncertainty. For interaction effects, there are no interactive effects among these four strategy types.

AbstractIn this chapter, we provide the main elements for implementing deep neural networks in Keras for binary, categorical, and mixed outcomes under feedforward networks as well as the main practical issues involved in implementing deep learning models with binary response variables. The same practical issues are provided for implementing deep neural networks with categorical and count traits under a univariate framework. We follow with a detailed assessment of information for implementing multivariate deep learning models for continuous, binary, categorical, count, and mixed outcomes. In all the examples given, the data came from plant breeding experiments including genomic data. The training process for binary, ordinal, count, and multivariate outcomes is similar to fitting DNN models with univariate continuous outcomes, since once we have the data to be trained, we need to (a) define the DNN model in Keras, (b) configure and compile the model, (c) fit the model, and finally, (d) evaluate the prediction performance in the testing set. In the next section, we provide illustrative examples of training DNN for binary outcomes in Keras R (Chollet and Allaire, Deep learning with R. Manning Publications, Manning Early Access Program (MEA), 2017; Allaire and Chollet, Keras: R interface to Keras’, 2019).

AbstractIn recent years, numerous landslides catastrophes have occurred, generating considerable financial losses and other tolls. The deformational and mechanical properties of sliding zone soil would be of primary significance to landslide research, as the sliding zone basically controls the initiation and mobility of the landslide. An in situ triaxial test was carried out on a sample from the sliding zone of the Riverside Slump 1# landslide, a subdivision of the Huangtupo landslide in the Three Gorges area of China. The test results indicate that a. the sliding zone exhibits low compressibility due to the high rock content (54.3%) and long-time consolidation by the overlying soil mass; b. only decaying creep occurs without abrupt failure, and a constitutive equation with both linear and nonlinear viscoplastic terms is deduced to accurately fit the test data; c. the surface with an orientation of 35° presents anisotropic traits in terms of displacement, possibly due to cracks that formed at similar orientations within the sample cube; and d. the creep behavior of the landslide may be closely related to the properties of the sliding zone soil. When a similar stress magnitude to that of the in situ stress environment is applied to the sample, the sliding zone soil behavior matches the landslide behavior. The test results indicate that the Huangtupo 1# landslide will continue to creep, as interpreted from the deformation and properties of the sample. However, unavoidable limitations of the test and extreme external factors, namely, unexpected rainfall and water fluctuation, cannot be ignored.

R&D cooperation and production cooperation are regarded as two key dimensions of collaborative arrangements in the innovation and production system. Different from prior studies focusing on performance outcomes, this study emphasizes the antecedents which have impacts on firms' decisions of R&D cooperation and production cooperation. The antecedents are identified and categorized into organizational characteristics (market orientation and technological capability), technical characteristics (technology clockspeed and technology-production fit), and relational characteristics (asset specificity). Through statistical analyses on survey data of Chinese manufacturing firms, this study finds that two factors including technology clockspeed and asset specificity have significant effects on firms' decisions of R&D cooperation, while technological capability, technology clockspeed, and technology-production fit are confirmed to have significant effects on decisions of production cooperation.

Chapter 3 investigates more precisely where in the Enquiry Kant finds Hume showing that we cannot know causal connections via pure concepts, and thus that we cannot know the principle of sufficient reason. Hume’s rejection of the principle of sufficient reason comes to a head at 4.13, and Hume returns to it at 12.29 note (d). 12.29 note (d) is directed not, as Hume pretends, against Lucretius’s principle Ex nihilo, nihil fit, but against the causal principle that Descartes, Locke, and Clarke had used to prove the existence of God. This is confirmed by reading 12.29 note (d) against the background of Bayle’s “Spinoza.” Descartes’s Ex nihilo, nihil fit is equivalent, for our purposes, to the principle of sufficient reason found in Wolff, Baumgarten, and the early Kant. The chapter addresses Michael Della Rocca’s argument that Hume failed in his attack on the principle of sufficient reason.

