Background Physiologist-led stress echocardiography (PLSE) services provide potential for expansion of SE services and increased productivity for cardiologists. There are however no published data on the feasibility of PLSE. We sought to assess the feasibility, safety and robustness of PLSE and cardiologist-led stress echocardiography (CLSE) for coronary artery disease (CAD) assessment. Methods Retrospective analysis of 898 patients undergoing PLSE or CLSE for CAD assessment using exercise or dobutamine stress over 24 months. PLSE involved 2 cardiac physiologists (exercise) or 1 physiologist plus 1 cardiac nurse (dobutamine). A cardiology registrar was present in the echocardiography department during PLSE in case of medical complications. CLSE involved 1 physiologist and 1 trainee cardiologist who analysed the study and reviewed findings with an imaging cardiologist. Sixteen-segment wall motion scoring (WMS, WMSI) analysis was performed. Feasibility (stressor, image quality, proportion of completed studies, agreement with imaging cardiologist analysis) and safety (complication rate) were compared for PLSE and CLSE. Results The majority of studies were CLSE (56.2%) and used dobutamine (68.7%). PLSE more commonly used exercise (69.2%). Overall, 96% of studies were successfully completed (>14 diagnostic segments in 98%, P = 0.899 PLSE vs CLSE). Commencement of PLSE was associated with an increase in annual SE’s performed for CAD assessment. Complication rates were comparably very low for PLSE and CLSE (0.8% vs 1.8%, P = 0.187). There was excellent agreement between PLSE and CLSE WMS interpretation of 480 myocardial segments at rest (κ = 0.87) and stress (κ = 0.70) and WMSI (ICCs and Pearson’s r >0.90, zero Bland–Altman mean bias). Conclusion This to our knowledge is the first study of the feasibility of PLSE. PLSE performed by well-trained physiologists is feasible and safe in contemporary practice. PLSE and CLSE interpretation of stress echocardiography for CAD agree very closely.

Polymerization of fibrin in the presence of pulmonary surfactant was recently noted to induce incorporation of phospholipids into the insoluble clot material, thereby effecting severe loss of surface activity (W. Seeger, A. Elssner, A. Gunther, H.-J. Kramer, and H. O. Kalinowski. Am. J. Respir. Cell Mol. Biol. 9: 213'220, 1993). In the present study, we investigated the influence of such incorporation of calf lung surfactant extract (CLSE) on the enzymatic cleavage of the fibrin network with the use of plasmin, trypsin, or elastase. Employing a fibrin-plate assay, the proteolytic release of radioactivity originating from 125I-labeled fibrinogen was assessed, and the pattern of split products was characterized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis technique. Surface activity of CLSE was measured in the pulsating bubble surfactometer. When incorporated into the fibrin clot, CLSE inhibited the cleavage of fibrin by all proteases in a dose-dependent manner without affecting the profile of scission products. Inhibition of plasmin-induced clot lysis was also noted on incorporation of CLSE into clotted plasma and on incorporation of dipalmitoylphosphatidylcholine into fibrin polymers. In contrast, corresponding concentrations of CLSE added to the incubation medium after preformation of the fibrin matrix did not substantially influence the kinetics of fibrinolysis. CLSE incorporation into the nascent fibrin clot resulted in complete loss of surface activity, but adsorption and surface tension-lowering properties were largely restored by subsequent plasmic clot lysis. Arising fibrin split products were shown to display similar inhibitory strength on CLSE surface activity compared with fibrinogen split products.(ABSTRACT TRUNCATED AT 250 WORDS

Rabbits exposed to hyperoxia develop surfactant deficiency, abnormal lung mechanics, and increased permeability to solute. We investigated whether replenishment of depleted alveolar surfactant by the intratracheal instillation of calf lung surfactant extract (CLSE) would mitigate the increase in alveolar permeability to solute. Twenty-eight rabbits were exposed to 100% O2 for 72 h and received intratracheal instillations of 125 mg CLSE (approximately 170 mumol dipalmitoyl phosphatidylcholine) at 24 and 48 h. The interlobar and intralobar distribution of CLSE was quantified by adding [14C]dipalmitoyl phosphatidylcholine liposes into the instillate and measuring the