Abstract. The tRNA modification gene in eukaryotes is relatively conservative. As an important modification gene, the TRDMT1 gene plays an important role in maintaining tRNA structural maintenance and reducing mistranslation of protein translation by methylation of specific tRNA subpopulations. Mouse and zebrafish TRDMT1 knockout experiments indicate that it may mediate growth and development through tRNA modification. However, there are no systematic reports on the function of tRNA-modified genes in livestock. In this study, Qinchuan cattle DNA pool sequencing technology was used. A G>C mutation in the −1223 bp position upstream of the TRDMT1 translation initiator codon was found. At this locus, the dual-luciferase assay indicated that different genotypes cause differences in transcriptional activity (P<0.05). Our experiment detected a natural genetic variation of a tRNA modification gene TRDMT1, which may provide potential natural molecular materials for the study of tRNA modification.

Анотація:
5-Azacytidine (5-azaC), a methyltransferase inhibitor and anticancer drug, can promote several cellular stress responses such as apoptosis, autophagy, and senescence. The action of 5-azaC is complex and can be modulated by dose, time of treatment, and co-administration with oxidants. Insulinoma is a rare pancreatic neuroendocrine tumor with limited chemotherapeutic options. In the present study, two cellular models of insulinoma were considered, namely NIT-1 and β-TC-6 mouse cells, to evaluate the effects of 5-azaC post-treatment during hydrogen peroxide-induced oxidative stress. 5-azaC attenuated the development of oxidant-induced senescent phenotype in both cell lines. No pro-apoptotic action of 5-azaC was observed in cells treated with the oxidant. On the contrary, 5-azaC stimulated an autophagic response, as demonstrated by the increase in phosphorylated eIF2α and elevated pools of autophagic marker LC3B in oxidant-treated β-TC-6 cells. Notably, autophagy resulted in increased necrotic cell death in β-TC-6 cells with higher levels of nitric oxide compared to less affected NIT-1 cells. In addition, 5-azaC increased levels of RNA methyltransferase Trdmt1, but lowered 5-mC and m6A levels, suggesting Trdmt1 inhibition. We postulate that the 5-azaC anticancer action may be potentiated during oxidative stress conditions that can be used to sensitize cancer cells, at least insulinoma cells, with limited drug responsiveness.

Eukaryotic 5-methylcytosine RNA methyltransferases catalyze the transfer of a methyl group to the fifth carbon of a cytosine base in RNA sequences to produce 5-methylcytosine (m5C). m5C RNA methyltransferases play a crucial role in the maintenance of functionality and stability of RNA. Viruses have developed a number of strategies to suppress host innate immunity and ensure efficient transcription and translation for the replication of new virions. One such viral strategy is to use host m5C RNA methyltransferases to modify viral RNA and thus to affect antiviral host responses. Here, we summarize the latest findings concerning the roles of m5C RNA methyltransferases, namely, NOL1/NOP2/SUN domain (NSUN) proteins and DNA methyltransferase 2/tRNA methyltransferase 1 (DNMT2/TRDMT1) during viral infections. Moreover, the use of m5C RNA methyltransferase inhibitors as an antiviral therapy is discussed.

