Tandem solar cells are potentially much more efficient than the silicon solar cells that currently dominate the market but require materials with different bandgaps. This thesis presents work on silicon nanocrystals (Si-NC) embedded in silicon carbide (SiC), which are expected to have a higher bandgap than bulk Si due to quantum confinement, with a view to using them in the top cell of a tandem cell. The strong photoluminescence (PL) of precursor films used to prepare Si-NC in SiC (Si-NC/SiC) was markedly reduced upon Si-NC formation due to simultaneous out-diffusion of hydrogen that passivated dangling bonds. This cannot be reversed by hydrogenation and leads to weak PL that is due to, and limited by, non-paramagnetic defects, with an estimated quantum yield of ≤5×10-7. Optical interference was identified as a substantial artefact and a method proposed to account for this. Majority carrier transport was found to be Ohmic at all temperatures for a wide range of samples. Hydrogenation decreases dangling bond density and increases conductivity up to 1000 times. The temperature-dependence of conductivity is best described by a combination of extended-state and variable-range hopping transport where the former takes place in the Si nanoclusters. Furthermore, n-type background doping by nitrogen and/or oxygen was identified. In the course of developing processing steps for Si-NC-based tandem cells, a capping layer was developed to prevent oxidation of Si-NC/SiC, and diffusion of boron and phosphorus in nanocrystalline SiC was found to occur via grain boundaries with an activation energy of 5.3±0.4 eV and 4.4±0.7 eV, respectively. Tandem cells with a Si-NC/SiC top cell and bulk Si bottom cell were prepared that exhibited open-circuit voltages Voc of 900 mV and short-circuit current densities of 0.85 mAcm-2. Performance was limited by photocurrent collection in the top cell; however, the Voc obtained demonstrates tandem cell functionality.

Анотація:

La spectrometrie de masse tandem est une methode microanalytique destructrice permettant en particulier l'analyse structurale de composes organiques et biologiques. La methode classiquement utilisee (cid) consiste a selectionner un faisceau d'ions avec un premier analyseur en masse, a l'exciter en lui faisant subir des collisions avec des atomes ou molecules d'un compose en phase gazeuse, et a analyser les ions fragments obtenus par dissociation des ions actives a l'aide d'un deuxieme analyseur. Une alternative, proposee en 1985, consiste a exciter les ions par collision avec une surface metallique (sid). L'application de cette methode a l'etude de la fragmentation de divers types de composes (alcane, peptide, polyaromatique hydrocarbone) montre qu'elle permet un meilleur controle de l'excitation de l'ion incident et un acces a des energies d'activation plus elevees. Elle presente toutefois un desavantage majeur: la neutralisation partielle du faisceau incident, qui conduit a une perte de sensibilite. Cette technique a ete etudiee sur trois appareils differents, en essayant differentes geometries d'optique ionique et differents types de surface (surface metallique en acier inoxydable, cuivre ou aluminium et surface semi-conductrice constituee d'une galette de microcanaux). Les resultats obtenus montrent une neutralisation elevee pour les surfaces metalliques, plus faible dans le cas de la surface en aluminium et encore plus faible dans celui de la galette a microcanaux. Toutefois, cette derniere solution semble peu adaptee a la gamme d'energie cinetique compatible avec les appareils a filtre quadripolaire utilises (quelques ev a 100ev), l'incidence rasante du faisceau limitant l'energie d'activation. L'ensemble des resultats obtenus permet d'etablir que cette technique d'activation peut permettre une extension du domaine d'application de la spectrometrie de masse tandem, en particulier pour l'analyse structurale de molecules de grande taille. La recherche de materiaux minimisant la neutralisation partielle semble toutefois indispensable

Анотація:

Grâce à leurs multiples sites de fonctionnalisation, les cyclopenténones sont des intermédiaires particulièrement utiles dans la synthèse de produits naturels d’intérêt thérapeutique. En particulier, les composés 4-aminocyclopenténones permettent l’accès au motif aminocyclopentitol présent dans des molécules bioactives telles que le peramivir, la pactamycine, ou encore la trehazoline. L’une des méthodes les plus efficaces pour accéder aux 4-aminocyclopenténones est la réaction de cyclisation d’aza-Piancatelli. Cette réaction est basée sur le réarrangement de 2-furylcarbinols en présence d’un nucléophile azoté suivant un mécanisme faisant intervenir une électrocyclisation 4π-conrotatoire. Au sein de notre laboratoire, un système catalytique simple, utilisant un complexe de calcium combiné à un sel d’ammonium, a été mis au point pour avoir accès à ces composés. Cette méthode présente de nombreux avantages : elle est efficace (rendements allant jusqu’à 98%), rapide (15 à 30 minutes), elle nécessite seulement 1 mol% de catalyseur dans des conditions commodes (solvants non distillés et réaction à l’air libre) sur une grande échelle (multi-grammes). Dans ce contexte, nous avons cherché à exploiter tout le potentiel de cette réaction en synthétisant des 2-furylcarbinols plus complexes dans le but de concevoir un accès direct à des squelettes de composés bioactifs. Nous nous sommes en particulier concentrés sur la synthèse totale de la jogyamycine. Par ailleurs, nous avons effectué la synthèse totale de la bruceolline D avec un rendement global de 16% sur 5 étapes. Nous avons également développé une nouvelle séquence réactionnelle impliquant une réaction d’aza-Piancatelli suivie d’une réaction d’hydroamination promue par des sels de cuivre qui fournit un large éventail de cyclopenta[b]pyrroles polyfonctionnels à partir de 2-furylcarbinols facilement accessibles incorporant un groupement alcyne. Par cette méthode, 42 cyclopenta[b]pyrroles ont été obtenus avec des rendements atteignant 98%. Une caractéristique originale de notre approche est liée à l'utilisation d'anilines ortho-substituées. En effet, dans ce cas, des atropisomères avec un excès diastéréomérique supérieur à 20:1 ont pu être obtenus, accompagnés par la création d'une chiralité axiale N-C lors de l'étape d'hydroamination. A ce jour, il n'existait aucun autre exemple de synthèse atropodiastéréosélective de cyclopenta[b]pyrroles. Au cours de nos recherches sur le champ d’application de la réaction, nous avons remarqué que les cyclopenta[b]pyrroles pouvaient se réarranger spontanément en cyclopenta[b]pyrrolines suivant une déaromatisation lorsque l’hexafluoroisopropanol était utilisé comme solvant, permettant d’obtenir 13 cyclopenta[b]pyrrolines avec des rendements variant de 44 à 73%. Nous nous sommes également intéressés à la fonctionnalisation du motif cyclopenta[b]pyrrole pour illustrer l'utilité de notre méthodologie, en combinant en particulier cette séquence réactionnelle avec des réactions de type Friedel-Crafts. L’un des objectifs du projet de thèse était le développement de la version énantiosélective de la réaction. Après la publication récente de trois articles utilisant des acides phosphoriques chiraux en tant que catalyseurs, nous avons envisagé une autre approche pour la synthèse asymétrique de 4-aminocyclopenténones, s’appuyant sur l’utilisation d’une copule chirale (sulfoxyde chiral) portée par l’aniline en collaboration, avec le Dr. Wencel-Delord et le Pr. Colobert (Université de Strasbourg). Ainsi nous avons pu générer les 4-aminocyclopenténones correspondantes avec d’excellents rendements et diastéréosélectivités Due to their various functionalizable sites, cyclopentenones are very useful intermediates for the synthesis of natural products of therapeutic value. In particular, 4-aminocyclopentenones enable the access to the aminocyclopentitol frameworks, which are present in a variety of bioactive molecules such as peramivir, pactamycin, or trehazolin. One of the most efficient methods to access 4-aminocyclopentenones is the aza-Piancatelli reaction. It is based on the rearrangement of 2-furylcarbinols in the presence of a nitrogen nucleophile following a mechanism involving a 4π-conrotatory electrocyclization. In our laboratory, a simple catalytic system using a calcium complex combined with an ammonium salt was developed to gain access to these compounds. This method has many advantages : it is effective (yields up to 98%), fast (15 to 30 minutes), it requires only 1 mol% of catalyst under pratical conditions (undistilled solvents without an inert atmosphere) on a large scale (multi-gram). In this context, we sought to extend the scope of this reaction by designing more complex 2-furylcarbinols in order to directly access skeletons of bioactive compounds. In particular, we focused on the total synthesis of jogyamycin. In addition, we achieved the total synthesis of bruceollin D with an overall yield of 16% over five steps. We also developed a new reaction sequence involving an aza-Piancatelli reaction followed by a hydroamination reaction promoted by a copper salt. This sequence provides a wide range of highly functionalized cyclopenta[b]pyrroles from readily-available 2-furylcarbinols substituted by an alkyne moiety. Following this method, 42 cyclopenta[b]pyrroles were obtained with yields up to 98%. An original feature of this transformation is related to the use of ortho-substituted anilines. Indeed, in this case, atropisomers with a diastereomeric excess superior to 20:1 could be obtained accompanied by the creation of an chiral N-C axis during the hydroamination step. To date, no other example of atropodiastereoselective synthesis of cyclopenta[b]pyrroles has been reported. During our investigations on the scope of the reaction, we noticed that cyclopenta[b]pyrroles underwent a rearrangement into cyclopenta[b]pyrrolines, following a dearomatization when hexafluoroisopropanol was used as solvent. This transformation led to 13 cyclopenta[b]pyrrolines with yields ranging from 44% to 73%. We also investigated the functionalization of the cyclopenta[b]pyrrole motif to illustrate the synthetic utility of our methodology, notably by combining this reaction sequence with a Friedel-Crafts reaction. One of the objectives of this Ph.D. was the development of an enantioselective version of the reaction. After the recent publication of three papers using chiral phosphoric acids as catalysts, we devised another approach for the asymmetric synthesis of 4-aminocyclopentenones, relying on the use of a chiral auxiliary (chiral sulfoxide) in collaboration with Dr. Wencel-Delord and Pr. Colobert (University of Strasbourg). Thus, we were able to provide the corresponding 4-aminocyclopentenones with excellent yields and diastereoselectivity