levels of activity in lung tissue. CLSE was nonuniformly distributed in the different lung lobes, the right lower lobe receiving more CLSE than the rest. Alveolar epithelial permeability to solute was assessed by instilling 10 ml isotonic saline, which contained a trace amount of [57Co]cyanocobalamin, in the right lower lobe and measuring the disappearance of the tracer from the alveolar saline and its appearance in the arterial blood during a 1-h period. CLSE treatment was associated with significantly increased 72-h survival in hyperoxia compared with saline-treated controls (number of survivors: 16/17 vs. 5/11, P less than 0.01). CLSE treatment significantly reduced the rate constant for the movement of cyanocobalamin out of the alveolar space (24 +/- 5 vs. 42 +/- 6 min-1 x 10(-3), P less than 0.01) and tracer appearance in the blood at the end of the study (7 +/- 1 vs. 34 +/- 13%, P less than 0.01) when compared with values in saline controls.(ABSTRACT TRUNCATED AT 250 WORDS)

Depression is a mental health disorder that affects millions of people worldwide. Plants and their products are widely used in the treatments of various diseases including depression for their therapeutic effectiveness with least or no side effects. The present study has therefore been conducted to evaluate the antidepressant like effects of Citrullus lanatus seed extract (CLSE) in the reserpine-induced depressed mouse model by assessing the phytoconstituents of CLSE, body weight, behaviour, histopathology of the adrenal gland and level of plasma corticosterone. Twenty-four adult male mice of Swiss strain were distributed into four groups of six each (n=6). Group I served as control while groups II, III and IV received reserpine (RES: 0.75mg/kg/BW/day) for 14 days, CLSE (300mg/kg/BW/day) only, for 28 days, and RES+CLSE [RES treatment for 14 days followed by CLSE treatment for 28 days], respectively. Behavioural alterations were analysed by food consumption, sucrose preference, eight-arm radial maze, and forced swim tests during the treatment. RES-induced depression caused marked alterations in the body weight, all behavioural parameters, histopathology of the adrenal cortex, and in the level of plasma corticosterone, compared with the control. Administration of CLSE only, did not cause significant alterations in all the studied parameters, compared with the control. However, CLSE administration in RES-treated mice resulted in significant restoration in all the studied parameters, similar to the control. The findings therefore suggest the antidepressive activity of the seeds of C. lanatus, that may emerge as one of the potential sources among the natural therapies of depression disorder.

We have previously demonstrated that instillation of a calf lung surfactant extract (CLSE) in rabbits after exposure to 100% O2 for 64 h mitigates the progression of lung pathology after return to room air (J. Appl. Physiol. 62: 756–761, 1987). In the present study, we investigated whether we could prevent or reduce the onset and development of hyperoxic lung injury by sequential instillations of CLSE during the hyperoxic exposure. Rabbits were exposed to 100% O2. CLSE (125 mg, approximately 170 mumol of phospholipid) was suspended in 10 ml of sterile saline and instilled intratracheally into their lungs, starting at 24 h in O2, a time at which no physiological or biochemical injury was detected, and at 24-h intervals thereafter. Control rabbits breathed 100% O2 and received either equal volumes of saline or no instillations at all. CLSE-instilled rabbits had higher arterial PO2 (Pao2) values throughout the exposure period and survived longer when compared with saline controls [120 +/- 4 vs. 102 +/- 4 (SE) h; n greater than or equal to 10; P less than 0.05]. At 72 h in O2, CLSE-instilled rabbits had significantly higher lavageable alveolar phospholipid levels (12.5 +/- 1.5 vs. 5 +/- 1 mumol/kg) and total lung capacities (41 +/- 2 vs. 25 +/- 3.5 ml/kg) and lower levels of alveolar protein (24 +/- 3 vs. 52 +/- 8 mg/kg), minimum surface tension (2 +/- 1 vs. 26.1 dyn/cm), and lung wet-to-dry weights (5.9 +/- 0.2 vs. 6.5 +/- 0.3). After 72 h in O2, lungs from both CLSE- and saline-instilled rabbits showed evidence of diffuse hyperoxic injury. However, atelectasis was less prominent in the former. We concluded that instillation of CLSE limits the onset and development of hyperoxic lung injury to the alveolar epithelium of rabbits.

