With the prevalence of online review websites, large-scale data promote the necessity of focused analysis. This task aims to capture the information that is highly relevant to a specific aspect. However, the broad scope of the aspects of the various products makes this task overarching but challenging. A commonly used solution is to modify the topic models with additional information to capture the features for a specific aspect (referred to as a targeted aspect). However, the existing topic models, either perform the full analysis to capture features as many as possible or estimate the similarity to capture features as coherent as possible, overlook the fine-grained semantic relations between the features, resulting in the captured features coarse and confusing. In this paper, we propose a novel Hierarchical Features-based Topic Model (HFTM) to extract targeted aspects from online reviews, then to capture the aspect-specific features. Specifically, our model can not only capture the direct features posing target-to-feature semantics but also capture the latent features posing feature-to-feature semantics. The experiments conducted on real-world datasets demonstrate that HFTMl outperforms the state-of-the-art baselines in terms of both aspect extraction and document classification.

This paper reports beetle pests common to flourmills targeted during a series of trapping studies over a two-year period in flourmill 1 and a one year period in flourmill 2. Objectives were (1) use pheromone-baited traps to detect T. castaneum (Herbst) and other pest species present for their distribution over space and time, (2) monitor T. castaneum activity before and after fumigations to assess efficacy of the treatment, and (3) correlate counts of T. castaneum via trap capture against direct T. castaneum counts from samples of the milled flour to assess the value of trap data to estimate relative size of the pest population. Traps were deployed in two different flourmills over two consecutive years. T. castaneum was the most commonly trapped beetle during both years in mill 1. In mill 2, Typhaea stercorea (L.) and Cryptolestes ferrugineus (Stephens) were both captured in higher numbers than T. castaneum. In mill 1, trap capture was higher overall during Year 2 for most of the species compared with capture during Year 1, likely due to a dust cover modification made for the pitfall trap used in Year 2. Trap capture was also evaluated by location within the mills and a significant difference was found in the capture of T. stercorea during both years in mill 1. T. castaneum captures were significantly reduced following most fumigations, which used methyl bromide in milling areas and phosphine in bulk-stored finished flour. However, in most cases trap catches showed that beetle populations were not eliminated. Trap captures after fumigation suggest either that the fumigations were not entirely effective, or that full grown adult beetles were entering the mill soon after fumigation. When captures of T. castaneum from traps in two spaces of mill 1 during Year 2 were compared with counts of beetles from samples of siftings collected in the finished flour, the correlation coefficients were nearly significant for both sets of traps.

eB4CAST, evidence-Based forecast C-capture, A-assemble, S-sustain, T-timelessness (eB4CAST), framework was developed from existing dissemination and implementation (D & I) constructs as a dissemination tool to promote community-based program usability and future application in targeted populations. eB4CAST captures and transforms research findings into a dissemination report that shows program need and impact to endorse program continuation and expansion. This is achieved through direct and indirect data collection of community factors and program impact that can showcase the need for program sustainability and potential for future dissemination sites. Testimonials, individual feedback, and program process and outcomes contribute to the direct data while data collected from census, county, and state databases and reports allow for indirect information to be captured and analyzed. Capturing data in the two levels allow eB4CAST to forecast program need and highlight program impact through a footprint. eB4CAST framework for dissemination tool creation is organized into four sections: Capture, Assemble, Sustainability, and Timelessness. Capture encompasses the collection of indirect and direct data related to intervention goals. Assemble is the compilation of the data into a visually appealing and easily understood media. Sustainability encourages the use of dissemination tools to provide forecast of program need and footprint of program impact back to community participants, program leaders, and key stakeholders to endorse program sustainability. Lastly, timelessness encourages cyclic movement through these constructs to continue program monitoring and data sharing to ensure timeless program evaluation and conformation to change in needs. The eB4CAST framework provides a systematic method to capture justification of program need and impact of community-based research that can be modified to fit diverse public health interventions providing a necessary D & I tool.