DNA, RNA and histone methylation is implicated in various human diseases such as cancer or viral infections, playing a major role in cell process regulation, especially in modulation of gene expression. Here we developed a convergent synthetic pathway starting from a protected bromomethylcytosine derivative to synthesize transition state analogues of the DNA methyltransferases. This approach led to seven 5-methylcytosine-adenosine compounds that were, surprisingly, inactive against hDNMT1, hDNMT3Acat, TRDMT1 and other RNA human and viral methyltransferases. Interestingly, compound 4 and its derivative 2 showed an inhibitory activity against PRMT4 in the micromolar range. Crystal structures showed that compound 4 binds to the PRMT4 active site, displacing strongly the S -adenosyl- l -methionine cofactor, occupying its binding site, and interacting with the arginine substrate site through the cytosine moiety that probes the space filled by a substrate peptide methylation intermediate. Furthermore, the binding of the compounds induces important structural switches. These findings open new routes for the conception of new potent PRMT4 inhibitors based on the 5-methylcytosine-adenosine scaffold. Abstract Phthalates are ubiquitous environmental contaminants and impact reproduction and epigenetic mechanisms. The use of diisobutyl phthalate (DIBP) has increased in the last decade. However, studies about DIBP's effects on female reproductive physiology are limited. Methylation of DNA and RNA are epigenetic processes that regulate gene expression, cell biology, and reproductive functions. How DIBP exposure regulates these epigenetic mechanisms in ovarian follicular cells remains to be unveiled. We hypothesize that DIBP at low doses alters the expression of RNA and DNA methylation regulators in bovine granulosa cells (GC). Bovine GC was isolated from small antral follicles (1-5 mm on surface diameter) from ovaries obtained at a slaughterhouse. Cells were cultured in vitro until reaching an average 70% confluence and treated with DIBP at levels detected in human blood (0 ng/mL, 1 ng/mL, 10 ng/mL, 100 ng/mL) combined with hormones that regulate ovarian folliculogenesis: FSH or FSH+IGF1. Cells were cultured for 24h for total RNA isolation (n = 5 pools) or for 48h for cell counting (n=5 pools). Relative mRNA abundance of RNA N6 methyladenine (m6A) methylation machinery (m6A writers: METTL3, METTL14; m6A erasers: ALKBH5, FTO), RNA 5-methylcytosine (m5C) methylation regulators (NSUN2, TRDMT1), and DNA methylation players (DNA methyltransferases: DNMT1, DNMT3A, DNMT3B; DNA demethylases: TET1, TET2, TET3) was determined via qPCR using 18S as housekeeping gene and expressed as 2-ΔΔCt. Cell proliferation was analyzed by cell counting using a MUSE cell analyzer. Data were analyzed via one-way ANOVA with GLM procedures of SAS for Windows. Normality was tested using Shapiro-Wilk test procedures and data considered not normal were transformed to log2 (Y + 0.6). Data sets that showed non-gaussian distribution after log-transformation was analyzed via the Kruskal-Wallis test using GraphPad Prism 9.2.0. Cell proliferation was not affected by DIBP treatments in combination with FSH or with FSH+IGF1. In GC treated with FSH in combination with DIBP, there was no significant effect on relative mRNA abundance of RNA or DNA methylation players. In combination with FSH+IGF1, GC treated with DIBP had lower (P &lt; 0.05) METTL14 relative mRNA abundance at 1 ng/mL and 10 ng/mL than at 0 ng/mL and 100 ng/mL. TRDMT1 relative mRNA abundance was lower (P &lt; 0.05) in the DIBP group at 10 ng/mL in comparison to groups 0 ng/mL and 1 ng/mL, but no 100 ng/mL. In addition, there was no significant effect on the relative mRNA abundance of DNA methylation regulators. In summary, GC treated with FSH+IGF1 and DIBP at low doses alters the gene expression of RNA methylation players in m6A and m5C RNA modifications in a non-monotonic dose-response, but does not alter mRNA abundance of DNA methylation regulators.

Fast and accurate acquisition of the outline of rural buildings on remote sensing images is an efficient method to monitor illegal rural buildings. The traditional object detection method produces useless background information when detecting rural buildings; the semantic segmentation method cannot accurately segment the contours between buildings; the instance segmentation method cannot obtain regular building contours. The rotated object detection methods can effectively solve the problem that the traditional artificial intelligence method cannot accurately extract the outline of buildings. However, the rotated object detection methods are easy to lose location information of small objects in advanced feature maps and are sensitive to noise. To resolve these problems, this paper proposes a two-stage rotated object detection network for rural buildings (TRDet) by using a deep feature fusion network (DFF-Net) and a pixel attention module (PAM). Specifically, TRDet first fuses low-level location and high-level semantic information through the DFF-Net and then reduces the interference of noise information to the network through the PAM. The experimental results show that the mean average precession (mAP), precision, recall rate, and F1 score of the proposed TRDet are 83.57%, 91.11%, 86.5%, and 88.74%, respectively, which outperform the R2CNN model by 15%, 15.54%, 4.01%, and 9.87%. The results demonstrate that the TRDet can achieve better detection in small rural buildings and dense rural buildings.