Surface activity and sensitivity to inhibition from phospholipase A2 (PLA2), lysophosphatidylcholine (LPC), and serum albumin were studied for a synthetic C16:0 diether phosphonolipid (DEPN-8) combined with 1.5% by weight of mixed hydrophobic surfactant proteins (SP)-B/C purified from calf lung surfactant extract (CLSE). Pure DEPN-8 had better adsorption and film respreading than the major lung surfactant phospholipid dipalmitoyl phosphatidylcholine and reached minimum surface tensions <1 mN/m under dynamic compression on the Wilhelmy balance and on a pulsating bubble surfactometer (37°C, 20 cycles/min, 50% area compression). DEPN-8 + 1.5% SP-B/C exhibited even greater adsorption and had overall dynamic surface tension lowering equal to CLSE on the bubble. In addition, films of DEPN-8 + 1.5% SP-B/C on the Wilhelmy balance had better respreading than CLSE after seven (but not two) cycles of compression-expansion at 23°C. DEPN-8 is structurally resistant to degradation by PLA2, and DEPN-8 + 1.5% SP-B/C maintained high adsorption and dynamic surface activity in the presence of this enzyme. Incubation of CLSE with PLA2 led to chemical degradation, generation of LPC, and reduced surface activity. DEPN-8 + 1.5% SP-B/C was also more resistant than CLSE to direct biophysical inhibition by LPC, and the two were similar in their sensitivity to biophysical inhibition by serum albumin. These findings indicate that synthetic surfactants containing DEPN-8 combined with surfactant proteins or related synthetic peptides have potential utility for treating surfactant dysfunction in inflammatory lung injury.

Monomolecular films of phospholipids in the liquid-expanded (LE) phase after supercompression to high surface pressures (π), well above the equilibrium surface pressure (πe) at which fluid films collapse from the interface to form a three-dimensional bulk phase, and in the tilted-condensed (TC) phase both replicate the resistance to collapse that is characteristic of alveolar films in the lungs. To provide the basis for determining which film is present in the alveolus, we measured the melting characteristics of monolayers containing TC dipalmitoyl phosphatidylcholine (DPPC), as well as supercompressed 1-palmitoyl-2-oleoyl phosphatidylcholine and calf lung surfactant extract (CLSE). Films generated by appropriate manipulations on a captive bubble were heated from ≤27°C to ≥60°C at different constant π above πe. DPPC showed the abrupt expansion expected for the TC-LE phase transition, followed by the contraction produced by collapse. Supercompressed CLSE showed no evidence of the TC-LE expansion, arguing that supercompression did not simply convert the mixed lipid film to TC DPPC. For both DPPC and CLSE, the melting point, taken as the temperature at which collapse began, increased at higher π, in contrast to 1-palmitoyl-2-oleoyl phosphatidylcholine, for which higher π produced collapse at lower temperatures. For π between 50 and 65 mN/m, DPPC melted at 48–55°C, well above the main transition for bilayers at 41°C. At each π, CLSE melted at temperatures >10°C lower. The distinct melting points for TC DPPC and supercompressed CLSE provide the basis by which the nature of the alveolar film might be determined from the temperature-dependence of pulmonary mechanics.

The sensor needs to maneuver to get better observability in Bearings-Only passive target tracking with single sensor which makes the observation time longer. Multisensor Bearings-Only passive target tracking can solve the problem using exchange data. So the constrained Least Square Estimation (CLSE) algorithm is proposed for Multisensor Bearings-Only passive target tracking. The constrained condition is introduced to the Least Square Estimation algorithm firstly. Then the eigenvector corresponding to the least eigenvalue of the matrix is used to overcome the shortcoming of Extend Kalman Filter algorithm which needs the initial value. Also the bias problem of Least Square Estimation is conquered. The simulation results show that the CLSE can gradually approach the Cramer-Rao Lower Bound and its precision is better than the Least Square Estimation algorithm. Finally the CLSE is proved to be a gradually, stable and almost unbiased estimation algorithm.

This study compared the shear bond strength (SBS) and micromorphology of composite resin to human dentin after pre-treatment with silica-modified aluminum oxide air abrasion. Forty-six molar teeth were treated with either Scotchbond Multi-Purpose (SCMP) or Clearfil SE Bond (CLSE) adhesive. Buccal surfaces were pre-treated with the CoJet air abrasion system (SB), and lingual surfaces were controls. The adhesion of light-cured resin composite to the treated dentin surface was evaluated with SBS. After debonding, substrate surfaces were examined with an optical microscope for failure analysis. In addition, 15 molar teeth were sectioned and randomly assigned to one of five groups, according to the dentin surface pre-treatment and adhesive type, and examined with high-vacuum scanning electron microscopy/energy dispersive X-rays (SEM/EDS). The type of adhesive had a significant effect on SBS (p = 0.000); CLSE had the highest values. SB did not affect SBS (p = 0.090). SEM/EDS revealed residual aluminum and/or silicon on all dentin surfaces after SB, except for the control. Treatment with 32% phosphoric acid in the SCMP adhesive decreased the amounts of aluminum and silicon compared to SB dentin only, whereas CLSE resulted in similar quantities of aluminum and silicon as air-abraded dentin. The results of this study indicate that CLSE might have a higher bond strength to dentin than SCMP. Pre-treatment with SB does not appear to affect bonding strength.

