O objectivo da pesquisa foi estudar os principais factores que influenciam no transbordo das águas das bacias de contenção na zona de estudo. A metodologia usada foi recurso bibliográfico e entrevistas do tempo de vivência na região (TV), da existência de eficiência no tratamento do lixo (ETL), descarte de resíduos nas bacias (DR), manutenção regular das bacias (MR), tratamento de esgotos sanitários (TES), consequências na construção em linhas de águas (CLA), uso de águas das bacias para fins domésticos (UAFD), consequências socioeconómicas (CSE) e benefícios (B). Se determinou também a capacidade volumétrica (CV) e capacidade de contenção (CC), assim como alguns parâmetros geométricos das bacias. Seis bacias de contenção (Bc) foram estudadas: Bc1,Bc2, Bc3, Bc4, Bc5, Bc6). Os resultados da TV foram divididos em 5 grupos (1-5, 6-10, 11-15, 16-20 e mais de 20 anos de vivência) num total de 93 pessoas. Para ETL, DR, MR, TES, CLA, UAFD e CSE a maior parte respondeu não existir e para B afirma existir. Quanto CV, CC os resultados indicam a Bc2 e Bc1 terem mais e Bc3 e Bc6 apresentaram menos. Os resultados dos parâmetros mostram para bacia Bc2 (39.118m2) e Bc5 e (8.774 m2). Para perímetros Bc4 registrou (1.450m) e Bc6 (484,8m). O coeficiente de compacidade indicou para Bc4 (3,016) e Bc5 (1,13). O factor de forma indicou no Bc6, (0,996), e Bc5 (0,04). O índice de circularidade foi no Bc3 (0,770) e Bc4 e Bc5 (0.1288). Conclui-se que existe influência de factores antropogénicos e naturais no transbordo das águas.

The relationship between sport result and pre-competition mental state of 109 boccia athletes was analyzed. Mental state was described by: athletic identity, self-esteem, self-efficacy for sports, hope for success, fear of failure, anxiety, and expectancy of success. Correlation analyses were made for all four boccia classes (BC1, BC2, BC3, and BC4) and revealed that only athletic identity was associated with sport result in class BC4. Four hierarchical multiple regression models (for BC1, BC2, BC3, and BC4 boccia classes) were created, with sport result as the dependent variable. Only the BC4 model was significant and included athletic identity, anxiety, self-efficacy for sports, and expectancy of success, which explained 49% of variance in sport result. BC4 class results indicate that psychological variables have a potential impact on sport performance in boccia, and the type and level of disability should be taken into account.

ABSTRACT Introduction: Although studies have been conducted in the athlete population during the pandemic, its impacts on Paralympic athletes are still unknown. Objective: To evaluate the mood and quality of life of Paralympic boccia players during the COVID-19 pandemic period. Method: Questionnaires assessing mood, quality of life, and demographic data were sent electronically via social media. The study included 43 athletes from all functional classes (BC1 = 8, BC2 = 12, BC3 = 11, and BC4 = 11) and four regions of Brazil (Northeast = 12, Southeast = 12, Central-West = 3, and South = 17). ANOVA, t test, and similar non-parametric tests were used, with the respective effect sizes (ES). Results: Regarding mood states, BC4 athletes differed from BC1 athletes in the depression subscale score (25.5 vs. 14.5, p = 0.026, ES=1.47). The fatigue subscale score differed between BC4 and BC1 athletes (17.8 vs. 10.4, p = 0.023, ES=1.38) and between BC4 and BC2 athletes (17.8 vs 10.3, p = 0.008, ES=1.32). BC4 athletes had higher total mood disturbance (TMD) values than BC1 (175 vs. 141, p = 0.025, ES=1.35) and BC2 (175 vs.141, p = 0.025, ES=0.97) athletes. Lower stress (18.5 vs.21.8, p = 0.027, ES=0.64) and TMD (148.0 vs 162.0; p = 0.044; ES= 0.53) values were observed among the national level athletes. In terms of quality of life, differences were found between BC4 and BC2 athletes in the psychological health domain (3.73 vs. 4.49, p = 0.024, ES=0.89) and between athletes from the Northeast and South in the environment domain (3.39 vs. 4.18, p=0.030, ES=0.44). Conclusion: BC4 athletes are more susceptible to negative mood and quality-of-life perceptions. Additionally, athletes from the Northeast region may have poorer perceptions of quality of life related to the environment in which they live. Level of evidence III; Retrospective study.

