Повний текст джерелаCell-penetrating peptides (CPPs) are short, cationic or amphipatic peptides, usually containing less than 30 amino acids, which are able to deliver various bioactive cargoes inside cells. Among the many CPPs described in the literature, the arginine-rich peptides have attracted particular attention. Several chemical modifications of the peptide backbone leading to different spatial distributions of the CPP functional groups, or the introduction of aliphatic chains have been made to enhance their internalization efficiency. The aim of this work was the synthesis of new cyclic CPPs containing a minimal cationic domain and their functionalisation with an aliphatic chain. We have synthesised a small library of fifteen new cyclic carriers including the classical CPPs Penetratin and R6W3 using native chemical ligation (NCL) in solution. The introduction of an N-terminal Cys residue and of a C-terminal thioester (or precursor) in the initial linear peptide sequence allowed the head-to-tail cyclisation. The efficiency of cargo delivery in CHO cells was measured by MALDI-TOF mass spectrometry. We found that cyclisation of CPPs improved their internalisation efficiency mostly by glycosaminoglycan-dependent endocytosis. Among the whole series of lipopeptides tested in this project, two sequences are distinguished by their remarkable ability to cross cellular membranes: the peptides [C12-R4] and [C12-R7]

Повний текст джерелаThe first part of this thesis gives a description of invariant subspaces for the backward shift generated by vector valued lacunary series and by a class of lacunary power series in H²(X), (where X is an Hilbert space). In particular, we show that these series f in H²(X) are cyclic vectors if and only if the queue of Taylor coefficients {f^(k), k>N} generates the whole space X. Analogs of this result are obtained for some functions whose spectrum is a finite union of lacunary sequences and in the polydisc. In the scalar case H², we give the criterion to have cyclicity of lacunary series for any power of the backward shift. In the second part, we prove in the vector-valued spaces L²(ℝ+, X) (where X is a finite dimensional Hilbert space) the cyclicity for the semi-group of left translations of some particular functions with support included in a lacunary system of intervals

Повний текст джерелаThis thesis is devoted to the study of uncertainty principles and approximation problems in Fourier analysis. It consists two parts.The first part focus on uncertainty principles in Fourier analysis. Using a technique recently invented by Avi Wigderson and Yuval Wigderson, we give a new proof of the classical Heisenberg uncertainty principle, hence answering several questions affirmatively posed by Wigderson & Wigderson. Also, we obtain some other new generalization on uncertainty principle, which illustrates the power of the new method.The second part is about approximation on weighted sequence spaces. We generalize an old result due to Douglas, Shapiro and shields on cyclic vectors of shift operator in sequence spaces, which asserts that if an element in l^2 spaces has a ``sparse" spectrum, then its shifts can not be concentrated on a proper subset, hence they must spread out in the whole space. This phenomenon can also be roughly considered as an uncertainty principle, and it is also true for p greater than 2 and false for 1
