Bovine tuberculosis (bTB) is diagnosed in naturally infected populations exposed to a wide variety of other pathogens. This study describes the cell-mediated immune responses of cattle exposed toMycobacterium aviumsubspeciesparatuberculosis(Map) andMycobacterium aviumsubspeciesaviumwith particular reference to routine antefmortemMycobacterium bovisdiagnostic tests. The IFN-γreleased in response to stimulated blood was found to peak later in the Map-exposed group and was more sustained when compared to the Maa-exposed group. There was a very close correlation between the responses to the purified protein derivatives (PPD) used for stimulation (PPDa, PPDb, and PPDj) with PPDa and PPDj most closely correlated. On occasion, in the Map-infected cattle, PPDb-biased responses were seen compared to PPDa suggesting that some Map-infected cattle could be misclassified asM. bovisinfected using this test with these reagents. This bias was not seen when PPDj was used. SICCT results were consistent with the respective infections and all calves would have been classed skin test negative.

Objective: To study the cardiovascular effect over 30 minutes following the end of fluid bolus therapy (FBT) with 20% albumin in patients after cardiac surgery. Design: Prospective observational study. Setting: Intensive care unit of a tertiary university-affiliated hospital. Participants: Twenty post-cardiac surgery mechanically ventilated patients with a clinical decision to administer FBT. Intervention: FBT with a 100 mL bolus of 20% albumin. Main outcome measures: Cardiac index (CI) response was defined by a  15% increase, while mean arterial pressure (MAP) response was defined by a  10% increase. Results: The most common indication for FBT was hypotension (40%). Median duration of infusion was 7 minutes (interquartile range [IQR], 3–9 min). At the end of FBT, five patients (25%) showed a CI response, which increased to almost half in the following 30 minutes and dissipated in one patient. MAP response occurred in 11 patients (55%) and dissipated in five patients (45%) by a median of 6 minutes (IQR, 6–10 min). CI and MAP responses coexisted in four patients (20%). An intrabolus MAP response occurred in 17 patients (85%) but dissipated in 11 patients (65%) within a median of 7 minutes (IQR, 2–11 min). On regression analysis, faster fluid bolus administration predicted MAP increase at the end of the bolus. Conclusion: In post-cardiac surgery patients, CI response to 20% albumin FBT was not congruous with MAP response over 30 minutes. Although hypotension was the main indication for FBT and a MAP response occurred in most of patients, such response was maximal during the bolus, dissipated in a few minutes, and was dissociated from the CI response.

Mantı, the traditional Turkish food, was subjected to modified atmosphere packaging (MAP) compositions of MAP 1 (80% CO2 + 20% N2), MAP 2 (40% CO2 + 60% N2), MAP 3 (60% CO2 + 40% N2) and control (packaged under atmospheric composition) to extend its refrigerated storage at 4°C. The physical, chemical and sensorial qualities of each package were assessed by analysing headspace gas composition, pH, water activity, 2-thiobarbituric acid (TBA), dry matter, lipid content and a sensory analysis of both cooked and raw mantı samples. The compositions of MAP samples (MAP 1, MAP 2 and MAP 3) resulted in the maximum storage time of 126 days versus 20 days in normal atmospheric packaging (control). In conclusion, 60% CO2 or either 80% CO2 with N2, as a make-up gas, should be implemented in the mantı process.

