Hillbilly Elegy has had a profound impact on contemporary views and opinions of Appalachia. In this review, the authors discuss this impact and provide key critiques that help readers make sense of the generalizations made in the book by placing Hillbilly Elegy in conversation with other contemporary Appalachian scholarship. The authors conclude that J. D. Vance has a right to tell his story but telling the story of the Appalachian people is more complex and nuanced than Vance acknowledges, and the authors caution readers to consider this when reading Hillbilly Elegy.

In 2016, J. D. Vance, a previously unknown author, surprisingly hit the bestseller’s list with his memoir, Hillbilly Elegy: A Memoir of a Family and Culture in Crisis. However, historically scholars have pushed back against the stereotypes that Vance portrays and have argued for a narrative that embraces the diversity of the Appalachian region. Using content analysis, this research study investigated the frequency and context of key terms related to Appalachia across the K-12 social studies standards of states labeled as being part of the region by the Appalachian Regional Commission (ARC). Results illustrate the lack of inclusion of these terms and that, when included, they do not represent the rich history or culture of the region. These results suggest that researchers, teacher educators, and teachers should consider the ways in which standards define curriculum on topics such as Appalachia and how these definitions interact with the powerful narratives being portrayed within our mainstream democracy. Guidance and suggestions for disrupting Appalachian stereotypes within social studies education are provided.

Cleveland digests, comparative in vitro kinase assays, secondary transfections, and immunoprecipitations indicated that focus formation was caused by rare transfection events that resulted in very high-level pp60c-src expression rather than by mutations of the transfected c-src genes. These results suggest that pp60v-src induced transformation is not a completely spurious activity which is unrelated to the function of pp60c-src but that it represents a perturbation of already existent molecular control processes involving pp60c-src.

Turbin angin adalah salah satu alternatif untuk mengurangi beban listrik di wilayah perkotaan. Di wilayah perkotaan terdapat gedung bertingkat dengan jumlah yang cukup banyak. Sehingga menjadi lokasi yang tepat untuk aplikasi turbin angin sekaligus mengurangi beban listrik. Tipe turbin yang tepat untuk aplikasi gedung bertingkat adalah turbin angin sumbu vertikal (VAWT). Salah satu jenis VAWT adalah turbin Savonius. Turbin angin Savonius konvensional memiliki kinerja yang rendah seperti koefisien daya dan torsi yang rendah dibandingkan dengan turbin angin jenis lain. Ini terjadi karena aliran angin dapat menyebabkan tekanan negatif pada salah satu sisi sudu. Untuk mengatasi masalah ini, turbin angin Savonius konvensional dikombinasikan dengan guide vane. Tujuan dari penelitian ini adalah untuk mempelajari pengaruh guide vane terhadap performa dan karakteristik pola aliran sekitar turbin angin Savonius. Model numerik dihitung menggunakan persamaan Navier-Stokes dengan model turbulen k-ε standar. Analisa menggunakan software ANSYS-Fluent R15. Simulasi dilakukan pada arah angin yang berbeda, antara lain: 0o, 30o, 60o pada kecepatan angin 2 m/s. Hasil penelitian menunjukkan bahwa guide vane mampu menambah laju aliran udara yang menuju sudu turbin dan meningkatkan performa turbin angin Savonius sebesar 22,2%. In this ever-expanding world, researchers are focused on changing how we consume energy. Progress has been made over time, but the devices such as low-temperature polymer electrolyte fuel cells and metal-air batteries that can replace the conventional fossil fuel engines still use noble metal catalysts,1 which hinders the mass production and long-term usage of the devices. Transition metal macrocyclic (MN4-type) catalysts are good alternatives for the noble metal catalysts due to their low-price and