Lyme disease is the leading tick-borne infection in the United States, caused by the pathogenic spirochete Borreliella burgdorferi, formerly known as Borrelia burgdorferi. Diderms, or bacteria with dual-membrane ultrastructure, such as B. burgdorferi, have multiple methods of transporting and integrating outer membrane proteins (OMPs). Most integral OMPs are transported through the β-barrel assembly machine (BAM) complex. This complex consists of the channel-forming OMP BamA and accessory lipoproteins that interact with the five periplasmic, polypeptide transport-associated (POTRA) domains of BamA. Another system, the translocation and assembly module (TAM) system, has also been implicated in OMP assembly and export. The TAM system consists of two proteins, the BamA paralog TamA which has three POTRA domains and the inner membrane protein TamB. TamB is characterized by a C-terminal DUF490 domain that interacts with the POTRA domains of TamA. Interestingly, while TamB is found in almost all diderms, including B. burgdorferi, TamA is found almost exclusively in Proteobacteria. This strongly suggests a TamA-independent role of TamB in most diderms. We previously demonstrated that BamA interacts with TamB in B. burgdorferi and hypothesized that this is facilitated by the BamA POTRA domains interacting with the TamB DUF490 domain. In this study, we utilized protein-protein co-purification assays to empirically demonstrate that the B. burgdorferi TamB DUF490 domain interacts with BamA POTRA2 and POTRA3. We also observed that the DUF490 domain of TamB interacts with the accessory lipoprotein BamB. To examine if the BamA-TamB interaction is more ubiquitous among diderms, we examined BamA-TamB interactions in Salmonella enterica serovar Typhimurium (St). Interestingly, even though St encodes a TamA protein that interacts with TamB, we observed that the TamB DUF490 of St interacts with BamA in this organism. Our combined findings strongly suggest that the TamB-BamA interaction occurs independent of the TamA component of the TAM protein export system.

Bile salt hydrolases (BSHs) are gut microbial enzymes that play a significant role in the bile acid modification pathway. Penicillin V acylases (PVAs) are enzymes produced by environmental microbes, having a possible role in pathogenesis or scavenging of phenolic compounds in their microbial habitats. The correct annotation of such physiologically and industrially important enzymes is thus vital. The current methods relying solely on sequence homology do not always provide accurate annotations for these two members of the cholylglycine hydrolase (CGH) family as BSH/PVA enzymes. Here, we present an improved method [binding site similarity (BSS)-based scoring system] for the correct annotation of the CGH family members as BSH/PVA enzymes, which along with the phylogenetic information incorporates the substrate specificity as well as the binding site information. The BSS scoring system was developed through the analysis of the binding sites and binding modes of the available BSH/PVA structures with substrates glycocholic acid and penicillin V. The 198 sequences in the dataset were then annotated accurately using BSS scores as BSH/PVA enzymes. The dataset presented contained sequences from Gram-positive bacteria, Gram-negative bacteria and archaea. The clustering obtained for the dataset using the method described above showed a clear distinction in annotation of Gram-positive bacteria and Gram-negative bacteria. Based on this clustering and a detailed analysis of the sequences of the CGH family in the dataset, we could infer that the CGH genes might have evolved in accordance with the hypothesis stating the evolution of diderms and archaea from the monoderms.

