RESUMO Objetivo: Avaliar a habilidade de ordenação temporal com diferentes testes, verificar a especificidade desses testes e comparar o resultado com o grau de facilidade/dificuldade relatado pelos sujeitos. Métodos: Os indivíduos realizaram avaliação audiológica básica e Teste Dicótico de Dígitos, para triagem do processamento auditivo, e foram submetidos ao Teste Padrão de Frequência (TPF) e ao Teste Padrão de Duração (TPD), nas versões de Taborga-Lizarro, Musiek e Auditec®, para avaliar a habilidade de Ordenação Temporal. Após, foi apresentada a Escala Visual Analógica (EVA), para que os sujeitos identificassem a dificuldade de cada teste. Resultados: Foram avaliados 33 sujeitos, 29 do gênero feminino e quatro do gênero masculino, com idade entre 17 e 27 anos. Houve maior número de indivíduos que atingiram valores normais no teste Auditec® e de Taborga-Lizarro, para o TPD e TPF. No teste de Musiek, houve um número de sujeitos com resultados normais muito próximos aos de sujeitos com alteração de processamento. Na distribuição do teste mais difícil, houve significância estatística para o teste de Musiek. Quanto à especificidade dos testes, o Auditec® mostrou-se melhor, porém, a análise foi realizada apenas em TPF. Conclusão: Embora os indivíduos não tenham apresentado alteração e queixa de processamento auditivo, houve diferença nos resultados dos testes de ordenação temporal. Quanto ao grau de dificuldade relatado para o teste de Musiek, verificou-se que houve influência deste fator nos resultados dos testes. Na análise de especificidade dos testes, pôde-se observar melhores resultados para o teste Auditec®.

Application of TP as Tipimon® or TPA as Maxim® at the young fruitlet stage significantly increased yield in three lychee (Litchi chinensis Sonn.) cultivars: `Mauritius', `Floridian', and `Kaimana'. Application of TP followed by TPA a week later increased yield more than did either substance alone. In all experiments, TPA increased fruit size and weight, relative to both controls and TP-treated trees. The increased yield did not prevent the increase in fruit size. Use of sprays of TPA may be an effective way of satisfying the market demand for large lychee fruit. Chemical names used: 2,4,5-trichlorophenoxypropionic acid (TP); 3,5,6-trichloro-2-pyridyloxyacetic acid (TPA).

Quality seedlings production is one of the most important steps in the cultivation of vegetable species. In this sense, the objective was to evaluate the influence of different environments and substrates on the production of “pimenta-de-cheiro” (Capsicum chinense Jacq.) seedlings in Boa Vista, Roraima. An experiment was installed in a completely randomized design (CRD), with treatmentsarranged in subdivided plots. Five environments were evaluated: AG - agricultural greenhouse; TV35 - red shading net with 35% shading (red ChromatiNet®); TV50 - red shading net with 50% shading (red ChromatiNet®); TP35 - silver shading net with 35% shading (ChromatiNet® silver) and TP50 - silver shading net with 50% shading (ChromatiNet® silver) and four substrates: O -OrganoAmazon®; OP - OrganoAmazon® and PuroHumus®; OPSE (OrganoAmazon®, PuroHumus®, soil and manure) and OPSEC (OrganoAmazon®, PuroHumus®, soil, manure and carbonized rice husk). On the TP35 and TP50 shading nets, the C. Chinense grown on the OP substrate showed high Dickson’s quality index (DQI). The combination agricultural greenhouse (EA) and OPSE substrate promoted superior quality Capsicum chinense seedlings. The substrates OPSEC and OPSE are more affordable alternatives for the production of Capsicum chinense seedlings. Shading nets, associated with the alternative substrates, do not favor the production of quality Capsicum chinense seedlings under Amazon savanna conditions.

The past number of years has seen a sharp rise in the gold price. This has led to renewed activity in the gold sector and especially in refractory ore-bodies that are not economically feasible at lower gold prices. There have, historically, been a total of eleven BIOX® plants commissioned world wide with eight currently in operation. The most recent plants to be commissioned were Bogoso and Jinfeng in 2007 and Kokpatas in Uzbekistan in 2008. Design of the Bogoso BIOX® plant was started by Minproc in 2005. The design throughput of the sulphide BIOX® circuit is 820 tpd concentrate at a feed sulphide sulphur grade of 20 %. The Bogoso BIOX® plant has the largest biooxidation reactors with a live capacity of 1,500 m3 each. Plant design on the Jinfeng project started in 2005 and the engineering was performed jointly by Ausenco and Nerin. The BIOX® plant has a design capacity of 790 tpd concentrate at 9.4 % sulphide sulphur. The Kokpatas plant will be the largest BIOX® plant in the world with a Phase 1 design concentrate treatment capacity of 1,069 tpd at 20 % sulphide sulphur. The throughput will be doubled during Phase 2. Interestingly, unlike other BIOX® Plant, this plant will use a resin-in-pulp circuit for gold recovery, but the design will allow the plant to be converted to Carbon-in-pulp if required. There are currently three BIOX® projects in various stages of design or construction. This includes two expansions to existing BIOX® operations and one new BIOX® project. There are a number of major R&D projects currently under investigation with some very positive results. Most R&D is focussed on the main capital and operating cost items in the process, but there are also projects focussed purely on process optimisation and improvement.

An elastomeric spinal disk prosthesis design (BioFI™) with vertebral interlocking anchors has been modified using an embedded TiNi wire array. Bioinert styrenic block copolymer (Kraton®) and polycarbonate urethane (Bionate®) thermoplastic elastomer (TPE) matrices were utilized. Fatigue resistant NiTi wire was pretreated to induce superelastic martensitic microstructure. Stent-like helical structures were produced for incorporation within homogenous TPE matrix. Composite prototypes were fabricated in a vacuum hot press using transfer moulding techniques. Implant prototypes were subject to axial compression using a BOSE ® ELF3400. The NiTi reinforced implants exhibited reduction in axial strain, compliance, and creep compared to TPE controls. The axial properties of the NiTi reinforced Bionate® BioFI™ implant best approximated those of a spinal disk followed by Kraton®-NiTi, Bionate® and Kraton® prototypes. An ovine lumbar segment biomechanical model was used to characterize the disk prosthesis prototypes. Specimens were subject to 7.5Nm pure moments in axial rotation, flexion-extension and lateral bending with a custom jig mounted on an Instron® 8874. The motion preserving ligamentous nature of this arthroplasty prototype was not inhibited by NiTi reinforcement. Joint stiffness for all prototypes was significantly less than the intact and discectomy controls. This was due to lack of vertebral anchor rigidity rather than BioFI™ motion segment matrix type or reinforcement. Implant stress profiles for axial compression and axial torsion conditions were obtained using finite element methods. The biomechanical testing and finite element modelling both support existing BioFI™ design specifications for higher modulus vertebral anchors, endplates and motion segment periphery with gradation to a low modulus core within the motion segment. This closer approximation of the native spinal disk form translates to improvements in prosthesis biomechanical fidelity and longevity. Axial compressive strain induced within a TiNi reinforced Kraton® BioFI™ was found to be linearly proportional to the NiTi helical coil electrical resistance. This neural network capability delivers opportunities to monitor and telemeterize in situ multiaxis joint structural performance and in vivo spine biomechanics.