Specific ecological interactions between insects and microbes have potential in the development of targeted pest monitoring or control techniques for the spotted wing drosophilid, Drosophila suzukii (Matsumura), an exotic invasive pest of soft fruit. To evaluate D. suzukii attraction to yeast species from preferred types of fruit, three yeasts were isolated from blackberry fruit and two yeasts from raspberry fruit and used to bait simple plastic bottle traps. Saccharomyces cerevisiae and Hanseniaspora uvarum were identified from blackberries, whereas a different H. uvarum strain was identified from raspberry. Yeast identification was based on sequence analysis of the D1/D2 domain of the large subunit 26S rRNA gene. Commercial baker’s yeast (S. cerevisiae) was similar or more effective for the capture of D. suzukii males and females than yeasts isolated from blackberry or raspberry when grown in sucrose. However, when grown in corn syrup, a strain of S. cerevisiae from blackberry captured the highest number of females and a strain of H. uvarum isolated from raspberry captured high numbers of males and females. Species of Candida, Hanseniaspora, and Pichia from a laboratory yeast collection did not outperform baker’s yeast in pairwise tests when grown in sucrose solution or yeast-peptone-dextrose medium. The raspberry strain of H. uvarum grown in corn syrup outperformed S. cerevisiae grown in sucrose, in terms of captures in baited traps under laboratory conditions. We conclude that yeast species, strain, and growth medium can have a marked influence on D. suzukii attraction to baited traps, a finding that could assist in the development of yeast-related monitoring or control techniques targeted at this pest.

Abstract Background: The introduction and use of next-generation sequencing (NGS) techniques have taken genomic research into a new era; however, implementing such powerful techniques in diagnostics laboratories for applications such as resequencing of targeted disease genes requires attention to technical issues, including sequencing template enrichment, management of massive data, and high interference by homologous sequences. Methods: In this study, we investigated a process for enriching DNA samples that uses a customized high-density oligonucleotide microarray to enrich a targeted 280-kb region of the NF1 (neurofibromin 1) gene. The captured DNA was sequenced with the Roche/454 GS FLX system. Two NF1 samples (CN1 and CN2) with known genotypes were tested with this protocol. Results: Targeted microarray capture may also capture sequences from nontargeted regions in the genome. The capture specificity estimated for the targeted NF1 region was approximately 60%. The de novo Alu insertion was partially detected in sample CN1 by additional de novo assembly with 50% base-match stringency; the single-base deletion in sample CN2 was successfully detected by reference mapping. Interferences by pseudogene sequences were removed by means of dual-mode reference-mapping analysis, which reduced the risk of generating false-positive data. The risk of generating false-negative data was minimized with higher sequence coverage (>30×). Conclusions: We used a clinically relevant complex genomic target to evaluate a microarray-based sample-enrichment process and an NGS instrument for clinical resequencing purposes. The results allowed us to develop a systematic data-analysis strategy and algorithm to fit potential clinical applications.

The success of long short-term memory (LSTM) neural networks in language processing is typically attributed to their ability to capture long-distance statistical regularities. Linguistic regularities are often sensitive to syntactic structure; can such dependencies be captured by LSTMs, which do not have explicit structural representations? We begin addressing this question using number agreement in English subject-verb dependencies. We probe the architecture’s grammatical competence both using training objectives with an explicit grammatical target (number prediction, grammaticality judgments) and using language models. In the strongly supervised settings, the LSTM achieved very high overall accuracy (less than 1% errors), but errors increased when sequential and structural information conflicted. The frequency of such errors rose sharply in the language-modeling setting. We conclude that LSTMs can capture a non-trivial amount of grammatical structure given targeted supervision, but stronger architectures may be required to further reduce errors; furthermore, the language modeling signal is insufficient for capturing syntax-sensitive dependencies, and should be supplemented with more direct supervision if such dependencies need to be captured.