Certain nonsteroidal anti-inflammatory drugs have been reported to elevate blood pressure in some hypertensive patients, who are either untreated or treated with antihypertensive agents. This study was undertaken to determine the effect of a selective cyclooxygenase-2 (COX-2) inhibitor, celecoxib, on the antihypertensive effects of the angiotensin II type 1 receptor (AT1) antagonist, losartan potassium. We studied the effect of oral treatment with losartan (30 mg/kg), celecoxib (3 mg/kg), and their combination on the mean arterial blood pressure (MAP), plasma renin activity (PRA), and plasma prostaglandin E2(PGE2) in male Sprague–Dawley rats with renovascular hypertension (RVH) induced by partial subdiaphragmatic aortic constriction. Treatment was continued for 7 days after aortic coarctation. Aortic coarctation led to significant increases in the MAP, PRA, and plasma PGE2. In RVH rats, losartan treatment caused a significant decrease of MAP with a significant increase in both plasma PGE2and PRA. Celecoxib caused a nonsignificant change in MAP with a significant decrease in the raised levels of plasma PGE2and PRA. Concomitant administration of celecoxib and losartan did not significantly affect the lowering effect of losartan on MAP with a subsequent significant decrease in the plasma PGE2and PRA in RVH rats. Therefore, celecoxib could be used in renin-dependent hypertensive patients who receive losartan, without fear of a rise in their blood pressure.

The purpose of this study was to examine the effect of Mindful Listening Instruction on Music Listening Sensitivity and Music Listening Enjoyment. The type of mindfulness investigated in this study was of the social-psychological type, which shares both commonalities with and distinctions from meditative mindfulness. Enhanced context awareness, openness to new information, situation in the present, awareness of novel distinctions, and awareness of multiple possible perspectives (cognitive flexibility) are components of social-psychological mindfulness. A pretest-posttest control group design was used for this study. Two different age groups of students were studied: fourth-grade students (N = 42) and undergraduate non-music major college students (N = 48). The fourth-grade participants in this study were selected from an elementary school in a large city in the Northeastern United States. The college students were selected from a large university in the Southeastern United States. Participants were randomized into either the experimental or control group. Gordon’s Intermediate Measures of Music Audiation and Advanced Measures of Music Audiation were used as a pretest for fourth-grade students and college students, respectively. The results showed no statistically significant differences between the experimental and control groups. Student demographical information was also collected and reported. The treatment consisted of 10 lessons for fourth-grade students. Five of the 10 lessons were used with the college students. For each age level, participants in both groups, Mindful Listening and Control, received instruction using listening-map-based and non-listening-map-based lessons from the Share the Music textbook series. Students in the Mindful Listening groups also received listening instructions designed to promote mindful listening. Music Listening Sensitivity was measured using the phrasing test from the Sensitivity portion of Gordon’s Music Aptitude Profile (MAP-P), as well as the researcher-created Anderson Test of Music Listening Sensitivity (ATMLS). Music Listening Enjoyment was measured using students’ ratings of their Listening Enjoyment after each lesson on a seven-point Likert-type scale. Results indicated that Mindful Listening Instruction yielded higher scores, which were statistically significant (at α = .05), for Music Listening Sensitivity (as measured by both the ATMLS and the MAP-P) and Music Listening Enjoyment for fourth-grade and college-student participants.

With modern culture moving towards convenience in terms of fresh produce, especially with meat products, science needs to constantly evolve to serve these customer needs. These needs however can sometimes be implemented too hastily without the proper assessment done for factors such as food safety. One such improvement is the use of modified atmosphere packaged minced meat. This form allows the minced meat to be kept for much longer than normal without freezing the product, providing the fast pasted consumer both the convenience of having fresh, unfrozen meat as well as an added shelf life. MAP works by disrupting the atmosphere within these packages, retarding the growth of the spoilage micro-organisms, thus causing them to require a longer time span to reach spoilage numbers. The problem however arises that although most of these techniques are tried and trusted on the products’ spoilage organisms, it does not take into consideration the effect MAP and the altered spoilage organism communities would have on a pathogen that might be present on the products. This study thus aimed to assess the effect of both factors on Escherichia coli O157:H7 (phase 1) and Staphylococcus aureus (phase 2) in minced meat kept at 5°C as well as trying to identify the major affecting factors. The two subject organisms were each inoculated into 2 different types of MAP packs and a non-atmosphere modified PVC overlaid minced meat sample at two differing concentrations of 105 and 102 to also assess the impact of high andlow initial pathogen presence. These packs were then analysed over a time period of 16 days to track the changing minced meat environment. APC, Pseudomonads, LAB and Enterobacteriaceae counts were all investigated along with the pathogenic counts. Apart from colony enumeration, the colour of the minced meat samples were also taken to determine the effect that these parameters have on the appearance of the product, as colour is often the first sensorial characteristic that determines the purchase of fresh meat products. pH was determined to ascertain the environmental changes occurring in the product and whether groups such as the LAB would change the environment to better suit their needs. Finally the atmospheric makeup was also measured to determine the effect of the MAP system and the change occurring in a closed system that could be attributed to the growth and respiration of the bacterial communities present. Apart from the main aim of the study, two additional studies were performed that arose during the planning and analyses of the two primary phases. Firstly the use of a quarter versus a full plate enumeration was studied to determine its accuracy as well as possibility of use in full studies to aid enumeration and decrease time and financial input. Here a direct comparison was done between the two techniques after which they were compared and assessed in their functionality for both homogenous and heterogeneous community enumeration on selective and non-selective media. The other secondary study focussed on the use of new technology for both the enumeration and tracking of genetically modified organisms in a variety of different environments. Here a bioluminescent imaging system was used on a genetically modified strain of E. coli to track its spread through minced meat, packaged either in a MAP or PVC overlaid pack, over 48 hours in an accelerated shelf life study. Enumeration of said organism was also undertaken whereby the intensity of emitted light would correspond to a defined count, enabling rapid enumeration of samples, whether overgrown or not. Copyright