In response to uncertainty of prospective technologies and how they might fit market demand, firms tend to establish R&D alliances. In this chapter the effect over time of continuation of underperforming R&D alliances on innovation performance during the pre-market stage is investigated. This stage is characterized by non-linearity, as expected outcomes and market demands are uncertain. Literature suggests that computational modeling in particular agent-based modeling can be used to investigate such non-linear processes. Agent based modeling starts with simple behavioral rules that develop into emergent system-level behaviors, and in that way controlled system level experiments are used to identify in an inductive way causal mechanisms that drive the system development. In this chapter's simulation model, an agent decides to continue its R&D alliance based on its strategic and cooperation objectives. After evaluating if the strategic goals is met, firms can decide about the extent to which to continue the R&D alliances if the strategic goal is not met. This is called persistency. The model is aimed to explain developmental paths and patterns of the co-evolution of alliances and technology. Despite suggestions to investigate non-linear processes in the pre-market phase by using an agent-based model, agent-based models so far do not focus on the impact of alliance continuation on innovation performance over the path of technology development. In previous research these paths mainly have been investigated in case and cross sectional studies but not in an agent based model. A base-line model is developed and the extent to which it reflects reality is analyzed in order to improve the model's performance.

Fibrin gel matrix organizes solid tumors into discrete cell nests and providesa provisional matrix for mature stroma generation. Fibrin deposits result fromlocal extravasation of plasma fibrinogen followed by extravascular coagulationand crosslinking. Unlike solid tumors, ascites tumor cells in body cavities grow in suspension, and are not envelopedin fibrin gel. The lack of fibrin gel in ascites tumors has several possible explanations: 1. Peritoneal wall blood vessels are impermeable to molecules as large as fibrinogen. 2. Fibrinogen leaks from peritoneal vessels, but is clotted to fibrin and never reaches the peritoneal cavity. 3. Fibrinogen leaks from vessels, reaches the peritoneal cavity, and is rapidly degraded. To distinguish among these possibilities we investigated the permselective properties of the peritoneal wall. We measured the influx of intravenously (iv) administered, fluorescein-labeled dextrans (FITC-D) of varying size into the peritonea of normal and tumor or inflammatory ascites-bearing mice and guinea pigs. FITC-D of MW 70-5,000 kD leaked from vessels and ∽ 10-fold more entered MOT or TA3/St tumor-associated or serotonin-induced inflammatory ascites than normal peritonea. In tumor bearing animals, 3-20% of injected FITC-D entered the peritoneum within 15-30 min., all of it intact by gel exclusion chromatography. We also investigated the influx of fluorescein-labeled fibrinogen (FITC-F). At 30-60 min after iv injection, FITC-F hadextravasated into the peritoneal wall as judged by fluorescence microscopy (tumor bearing ≫ normal animals). Moreover, monoclonal antibodies reactivewith fibrin, but not fibrinogen, stained such deposits by immunohistochemistry in guinea pigs bearing line 1 or line 10 ascites tumors. Following iv injection of 125l-fibrinogen, as much as 11% of injected radioactivity accumulated in tumor ascites fluid over1-3 hrs (versus ∽1% in normal controls). In both, only ∽half was clottableand 3040% was of low MW (≤.10 kD). In contrast, iv 125I-albumin also entered the peritoneal cavity at an enhanced rate but showed no evidence of degradation. We conclude that fibrinogen extravasates from peritoneal vessels,thatleakage is increased in ascites tumor-bearing animals, and that fibrinogen clots, at least in part, before it enters the peritoneal cavity. Also, fibrinogen undergoes selective degradation and much of the fibrinogen that does enter the peritoneum represents degradation products of fibrinogen/fibrin. The resulting lack of peritoneal fibrin gel matrixfavors the suspension pattern of ascites, as compared with solid tumor growth.

Platelets may become activated in a number of clinical disorders and participate in thrombus formation. We have developed a direct test for activated platelets in whole blood that utilizes dual-color flow cytometry and requires no washing steps. Platelets were distinguished from erythrocytes and white blood cells in the flow cytometer by labeling the platelets with biotin-AP1, an antibody specific for membrane glycoprotein lb, and analyzing the cells for phycoerythrin-streptavidin fluorescence. Membrane surface changes resulting from platelet activation were detected with three different FITC-labeled monoclonal antibodies: 1) PAC1, an antibody specific for the fibrinogen receptor on activated platelets; 2) 9F9, which binds to the D-domain of fibrinogen and detects platelet-bound fibrinogen; and 3) S12, which binds to an alpha-granule membrane protein that associates with the platelet surface during secretion. Unstimulated platelets demonstrated no PAC1, 9F9, or S12-specific fluorescence, indicating that they did not bind these antibodies. Upon stimulation with agonists, however, the platelets demonstrated a dose-dependent increase in FITC-fluorescence. The binding of 9F9 to activated platelets required fibrinogen. Low concentrations of ADP and epinephrine, which induce fibrinogen receptors but little secretion, stimulated near-maximal PAC1 or 9F9 binding but little S12 binding. On the other hand, a concentration of phorbol myristate acetate that evokes full platelet aggregation and secretion induced maximal binding of all three antibodies. When blood samples containing activated and non-activated platelets were mixed, as few as 0.8% activated platelets could be detected by this technique. There was a direct correlation between ADP-induced FITC-PAC1 binding and binding determined in a conventional 125I-PAC1 binding assay (r = 0.99; p < 0.001). These studies demonstrate that activated platelets can be reliably detected in whole blood using activation-dependent monoclonal antibodies and flow cytometry. This method may be useful to assess the degree of platelet activation and the efficacy platelet inhibitor therapy in thrombotic disorders.