Individuals play various roles in maintaining social integrity of mammalian populations. However, many models developed for managing wildlife resources assume that all individuals are equal. Killer whales are social animals that rely on relationships within and among family groups for survival. In the northeastern Pacific, fish-eating, ‘resident’ killer whale populations are composed of matrilines from which offspring do not disperse. We analysed the influence of various individuals' age, sex and matrilineal affiliation on their position in a social network. Here, we show that some matrilines appeared to play more central roles than others in the network. Furthermore, juvenile whales, especially females, appeared to play a central role in maintaining network cohesion. These two key findings were supported subsequently by simulating removal of different individuals. The network was robust to random removals; however, simulations that mimicked historic live-captures from the northeastern Pacific were likely to break the network graph into isolated groups. This finding raises concern regarding targeted takes, such as live-capture or drive fisheries, of matrilineal cetaceans.

This paper considers methods for taxing the income of the affluent. Much of this income is unrealized capital gains that escapes tax. Conventional individual income tax changes cannot capture this income and corporate taxes cannot target the wealthy. Other options are estate and gift taxes, taxation of gains on an accrual basis, and a wealth tax. Accrual taxation of capital gains most closely captures untaxed income, can be targeted to the wealthy, and appears to be feasible. If wealth and accrual taxation are deemed too difficult, a combination of conventional changes and taxing gains at death are options.

In countries where the government encourages “donations” and “gifts” from private companies, FDI firms are more likely than their domestic counterparts to engage in corrupt forms of political influence, known as State Captures. Different types of foreign investors engage in particular types of corruption, depending on what competitive advantage they will get out of it. FDI firms with local partners are more likely to engage in State Capture. Larger multinational firms with headquarters overseas rely much less on State Capture, yet are more likely to resort to political corruption and bribery, kickbacks, in their dealing with foreign governments. Though quite often, foreign direct investors might claim that they are specifically targeted for bribes by local governments, it has been found that there is no evidence that FDI firms pay higher overall bribes than domestic firms, even though they are more likely to engage in specific forms of corruption.

Immunoglobulin (Ig)/T-cell receptor (TCR) gene rearrangement-based molecular minimum residual disease (MRD) monitoring has become one of the most powerful prognostic indicators for patients with acute lymphoblastic leukemia (ALL). Infant ALL has a poor prognosis and requires therapeutic development based on accurate MRD assessment. However, there are some difficulties associated with the highly sensitive detection of MRD based on Ig/TCR gene rearrangements in infant ALL cases, compared to other B-ALL cases, because of the immature Ig/TCR gene rearrangement patterns and their oligoclonality. Since KMT2A(MLL) gene rearrangements are identified in infant ALL at a high frequency, the use of KMT2A rearranged sequences is an alternative method for MRD detection in KMT2A rearranged ALL cases. However, identifying the genomic breakpoints in KMT2A rearrangements by conventional methods is laborious and difficult in routine practice. To detect molecular MRD markers in infant ALL cases, we designed capture probes covering the coding and recognition signal sequences of the V, D, and J genes of the Ig/TCR loci and common genomic breakpoints in KMT2A, AFF1 (AF4), MLLT3 (AF9), MLLT1 (ENL), MLLT10 (AF10), and ELL. 88 infant ALL cases from the JPLSG MLL-10 study, including 73 cases with KMT2A rearrangements, were analyzed by high-throughput target-capture sequencing on the Illumina platform. The sequenced data were analyzed with the Ig/TCR recombination analysis tool Vidjil and the Genomon pipeline for detection of KMT2A rearrangements. Although 40% of cases in this study were marker-negative in the conventional marker screening analysis, clonal Ig/TCR targets for MRD measurements were identified in more than 80% of the cases by target-capture sequencing. Ig / TCR rearrangements detected in infant ALL were unique and related to age at diagnosis and type of KMT2A rearragement. KMT2A-rearranged cases showed a significantly higher frequency of incomplete rearrangements of IGH and TCRD compared to KMT2A-negative infant ALL cases. The target-capture method also showed an increased sensitivity of Ig/TCR-based MRD measurements even in cases where MRD measurement was possible. Genomic breakpoints of KMT2A rearrangements were detected in 80% of KMT2A-rearranged cases, in which KMT2A was most frequently rearranged with AFF1, followed by MLLT1, MLLT3, and MLLT10. Other rare fusion partners of the KMT2A gene were also captured by probes for KMT2A sequences. The determined patient-specific genomic breakpoint sequences could be used for monitoring the MRD of KMT2A-rearranged cases. Thus, target-capture sequencing is an easy and useful method for screening MRD markers in infant leukemia cases. Disclosures No relevant conflicts of interest to declare.