Abstract Lyme disease is the most common vector-borne illness in the United States with 400,000 estimated new cases annually. Borrelia burgdorferi (Bb), the causative agent, causes severe clinical manifestations, but lacks classical virulence factors. The primary cause for patient symptomology is unknown. Peptidoglycan (PG) serves as both a protective barrier from osmotic pressure and a pathogen associated molecular pattern (PAMP). In diderms, this structure resides between the outer and inner membrane. Bb is an atypical Gram-negative bacterium. It contains L-ornithine in its PG peptide stem and lacks a PG recycling pathway. The collective results are an atypical pathogen that sheds unusual PG fragments into its extracellular environment. We have recently shown that these PG fragments reside in the human host, well after acute infection in Lyme arthritis patients. Also, when injected into a mouse, Bb PG is capable of causing acute arthritis that mimics that of Lyme arthritis. Here, we probed the pathogenic properties of Bb PG chemistry. Comparative RNA-seq analysis from human PBMCs stimulated with PG fragments isolated from different bacteria demonstrate a surprising and unique immunological response that appears to be dictated by the chemical features unique to Bb PG. We also provide evidence that Bb PG is not alone in eliciting this unique response. Our recent, unpublished studies show that NapA— Neutrophil Attracting Protein A— is physically linked to Bb PG and contributes to this atypical PG response. Using molecular and cellular studies, including a novel microfluidic chamber, we show that the unique PG-NapA linkage augments the production of IL-17 and recruits the frontline of the innate immune system in response towards this unusual PAMP.

Lėtinė obstrukcinė plaučių liga (LOPL) yra dažna sergamumo ir mirtingumo priežastis visame pasaulyje. Patiriami kvėpavimo simptomai, apribota kasdienė veikla ir pomėgiai keičia gyvenimo kokybę. Nevaldomi ligos simptomai, paūmėjimai tarsi sukuria užburtą ratą ligai sparčiau vystytis. Ligos valdymui tenka reikšminga sveikatos priežiūros išlaidų dalis. Todėl būtina savalaikiai optimizuoti LOPL gydymą, valdyti ligos simptomus, retinti paūmėjimus, sulėtinti ligos vystymąsi ir užkirsti kelią komplikacijoms. Pernelyg didelis trumpo veikimo β2 agonistų vartojimas yra susijęs su ligos sunkumu ir simptomais, padidėjusia paūmėjimų rizika ir yra ženklas, kad nuolatinis medikamentinis gydymas turi būti peržiūrimas ir optimizuojamas. Nesant dažnų paūmėjimų, bet esant išreikštiems simptomams, tikslingas gydymas ilgo veikimo muskarino receptorių antagonisto / ilgo veikimo β2 agonisto deriniu. Vienas naujausių LOPL gydymui skiriamų derinių yra aklidinio bromidas ir formoterolio fumaratas, kurio veiksmingumas ir saugumas įrodytas didelės apimties tyrimais. Diderma is one of the most abundant species in Myxomycetes. Based on the study of morphology and phylogeny, two new species—Diderma liaoningensis (is characterized by grouped sporocarps, the sporocyst with dehiscence lines, capillitium with large brown swelling, spore encircled by a pale line) and Diderma verrucocapillitia (is characterized by depressed plasmodiocarps, verrucose capillitium, red-brown spores)—collected from Liaoning province and Jilin province were introduced. Detailed descriptions and illustrations are presented as well as comparisons to similar species. Phylogenetic analyses were carried out using Maximum Likelihood and Bayesian inference based on small subunit ribosomal RNA sequences (SSU rRNA) of Diderma and related genera. These analyses confirmed the placement of two new species in the genus Diderma. A key to 19 species of Diderma from China is also provided. Holotype specimens have been housed in the Herbarium of the Mycological Institute of Jilin Agricultural University (HMJAU), Changchun, China.