Partially fertile synthetic amphiploids (2n = 8x = 56) of Canada wildrye × slender wheatgrass F1 hybrid were crossed to slender wheatgrass and tetraploid lines resembling slender wheatgrass were recovered after four backcrosses. The BC1 progeny was hexaploid, while the BC2 and BC3 progenies were aneuploids with chromosome numbers ranging from 31 to 38. Tetraploid lines were recovered in the BC4 generation along with 29- and 30-chromosome types. Multivalents (mostly trivalents) resulting from intergenomic pairing were fequently encountered in BC1 and BC2 plants. The multivalent frequencies declined with each succeeding backcross. Pairing in the 28-chromosome BC4 plants was completely regular with 14 bivalents. Intergenomic pairing in the backcross progeny promoted introgression from Canada wildrye to slender wheatgrass.Key words: Elymus, intergenomic pairing, trivalents, recombination.

In this study, T. medium (zigzag clover, 2n = 80 and T. pratense (red clover, 2n = 28) were hybridized and backcrossed four times to T. pratense (BC1, BC2, BC3and BC4), and the seed production and vigor of the backcross progenies were assessed under field conditions. Both female and pollen fertility of the backcross progenies increased as the backcross generations advanced. Female and pollen fertility of BC4 were 21.3 and 65.3%, respectively. When honeybees were used as pollinators, almost all BC4 plants produced mature seeds in the field. Most of the BC1, BC2, and BC3 plants died by the end of the fourth year in the field, whereas 61% of the BC4 plants survived for 4 yr. The number of chromosomes in 79% of the BC4 plants was more than 28, which is greater than that of T. pratense. Differences in leaflet size and shape between BC4 plants and T. pratense suggest that genes from T. medium were expressed in the BC4 plants. These results indicate that BC4 plants may be useful for the improvement of the persistence of T. pratense. The main aim of this work was to verify the metabolic and functional aptitude of 15 vaginal strains belonging to Lactobacillus crispatus, Lactobacillus gasseri, and Limosilactobacillus vaginalis (previously Lactobacillus vaginalis), already characterized for their technological and antimicrobial properties. In order to evaluate the metabolic profile of these vaginal strains, a phenotype microarray analysis was performed on them. Functional parameters such as hydrophobicity, auto-aggregation, deconjugation of bile salts, adhesion to an intestinal cell line (Caco-2), and a simulated digestion process were evaluated for these strains. A good number of these strains showed hydrophobicity values higher than 70%. Regarding the auto-aggregation assay, the most promising strains were L. crispatus BC9 and BC1, L. gasseri BC10 and BC14, and L. vaginalis BC17. Moreover, L. crispatus BC4, BC6, BC7, and BC8 were characterized by strong bile salts hydrolase activity (BHS). In addition, L. crispatus BC8 and L. vaginalis BC17 were characterized by a medium ability to adhere to Caco-2 cells. Data related to digestion process showed a strong ability of vaginal lactobacilli to withstand this stress. In conclusion, the data collected show the metabolic versatility and several exploitable functional properties of the investigated vaginal lactobacilli.