We use data from Hungary to establish two results about the relationship between the government and the media. (i) We document large advertising favors from the government to connected media, and large corruption coverage favors from connected media to the government. Our empirical strategy exploits sharp reallocations around changes in media ownership and other events to rule out market‐based explanations. (ii) Under the assumptions of a structural model, we distinguish between owner ideology and favor exchange as the mechanism driving favors. We estimate our model exploiting within‐owner changes in coverage for identification and find that both mechanisms are important. These results imply that targeted government advertising can meaningfully influence content. Counterfactuals show that targeted advertising can also influence owner ideology, by making media ownership more profitable to pro‐government connected investors. Our results are consistent with qualitative evidence from many democracies and suggest that government advertising affects media content worldwide.

Abstract The KMT2A gene is rearranged in pediatric and adult acute lymphoblastic leukemia (ALL) and acute myeloid leukemia (AML). By fusing with a number of different partner genes, the KMT2A gene generates chimeric proteins with oncogenic abilities. Due to the promiscuity of the gene, routine genetic analysis of KMT2A-rearranged leukemia is time-consuming as it often involves chromosome and FISH analyses, RT-PCR or southern blot. We have thus developed a captured-based next generation sequencing approach to detect KMT2A fusion genes, especially useful when routine methods indicate a rearrangement but cannot identify the specific fusion partner. First, we analyzed bone marrow samples from two AML cases with deletions of the distal part of the KMT2A gene and, as a control, a pre B-ALL with a known KMT2A-AFF1 fusion. The capture was carried out using custom hybridization probes (SureSelect, Agilent) encompassing the entire KMT2A gene. Sequencing was performed on the Ion Torrent PGM, a sequencing instrument well suited for routine diagnostics given its rapid turn-around time. Putative fusions were identified from sequencing reads connecting an exon within KMT2A to an exon from any other gene and the results were confirmed by RT-PCR. As comparison we performed the standard RNA-sequencing protocol using Ion Proton. In the two patients with unknown KMT2A-rearrangements, the capture approach identified 3034 and 2118 reads over a KMT2A-MLLT4 gene fusion, while only 4 and 2 reads, were detected with the standard RNA-sequencing protocol (Table 1). Similarly, RNA-sequencing and the capture approach detected in the control a KMT2A-AFF1 fusion, with a sequencing depth of 4 and 144 reads, respectively. Interestingly, with the capture approach we obtained 5032 reads encompassing the reciprocal fusion, AFF1-KMT2A, underlining the importance of the reciprocal transcript in this entity. Hence, the KMT2A capture approach resulted in a 5000-35000-fold enrichment of the KMT2A fusions compared to standard RNA-sequencing. Although the standard RNA-sequencing detected reads covering the fusion genes, it yielded far too low sequencing depth to be of diagnostic use. We then performed the KMT2A capture approach on three additional cases with distal KMT2A deletions. In one case, we detected a KMT2A-ARHGEF12 fusion, resulting from a 20Mb interstitial deletion (Table 1). This aberration is missed by standard cytogenetics and consequently likely under-diagnosed. However, in two other cases no KMT2A-fusions were identified. In these cases, the 11q-deletion was present only in a proportion of cells and in one, the deletion was absent at relapse, indicating that it was not likely the primary event in this leukemia. These results demonstrate the importance of establishing if a suspected KMT2A-rearrangement truly generates a KMT2A-fusion. In summary, the hybridization-based capture sequencing approach efficiently detects KMT2A-fusion transcripts and reliably identifies canonical and cryptic KMT2A-rearrangements, generating significantly higher number of fusion breakpoint reads, as compared to RNA-sequencing. Abstract So far 24 species of terrestrial small mammals are known from Făgăraş Piedmont, one of them (Chionomys nivalis) requiring further confirmation, as its presence at this elevation is put under question by the authors. In 2010 and 2011 three field campaigns were carried out in Făgăraş Piedmont, which targeted the abundance and structure of terrestrial small mammal communities in different habitats from three localities, using the capture-mark-release method. 61 transects of traps were installed in 9 types of habitats. 491 small mammals were captured belonging to 14 species, three shrews and 11 rodents. The highest densities were recorded in cultures and unused land, but specific diversity is not correlated with abundance.

