In the present study, a theoretical analysis of hydrogen bond formation of ethylene glycol, thioglycol, dithioglycol with single water molecule has been performed based on structural parameters of optimized geometries, interaction energies, deformation energies, orbital analysis and charge transfer. ab initio molecular orbital theory (MP2) method in conjunction with 6-31+G* basis set has been employed. Twelve aggregates of the selected molecules with water have been optimized at MP2/6-31+G* level and analyzed for intramolecular and intermolecular hydrogen bond interactions. The evaluated interaction energies suggest aggregates have hydrogen bonds of weak to moderate strength. Although the aggregates are primarily stabilized by conventional hydrogen bond donors and acceptors, yet C-H···O, S-H···O, O-H···S, etc. untraditional hydrogen bonds also contribute to stabilize many aggregates. The hydrogen bonding involving sulfur in the aggregates of thioglycol and dithioglycol is disfavoured electrostatically but favoured by charge transfer. Natural bond orbital (NBO) analysis has been employed to understand the role of electron delocalizations, bond polarizations, charge transfer, etc. as contributors to stabilization energy.

The kinetics and mechanism of reduction of N-(2-hydroxyethyl) ethylenediaminetriacetatoiron (III) complex (hereafter [Fe(III)HEDTAOH2]) by thioglycol (hereafter RSH) has been studied spectrophotometrically in a bicarbonate buffer medium. The study was carried out under pseudo-first order conditions of an excess of thioglycol concentration at 28 ± 1℃, I = 0.44 mol dm-3 (KNO3) and λmax = 490 nm. The reaction is first order in [Fe(III)HEDTAOH2] and half order in [RSH] and a stoichiometric mole ratio of [Fe(III)HEDTAOH2]: RSH is 2:1. Reaction rates increased with increase in ionic strength (I) and dielectric constant (D) of the reaction medium of the reaction. The reaction displayed positive primary salt effect, which suggests the composition of activated complex are likely charged reactants ions. Test for possibility of an intermediate complex formation shows negative as Michaelis-Menten plot was linear with very negligible intercept. Based on the findings, outer-sphere mechanism is proposed for the reaction. The experimental rate law obtained is; - = k2 [Fe(III)HEDTAOH2][RSH]½

Selected dithiols were converted into α,ω-dipropargyl sulfides and combined with thioglycol-derived diazides under Huisgen–Sharpless–Meldal reaction conditions to give a new class of 1,2,3-triazole-linked sulfur-rich crown ethers.

On the basis of physico-chemical analyses, a cis-octahedral environment around Ti (IV) is proposed.

Creating a multi-step method for synthesizing heterocyclic compounds is the goal of this research. The first step involves reacting 4,5-dichloro-2- phenylenediamine with 3- amino- 4-“hydroxybenzoic acid to produce an imidazole derivative (1), which then reacts with salicyldehyde to produce the azo compound (2). Two, a Schiff base derivative is produced via a reaction with p-chloro aniline (23). Final steps involve reactions (3) with chloroacetyl chloride, alanine, sodium azide, thioglycol acid, anthranilic acid, and maleic anhydride, yielding beta-lactam derivatives (4), imidazolidine (5), tetrazol(6), thiazolidine (7), hydroquinazoline”(8), and oxazepine (9), respectively. The spectroscopic techniques of fourier transform infrared (FTIR) and proton nuclear magnetic resonance (1HNMR) have been used to identify all of the compounds that have been prepared; next, the compounds’ biological effects on two types of cancer have been investigated.