Abstract The nucleotide sequence of partial cDNA clones coding for the core protein of a human polymorphic epithelial mucin has recently been obtained, this mucin consists of a highly conserved 60 bp tandem repeat and the amino acids commonly found are PDTRPAPGSTAPPAHGVTSA. We synthesized three peptides, 1) P1.24 containing the 20 amino acids and four amino acids (PDTR) of the adjoining repeat; 2) P1.15 consisting of the first fifteen (PDTRPAPGSTAPPAH) and P1.09 the second nine amino acids (GVTSAPDTR) of peptide P1.24. The reactivities of the synthetic peptides with mAb known to react with breast cancer (BC1, BC2, BC3, HMFG-1, 3E1.2, and RCC-1) were studied. The synthetic peptide, P1.24, corresponding to the antigenic sequence predicted from the tandem repeat reacted with antibodies BC1, BC2, and BC3 (known to react with human milk mucin and mucin expressed in breast cancer) and the antibody HMFG-1 which was used to select the cDNA clones. In addition, the epitopes recognized by BC1, BC2, and BC3 appear to be in the same region of the molecule represented by their reactions with the nine amino acids in peptide P1.09 (GVTSAPDTR). By contrast, other antibodies such as 3E1.2 which reacts only weakly with components of human milk, and RCC-1 that detects a low Mr component (95 kDa) in breast cancer, had no specific reaction with the synthetic peptides, indicating that their epitopes are distinct from those of BC1, BC2, BC3, and HMFG-1. Inasmuch as the antibodies HMFG-1, BC1, BC2, and BC3 react with the fully processed milk mucin, it is likely that some of the peptide is exposed, even in the fully glycosylated molecule. Identification of the different epitopes could lead to the development of "second generation" mAb with enhanced specificity for breast carcinoma using the appropriate synthetic peptides as immunogens.

Vaginal lactobacilli offer protection against recurrent urinary and vaginal infections. The precise mechanisms underlying the interaction between lactobacilli and the host epithelium remain poorly understood at the molecular level. Deciphering such events can provide valuable information on the mode of action of commensal and probiotic bacteria in the vaginal environment. We investigated the effects exerted by five Lactobacillus strains of vaginal origin (Lactobacillus crispatus BC1 and BC2, Lactobacillus gasseri BC9 and BC11 and Lactobacillus vaginalis BC15) on the physical properties of the plasma membrane in a cervical cell line (HeLa). The interaction of the vaginal lactobacilli with the cervical cells determined two kinds of effects on plasma membrane: (1) modification of the membrane polar lipid organisation and the physical properties (L. crispatus BC1 and L. gasseri BC9); (2) modification of α5β1 integrin organisation (L. crispatus BC2, L. gasseri BC11 and L. vaginalis BC15). These two mechanisms can be at the basis of the protective role of lactobacilli against Candida albicans adhesion. Upon stimulation with all Lactobacillus strains, we observed a reduction of the basal oxidative stress in HeLa cells that could be related to modifications in physical properties and organisation of the plasma membrane. These results confirm the strictly strain-specific peculiarities of Lactobacillus and deepen the understanding of the mechanisms underlying the health-promoting role of this genus within the vaginal ecosystem.

Parental, F1, F2, BC1, BC2, BC11, BC12, BC21, BC22, BC1 self-pollinated and BC2 self-pollinated generations of three crosses involving six cultivars of durum wheat (Triticum turgidum var. durum) were studied for flag leaf length under normal and late sown environments to analyse the nature of gene effects. For most crosses the 10-parameter model was the best descriptor of the data to account for the variability in generation means of this trait but in cross HI 8062×JNK-4W-128 the 6-parameter model was the best (normal sown) while in cross Raj 911×DWL 5002 the 3-parameter model was the best (late sown). Of the epistatic interactions, dominance×dominance (l) and dominance×dominance×dominance (z) played significantly greater roles in controlling the inheritance of this trait. Absolute totals of non-fixable gene effects were much higher than the fixable gene effects in all the crosses in both the environments, indicating the greater roles of non-additive effects in controlling the inheritance of flag leaf length in durum wheat cultivars. Significant partial dominance was observed frequently in most of the crosses. Significant heterosis was attributed to combined effects of dominance×dominance (l), additive×additive×dominance (x) and dominance×dominance×dominance (z) epistatic interactions in the cross Cocorit 71×A-9-30-1 under late sown environment. Biparental mating and/or diallel selective mating, which exploit both fixable and non-fixable components, have been suggested for the improvement of this trait in durum wheat cultivars.

