Повний текст джерелаDue to their various functionalizable sites, cyclopentenones are very useful intermediates for the synthesis of natural products of therapeutic value. In particular, 4-aminocyclopentenones enable the access to the aminocyclopentitol frameworks, which are present in a variety of bioactive molecules such as peramivir, pactamycin, or trehazolin. One of the most efficient methods to access 4-aminocyclopentenones is the aza-Piancatelli reaction. It is based on the rearrangement of 2-furylcarbinols in the presence of a nitrogen nucleophile following a mechanism involving a 4π-conrotatory electrocyclization. In our laboratory, a simple catalytic system using a calcium complex combined with an ammonium salt was developed to gain access to these compounds. This method has many advantages : it is effective (yields up to 98%), fast (15 to 30 minutes), it requires only 1 mol% of catalyst under pratical conditions (undistilled solvents without an inert atmosphere) on a large scale (multi-gram). In this context, we sought to extend the scope of this reaction by designing more complex 2-furylcarbinols in order to directly access skeletons of bioactive compounds. In particular, we focused on the total synthesis of jogyamycin. In addition, we achieved the total synthesis of bruceollin D with an overall yield of 16% over five steps. We also developed a new reaction sequence involving an aza-Piancatelli reaction followed by a hydroamination reaction promoted by a copper salt. This sequence provides a wide range of highly functionalized cyclopenta[b]pyrroles from readily-available 2-furylcarbinols substituted by an alkyne moiety. Following this method, 42 cyclopenta[b]pyrroles were obtained with yields up to 98%. An original feature of this transformation is related to the use of ortho-substituted anilines. Indeed, in this case, atropisomers with a diastereomeric excess superior to 20:1 could be obtained accompanied by the creation of an chiral N-C axis during the hydroamination step. To date, no other example of atropodiastereoselective synthesis of cyclopenta[b]pyrroles has been reported. During our investigations on the scope of the reaction, we noticed that cyclopenta[b]pyrroles underwent a rearrangement into cyclopenta[b]pyrrolines, following a dearomatization when hexafluoroisopropanol was used as solvent. This transformation led to 13 cyclopenta[b]pyrrolines with yields ranging from 44% to 73%. We also investigated the functionalization of the cyclopenta[b]pyrrole motif to illustrate the synthetic utility of our methodology, notably by combining this reaction sequence with a Friedel-Crafts reaction. One of the objectives of this Ph.D. was the development of an enantioselective version of the reaction. After the recent publication of three papers using chiral phosphoric acids as catalysts, we devised another approach for the asymmetric synthesis of 4-aminocyclopentenones, relying on the use of a chiral auxiliary (chiral sulfoxide) in collaboration with Dr. Wencel-Delord and Pr. Colobert (University of Strasbourg). Thus, we were able to provide the corresponding 4-aminocyclopentenones with excellent yields and diastereoselectivity