promising electrocatalytic activity.2 Thus, the aim of this study was to develop the bifunctional ORR/OER electrocatalysts using mixed transition metal phthalocyanine-modified multi-walled carbon nanotubes (MWCNT) by simple pyrolysis method.3 Among the prepared bifunctional catalysts, FeCoN-MWCNT showed the superior electrocatalytic ORR activity in alkaline media with half-wave potential of 0.86 V vs RHE and FeNiN-MWCNT catalyst exhibited the exceptional OER activity with E j=10 mA cm–2 of 1.58 V (see Fig. 1a, b). Physical characterization of the prepared catalysts was investigated with (scanning) electron microscopy, X-ray diffraction, X-ray photoelectron spectroscopy and Raman spectroscopy. Further, catalysts were tested in anion-exchange membrane fuel cells (AEMFCs) as cathode material and very good peak power density of 692 mW cm–2 was obtained for FeCoN-MWCNT (see Fig. 1c). FeNiN-MWCNT was tested in anion-exchange membrane electrolyzer (AEMEL) and showed comparable results with state-of-the-art RuO2 catalysts (Fig. 1d). L-valine L-valinium perchlorate monohydrate single crystal has been synthesized by slow evaporation technique at room temperature. The crystal structure and space group of the crystal were confirmed by single crystal X-ray diffractometer. Optical behavior of the crystal was analyzed by using UV-visible spectrophotometer. Thermal stability was discussed by using thermo gravimetric analysis. Mechanical strength of the grown crystal was studied using Vickers microhardness tester. Nonlinear optical property was explored to confirm the second harmonic generation efficiency of the grown crystal. As our society seeks increased energy security and resilience, the need for efficient, cost-effective, carbon-free hydrogen generation will continue to grow. Proton exchange membrane (PEM) water electrolysers are a promising technology to produce “green” hydrogen, however, at present, their large-scale commercialization is hindered by the high capital costs of critical components. The anodic membrane electrode assembly requires large quantities of iridium (Ir) to facilitate the kinetically demanding oxygen evolution reaction (OER). Owing to its scarcity, iridium is a severe bottleneck and expense in the widespread implementation of these technologies. The iridium content in electrolyser anodes can be lowered substantially by designing catalysts which consist of finely dispersed nanoparticles of iridium dioxide affixed onto suitable support materials. This approach enhances the active surface area and operational lifetime of these electrocatalysts towards the OER while utilising a fraction of the otherwise required iridium. To reap these benefits, the support needs to be a high surface area, corrosion-resistant material that is a good electronic conductor1,2. Metal oxides have dominated as oxygen evolution support materials for PEM electrolysis; however, they typically need to be doped with an impurity to demonstrate sufficient electronic conductivity. Iridium oxides supported on antimony−doped tin oxide (ATO) support have been found to behave as high-performing OER electrocatalysts in acidic conditions3,4. While there is no doubt that existing studies prove a link between the presence of ATO support and the improved performance of ATO-supported iridium oxides versus unsupported catalysts, an in-depth characterisation of the charge transfer between the ATO support and deposited IrO2 nanoparticles could prove useful in understanding the reason for these performance enhancements on the basis of the electronic properties of the catalyst and the support. In this study, we investigated the performance of highly dispersed IrO2 nanoparticles, supported on doped tin oxide supports, consisting of various metallic dopants and iridium loadings. The electronic interactions between the Ir-phase and the support were studied by measuring the valence band spectra of the prepared materials with X-ray and ultraviolet photoelectron spectroscopy, and Hall measurements were utilised to understand the nature of electronic