Multiple myeloma is a plasma cell neoplasm characterized by clonal immunoglobulin V(D)J signatures and oncogenic immunoglobulin gene translocations. Additional subclonal genomic changes are acquired with myeloma progression and therapeutic selection. PCR-based methods to detect V(D)J rearrangements can have biases introduced by highly multiplexed reactions and primers undermined by somatic hypermutation, and are not readily extended to include mutation detection. Here, we report a hybrid-capture approach (CapIG-seq) targeting the 3′ and 5′ ends of the V and J segments of all immunoglobulin loci that enable the efficient detection of V(D)J rearrangements. We also included baits for oncogenic translocations and mutation detection. We demonstrate complete concordance with matched whole-genome sequencing and/or PCR clonotyping of 24 cell lines and report the clonal sequences for 41 uncharacterized cell lines. We also demonstrate the application to patient specimens, including 29 bone marrow and 39 cell-free DNA samples. CapIG-seq shows concordance between bone marrow and cfDNA blood samples (both contemporaneous and follow-up) with regard to the somatic variant, V(D)J, and translocation detection. CapIG-seq is a novel, efficient approach to examining genomic alterations in myeloma.

While the relatives of those holding formal leadership positions are more likely to receive benefits in some programs, we argue that the welfare consequences of elite capture appear small: eliminating elite capture entirely would improve the welfare gains from these programs by less than one percent.

N terminomics is a powerful strategy for profiling proteolytic neo-N termini, but its application to cell surface proteolysis has been limited by the low relative abundance of plasma membrane proteins. Here we apply plasma membrane-targeted subtiligase variants (subtiligase-TM) to efficiently and specifically capture cell surface N termini in live cells. Using this approach, we sequenced 807 cell surface N termini and quantified changes in their abundance in response to stimuli that induce proteolytic remodeling of the cell surface proteome. To facilitate exploration of our datasets, we developed a web-accessible Atlas of Subtiligase-Captured Extracellular N Termini (ASCENT; http://wellslab.org/ascent). This technology will facilitate greater understanding of extracellular protease biology and reveal neo-N termini biomarkers and targets in disease.

Many bird species avoid traditional traps such as mist nets, thus alternative trapping methods are often needed. The Rufous Hornero (Furnariidae: Furnarius rufus) is one such species that was captured so far using conventional mist netting in rural areas or using nest-targeted traps, which risk nest damage and abandonment. Here we describe the novel rise-up mist-netting (RUM) protocol and its catchability for the wary and territorial Rufous Hornero in an urban area. The RUM trap allowed us quickly capturing 40 Rufous Hornero individuals even in paved parking lots and streets during the bird’s breeding period. The RUM is an active and targeted protocol that demands a single and quickly trainable field assistant. Therefore, it should also enable the capture of many understudied territorial but wary species. Moreover, it has enough mobility to fit studies in urban areas, and whenever researchers may deal with limited space and, at the same time, should reduce negative visual impacts to the general public of birds’ capturing and banding procedures.