This master's thesis focuses on the dynamic presentation of still photography using a depth map. This text presents an algorithm that describes the process of creating a spatial model which is used to render input photography so that the movement of virtual camera creates parallax effect due to depth in image. The thesis also presents an approach how to infill the missing data in the model. It is suggested that a guided texture synthesis is used for this problem by using rendering outputs of the model themselves as guides. Additional information in model allows the virtual camera to move more freely. The final result of the camera movement can be saved to simple video sequence which can be used for presenting the input photography.

iv In recent years, outbreaks of infections associated with raw and minimally processed fruits and vegetables have been reported. Possible sources for contamination are irrigation water, manure, wash water, handling by workers and contact with contaminated surfaces. Pathogens can occur on raw and minimally processed produce at populations ranging from 103 to 109 CFU/g and able to survive and sometimes grow under various storage conditions. The objective of this study was to analyse the growth/survival of Salmonella Enteritidis at spot-inoculated or stem-injected cherry tomatoes during passive modified atmosphere packaging (MAP), controlled atmosphere (CA) and air storage at 7 and 22°
C. Low density polyethylene (LDPE) with a package size of 10x10 cm2 for 25±
2 g tomatoes was used for MAP storage in which the gas composition equilibrated to 6% O2/ 4% CO2 and a carbon dioxide incubator was used for CA storage in which the CO2 level was monitored and maintained as 5% through the term of storage at 7 and 22°
C. During the research, the effect of ozone treatment (5-30 mg/L ozone gas for 0-20 min) was also considered for surface sanitation. The results demonstrate that S.Enteritidis can survive and/or grow during the storage of tomatoes depending on the location site of the pathogen on fruit, suspension cell density and storage temperature. During MAP, CA and air storage, S.Enteritidis with initial population of 7.0 log10 CFU/tomato survived on tomato surfaces with an approximate decrease of 4.0-5.0 log10 CFU/tomato in population within the storage period
however, in the case of initial population of 3.0 log10 CFU/tomato, cells died completely on day 4 during MAP storage and on day 6 during CA and air storage. The death rate of S.Enteritidis on the surfaces of tomatoes that were stored in MAP was faster than that of stored in air. Storage temperature was effective on the survival of S.Enteritidis for the samples stored at ambient atmosphere
cells died completely on day 6 at 7°
C and on day 8 at 22°
C. Stem scars provided protective environments for Salmonella
an approximate increase of 1.0 log10 CFU/tomato in stem-scar population was observed during MAP, CA and air storage at 22°
C within the period of 20 days. Cells survived with no significant change in number at 7°
C. The development of the microbial association in tomatoes was dominated by lactic acid bacteria (LAB). The pH values of the tomatoes changed approximately from 4.0 to 3.0 during the storage period. LAB grew well under all atmospheric conditions with or without the presence of S.Enteritidis. Gaseous ozone treatment has bactericidal effect on S.Enteritidis, inoculated on the surface of the tomatoes. 5 mg/L ozone gas treatment was not effective. 30 mg/L ozone gas treatment affected surface color.