Cranes are often the most conspicuous machines on a construction site. This is due to their large size, in addition to the important role they have in transporting heavy payloads vertically and horizontally. There are two major families of construction cranes: tower cranes and mobile cranes. Mobile cranes that are mounted on tracks are a subgroup referred to as ``crawler cranes''. Crawler cranes are widely used on construction sites, and are a backbone of the United States construction industry, thus a detailed study of these cranes' behavior is essential. This research studies the tip-over stability of crawler cranes in heavy-lifting applications. Two major applications are discussed: crawler cranes using movable counterweights and crawler cranes in tandem lifting.

Les structures protéiques peuvent être divisées en répétitives et apériodiques, les structures apériodiques correspondant pour la plupart à des protéines globulaires. Les protéines répétitives (PRs) contiennent des unités de répétitions adjacentes, appelées séquences répétées en tandem (TRs). Les PRs sont abondantes et ont une importance fonctionnelle fondamentale. De plus de nombreuses études ont démontré l'implication des TRs dans les pathologies humaines. Ainsi, la découverte des PRs et la compréhension de leur relation séquence-structure-fonction, offrent des perspectives de recherche prometteuses.Le développement d’initiatives en génomique structurale, combiné à une meilleure adaptation des techniques de cristallographie et de RMN à l’étude des protéines non globulaires, a permis d’élucider la structure d’un nombre croissant de PRs, d’où la nécessité de mettre en place un système de classification. Les structures répétitives ont été réparties en cinq classes, principalement fondées sur la longueur des TRs: Classe I - agrégats cristallins; Classe II - structures fibreuses; Classe III - structures allongées, dont la stabilité dépend des interactions qui s’établissent entre les motifs répétés. Classe IV - structures répétitives fermées ; Classe V - structures en collier de perles. Les efforts de ces dernières années ont abouti au développement d’outils bioinformatiques utiles à la détection et l'analyse d'éléments répétitifs présents au sein des structures protéiques (3D TRs). En fonction des caractéristiques des répétitions, certaines méthodes fonctionnent mieux que d'autres, mais, jusqu’à présent, aucune ne permettait de couvrir toute la gamme des répétitions. Ce constat nous a incités à développer une nouvelle méthode, appelée détecteur de protéines en tandem (TAPO). TAPO exploite les périodicités des coordonnées atomiques ainsi que d'autres types de représentation structurale, comprenant les chaînes générées par un alphabet conformationnel, les cartes de contact entre résidus, et les arrangements en vecteurs d'éléments de structure secondaire. Actuellement, sept scores, issus des caractéristiques analysées par TAPO, sont combinés à l’aide d’une Machine à Vecteur Support pour produire un score final permettant de différencier les protéines renfermant ou non des 3D TRs. En atteignant 94% de sensibilité et 97% de spécificité pour la référence actuelle, TAPO présente des performances améliorées par rapport aux autres méthodes de pointe. Le développement de TAPO offre de nouvelles opportunités pour l’analyse à grande échelle des protéines renfermant des 3D TRs. Ainsi, notre analyse de la base de données PDB, à l’aide de TAPO, a montré que 19% des protéines contiennent des 3D TRs. L'analyse à grande échelle des structures 3D TRs dans PDB nous a également permis de découvrir plusieurs nouveaux types de structures répétitives, absents de la classification existante et dont certains sont décrits ici.Nous avons entrepris une analyse complète des 3D TRs constitutifs du Rossmann Fold (RF). Notre intérêt pour les RFs a été suscité par le fait que de nombreuses protéines RFs représentent un cas ambigüe vis à vis des structures répétitives et non répétitives. A priori, les unités hélice α - feuillet β des RFs devraient avoir une forte tendance à s’empiler et donc, à former des structures répétitives. Afin de déterminer la fréquence à laquelle les RFs forment de longues unités de répétition empilées, nous avons sélectionné, à l’aide de TAPO, des structures contenant des RFs et les avons classées. Notre analyse montre que les RFs typiques ne peuvent pas être clairement définis comme des structures répétitives mais plutôt comme des unités de structures globulaires, comptant au plus trois répétitions α-β. Des éléments de discussion seront proposés pour tenter d’expliquer cette observation surprenante In general, protein structures can be divided into: repetitive and aperiodic structures. Most of the aperiodic structures are globular proteins. The repetitive proteins contain arrays of repeats that are adjacent to each other, called Tandem Repeats (TRs). Proteins containing TRs are abundant and have fundamental functional importance. Numerous studies demonstrated the involvement of such TR-containing proteins in human diseases. Furthermore, genetic instability of these regions can lead to emerging infection threats. Additionally, TR-containing structures have generated significant interest with respect to protein design as they can make excellent scaffolds for specific recognition of target molecules. Therefore, the discovery of these domains, understanding of their sequence–structure–function relationship promises to be a fertile direction for research.The growth of structural genomics initiatives, in combination with improvements in crystallographic and NMR techniques aimed at non-globular proteins, has resulted in an increase in structurally elucidated TR proteins. This has necessitated the development of classification schemes. Structural repeats were broadly divided into five classes mainly based on repeat length; Class I – crystalline aggregates; Class II – fibrous structures such as collagen; Class III – elongated structures where the repetitive units require each