Įvadas Nors kolonoskopinių gaubtinės žarnos perforacijų dažnis nėra didelis, tačiau jos gali lemti sunkias komplikacijas ar net mirtį. Šios studijos tikslas buvo įvertinti kolonoskopinių gaubtinės žarnos perforacijų dažnį ir jų chirurginio gydymo rezultatus.Metodai Retrospektyviai buvo peržiūrėti duomenys pacientų, kuriems atliekant diagnostinę ar gydomąją kolonoskopiją Nacionaliniame vėžio institute nuo 2007 m. sausio 1 d. iki 2016 m. gruodžio 31 d. įvyko gaubtinės žarnos perforacijos. Analizuoti pacientų demografiniai rodikliai, kolonoskopijos duomenys, informacija apie jų gydymą ir gydymo rezultatus.Rezultatai Iš viso Nacionaliniame vėžio institute minėtu laikotarpiu buvo atlikta 16 186 kolonoskopijos. Bendras kolonoskopinių gaubtinės žarnos perforacijų dažnis siekė 0,14 % (23 iš 16 186 pacientų). Iš šių 23 perforacijų 20 atvejų buvo taikytas chirurginis gydymas. Daugiausiai perforacijų įvyko riestinėje žarnoje (12 atvejų). 11 atvejų defektas žarnoje buvo užsiūtas, 6 atvejais atlikta žarnos rezekcija su pirmine anastomoze. Viena mirtis įvyko dėl ūmios bronchopneumonijos išsivysčius dauginiam organų disfunkcijos sindromui, kiti du pacientai, kuriems pasireiškė pilvo ertmės sepsis, mirė nuo kardiopulmoninio nepakankamumo.Išvados Jeigu kolonoskopinės gaubtinės žarnos perforacijos atvejais pavyktų išvengti operacijos ir ją lydinčių sunkių komplikacijų, tai leistų reikšmingai sumažinti kolonoskopijos komplikacijos padarinius. Visus pacientus prieš procedūrą būtina įspėti apie galimas komplikacijas, o gydytojai turi būti itin atidūs, jei pacientas priklauso didelės rizikos grupei.

Triburio vožtuvo nesandarumas (TVN) − sąlyginai dažna, progresuojanti ir sunki liga, susijusi su prasta gyvenimo kokybe bei padidėjusiu mirštamumu. Antrinis, arba funkcinis TVN, sudaro iki 90 proc. atvejų. Jis vystosi, kai padidėjus spaudimui ir (ar) atsiradus tūrio perkrovai, dešinysis skilvelis (DS) ima plėstis, sąlygodamas triburio vožtuvo (TV) žiedo dilataciją bei sutrikusią burių koaptaciją. Dažniausiai antrinį TVN sukelia kairiosios širdies vožtuvų ligos, plautinė hipertenzija, ilgalaikis prieširdžių virpėjimas, DS išemija ar kardiomiopatijos. TVN neretai ilgą laiką nesukelia jokių simptomų, o pirmieji požymiai atsiranda dėl kairiosios širdies vožtuvų disfunkcijos ar širdies nepakankamumo. Auksiniu standartu TVN diagnostikoje laikomas echokardiografinis tyrimas. Jo metu išsamiai vertinama vožtuvo morfologija, pažeidimo laipsnis, etiologija, nustatoma kitų širdies struktūrų geometriniai bei funkciniai pokyčiai. Vertinant funkcinio TVN prognozę, echokardiografijos metu svarbu atlikti dešiniosios širdies ertmių dydžių matavimus bei DS funkcijos įvertinimą. Pirmaeilis funkcinio TVN chirurginis gydymas yra anuloplastika − vožtuvo žiedo korekcija, išsauganti nuosavą triburį vožtuvą. Vožtuvo protezavimas turėtų būti svarstomas, kai nustatomas didelis žiedo išsiplėtimas arba reikšmingas TV burių nutempimas į DS ertmę. Vystantis minimaliai invazinių procedūrų metodams, didelės operacinės rizikos pacientams atsiranda vis daugiau naujų vožtuvinių širdies ligų gydymo alternatyvų. Kateterinis funkcinio TVN gydymas turėtų būti svarstomas pasireiškus simptomams, kai pacientams, turintiems tinkamą TV anatomiją, kai negalima taikyti chirurginio gydymo.

Abstract A new species of Diderma (Physarales, Didymiaceae) was recorded in the Mata Estrela Private Nature Reserve, Rio Grande do Norte state. It is characterized by sessile sporangia, white globose columella, capillitium irradiating from the peridium, forming three layers, the middle part strongly calcareous, the outer layer cartilaginous and the inner layer membranous. A key to species of sessile sporangia Diderma with triple peridium is presented.