Повний текст джерелаIn general, protein structures can be divided into: repetitive and aperiodic structures. Most of the aperiodic structures are globular proteins. The repetitive proteins contain arrays of repeats that are adjacent to each other, called Tandem Repeats (TRs). Proteins containing TRs are abundant and have fundamental functional importance. Numerous studies demonstrated the involvement of such TR-containing proteins in human diseases. Furthermore, genetic instability of these regions can lead to emerging infection threats. Additionally, TR-containing structures have generated significant interest with respect to protein design as they can make excellent scaffolds for specific recognition of target molecules. Therefore, the discovery of these domains, understanding of their sequence–structure–function relationship promises to be a fertile direction for research.The growth of structural genomics initiatives, in combination with improvements in crystallographic and NMR techniques aimed at non-globular proteins, has resulted in an increase in structurally elucidated TR proteins. This has necessitated the development of classification schemes. Structural repeats were broadly divided into five classes mainly based on repeat length; Class I – crystalline aggregates; Class II – fibrous structures such as collagen; Class III – elongated structures where the repetitive units require each other for structural stability such as solenoid proteins; Class IV – closed repetitive structures, such as TIM-barrels and Class V – bead on a string structures such as tandems of Ig-fold domains. Despite this progress, the majority of bioinformatics approaches have focused on non-repetitive globular proteins.In recent years, efforts have been made to develop bioinformatics tools for the detection and analysis of repetitive elements in protein structures (3D TRs). Depending on the size and character of the repeats, some methods perform better than others, but currently no best approach exists to cover the whole range of repeats. This served as a motivation for the development of our method called the TAndem PrOtein detector (TAPO). TAPO exploits, periodicities of atomic coordinates and other types of structural representation, including strings generated by conformational alphabets, residue contact maps, and arrangements of vectors of secondary structure elements. Currently, seven feature based scores produced by TAPO are combined using a Support Vector Machine, producing a score to enable the differentiation between proteins with and without 3D TRs. TAPO shows an improved performance over other cutting edge methods, achieving 94% sensitivity and 97% specificity on the current benchmark. The development of TAPO provided new opportunities for large scale analysis of proteins with 3D TRs. In accordance with our analysis of PDB using TAPO, 19% of proteins contain 3D TRs. The large scale analysis of the 3D TR structures in PDB also allows us to discover several new types of TR structures that were absent in the existing classification. Some of them are described in the thesis manuscript. This suggests that TAPO can be used to regularly update the collection and classification of existing repetitive structures. In particular, a comprehensive analysis of 3D TRs related to Rossmann Fold (RF) was undertaken. Our special interest in RFs was based on the observation that many proteins with RFs represent borderline cases between repetitive and non-repetitive structures. In principle, α-helix-β-strand units of RFs should have a strong potential to stack one over the other, forming repetitive structures. To probe the question of how frequently RFs form long arrays of stacked repeats, we selected by using TAPO known RF-containing structures and classified them. Our analysis shows that typical RFs cannot be clearly defined as repetitive, rather they are part of globular structures with up to 3 αβ-repeats. We provide some explanations for this surprising observation

It has been shown that the hydroxyalkylated dihydropyran unit, obtained frome this reaction, opens the access to different molecules from the Styryllactones’ family. It has been shown that the use of the paraformaldehyde during the process, makes possible the synthesis of natural products bearing a double bond adjacent to the heteroatom of the cycle. This tandem reaction has also been employed for the synthesis of the laulimalide’s C₁₅-C₂₇ fragment and three analogs. Finally, some tests have been managed for the second C₁-C₁₄ fragment’s synthesis

Повний текст джерелаThe main purpose of this work was to develop new methodologies for the synthesis of molecules of biological interest, such as azafluorenones and precursors of variolin B analogs. First, we synthesized diarylketones, precursors for our targets, via a deprotocupration-aroylation sequence. Then, we involved them in a pallado-catalyzed coupling step: some substrates underwent C-H activation to provide azafluorenones. Next, we turned our attention to the synthesis of substituted azafluorenones. The latter were synthesized by a tandem process combining Suzuki coupling or Heck coupling with intramolecular cyclization catalyzed by palladium. Some of these molecules were biologically evaluated and showed good biological activities: cytotoxic, antimalarial, antibacterial and antifungal. Finally, we succeeded in synthesizing precursors of variolin analogues in only three steps from a commercial product

