Tulsi (Holy basil, Ocimum tenuiflorum L., Lamiaceae), native to Asia, has become globalised as the cultural, cosmetic, and medicinal uses of the herb have been popularised. DNA barcoding, a molecular technique used to identify species based on short regions of DNA, can discriminate between different species and identify contaminants and adulterants. This study aimed to explore the values associated with Tulsi in the United Kingdom (UK) and authenticate samples using DNA barcoding. A mixed methods approach was used, incorporating social research (i.e., structured interviews) and DNA barcoding of Ocimum samples using the ITS and trnH-psbA barcode regions. Interviews revealed the cultural significance of Tulsi: including origins, knowledge exchange, religious connotations, and medicinal uses. With migration, sharing of plants and seeds has been seen as Tulsi plants are widely grown in South Asian (SA) households across the UK. Vouchered Ocimum specimens (n = 33) were obtained to create reference DNA barcodes which were not available in databases. A potential species substitution of O. gratissimum instead of O. tenuiflorum amongst SA participants was uncovered. Commercial samples (n = 47) were difficult to authenticate, potentially due to DNA degradation during manufacturing processes. This study highlights the cultural significance of Tulsi, despite a potential species substitution, the plant holds a prestigious place amongst SA families in the UK. DNA barcoding was a reliable way to authenticate Ocimum species.

Ocimum traditionally known as Holy Basil or Tulsi is an available herb plenty across the country. Traditionally, it is used against a number of human diseases. In this present study, the metabolites present in the ethanolic extracts of nine Ocimum taxa, O. tenuiflorum L. (Green and purple type) two morphotypes of O. basilicum L., (Babu and Marua tulsi) two morphotypes of O. gratissimum L. (Ram and Ajowan tulsi) and each one from O. americanum L. (Bon tulsi), O. × africanum Lour. (Lebu tulsi), and O. kilimandscharicum Guerke.grown naturally in Dakshin Dinajpur district, West Bengal, India were identified using Gas chromatography–mass spectrometry (GC-MS). Among the identified metabolites, carbohydrates, aliphatic alcohols, aliphatic acids, fused ring aromatic hydrocarbon, amino acids, phenolic compounds, quinone, steroids, terpenoids and vitamin E were the chief constituents. The occurrence of these metabolites describes the high biological activity of Ocimum species. Furthermore, in vitro antibacterial activities were also identified against four bacterial strains, Staphylococcus aureus (MTCC 96), Bacillus cereus (MTCC 1305), Proteus vulgaris (MTCC 1771) and Escherichia coli (MTCC 2939). Tested bacterial strains were taken from the Institute of Microbial Technology, Chandigarh, India The results indicated that the ethanolic extracts of all the nine Ocimum taxa have satisfactory minimum inhibitory concentration (MIC) values against the tested microorganisms. O. tenuiflorum has the highest (91.03 %) metabolic content whereas O. × africanum has the minimum. This would definitely serve as a scientific basis of the traditional use of basil against human ailments.

The ligand-activated transcription factor peroxisome proliferator-activated receptor (PPAR) has become a major target for type 2 diabetes. It belongs to the nuclear receptor superfamily, which controls the expression of proteins involved in glucose metabolism, lipid metabolism, adipocyte proliferation and differentiation, and insulin sensitivity. Ocimum tenuiflorum, often known as Krishna tulsi, is the most sacred herb in India. It was utilized for a variety of medicinal purposes. Therefore, it is of interest to document the molecular docking analysis data of PARγ modulators from Ocimum tenuiflorum. Four of the twenty substances (rosmarinic acid, permethrin, luteolin, and isosakuranetin) have a considerable binding affinity for the PPARγ. These phytochemicals are a source of potential anti-diabetic medicines.