Two specimens of a rare species Diderma aurantiacum collected in the Sikhote-Alin Nature Reserve and the territory of the Land of the Leopard National Park (Primorye Territory, Russian Federation) were found during revision of the myxomycete collection of the Mycological herbarium of the Komarov Botanical Institute (LE). These specimens are the first records of this species outside the Japanese islands. Diderma aurantiacum is characterized by a thick cartilaginous peridium of dull orange color that breaks into petal-like fragments during dehiscence, a small hemispherical columella, weakly branched dark capillitial threads with pale tips, and warted spores with darker warts arranged in small groups. The color photographs of sporocarp structures and SEM images of spores and capillitium are published for the first time. The partial nucleotide sequences of nrSSU (18S rDNA) and EF1α genes of specimens from the Russian Far East differed clearly from these of other Diderma species.

L'enveloppe bactérienne est essentielle à de nombreuses fonctions cellulaires, comme le maintien de l'intégrité structurelle, la division cellulaire, la motilité, l'absorption des nutriments et la communication. Elle joue également un rôle crucial dans la pathogénicité bactérienne, faisant de son assemblage et de son maintien un domaine de recherche clé pour le développement de nouveaux antimicrobiens. Les enveloppes bactériennes présentent une dichotomie unique entre les structures monodermes (membrane unique) et didermes (double membrane). Traditionnellement, cette distinction correspond aux bactéries à Gram-positif (monodermes) et à Gram-négatif (didermes). On considérait initialement que les bactéries didermes avaient évolué à partir des monodermes par complexification progressive. Cependant, des études phylogénomiques récentes montrent que le dernier ancêtre commun bactérien (LBCA) était probablement déjà un diderme, et que la transition vers une structure monoderme s'est produite via des pertes indépendantes de la membrane externe (ME). Ces transitions ont principalement eu lieu chez les Terrabacteries, un sous-domaine regroupant des phylums comme les Cyanobactéries, Firmicutes et Actinobactéries. Les raisons de ces pertes d'ME chez les Terrabacteries restent floues, mais pourraient être liées à des mécanismes de biogenèse de la ME encore inconnus. Dans ma thèse, j'ai étudié la biogenèse de la ME sur deux modèles de Terrabacteries. Le premier est Veillonella parvula, une bactérie diderme du microbiome humain appartenant aux Firmicutes, mieux connus pour leurs modèles monodermes (Bacillus, Staphylococcus). J'ai caractérisé deux gènes (lap1 et lap2), associés aux didermes dans une analyses bioinformatique précédemment réalisée par le laboratoire, et montré leur rôle dans la biogenèse de la ME. J'ai montré que Lap1 et Lap2 sont localisés respectivement à la membrane cytoplasmique et à la ME. De manière surprenante, les mutations de Lap1-2 ont été compensées par des gènes de biosynthèse des lipides, y compris PlsA, une enzyme récemment découverte de synthèse d'étherlipides. Bien que je n'aie pas pu établir un lien direct entre plsA et la biogenèse de la ME, j'ai pu démontrer son rôle dans la résistance au stress oxydatif chez les bactéries. J'ai aussi développé un système CRISPR/Cas9 ciblant PlsA en profitant d'un test colorimétrique différentiel pour le WT et mutant de délétion plsA. De plus, j'ai identifié un nouveau transporteur putatif de glycérophospholipides et montré qu'il devenait essentiel en absence des protéines AsmA-like, suggérant un rôle dans le transport de ces lipides. J'ai également constaté que les Terrabacteria possédaient souvent une seule copie des protéines AsmA-like, tandis que les Gracilicutes en avaient plusieurs, suggérant l'existence d'autres transporteurs de glycérophospholipides. Le second modèle que j'ai étudié est Deinococcus radiodurans, une bactérie appartenant au phylum Deinococcota, dont l'enveloppe se distingue des modèles didermes classiques. J'ai retracé l'évolution du transporteur Lpt chez les Deinococcota, montrant qu'il