ABSTRACT This report describes the distribution of conventional nuclear localization sequences (NLS) and of a beta-stranded so-called nuclear localization motif (NLM) in the two proteins (VP1, 82 kDa; VP2, 63 kDa) forming the T=1 icosahedral capsid of the parvovirus minute virus of mice (MVM) and their functions in viral biogenesis and the onset of infection. The approximately 10 VP1 molecules assembled in the MVM particle harbor in its 142-amino-acid (aa) N-terminal-specific region four clusters of basic amino acids, here called BC1 (aa 6 to 10), BC2 (aa 87 to 90), BC3 (aa 109 to 115), and BC4 (aa 126 to 130), that fit consensus NLS and an NLM placed toward the opposite end of the polypeptide (aa 670 to 680) found to be necessary for VP2 nuclear uptake. Deletions and site-directed mutations constructed in an infectious MVM plasmid showed that BC1, BC2, and NLM are cooperative nuclear transport sequences in singly expressed VP1 subunits and that they conferred nuclear targeting competence on the VP1/VP2 oligomers arising in normal infection, while BC3 and BC4 did not display nuclear transport activity. Notably, VP1 proteins mutated at BC1 and -2, and particularly with BC1 to -4 sequences deleted, induced nuclear and cytoplasmic foci of colocalizing conjugated ubiquitin that could be rescued from the ubiquitin-proteasome degradation pathway by the coexpression of VP2 and NS2 isoforms. These results suggest a role for VP2 in viral morphogenesis by assisting cytoplasmic folding of VP1/VP2 subviral complexes, which is further supported by the capacity of NLM-bearing transport-competent VP2 subunits to recruit VP1 into the nuclear capsid assembly pathway regardless of the BC composition. Instead, all four BC sequences, which are located in the interior of the capsid, were absolutely required by the incoming infectious MVM particle for the onset of infection, suggesting either an important conformational change or a disassembly of the coat for nuclear entry of a VP1-associated viral genome. Therefore, the evolutionarily conserved BC sequences and NLM domains provide complementary nuclear transport functions to distinct supramolecular complexes of capsid proteins during the autonomous parvovirus life cycle.

Des inhibiteurs du complexe bc1 de la chaîne respiratoire mitochondriale ont été développés comme agents antimicrobiens pour lutter contre des pathogènes de l'Homme et de plantes. Ces drogues ciblent les poches catalytiques Qo et Qi formées par le cytochrome b. La comparaison de séquences de cette protéine montre que les sites Qo et Qi sont bien conservés entre les organismes mais qu'il existe toutefois des variations qui pourraient expliquer leur différence de sensibilité aux drogues. A l'aide du modèle levure S. cerevisiae, nous avons étudié les déterminants de la résistance/sensibilité naturelle à deux antipaludiques se liant au site Qo de Plasmodium: l'atovaquone et RCQ06. Nous avons notamment montré que le résidu 275 joue un rôle clé dans ce phénomène. Une approche similaire est actuellement utilisée pour identifier les facteurs de la sensibilité différentielle à deux drogues ciblant le site Qi des oomycètes. Malheureusement, des cas de résistance acquise à ces antimicrobiens ont été rapportés et ont pour origine des mutations dans le cytochrome b. De ce fait, de nouvelles molécules sont requises pour court-circuiter ces résistances. Au cours de ma thèse, nous avons mis au point un test qui permet de cribler des molécules capables d'inhiber la fonction respiratoire. Nous avons ainsi pu identifier un nouvel inhibiteur du complexe bc1 : D12. Nous avons ensuite déterminé le mode de liaison de cette molécule ainsi que celui d'un composé capable d'inhiber la prolifération de Plasmodium, HDQ, grâce à une collection de mutants des poches catalytiques. HDQ s'est avéré être un inhibiteur du site Qi. Il pourrait être utilisé avec un inhibiteur du site Qo afin de limiter l'apparition de mutations de résistance. D12 est un inhibiteur du site Qo qui est capable notamment de court-circuiter la mutation de résistance à des fongicides du site Qo G143A. Cette dernière a été trouvée chez de nombreux phytopathogènes, mais n'est cependant pas apparue chez des champignons possédant un intron immédiatement après le codon codant pour la glycine 143. En utilisant la levure, nous avons montré que la mutation empêche l'épissage de l'intron en altérant la structure exon/intron. Nous avons également identifié des mécanismes de " by-pass " qui permettent de restaurer la fonction respiratoire du mutant et qui pourraient apparaître chez les pathogènes. Les mutants créés au cours de ma thèse pourront aider à identifier, concevoir et caractériser de nouveaux antimicrobiens et à étudier l'apparition de mutations de résistance.