Ocimum tenuiflorum L. is found throughout semitropical and tropical parts of Southeast Asia. In Nepal, O. tenuiflorum L. is popular with two variants: Krishna Tulsi consisting of purple-colored leaves and Sri Tulsi consisting of green-colored leaves. O. tenuiflorum L. is considered the queen of herbs and is a traditionally and clinically proven medicinal herb for its application and efficacy. However, no commercial pharmaceutical preparations of O. tenuiflorum L. are available using effervescent vehicles. Therefore, the present study aimed to compare the antioxidant activity of leaves from the two varieties of O. tenuiflorum L. and formulate and evaluate the quality parameters of effervescent granules of the potent extract. The antioxidant activity of O. tenuiflorum L. ethanolic extracts was evaluated by DPPH radical scavenging assay at 1, 10, and 100 µg/mL concentrations, where ascorbic acid was used as the positive control. The antioxidant activity of purple-leafed O. tenuiflorum L. was found to be higher than that of green-leafed O. tenuiflorum L. Therefore, effervescent granules of the ethanolic extract of purple-leafed O. tenuiflorum L. were formulated using the pharmaceutical excipients tartaric acid, citric acid, and sodium bicarbonate and the quality parameters of the granules were evaluated. The formulated granules met the quality parameters assessed from the angle of repose, bulk density, tapped density, Carr’s Index, Hausner’s ratio, effervescent cessation time, and stability studies. Thus, the formulated effervescent granules of O. tenuiflorum L. can be used for therapeutic purposes or as a functional food.

Abstract Tulsi, also known as holly basil, are well known for their medicinal, pharmacological, and health benefits. However, the sensory profiles of food or beverages made from this plant have never been reported before. This study aimed to define the sensory characteristics of beverages made from tulsi leaves using the Quantitative Descriptive Analysis (QDA) method. Fourteen trained panelists were selected and trained, three samples of tulsi beverages were evaluated and the tests were replicated twice. In total, 27 sensory attributes were generated. The dominant attributes include yellow, brown and transparent colors for the visual aspect, representing tea-like beverage. Moreover, both the odor and flavor of basil, mint and grassy were also perceived as key attributes for the tulsi beverages. In addition, the tulsi beverages were also characterized by strong astringent taste and aftertaste. In general, the test was reliable, however, additional training is needed to train the panelists intensively for the selected attributes. The results obtained from this research can be used as a preliminary study to develop new functional food products made from tulsi leaves by considering their sensory profiles.

Aims: To Analysis of genetic variability in Tulsi (Ocimum sanctum) genotypes by using RAPD & SSR Markers. Place and Duration of Study: Department of Plant Biotechnology at K.K.Wagh College of Agricultural Biotechnology, Nashik. Methodology: Ocimum tenuiflorum Linn., commonly known as Tulsi, is an aromatic plant with significant traditional and medicinal value. To assess the genetic diversity and relatedness of six Tulsi genotypes (Krishna, Ram, Lavangi, Pandharpuri, Daisil, and Kapoori), molecular techniques were employed. The genotypes were collected from Nagarjuna Medicinal and Aromatic Plant Park at Dr. P.D.K.V. Akola. DNA isolation was performed using alcohol fixation without liquid nitrogen, and the genotypes were analyzed using RAPD and SSR primers for molecular characterization. Results: Genetic diversity analysis of six Tulsi genotypes (Krishna, Ram, Lavangi, Pandharpuri, Daisil, and Kapoori) was performed using RAPD and SSR markers. Five RAPD primers produced 15 bands, with 11 bands showing polymorphism (73.3%) and 4 bands showing monomorphism (26.7%). The PIC value ranged from 0.28 to 0.49 (average: 0.40). Four SSR primers generated 9 bands, with 8 bands showing polymorphism and 1 band showing monomorphism. The PIC value ranged from 0.24 to 0.57 (average: 0.39). The Jaccard coefficient revealed moderate to high similarity in RAPD (0.40 to 0.73) and SSR (0.44 to 0.88) analyses. The UPGMA dendrogram separated the genotypes into two main clusters. Cluster 1 included Krishna, Lavangi, Ram, Pandharpuri, and Daisil Tulsi, while cluster 2 consisted of Kapoori Tulsi. The SSR dendrogram also formed two clusters, with Krishna, Lavangi, Ram, Daisil, and Kapoori genotypes in cluster 1, and Pandharpuri Tulsi showing dissimilarity and forming cluster 2.