Reducing water use could impact existing sewer systems but this is not currently well understood. This work describes a new flow and wastewater quality model developed to investigate this impact. SIMDEUM WW® was used to generate stochastic appliance-specific discharge profiles for wastewater flow and concentration, which were fed into InfoWorks® ICM to quantify the impacts within the sewer network. The model was validated using measured field data from a sewer system in Amsterdam serving 418 households. Wastewater concentrations of total suspended solids (TSS), chemical oxygen demand (COD), total Kjeldahl nitrogen (TKN) and total phosphorus (TPH) were sampled on an hourly basis, for one week. The results obtained showed that the InfoWorks® model predicted the mass flow of pollutants well (R-values 0.69, 0.72 and 0.75 for COD, TKN and TPH respectively) but, due to the current lack of a time-varying solids transport model within InfoWorks®, the prediction for wastewater concentration parameters was less reliable. Still, the model was deemed capable of analysing the effects of three water conservation strategies (greywater reuse, rainwater harvesting and water-saving appliances) on flow, nutrient concentrations, and temperature in sewer networks. Results show through a 62% reduction in sewer flow, COD, TKN and TPH concentrations increased by up to 111%, 84% and 75% respectively, offering more favourable conditions for nutrient recovery.

Objective: Evaluation of postoperative sensitivity in posterior teeth restored with CIMMO DTA® liner, in composite resin restorations. Method: Randomized, double-blind clinical trial. There were 20 male participants, with an average age of 20 years. Totaling 70 treated teeth, divided into two groups: Group R (Control n=35), treated with standard restorative procedure. Group D (Study n=35), treated using CIMMO DTA® cement, as an indirect liner, the assessment was performed by a masked examiner. Sensitivity tests were performed before the restorative procedure, at 48 hours, 7, 15 and 30 days after the procedure. The analysis included the pain caused by thermal stimulation: Cold Test (FT); à Horizontal (TPH) and vertical (TPV) percussion; Bite Test (TM) and Pain Decline Time (DD) is measured. Measured through Visual Analog Scale (EVA). Results: All participants returned for postoperative sensitivity tests. Group D had lower pain sensitivity than group R: on the second day, for TPH with p=0.041; TPV with p=0.036; on the fifteenth day, for TPH with p=0.043; TM p=0.021 and on the thirtieth day, for TF with p=0.011. Conclusion: CIMMO DTA® cement, as an indirect liner in dental restorations, decreased postoperative sensitivity.

Recently, two oral-administered peptide pharmaceuticals, semaglutide and octreotide, have been developed and are considered as a breakthrough in peptide and protein drug delivery system development. In 2019, the Food and Drug Administration (FDA) approved an oral dosage form of semaglutide developed by Novo Nordisk (Rybelsus®) for the treatment of type 2 diabetes. Subsequently, the octreotide capsule (Mycapssa®), developed through Chiasma’s Transient Permeation Enhancer (TPE) technology, also received FDA approval in 2020 for the treatment of acromegaly. These two oral peptide products have been a significant success; however, a major obstacle to their oral delivery remains the poor permeability of peptides through the intestinal epithelium. Therefore, gastrointestinal permeation enhancers are of great relevance for the development of subsequent oral peptide products. Sodium salcaprozate (SNAC) and sodium caprylate (C8) have been used as gastrointestinal permeation enhancers for semaglutide and octreotide, respectively. Herein, we briefly review two approved products, Rybelsus® and Mycapssa®, and discuss the permeation properties of SNAC and medium chain fatty acids, sodium caprate (C10) and C8, focusing on Eligen technology using SNAC, TPE technology using C8, and gastrointestinal permeation enhancement technology (GIPET) using C10.

In order to evaluate in vitro the suitability of various prosthetic materials for endothelial seeding, human endothelial cells derived from the umbilical vein were placed in direct contact with a variety of polymers. As a control, endothelial cells were cultured in the absence of material. After 24, 48, 72 and 96 hours, the cells were counted and a viability test with neutral red was performed. Assays of 6-keto prostaglandinFla (6-keto-PGF1a), tissue plasminogen activator (tPA) and plasminogen activator inhibitor 1 (PAI-1) were carried out on the supernatants. The cell counting technique demonstrated growth inhibition in the cell populations in contact with Woven Dacron® and Double Velour Dacron® in comparison with control cell cultures. Vital staining with neutral red, always sharply positive in the controls, was weak in the cells placed in contact with the materials. The 6-keto-PGF1a concentration in the supernatant was similar to the control level in the populations in contact with Woven Dacron® and Double Velour Dacron®. The tPA synthesis was higher in the cells exposed to the assayed materials compared to control values, while the PAI-1 concentration in the supernatant was lower in the cultures in contact with all materials.

In this study, the use of spray-drying technology for encapsulating Flavourzyme® (protease–peptidase complex) was evaluated to overcome the limitations (low encapsulation efficiency and no large-scale production) of other encapsulation processes. To the best of our knowledge, spray drying has not been applied previously for the immobilization of this enzyme. Firstly, bovine serum albumin (BSA), as a model protein, was encapsulated by spray drying in chitosan and tripolyphoshate (TPP) cross-linked-chitosan shell matrices. The results showed that the chitosan–TPP microcapsules provided a high encapsulation efficiency and better protein stability compared to the non-crosslinked chitosan microcapsules. The effect of enzyme concentration and drying temperature were tested during the spray drying of Flavourzyme®. In this regard, an activity yield of 88.0% and encapsulation efficiency of 78.6% were obtained with a concentration of 0.1% (v/v) and an inlet temperature of 130 °C. Flavourzyme®-loaded chitosan microcapsules were also characterized in terms of their size and morphology using scanning electron microscopy and laser diffractometry.

This study was designed to evaluate the flavonoid, saponin (TSC) and phenolic (TPC) contents and in vitro antioxidant activity of the crude (CME) and dry extracts and fractions of Medicago lupulina L. aerial parts. A validated RP-HPLC method led to quantitation of flavonols (kaempferol, quercetin and myricetin) and flavones (apigenin and luteolin) in the hydrolyzed extract. TSC and TPC were assayed spectrophotometrically at 560 and 760 nm, respectively. The antioxidant activity of the CME and the dry fractions were followed in vitro by DPPH free radical and ferric reducing antioxidant power (FRAP) methods. The flavonoid content of CME was 1.27 mg/g dw. The prevailing flavonoids were luteolin and myricetin, at concentrations of 0.37 and 0.36 mg/g dw, respectively. TSC and TPC were detected in CME at the level of 90.4 mg ESE/g dw, and 12.9 mg GAE/g dw, respectively. In the DPPH and FRAP tests, the CME exhibited antioxidant capacity with TEAA and FRAP values of 45.4 μmol Trolox®/g dw and 0.2 mmol Fe2+/g dw, respectively. The diethyl ether dry fraction was the most valuable one, showing the highest antioxidant activity (TEAA = 726.1 μmol Trolox®/g dw, FRAP = 2349.4 μmol Fe2+/g dw) that was in accordance with its high TPC (162.4 mg/g dw).