Toxoplasmosis is a highly prolific and neglected disease, and current treatments fail to adequately address the challenges it presents. As such development of new more efficacious treatments, tailored to the complex nature of the infection, is required. The causative parasite Toxoplasma gondii is a resilient and resourceful organism, adept at invasion, avoidance and survival within its host. The cytochrome bc1 complex has been proven to be a validated target for the treatment of a number of apicomplexan parasitic diseases, including both malaria and toxoplasmosis. Recent work has also identified overexpression of this complex within a strain expressing the difficult to treat bradyzoite phenotype, which supported by findings that treatment with known bc1 inhibitor atovaquone shows reduction in cyst burden, suggests this may be a particularly promising target for an effective systemic parasite treatment. Within discusses the design, synthesis and analysis of a number of cytochrome bc1 inhibitors across several novel scaffolds. Designs focused on improving deficiencies identified in previous inhibitors. Successful synthesis of a number of these designs and biological evaluation identified several submicromolar examples. After preliminary ADMET evaluation of the synthesised scaffolds the tetrahydroquinolone scaffold was selected for further development. Optimisation of the synthesis to the tetrahydroquinolones (THQ's) resulted in access to a library of analogues. From these analogues compound 169 was selected for further analysis, characterisation and ultimately progressed to early stage in vivo assays, in which it demonstrated exceptional potency and efficacy.

Protein-protein interactions (PPIs) play a major role in cellular function, mediating signal processing and regulating enzymatic activity. Understanding how proteins interact is essential for predicting new binding partners and engineering new functions. Mutational analysis is one way to study the determinants of protein interaction. Traditionally, the biophysical study of protein interactions has been limited by the number of mutants that could be made and analyzed, but advances in high-throughput sequencing have enabled rapid assessment of thousands of variants. The Keating lab has developed an experimental protocol that can rank peptides based on their binding affinity for a designated receptor. This technique, called SORTCERY, takes advantage of cell sorting and deep-sequencing technologies to provide more binding data at a higher resolution than has previously been achievable. New computational methods are needed to process and analyze the high-throughput datasets. In this thesis, I show how experimental data from SORTCERY experiments can be processed, modeled, and used to design novel peptides with select specificity characteristics. I describe the computational pipeline that I developed to curate the data and regression models that I constructed from the data to relate protein sequence to binding. I applied models trained on experimental data sets to study the peptide-binding specificity landscape of the Bc1-xL, Mc1-1, and Bf1-1 anti-apoptotic proteins, and I designed novel peptides that selectively bind tightly to only one of these receptors, or to a pre-specified combination of receptors. My thesis illustrates how data-driven models combined with high-throughput binding assays provide new opportunities for rational design.