s'est dupliqué avec un complexe prédit pour être impliqué dans le transport du lipopolysaccharide (LPS), et l'autre possédant un substrat encore inconnu. J'ai également montré que les Deinococcota ont perdu le LPS trois fois indépendamment. En étudiant les transporteurs Lpt chez D. radiodurans, j'ai découvert que le complexe impliqué autrefois dans le transport des LPS n'était pas important pour cette bactérie, tandis que la suppression du second complexe entraînait des défauts sévères. J'ai proposé que ce dernier soit impliqué dans l'exportation des lipoprotéines, basé sur des analyses de mutations compensatoires, la cryo-EM et la spectrométrie de masseThe bacterial envelope is essential for numerous cellular functions, including maintaining structural integrity, cell division, motility, nutrient uptake, and communication. It also plays a key role in bacterial pathogenicity, making its assembly and maintenance a critical area of research for new antimicrobial development. Bacterial envelopes exhibit a significant dichotomy, unique to the bacterial domain, between monoderm (single membrane) and diderm (double membrane) structures. Traditionally, this divide has been classified as 'Gram-positive' (monoderm) and 'Gram-negative' (diderm). Historically, it was thought that diderm bacteria evolved from monoderms through a gradual increase in complexity. However, recent phylogenomic studies challenge this view, suggesting that the last bacterial common ancestor (LBCA) was already a diderm, and that the shift to a monoderm architecture occurred through multiple independent losses of the outer membrane (OM). Interestingly, all these diderm-to-monoderm transitions are confined to the Terrabacteria subdomain, which includes phyla such as Cyanobacteria, Firmicutes, and Actinobacteria. The reasons for these OM losses specifically in Terrabacteria remain unknown but may be linked to unknown mechanisms unique to the OM biogenesis in this group. In my thesis, I studied the outer membrane biogenesis of two Terrabacteria model. The first model is Veillonella parvula, a diderm from the human microbiome that belongs to the Firmicutes which are better known for their monoderm models (Bacillus, Staphylococcus). I characterized two genes of unknown function (lap1 and lap2) which were previously associated to diderms in a bioinformatical analyses and found that they were involved in OM biogenesis and localized at the cytoplasmic membrane and OM respectively. Surprisingly, the deletions mutant of Lap1-2 was suppressed by lipid biosynthesis genes including a recently identified ether-lipid synthesis enzyme PlsA. While I could not associate the function of plsA in OM biogenesis, I could demonstrate its function in oxidative stress resistance for the first time in Bacteria. I also used a differential colorimetric assay between the WT and plsA deletion mutants to develop CRISPR/Cas9 using plsA as a target. I also identified a putative a new type of glycerophospholipid transporter and obtained preliminary data suggesting it becomes essential in a genomic background devoid of all copies of the recently discovered glycerophospholipid transporter family AsmA-like supporting its putative role. I also showed that AsmA-like proteins have higher copy numbers in Gracilicutes while Terrabacteria often have a single copy or none suggesting the existence of more glycerophospholipid transporters than previously known. Interestingly the newly identified putative transporter was more abundant in Terrabacteria. The second model I studied is Deinococcus radiodurans, a bacterium belonging to the phylum Deinococcota and whose envelope differs significantly from classical diderm models. I was able to retrace the evolutionary history of the lipopolysaccharide transporter Lpt in Deinococcota as I showed that it duplicated in this phylum with one complex predicted to be involved in LPS transport while the other was predicted to possess an unknown substrate. I showed that Deinococcota have lost LPS three times independently and studied the function of both Lpt transporters in D. radiodurans which has lost LPS. I found that the complex which used to be involved in LPS transport was not crucial to the physiology of D. radiodurans while the other complex led to grave defects in the bacterium when deleted. I suggested that this second complex is involved in lipoprotein export to the surface based on suppressor assays, cryo-EM and mass spectrometry