Повний текст джерелаIt is well known that eutrophicated lakes and water reservoirs can be a serious concern to wildlife, domestic livestock and human health since cyanobacterial blooms are prone to occur. Some cyanobacteria (blue-green algae) species which form bloom may biosynthesize a family of hepatotoxic peptides named microcystins under certain circumstances, related specially to nitrogen: phosphorus ratio, temperature, pH and light intensity of the water body. There are at least 70 variants of microcystins with different toxicity, DL50 ranging from 50 to 500 µg.kg-1 (in mouse, i.p.). Acute exposure provoke hyperphosphorylation of the cytoskeleton, in liver cells, leading to cellular disruption with haemorrhage and the long term exposure can cause tumor promotion by inactivation of serine/threonine phosphatases 1 and 2A (PP1 and PP2A). Therefore, it is very important to determine and quantify microcystin variants present in water reservoirs. There are several methods available to evaluate qualitative and quantitatively microcystin and its variants in water reservoir and estimate human risks. Such methods comprise biological, biochemical and physico-chemical assays. In this work, we have isolated and analyzed hepatotoxins by High Performance Liquid Chromatography (HPLC) with photodiode array (PDA) detector and their structures were elucidated by Tandem - MS/MS with eletrospray ionization (ESI-MS/MS). MC-RR, MC-LR, MC-YR and MC-hRhR were determined in samples collected at Billings reservoir and [Asp3]MC-LR was found in the strain Microcystis aeruginosa BCCUSP 235. The procedures developed herein can be a powerful tool for isolation and determination of microcistin and its derivatives founded in water reservoirs. Also, we have described for the first time a new variant of microcistin: MC-hRhR.

Повний текст джерелаSince the 70s, organometallic catalysis profoundly influenced organic chemistry. The richness of the reactivity of transition metals in homogeneous phase motivates the realization of extensive research to « heterogenize » reactions of homogeneous catalysis. Transposing the reactions to heterogeneous systems, we can take advantage of the first (selectivity, efficiency) and second (green chemistry, recycling of the catalyst). Therefore, the heterogeneous metal catalysis is an alternative and area of fertile and innovative research for the development of « one pot » reaction in accordance with green chemistry. My thesis work is part of this problem through the development of synthetic methodologies involving heterogeneous metallic multitask catalysts. We have developed a method to generating heteregeneous multi-metallic catalysts Pd-M (with M = Au, Cu). The nanoparticles were obtained by impregnation of metallic precursors onto the support followed by reducing the metal ion under a hydrogen atmosphere. The crystal structures of nanoparticles were characterized by combining different experimental techniques such as : TEM, HRTEM, XPS and XRD. The catalytic activity and recyclability of heteregeneous bimetallic catalysts has been compared to monometallic catalysts and studied in : (i) Sonogashira coupling reactions, (ii) Sonogashira coupling/cyclization, (iii) Sonogashira coupling/Click and (iv) Click/CH arylation

Повний текст джерелаElaboration of synthetic methods based on metal-catalyzed reactions has been a hot topic in organic chemistry. Despite good efficiency, catalysis proceeding homogeneously, are limited in the operation of recovering/recycling of the catalysts. An important stress was placed to design catalysis, offering both the efficiency of homogeneous catalysts and the recyclability of heterogeneous catalysts. In this context, metal nanoparticles merged as a key tool, due to their unique physical and chemical properties. Notably, Au NPs have shown remarkable catalytic activity in the oxidation of activated alcohols under O2 atmosphere. Since now, the access to more complex molecules is the next step forward for this field, we envisioned multicatalytic roads, based on the oxidation of activated alcohols via supported Au NPs. Our choice of using solid catalysts was relevant, since nanostructured catalysts for which the fraction of active sites are located on the surface, limit the risk of cross-quenching. The latter carbonyl formed, could be further converted in situ, via tandem protocol. Herein, we developed novel, atom- and step-economical bicatalytic one-pot processes, to access substituted chromenes/quinolines (53-93%) by tandem oxidation/hetero-Michael addition/aldolisation combining nanocatalysis and base catalysis, ortho-THCs (50-81%) via tandem oxidation/arylation/cyclisation combining nanocatalysis and supported catalysts and a tandem cascade oxidation/hydrolysis to access HMLA (86%, sel 93%). A large panel of products of biological activity relevance, pertaining to the fragrance chemistry or aiming in some cases, pre-industrial scalability via continuous flow applications