other for structural stability such as solenoid proteins; Class IV – closed repetitive structures, such as TIM-barrels and Class V – bead on a string structures such as tandems of Ig-fold domains. Despite this progress, the majority of bioinformatics approaches have focused on non-repetitive globular proteins.In recent years, efforts have been made to develop bioinformatics tools for the detection and analysis of repetitive elements in protein structures (3D TRs). Depending on the size and character of the repeats, some methods perform better than others, but currently no best approach exists to cover the whole range of repeats. This served as a motivation for the development of our method called the TAndem PrOtein detector (TAPO). TAPO exploits, periodicities of atomic coordinates and other types of structural representation, including strings generated by conformational alphabets, residue contact maps, and arrangements of vectors of secondary structure elements. Currently, seven feature based scores produced by TAPO are combined using a Support Vector Machine, producing a score to enable the differentiation between proteins with and without 3D TRs. TAPO shows an improved performance over other cutting edge methods, achieving 94% sensitivity and 97% specificity on the current benchmark. The development of TAPO provided new opportunities for large scale analysis of proteins with 3D TRs. In accordance with our analysis of PDB using TAPO, 19% of proteins contain 3D TRs. The large scale analysis of the 3D TR structures in PDB also allows us to discover several new types of TR structures that were absent in the existing classification. Some of them are described in the thesis manuscript. This suggests that TAPO can be used to regularly update the collection and classification of existing repetitive structures. In particular, a comprehensive analysis of 3D TRs related to Rossmann Fold (RF) was undertaken. Our special interest in RFs was based on the observation that many proteins with RFs represent borderline cases between repetitive and non-repetitive structures. In principle, α-helix-β-strand units of RFs should have a strong potential to stack one over the other, forming repetitive structures. To probe the question of how frequently RFs form long arrays of stacked repeats, we selected by using TAPO known RF-containing structures and classified them. Our analysis shows that typical RFs cannot be clearly defined as repetitive, rather they are part of globular structures with up to 3 αβ-repeats. Ce manuscrit décrit l’utilisation d’une réaction tandem asymétrique hétéro Diels-Alder/allylboration dans la synthèse de plusieurs produits naturels. Le motif hydroxyalkyl-dihydropyrane obtenu a permis d’accéder, selon l’aldéhyde engagé dans la réaction d’allylboration, à différentes molécules appartenant à la famille des Styryllactones. Il a été montré que l’utilisation du paraformaldéhyde permet d’obtenir des structures contenant une double liaison en α de l’hétéroatome du cycle, ce qui ouvre la voie à la synthèse d’autres molécules naturelles. Cette réaction tandem a également permis de synthétiser le fragment C₁₅-C₂₇ du macrocycle laulimalide ainsi que trois analogues stéréochimiques et structuraux. Enfin, des essais ont été menés pour synthétiser le deuxième fragment C₁-C₁₄ du laulimalide He utilisation of an hetero Diels-Alder/allylboration asymmetric tandem reaction for the synthesis of several natural products, has been studied. It has been shown that the hydroxyalkylated dihydropyran unit, obtained frome this reaction, opens the access to different molecules from the Styryllactones’ family. It has been shown that the use of the paraformaldehyde during the process, makes possible the synthesis of natural products bearing a double bond adjacent to the heteroatom of the cycle. This tandem reaction has also been employed for the synthesis of the laulimalide’s C₁₅-C₂₇ fragment and three analogs. Finally, some tests have been managed for the second C₁-C₁₄ fragment’s synthesis

Le but principal de ce travail a été de développer de nouvelles méthodologies pour la synthèse de molécules d'intérêt biologique, telles que des azafluorénones, et des précurseurs d'analogues de la variolines B. Tout d'abord, nous avons synthétisé des diarylcétones, précurseurs pour nos cibles, par une séquence déprotocupration-aroylation. Ensuite, nous les avons impliquées dans une étape de couplage pallado-catalysé : les substrats ont subi une C-H activation pour aboutir aux azafluorénones. Nous nous sommes tournés par la suite vers la synthèse d'azafluorénones substituées. Ces dernières ont été synthétisées par un processus auto-tandem en combinant le couplage de Suzuki ou le couplage de Heck avec la cyclisation intramoléculaire catalysée par le palladium. Certaines de ces molécules ont été évaluées pour leurs propriétés biologiques et ont révélé des bonnes activités cytotoxique, antimalariale, antibactérienne et antifongique. Enfin, nous avons réussi à synthétiser des précurseurs d'analogues de variolines en seulement trois étapes à partir d'un produit commercial The main purpose of this work was to develop new methodologies for the synthesis of molecules of biological interest, such as azafluorenones and precursors of variolin B analogs. First, we synthesized diarylketones, precursors for our targets, via a deprotocupration-aroylation sequence. Then, we involved them in a pallado-catalyzed coupling step: some substrates underwent C-H activation to provide azafluorenones. Next, we turned our attention to the synthesis of substituted azafluorenones. The latter were synthesized by a tandem process combining Suzuki coupling or Heck coupling with intramolecular cyclization catalyzed by palladium. Some of these molecules were biologically evaluated and showed good biological activities: cytotoxic, antimalarial, antibacterial and antifungal. Finally, we succeeded in synthesizing precursors of variolin analogues in only three steps from a commercial product