conductivity in the prepared materials. The uncovered physicochemical and electronic characteristics are related to the electrochemical performance of these materials towards the OER. These novel findings contribute insights which can be used as the basis for the rational design of next-generation oxide-supported Ir-based catalysts with tunable electronic properties for sustainable PEM electrolysis applications. Proton exchange membrane fuel cells (PEMFCs), in their low temperature (LT, 60-80 °C) and high temperature (HT, 120-200 °C) variants, can be used for broad applications in the automotive and stationary sector. However, the most commonly used PEMFC catalyst, consisting of platinum nanoparticles supported on a carbon black, suffers from degradation under the relevant working conditions e.g. low pH environment and potentials in the range of 0.6-1.5 V vs. RHE. These processes include carbon corrosion, Pt dissolution and detachment as well as agglomeration.[1-3] Especially, carbon corrosion leads to an increase of the support hydrophilicity, decreased conductivity and loss of Pt particles resulting in an overall performance loss of PEMFCs.[1] Different studies already reported a positive effect of metal oxide-carbon composite supports using TiO2-Vulcan®XC-72[1,2], SnO2-Vulcan®XC-72[2,3], or fluorine-doped SnO2 on reduced graphene oxide[4] on the carbon and Pt stability. Moreover, Ruiz Camacho et al. showed higher ORR activity for Pt/TiO2-Vulcan and Pt/SnO2-Vulcan compared to Pt/C and a lower potential for CO oxidation during stripping experiments.[2] This can be beneficial for PEMFC operation with reformate. To further push the activity and stability of Pt/metal oxide-carbon catalysts homogeneous distribution of metal oxide and Pt nanoparticles is necessary. In contrast to previous studies, that uses low surface area Vulcan®XC-72[1-3] the implementation of high surface area Black Pearls (BPs) can enable more homogenous distribution of metal oxides and Pt nanoparticles which can positively impact the activity. In this comparative study, Pt/metal oxide-carbon catalysts using SnO2 and TiO2 nanoparticles on BPs are analyzed towards their physical properties and electrochemical ORR activity and stability. Metal oxide/carbon composites were fabricated by deposition of 50 wt.% commercial SnO2 or TiO2 nanoparticles on Black Pearls® 2000. Thermogravimetric analysis (TGA) reveals the successful deposition of metal oxides TiO2 (41 wt.%) and SnO2 (47 wt.%) on BP. Next, deposition of 40 wt.% Pt nanoparticles with diameters between 1-2 nm on the metal oxide-BP composites is done. Transmission electron microscope (TEM) images display successful deposition of Pt with uniform distribution of Pt for both composite catalysts in Figure 1 a) and c). The elemental mapping of Pt and Sn or Ti, using scanning TEM with energy dispersive spectroscopy (EDS) for analysis of the interaction between metal oxide displays homogenous distribution of Pt over the metal oxide-BP supports (Fig 1, b), d)). In the case of Pt/SnO2-BP also uniform Sn distribution is observed whereas for Pt/TiO2-BP partial agglomeration of TiO2 is found. Further analysis of Pt and metal oxides will be given using high resolution-TEM for analysis of lattice distance and ICP-MS for determination of Pt content. Moreover, electrochemical characterization using rotating ring disc electrode will be carried out for comparison of ORR activity and selectivity. Furthermore, an accelerated stress test including 5000 cycles in the range of 0.6-1.5 V vs. RHE in N2-saturated 0.1 mol L-1 HClO4 is applied to analyze the overall catalyst stability. IL-37 is also one of the IL-1 cytokines activated by caspase-1 acting as natural inhibitor of inflammation (3).ObjectivesIL-37 has pathogenetic roles for certain inflammatory diseases. We aimed to investigate serum IL-37 levels and its relationship with clinical and laboratory features of disease.Methods58 adult patients diagnosed with FMF according to Tel Hashomer criteria were included. Thirty subjects were served as healthy control