Повний текст джерелаThis thesis focuses on the characterization of the structural defects on a MBE-made GaP/Si pseudo-substrate. The main purpose concerns the structural quality of GaP epitaxial layer on Si substrate, as a cornerstone for the development of high-efficiency solar cell with relatively low cost. Firstly, the study focuses on the characterization of the distribution of dislocations in GaP on Si (001) vicinal substrate, with an advanced submicrometer-beam X-ray scattering method, K-Map. The local tilt and strain information are obtained through an analysis of the complex 5D dataset. This study reveals an anisotropic distribution of the dislocations along different crystallography directions, linked to the steps on the surface of Si substrate, and an inhomogeneous distribution of the dislocations, linked to the tendency to form bunches. The second part of the study is on the growth and characterization ofCIGS on GaP/Si, which is used to explore tandem junctions associating single crystalline silicon bottom cell. The CIGS is observed to be epitaxially grown on the GaP/Si pseudo substrate, combining the results collected from XRD, EDX and HRTEM. A first try of a CIGS/GaP/Si solar cell is realized on a non-optimized GaP/Si pseudo-substrate. The obtained EQE is similar to the one obtained with the CIGS solar cell grown on a traditional glass substrate with the optimised deposition parameters

Повний текст джерелаThis work focuses on the behavior in space environment of a new photovoltaic solar cell technology: the III-V//Si tandems (2- and 3-junction), obtained by direct bonding. These cells have been exposed to electron and proton irradiations and tested in two types of environment: a) normal irradiance, 1 sun, and 300K room temperature, NIRT condition (Earth orbits) and b) low irradiance, 0.03 sun, and 120K low temperature, LILT condition (deep space). In a preliminary stage, a comparative study was conducted on 2 solar simulators, respectively equipped with a flash lamp and LED lamps, in order to ensure the reliability and reproducibility of the measurements of these multi-junctions. For the flash simulator, a tandems characterization method for I-V under 1 sun that dispense the use of isotype reference cells has been adopted, based on EQE and flash spectrum measurements. For the LED simulator, mounted in-situ on the irradiation beam, a spectrum optimization was performed in order to approach the low irradiance reference, i.e. ~3% AM0. This comparative study also allowed to establish the validity of the extrapolation by calculating I-V measurements under 1 sun towards low irradiances.Then, the compatibility of this tandem III-V//Si technology with thermal cycling on the one hand and irradiances on the other hand has been demonstrated. The bonding interface maintains its mechanical and electrical integrity face to these constraints. The impact of the irradiations on the cell performances has revealed certain similarities at 300 K and 120 K: - a marked decrease in the short-circuit current (linked to the decrease in the diffusion length) - a smaller decrease in the open-circuit voltage (generation type defects). Due to the series connection of the sub-cells, the degradation of the limiting Si (low intrinsic resistance to irradiation) dominates the behavior of the multi-junction. It has been shown that the addition of an increasing number of cells on the Si results in an increased sensitivity to irradiation; indeed, the tandem configuration restricts the absorption band of the Si to the near infrared, the spectral part most affected by the decrease in diffusion length. The use of a model based on the IQE allowed the qualification of this diffusion length degradation of the Si in tandem, as well as the damage coefficient. Unlike electrons, 1 MeV proton irradiations are at the origin of a non-homogeneous degradation in Si; by EQE measurements coupled with simulation, we have correlated this non-homogeneous degradation in Si with the position of the corresponding Bragg peak.For the low-temperature study, a linear increase in efficiency was observed up to ~150K; and below this, anomalies of I V characteristics were detected; of "S-like shape" and "flat spot" type, these defects affect the FF and thus the efficiency. Reported in the literature, these effects are characteristic of LILT conditions, and are often related to changes in the metal/semiconductor interfaces. Although significant, the LILT end-of-life electrical performance degradation of III-V//Si has been shown to be more predictable than that of III-V/Ge LILT (statistical dispersion). We have also shown that a 300 K annealing after irradiation at 120 K leads to a marked healing of the short-circuit current; this underlines the importance of in-situ characterizations to quantify cell aging under operating conditions. The Displacement Damage Dose (DDD) approach was applied for 1 MeV electrons and protons in order to compare the rate of induced degradation. This approach allows to predict the degradation of these cells whatever the fluence, particles and energy, for a space mission at 300 K