T cell antigen receptor δ (Tcrd) variable region exons are assembled by RAG-initiated V(D)J recombination events in developing γδ thymocytes. Here, we use linear amplification–mediated high-throughput genome-wide translocation sequencing (LAM-HTGTS) to map hundreds of thousands of RAG-initiated Tcrd D segment (Trdd1 and Trdd2) rearrangements in CD4−CD8− double-negative thymocyte progenitors differentiated in vitro from bone marrow–derived hematopoietic stem cells. We find that Trdd2 joins directly to Trdv, Trdd1, and Trdj segments, whereas Trdd1 joining is ordered with joining to Trdd2, a prerequisite for further rearrangement. We also find frequent, previously unappreciated, Trdd1 and Trdd2 rearrangements that inactivate Tcrd, including sequential rearrangements from V(D)J recombination signal sequence fusions. Moreover, we find dozens of RAG off-target sequences that are generated via RAG tracking both upstream and downstream from the Trdd2 recombination center across the Tcrd loop domain that is bounded by the upstream INT1-2 and downstream TEA elements. Disruption of the upstream INT1-2 boundary of this loop domain allows spreading of RAG on- and off-target activity to the proximal Trdv domain and, correspondingly, shifts the Tcrd V(D)J recombination landscape by leading to predominant V(D)J joining to a proximal Trdv3 pseudogene that lies just upstream of the normal boundary.

V prispevku se razpravlja o tem, ali bi slovenski stavek Še pogleda me ne lahko služil kot zgled za to, da je mogoče v slovenščini aktualno sedanjost izraziti z dovršnim sedanjikom. Čeprav ni mogoče trditi, da je vidsko obnašanje v vseh slovanskih jezikih enotno, pa se zdi, da stara Miklošičeva trditev, da je odgovor na vprašanje Kaj počneš? v slovanskih jezikih praviloma nedovršni sedanjik, še vedno drži. Dovršni sedanjik ima v slovenščini vrsto pomenov, vendar nima pomena aktualne sedanjosti. Kot navajata slovnica avtorjev Bajec, Kolarič, Rupel in Toporišičeva slovnica, imajo nekateri pomeni dovršnega sedanjika modalno komponento sem pripravljen, sem sposoben, nameravam narediti. Modalno komponento ima verjetno tudi ti. prasens effectivum oz. performativna raba tipa obljubim, prisežem. Mogoče je torej trditi, da ima dovršni sedanjik v Še pogleda me ne modalni pomen – ‘še pogledati me noče’, za kar najdemo paralele tudi v drugih slovanskih jezikih, med drugim tudi v starem novgorodskem dialektu (po Zaliznjaku prezens naprasnogo ožidanija).

Objetivo: Avaliar a eficácia de um programa ludomotor de soltar pipas, voltado para desenvolver a velocidade de reação. Método: Este estudo foi desenvolvido utilizando-se os testes de reação motora (TRM), composto de três provas específicas: Tempo Reação Simples (TRS), Tempo de Reação com Discriminação (TRD) e Tempo de Reação com Discriminação para Tarefa Complexa (TRDTC). Resultados: Após 65 sessões do programa ludomotor, com duração de 45 minutos cada uma, mostrou-se, em relação ao TRS, TRD e TRDTC, uma significância estatística, em que foi identificada a importância da intervenção proposta. Conclusão: Verificou-se que a intervenção desenvolvida neste trabalho refinou as habilidades perceptivo-motoras dos alunos, o que se refletiu no desenvolvimento da coordenação visomotora.