subjects. Demographic, genetic, clinical and laboratory features and treatment responses of patients were recorded. Twenty-nine patients were colchicine responsive whereas 29 were colchicine refractory. Serum IL-37 levels were measured by ELISA from blood samples obtained at attack free periods.ResultsPeritonitis was the most common attack type (81%) followed by fever (80%) and arthritis (67%). There was no difference between FMF patients and healthy subjects for their serum IL-37 levels. A negative correlation was found between IL-37 values ​​and erythrocyte sedimentation rate in FMF patients (r: -0,31; p:0.015). IL -37 level was found to be significantly lower in patients who suffer from arthritis (median [IQR] 119 [396] ng/L vs 53 [164] ng/L, p= 0.03), myalgia (147 [364] ng/L vs 53 [84] ng/L, p= 0.05) or skin rash (102 [360] ng/L vs 54 [130] ng/L, p= 0.05) compared to those who did not have these attacks.ConclusionAlthough there was no difference in serum IL-37 levels between FMF patients and healthy subjects, IL-37 seem to be associated with musculoskeletal and skin attacks of FMF. Background:OMERACT recommends three “core set” MRI sequences with an optional cartilage-dedicated sequence to perform Rheumatoid Arthritis (RA) MRI scoring (RAMRIS) (1). Dixon method allows the production of four different images from a single MRI sequence.Objectives:To test a short MRI protocol based on a single Dixon sequence to assess disease activity and cartilage in hands of patients with early RA.Methods:Twenty-four patients (16 women, mean age 45.7 years old) with early DMARD-naive RA meeting the ACR/EULAR 2010 criteria were prospectively included. Both hands of each patient were imaged with MRI including contrast-enhanced T1-weighted Dixon and OMERACT “core set” MRI sequences and with conventional radiography.Two musculoskeletal radiologists (R1 and R2) separately assessed disease activity according to RAMRIS scoring system based on the Dixon images (contrast-enhanced T1-weighted Dixon water-only images to score synovitis, tenosynovitis and osteitis and contrast-enhanced T1-weighted Dixon fat-only images to score erosions) and the three OMERACT “core set” MRI sequences (contrast-enhanced fat-saturated T1-weighted images to score synovitis and tenosynovitis, fat-saturated T2-weighted images to score osteitis and T1-weighted images without contrast-material injection to score erosions).One radiologist (R1) separately measured the thickness of the cartilage in the joints corresponding to those assessed by the Sharp/van der Heijde modified scoring method on contrast-enhanced T1-weighted Dixon out-of-phase images and radiographs (2).RAMRIS scoring and measurement of the cartilage thickness were repeated by R1 to assess intra-observer agreement. Statistical analysis was based on intra-class correlation coefficients (ICC) with 95% confidence interval to assess inter-technique, intra-observer and inter-observer agreement. The strength of agreement was interpreted as follows: ≤0, poor; 0.01-0.20, slight; 0.21-0.40, fair; 0.41-0.60, moderate; 0.61-0.80, substantial and ≥0.81, excellent.Results:Agreement between total RAMRIS scores obtained with the Dixon water- and fat-only images and total RAMRIS scores obtained with the OMERACT sequences was excellent for R1 (0.94; 0.86-0.97) and R2 (0.91; 0.81-0.96). Intra-observer agreement was excellent with Dixon images (0.97; 0.92-0.98) and OMERACT sequences (0.96; 0.90-0.98). Inter-observer agreement was excellent with Dixon images (0.92; 0.82-0.96) and OMERACT sequences (0.93; 0.85-0.97).Agreement between the measures of cartilage thickness on the Dixon out-of-phase images and the measures of cartilage thickness on radiographs was substantial (0.71; 0.66-0.75). Intra-observer agreement was excellent with Dixon out-of-phase images (0.94; 0.93-0.95) and radiographs (0.93; 0.92-0.94).Conclusion:An MRI protocol based on a single contrast-enhanced T1-weighted Dixon sequence allows reproducible RAMRIS scoring and measurement of the cartilage thickness. It served as a forum to disseminate the latest