Повний текст джерелаDisease-specific compounds (biomarkers) are analyzed in clinical laboratories to assist with diagnosing diseases. This thesis describes development and validation of liquid chromatography tandem mass spectrometry (LC-MS/MS) based tests for diagnosing a diverse group of endocrine and metabolic diseases. The analytical methods used on-line and off-line sample extraction and analytical derivatization as means of enhancing the analytical sensitivity, specificity and clinical utility. All developed methods were extensively validated and reference intervals for the biomarker concentrations were established in blood samples of healthy adults and children. Advantages of the LC-MS/MS as an analytical technique include possibility of simultaneous measurement of multiple analytes and ability of confirming their identity. In this thesis we proposed and evaluated approaches for the assessment of the specificity of analysis in the methods that use tandem mass spectrometry detection. To enhance throughput of the LC-MS/MS tests for the biomarkers that have endogenous or exogenous isomers an approach was developed for quantitation of isomers from unresolved chromatographic peaks. Using methods developed in this thesis we performed a study of the steroidogenesis in ovarian follicles of healthy women and women with polycystic ovary syndrome (PCOS). Obtained data on the steroid concentrations and associations between the steroid metabolites in the pathway would be helpful for better understanding of the ovarian pathophysiology. Повний текст джерелаNucleotides, term including nucleoside mono-, di- and triphosphates, are endogenous compounds playing various roles in biology. They are components of nucleic acids, provide energy to metabolic reactions and act as carriers or second messenger. The study of endogenous nucleotides has become of great interest in physiological and pathological conditions. We developed a method for the quantification of endogenous nucleotides, using an on-line extraction on a WAX column coupled with LC-MS/MS. Analytical separation is performed on a Hypercarb column, without ion pairing agent in the mobile phase. The use of a triple quadrupole mass spectrometer following positive mode ionization allows the unambiguous identification of nucleotides presenting the same mass. Extraction and separation of nucleotides are achieved within 20 min and the method including re-equilibration of the two columns within 37 min. The method was validated for the quantification of nucleoside mono- and triphosphates, and could be applied to series of more than twenty biological samples. Secondly, in a study based on design of experiments, pre-analytical parameters influencing results of intracellular nucleotides were compared in four cell lines. We demonstrated that optimal pre-analytical parameters depend on cell lines. This clearly highlights the importance of pre- analytical conditions for the quantification of intracellular nucleotides to be as representative as possible of the real levels in cells. Then, thanks to experience acquired during the development and the validation of the analytical method, scientific collaborations have been established with several cancer research teams. For example, implication of nucleotide metabolism in replicative stress induced by oxidative stress or in the metabolic reprogramming in cancer cells was studied. Results obtained by our analytical approach were complementary to those obtained by other techniques. To conclude, our work consisted on the study of the entire workflow for the analysis of endogenous nucleotides in various biological samples

Повний текст джерелаToday, the growth of protein sequencing data significantly exceeds the growth of capacities to analyze these data. In line with this data deluge and urgent needs in new bioinformatics tools our work deals with the development of new algorithms to better understand the sequence-structure-function relationship. Proteins contain a large portion of periodic sequences representing arrays of repeats that are directly adjacent to each other, so called tandem repeats (TRs). TRs occur at least in 14% of all proteins. Highly divergent, they range from a single amino acid repetition to domains of 100 or more repeated residues. Numerous studies demonstrated the fundamental functional importance of such TRs and their involvement in human diseases, especially cancers. Here we show the importance of integrating several TR detectors to get the most complete set of TRs in proteomes. We designed an appropriate pipeline and developed a filter to speed the process as well as a new scoring module to select relevant structured TRs. In addition, we undertook a large scale analysis of TRs in 94 proteomes. This large scale analysis allowed us to update previous census of TR showing that TRs occurs in 64% of all proteins and leads to a better understanding of TR in terms of their characteristics, composition and implication in human disease