Mitochondria contain their own genetic system, encoding key subunits of the oxidative phosphorylation system. These subunits are expressed by an organelle-specific gene expression machinery. This work revealed a number of fundamental aspects of mitochondrial gene expression and provides evidence that this process is organized in a unique and organelle-specific manner which likely evolved to optimize protein synthesis and assembly in mitochondria. Most importantly, improving the experimental handling of ribosomes we could show that mitochondrial ribosomes are organized in large assemblies that we termed MIOREX complexes. Ribosomes present in these complexes organize gene expression by recruiting multiple factors required for post-transcriptional steps. In addition, we could reveal mechanisms by which ribosome-interactor complexes modulate and coordinate the expression and assembly of the respiratory chain subunits. For example we showed that the Cbp3-Cbp6 complex binds to the ribosome in proximity to the tunnel exit to coordinate synthesis and assembly of cytochrome b. This location perfectly positions Cbp3-Cbp6 for direct binding to newly synthesized cytochrome b and permits Cbp3-Cbp6 to establish a feedback loop that allows modulation of cytochrome b synthesis in response to assembly efficiency. Likewise the interaction of the membrane-anchor proteins Mba1 and Mdm38 with the tunnel exit region enables them to participate in the translation of the two intron-encoding genes COX1 and COB in addition to their role in membrane insertion.  In summary, work presented in this thesis shows that mitochondrial gene expression is a highly organized and regulated process. The concepts and technical innovations will facilitate the elucidation of many additional and important aspects and therefore contribute to the general understanding of how proteins are synthesized in mitochondria.

In recent years investigation of the biological mode of action of the antimalarial compound atovaquone has validated the Plasmodium falciparum bC1 protein complex as a target for the development of novel antimalarials. This project is concerned with the design, synthesis and development of quinolone based antimalarial compounds targeting the Qo site of the Plasmodium falciparum bC1 protein complex. -......, ..•. : - A small array of quinolone-based compounds, substituted at the 3 and 7 positions, has been developed utilising the Gould-Jacobs methodology. The synthesised compounds express activity as low as 0.46 nM versus Plasmodium falciparum malaria parasites in vitro. Activity at the bC1 protein complex has been confirmed and selectivity for the parasite protein over the mammalian equivalent has been demonstrated. Poor solubility has been highlighted as a potential issue; however steps have been taken towards the ,/ development of carbamate prod rugs with encouraging results. Docking studies performed in silico, using the crystallised structure of yeast bC1 protein, demonstrate a key role for residues His181 and Glu272 in the recognition of high potency inhibitors and have aided in the rationalisation of the observed biological activities. The orientation of the quinolone group in the active site and interaction of the inhibitors with the hydrophobic channel leading to the active site have both been shown to be of significance for biological potency. Computer lead design of a new chemotype has resulted in the development of a second array of quinolone based compounds synthesised by a divergent method based on the Huisqen 1,3-dipolar cycloaddition. Docking studies predicted the synthesised compounds to be potent inhibitors of Plasmodium falciparum bC1. However, in vitro results for the array proved disappointing, an outcome attributed to a failure to account for membrane permeability and the physical location of the targeted bC1 protein complex.

The aim of this work was to study the inheritance of the total tocopherol content (TTC) in backcrosses of sunflower breeding lines. Inheritance in the BC1 generation with a highly significant parent is carried out according to the type of intermediate and overdominance (h/d from -0.53 to 2.81), with low significance – according to the type of intermediate and negative overdominance (h/d from -0.59 to -1, 48). A significant positive correlation between the values of the recurrent parent and BC1, equal to 0.8, indicates the observed maternal effect. In the I1BC1 generation, positive and negative transgression was found.