Background: Ampicillin (AMP) is frequently administered parenterally in critically ill patients with meningitis or endocarditis. Many of them require parallel infusion of total parenteral nutrition (TPN) admixtures. The aim of the study was to determine the physicochemical stability of AMP in TPN admixtures. Methods: AMP was added to two formulations of TPN admixtures differing in the lipid emulsion (Lipofundin® MCT/LCT 20% or LIPIDem®). Samples were stored at 4 ± 1 °C with light protection, and at 25 ± 1 °C with and without light protection to assess the impact of temperature and light on formulation stability. Every 24 h the pH, zeta potential, mean droplet diameter (MDD) of a lipid emulsion, and AMP concentration using HPLC method were determined. The assessment of stability and compatibility of TPN admixtures with vitamins and trace elements was carried out immediately after preparation and after 24 h of storage. Results: The addition of AMP as well as vitamins and trace elements to the TPN admixtures did not affect their physical stability. An increase in the pH value of approx. 0.6 and reduction of zeta potential were observed. The MDD of the lipid emulsions was below the limit of 500 nm (dynamic light scattering (DLS) method) and no fat droplets greater than 525 nm were observed (light diffraction (LD) method). The content of AMP after the first 24 h was within the acceptable limit of 90% for TPN admixtures stored at 4 ± 1 °C and 25 ± 1 °C with light protection. Conclusions: The results showed that co-administration of AMP in the same bag with TPN admixture at the tested dose is possible when used ex tempore and with light protection.

Background HIV and Syphilis are common STIs, which have become a concern and burden on healthcare systems, as many infections go untreated and lead to potentially serious complications. HIV is usually diagnosed with Western blot, PCR, and p24 antigen testing. Whereas, Syphilis is mainly diagnosed through clinical findings and serologic testing. The Medical Commission Department (MC) under MOPH is responsible for screening all newcomers to Qatar, aiming to keep the country free from serious infectious diseases. Objective We aimed to evaluate the diagnostic efficiency of the protocols used in the MC for screening HIV and Syphilis infections. Methods We conducted a retrospective study of samples analyzed by 4th Generation ARCHITECT® HIV Ag/Ab Combo and Rapid Plasma Reagin (RPR) between January to December 2019. ARCHITECT® HIV Ag/Ab Combo positive samples were confirmed by INNO-LIA™ HIVI/II and RT-PCR. RPR-reactive samples were confirmed by ARCHITECT® Syphilis Treponema pallidium Antibody (Syphilis TPA) assay. Results For HIV, data were collected from 585,587 individuals, of which 595 (0.1%) were positive by the ARCHITECT® HIV Ag/Ab Combo (Analyzer A). When all initially positive sera were re-tested on newly collected blood samples using different ARCHITECT® HIV Ag/Ab Combo analyzer (analyzer B), 99.8% (594/595) of samples were also positive, suggesting high reproducibility. The positive predictive value (PPV) between ARCHITECT® HIV Ag/Ab Combo and the INNO-LIA™ HIVI/II confirmatory assay was 31.8%. The PPV between ARCHITECT® HIV Ag/Ab Combo and HIV-PCR assay was 26.8%. Retrospective data for Syphilis were collected from a total of 97,298 individuals who visited the MC, of which 198 (0.20%) were initially positive by RPR. The PPV between RPR and Syphilis TPA confirmatory assay was 36.6%. Conclusion Despite the high rate of false positivity using ARCHITECT® HIV Ag/Ab Combo and RPR screening assays, both assays have proven to be highly effective as screening testing methods.

Analisis parameter dalam leachate memerlukan analisis khusus maka diperlukan suatu kegiatan monitoring sederhana terhadap kualitas leachate yang dihasilkan oleh suatu TPA. Kegiatan monitoring dapat dilakukan dengan melakukan pengukuran terhadap parameter fisika, kimia dalam leachate. Response Surface Methodology di dalam penelitian ini menggunakan Minitab® 17. Minitab® 17 adalah salah satu program komputer yang dirancang untuk melakukan pengolahan statistik. Hal ini mempermudah user dalam memahami hubungan antara parameter. Tujuan Penelitian ini adalah untuk mengetahui adanya korelasi Antara Parameter Fisika dan Kimia dalam leachate Parameter fisika seperti Suhu dan TSS. Parameter biologi seperti E.Coli. Dari hasil penelitian terdapat korelasi antara Suhu, pH dan TSS Terhadap, E.coli. Setiap parameter saling berkorelasi dan menunjukan suatu hubungan yang positif yaitu bila salah satu parameter meningkat maka parameter lain juga akan meningkat, dan terdapat nilai koefisien determinasi R-Squared yang tinggi untuk semua hubungan fungsional. Berdasarkan hasil penelitian diperoleh suatu parameter monitoring lain yang jauh lebih sederhana, mudah di aplikasikan di TPA yaitu Suhu, pH dan TSS untuk mengetahui nilai konsentrasi dan E.Coli dengan Response Surface Methodology dan menggunakan aplikasi Minitab® 17.

Background and aims High consumption of red chili pepper has been shown to be a risk factor for gallbladder cancer (GBC) in Chilean women. However, genetic factors in addition to this and other environmental factors may also be associated with an increased risk of GBC. We aimed to examine the associations of cytochrome P450 1A1 (CYP1A1), glutathione S-transferase class mu (GSTM1), and tumor protein p53 (TP53) polymorphisms with the risk of GBC in Chilean women. Methods A hospital-based case-control study of 57 patients with GBC, 119 patients with gallstones, and 70 controls was conducted. The statuses of the polymorphisms CYP1A1 rs4646903, CYP1A1 rs1048943, and TP53 rs1042522 were assayed using Custom Taqman® SNP Genotyping Assays and TaqMan®SNP Genotyping Assay. GSTM1 deletion polymorphism was detected by allele-specific PCR analysis. Results No significant differences in the genotypic or allelic frequencies in the CYP1A1, GSTM1, and TP53 polymorphisms were found between the three groups. Conclusion These genetic variants were not related to an increased risk of GBC in Chilean women. Other polymorphisms, such as red-chili-pepper-related polymorphisms, may contribute to the development of GBC in Chilean women.

Butyrate affects cell proliferation, differentiation, and motility. Butyrate inhibits histone deacetylase (HDAC) activities and induces cell-cycle arrest and apoptosis. TP53 is one of the most active upstream regulators discovered by ingenuity pathways analysis (IPA) in our RNA-sequencing data set. TP53 signaling pathway plays key role in many cellular processes. TP53 pathway and their involvement in cellular functions modified by butyrate treatment were scrutinized in this report by data mining the RNA-sequencing data using IPA (Ingenuity System®). The TP53 mechanistic pathway targets more than 600 genes. Downstream analysis predicted the activation of the TP53 pathway after butyrate treatment. The data mining also revealed that nine transcription factors are downstream regulators in TP53 signaling pathways. The analysis results also indicated that butyrate not only inhibits the HDAC activities, but also regulates genes encoding the HDAC enzymes through modification of histones and epigenomic landscape.

Due to the deliberate disposal of industrial waste, a great amount of petroleum hydrocarbons pollute the soil and aquatic environments. Bioremediation that depends on the microorganisms in the removal of pollutants is more efficient and cost-effective technology. In this study, five rhizobacteria were isolated from Phragmites australis roots and exposed to real wastewater from Al-Daura refinery with 70 mg/L total petroleum hydrocarbons (TPH) concentration. The five selected rhizobacteria were examined in a biodegradation test for seven days to remove TPH. The results showed that 80% TPH degradation as the maximum value by Sphingomonas Paucimobilis as identified with Vitek® 2 Compact (France).