Ocimum tenuiflorumL., holy basil “Tulsi”, is an important medicinal plant that is being grown and traditionally revered throughout Indian Subcontinent for thousands of years; however, DNA sequence-based genetic diversity of this aromatic herb is not yet known. In this report, we present our studies on the phylogeography of this species usingtrnL-trnFintergenic spacer of plastid genome as the DNA barcode for isolates from Indian subcontinent. Our pairwise distance analyses indicated that genetic heterogeneity of isolates remained quite low, with overall mean nucleotidep-distance of5×10-4. However, our sensitive phylogenetic analysis using maximum likelihood framework was able to reveal subtle intraspecific molecular evolution of this species within the subcontinent. All isolates except that from North-Central India formed a distinct phylogenetic clade, notwithstanding low bootstrap support and collapse of the clade in Bayesian Inference. North-Central isolates occupied more basal position compared to other isolates, which is suggestive of its evolutionarily primitive status. Indian isolates formed a monophyletic and well-supported clade withinO. tenuiflorumclade, which indicates a distinct haplotype. Given the vast geographical area of more than 3 million km2encompassing many exclusive biogeographical and ecological zones, relatively low rate of evolution of this herb at this locus in India is particularly interesting.

Ocimum sanctum L. (Lamiaceae) sin. Ocimum tenuiflorum L. or Tulsi basil is a plant originating from tropical and subtropical areas of India. It is used in both the traditional and official medicine in India. Tulsi is a type of basil that is insufficiently explored and studied in Europe. The goal of this paper is to determine the chemical composition, antioxidative, and antimicrobial activity of the essential oil Ocimum sanctum L. grown in Serbia. The quantity of essential oil in 100 g of herb (v/w) is 0.68%, with 41 components identified in the tested essential oil. The most represented chemical group are sesquiturpene hydrocarbonates with 80.47%. Other groups were much less represented. Sesquiturpene hydrocarbonate ?-cariophyllene is a predominant component in the essential oil with 63.80%. The quantity of tested essential oil needed to achieve 50% of inhibition of DPPH radicals is 0.35 ?g/ml, and it has high potential to neutralize free radicals. The essential oil exhibited antibacterial activity to all tested strains of bacteria, both Gram-positive and Gram-negative. It affected all strains in an inhibitory way in the interval 0.34-41.50 ?l/ml, and in a bactericide way within the range 22.50-124.5 ?l/ml. This study aimed to characterize the morphological and anatomical traits of six Ocimum species namely Ocimum tenuiflorum L. Syn. O. Sanctum L. (green var. known as Radha Tulasi), O. tenuiflorum L. (purple var. known as Krishna Tulasi), O. basilicum L.( green var. known as Sugandhi Tulasi), O. basilicum var. pupurascens L.( purple var. known as Durlabha Tulasi), O. canum Sims. (Kanaka Tulasi) and O. gratissimum L. (Rama Tulasi). Chlorophyll estimation of young and matured leaves was also estimated. Cholorophyll a, Chl. b and total chlorophyll was maximum in both young and matured leaves of O. gratissimum L. compared to other species. Carotenoid pigments was maximum of 203.94 mg/ml in young leaves of O. gratissimum L. and 244.92 mg/ml in matured leaves of O. sanctum L. Comparative morphological characters and anatomical variation were studied for identification of the desired species in drug manufacturing industries before making formulations. This study will certainly help to ensure the quality of the crude drug as these are collected by untrained workers and supplied in dry condition to the drug manufacturing industries. Therefore, there is a chance of adulteration because of vested interest or ignorance as many plants are known by common names or in vernacular names. It was found that the six species can be identified and authenticated using these experimental findings before preparation of compound Ayurveda, Siddha and Unani (ASU) drugs in Indian System of Medicines.