Objectives: Investigate the relationship between thoughtfully reflective decision-making (TRDM) and computer-focused cyber deviance involvement and computer-focused cybercrime victimization. Method: Survey data collected from samples of 1,039 employees and 418 students at a large private university were analyzed using ordinary least squares and negative binomial regression to test the effects of TRDM on computer-focused cyber deviance involvement and victimization. Results: TRDM reduces computer-focused cyber deviance involvement and computer-focused cybercrime victimization across measures and samples. The sensitivity analyses also indicated that TRDM is a more robust predictor of cyber deviance involvement than victimization. The results from moderation analyses showed that, whereas protective effects of TRDM are invariant across genders, they are less salient among older employees for the scenario-based measure of cybercrime victimization. Conclusions: Individual-level cognitive decision-making processes are important in predicting computer-focused cyber deviance involvement and victimization. These results can inform the development of targeted institutional and criminal justice policies aimed at reducing computer-focused cybercrime.

The radial temperature difference calculation of a steam drum wall is an important step in the process of calculating the low-cycle fatigue lifetime of a steam drum. So the German Standard TRD301 recommended a temperature difference computing method based on the simplified solution for the radial temperature of a steam drum wall. In order to validate the feasibility for the application of this computing method to solving the same problem of a supercharged boiler, on the basis of the reasonable simplifications to the unstable heat conduction problem across a steam drum wall, its heat-conduction equations and definite conditions were determined, then the analytical solution for the radial temperature of a steam drum wall, together with its approximate and simplified solutions, is derived. The running example of one super-charged boiler was calculated and analyzed by use of the radial temperature difference computing methods based on the approximate and simplified solutions respectively. The com-puted results show that the German Standard computing method of TRD301 has great limitations in the same problem calculation of a supercharged boiler, and it can’t apply to the researches on the same problem and its relevant problems of a supercharged boiler as a universal computing method.

V prispevku predstavljamo izhodišča in smernice poteka razvoja naprednega orodnega jekla za najzahtevnejše aplikacije v industriji nožev, ki se izvaja v podjetju SIJ Ravne Systems v povezavi z Univerzo v Mariboru Fakulteto za strojništvo. Razvoj je na eni strani povezan z identifikacijo ključnih lastnosti orodnega jekla, ki predstavljajo konkurenčno prednost v primerjavi z ostalimi konvencionalnimi jekli in to so: žilavost, trdota, trdnost in rezilna sposobnost. Po drugi strani pa razvoj predstavlja tudi vpeljavo ustrezne tehnologije izdelave in naknadne toplotne oziroma mehanske obdelave orodnega jekla, kar omogoča doseganje ključnih lastnosti izdelka – nožev.

Potrošniki v zadnjih nekaj letih vedno pogosteje od podjetij zahtevajo, da trajnostno poslujejo in proizvajajo trajnostne izdelke. Zato lahko na trgu vedno pogosteje najdemo trajnostne izdelke. Namen raziskave je bil ugotoviti, kakšen odnos imajo študenti Univerze v Novem mestu do trajnostnih izdelkov. Raziskava je bila izvedena novembra in decembra 2021, uporabili smo tehniko anketiranja. Rezultati so pokazali, da so študentom Univerze v Novem mestu trajnostni izdelki všeč. Razlik med spoloma in starostnimi skupinami v pogostosti poseganja po trajnostnih izdelkih ni. Prav tako ne moremo trditi, da študenti Univerze v Novem mestu čutijo pritisk družbe k kupovanju trajnostnih izdelkov.