This paper describes the application of poly(ether-block-amide) polymers, so-called Pebax, in fused filament fabrication (FFF). Pebax® is a thermoplastic elastomer (TPE), a copolymer based on rigid polyamide and soft polyether blocks. By variation of the blocks, unique properties such as soft or rigid behaviour are tailored without additional additives and plasticisers. Pebax®Rnew® polyamide blocks are bio-based and made from castor beans that allow the design of sustainable applications. In this study, two types of Pebax were selected, processing parameters were characterised, filaments were extruded and applied to FFF printing, and the final mechanical characteristics were determined. Both types were suitable for FFF processing with improved process stability due to less shear thinning and good mechanical performance. The connection strength between the grades was also described in the design context for complex parts with tailored soft or hard regions. Combining the two materials in one design is a promising concept, and the adhesion strength is close to the strength in the Z-direction of the flexible Pebax®Rnew®35R53 grade.

Introduction ITP is an autoimmune disorder characterized by platelet destruction and impaired production. Some patients with ITP are refractory or unresponsive to existing therapies, indicating significant unmet medical needs. An assessment of current real-world treatment patterns among patients with ITP should enhance understanding of the ITP disease population and current unmet need. Here we present real-world data describing the ITP population in the United States in terms of demographic and clinical characteristics and use of available ITP therapies. Methods This was a longitudinal, retrospective, observational cohort study utilizing data from the Optum® Clinformatics® claims database. Patients with newly diagnosed ITP were included; defined as patients with ≥1 inpatient claim or ≥2 outpatient claims within 90 days of ITP-related diagnosis codes (International Classification of Diseases [ICD], Ninth and Tenth Revision; ICD-9: 287.3x; ICD-10: D69.3x or D69.4x) between October 1, 2015 and March 31, 2020. The index date was the date these criteria were met. Patients were excluded if they had <12 months continuous coverage or any record of thrombopoietin receptor agonist (TPO-RA) use before the index date. All patients were required to have ≥1 prescription during follow-up. Primary ITP was defined as patients who met the inclusion criteria and also had no ICD codes for secondary ITP, and no causes of secondary ITP in an identified time period prior to the index date. Here we describe clinical characteristics and medication use among newly diagnosed patients with primary ITP and the proportion of patients who went on to develop persistent (90 days to <12 months) or chronic (≥12 months) ITP. Results There were 19,376 newly diagnosed patients who met inclusion criteria; 15,798 (82%) had primary ITP and 3,578 (18%) had secondary ITP. Among patients with primary ITP, median (min-max) duration of follow-up was 16 (0-55) months and 61% had ≥12 months of follow-up (median duration: 26 months). At baseline, mean (standard deviation [SD]) age was 67 (16) years and 53% of patients were male. Mean (SD) Charlson Comorbidity Index (CCI) was 0.94 (1.41) and 11% of patients fell into a CCI category ≥3. The most common comorbid conditions were hypertension (60%), cardiovascular disease (37%), diabetes (29%), and anemia (27%). The most commonly used concomitant medications were lipid-lowering medications (44%), pain medications (24%), corticosteroids (23%), and anticoagulants (11%). During follow-up, use of these medications increased (lipid-lowering medications [46%], pain medications [38%], corticosteroids [39%], and anticoagulants [15%]). The most commonly used ITP medications during follow-up were corticosteroids (33%), rituximab (3.3%), TPO-RA (2.8%), immunosuppressants (2.6%), intravenous immunoglobulin (2.5%), and immunomodulators (2.4%). There were 1935 (12%) deaths during follow-up, and 87 (0.6%) patients underwent a splenectomy. Of the patients with ≥12 months of follow-up (n=9610), 16% and 32% developed persistent ITP and chronic ITP, respectively. Among the patients with persistent/chronic ITP and ≥12 months of follow-up, 242 (5.3%) received TPO-RA during follow-up. Of these, 93% concomitantly received ≥1 additional ITP treatment-related medication (TRM), 73% of the patients who received TPO-RA and ≥1 other TRM received ≥2 additional ITP TRMs, and 68% of patients who received TPO-RA and ≥2 other TRMs received ≥3 additional ITP TRMs. Of the 242 patients with ≥12 months of follow-up and persistent/chronic ITP who received TPO-RA, 55 (23%) either received an additional TPO-RA therapy or received another newly initiated ITP-related medication ≥30 days after starting TPO-RA. Almost half of the patients with primary ITP developed persistent/chronic disease. However, only a small percentage of these patients in the Optum® Clinformatics® claims database with no history of TPO-RA use received TPO-RA during follow-up. The majority of patients treated with TPO-RA received ≥1 other ITP TRM during follow-up and nearly 1 in 4 persistent/chronic patients receiving TPO-RA required ≥1 additional TPO-RA or switched to another ITP medication. Conclusion Even with the availability of many ITP drug therapies, there is still a considerable current unmet need for effective treatments among patients with primary ITP. Tubular composites are widely used in many industrial applications, and there is need to use new material and reliable manufacturing processes to improve the performance and process aspects. The current research presents a detailed study to understand the flexure response of rectangular tubular composites based on thin ply carbon fibres and Elium® resin. Another aim was to understand the failure mechanisms of novel tubular thermoplastic composite systems and carry out a baseline comparison with Epoxy-based tubular systems. In the current research, a bladder-assisted resin transfer moulding process was used to manufacture hollow thermoplastic composite tubes, and the bending behaviour of thin ply carbon (TPC) composite parts with novel Elium® (EL) and Epoxy (EP) resin as the matrix material was studied using a detailed experimental study. A testing method with optimized support span and a saddle was used to carry out three-point bending tests on the tubular composite structures. The TPC/EL composite tubes have shown 10% higher bending strength, with a noticeable increase in deformation due the presence of extended plasticity attributes for acrylic Elium resin. Failure mechanisms studied with the detailed microscopic investigation have shown severe catastrophic failure for epoxy-based composite tubes; however, acrylic Elium®-based composite tubes have shown different damage modes such as fibre splitting, resin infragmentation, and fibre resin-interfacial cracking.

The application of processing waste by-products along with probiotics is an interesting choice to confer potential functional aspects to food products. This study was designed to investigate the nutritional capacity of freeze-dried mango peel powder (MPP) and banana peel powder (BPP) in the presence of a mixture of three probiotic species (1% of each of three probiotics (Lacticaseibacillus casei (431®), Lacticaseibacillus rhamnosus (LGG®) and Bifidobacterium subsp. Lactis (Bb-12®)) as sources of additional nutrients and prebiotics in fresh and rehydrated freeze-dried (RFD) yogurts for 28 days of refrigerated storage. The net count of probiotics in yogurt fortified with MPP and BPP increased by at least 1 log CFU/g after 4 weeks of refrigerated storage. Adding fruit peel powder (FPP) significantly (p < 0.05) increased fat, ash, and protein contents in both fresh and RFD yogurts in comparison with the control yogurt. Similarly, the total phenolic contents (TPC) and antioxidant activity (AOA) was enhanced significantly (p < 0.05). The TPC reached 2.27 ± 0.18 and 2.73 ± 0.11 mg GAE/g in RFD enriched with BPP and MPP compared to a TPC of 0.31 ± 0.07 mg GAE/g in the control. Additionally, yogurt samples enriched with BPP (Y-5) and MPP (Y-6) demonstrated 12% more sugar contents than non-fortified yogurts (Y-1). Higher titratable acidity and lower pH values were also recorded in the RFD yogurt. Significant differences (p < 0.05) in the color parameters were detected in both fresh and RFD yogurts with reduced brightness (L*) and increased redness (a*) of the product. These findings demonstrated the suitability of MPP and BPP in yogurt formulations to optimize the advantages of such synbiotic products with higher availability of phenolic compounds.