Background: Cholestasis is a frequent and severe condition during childhood. Genetic cholestatic diseases represent up to 25% of pediatric cholestasis. Molecular analysis by targeted-capture next generation sequencing (NGS) has recently emerged as an efficient diagnostic tool. The objective of this study is to evaluate the use of NGS in children with cholestasis. Methods: Children presenting cholestasis were included between 2015 and 2020. Molecular sequencing was performed by targeted capture of a panel of 34 genes involved in cholestasis and jaundice. Patients were classified into three categories: certain diagnosis; suggested diagnosis (when genotype was consistent with phenotype for conditions without any available OMIM or ORPHANET-number); uncertain diagnosis (when clinical and para-clinical findings were not consistent enough with molecular findings). Results: A certain diagnosis was established in 169 patients among the 602 included (28.1%). Molecular studies led to a suggested diagnosis in 40 patients (6.6%) and to an uncertain diagnosis in 21 patients (3.5%). In 372 children (61.7%), no molecular defect was identified. Conclusions: NGS is a useful diagnostic tool in pediatric cholestasis, providing a certain diagnosis in 28.1% of the patients included in this study. In the remaining patients, especially those with variants of uncertain significance, the imputability of the variants requires further investigations.

Magnetic Fe3O4 nanoparticles (NPs) and their surface modification with therapeutic substances are of great interest, especially drug delivery for cancer therapy, including boron-neutron capture therapy (BNCT). In this paper, we present the results of boron-rich compound (carborane borate) attachment to previously aminated by (3-aminopropyl)-trimethoxysilane (APTMS) iron oxide NPs. Fourier transform infrared spectroscopy with Attenuated total reflectance accessory (ATR-FTIR) and energy-dispersive X-ray analysis confirmed the change of the element content of NPs after modification and formation of new bonds between Fe3O4 NPs and the attached molecules. Transmission (TEM) and scanning electron microscopy (SEM) showed Fe3O4 NPs’ average size of 18.9 nm. Phase parameters were studied by powder X-ray diffraction (XRD), and the magnetic behavior of Fe3O4 NPs was elucidated by Mössbauer spectroscopy. The colloidal and chemical stability of NPs was studied using simulated body fluid (phosphate buffer—PBS). Modified NPs have shown excellent stability in PBS (pH = 7.4), characterized by XRD, Mössbauer spectroscopy, and dynamic light scattering (DLS). Biocompatibility was evaluated in-vitro using cultured mouse embryonic fibroblasts (MEFs). The results show us an increasing of IC50 from 0.110 mg/mL for Fe3O4 NPs to 0.405 mg/mL for Fe3O4-Carborane NPs. The obtained data confirm the biocompatibility and stability of synthesized NPs and the potential to use them in BNCT.

Background and objectivesImage-guided intervention of the suprascapular nerve is a reported treatment to manage chronic shoulder joint pain. The suprascapular nerve is conventionally targeted at the suprascapular notch; however, targeting of its branches, the medial and lateral trunks, which are given off just posterior to the notch has not been considered. Since the lateral trunk supplies the posterior supraspinatus and articular branches to the glenohumeral joint capsule, while the medial trunk provides motor innervation to the anterior region, it may be possible to preserve some supraspinatus activation if the medial trunk is spared. The main objective was to investigate whether midpoint between suprascapular and spinoglenoid notches is the optimal target to capture articular branches of lateral trunk while sparing medial trunk.MethodsIn 10 specimens, using ultrasound guidance, one 17 G needle was placed at the suprascapular notch and a second at midpoint between suprascapular and spinoglenoid notches. The trunks and needles were exposed in the supraspinous fossa, digitized and modeled in 3D. Lesion volumes were added to the models to asses medial and lateral trunk capture rates. Mean distance of needle tips to origin of medial trunk was compared.ResultsConventional notch technique captured both lateral and medial trunks, whereas a midpoint technique captured only lateral trunk. Mean distance of needles from the origin of medial trunk was 5.10±1.41 mm (notch technique) and 14.99±5.53 mm (midpoint technique).ConclusionsThe findings suggest that the midpoint technique could spare medial trunk of suprascapular nerve, while capturing lateral trunk and articular branches. Further clinical investigation is required.