The antioxidant activities of four genotypes of basil species were selected for biochemical studies, viz., Ocimum sanctum L. (Rama Tulsi), Ocimum tenuiflorum L. (Krishna Tulsi), Ocimum gratissimum L. (Van Tulsi), and Ocimum basilicum L. (Babui Tulsi) for the antioxidant properties. Ocimum L. is considered useful in the treatment of respiratory system disorders, reduction of blood glucose level and is regarded as an 'adaptogen' or anti-stress agent. The phytochemical analysis of the plant leaves extract to the estimation of standard biochemical methods such as peroxidase, superoxide dismutase (SOD), polyphenol oxidase (PPO), catalase activity, antioxidant, flavonoids, total phenols, protein as well as total carbohydrates, starch and reducing and non-reducing sugar. The biochemical studies revealed that the starch (98 mg/gfw), the content was found to be higher in O. gratissimum. Similarly the other parameters such as total sugar (159 mg/gfw), non-reducing sugar (61mg/gfw), Peroxidase (142 U/mL), Catalase (188 U/mL), SOD (92.47 U/mL), PPO (462 U/mL) were found to be higher in O. sanctum. On the other hand, soluble protein (62 mg/gfw), total flavonoids (325 mg/gfw) and total flavonols (486 mg/gfw) were found to be higher in O. tenuiflorum. Total antioxidants (136.45 mg/gfw), reducing sugar (108 mg/gfw) and phenol (479 mg/gfw) were found to be higher in O. basilicum. The result shows that the best genotype among the four follows the trends O. sanctum > O. basilicum > O. tenuiflorum > O. gratissimum. On the basis of the result, the two genotypes can be recommended for medicinal purpose in the pharmaceutical application.

The objective of this study was to gather data, create a database of the compounds present in Ocimum tenuiflorum (O. tenuiflorum), and gather related literature on the compounds found. A thor-ough literature search was performed to gather in-formation on compounds present in O. tenuiflorum, including chemical structures, relative abundance, presence in different plant parts, and availability from chemical supply vendors. The compounds’ chemical structures were refined using Discovery Studio Visualizer and Chimera software for future in-silico docking studies. The structures with cleaned structural geometry were obtained through D.S. Vis-ualizer for docking in the future. From the literature search of previously presented articles, it was found that methyl eugenol had the greatest percent com-position in O. tenuiflorum. After searching the Pro-tein Data Bank, COX-1, COX-2, and NF Kappa B were found to be the main protein targets of O. ten-uiflorum compounds in the arachidonic acid inflamematory pathway. Thus, the anti-inflammatory proper-ties of O. tenuiflorum have been analyzed in this ar-ticle for future in silico docking.

AbstractAcross the three domains of life, circadian clock is known to regulate vital physiological processes, like, growth, development, defence etc. by anticipating environmental cues. In this work, we report an integrated network theoretic methodology comprising of random walk with restart and graphlet degree vectors to characterize genome wide core circadian clock and clock associated raw candidate proteins in a plant for which protein interaction information is available. As a case study, we have implemented this framework in Ocimum tenuiflorum (Tulsi); one of the most valuable medicinal plants that has been utilized since ancient times in the management of a large number of diseases. For that, 24 core clock (CC) proteins were mined in 56 template plant genomes to build their hidden Markov models (HMMs). These HMMs were then used to identify 24 core clock proteins in O. tenuiflorum. The local topology of the interologous Tulsi protein interaction network was explored to predict the CC associated raw candidate proteins. Statistical and biological significance of the raw candidates was determined using permutation and enrichment tests. A total of 66 putative CC associated proteins were identified and their functional annotation was performed.