The intensification of total phenolic compound (TPC) extraction from blood orange peels was optimized using a novel green infrared-assisted extraction technique (IRAE, Ired-Irrad®) and compared to the conventional extraction using a water bath (WB). Response surface methodology (RSM) allowed for the optimization of ethanol concentration (E), time (t), and temperature (T) in terms of extracted TPC and their antiradical activity, for both WB extraction and IRAE. Using WB extraction, the multiple response optimums as obtained after 4 h at 73 °C and using 79% ethanol/water were 1.67 g GAE/100 g for TPC and 59% as DPPH inhibition percentage. IRAE increased the extraction of TPC by 18% using 52% ethanol/water after less than 1 h at 79 °C. This novel technology has the advantage of being easily scalable for industrial usage. HPLC analysis showed that IRAE enhanced the recovery of gallic acid, resveratrol, quercetin, caffeic acid, and hesperidin. IR extracts exhibited high bioactivity by inhibiting the production of Aflatoxin B1 by 98.9%.

Two distinct types of trans fatty acids (TFA) are found in the diet. Industrial TFA such as elaidic acid (EA) have deleterious effects on metabolic risk factors, and oppositely ruminant TFA including trans-palmitoleic acid (TPA) may have beneficial effects. The objective is to evaluate the taste preference between EA, TPA, lecithin or water. In this study, 24 female C57BL/6 mice were microchipped and placed in two separate IntelliCages®. Nano encapsulated TFA or lecithin were added to drinking water in different corners of the cage with normal diet. The study was carried out over 5 weeks, during which mice were exposed to water only (weeks 1 and 3), TFA or lecithin (week 2), and EA or TPA (weeks 4 and 5). Mice weights, corner visits, nose pokes (NP), and lick number were measured each week. The results demonstrated that mice consume more TFA, either EA or TPA, compared with lecithin. In addition, the mice licked more EA compared with TPA in one cage; conversely, in the other cage they licked more TPA compared with EA. However, when TFA positions were swapped, mice had equal licks for EA and TPA. In sum, mice preferred TFA, in equal matter compared with controls; therefore, the results demonstrate the potential for TFA-type substitution in diet.

Background: Molecular diagnosis has become an established tool in the characterisation of adult soft-tissue sarcomas (STS). FoundationOne® Heme analyses somatic gene alterations in sarcomas via DNA and RNA-hotspot sequencing of tumour-associated genes. Methods: We evaluated FoundationOne® Heme testing in 81 localised STS including 35 translocation-associated and 46 complex-karyotyped cases from a single institution. Results: Although FoundationOne® Heme achieved broad patient coverage and identified at least five genetic alterations in each sample, the sensitivity for fusion detection was rather low, at 42.4%. Nevertheless, potential targets for STS treatment were detected using the FoundationOne® Heme assay: complex-karyotyped sarcomas frequently displayed copy-number alterations of common tumour-suppressor genes, particularly deletions in TP53, NF1, ATRX, and CDKN2A. A subset of myxofibrosarcomas (MFS) was amplified for HGF ( n = 3) and MET ( n = 1). PIK3CA was mutated in 7/15 cases of myxoid liposarcoma (MLS; 46.7%). Epigenetic regulators (e.g. MLL2 and MLL3) were frequently mutated. Conclusions: In summary, FoundationOne® Heme detected a broad range of genetic alterations and potential therapeutic targets in STS (e.g. HGF/MET in a subset of MFS, or PIK3CA in MLS). The assay’s sensitivity for fusion detection was low in our sample and needs to be re-evaluated in a larger cohort.

Allele frequencies for the 15 STR loci in the AmpFlSTR® Identifiler® and statistical parameters were estimated from a sample of 233 unrelated individuals from different area of Nepal. A total of 161 alleles were found, with corresponding allelic frequencies ranging from 0.0021 to 0.4077. The MP, PD, PIC, PE, TPI, Ho and He ranged from 0.0282 to 0.1511, 0.8489 to 0.9717, 0.6478 to 0.8700, 0.4546 to 0.7631, 1.7651 to 4.3148, 0.7167 to 0.8841 and 0.7009 to 0.8833 respectively. Deviations from the Hardy–Weinberg Equilibrium were observed for D16S539, D18S51, D21S11 and TPOX Markers. One new allele has been detected in the process. Eudraginated polymer blends: A potential oral controlled drug delivery system for theophylline Sustained release (SR) dosage forms enable prolonged and continuous deposition of the drug in the gastrointestinal (GI) tract and improve the bioavailability of medications characterized by a narrow absorption window. In this study, a new strategy is proposed for the development of SR dosage forms for theophylline (TPH). Design of the delivery system was based on a sustained release formulation, with a modified coating technique and swelling features aimed to extend the release time of the drug. Different polymers, such as Carbopol 71G (CP), sodium carboxymethylcellulose (SCMC), ethylcellulose (EC) and their combinations were tried. Prepared matrix tablets were coated with a 5 % (m/m) dispersion of Eudragit (EUD) in order to get the desired sustained release profile over a period of 24 h. Various formulations were evaluated for micromeritic properties, drug concentration and in vitro drug release. It was found that the in vitro drug release rate decreased with increasing the amount of polymer. Coating with EUD resulted in a significant lag phase in the first two hours of dissolution in the acidic pH of simulated gastric fluid (SGF) due to decreased water uptake, and hence decreased driving force for drug release. Release became faster in the alkaline pH of simulated intestinal fluid (SIF) owing to increased solubility of both the coating and matrixing agents. The optimized formulation was subjected to in vivo studies in rabbits and the pharmacokinetic parameters of developed formulations were compared with the commercial (Asmanyl®) formulation. Asmanyl® tablets showed faster absorption (tmax 4.0 h) compared to the TPH formulation showing a tmax value of 8.0 h. The Cmax and AUC values of TPH formulation were significantly (p < 0.05) higher than those for Asmanyl®, revealing relative bioavailability of about 136.93 %. Our study demonstrated the potential usefulness of eudraginated polymers for the oral delivery of the sparingly soluble drug theophylline.

We present a unique case of a thigh compartment syndrome that occurred after overnight catheter delivered Tissue plasminogen activator (tPA) thrombolysis of an acutely thrombosis femoral-to above knee popliteal artery Propaten® PTFE (WL Gore & Associates, Flagstaff, AZ) bypass graft. The condition was treated by emergency fasciotomy and the patient recovered uneventfully.