Prenant son depart dans une pensee du signe qui privilegie le realisme pictural, la science iconologique de diderot cherche a etablir la cricularite des notions d'imitation et de signification, de lisible et de visible faisant recours au mythe des origines figurees du langage qui a pour nom "hieroglyphe". Cependant un tel cadre conceptuel se trouve contredit par la pratique d'ecriture des images qui decouvre une tension entre voir et savoir, entre lisibilite et visualite des images parce qu'intervient, dans la relation a l'image, la dimension du fantasme et l'economie d'un desir. Nous connaissons l'écrivain Diderot, le philosophe, le directeur de l'Encyclopédie, l'esthéticien, voire le scientifique; sur un autre plan, Diderot auteur satirique demeurait, en grande partie, à découvrir. Nous nous proposons de montrer l'ampleur et la diversité des liens qui unissent Diderot à la satire en expliquant ce que représente la satire pour Diderot, en montrant dans quelle mesure et à quelles conditions Diderot est un auteur satirique, et de quelle manière il satirise. Après avoir défini la satire et après avoir rappelé ce que la notion recouvre historiquement comme genres et comme formes, nous étudions les sens que prend le terme chez Diderot et les rapports de celui-ci avec les auteurs satiriques canoniques. Nous abordons ensuite l'oeuvre satirique de Diderot selon trois axes. Entre l'abstraction et le réalisme le plus cru, l'auteur satirique possède une manière qui lui est propre de représenter son milieu. Dans cette perspective, nous analysons les représentations satiriques chez Diderot d'après les notions d'abstraction (allégories, rêves, animalisation), de monde inverse (l'utopie) et de théâtralisation du monde. Le deuxième axe de notre étude s'attache aux textes de fiction. Outre les poésies satiriques, nous examinons les oeuvres les plus connues de Diderot sous l'angle de l'esthétique comico-sérieuse (les Satires première et seconde), de la ménippée (Les Bijoux indiscrets, Jacques le Fataliste) et sous l'angle de la satire romanesque (La Religieuse). Le troisième et dernier volet de la thèse porte sur les pratiques polémiques (lettres ouvertes, apologies) et pamphlétaires de Diderot, ainsi que sur l'usage satirique qu'il fait du paradoxe.

La musique est presente dans toute l'oeuvre de diderot. Ses ecrits sur la musique sont d'abord un temoignage sur la vie musicale de l'epoque. Ils constituent un document sur le role social de la musique, sur les lieux et les modalites de la vie musicale, et enfin sur la pedagogie de la musique. Ces ecrits sont egalement l'echo de tous les debats theoriques et esthetiques qui ont agite la vie musicale du siecle. Diderot, dans un premier temps, a ete un adepte de la theorie ramiste, et donc d'une approche physico-mathematique du phenomene musical. Il a contribue a vulgariser la theorie ramiste, mais s'en est ecarte rapidement. Sa contribution essentielle consiste en fait a degager l'esthetique musicale des schemas herites de la periode classique, fondes sur la notion d'imitation ("mimesis") et sur une conception etroitement linguistique de la musique : la musique est un langage, elle parle, elle signifie comme le langage verbal. Diderot, parti de ces presupposes theoriques, aboutit a une esthetique qui decrit la musique a partir d'elle-meme et de sa structure sensible, et non pas a partir d'un modele qui lui est etranger. His writings about music are a testimony about the musical life of the 18th century. They are a document about the hows and wheres of musical life, and about musical pedagogy. At first, diderot was diderot an adept of rameau's theory, and also of a physico-mathematical approach to the musical phenomen. He contributed to the popularization of rameau's theory, but later he diverges from it. His most important contribution was freeing the musical esthetics from the schemas inheritated from the classical times and based on the a linguistic conception of music. Music is a language, it "speaks", it signifies. Getting rid of thes theoretical assumptions, diderot proposes an esthetics which describes music from the point of view of its sensitive perception, without imparting the model of linguistic description