Abstract Context Lethal control of predators is often undertaken to protect species of conservation concern. Traps are frequently baited to increase capture efficacy, but baited traps can potentially increase predation risk by attracting predators to protected areas. This is especially important if targeted predators can escape capture due to low trap success. Snake traps using live mouse lures may be beneficial if traps effectively remove snakes in the presence of birds and do not attract additional snakes to the area. Aims The present study evaluated whether mouse-lure traps in areas occupied by birds (simulated by deploying bird-lure traps) could influence predation risk from an invasive snake on Guam. Methods Snake traps were used, with Japanese quail (Coturnix japonica) as a proxy for predation risk, to assess if an adjacent trap with a mouse (Mus musculus) would attract brown treesnakes (Boiga irregularis) to a focal area and increase contact between an invasive snake and avian prey. Catch per unit effort (CPUE) at stations containing either a bird-lure trap, mouse-lure trap or pair of traps (i.e. one bird-lure and one mouse-lure trap) was evaluated. Key results Bird-lure traps paired with mouse-lure traps did not differ in CPUE from isolated bird-lure traps. At paired stations, CPUE of snakes in mouse-lure traps was 2.3× higher than bird-lure traps, suggesting mouse lures were capable of drawing snakes away from avian prey. Bird-lure traps at paired stations experienced a decay in captures over time, whereas CPUE for isolated bird-lure traps increased after 9 weeks and exceeded mouse-lure traps after 7 weeks. Conclusions Mouse lures did not increase the risk of snakes being captured in bird-lure traps. Instead, mouse-lure traps may have locally suppressed snakes, whereas stations without mouse-lure traps still had snakes in the focal area, putting avian prey at greater risk. However, snakes caught with bird lures tended to be larger and in better body condition, suggesting preference for avian prey over mammalian prey in larger snakes. Implications Strategic placement of olfactory traps within areas of conservation concern may be beneficial for protecting birds of conservation concern from an invasive snake predator.

The early diagnosis and treatment of cholangiocarcinoma (CCA) remain a challenge worldwide. Genetic testing promises to solve these problems. Due to the different mutation landscapes across populations and the paucity of sequencing data of Chinese patients with CCA, the existing mutation landscape is insufficient to reflect the mutation characteristics of Chinese patients. Thus, we retrospectively analyzed 72 Chinese patients with CCA who had received genetic testing of targeted capture sequencing. A total of 2152 somatic mutations were detected in 56 (77.78%) patients, of which, the frequently mutated driver genes were TP53 (27.78%), KMT2D (23.81%), KMT2C (20.63%), BCOR (18.06%), APC (15.28%), BAP1 (13.89%), ARID1A (12.50%), NF1 (12.50%), PIK3CA (12.50%), KRAS (11.11%), and LRP1B (11.11%). Most mutations were enriched in NRF2, TP53, and TGF-Beta oncogenic signaling pathways and cadherin repeat domains which were associated with intercellular adhesion. Based on cancer-related public databases and multiple protein function prediction algorithms, we identified 118 novel pathogenic or likely pathogenic somatic mutations and 77 actionable alterations. Molecular analysis of tumors from a precision oncology perspective can provide potential targets for early diagnosis and treatment of CCA and assist physicians in clinical decision making.