When provided with spatial niches by growth in static nutrient medium, Pseudomonas fluorescens diversifies through adaptive radiation into several well-defined phenotype classes. One of these classes, named wrinkly spreader (WS) for its morphology on agar medium, forms a biofilm at the air-liquid interface through mutations in one of several loci including the genes wspF and awsX. These genes code for negative regulators of di-guanylate cyclases (DGCs). These DGCs catalyse synthesis of cyclic-di-GMP, a second messenger, overproduction of which effects physiological changes leading to overproduction of a cellulose polymer and the WS phenotype. Intriguingly, a diverse range of wspF mutations leads to diversity both in colony morphology and strain fitness.In this study, I investigate genetic and fitness diversity in wrinkly spreaders with the aim of identifying the causal factors that link genetic diversity and physiological factors with diversity in fitness. I approach the subject from several directions, examining the historical context of genetic diversity in wspF and awsX, distribution of control over output in the Wsp pathway and overall fitness effects of different causal factors. I investigate the genetic basis of wrinkly spreader evolution through generation of a large number of novel WS strains and exploration of the distribution of mutations in the wspF and awsX genes. In combination with this I calculate estimates of the past rates of mutation in these genes, derived from a phylogenetic investigation of a group of orthologues. I examine the response of the Wsp pathway to change in WspF function through a novel computational analysis that is capable of revealing valuable information on control in a biological system based purely on model structure. In addition I show how this analysis can be refined through specification of broad estimates of system parameters, thereby avoiding issues related to over-reliance on specific parameter values. Finally, I investigate the fitness implications of these factors, as well as a variety of others, through assays of fitness in a group of WS strains combined with machine learning analyses of predictive relationships between protein and mutation characteristics and experimentally measured strain fitness, and consider the implications of this analysis in the context of intermediate physiological effects.I find that mutations in the WspF protein that lead to the WS phenotype tend to be located in regions of historically strong conservation, the first time that any such pattern to WS mutations has been identified. Mutations in AwsX, on the other hand, do not fit such a pattern. Computational analysis of the Wsp pathway shows that, regardless of model parameters, pathway output is always more sensitive to changes in methylesterase activity by WspF than to changes in phosphorylation of WspF, which may explain the greater frequency of mutations fixed in vivo seen in the methylesterase domain. Despite these patterns, none of a wide range of mutation and sequence-based biochemical characteristics, including local rates of past evolution and size and position of mutations, exhibited any predictive power over WS fitness. Overall, the findings in this study point towards an essential role for complex pleiotropic effects in strongly modulating the fitness effect of different mutations in wspF.

High-precision optical systems are generally tested using interferometry, since it often is the only way to achieve the desired measurement precision and accuracy. Interferometers can generally measure a surface to an accuracy of one hundredth of a wave. In order to achieve an accuracy to the next order of magnitude, one thousandth of a wave, each error source in the measurement must be characterized and calibrated.Errors in interferometric measurements are classified into random errors and systematic errors. An approach to estimate random errors in the measurement is provided, based on the variation in the data. Systematic errors, such as retrace error, imaging distortion, and error due to diffraction effects, are also studied in this dissertation. Methods to estimate the first order geometric error and errors due to diffraction effects are presented.Interferometer phase modulation transfer function (MTF) is another intrinsic error. The phase MTF of an infrared interferometer is measured with a phase Siemens star, and a Wiener filter is designed to recover the middle spatial frequency information.Map registration is required when there are two maps tested in different systems and one of these two maps needs to be subtracted from the other. Incorrect mapping causes wavefront errors. A smoothing filter method is presented which can reduce the sensitivity to registration error and improve the overall measurement accuracy.Interferometric optical testing with computer-generated holograms (CGH) is widely used for measuring aspheric surfaces. The accuracy of the drawn pattern on a hologram decides the accuracy of the measurement. Uncertainties in the CGH manufacturing process introduce errors in holograms and then the generated wavefront. An optimal design of the CGH is provided which can reduce the sensitivity to fabrication errors and give good diffraction efficiency for both chrome-on-glass and phase etched CGHs.