TPL (Tachypleus plasma lectin)-1 was purified by using a Sepharose column and TPL-2 was purified from an LPS–Sepharose (LPS coupled to Sepharose matrix) affinity column, as described previously [Chiou, Chen, Y.-W., Chen, S.-C., Chao and Liu (2000) J. Biol. Chem. 275, 1630–1634] and the corresponding genes were cloned [Chen, Yen, Yeh, Huang and Liu (2001) J. Biol. Chem. 276, 9631–9639]. In the present study, TPL-1 and -2 were produced in yeast, and the recombinant proteins secreted into the media were purified and characterized. The proteins show specific PGN (peptidoglycan)- and LPS-binding activity, suggesting a role in trapping Gram-positive and Gram-negative bacteria respectively in innate immunity. Using BIAcore® assays, the dissociation constant for the TPL-1–PGN complex was measured as 8×10−8 M. Replacement of Asn74, the N-glycosylation site of TPL-1, with Asp abolishes the PGN-binding affinity, whereas the unglycosylated TPL-2 N3D mutant retains LPS-binding activity. DTT (dithiothreitol) treatment to break disulphide linkages abrogates TPL-2 activity but does not interfere with TPL-1 function. Cys4 in TPL-2 may form an intermolecular disulphide bond, which is essential for activity. As a result, the TPL-2 C4S mutant is inactive and is eluted as a monomer on a non-reducing gel. TPL-2 C6S is active and forms a non-covalently linked dimer. A model describing TPL-2 binding with LPS is proposed. These two plasma lectins that have different ligand specificities can be used for the detection and discrimination of bacteria and removal of endotoxins.

Objectives To evaluate long-term outcome using the BioMedtrix™ TPLO Curve® plate in dogs with cranial cruciate ligament disease (CrCLd) treated by tibial plateau leveling osteotomy (TPLO). Study design Retrospective case study. Animals Dogs (n = 323, 337 stifles). Methods Medical records were searched for dogs presented with CrCLd and treated by TPLO with the BioMedtrix™ TPLO Curve® plate for 3.5 mm screws between March 2018 and December 2020. Tibial plateau angles (TPA) were measured on preoperative, postoperative, and follow-up radiographs. Changes in TPA between postoperative and follow-up radiographs (ModTPA) were calculated. Radiographic bone healing was scored. Complications were evaluated. Surgical site infections (SSI) were compared to a control group of dogs treated with the Synthes™ TPLO plate between January and December 2017. Owners of both groups were contacted by telephone at least 1 year postoperatively. Results The BioMedtrix™ group was composed of 237 dogs (248 stifles), the control group was composed of 86 dogs (89 stifles). In the BioMedtrix™ group, radiographic follow-up was performed at a median of 48 days. Average ModTPA was 1.2°. Bone healing was graded as complete, good, poor, and none in 18%, 62%, 20%, and 0%, respectively. At a median of 786 days postoperatively, minor and major postoperative complications were observed in 6 (2.4%) and 32 (12.9%) cases in this group, respectively. During the first year following the surgery, 23 (9.3%) and 12 (13.5%) cases suffered a SSI, of which 12 (4.8%) and 7 (7.9%) had their implant explanted in the BioMedtrix™ group and the control group, respectively. There was no significant difference between groups for SSI and implant explantations (p = 0.31 and p = 0.29, respectively). Conclusion The BioMedtrix™ TPLO Curve® plate provided a reliable fixation system for osteotomy healing after TPLO. Bone healing and long-term complication rates were similar to previous studies using other implants. SSI rates were similar between the BioMedtrix™ group and the control group. The antimicrobial HyProtect® coating of the plate did not reduce SSI in this study. Clinical significance The BioMedtrix™ TPLO Curve® plate can be safely used for TPLO. The value of the antimicrobial HyProtect® coating of the plate may be questioned, as SSI rate was not lower in this study compared to the control group or previous reports.

The use of biostimulants in modern agriculture has rapidly expanded in recent years, owing to their beneficial effects on crop yield and product quality, which have come under the scope of intensive research. Accordingly, in the present study we appraised the efficacy of two plant-derived biostimulants, the legume-derived protein hydrolysates Trainer®® (PH), and the tropical plant extract Auxym®® (TPE) on two lettuce cultivars (green and red salanova®®) in terms of morpho-physiological and biochemical traits (primary and secondary metabolites). The two cultivars differed in their acquisition capacity for nitrate and other beneficial ions, their photosynthetic and transpiration rates, and their ability to synthetize and accumulate organic acids and protective metabolites. The biostimulant effect was significant for almost all the parameters examined but it was subjected to significant cultivar × biostimulant interactions, denoting a cultivar-dependent response to biostimulant type. Notwithstanding this interaction, biostimulant application could potentially improve the yield and quality of lettuce by stimulating plant physiological processes, as indicated by the SPAD index (leaf chlorophyll index), ACO2 (assimilation rate), E (transpiration), and WUEi (intrinsic water use efficiency), and by increasing concurrently the plant mineral content (total N, K, Ca, Mg) and the biosynthesis of organic acids (malate, citrate), phenols (caffeic acid, coumaroyl quinic acid isomer 1, dicaffeoylquinic acid isomer 1), and flavonoids (quercetin-3-O-glucuronide, quercetin-3-O-glucoside). Biostimulant action may facilitate the bio-enhancement of certain lettuce cultivars that are otherwise limited by their genetic potential, for the accumulation of specific compounds beneficial to human health.

Estudou-se o desenvolvimento na área de cria de 14 núcleos com quatro favos a partir da divisão de nove colméias de abelhas Apis mellifera africanizada, distribuídos em dois tratamentos: TPL - núcleos alimentados com açúcar invertido + 3,5ml do suplemento de aminoácido vitamínico (Promotor L®), composto por seis núcleos e o TAI - núcleos alimentados com açúcar invertido, composto por oito núcleos. Os núcleos foram alimentados semanalmente em alimentadores individuais tipo bandeja, e as avaliações realizadas em quatro períodos, totalizando 74 dias Os tratamentos não apresentaram diferença significativa, sendo que, numericamente, o TPL apresentou área de cria inferior ao TAI (233,63 vs. 273,02cm², respectivamente). Quanto aos períodos o quarto período foi superior (P<0,05) ao primeiro e ao segundo, sendo que o terceiro período não apresentou diferença significativa (P<0,05) em relação aos demais. A utilização do TAI foi economicamente mais favorável em relação ao TPL em R$0,21 em relação ao custo para produção de 1kg de alimento.