V prispevku smo z lastnoimenskega stališča analizirali francoske prevode slovenskih turističnih brošur, ki sta jih izdali Slovenska turistična organizacija ali Vlada Republike Slovenije, Pisarna za odnose z javnostjo in mediji.Posvetili smo se imenom bitij (skupina je pričakovano manj obsežna) in zlasti zemljepisnim imenom. Medtem ko pri prevajanju imen bitij nismo zaznali večjih zagat, ne moremo trditi enako za zemljepisna imena. Podobno kot imena bitij tudi enobesedna zemljepisna poimenovanja ne povzročajo večjih zagat. Te se pojavljajo pri večbesednih ze mljepisnih poimenovanjih.Ker gre pri turističnih brošurah za pomemben vidik predstavitve države navzven, bi pričakovali, da bodo dobili prevajalci podrobnejše smernice za prevajanje lastnih imen. Upamo, da v pričujočem prispevku na dovolj jasen način osvetlimo prevajalske zagate in s tem koga spodbudimo k nastanku omenjenih smernic oziroma priporočil.

Recently, Youssef developed the fractional order generalized thermoelasticity (FOGTE) in the context of extended thermoelasticity (ETE). In this work, we extended the concept of fractional calculus into the temperature rate dependent thermoelasticity (TRDTE) and introduced the unified form of the two cases. Upon introducing the electromagnetic field with magnetic monopoles and considering the geometrical nonlinearity, we proposed a fractional order generalized electro-magneto- thermo-elasticity (FOGEMm-poleTEg-non) with magnetic monopoles (m-pole) and geometrical nonlinearity (g-non). To deal with multi-physics problems using numerical methods, we obtained a generalized variational theorem by using the semi-inverse method.

High availability is important for large scale distributed systems. Replication provides effective ways to enhance performance, high availability and fault tolerance in distributed systems. An efficient and effective replication technique is the key to improve the availability performance. Data and processes can be replicated for failures recovery. There are currently projects successfully implemented in two-replica distribution technique (TRDT) or primary–backup technique. However, these projects have their weaknesses of increasing cost overhead and inherit irrecoverable scenarios from TRDT such as double faults when both copies of replicated components are damaged. The authors propose the Neighbor Replica Distributed Technique (NRDT) availability prediction model. Focusing on improving high availability in which it predicts future expectation of interdependent server’s availability in a distributed online system over an extended period of time. The results and discussion are explored further in the article.

Prispevek skuša ovrednotiti vlogo kovin v Heziodovih pesnitvah Teogonija ter Dela in dnevi. Zlato je pri Heziodu božanska kovina – le bogovi ga nosijo, zlati, najboljši izmed rodov, je prav tako bogovom najbližji. V obeh pesnitvah redko omenjeno srebro je slabše od zlata in ni več atribut bogov. S srebrom Heziod opisuje vrtince rek, visoke stebre v podzemlju, nimfe. Tako kot je srebro manj vredno od zlata, je tudi srebrni rod manj vreden od zlatega. Bronu, prvi zlitini, pripiše pesnik lastnosti trdote in moči, iz njega sta izdelana orodje in orožje, s katerim bronasti rod bojuje vojne. Železo se v pesnitvah pojavi kot kovina iz katere sta izdelana orodje in orožje, železni rod, najslabšega izmed vseh rodov, pestita žalost in težko delo. Kljub temu, da je izdelava železnih predmetov spodbudila velik tehnološki napredek, pomeni ta kovina pesniku sinonim za trdo delo, napore in bedo – skratka popolno nasprotje zlatemu, tj. božanskemu.

Če si izposodim besede Janeza (Ivana) Samse v članku iz leta 1916, objavljenem v reviji Dom in svet in namenjenem predstavitvi komaj iz danega grško-slovenskega slovarja, bi si upala trditi, da je grško-slovenski slovar tako redek podvig, »kakor cvetje bajne aloe, ki se pojavi menda vsakih sto let«. Vsi tisti, ki smo staro grščino začeli študirati že v gimnazijskih letih, smo imeli za nepogrešljivega sopotnika tudi prvi in edini grško-slovenski slovar, ki smo ga po domače imenovali kar »Dokler« in ki je izšel daljnega leta 1915. Dijaki, ki smo prevajali grška besedila v gimnaziji, smo se večkrat tudi posmehovali že zastarelemu slovenskemu jeziku, ki smo ga zasledili v slovarju. Naš stari »Dokler« pa je ostajal kot steber sredi dolgih popoldnevov, ki smo jih preživeli ob grških besedilih, in bil za vse nas pravi sopotnik pri odkrivanju in razčlenjevanju dolgih grških stavkov ter spoznavanju nepogrešljivih zakladov grške modrosti.