São biossintetizadas em condições específicas por cianobactérias, principalmente em corpos d\'água eutrofizados. Apresentam organotropismo ao fígado e DL50 entre 50-500 µg.kg-1 peso (rato, i.p.). A exposição aguda pode impactar a saúde humana ao desarranjar o parênquima hepatocelular por hiperfosforilação do citoesqueleto e produzir intenso sangramento, podendo levar à morte por choque hipovulêmico um homem adulto e são promotoras tumorais, quando ocorre exposição crônica, por inibição de fosfatases 1 e 2A. Os congêneres das microcistinas apresentam toxicidades distintas e é importante caracterizá-los em corpos d\'água para se estimar o risco à saúde humana. Na literatura, não há convergência quanto à forma de extração das toxinas, preparo das amostras para análise e condições cromatográficas para o isolamento e quantificação. Neste trabalho, diversas análises foram conduzidas para se determinar as melhores formas de extração das toxinas, clean up da amostra e condições cromatográficas para amostras de campo ou cultura de cianobactérias. A extração hidro-alcoólica (MeOH:H2O, 3:1, v/v) parece ser a melhor forma de extração, seguida de partição líquido-líquido para amostras de campo (florações) e extração em fase sólida (cartuchos C18) para algas cultivadas em laboratório. Por meio de isolamento por cromatografia líquida de alta eficiência (HPLC) com detectores UV-VIS e pela análise do padrão de fragmentação das moléculas por espectrometria de massas (MS) com ionização por eletrospray em Tandem (ESI-MS/MS) determinou-se a presença das variantes de microcistinas MC-RR, MC-LR, MC-YR e MC-hRhR na amostra de água do Reservatório Billings e [Asp3]MC-LR na cepa Microcystis aeruginosa BCCUSP 235. Este método parece ser adequado para a extração e caracterização de microcistinas presente em corpos d\'água e em cultura de laboratório e a variante MC-hRhR foi caracterizada pela primeira vez na literatura mundial. It is well known that eutrophicated lakes and water reservoirs can be a serious concern to wildlife, domestic livestock and human health since cyanobacterial blooms are prone to occur. Some cyanobacteria (blue-green algae) species which form bloom may biosynthesize a family of hepatotoxic peptides named microcystins under certain circumstances, related specially to nitrogen: phosphorus ratio, temperature, pH and light intensity of the water body. There are at least 70 variants of microcystins with different toxicity, DL50 ranging from 50 to 500 µg.kg-1 (in mouse, i.p.). Acute exposure provoke hyperphosphorylation of the cytoskeleton, in liver cells, leading to cellular disruption with haemorrhage and the long term exposure can cause tumor promotion by inactivation of serine/threonine phosphatases 1 and 2A (PP1 and PP2A). Therefore, it is very important to determine and quantify microcystin variants present in water reservoirs. There are several methods available to evaluate qualitative and quantitatively microcystin and its variants in water reservoir and estimate human risks. Such methods comprise biological, biochemical and physico-chemical assays. In this work, we have isolated and analyzed hepatotoxins by High Performance Liquid Chromatography (HPLC) with photodiode array (PDA) detector and their structures were elucidated by Tandem - MS/MS with eletrospray ionization (ESI-MS/MS). MC-RR, MC-LR, MC-YR and MC-hRhR were determined in samples collected at Billings reservoir and [Asp3]MC-LR was found in the strain Microcystis aeruginosa BCCUSP 235. The procedures developed herein can be a powerful tool for isolation and determination of microcistin and its derivatives founded in water reservoirs. Also, we have described for the first time a new variant of microcistin: MC-hRhR.

Depuis les années 70, la catalyse organométallique a profondément influencé la chimie organique. La richesse de la réactivité des métaux de transition en phase homogène motive la réalisation de nombreux travaux de recherche visant à « hétérogénéiser » des réactions de catalyse homogène. En transposant ces réactions à des systèmes hétérogènes, on peut bénéficier des avantages de la première (sélectivité, efficacité) et de la seconde (chimie verte, recyclabilité du catalyseur). Dès lors, la catalyse métallique hétérogène constitue une alternative et un domaine de recherche fertile et novateur pour le développement de réactions « one pot » en accord avec la chimie verte. Mon travail de thèse s'inscrit dans cette problèmatique par le développement de méthodologies de synthèse faisant intervenir des catalyseurs bimétalliques hétérogènes multitâches. Nous avons développé une méthode simple et rapide permettant de générer des catalyseurs hétérogène bimétallique Pd-M (avec M = Au, Cu). Les nanoparticules ont été obtenues par imprégnation des précurseurs métalliques sur le support suivie par la réduction des métaux. Les structures cristallines ont été caractérisées en combinant plusieurs techniques tels que : MET, HRMET, XPS et DRX. L'activité catalytique et la recyclabilité des systèmes bimétalliques et monométalliques ont été comparées et étudiées pour des réactions de : (i) Sonogashira, (ii) Sonogashira/cyclisation, (iii) Sonogashira/click et (iv) Click/CH arylation Since the 70s, organometallic catalysis profoundly influenced organic chemistry. The richness of the reactivity of transition metals in homogeneous phase motivates the realization of extensive research to « heterogenize » reactions of homogeneous catalysis. Transposing the reactions to heterogeneous systems, we can take advantage of the first (selectivity, efficiency) and second (green chemistry, recycling of the catalyst). Therefore, the heterogeneous metal catalysis is an alternative and area of fertile and innovative research for the development of « one pot » reaction in accordance with green chemistry. My thesis work is part of this problem through the development of synthetic methodologies involving heterogeneous metallic multitask catalysts. We have developed a method to generating heteregeneous multi-metallic catalysts Pd-M (with M = Au, Cu). The nanoparticles were obtained by impregnation of metallic precursors onto the support followed by reducing the metal ion under a hydrogen atmosphere. The crystal structures of nanoparticles were characterized by combining different experimental techniques such as : TEM, HRTEM, XPS and XRD. The catalytic activity and recyclability of heteregeneous bimetallic catalysts has been compared to monometallic catalysts and studied in : (i) Sonogashira coupling reactions, (ii) Sonogashira coupling/cyclization, (iii) Sonogashira coupling/Click and (iv) Click/CH arylation

A more detailed knowledge of the inhibition profiles for new chemical entities with respect to the major cytochrome P450 enzymes is prerequisite for their registration. The use of recombinant P450 enzymes is intended to limit interactions with other minor P450 enzymes and their reductases that are present in human liver microsomes. Using combined substrates (phenacetin, diclofenac, S-mephenytoin, bufuralol and midazolam) with combined recombinant P450 enzymes (CYP1A2, 2C9, 2C19, 2D6, and 3A4), a high-throughput LC/MS/MS method for the determination of IC50 values using six concentrations for up to thirteen compounds in a single assay has been developed. Kinetic analysis of each enzyme-substrate pair under single and combined conditions highlighted the poor selectivity of phenacetin. Subsequent manipulation of relative P450 enzyme concentrations reduced the cross-reactivity of this substrate. IC50 determinations with α-naphthoflavone (0.04µM), sulphapenazole (0.26µM), tranylcypromine (9µM), quinidine (0.02µM) were also similar under single and combined conditions. Further evaluation of the assay was carried out using 11 known inhibitors of cytochromes P450 and 52 new drug candidates. Using the high sensitivity and selectivity offered by tandem mass spectrometry in conjunction with hydrophilic interaction liquid chromatography (HILIC), a method capable of measuring nanomolar levels of unconjugated or free metanephrines in plasma has been developed. Elevated levels of metanephrine, normetanephrine and 3-methoxytyramine can be indicative of a rare tumour of the chromaffin cells termed phenochromocytoma. Limits of quantitation for metanephrine (0.02nmol/L), normetanephrine (0.21nmol/L) and 3-methoxytryaramine (0.14nmol/L) are within or below published reference ranges where documented. Linearity of response has been shown to at least 10.0 nmol/L for all analytes with intra- and inter-assay variability shown to be <15%CV across the range of expected physiological concentrations.