Objective The HPLC-PDA method was developed and validated for vicenin-2, orientin, cynaroside, betulinic acid, genistein, with syringic acid, rosmarinic acid, eugenol, carnosic acid, oleanolic acid, ursolic acid, luteolin, apigenin for quantification and confirmed using a novel ESI -MS/MS method in the Ocimum samples. Method The methodology parameters were developed on the RP-C18 column with a gradient elution of 1 mL/min flow rate for 0.1% o-phosphoric acid and acetonitrile at 210 and 340 nm wavelengths. Result The validation data for 13 bioactive compounds showed good linearity (r2> 0.99) with sensitive LOD (0.034-0.684 µg/mL) and LOQ (0.100-2.068 µg/mL) with recoveries (83.66-101.53%).The results were found precise (RSD,<5.0%) and accurate (RE, -0.60-1.06) for the quantification. The method performance was verified and found robust by analyzing ten samples of O. tenuiflorum from the ten geographical states of India (RSD, < 5.0%). Conclusion The validated HPLC-PDA method was found selective and suitable for analyzing thirteen compounds in O. tenuiflorum and twelve cultivars from the Ocimum genus as a quality control tool. This method can be used in routine analysis as an inexpensive alternative to advanced techniques. Highlights This work is the first to report for vicenin-2, orientin, cynaroside, betulinic acid, and genistein, with simultaneous analysis of eight bioactive compounds in the Ocimum genus.

Aims: Species of the family Lamiaceae possess a rich tradition of use for flavoring and medicinal purposes. This paper focusses on the nutritional and thermal characteristics of the seeds from eight species belonging to this family: Gmelina arborea Roxb. ex Sm., Hyptis suaveolens (L.) Poit., Leonotis nepetifolia (L.) R.Br., Ocimum americanum L., Ocimum sanctum L. (Rama Tulsi), Ocimum tenuiflorum L. (Krishna Tulsi), Origanum vulgare L. and Tectona grandis L.f. Methodology: The oil, starch, total polyphenol, flavonoid and mineral contents for aforementioned seeds were determined. Fourier-transform infrared (FTIR) spectroscopy was used to assess the phytoconstituents. Thermogravimetric/derivative thermogravimetric analyses (TG/DTG) and differential scanning calorimetry (DSC) analyses were performed to analyze the decomposition patterns. Results: The concentrations of oil, starch, total polyphenol, flavonoids and minerals for the seeds from the eight plants under study ranged from 11.8 to 50.4%, from 0.22 to 1.84%, from 295 to 5842 mg/kg, from 1660 to 12680 mg/kg and from 11756 to 33927 mg/kg, respectively. Unsaturated oils, polyphenols and lignin were recognized by vibrational spectroscopy. The sequence of thermal effects in the seed pyrolysis process above 100°C have been put in relation to seed protein crystallization (endotherm at 200°C), oxidation reactions and degradation of hemicellulose and other fiber components (at around 300°C), and decomposition of polyunsaturated (at 357°C) and mono-unsaturated (at 391°C) triglycerides. Conclusion: Lamiaceae seeds are potential food alternatives to cereals.

Stable silver nanoparticles of various sizes are synthesised using the aqueous extract of Tulsi ( Ocimum tenuiflorum) as the reducing agent as well as a stabilising agent under the influence of light of wavelength ranging from 200 to 900 nm. The formation of silver nanoparticles is monitored using a UV-Visible spectrophotometer and the particle shape, size and stability are analysed using scanning electron microscopy, dynamic light scattering and zeta potential measurement techniques. It is observed that the size of the silver nanoparticles decreases on increasing the wavelength of irradiating light. The silver nanoparticles show antimicrobial activity against gram-positive ( Bacillus subtilis), as well as gram-negative ( Escherichia coli) bacteria. The antimicrobial activity shows an inverse relationship with the size of the silver nanoparticles. The bio-synthesised silver nanoparticles are successfully embedded in a polymeric film to act as an active antimicrobial film that can potentially release the active constituents.