Halide ions are a type of anions that are commonly found in the environment, have important physiological functions, and are used in industry. However, their high content is harmful to the environment and human health, largely depending on the status of the endogenous microbiota. The aim of this work is to research the features of the influence of halides on Bifidobacterium bifidum growth in vitro. The dependence of biomass accumulation in the stationary phase on the content of sodium and potassium halides in thioglycol medium at different concentrations (0,001, 0,01, 0,1 and 1 M) was studied. Empirical values were fitted with dose-response model curves. Two-factor PERMANOVA was shown that the shape of the curves significantly (p<0,01) depends on the type of anion, type of cation, salt concentration and the interaction of factors “type of anion / salt concentration”. Moreover, the biomass of bacteria is often lower in the presence of potassium salts than similar sodium ones. As a result, model curves for different salts has various form (logistics, exponential and hyperbolic). Fluorides and chlorides inhibited biomass growth at all concentrations compared to control. Bromides and iodides at 0,001–0,01 M stimulated the accumulation of biomass to 20–50%. This effect can be used in the food industry, agriculture and healthcare. At a concentration of 0,001 M, a significant regression relation was shown between the atomic mass of the halogen and the fitted curve value of biomass. This dependence probably demonstrates effect atomic radius or/and electronegativity halogens for their penetration into the cell and interaction with cellular targets. The result dependencies do not correspond with state sanitary standards for halide ions. These regulations for fluorides (~10–4 M) and chlorides (10–2 M) roughly match to the survival limit of bifidobacteria. However, more strong limits for bromides (~10–6 M) and iodides (~10–6 M) do not correlate with the biological response of bifidobacteria. In addition, the sanitarian standards halogenids do not take into account the modifying effect of cations which can be important.

High concentration of sepsis-associated aromatic microbial metabolites (AMM) stands as a prognostically unfavorable factor, indicating the progression of multiple organ dysfunction and an increased risk of death in patients with sepsis and septic shock. This study is based on a hypothesis that excess of sepsis-associated AMM in patients with sepsis is caused by metabolic alterations (dysfunction) in the intestinal microbiota.The aim of this study was to compare the potential of normobiota and pathobiota to bio-transform sepsis-associated metabolites of aromatic amino acids tyrosine and phenylalanine, such as phenyllactic acid(PhLA) and 4-hydroxyphenyllactic acid (4-HPhLA).Materials and methods. Samples of intestinal contents of patients with septic shock (N=10, pathobiota) and healthy volunteers (N=9, normobiota) were placed in test tubes with the omnipurpose thioglycol medium. The clinical model of excessive inflow of sepsis-associated AMM into the intestine (for example, from blood or sites of inflammation) was reproduced in the in vitro experiment by adding PhLA or 4-HPhLA in clinically significant concentrations (25 mkM) into each test tube with pathobiota and normobiota. After incubation in a thermostat (37°, 24 hours), AMМ concentrations were measured in the samples with pathobiota and normobiota using GC-MS analysis.Results. Concentration of AMM decreased within 24 hours in the tubes with normobiota after PhLA or 4-HPhLA were added. In the tubes with pathobiota, no decrease in AMM concentrations was documented after loading with PhLA or 4-HPhLA. Concentrations of PhLA (P=0.002) and 4-HPhLA (P0.001) were statistically significantly higher in pathobiota samples compared to normobiota.Conclusion. The in vitro experiment demonstrates that after excessive load with sepsis-associated metabolites (PhLA, 4-HPhLA), the microbiota of healthy people is capable to bio-transform such metabolites to the end products of microbial metabolism, while pathobiota of septic patients exhibits altered biotransformational potential. This data demonstrate that microbiota dysfunction may contribute to the pathogenesis of sepsis.