L'élaboration de méthodes de synthèse, basées sur l’utilisation d’espèces métalliques a été un sujet de tous les instances en chimie organique. Malgré l’efficacité des métaux utilisés en catalyse homogènes, leurs procédures de recyclage restent limitées. Ce pourquoi, une contrainte supplémentaire a été placée dans la conception de catalyseurs, pouvant offrir à la fois l'efficacité de la catalyse homogène et le recyclage de l’hétérogène. Dans ce contexte, les nanoparticules métalliques sont apparues comme objet phare, en raison de leurs propriétés physico-chimiques inégalées. On a découvert que les nanoparticules de métaux nobles présentaient des propriétés catalytiques similaires dans certains cas, aux complexes monoatomiques. De plus, les Au NPs ont montré une activité catalytique remarquable dans l'oxydation d’alcools activés sous O2. Nous avons donc envisagé des procédures multicatalytiques, basées sur les NPs d’Au. Notre choix d'utiliser des catalyseurs solides était pertinent, puisque les nano-catalyseurs, pour lesquels la fraction de sites actifs se trouve en surface, limitent les risques de cross-quenching. Ici, nous présentons trois nouveaux procédés bicatalytiques permettant l’accès, à des chromenes/quinoléines (53-93%) via une oxydation / Michael Addition/ aldolisation, combinant nanocatalyse et catalyse basique, l’accès à des ortho-THC (50-81%) via oxydation / arylation / cyclisation, combinant nanocatalyse et catalyse supportée, ainsi qu’une une oxydation / hydrolyse en cascade, pour accéder à l’HMLA (86%, sel 93%), un grand panel de produits d'activité biologique reconnue, utilisé en parfumerie ou visant une pré-industrialisation via la chimie en flux continu Elaboration of synthetic methods based on metal-catalyzed reactions has been a hot topic in organic chemistry. Despite good efficiency, catalysis proceeding homogeneously, are limited in the operation of recovering/recycling of the catalysts. An important stress was placed to design catalysis, offering both the efficiency of homogeneous catalysts and the recyclability of heterogeneous catalysts. In this context, metal nanoparticles merged as a key tool, due to their unique physical and chemical properties. Notably, Au NPs have shown remarkable catalytic activity in the oxidation of activated alcohols under O2 atmosphere. Since now, the access to more complex molecules is the next step forward for this field, we envisioned multicatalytic roads, based on the oxidation of activated alcohols via supported Au NPs. Our choice of using solid catalysts was relevant, since nanostructured catalysts for which the fraction of active sites are located on the surface, limit the risk of cross-quenching. The latter carbonyl formed, could be further converted in situ, via tandem protocol. Herein, we developed novel, atom- and step-economical bicatalytic one-pot processes, to access substituted chromenes/quinolines (53-93%) by tandem oxidation/hetero-Michael addition/aldolisation combining nanocatalysis and base catalysis, ortho-THCs (50-81%) via tandem oxidation/arylation/cyclisation combining nanocatalysis and supported catalysts and a tandem cascade oxidation/hydrolysis to access HMLA (86%, sel 93%). Cette thèse porte sur l'analyse de de défauts structuraux d'un pseudo-substrat GaP/Si. L'objectif principal concerne la qualité structurale de la couche épitaxiale de GaP sur un substrat de Si, en tant que pierre angulaire du développement d'une cellule solaire tandem à haut rendement avec des dispositifs bas-couts (jonctions silicium et CIGS) et une couche intermédiaire Ill-V. Tout d'abord, l'étude porte sur la caractérisation de la distribution des dislocations dans le substrat vicinal GaP sur Si(001), avec une méthode de diffusion des rayons X ayant une résolution sous-micrométrique, appelée K-Map. Les informations locales d'inclinaison et de déformation sont obtenues par une analyse de l'ensemble de données complexes 5D. Cette étude révèle une distribution anisotrope des dislocations dans différentes directions cristallographiques, liée aux marches à la surface du substrat de Si, et une distribution non homogène des dislocations, liée à la tendance à la formation de regroupements de marches. La deuxième par1ie de l'étude porte sur la croissance et la caractérisation de CIGS sur GaP/Si, en vue de développer des cellules solaires tandems associant une cellule du dessous en silicium monocristallin et une cellule du dessus en CIGS. On observe (par XRD, EDX et HRTEM) que le CIGS est déposé par croissance épitaxiale sur le pseudo substrat GaP/Si et présente une qualité structurale proche d'un monocristal. Le premier essai, non optimisé, d'une cellule solaire simple jonction de CIGS sur pseudo-substrat GaP/Si, permet d'obtenir un EQE très encourageant, similaire à celui obtenu avec la même cellule déposée sur substrat de verre avec des paramètres de dépôts optimisés This thesis focuses on the characterization of the structural defects on a MBE-made GaP/Si pseudo-substrate. The main purpose concerns the structural quality of GaP epitaxial layer on Si substrate, as a cornerstone for the development of high-efficiency solar cell with relatively low cost. Firstly, the study focuses on the characterization of the distribution of dislocations in GaP on Si (001) vicinal substrate, with an advanced submicrometer-beam X-ray scattering method, K-Map. The local tilt and strain information are obtained through an analysis of the complex 5D dataset. This study reveals an anisotropic distribution of the dislocations along different crystallography directions, linked to the steps on the surface of Si substrate, and an inhomogeneous distribution of the dislocations, linked to the tendency to form bunches. The second part of the study is on the growth and characterization ofCIGS on GaP/Si, which is used to explore tandem junctions associating single crystalline silicon bottom cell. The CIGS is observed to be epitaxially grown on the GaP/Si pseudo substrate, combining the results collected from XRD, EDX and HRTEM. A first try of a CIGS/GaP/Si solar cell is realized on a non-optimized GaP/Si pseudo-substrate. The obtained EQE is similar to the one obtained with the CIGS solar cell grown on a traditional glass substrate with the optimised deposition parameters