Повний текст джерелаPhotovoltaic (PV) solar energy consists on the ability of certain materials to convert the photon energy into electric current. The development of PV conversion systems in the past thirty years has led to considerable improvements in terms of cost and performance in the field of renewable energies.A multi-junction (MJ) cell, based on III-V semiconductor materials, is a stack of sub-cells with decreasing gaps which notably allows wider use of the solar spectrum. Exposing these PV cells to a concentrated solar flux can significantly increase the electrical power generated, and therefore substantially lower the cost of electricity yielded.The world record is currently held by the partnership Soitec / Fraunhofer ISE with an efficiency of 46.0 % measured on a four-junction cell GaInP/GaAs//InGaAsP/InGaAs for a concentration ratio of 508 X (where 1 X = 1 sun = 1 kW/m²).The objective of the work in this thesis is to propose an alternative to existing cells, easier to implement with monolithic MJ cells grown on a GaSb substrate for solar concentrations of 1 000, which corresponds to a direct irradiance of 1 MW/m². This type of cell, due to the good complementary of the material gaps and its favorable band alignments, is a realistic and original alternative to existing cells for use under highly concentrated solar flux.To better understand the optimal multijunction III-Sb cell, the work carried out consisted on the manufacturing and characterization of the three sub-cells independently.These three epitaxial samples are Al0,9Ga0,1As0,07Sb0,93 (Top cell), the Al0,35Ga0,65As0,03Sb0,97 (Middle cell) and GaSb (Bottom cell) having as respective gaps 1.6 eV, 1.22 eV and 0.726 eV at 300 K.The work presented in this thesis is:- The establishment of all the technological steps required to manufacture the cells (metal deposition, wet and dry plasma etching ...).- The characterization of metallization by TLM structure (Transmission Line Method) with the best result being a three-layer metallization Cr/Pd/Au (30/30/30 nm) on a GaSb P-type substrate.- The characterization under dark of current-voltage electrical parameters of PV cells at room temperature and in function of the temperature.- The thermal characterization by measuring the thermal conductivity of the materials and a surface temperature mapping in function of the concentrated solar flux in realistic conditions.- The electro-optical characterization by spectral response, from which we calculated the external quantum efficiency which is the ratio between the amount of electrons created and the amount of incident photons.- The characterization under 1 sun illumination (1 000 W/m²) in a solar simulator and in realistic conditions of which we compared the electrical parameters.- The characterization of solar cells under (highly) concentrated solar flux in the PROMES laboratory.The best efficiencies for Bottom, Middle and Top PV cells respectively are 4.6 % for 40 X (close to the state of the art), 8.2 % for 96 X and 5.4 % for 185 X (world first for these quaternary materials).This work was cofounded by the Ministry of Education and Research (ED Research grant) and Labex SOLSTICE

Повний текст джерелаAn evolutive tandem repeat consists in a series of almost contiguous copies, such that each copy is strongly similar to its predecessor and its successor. There is no model and the first and the last copy can be completely different. It differs from a ``classical'' tandem repeat that considers a model and a radius around this model. This ``new'' type of repeat has been pointed out during the assembly of the human genome. In this manuscript, I first define, in a formal way, the evolutive tandem repeats from observations achieved in biological sequences. After having carried out tests using well known existing softwares, I conclude that no one was able to detect it correctly and that they are not part of the sequences that have been submitted to databanks. I therefore designed algorithms that are able to locate this specific type of repeats: a first algorithm, quadratric in the length of the sequence, then a linear one. These algorithms have been implemented in order to provide biologists with tools that detect efficiently these repeats in real large sequences, such as human chromosomes. Several parallelisation methods are presented too. The results of the carried out tests on chromosomes show the existence of such repeats in genomes