developments in relevant topics such as Advanced Batteries, Fuel Cells, Photoelectrochemical Cells, Hydrogen Production and Storage, among others. The Event was hosted by the Mexican Hydrogen Society (MHS). Founded in 1999, the MHS is nowadays the leading organization within Mexico bringing together Research Groups, Government Agencies and Industry partners to promote R&D related to hydrogen technologies. Plenary lectures were delivered by Prof. Nicolas Alonso-Vante from University of Poitiers (France), Prof. Perla B. Balbuena from Texas A&M University (USA), Prof. Bruce Parkinson from University of Wyoming (USA), Prof. Krishnan Rajeshwar from University of Texas At Arlington (USA), Prof. James J. ABSTRACT We previously produced a heavy-chain-only antibody (Ab) VH domain (VHH)-displayed phage library from two alpacas that had been immunized with ricin toxoid and nontoxic mixtures of the enzymatic ricin toxin A subunit (RTA) and binding ricin toxin B subunit (RTB) (D. J. Vance, J. M. Tremblay, N. J. Mantis, and C. B. Shoemaker, J Biol Chem 288:36538–36547, 2013, https://doi.org/10.1074/jbc.M113.519207 ). Initial and subsequent screens of that library by direct enzyme-linked immunosorbent assay (ELISA) yielded more than two dozen unique RTA- and RTB-specific VHHs, including 10 whose structures were subsequently solved in complex with RTA. To generate a more complete antigenic map of ricin toxin and to define the epitopes associated with toxin-neutralizing activity, we subjected the VHH-displayed phage library to additional “pannings” on both receptor-bound ricin and antibody-captured ricin. We now report the full-length DNA sequences, binding affinities, and neutralizing activities of 68 unique VHHs: 31 against RTA, 33 against RTB, and 4 against ricin holotoxin. Epitope positioning was achieved through cross-competition ELISAs performed with a panel of monoclonal antibodies (MAbs) and verified, in some instances, with hydrogen-deuterium exchange mass spectrometry. The 68 VHHs grouped into more than 20 different competition bins. The RTA-specific VHHs with strong toxin-neutralizing activities were confined to bins that overlapped two previously identified neutralizing hot spots, termed clusters I and II. The four RTB-specific VHHs with potent toxin-neutralizing activity grouped within three adjacent bins situated at the RTA-RTB interface near cluster II. These results provide important insights into epitope interrelationships on the surface of ricin and delineate regions of vulnerability that can be exploited for the purpose of vaccine and therapeutic development.

Pumped-storage power technology is currently the only available energy storage technology in the grid net, and its reliability is receiving attention increasingly. However, when a pump-turbine unit undergoes runaway transitions, hydraulic fluctuations intensively affect the reliable operation of a pumped-storage power station. To reduce hydraulic fluctuations, this study investigated the formation mechanism of hydraulic fluctuations and explored its influence factors. In this study, a developed one-dimensional and three-dimensional (1 D-3D) coupling simulation method was adopted. Transient runaway transitions of a pump-turbine with three different inertias (0.5 J, 1 J, and 2.0 J) at three different guide vane openings (21°, 15°, and 12°, respectively) were simulated and compared. The results suggest that, at smaller guide vane openings (15° and 12°), water hammer owing to the increase in rotational speed is the primary unstable issue compared to the pulsation of radial hydraulic exciting forces on the runner. However, at a larger guide vane opening (21°), the latter owing to the back-flow near the runner inlet is the primary unstable issue. Moreover, it is found that a sufficiently large inertia improves the hydraulic fluctuations of the pump-storage power station, particularly in reducing the pulsation of radial hydraulic exciting loads on the runner. The findings of this study provide a valuable reference for determining suitable rotor inertia.