Abstract Glyphosate has long be suspected to promote endocrine disruptive and even carcinogenic effects, but little is known about its effect at different concentrations on normal and on cancer thyroid cells. Since the endocrine system responds to low chemical concentrations and abnormal cells may have different sensitivity, it is essential to determine the effects of this compound at different levels, including the Acceptable Occupational Exposure Level (AOEL) and the Acceptable Daily Intake (IDA) doses. We used two human thyroid-derived cell lines strains, Nthy-ori 3-1 (from thyroid normal follicular cells) and TPC-1 (from papillary carcinoma) to test 15 different concentrations of Roundup® Original DI, ranging from 6.5 µg/L to 6.500 µg/L, including AOEL and IDA according to the Technical Note Process No. 25351. 056754/2013-17 of the National Health Surveillance Agency (ANVISA) in Brazil. Trypan blue assay and Cell Counting Kit – 8 (CCK-8) were employed to evaluate cell viability and cytotoxicity. Exposures were made for 24 and 48 hours in technical and biological triplicates. We observed a decrease in the number of viable cells at the 24h exposure compared to 48h in both strains. Exposure of Nthy-ori 3-1 and TPC-1 cells for 24h at the AOEL concentration (160µg/L) caused death of 52% and 58% of the cells, respectively, whereas IDA (830µg/L) resulted in 43% e 58% cell death. These data evidence an important disruptive effect of Roundup Original DI® at the acceptable concentrations. After 48h of exposure, AOEL and IDA concentrations caused a lower number of Nthy-ori 3-1 (19% e 15%) and TPC-1 (29% e 23%) dead cells, respectively, suggesting both thyroid strains have compensatory mechanisms that protect them from longer exposures to the herbicide. The CCK-8 assay, on the contrary, showed a proliferative effect on small concentrations: 6.5 µg/L increased the number of viable Nthy-ori 3-1 and TPC-1 cells to 113% and 118%, respectively, at 24h, and this stimulatory effect persisted after 48h of exposure. In contrast, AOEL and IDA concentrations produced a reduction in the number of both strains cells, likewise observed with the Trypan blue assay. Our data demonstrate that thyroid cells exposed to Roundup Original DI® present a non-monotone dose-response curve, which is a well-known characteristic of an endocrine disruptor. The proliferative effect produced on thyroid cells, especially TPC-1 mutant cells, by very low concentrations of this glyphosate-based herbicide widely used around the world, suggests that it may play a role in the increasing incidence rate of thyroid nodules and/or cancer that have been observed worldwide. During therapeutic plasma exchange (TPE), a large volume of a patient’s plasma is separated from the cellular components of the blood and replaced with appropriate fluids to remove circulating plasma components and disease mediators such as antibodies, cholesterol, toxins and drugs. Automated TPE systems can be categorised into two distinct groups based on the method of plasma separation: centrifugal TPE (cTPE) and membrane filtration TPE (mTPE). While technical and performance parameters for various TPE systems have been reported previously, the recent publication of several head-to-head studies directly comparing the Spectra Optia® cTPE system (Terumo BCT, Lakewood, CO, US) with mTPE systems has increased the understanding of their performance. This review summarises the comparative information between the systems. Although both device types are well tolerated, the majority of technical variables favour the cTPE system over mTPE systems, with a low risk of clotting events and benefits in plasma removal efficiency and hence procedure time. Platelet loss with the cTPE system was also shown to be similar or reduced compared with mTPE systems. Finally, flow rate requirements limit the use of peripheral venous access with mTPE devices, but not cTPE devices. Conclusively, several important differences exist between cTPE and mTPE systems that may influence treatment decisions.

Because of a waste problem in plastic hose packaging of daily chemical products, this article recommends an improvement to the structure and material of packaging design for these products. Through the use of a Ziploc®-type structure, this re-design achieves the needs for making full use of package contents and for package reloading functionality. TPE rubber replaces traditional PE plastic, making the improved structure more reasonable to produce, while promoting environmental protection.

Nitrogen (N) fertilizers play a crucial role in agriculture, representing a powerful tool for farmers for increasing yields throughout the seasons under both optimal and suboptimal conditions. At the same time, their synthetic/chemical nature could have several influences on ecosystems and human health. For this reason, there is an urgent need to find new and more sustainable means of production to increase plant productivity and optimize nitrogen use. An experiment was conducted in a plastic tunnel to assess the response of baby lettuce crop to the foliar application of three plant biostimulants (PBs): Legume-derived protein hydrolysate (LDPH) ‘Trainer®’, tropical plant extract (TPE) ‘Auxym®’ and seaweed extract (SwE) from Ecklonia maxima ‘Kelpak®’ under different N rates of 0, 10, 20 and 30 kg N·ha−1. The responses of baby lettuce plants were assessed in terms of yield, growth parameters and physicochemical composition of the leaves. The fresh yield of baby lettuce in both biostimulant-treated and untreated plants was positively affected by increasing N rates from 0 to 20 kg N·ha−1, reaching a plateau thereafter indicating luxury N conditions at 30 kg N·ha−1. However, high N fertilizer application (20 and especially 30 kg N·ha−1) resulted in undesirable decreases in antioxidant activities and total ascorbic acid (TAA). Under non-fertilized regimens, foliar PBs application boost growth and yield of baby lettuce in comparison to non-treated plants. Foliar spray with LDPH and especially SwE elicited significant increases in marketable fresh yield (averaging 14%, 6% and 7% at 10, 20 and 30 kg N·ha−1, respectively) compared to TPE and untreated plants. Improved agronomical performance of baby lettuce under optimal (10 kg N ha−1) and especially suboptimal N regimens (0 kg N ha−1) was associated with increasing photochemical efficiency and a better activity of photosystem II (higher Soil Plant Analysis Development-SPAD index and chlorophyllous pigments biosynthesis). The application of LDPH enhanced antioxidant capacity and TAA in baby lettuce leaf and did not increased nitrate content as recorded in SwE and TPE treatments. Overall, plant biostimulants may be considered as a sustainable tool of production to increase leafy vegetable productivity in low fertility soils.

Tomato potato psyllid (TPP), Bactericera cockerelli (Sulc) (Hemiptera: Triozidae), is the vector for “Candidatus Liberibacter solanacearum” (CLso), which causes the serious disease “zebra chip” (ZC) of potatoes. Between 2016 and 2019, a reduced-insecticide approach to control TPP was evaluated. We compared a standard-insecticide weekly spray regime that included Integrated Pest Management (IPM)-compatible insecticides plus JMS Stylet Oil® (JMS) as a wetting agent, with a reduced-insecticide regime where we used the oil on its own on alternate weeks with the insecticide/oil mixtures. Spray programme start dates were determined by: (1) crop scouting; (2) sticky-trap monitoring; and (3) degree-day calculation. Crop scouting combined with a sticky-trap action threshold and degree-day data was an effective method for determining when to start spraying. The most substantial reduction in insecticides was achieved by alternating weekly insecticides with the oil formulation on its own. Sub-samples of TPP from sticky traps situated in the trials tested for CLso confirmed the presence of the bacteria in (commonly known as ‘hot’) TPP throughout the trials. The reduced-treatment approach gave statistically similar levels of ZC to the standard insecticide spray programme.

Intra-articular administration of anti-inflammatory drugs is a strategy that allows localized action on damaged articular cartilage and reduces the side effects associated with systemic drug administration. The objective of this work is to prepare injectable thermosensitive hydrogels for the long-term application of dexamethasone. The hydrogels were prepared by mixing chitosan (CS) and Pluronic-F127 (PF) physically. In addition, tripolyphosphate (TPP) was used as a crosslinking agent. Chitosan added to the mix increased the gel time compared to the pluronic gel alone. The incorporation of TPP into the material modified the morphology of the hydrogels formed. Subsequently, MTS and Live/Dead® experiments were performed to investigate the toxicity of hydrogels against human chondrocytes. The in vitro releases of dexamethasone (DMT) from CS-PF and CS-PF-TPP gels had an initial burst and took more time than that from the PF hydrogel. In vivo studies showed that hydrogels retained the fluorescent compound longer in the joint than when administered in PBS alone. These results suggest that the CS-PF and CS-PF-TPP hydrogels loaded with DMT could be a promising drug delivery platform for the treatment of osteoarthritis.