We investigated alterations in pH on the surface properties of natural lung surfactant and the calf lung surfactant extract (CLSE), suspended in 10 mM N-2-hydroxyethylpiperazine-N′-2-ethanesulfonic acid, using a pulsating bubble surfactometer. Increasing the pH value of the medium to > 7.4 decreased the ability of CLSE, but not of natural lung surfactant mixtures (2 mg phospholipid/ml), to achieve a low minimum surface tension during dynamic compression and enhanced their sensitivity to albumin inactivation. These detrimental effects on surface tension were reversed by addition of surfactant protein A (SP-A; 3% by weight) or by increasing the lipid concentration to 4 mg/ml. SP-A-induced lipid aggregation at pH 10 was not different than at pH 7.4. Alkalinization impaired the ability of CLSE to restore normal lung mechanics in excised surfactant-deficient rats lungs. These results indicate that cooperation between SP-A and the hydrophobic surfactant proteins has an important role in achieving low minimum surface tension at pH > or = 7.6.

Cette thèse traite l'inférence statistique de quelques processus de diffusion affine dans \( \R^m_+ \times \R^n \), avec m,n\in\N. Cette sous-classe de diffusions, notée par \textit{AD}(m,n), est appliquée à la tarification des options sur obligations et des actions, ce qui est illustré pour les modèles de Vasicek, Cox-Ingersoll-Ross (CIR) et Heston. Dans cette thèse, nous considérons deux différents modèles: le premier lorsque \( m = 1 \) et \( n \in \mathbb{N} \) et le deuxième lorsque \( m = 2 \) et \( n = 1 \). Pour le modèle \mathit{AD}(1, n), nous introduisons, au Chapitre 2, un résultat de classification où nous distinguons trois cas différents : sous-critique, critique et surcritique. Ensuite, nous étudions la stationnarité et l'ergodicité de sa solution sous certaines hypothèses sur les paramètres du drift. Pour le problème d'estimation paramétrique, nous utilisons deux méthodes différentes : l'estimation par maximum de vraisemblance (MLE) et l'estimation des moindres carrés conditionnels (CLSE). Au Chapitre 2, nous présentons l'estimateur obtenu par la méthode MLE basée sur des observations en temps continu et nous étudions sa consistance et son comportement asymptotique dans des cas ergodiques et non-ergodiques particuliers. Au Chapitre 3, nous présentons l'estimateur obtenu par la méthode CLSE basée sur des observations en temps continu puis discret avec haute fréquence et horizon infini et nous étudions sa consistance et son comportement asymptotique dans des cas ergodiques et non-ergodiques particuliers. Il est à noter ici que nous obtenons les mêmes résultats asymptotiques que dans le cas continu sous des hypothèses supplémentaires sur le pas de discrétisation \( \Delta_N \). Au Chapitre 4, nous étudions le modèle \mathit{AD}(2, 1), également appelé modèle de double Heston. Dans un premier temps, nous introduisons sa classification suivant les cas sous-critique, critique et surcritique. Dans un second temps, nous établissons les théorèmes de stationnarité et d'ergodicité y associés. Dans la partie statistique de ce chapitre, nous étudions les estimateurs par la méthode MLE et la méthode CLSE du modèle de double Heston en se basant sur des observations en temps continu dans le cas ergodique et nous introduisons les théorèmes de consistance et de normalité asymptotique pour chaque estimateur obtenu
This thesis deals with statistical inference of some particular affine diffusion processes in the state space \R_+^m\times\R^n, where m,n\in\N. Such subclass of diffusions, denoted by \mathit{AD}(m,n), is applied to the pricing of bond and stock options, which is illustrated for the Vasicek, Cox-Ingersoll-Ross (CIR) and Heston models. In this thesis, we consider two different cases : the first one is when m=1 and n\in\N and the second one is when m=2 and n=1. For the \mathit{AD}(1,n) model, we introduce, in Chapter 2, a classification result where we distinguish three different cases : subcritical, critical and supercritical. Then, we study the stationarity and the ergodicity of its solution under some assumptions on the drift parameters. For the parameter estimation problem, we use two different methods: the maximum likelihood estimation (MLE) and the conditional least squares estimation (CLSE). In Chapter 2, we present the