Structural and functional properties of the human tendon tissue complicate process of tendon-based grafts preserving. In the process of preservation, it is necessary to maintain mechanical and functional parameters of the tendon tissue as well as to provide sterility and biological safety of the graft. One could conclude effectiveness of techniques, combining tendon conservation at low temperatures and sterilization with the supercritical carbon dioxide.Aim of work: to find optimal conditions for allogeneic tendon grafts sterilization with supercritical carbon dioxide.Materials and methods. Allogeneic tendons were prepared from the tissue donors, following the rules of asepsis and antisepsis. After quarantine and confirmation of the absence of blood-borne infections the tendon grafts were divided into 3 groups: in the control group the tendons were not subjected to cryopreservation and sterilization procedures; in 2 experimental groups tendons were treated with a cryoprotector 10 % dimethyl sulfoxide (DMSO) solution and sterilized with supercritical carbon dioxide with slow gas discharge (group 2) or with fast gas discharge (group 3); the duration of sterilization in both groups varied from 1 to 12 hours. The toxicity of the grafts was assessed in human mesenchymal multipotent stromal cells (MMSC) culture on the 3rd and 7th days. The sterility of the grafts was confirmed on the 7th and 14th day by bacteriological culture on thioglycol medium and Saburo broth. Mechanical tests were performed on testing machine LLOYD Instruments LR5K Plus with the tensile speed 5 mm/min.Results. Histological analysis showed, that in the group with slow gas discharge collagen fibers retained their integrity and topography and contained only local minor tears; in all terms of treatment, the tendon grafts were sterile and nontoxic. On the contrary, in the group with fast gas discharge there was marked damage of the collagen fibers and growth of bacterial and fungal flora was observed in the culture. The grafts of the control group and the group, where sterilization with supercritical carbon dioxide was performed with slow gas discharge for 1-12 hours, were used to evaluate mechanical characteristics. In the preserved tendons the stiffness and ultimate strain did not significantly differ from the similar values in the control (p > 0.05), on the contrary, the level of ultimate strain in all experimental specimens was significantly reduced by 1.5-2.1 times comparing to the control. Young's modulus and load at rupture in the control grafts and grafts, sterilized for 1 hour, had similar values, whereas in grafts, sterilized from 3 to 12 hours, these parameters were 1.4-2.1 times lower (p < 0.05).Conclusions. The suggested technique of tendon preservation, using cryoprotector DMSO and sterilization with the supercritical carbon dioxide, allows to obtain sterile and non-toxic grafts. The structure of cells and fibers of the preserved tendons did not significantly disturbed. For supercritical carbon dioxide sterilization of tendon grafts the most optimal processing time is 1 hour.

BackgroundIn Oriental countries, extracts of Schisandra chinensis have been used as an anti‐fatigue and tonic agent. Because inflammatory and oxidative stress are related to chronic fatigue syndrome, it was hypothesized that Schisandra chinensis has anti‐inflammatory constituents. The effects of new compounds isolated from dry fruits of Schisandra chinensis were studied, to elucidate the anti‐fatigue effect of Schisandra chinesis.METHODSFrom an extract of Schisandra chinensis fruits, SC‐3162 was isolated. Inductions of inducible nitric oxide synthase and cyclooxygenase 2 by lipopolysaccharide were quantified by Western blotting in peritoneal macrophages isolated from thioglycol‐treated mice. A peritonitis animal model was used to test the in vivo efficacy of SC‐3162.RESULTSSC‐3162 inhibited the inductions of inducible nitric oxide synthase and cyclooxygenase 2 in peritoneal macrophages isolated from thioglycol‐treated mice at the mRNA and protein levels. lipopolysaccharide ‐induced productions of nitric oxide and prostaglandin E2 were inhibited by SC‐3162. Furthermore, SC‐3162 inhibited the lipopolysaccharide‐induced activations of p38 MAPK, but not that of ERK and JNK in mouse peritoneal macrophages. In addition, SC‐3162 strongly inhibited the accumulation of neutrophils, eosinophils, and basophils in the peritoneal cavity in the lipopolysaccharide‐induced peritonitis model,.CONCLUSIONSC‐3162 suppresses inflammatory responses in peritoneal macrophages and could be a potential therapeutic agent for treatment of inflammatory diseases.

Currently, blood transfusion should be considered as a tissue transplantation operation with all the consequences - possible rejection of cellular and plasma components of blood, development of allosensitization to antigens of blood cells and plasma proteins, as well as, in the case of an immunodeficient patient, the possible development of a life-threatening reaction "Transplant against host". Erythrocyte mass is a transfusion medium containing at least 70% erythrocytes, which is optimal in the treatment of anemic syndrome. At equal volumes compared to whole blood, the erythrocyte mass contains a larger number of erythrocytes, but significantly less citrate, products of cell decay, cellular and protein antigens, and antibodies. Transfusions of erythrocyte mass occupy a prominent place in transfusion therapy, which aims to fill the deficiency of erythrocytes in anemic conditions. We studied 5 samples of preserved erythrocyte mass from clinically healthy horses. Blood was collected by a closed method followed by separation of the erythrocyte mass by centrifugation. The mass was stored in a refrigerator at a temperature of 2-6°C for 30 days. Sowing of each sample was carried out in the thickness of the nutrient medium. Cultures in a thioglycol medium and control tubes were incubated in a thermostat at a temperature of 20-25°C and at a temperature of 30-35°C, with a Sabouraud medium at a temperature of 20-25°C.