Le problème de la guerre dans la pensée de Diderot est abordé successivement sous 3 angles, philosophique, technique, politique ; sont pris en compte d'une part les écrits personnels de Diderot, y compris les articles qu'il a lui-même rédigés pour l'encyclopédie, d'autre part le Dictionnaire raisonné dans son ensemble, là où Diderot en a assumé la direction. La 1ère partie rappelle ce qui a été au 18e, pour lui et ses contemporains l'expérience de la guerre ; elle fait le point sur la pensée religieuse et philosopphique de la guerre avant lui et autour de lui, analyse les thèmes et débats qui apparaissent à ce propos dans l'Encyclopédie, souligne l'intérêt porté par Diderot dans ses oeuvres de vieillesse à la condition du soldat. La dénonciation de la guerre par le philosophe soutient enfin le goût du critique d'art pour la peinture de batailles : le choc émotif devait être pour le spectateur-citoyen une incitation à la réflexion. Si Diderot ne s'est pas personnellement beaucoup intéressé à la technique militaire, l'art de la guerre occupe une part non négligeable dans l'Encyclopédie. Présentant dans un premier temps la formation artistique de Diderot, elle répertorie les lectures effectuées par le salonnier et souligne l'importance de l'Encyclopédie dans la constitution de ses réflexions artistiques. Sont ensuite étudiés les neuf Salons de Diderot et ses deux « essais sur la peinture ». Montrant tout d'abord comment Diderot se positionne par rapport au discours artistique de son temps, cette thèse met ensuite l'accent sur l'originalité des Salons, tant sur les plans thématique et théorique que sur le plan formel. Enfin est analysée l'influence des réflexions picturales de Diderot sur ses œuvres romanesques et dramatiquesThis thesis entitled, « Diderot seen as an art critic », highlights the interactions of Diderot's writings on art and his literary works. Firstly, Diderot's artistic formation is presented, with an exhaustive repertoire of readings made by this noted « Salon » art critic and emphasizes the major role of the Encyclopaedia in the shaping of his artistic opinions and judgments. The following sequence examines in depth Diderot's nine « Salons » as well as his two « essays on painting ». At the outset, Diderot's status (position) is perceived in relationship with the artistic way of thinking (reasoning) of his time. The thesis then emphasizes the originality of the Salons, both on a thematic and theoretical level as well as on the formal level. Le travail voudrait établir le rapport entre Diderot et la satire. En s’appuyant sur le dernier Diderot, cette étude entend mettre en lumière l’un des paradoxes qui caractérise le siècle des Lumières. En établissant des parallèles, des liens entre les satiriques latins (Horace, Pétrone, Juvénal, Perse) et Diderot, il est démontré que la satire diderotienne, à l’image du phénix, se renouvelle à l’intérieur du genre. Sous l’égide de ces moralistes, en l’occurrence Horace et Pétrone, Diderot tente de sauver un genre qui cristallise les énergies au temps des Lumières malgré la place mineure qui lui est accordée dans l’échelle des genres. En imitant ses pères spirituels, Diderot bouleverse la satire régulière sur les plans énonciatif, moral, rhétorique. Ce travail fait au sein du genre permet à celui-ci de se moderniser, de s’adapter à son temps et de résister à tous ses détracteurs qui avaient programmé sa fin. En somme, à travers la satire, on découvre cette fois-ci une autre facette de Diderot qui ne théorise plus sur le drame bourgeois mais sur la satire en tant que genre moderneThe work would aim at establishing