Abstract The fundamental equations of the two dimensional generalized thermoelasticity (L-S model) with one relaxation time parameter in orthotropic elastic slab has been considered under effect of rotation. The normal mode analysis is used to the basic equations of motion and heat conduction equation. Finally, the resulting equations are written in the form of a vector-matrix differential equation which is then solved by the eigenvalue approach. The field variables in the space time domain are obtained numerically. The results corresponding to the cases of conventional thermoelasticity CTE), extended thermoelasticity (ETE) and temperature rate dependent thermoelasticity (TRDTE) are compared by means of graphs.

V njuni koncepciji družbe primata nima menjava, temveč dolg: temeljno družbeno razmerje je torej razmerje med upnikom in dolžnikom. Nujni pogoj tega razmerja je spomin, ki zagotavlja, da dolg ni pozabljen. Vendar ta spomin ni samoumeven, ampak ga je bilo treba človeku šele narediti. To »predzgodovinsko delo človeka« Deleuze in Guattari interpretirata kot osrednji napor »primitivnega sociusa«, ki sestoji iz vseh krutih tehnik vpisovanja v telo, kar imenujeta »sistem krutosti«. Avtorja se opreta na Nietzschejevo Genealogijo morale in trdita, da se besedni spomin vzpostavi na podlagi »aktivne sposobnosti pozabe« in s »potlačitvijo biološkega spomina«. Če je sredstvo vzpostavitve besednega spomina sistem krutosti in njen agens aliansa oziroma primitivni socius sam, kaj točno je potlačitev in kaj je tisto, kar je potlačeno? The present work is concerned with an electro-magneto-thermoelastic coupled problem for a homogeneous, isotropic, thermally and electrically conducting semi-infinite solid medium in two-dimensional space where an initial magnetic field with constant intensity acts parallel to the plane boundary of the half-space. The surface of the half-space taking as traction free which is subjected to a thermal shock. The modified Ohm's law including the temperature gradient and charge density effect to the governing equations of the generalized thermoelasticity under the temperature rate dependent thermoelasticity (TRDTE) proposed by Green and Lindsay (GL model) has been introduced. Normal mode analysis together with eigenvalue approach technique is used to obtain the general solutions for the physical quantities. Numerical results for the physical quantities are illustrated graphically and analyzed. The graphical results indicate that the effect of the coefficient connecting the temperature gradient and the electric current density of the modified Ohm's law on the plane waves is very pronounced. Comparison are made with the results obtained in the absence of the above coefficient.