The recent development of a hydroxy-directed Diels-Alder reaction, which enables control of both regio- and diastereoselectivity, has prompted an investigation of potential applications. Extending the methodology to more complicated dienes, such as those created via the tandem oxy-Cope/Ene reaction, offers the potential for rapid construction of polycyclic molecules with significant stereochemical complexity.* The simplicity and efficiency of this technique has also inspired its employment as the primary disconnection in the production of a library of analogues of the unsaturated dialdehyde Isovelleral.* Also examined herein is the recently discovered tandem oxy-Cope/Ene/Claisen reaction; a highly diastereoselective method for generating trans -decalin systems containing quaternary carbon centers. A polymerase chain reaction (PCR) multiplex assay capable of amplifying 13 rapidly mutating Y chromosome short tandem repeats (RM Y-STRs) simultaneously was developed and optimised. This multiplex assay which was termed RM-Yplex is the first to include all 13 RM Y-STRs including DYF387S1, DYF399S1, DYF403S1a/b, DYF404S1, DYS449, DYS518, DYS526a/b DYS547, DYS570, DYS576, DYS612, DYS626 and DYS627. A developmental validation was performed following the Scientific Working Group for DNA Analysis Methods (SWGDAM) revised guidelines. Robustness and limitations of the assay were demonstrated through a range of studies including reproducibility, sensitivity, specificity, stability and mixture studies. Appropriate controls were used during the studies that included a number of male and female commercial controls including, 2800M, 9948 and Taqman male controls and 9947A female control. An allelic ladder was developed for the assignment of the alleles. This was done by choosing samples with different alleles, amplifying them and then adjusting the volumes of amplified products in a mixture. The developed mixtures were used to balance the composite ladder. Multiple alleles of the various loci included in the ladder were sequenced. Reference haplotypes were developed for the 5 male samples included in the Y chromosome Standard Reference Material 2395 (SRM2395) using RM-Yplex. The International Society of Forensic Genetics (ISFG) recommendations were followed for adopting allele nomenclature. As part of developmental validation, the assay was included in an external proficiency trial which was concluded successfully. An internal validation of RM-Yplex was carried out at the Department of Forensic Sciences and Criminology Laboratory, Dubai where apart from other studies; application of the assay was demonstrated using non-probative forensic casework samples. The value of RM-Yplex was demonstrated for differentiating close male relatives in a case where a previously used Y-STR multiplex assay had shown identical haplotypes for those individuals. 1160 male individual samples were analysed in this study including UAE, other Arabian Peninsula populations as well as two South Asian populations residing in United Arab Emirates. RM-Yplex haplotypes have extremely high power of discrimination. The haplotype diversity for RM-Yplex haplotype is much more than the existing commercial Y-STR assays. Population studies have been carried out for the Arab, Indian and Pakistani populations. AMOVA was conducted for determining the apportionment of diversity and pairwise FST’s were estimated between populations. These have shown a marked homogeneity within the UAE Arab sub-populations. MDS plots of pairwise FST’s indicated that populations were not grouped significantly in accordance with the geographical locations. A network analysis showed the extent of distribution of haplotypes of various populations and their relationships. A highly sensitive and reliable RM-Yplex multiplex assay has been thus developed, which is expected to help genetic populations studies and forensic casework.

Introduction. Myriocin (Myr) is an antifungal compound isolated in the 1972s from Myriococcum albomyces. Myr specifically inhibits serine palmitoyl transferase, the first enzyme involved in the de novo synthesis of Ceramide (Cer), which Intracellular accumulation yields inflammation and apoptosis. Targeting Cer biosynthesis, thus, may be a promising therapeutic treatment for all sphingolipid-related diseases. To test the efficacy of Myr as pharmacological tool able to drive the clinical outcome and rationalize the dose-effect relationship, Myr levels need to be accurately measured in minute amount of biological samples. Since the few available methods are inadequate, this study developed and validated the first method for the measurement of Myr by LC-MS/MS. Methodology. Internal standard 14-OH-Myr (custom synthetized) and SPE were used for sample preparation and purification. A Dionex Ultimate 3000 UPLC coupled to an ABSciex 3200QTrap working in the MRM mode to monitor the analytical transitions (-400.4 →-104.0 for Myr; -402.4 → -104.0 for the IS) were used for analysis. Separation on Inertsil ODS-3 column (3μm, 3.0 x 150mm) lasted 9 min using a 50-100% 0.1% formic acid-acetonitrile gradient. As preliminary study, we measured, in C57 Black6 mice, Myr administrated by intra-tracheal and intravitreal routes, two easy and common administration routes employed for lung and retina related diseases. In addition, for the intra-tracheal administration was used also Myr loaded in Solid Lipid Nanoparticles (Myr-SLN). The dose administrated were 4.69 nmoles (N. mice=6) of Myr-DMSO and 3.15 nmoles (N. mice=4) of Myr-SLN for lung study and 1.87 nmoles (N. mice=3) of Myr-DMSO for retina study. After sacrifice, we measured Myr levels (LC-MS/MS) in lung and retina at 24hrs and 4 hrs from administration, respectively. In the same lung samples, we carried out dose-effect study quantifying Cer content, as a direct “readout”. For “in vitro” uptake study of Myr we used the human C38 cell line as a model system. Cells were seeded into P100 or 6 multi-wells plate and after 24 h Myr was added into LHC-8 media at 10 nmol/mL. The time points checked were 5,10,30,60,120 min, cells were collected, washed and analysed. As a mouse model of Retinitis Pigmentosa (RP) we chose Tvrm4 strain in which it is known that the exposure to high light originate the disease. As a preliminary validation of this model we tested the Cer increase (by LC-MS/MS) and their eventual inhibition by Myr-DMSO treatment (1.87 nmoles; N. mice=7). Results Method validation. As low as 20 fmoles can be detected on-column and quantified with ±15% precision and within ±20% of target level; the method was linear up to 25 pmoles in a typical 150 microL extracted. Examined samples (n>50; median level 0.25 pmoles) include organs of animal models, cultured human cells, Myr loaded into Solid Lipid Nanoparticles as an innovative pharmaceutical formulation (Myr-SLN). Dose-effect in mice lung. The levels of Myr in lung homogenates were 11.0 ± 1.96 pmoles (mean ± SEM) and 4.36 ± 1.16 pmoles (mean ± SEM) for Myr-DMSO and Myr-SLN respectively. Both DMSO-Myr, and SLN-Myr produced a significant decrease in Cer levels vs the respective control. IV Set-up of Myr measurement in mouse retina. The levels of Myr in retina homogenates of wild type mice after intravitreal injection were 0.24 ± 0.08 mean ± SEM for Myr-DMSO. In vitro uptake in C38 cells. The “in vitro” study of Myr-DMSO uptake in C38 cells already showed at the first observation point (5 min) the peak of Myr intracellular concentration (527 pmols/mg prot) followed by two different kinetics order of Myr disappearance. A fast kinetic was featured between 5-10 min, followed by slower trend for the other checked time points (10, 30, 60, 120 min of observation).The lower intracellular concentration of Myr was reached after 120 min (103 pmol/mg prot). The reported Myr concentrations corresponded to about 7-34 pmol/well against the nanomoles added into the media (10 nmol/mL, i.e. 30 nmol/well). Validation of TVRM4 model of Retinitis pigmentsa. In Tvrm4 mice control (DMSO) the light exposure induced a 60% increase of the total Cer (1934 pmol/mg prot; SEM= ± 110) compared with the total Cer content in wild type mice (1209 pmol/mg prot; SEM= ± 154). Overall, injection of Myr-DMSO induced a 28% reduction in mean Cer content, from 1934 pmol/mg prot (SEM= ± 110) to 1394 pmol/mg prot (SEM= ± 107) (p<0,05). Conclusions. We have optimized original mass spectrometric and chromatographic conditions for Myr quantification, to obtain satisfactory resolution and sensibility for the intended use. The method here presented proved to be suitable for dose-efficacy studies in lung mice and demonstrated the activity of this molecule in decreasing CEr levels. These preliminary results open a novel scenario on the activity and metabolism of this molecule, in the view of its use as candidate drug in a number of different sphingolipid-related diseases. We validated the accuracy of Myr administration by intravitreal route. We developed the extraction procedure of Myr from minute amounts of tissue such as a single retina (1 mg) proving a good accuracy of the measurement. The “in vitro” study on Myr uptake highlights an unexpected behaviour of the molecule, with intracellular amounts ranging between 1000-4000 fold lower than the one incubated. As it appears, only pmoles of Myr were found into the cells against nanomols added into the media, in accordance also with the “in vivo” observations. It will be necessary additional pharmacokinetics and biochemical studies to clarify the mechanistic basis by which Myr crosses the cellular membrane and achieve its activity. We demonstrated the validity of a new mouse model of RP by by following the behavior of all the Cer fingerprint. The perspective is to optimize a procedure able to assess, starting from a unique mouse retina, both Myr and Cer by LC-MS/MS, and therefore to allow dose-effect studies.