A associação de diferentes espécies de bactérias endofíticas com a rizosfera das plantas hospedeiras podem estimular o crescimento, desenvolvimento e aclimatação, oferecendo maior quantidade de mudas, além de reduzir o ciclo, proporcionando retorno econômico ao produtor. O objetivo deste estudo foi avaliar o efeito da introdução de quatro isolados bacterianos endofíticos produtores de auxina “BA - B. agrestis (10,820 mg mL-1) isolado de plantas de Pera (Pyrus spp.); BC1 - B. cereus cepa 1 (7,812 mg mL-1), BC2 - B. cereus estirpe 2 (5,715 mg mL-1), BT - B. thuringiensis (17.926 mg mL-1) isolados da cultivar de bananeira Prata Anã (AAB)” através da inoculação no sistema radicular de três cultivares de bananeira “Prata Anã (AAB), Grande Naine (AAA), BRS Princesa (AAAB)” na fase de aclimatação. O experimento foi realizado entre o meses de Maio a Agosto de 2019 em casa de vegetação sob ambiente protegido no Laboratório de Cultura de Tecidos Vegetais do Departamento de Agricultura (DAG) da Universidade Federal de Lavras (UFLA), localizada no município de Lavras, Minas Gerais, Brasil. As características morfológicas relacionadas ao desenvolvimento das plantas (altura total e diâmetro do pseudocaule) foram avaliados durante todo o período de aclimatação. Após 90 dias de transplante e aclimatação, foram avaliados o comprimento da raiz, número de folhas, massa seca (raiz, pseudocaule, folha), área foliar, concentração de carbono interno, condutância estomática, taxa de fotossíntese, taxa de transpiração, temperatura foliar e clorofila. As bactérias apresentaram resultados diferentes em relação as cultivares estudadas. Considerando a morfologia e características fisiológicas observadas neste estudo, a bactéria B. thuringiensis para as cultivares Prata Anã e Grande Naine e a bactéria B. agrestis para a cultivar BRS Princesa são recomendados para o processo de aclimatação das mudas de banana, pois estimularam o crescimento das plantas, aumentando a massa seca, além de promover o crescimento das raízes. Dessa forma, melhoraram os aspectos fisiológicos e reduziram o período de aclimatação das plantas de bananas.

Objective: The aim of this study is to compare the effects of resistance training (RT) on muscle strength (MS), body composition (BC), and anxiety indicators in eutrophic, overweight, and obese breast cancer survivors (BCS). Methodology: Twentysix BCS women (56.07±6.94 years old; body mass index [BMI]: 68.29±11.18 kg), who were undergoing hormone therapy, performed 8 weeks of RT once a week. The BCS were divided into three groups as follows: eutrophic (BMI ≤ 24.4 [23.72±1.41], n=9), overweight (BMI 25–29.9 [27.21±1.26], n=9), and obese (BMI ≥ 30 [33.37±2.06], n=8). For the MS, the 10-repetition maximum test (normalized 10-RM/BM) on the bench press (BP) and the leg press (LP); for BC, the dual-energy X-ray absorptiometry; and for anxiety, the state-trait anxiety inventory were performed. For overtime analysis, the paired t-test for MS and BC and the Wilcoxon test for anxiety indicators were conducted. For analyses between groups, the mean difference (∆ [post-baseline]) was calculated using the one-way analysis of variance for MS and BC and the Kruskal– Wallis test for anxiety indicators. Results: There were no differences between the groups. All groups improved on the BP (p≤0.001) and the LP (p<0.001) overtime (eutrophic [BP, ∆=0.050±0.03; LP, ∆=0.401±0.10], overweight [BP, ∆=0.069±0.03; LP, ∆=0.406±0.15], and obese [BP, ∆=0.037±0.02; LP, ∆=0.375±0.11]). In the BC analyses, it was verified and improved only for the eutrophic BCS in the fat percentage (∆=-1.122±1.11, p=0.016) and lean mass (∆=0.650±0.78, p=0.036). There was a significant reduction in the state-trait anxiety in the eutrophic (∆=-7.444±8.13, p=0.030) and obese (∆=-9.125±9.70, p=0.042) groups. Conclusions: All BCS groups improved their MS. The eutrophic BCS may have a better response in BC compared with overweight and obese BCS. With regard to state-trait anxiety, the eutrophic and obese BCS groups showed better response than the overweight BCS group. The results suggest that the eutrophic BCS can present improvements in more components with a weekly session of RT; however, further studies should be performed.