Porphyrins consist of twenty-atom rings containing four nitrogen atoms and can be used as sensor to detect odours and gases.

This thesis investigates whether or not porphyrins can be used as functional materials on grid gate devices.  Drops of PVC embedded porphyrins were deposited on the surface of a grid gate which is a Metal Oxide Semiconductor (MOS) capacitor. In order to detect the gas sensing properties of the porphyrins a light addressable method called Scanning Light Pulse Technique (SLPT) has been used.

A central challenge facing firms is how to incentivize employees. While objective, output-based incentives may be theoretically ideal, in practice they may lead employees to reduce effort on non-incentivized outcomes and may fail in settings where effort is weakly tied to output. We study the effect of subjective incentives (manager performance evaluation) and objective incentives (test score-based) relative to no incentives for teachers using an RCT in 230 Pakistani schools. First, we show that subjective and objective incentives both increase test scores and have similar magnitude effects. However, objective incentives decrease non-test score student outcomes relative to subjective incentives. Second, we show that teachers’ effort response is very different under each scheme, with attendance increasing under subjective and teaching quality decreasing under objective. Finally, we rationalize these effects through the lens of a moral hazard model with multi-tasking. We use within-treatment variation to isolate the causal effect of contract noise and distortion and show that these channels explain most of our reduced form effects.

The plant hormone auxin is involved in numerous developmental processes, including leaf and fruit development. The tomato (Solanumlycopersicum) gene ENTIRE (E) encodes an auxin-response inhibitor from the Aux/IAA family. While most loss-offunction mutations in Aux/IAA genes are similar to the wild type due to genetic redundancy, entire (e) mutants show specific effects on leaf and fruit development. e mutants have simple leaves, in contrast to the compound leaves of wild type tomatoes. In addition, e plants produce parthenocarpic fruits, in which fruit set occurs independently of fertilization. The aim of this research program was to utilize the e mutation to identify and characterize genes that mediate the specific effect of auxin in leaf and fruit development. The specific objectives of the project were to: 1. Characterize and map modifiers of the e leaf phenotype. 2. Characterize and map suppressors of the e fruit phenotype. 3. Dissect the developmental specificity of the E gene. 4. Examine the effect of fruit-overexpression of identified genes on fruit set and seed production. To identify mediators of auxin in leaf development, we mainly focused on one mutant, crawling elephant (crel, previously called t282), which showed substantial suppression of the e phenotype and other auxin-relatedphenotypes. We have identified the CREL gene as a homolog of the Arabidopsis VRN5 gene, involved in recruiting polycomb silencing complexes to specific targets. We showed that CREL affects auxin sensitivity in tomato. Suppressors of the e fruit phenotype have been further characterized and selected for more profound effects. Expression profiling by RNAseq was used to analyze the effect of e as well as crel on gene expression in leaves and fruits. This analysis has identified putative E and CREL targets. We have initiated studies to assess the role of some of these targets in flower and fruit development. The research has identified potential mediators of auxin response in leaf, flower and fruit development.