Ce travail s’intéresse au comportement en environnement spatial d’une nouvelle technologie de cellules solaires photovoltaïque : les tandems III-V//Si (2- et 3-jonctions), obtenues par collage direct. Ces cellules ont été exposé à des irradiations électrons et protons et testé dans deux type d’environnement : a) irradiance normale, 1 soleil, et température ambiante 300K, condition NIRT (orbites terrestres) et b) basse irradiance, 0,03 soleil, et basse température 120K, condition LILT (espace lointain). Dans une étape préliminaire une étude comparative a été mené sur 2 simulateurs solaires, équipés respectivement d’une lampe flash et de lampes LED, afin d’assurer la fiabilité et la reproductibilité des mesures de ces multi-jonctions. Pour le simulateur flash, une méthode de caractérisation pour tandems I-V sous 1 soleil qui s’affranchit de l’utilisation de cellules de référence isotype a été adoptée, en se basant sur des mesures d’EQE et des mesures du spectre du flash. Pour le simulateur LED, monté in-situ sur le faisceau d’irradiation, une optimisation du spectre a été effectuée afin de se rapprocher de la référence à basse irradiance, soit ~3% AM0. Cette étude comparative a également permis d’établir la validité de l’extrapolation par le calcul de mesures I-V sous 1 soleil vers les basses irradiances.Ensuite, la compatibilité de cette technologie tandem III-V//Si avec d’un part le cyclage thermique et d’autre part les irradiations a été démontrée, l’interface de collage maintient son intégrité mécanique et électrique face à ces contraintes. L’impact des irradiations sur les performances cellules a révélé certaines similitudes à 300 K et 120 K : - une décroissance marquée du courant de court-circuit (liée à la diminution de la longueur de diffusion) - une diminution plus faible de la tension de circuit-ouvert (défauts de type génération). Du fait de la connexion en séries des sous-cellules, la dégradation de la limitante Si (faible résistance intrinsèque aux irradiations) domine le comportement de la multi-jonction. Il a été démontré que l’ajout d’un nombre croissant de cellules sur le Si se traduit par une sensibilité accru aux irradiations; en effet, la configuration tandem restreint la bande d’absorption du Si au proche infrarouge, partie spectrale la plus affectée par la baisse de longueur de diffusion. L’utilisation d’un modèle basé sur l’IQE a permis de quantifier cette dégradation de longueur de diffusion dans le Si en tandem, ainsi que le coefficient de dommage. A la différence des électrons, les irradiation aux protons 1 MeV sont à l’origine d’une dégradation non-uniforme dans le Si ; par des mesures EQE couplées à de la simulation, nous avons corrélé cette dégradation non-homogène dans le Si avec la position du pic de Bragg correspondant.Pour l’étude basse température, une augmentation linéaire de l’efficacité a été observée jusqu’à ~150K ; et en deçà, des anomalies de caractéristiques I-V ont été détectées ; de type « S like shape » et « flat spot » ces défauts affectent le FF et donc le rendement. Documentées dans la littérature, ces effets sont caractéristiques des conditions LILT, et souvent liées à des modifications des interfaces métal/semi-conducteur. Bien qu’importante, la dégradation des performances électriques fin de vie LILT des III-V//Si s’avère être plus prédictible que celles des III-V/Ge LILT (dispersion statistique). Nous avons également démontré qu’un passage à 300 K, après irradiation à 120 K, entraine une guérison marquée du courant de court-circuit; ceci souligne l’importance des caractérisations in-situ pour quantifier le vieillissement cellule en conditions de fonctionnement. L’approche DDD « Displacement Damage Dose » a été appliqué pour les électrons et protons 1 MeV afin de comparer le taux de dégradation induit. Cette approche permet de prédire la dégradation de ces cellules quel que soit la fluence, les particules et l’énergie, pour une mission spatiale à 300 K This work focuses on the behavior in space environment of a new photovoltaic solar cell technology: the III-V//Si tandems (2- and 3-junction), obtained by direct bonding. These cells have been exposed to electron and proton irradiations and tested in two types of environment: a) normal irradiance, 1 sun, and 300K room temperature, NIRT condition (Earth orbits) and b) low irradiance, 0.03 sun, and 120K low temperature, LILT condition (deep space). In a preliminary stage, a comparative study was conducted on 2 solar simulators, respectively equipped with a flash lamp and LED lamps, in order to ensure the reliability and reproducibility of the measurements of these multi-junctions. For the flash simulator, a tandems characterization method for I-V under 1 sun that dispense the use of isotype reference cells has been adopted, based on EQE and flash spectrum measurements. For the LED simulator, mounted in-situ on the irradiation beam, a spectrum optimization was performed in order to approach the low irradiance reference, i.e. ~3% AM0. This comparative study also allowed to establish the validity of the extrapolation by calculating I-V measurements under 1 sun towards low irradiances.Then, the compatibility of this tandem III-V//Si technology with thermal cycling on the one hand and irradiances on the other hand has been demonstrated. The bonding interface maintains its mechanical and electrical integrity face to these constraints. The impact of the irradiations on the cell performances has revealed certain similarities at 300 K and 120 K: - a marked decrease in the short-circuit current (linked to the decrease in the diffusion length) - a smaller decrease in the open-circuit voltage (generation type defects). Due to the series connection of the sub-cells, the degradation of the limiting Si (low intrinsic resistance to irradiation) dominates the behavior of the multi-junction. It has been shown that the addition of an increasing number of cells on the Si results in an increased sensitivity to irradiation; indeed, the tandem configuration restricts the absorption band of the Si to the near infrared, the spectral part most affected by the decrease in diffusion length. The use of a model based on the IQE allowed the qualification of this diffusion length degradation of the Si in tandem, as well as the damage coefficient. Unlike electrons, 1 MeV proton irradiations are at the origin of a non-homogeneous degradation in Si; by EQE measurements coupled with simulation, we have correlated this non-homogeneous degradation in Si with the position of the corresponding Bragg peak.For the low-temperature study, a linear increase in efficiency was observed up to ~150K; and below this, anomalies of I V characteristics were detected; of "S-like shape" and "flat spot" type, these defects affect the FF and thus the efficiency. Reported in the literature, these effects are characteristic of LILT conditions, and are often related to changes in the metal/semiconductor interfaces. Although significant, the LILT end-of-life electrical performance degradation of III-V//Si has been shown to be more predictable than that of III-V/Ge LILT (statistical dispersion). We have also shown that a 300 K annealing after irradiation at 120 K leads to a marked healing of the short-circuit current; this underlines the importance of in-situ characterizations to quantify cell aging under operating conditions. The Displacement Damage Dose (DDD) approach was applied for 1 MeV electrons and protons in order to compare the rate of induced degradation. This approach allows to predict the degradation of these cells whatever the fluence, particles and energy, for a space mission at 300 K