The implementation of the INR system has been assessed in a group of laboratories from over fifty countries. These centres participate in the World Health Organisation International Quality Control Programme in Blood Coagulation. Prothrombin times are performed on various lyophilised human coumarinised plasmas using both the local thromboplastin reagent and British Comparative Thromboplastin (BCT) reference preparation. During 1985 and 1986 a programme of education has been undertaken whereby instructions in the derivation of INR values have been given. The calculation of INR has been shown by means of a worked example and a chart for the conversion of the prothrombin ratio with BCT to INR was included. Prior to this, prothrombin results were expressed as British Corrected Ratios (BCR) with a minority of centres using INR. In 1984 the CV varied between 23.7% - 36.8% for results with local reagents, but between 8.2% - 14.3% with BCT. This suggested that conversion into INR/ BCR was unsatisfactory. The table shows the impact of the continuing educational programme.The number of laboratories expressing results as INR has steadily increased and the CV is now approaching that obtained with BCT. The improvement in CV suggests that the general application of the INR system will help improve inter-laboratory standardisation of oral anticoagulant control and should be encouraged.

1. Abstract: Over the past 25 years, vine cacti of the genera Hylocereus and Selenicereus have been introduced into Israel and southern California as new exotic fruit crops. The importance of these crops lies in their high water use efficiency and horticultural potential as exotic fruit crops. Our collaboration focused on the cytological, molecular and evolutionary aspects of vine cacti polyploidization to confront the agricultural challenge of genetic improvement, ultimately to improve success of vine cacti as commercial fruit crop plants. More specifically, we worked on the: 1- Identification of the putative ancestor(s) of the tetraploid H. megalanthus; 2- Determination of the number of origins of H. megalanthus (single vs. multiple origins of polyploidy); 3- Cytogenetic analysis of BC1 and F1 hybrids; 4- Determination of important agricultural traits and the selection of superior hybrids for cultivation. The plant material used in this study comprised interspecific Hylocereus F1 and first backcross (BC1) hybrids, nine Hylocereus species (58 genotypes), nine Selenicereus species (14 genotypes), and four Epiphyllum genotypes. Two BC1 hexaploids (BC-023 and BC-031) were obtained, a high ploidy level that can be explained only by a fertilization event between one unreduced female gamete from the triploid hybrid and a balanced gamete from the pollen donor, the diploid H. monacanthus. These findings are scientific evidence that support the possibility that “hybridization followed by chromosome doubling” could also occur in nature. Cytomixis, the migration of chromatin between adjacent cells through connecting cytoplasmatic channels, was observed in vine cacti hybrids and may thus imply selective DNA elimination in response to the allopolyploidization process. Evidence from plastid and nrDNA internal transcribed spacers (ITS) sequences support the placement of H. megalanthus within a monophyletic Hylocereus group. Furthermore, both plastid and ITS datasets are most consistent with a conclusion that this tetraploid species is an autopolyploid, despite observations that the species appears to be morphologically intermediate between Hylocereus and Selenicereus. Although the possibility of very narrow allopolyploidly (i.e., derivation from parents that are barely diverged from each other such as closely related species in the same genus) cannot be ruled out entirely based on our data (in part due to the unavailability of Hylocereus species considered to be morphologically the closest relatives of H. megalanthus), the possibility of H. megalanthus representing an intergeneric cross (i.e., Hylocereus × Selenicereus) seems extremely unlikely. Interestingly, the process of homogenization of ITS sequences (concerted evolution) is either incomplete or lacking in both Hylocereus and Selenicereus, and the inclusion of several artificial hybrids in the molecular study revealed the potential for biparental plastid inheritance in Hylocereus. The most important agricultural implication of this research project was the information collected for F1 and BC1 hybrids. Specifically, this project concluded with the selection of four superior hybrids in terms of fruit quality and potential yields under extreme high temperatures. These selected hybrids are self-compatible, avoiding the need for hand cross pollination to set fruits, thus reducing manpower costs. We recently offered these hybrids to growers in Israel for prioritized rapid evaluation and characterization.