The general goal was to identify, map, and tag, with DNA markers, segments of chromosomes of a wild species (wild emmer wheat, the progenitor of cultivated wheat) determining the number, chromosomal locations, interactions, and effects of genes that control quantitative traits when transferred to a cultivated plant (bread wheat). Slight modifications were introduced and not all objectives could be completed within the human and financial resources available, as noted with the specific objectives listed below: 1. To identify the genetic contribution of each of the available wild emmer chromosome-arm substitution lines (CASLs) in the bread wheat cultivar Bethlehem for quantitative traits, including grain yield and its components and grain protein concentration and yield, and the effect of major loci affecting the quality of end-use products. [The quality of end-use products was not analyzed.] 2. To determine the extent and nature of genetic interactions (epistatic effects) between and within homoeologous groups 1 and 7 for the chromosome arms carrying "wild" and "cultivated" alleles as expressed in grain and protein yields and other quantitative traits. [Two experiments were successful, grain protein concentration could not be measured; data are partially analyzed.] 3. To derive recombinant substitution lines (RSLs) for the chromosome arms of homoeologous groups 1 and 7 that were found previously to promote grain and protein yields of cultivated wheat. [The selection of groups 1 and 7 tons based on grain yield in pot experiments. After project began, it was decided also to derive RSLs for the available arms of homoeologous group 4 (4AS and 4BL), based on the apparent importance of chromosome group 4, based on early field trials of the CASLs.] 4. To characterize the RSLs for quantitative traits as in objective 1 and map and tag chromosome segments producing significant effects (quantitative trait loci, QTLs by RFLP markers. [Producing a large population of RSLs for each chromosome arm and mapping them proved more difficult than anticipated, low numbers of RSLs were obtained for two of the chromosome arms.] 5. To construct recombination genetic maps of chromosomes of homoeologous groups 1 and 7 and to compare them to existing maps of wheat and other cereals [Genetic maps are not complete for homoeologous groups 4 and 7.] The rationale for this project is that wild species have characteristics that would be valuable if transferred to a crop plant. We demonstrated the sequence of chromosome manipulations and genetic tests needed to confirm this potential value and enhance transfer. This research has shown that a wild tetraploid species harbors genetic variability for quantitative traits that is interactive and not simply additive when introduced into a common genetic background. Chromosomal segments from several chromosome arms improve yield and protein in wheat but their effect is presumably enhanced when combination of genes from several segments are integrated into a single genotype in order to achieve the benefits of genes from the wild species. The interaction between these genes and those in the recipient species must be accounted for. The results of this study provide a scientific basis for some of the disappointing results that have historically obtained when using wild species as donors for crop improvement and provide a strategy for further successes.

Review question / Objective: We aim to assess the antidepressant effects of statin therapy among patients complicated with moderate to severe depression. Condition being studied: Depression is one of the major causes of disability worldwide, and major depressive disorders (MDD) contribute to a significant heavy disease burden, which is expected to be second by 2050, only to heart disease. Despite great improvement in therapy, the treatment efficacy remains low. Therefore, alternative therapies have been intensely investigated. A substantial body of researches have suggested that inflammation is one of the operative pathways between MDD and increased risk of somatic comorbidities, and some specific depressive symptoms. Depression occurs in most patients with cardiac and cerebrovascular disease due to the long-term effects, and depression increases the risk of cardiovascular disease in the population as a whole and in patients with coronary artery disease or stroke. Several observational studies have demonstrated reduced rates of depression among patients taking statins, which may be related to its anti-inflammatory effect. However, whether statin improves the depressive symptoms and its associated mechanism is still mixed. Furthermore, there is little evidence about statin treatment effect in those with moderate to severe depression. In addition, whether the effect of statin treatment on depressive symptom changes with time or is affected by baseline depression severity or percentage change of lipid levels has not been explored in previous studies.

Parental involvement programs aim to strengthen school-home relations with the goal of improving children’s educational outcomes. We examine the effects of a parental involvement program in Mexico, which provides parent associations with grants and information. We separately estimate the effect of the grants from the effect of the information using data from two randomized controlled trials conducted by the government during the rollout of the program. Grants to parent associations did not improve educational outcomes. Information to parent associations reduced disciplinary actions in schools, mainly by increasing parental involvement in schools and changing parenting behavior at home. The divergent results from grants and information are partly explained by significant changes in perceptions of trust between parents and teachers. Our results suggest that parental involvement interventions may not achieve their intended goal if institutional rules are unclear about the expectations of parents and teachers as parents increase their involvement in schools.