a relationship between Diderot and Satire. Based on the last Diderot, this study intends to shed light on one of the paradoxes which characterizes the Age of Enlightenment. By drawing parallels, links between the Latin satirics (Horace, Petronius, Juvenal, Persia) and Diderot, it is demonstrated that the Diderotian Satire, like the Phoenix, is renewed within the genre. Under the aegis of the moralists, in this case Horace and Petrone, Diderot tries to save a genre that crystallizes energies in the times of Enlightenment and this, despite of the insignificant position granted to it in the genre scale. By imitating his spiritual fathers, Diderot upsets regular satire on enunciative, moral and rhetorical levels. This work done within the genre allows it to modernize, to adapt to its time and resist all its critics who had planned its end. In short, through satire, we discover this time a new facet of Diderot, which does not speculate on bourgeois drama but on satire as a modern genre

Per pastaruosius dešimtmečius, kada projektavimo srityje pradėta intensyviai taikyti CAD sistemas, atsirado palankios sąlygos kaupti ir saugoti projektinę informaciją. Įvairios projektavimo įmonės – pradedant sunkiąja pramone, baigiant baldų dizaino ir gamybos – saugo visus savo projektus: dokumentaciją, aprašymą ir, žinoma, inžinerinę informaciją – detales, duomenų bazėse. Toks darbo pobūdis leidžia lengvai grįžti ir pakoreguoti seną projektą, taip pat pasimokyti iš klaidų. Inžinieriai, dizaineriai kuria didelės vertės detalių 3D modelius, brėžinius. Pradėdami naują projektą jie gali pasinaudoti sukaupta patirtimi ir inžinieriniais sprendimais, kurie buvo taikomi kuriant ankstesnius modelius. Dažnai naudojamos detalės galiausiai tampa standartinėmis įmonės viduje. Pavyzdžiui, baldus gaminanti įmonė gali turėti ištobulintą tvirtinimo elemento šabloną, kurį šiek tiek pakoreguotą naudoja gamindama visą savo produkciją. Tokį efektyvumą įmonė gali pasiekti turėdama griežtai suorganizuotą dokumentaciją. Priešingu atveju inžinierius, kaskart dirbdamas su nauju projektu, pats to nežinodamas, vis iš naujo atranda jau duomenų bazėje esantį detalės dizainą. Todėl inžinieriai vis daugiau laiko skiria esamų detalių paieškai. Remiantis tyrimais, daugiau nei pusė darbo laiko yra skiriama reikiamos informacijos paieškai. Toks darbas nėra efektyvus, todėl yra didelis poreikis dizaino paieškos sistemoms.

Iš pilotuojamų orlaivių duomenų gauti tradiciniai tyrimai yra brangūs, o padarytų nuotraukų skiriamoji geba dažnu atveju yra ribota. Pagrindinis nepilotuojamų orlaivių (angl. Unmanned Aerial Vehicle, UAV) sistemos, veikiančios kaip fotogrametrinio jutiklio platforma, pranašumas, palyginti su tradicine pilotuojama orlaivių sistema, yra didelis lankstumas, leidžiantis gauti didelės raiškos nuotraukas iš netradicinių pozicijų ir maža kaina. Šiais laikais, matuojant nedidelius objektus, neekonomiška naudoti pilotuojamus orlaivius, todėl tobulinamos skaitmeninės fotokameros integruojamos į UAV programinę įrangą patogiam nuotraukų apdorojimui. UAV sistemos naudojamos įvairių objektų ortofotografinių aukščių modeliams (angl. Digital Elevation Model, DEM) formuoti, topografiniams planams gaminti. Tiriamajame darbe pagal UAV nuotraukas ir geodezinius matavimus tacheometru sukurtas 5000 m2 teritorijos paviršiaus DEM, kuris panaudotas topografiniam planui sudaryti. Vadovaujantis geodezijos ir kartografijos techninių reikalavimų (GKTR) reglamente pateiktais reikalavimais, išanalizuotas topografinio žemėlapio, sudaryto įprastiniais geodeziniais metodais, tikslumas ir pagal UAV nuotraukas, padarytas DJI PHANTOM 4 PRO su integruota 8,8 mm židinio nuotolio objektyvo FC6310 fotokamera. Rezultatas parodė, kad topografinių planų sudarymas, kombinuojant įprastinius geodezinius matavimus su fotogrametriniais metodais naudojant UAV, suteikia kur kas detalesnį vaizdą, tačiau būtina atsižvelgti į teritorijos užstatymo tankumą. DTM gali būti iškraipytas ir neatitikti realybėje esančio paviršiaus. Tačiau gautas planinis ir aukščių tikslumas atvirose vietovėse atitiko GKTR pateiktus reikalavimus.