estimator obtained by the MLE method based on continuous time observations and we study its consistency and its asymptotic behavior in ergodic and particular non-ergodic cases. In Chapter 3, we present the estimator obtained by the CLSE method based on continuous then discrete time observations with high frequency and infinite horizon and we study its consistency and its asymptotic behavior in ergodic and particular non-ergodic cases. It is worth to note here that we obtain the same asymptotic results in both discrete and continuous sets under additional assumptions on the discretization step \Delta_N. In Chapter 4, we study the \mathit{AD}(2,1) model, called also double Heston model, we introduce first its classification with respect to subcritical, critical and supercritical case and we establish the relative stationarity and ergodicity theorems. In the statistical part of this chapter, we study the MLE and the CLSE of the ergodic double Heston model based on continuous time observations and we introduce its consistency and asymtotic normality theorems for each estimation method

O gênero Erythrina está presente mundialmente nas regiões tropicais e subtropicais. Estudos fitoquímicos utilizando vários órgãos dessas plantas, demostraram a presença de alcaloides, flavonoides, pterocarpanos e triterpenóides. Várias espécies são encontradas no Brasil, dentre elas Erythrina falcata e Erythrina crista-galli, conhecidas popularmente como “corticeira” e utilizadas medicinalmente devido à ação sedativa, ansiolítica, anti-inflamatória e antihipertensiva. Este trabalho objetivou estudar a composição química de extratos vegetais obtidos de folhas de E. falcata e E. crista-galli e ainda, avaliar o potencial hipotensor in vivo de extratos de E. falcata. Inicialmente, após coleta do material, as folhas foram selecionadas, submetidas à secagem e trituradas. Então, foram submetidas à extração por maceração exaustiva utilizando etanol 40% (v/v) e por infusão utilizando água a 100 °C. Para a caracterização em termos de fenólicos totais e conteúdo de flavonoides, os extratos foram quantificados por espectrofotometria. Para o ensaio cromatográfico, os extratos foram analisados por CLAE em sistema de fase reversa, com fase móvel consistindo de acetonitrila:água em eluição por gradiente e fluxo de 1,0 mL/min. Para a análise por CLUEESI- MS, a fase móvel foi composta de mistura de acetonitrila:metanol (4:1) e ácido fórmico pH 3,0, com eluição por gradiente e fluxo de 0,2 mL/min. A detecção por espectrometria de massas foi conduzida a partir das seguintes condições: N2 como nebulizante; energia de colição 4.0 eV; temperatura da fonte do eletrospray e do gás de solvatação 100°C e 120°C, respectivamente; voltagem do capilar 3000V; voltagem do cone 40V. Os espectros de massas foram obtidos na faixa de m/z 200-800. A avaliação dos efeitos hemodinâmicos foi realizadaem ratos wistar normotensos, anestesiados com uretana (1,4 mg/Kg), via canulação da artéria carótida (para a verificação da PAS, PAD e FC) e veia jugular (para administração do extratose drogas). A avaliação do potencial hipotensor da E. falcata foi investigada através da realização de uma curva crescente de administração dos extratos e os possíveis mecanismos de ação envolvidos foram analisados através da administração de diferentes drogas sendo elas: L-NAME (30 mg/kg); losartana (10 mg/Kg); hexametônio (20mg/Kg) e propranolol (5mg/kg). Os teores de fenólicos totais para E. falcata e E. crista-galli estiveram na faixa de 1,3193 – 1,4989 mgEAG/mL para os extratos obtidos por maceração e de 0,8771 – 0,9506 mgEAG/mL para as infusões. Em flavonoides totais, os conteúdos estiveram na faixa de 7,7829 – 8,1976 ER mg/g para os extratos obtidos por maceração e 9,3471 – 10,4765 ER mg/g para as infusões. Na determinação por CLUE-MS, os dados de íon molecular e fragmentos de massa indicaram a composição predominante em alcaloides, sugerindo-se os componentes erythristemine, 11β-methoxyglucoerysodine, erysothiopine, 11β- hydroxyerysodine–glicose e 11-hydroxyerysotinone-ramnosídeo. O extrato aquoso da E. falcata mostrou-se um potente hipotensor dose-dependente, causando uma queda na PAS de 23 a 35% e na PAD de 32 a 49% para ambos os extratos estudados, sem influenciar a FC, podendo este efeito estar relacionado com a via dos receptores β-adrenérgicos.