Abstract Background Scant PK data are available with CZA-ATM in combination. Occurrence of ALT/AST elevations are common with ATM, and it is unknown if it is exacerbated by use of CZA-ATM. This analysis of COMBINE sought to describe the popPK of CZA-ATM and to assess the association between ATM exposures and ALT/AST elevations. Methods COMBINE was a Phase I study of 48 healthy subjects aged 18-45 years (NCT03978091). Subjects were enrolled into 1 of 6 Cohorts (Table 1). Drug(s) were administered for 7 D and intensive plasma and urine PK sampling was performed. PopPK models were developed for ceftazidime (CAZ), avibactam (AVI), and ATM. Empirical Bayesian estimates from the ATM PopPK model were used to simulate ATM day 1 (D1) exposures. Associations between D1 ATM exposures and highest observed ALT/AST were assessed with curvilinear regression (CR) and generalized linear models (GLM). Results Of enrolled 48 subjects, 35% had ALT/AST elevations; 94% occurred in ATM/CZA-ATM Cohorts. Two severe study product related ALT/AST AEs were observed with in the continuous infusion (CI) ATM Cohort. All subjects with ALT/ATS elevations were asymptomatic with no other signs of liver injury. In the ATM PopPK model, CZA-ATM administration reduced ATM non-renal clearance (CLNR) by 1 L/hr (16% of total CL) (Table 2). Administration of CAZ-ATM had a negligible effect on total CAZ CL in the CAZ PopPK model and CZA-ATM was not a covariate in final AVI PopPK model. In the CR, no ATM exposure-ALT/AST associations were identified in overall analyses. Associations (R2=0.18-0.22, p< 0.05) between D1 AUC and ALT/AST were observed in CR analyses restricted to the intermittent infusion (II) ATM Cohorts (Figure 1). In GLM, D1 AUC was significantly associated with ALT/AST in the ATM II Cohort analyses. Administration of CZA-ATM was not associated with ALT/AST in the GLM analyses. Figure 1 Conclusion Administration of CZA-ATM was found to reduce ATM CL, resulting in higher daily ATM AUCs, but did not exacerbate AST/ALT elevations relative to ATM alone. The observed association between ATM AUC and ALT/AST elevations in subjects who received II ATM suggest the risks vs benefits of using II of ATM 8 g/daily vs 6 g/daily with CZA should be considered. Use of CI ATM should be used with caution given the 2 severe ALT/ASTs in the CI ATM Cohort. The increasing demand to reduce fuel burn, hence CO2 emissions, from the gas turbine requires efficient diffusion to reduce the system pressure loss in the combustor. However, interactions between prediffuser and combustor can have a significant effect on diffuser performance. For example, the consequence of increased fuel injector flow at a dump gap set using conventional design guidelines has been shown (Walker, A. D., Carrotte, J. F., and McGuirk, J. J., 2007. “Compressor∕Diffuser∕Combustor Aerodynamic Interactions in Lean Module Combustors,” ASME Turbo Expo 2007—Power for Land Sea and Air, Paper No. GT2007-27872) to introduce a destabilizing interaction between fuel injector and upstream components. The present paper concentrates on examining the effects of increased dump gap. Dump gap ratios of 0.8, 1.2, and 1.6 were employed, with each test utilizing the same inlet guide vane, compressor rotor, integrated outlet guide vane (OGV)∕prediffuser, and dump geometry. The flow fraction of compressor efflux entering the combustor cowl was set to be representative of lean combustors (50–70%). Measurements were made on a fully annular rig using a generic flame tube with metered cowl and inner∕outer annulus flows. The results demonstrate that, with fixed cowl flow, as the dump gap increases, component interactions decrease. At a dump gap ratio of 0.8, the proximity of the flame tube influences the prediffuser providing a beneficial blockage effect. However, if increased to 1.2, this beneficial effect is weakened and the prediffuser flow deteriorates. With further increase to 1.6, the prediffuser shows strong evidence of separation. Hence, at the dump gaps probably required for lean module injectors, it is unlikely the prediffuser will be influenced beneficially by the flame tube blockage; this must be taken into account in the design. Furthermore, with small dump gaps and high cowl flow fraction, the circumferential variation in cowl flow can feed upstream and cause OGV∕rotor forcing. At larger dump gaps, the circumferential variation does not penetrate upstream to the OGV, and the rotor is unaffected. The optimum dump gap and prediffuser design for best overall aerodynamic system performance from rotor through to feed annuli is a compromise between taking maximum advantage of upstream blockage effects and minimizing any 3D upstream forcing.