The aim of this study was to investigate the acute effects of transdermal 17β-estradiol (Estraderm®) on plasma levels of coagulatory and fibrinolytic factors in postmenopausal normotensive and hypertensive women. Eleven normotensive and 13 hypertensive women were included in this placebo-controlled crossover study. In a randomized order each subject was treated with a patch of 100 yig 17β-estradiol or placebo for 24 hours. Serum levels of tissue type plasminogen activator (tPA) activity, plasminogen activator inhibitor I (PAI-1) activity, tPA antigen, PAI-I antigen, FVII, FX, and fibrinogen were assayed after both treatments. There was no significant difference in serum levels of hemostatic variables after treatment with estrogen compared to levels after placebo treatment in either of the groups. Nor was there any measurable difference when comparing hypertensive and normotensive subjects.

Consumption of cocoa (Theobroma cacao L.) and cocoa products is associated with numerous health benefits due to their high levels of polyphenols and antioxidant capacities. In this study, changes of total phenolic contents (TPC) and antioxidant capacities (AC) of raw cocoa powder at four maturity stages, under different fermentation methods and fermentation duration of cocoa beans were investigated. The TPC and AC were measured using Folin–Ciocalteu and Trolox equivalent antioxidant capacity (TEAC)/ABTS assay, respectively. In term of maturity stages, the powder of unfermented cocoa beans harvested from the stage one contained significantly lower levels of TPC (6.39 ± 0.02 g CE/100 g DM) and AC (26.82 ± 0.13 mol TE/100 g DM) than those from the beans harvested from the stage two, three and four. For all maturity stages studied, fermenting cocoa beans increased antioxidant capacities of the raw cocoa powder. Cocoa fermentation using the commercial enzyme Pectinex® Ultra SP-L resulted in lower TPC but higher AC in comparison to those treated without enzymes, however prolonged fermentation time in cocoa beans significantly reduced TPC and AC in the powder. Strong correlations between TPC and AC of fermented samples with (R = 0.923) and without enzyme supplement (R = 0.942) were obtained. Two-way anova analyses showed that changes of TPC and AC of cocoa beans were dependent on maturity stages, fermentation methods and fermentation duration. As a conclusion, fermentation of cocoa beans harvested at the maturity stage two and three was found to be optimum for the high levels of AC and TPC attainment; fermented beans with commercial enzyme could be utilized to reduce labor cost by shortening the fermentation duration.

Zusammenfassung Gegenstand und Ziel Tonometer messen den Augeninnendruck (IOD) nur indirekt, sodass es zu Messungenauigkeiten kommt. Alle Geräte müssen daher durch vergleichende manometrische Messungen für die einzelnen Tierarten kalibriert werden. Im Rahmen dieser Studie wurde das Applanationstonometer Tono-Pen Avia® Vet™ (TPA) für Hund und Katze kalibriert und ein Korrekturfaktor berechnet. Anschließend wurden anhand einer augengesunden Population Referenzwerte bestimmt und der Einfluss von Alter, Körpergewicht und Geschlecht untersucht. Material und Methoden Für die manometrische Studie erfolgte bei jeweils 10 enukleierten Hunde- und Katzenaugen eine vergleichende Messung des IOD mit einem Manometer und dem TPA über den Druckbereich von 5–60 mmHg. Anhand der mittleren prozentualen Abweichung zwischen dem mit dem Manometer und dem TPA gemessenen IOD wurden Korrekturfaktoren abgeleitet. Zur Ermittlung von Referenzwerten wurde bei augengesunden Tieren der IOD mit dem TPA gemessen. Die statistische Analyse eines Einflusses von Alter, Körpergewicht und Geschlecht der Tiere auf den IOD erfolgte mit gemischten Modellen. Ergebnisse Der TPA unterschätzt bei steigendem Druck den IOD zunehmend. Durch Multiplikation des mit dem TPA gemessenen IOD bei Hunden und Katzen mit dem Faktor 1,5 lässt sich der manometrisch bestimmte, reale IOD berechnen. Es wurden 94 Hunde (188 Augen) und 64 Katzen (128 Augen) untersucht. Der Referenzbereich für den mit dem TPA gemessenen IOD beträgt für Hunde 9–18 mmHg und für Katzen 9–20 mmHg. Bei beiden Tierarten hat das Alter einen signifikanten (p = 0,001 bzw. p = 0,008) Einfluss auf den IOD: Mit höherem Alter sinkt der IOD. Schlussfolgerung Der TPA unterschätzt den wahren IOD bei steigendem Druck. Die Nutzung des berechneten Korrekturfaktors ist bei der Umrechnung des tonometrisch gemessenen IOD in den wahren IOD sinnvoll. Klinische Relevanz Der TPA eignet sich gut für die Messung des IOD bei Hunden und Katzen.

SummaryVarious assays have been developed for quantitation of soluble fibrin or fibrin monomer in clinical plasma samples, since this parameter directly reflects in vivo thrombin action on fibrinogen. Using plasma samples from healthy blood donors, patients with cerebral ischemic insult, patients with septicemia, and patients with venous thrombosis, we compared two immunologic tests using monoclonal antibodies against fibrin-specific neo-epitopes, and two functional tests based on the cofactor activity of soluble fibrin complexes in tPA-induced plasminogen activation. Test A (Enzymun®-Test FM) showed the best discriminating power among normal range and pathological samples. Test B (Fibrinostika® soluble fibrin) clearly separated normal range from pathological samples, but failed to discriminate among samples from patients with low grade coagulation activation in septicemia, and massive activation in venous thrombosis. Functional test C (Fibrin monomer test Behring) displayed good discriminating power between normal and pathological range samples, and correlated with test A (r = 0.61), whereas assay D (Coa-Set® Fibrin monomer) showed little discriminating power at values below 10 μg/ml and little correlation with other assays. Standardization of assays will require further characterization of analytes detected.

Absztrakt: Bevezetés: Nemzetközi tapasztalatok alapján a COVID–19-fertőzött, kórházban kezelt betegek több mint 30%-ában vénás, artériás és microvascularis thrombosis, disszeminált intravascularis véralvadási zavar, illetve thromboembolia alakul ki. Ennek laboratóriumi jellemzői a magas D-dimer- és fibrinogénszint, valamint a megnyúlt prothrombinidő és aktivált parciális thromboplastinidő. Módszer: Az esetbemutatások során intenzív osztályon kezelt, COVID–19-fertőzött betegek (n = 3) ClotPro® trombolasztométeres (EX-test, IN-test, FIB-test, RVV-test és TPA-test) eredményeit értékeltük a klinikai állapottal összefüggésben. Eredmények: A ClotPro®-paraméterek procoagulatiót, hypercoagulatiót, fibrinolysist vagy annak „shut down” megnyilvánulását jelezték, és összhangban voltak a hagyományos alvadási tesztekkel, illetve a kompenzált disszeminált intravascularis coagulopathia meghatározásával. Következtetés: Eredményeink magyarázatot adhatnak a vénás thromboemboliás szövődményekre, támogatják a terápiás alvadásgátló kezelés és a thrombocytaaggregáció-gátlás fontosságát. A gyógyszer típusának, adagolási algoritmusának, optimális időtartamának meghatározásához intervenciós klinikai vizsgálatok szükségesek, ezek engedélyeztetése folyamatban van.