This paper is the second of two in which a methodology is presented that provides a structured way of solving Dimensioning and Tolerancing (D&T) problems on new product design. In the previous paper, a methodology was developed for representing the D&T problems of a product in a matrix form known as a Dimensional Requirements/Dimensions (DR/D) matrix. In this paper a comprehensive strategy is presented for determining the values of dimensions and tolerances by satisfying all the relationships represented in DR/D matrix. The comprehensive strategy presented here includes: strategy for separating D&T problems into groups, for determining an optimum solution order for coupled functional equations and tolerance allocation strategies for solving different types of D&T problems. A number of commonly used cost minimization strategies such as the use of standard parts, preferred sizes, preferred fits, and preferred tolerances have also been incorporated into the proposed methodology. The methodology is interactive and suitable for use in a Concurrent Engineering environment.

Abstract Proliferation of plastic in parts, and the ability to mold such parts of great complexity at little cost penalty, has resulted in growing use of integral attachments (or so called snap-fits) in designs. The diversity of integral attachment features and the potential applications of this assembly method have made it appear that design possibilities may be unbounded. This paper extends an “attachment level” methodology first articulated by researchers at General Motors by enumerating all possible design options for 2-D (rectilinear) and 3-D (prismatic) polyhedral as well as 2-D and 3-D curvilinear parts using a recently developed hierarchical classification scheme. It was found that there are only 343 possible interface combinations that will allow attachment, of which only 150 are totally unique if one recognizes the interchangeability of attachment features between mating components comprising the assembly. Thus, for a seemingly infinite or unbounded group of possible real integral attachment applications, the choices are, in fact, limited to a quite manageable number. This allows designers to truly optimize a design by considering literally every possibility after working down through the classification hierarchy.

Abstract Cylindrical surface and its tolerance verification play important role in machining process. Although there exists many approaches that can fit the maximum, minimum and minimum zone cylinders, the cylinder fitting problems can even be further simplified. The methodology developed in this paper seeks to reduce the number of parameters used in cylinder fitting model using the projection model. The model evaluates 3-D tolerance specifications by introducing the optimal direction of projection such that the 2-D point projected onto this direction has optimal tolerance specifications (maximum, minimum and minimum zone circles). Besides, a global optimum solver, Particle Swarm Optimization (PSO) is applied to avoid the problem of finding local optimum. The proposed simplified method shows consistent results when compared with the results from literature.

An experimental study has been carried out to measure the pressure drop in a 271-pin fuel assembly of a liquid metal reactor. The rod pitch to rod diameter ratio (P/D) of the fuel assembly is 1.2 and the wire lead length to rod diameter ratio (H/D) is 24.84. Measurements are made for five different sections in a fuel assembly; inlet orifice, fuel assembly inlet, wire-wrapped fuel assembly, fuel assembly outlet and fuel assembly upper region. A series of water experiments have been conducted changing flow rate and water temperature. It is shown that the pressure drops in the inlet orifice and in the wire-wrapped fuel assembly are much larger than those in other regions. The measured pressure drop data in a wire-wrapped fuel assembly region is compared with the existing four correlations. It is shown that the correlation proposed by Cheng and Todreas fits the best with the present experimental data among the four correlations considered.

Abstract The intent of this paper is to give the background and details of a project using the concepts of concurrent engineering and design for manufacturing in an educational framework. The project involves the design and production of parts that have simple 2-d shapes with slots. These can be fit together to create a variety of 3-d shapes and figures. These are like children’s toy “puzzle” pieces (building blocks) that can be disassembled and reassembled into new figures over and over. The parts have been produced using the machined mold on a small injection molding machine. This paper discusses the execution of the project from the concept to the design to the production stages. Design for Manufacturing education will be highlighted through this interesting example project.