The genus Erythrina is present worldwide in tropical and subtropical regions. Phytochemical studies using various organs of these plants demonstrated the presence of alkaloids, flavonoids, pterocarpans and triterpenoids. Several species are found in Brazil, among them Erythrina falcata and Erythrina crista-galli, which are popularly known as “corticeira” and. used medicinally due to it action as sedative, anxiolytic, anti-inflammatory and antihypertensive. The present study aimed to investigate the chemical composition of extracts obtained from leaves of E. falcata and E. crista-galli, and still, to evaluate the hypotensive potential in vivo of E. falcata extracts. Initially, after collection of material plant, the leaves were selected, submitted to dryness and powdered. Then, submitted to extration by exhaustive maceration using ethanol 40% (v/v) and by infusion using water at 100 °C. For characterization in terms of phenolics and flavonoid content, the extracts were quantified by spectrophotometry. For chromatographic assay, the extracts were analysed by HPLC in reversed phase system, with a mobile phase consisting of acetonitrile:water in gradient elution and flow rate of 1.0 mL/min. For analysis by UPLC-ESI-MS, the mobile phase consisted in a mixture of acetonitrile:methanol (4:1) and 0.1% formic acid pH 3.0, with a elution by gradient and flow rate of 0.2 mL/min. The MS detection was performed following the conditions: N2 as nebulizing; collision energy 4.0 eV; temperature of electrospray source and desolvation gas 100°C and 120°C, respectively; capillary voltage 3000V; sample cone 40V. Mass spectra were recorded in the range of m/z 200-800. The evaluation of the hemodynamic effects was performed in normotensive Wistar rats, anesthetized with urethane (1.4 mg/kg) by cannulation of the carotid artery (for verification of SBP, DBP and HR) and jugular vein (for administration of the extracts and drugs). The hypotensive potential of E. falcata was investigated by conducting a growing curve administration of the extracts and the possible mechanisms involved were analyzed by administering different drugs such as: L-NAME (30 mg/kg); losartan (10 mg/kg); hexamethonium (20 mg/kg) and propranolol (5 mg/kg). The total phenolics content for E. falcata and E. crista-galli was from 1.3193 to 1.4989 mgGAE/mL for maceration and 0.8771 to 0.9506 mgGAE/mL for infusions. In total flavonoid, the content was from 7.7829 to 8.1976 mg RE/g for maceration and 9.3471 to 10.4765 RE mg/g for infusions. The molecular ions and mass fragments obtained by UPLCMS indicated the predominant composition in alkaloids, suggesting the components erythristemine, 11β-methoxyglucoerysodine, erysothiopine, 11β-hydroxyerysodine-glucose and 11-hydroxyerysotinone-rhamnoside. The aqueous extract of E. falcata showed to be a potent dose-dependent hypotensive, decreasing the SBP in 23 to 35% and DBP in 32 to 49% for both extracts, without influence in HR, and this effect may be due to the route of β- adrenergic receptors.

O maracujá, nome popular atribuído ao fruto das diversas espécies do gênero Passiflora, da família Passifloraceae, é amplamente comercializado e consumido no mundo, sendo o Brasil um dos maiores produtores do fruto. Alguns estudos apontam possível toxicidade relacionada às espécies de Passiflora, principalmente P. incarnata. No entanto, há pouco conhecimento acerca das espécies P. edulis e P. alata, sobretudo em relação à polpa e sementes. Os extratos da polpa e das sementes dos frutos dessas duas espécies de \"maracujá\", Passiflora alata e Passiflora edulis, foram estudados com o objetivo de identificar alcaloides harmânicos, pelo preparo das amostras por extração por sorção em barra magnética recoberta com polidimetilsiloxano (SBSE-PDMS) e SBSE recoberta com polietilenoglicol silicone (SBSE-EG Silicone) e análise por cromatografia líquida de alta eficiência com detector por fluorescência (CLAE-Flu) e cromatografia líquida de ultra eficiência acoplada à espectrometria de massas sequencial (CLUE-EM/EM). A análise dos alcaloides harmana e harmina nos extratos da polpa de P. alata foi feita por meio do método de adição de padrão e mostrou menor quantidade destes alcaloides, em comparação com os resultados da análise dos extratos da polpa dos frutos de P. edulis, no trabalho de Pereira e colaboradores. As análises CLUE-EM e CLUE-EM/EM possibilitaram a identificação dos alcaloides nos extratos: nas sementes de P. alata, os alcaloides harmana, harmina, harmol, harmalol e harmalina foram identificados, enquanto que na polpa, harmana e harmina tiveram a confirmação da sua presença. Nos extratos da polpa dos frutos de P. edulis observou-se os alcaloides harmana, harmina e harmalina. E nas sementes de P. edulis harmina foi encontrada, porém há indícios da presença de harmana. A literatura sobre as barras de SBSE-EG Silicone Twister&®; não relata nenhum estudo relacionado ao seu uso para extração e concentração de alcaloides harmânicos nos extratos de P. alata e P. edulis. Por isso foi proposto inicialmente a aplicação do planejamento fatorial fracionário para otimização do método de extração, utilizando os padrões comerciais dos alcaloides harmana e harmina. O planejamento experimental revelou as variáveis principais e seus níveis de importância, e a partir destes resultados foi realizado o estudo cinético dos tempos de extração e de dessorção das barras de SBSE-EG Silicone. Porém, os resultados mostraram que as barras de SBSE-EG Silicone não são adequadas para a extração dos alcaloides harmana e harmina, uma vez que a recuperação obtida foi baixa, na ordem de 30%.