Izvleček Namen: V članku raziskujemo v literaturi pogosto spregledano vprašanje kolektivnega psihološkega opolnomočenja uporabnikov spletnih zdravstvenih skupnosti. Izhajajoč iz teorij opolnomočenja s področja psihologije skupnosti, preverjamo, kateri dejavniki, ki obenem odražajo pomembne lastnosti spletnih zdravstvenih skupnosti, so povezani s kolektivnim psihološkim opolnomočenjem njihovih uporabnikov. Metode: Vlogo štirih dejavnikov kolektivnega opolnomočenja smo analizirali s pomočjo multiple linearne regresije na podatkih, zbranih konec leta 2010 s spletno anketo na neverjetnostnem vzorcu (n = 235) aktivnih sodelujočih na forumih največje slovenske spletne zdravstvene skupnosti Med.over.net, ki je vključeval 8,5% moških, 49,7% vsaj visoko izobraženih in 41,5% poročenih anketirancev, ki so bili v povprečju stari 35,1 leta (SD = 9,1). Rezultati: Ugotavljamo, da se predstavljeni teoretični model dejavnikov ustrezno prilega podatkom (F = 8,65, df = 8, p < 0,001) in z njim lahko pojasnimo 23,4% variabilnosti občutka kolektivnega opolnomočenja. Občutek pripadnosti spletni skupnosti (β = 0,279, p < 0,001), vključenost v organizacijske aktivnosti skupnosti (0,194, 0,001) in zaznana participacija spletne skupnosti v širšem okolju (0,157, 0,02) vplivajo na kolektivno opolnomočenje uporabnikov spletne zdravstvene skupnosti Med.over.net, medtem ko tega ni mogoče trditi za intenzivnost participacije v forumskih razpravah (0,029, 0,65). Zaključek: Za povečevanje kolektivne komponente psihološkega opolnomočenja uporabnikov spletnih zdravstvenih skupnosti je treba v prvi vrsti graditi na kakovosti odnosov med člani, vključevanju članov v strateške odločitve o skupnosti in na vključenosti skupnosti v širše družbeno okolje, saj sama participacija uporabnikov v spletnih skupnosti še ne zagotavlja njihovega višjega kolektivnega opolnomočenja.

Abstract Background: Recent data showed that gamma delta T cells can act as a vast functional setting of immune defense against tumors. Our previous study found that persist clonally expanded TRDV4 T cells might be relatively to better outcome for the patient with T-cell acute lymphoblastic leukemia (T-ALL) after hematopoietic stem cell transplantation (HSCT). However, little is known, the distribution and clonality of TRDV repertoire of gamma delta T cells in peripheral blood (PB) and its potential effect for clinical outcome in acute myeloid leukemia (AML) patients. Methods: Gamma delta T cells were sorted from peripheral blood of 30 patients with untreated de novo AML (AML-M0 6 cases, M1 1 case, M2 5 cases, M3 10 cases, M4 3 cases and M5 5 cases) and were observed the distribution of TRDV repertoire, while 12 healthy individuals served as normal controls. Then the TRDV repertoire were further analyzed by Genescan technique to determine the size of complementarity determining region 3 (CDR3) of TRDV (1-8) subfamilies genes and to evaluate the clonality of the detectable subfamilies of gamma delta cells. The binary Logistic regression analysis was used to explore the associations between clonal expansion of gamma delta T cells and outcome of AML patients. Results: The frequencies of TRDV1, TRDV3, TRDV4 and TRDV6 in gamma delta T cells from AML patients were significantly lower than that from healthy individuals (HI) (Figure A). The most frequently expanded TRDV subfamilies in AML patients were TRDV8 (56.67%) and TRDV4 (40%). Furthermore, the most frequently oligoclonally expanded TRDV subfamilies from AML patients were TRDV2 and TRDV4 subfamilies, and the clonal expansion frequencies were significant higher than that from healthy individuals, however, a significant lower clonal expansion frequency of TRDV1 was observed in AML patients (Figure B). Importantly, oligoclonalities of TRDV4 and TRDV8subfamilies were the independent protective factors for complete remission. In addition, the oligoclonal expansion frequencies of TRDV5 and TRDV6 subfamilies from the recurrence cases were significantly higher than that from non-recurrence cases. Significantly, oligoclonal TRDV5 and TRDV6subfamilies were found in the patients who developed AML relapse in different time point after first complete remission. Conclusions: The distribution and clonality of TRDV subfamilies significantly changed in gamma delta T cells from AML patients. Clonally expanded TRDV4 and TRDV8 T cells might contribute to immune response against AML, while oligoclonal TRDV5 and TRDV6 may be accompanied to patients who underwent AML relapse, the function of such oligoclones of gamma delta T cells is needed further investigation. Overall, these TRDV gamma delta T cell clones might be a potential immune biomarker for AML outcome.