\"Maracujá\" is the popular name given to the fruit of several species of Passiflora genus, from Passifloraceae family, it is widely commercialized and consumed around the world, and Brazil is one of the largest producers of this fruit. Some studies pointed out the possible toxicity related to Passiflora species, mainly P. incarnata. Although, there is a lack of knowledge about the P. edulis and P. alata species, especially with regards to the pulp and seeds. The extracts of pulp and seeds from the \"maracujá\" species Passiflora alata and Passiflora edulis, were studied in order to identify harman alkaloids. The samples were prepared by extraction with sorptive stir bar coated with polydimethylsiloxane (SBSE-PDMS) and SBSE coated with polyethylene glycol silicon (SBSE-EG Silicone). The samples were analyzed by high performance liquid chromatography with fluorescence detector (HPLC-Flu), and ultra-high pressure liquid chromatography coupled to tandem mass spectrometry (UHPLC-MS/MS). Harmane and harmine alkaloids in P. alata pulp extracts were analyzed using the standard addition method and the results showed a lower amount of these alkaloids, compared with the test results for the extracts from the P. edulis pulp in the work of Pereira et al. UHPLC-MS and UHPLC-MS/MS analysis enabled to identify the alkaloids amount present in the extracts. In the P. alata seeds extract the following alkaloids were identified harmane, harmine, harmol, harmalol and harmaline, while in the pulp extract, harmane and harmine were confirmed. In the extracts of P. edulis pulp the alkaloids identified were harmane, harmine and harmaline. And in the P. edulis seeds extract the harmine alkaloid was found, some indications of the presence of harmana were observed. The literature about SBSE-EG Silicone Twister® bars reports no study related to their use for extraction and concentration of harman alkaloids in P. alata and P. edulis. Thus, it was initially proposed the application of fractional factorial design to optimize the extraction method using commercial standards of harmane and harmine alkaloids. The experimental design revealed the main variables and their importance levels, and from these results kinetic studies were performed for the extraction and desorption times of SBSE-EG Silicone bars. However, the results showed SBSE-EG Silicone bars are not suitable for the extraction of harmane and harmine alkaloids, since the recovery obtained was low, on the order of 30%.

Over the past two decades, the readership of poetry has declined to the point that the art form is seldom engaged with by the public. I argue that reading poetry requires a skillset that must be learned, practiced, and refined. While close reading is traditionally trained in college classrooms, such spaces cannot reach broad audiences. To address this dearth, I have developed a web app that applies interactive learning strategies, through a series of exercises, to cultivate expert reading practices in novice users. Close Reading will guide users through poems by Robert Frost. With each poem, users will progress through exercises grounded in the practices of expert readers. For instance, users will block poems into sections to allow a chunking of the material, slowing down novice reading speeds. Another exercise cognitively models the act of reading by displaying the sequential thoughts of a reader making sense of a work. Furthermore, Socratic questioning will attempt to stimulate an internal dialogue to foster focus and interpretation. These exercises will build on one another and attempt to replicate pedagogical processes observed in the classroom. Performing these pedagogical exercises will provide a resource for developing the skillset necessary for poetry appreciation. This ambitious digital humanities project experiments with a new venue for pedagogy and poetry, promoting an engagement with the public frequently neglected in academic work.

Resumo: O Brasil é o maior produtor mundial de citrus e o principal exportador de suco. Dentre as espécies cultivadas no Brasil, há a predominância de laranjas Pera e tangerinas Murcote. Porém, os agricultores brasileiros tem enfrentado a incidência de várias pragas agrícolas na cultura de citrus, de forma que, para manter a produção e a qualidade, tem sido necessário aplicar agrotóxicos de forma constante e cada vez maior. Por outro lado, devido ao risco resultante da exposição dos consumidores aos resíduos de agrotóxicos presentes nos alimentos, agencias governamentais reguladoras tem estabelecido limites máximos de resíduos (LMR) para todos os agrotóxicos. Neste contexto, três métodos para a determinação de resíduos dos principais agrotóxicos (aldicarbe, difenoconazol, diflubenzurom, diurom, imidacloprido e tiofanato metílico) aplicados no cultivo de laranjas Pera e tangerinas Murcote foram desenvolvidos e validados. Vários métodos de preparo de amostras foram avaliados e o método QuEChERS foi o que se mostrou mais eficiente, rápido e simples. Foram utilizadas diferentes técnicas cromatográficas para a validação do método: cromatografia líquida de alta eficiência com detecção por arranjo de diodos (CLAE-DAD), cromatografia líquida acoplada a espectrometria de massas em série (CL-EM-EM) e cromatografia líquida de ultra eficiência com detecção por arranjo de diodos (CLUE-DAD). Os parâmetros analíticos mostraram que o método proposto extrai de maneira satisfatória os agrotóxicos das matrizes, permitindo sua detecção por todas as técnicas analíticas utilizadas. Entretanto, devido a complexidade da matriz estudada foi necessário fazer a calibração na matriz. As recuperações obtidas foram de 79 a 124% com CLAE-DAD, de 83 a 119% com CL-EM-EM e de 78 a 113% com CLUEDAD, com coeficientes de variação menores que 15% para todas as tecnicas cromatograficas. Os limites de quantificacao obtidos mostraram que os métodos podem ser utilizados para a detecção dos agrotóxicos em concentrações abaixo dos LMR estabelecidos pela legislação brasileira e dos EUA. Com a CLUE-DAD obtiveram-se os menores tempos de análise, 7 min, e a CL-EM-EM foi a técnica cromatografica mais seletiva e com a melhor detectabilidade (4 mg L) utilizada neste trabalho. O método foi aplicado a amostras coletadas no comercio da cidade de Campinas-SP e o agrotóxico encontrado com maior frequência foi o imidacloprido, mas em concentrações abaixo dos LMR
Abstract: Brazil is the world's largest producer of oranges and the largest exporter of juice. Among the species grown in Brazil, there is a predominance of Pera oranges and Murcote tangerines. Brazilian agriculturist have faced several diseases during orange and tangerine cultivation that demand constant and even expensive applications of pesticides to mantain production and quality. On the other hand, because of potential health risk to the consumers, resulting from acute and/or chronic dietary exposure, government regulatory agencies have established maximum residue limits (MLR) for all the pesticides. Thus, this work developed and validated three methods to determine the main pesticides applied to Pera orange and Murcote tangerine cultivation in Brazil: aldicarb, difenoconazole, diflubenzuron, diuron, imidacloprid and thiophanate-methyl. Several sample preparation methods were evaluated and the QuEChERS method was the more suitable, rapid and simplest extraction method for the matrices studied. The methods were validated using different chromatographic techniques: high performance liquid chromatography ¿ diode array detection (HPLC-DAD), liquid chromatography ¿ tandem mass spectrometry (LC-MSMS) and ultra-performance liquid chromatography ¿ diode array detection (UPLC-DAD). The analytical parameters demonstrate that the proposed methodologies satisfactorily extract the pesticides from the matrices, allowing detection using all the quantification techniques employed. Because of the complexity of the matrices studied, it was necessary to employ matrix-matched calibration. When using HPLC-DAD recoveries of 79 to 124% were obtained, with LC-MS-MS the recoveries were 83 to 119% and when using UPLC they were 78 to 113%, with coefficients of variation below to 15% for all chromatographic techniques. These limits of quantification show that the methods developed can be used to detect these pesticides at concentrations below the MRL established by Brazilian and USA legislation. UPLC-DAD had a more rapid analysis time, 7 min, and LC-MS-MS had the best selectivity and detectivity (4 mg L). The methods were applied to samples collected in markets of Campinas city, and the pesticide most frequently found was imidacloprid, with concentrations well below the listed MRL