En recherche clinique ou environnementale, les biomarqueurs protéiques présentent un intérêt croissant. Bien que les immuno-dosages restent les méthodes de référence pour leur quantification, les récentes avancées en spectrométrie de masse (MS) font de cette technique une alternative crédible à l'ELISA. Ce travail apporte quelques éléments méthodologiques pour repousser certaines limitations de la MS. D'abord, deux dosages ont été proposés. Celui réalisé chez G. fossarum représente le premier exemple de dosage de la Vitellogénine chez un invertébré par LC-MS/MS. L'un des défis de la méthode présentée est de doser spécifiquement une protéine dans un organisme dont le génome est majoritairement inconnu. Le second dosage concerne les peptides contenant une méthionine. Nous avons développé un protocole d'oxydation des méthionines afin de s'affranchir du biais lié à leur oxydation partielle. Cette méthode a ensuite été appliquée à une protéine impliquée dans la maladie d'Alzheimer (l'Apolipoprotéine E4) dans une cohorte de 673 plasmas. Ce dosage est à ce jour l'une des plus grandes études réalisées par LC-MS/MS et montre toute la robustesse de cette approche. Enfin, l'influence de la phase mobile sur la sensibilité des dosages de peptides a été étudiée : d'abord en phase inverse, où le méthanol est une bonne alternative à l'acétonitrile ; ensuite en HILIC, où les difficultés liées à l'étude d'ions multichargés en milieu majoritairement organique ont été abordées. Les problèmes liés à la capacité de charge des colonnes ont également été soulevés. La chromatographie HILIC reste prometteuse pour la quantification de peptides puisqu'un facteur dix en sensibilité peut être apporté.

Disease-specific compounds (biomarkers) are analyzed in clinical laboratories to assist with diagnosing diseases. This thesis describes development and validation of liquid chromatography tandem mass spectrometry (LC-MS/MS) based tests for diagnosing a diverse group of endocrine and metabolic diseases. The analytical methods used on-line and off-line sample extraction and analytical derivatization as means of enhancing the analytical sensitivity, specificity and clinical utility. All developed methods were extensively validated and reference intervals for the biomarker concentrations were established in blood samples of healthy adults and children. Advantages of the LC-MS/MS as an analytical technique include possibility of simultaneous measurement of multiple analytes and ability of confirming their identity. In this thesis we proposed and evaluated approaches for the assessment of the specificity of analysis in the methods that use tandem mass spectrometry detection. To enhance throughput of the LC-MS/MS tests for the biomarkers that have endogenous or exogenous isomers an approach was developed for quantitation of isomers from unresolved chromatographic peaks. Using methods developed in this thesis we performed a study of the steroidogenesis in ovarian follicles of healthy women and women with polycystic ovary syndrome (PCOS). Obtained data on the steroid concentrations and associations between the steroid metabolites in the pathway would be helpful for better understanding of the ovarian pathophysiology. Potential biomarkers of PCOS were identified in the thesis; further studies will be necessary to confirm their clinical utility.

Cette étude a donc d’abord développé un système miniaturisé du couplage en ligne de l’extraction sur phase solide à la microchomatographie en phase liquide associée à la détection par spectrométrie de masse pour l’analyse des pesticides polaires (ammélide, amméline, DAA, DIA, DEA et atrazine) contenus dans des eaux. L’analyse a été permise à partir d’une phase stationnaire en carbone graphitisé poreux (PGC). Ce couplage a été mis en œuvre pour la préconcentration et l'analyse des microcystines. Une culture de masse de cyanobactéries a permis de récupérer et d'isoler plusieurs microcystines. Un support d'immunoextraction basé sur les anticorps anti-microcystine LR a été développé. Ce système est utilisé pour l'étude du comportement des microcystines sur ce support. Les informations sur les types d’interactions mises en jeu lors de l’immunopréconcentration des microcystines ont été étudiées. L'application d'échantillons réels a été réalisée pour l’extraction d’autres microcystines.

Les nucléotides, terme regroupant les nucléosides monophosphate, diphosphate et triphosphate, sont impliqués dans de nombreux processus cellulaires. Ils représentent les éléments constitutifs des acides nucléiques, fournissent de l'énergie à des réactions métaboliques, jouent le rôle de transporteurs et seconds messagers. L'exploration des pools nucléotidiques apparaît indispensable afin de connaître leur rôle précis dans des situations physiologiques ou pathologiques. Nous avons développé une méthode de dosage des nucléotides endogènes (formes mono-, di- et triphosphate) par extraction en ligne sur colonne WAX couplée à la LC-MS/MS. La séparation analytique est réalisée sur colonne Hypercarb, sans agent de paire d'ions dans la phase mobile. Grâce à l'utilisation d'un triple quadripôle et une ionisation en mode positif, les nucléotides endogènes sont identifiés sans équivoque, y compris ceux possédant la même masse molaire. L'extraction et la séparation des nucléotides sont réalisées en 20 min. L'ensemble de la méthode, en comptant la ré- équilibration des colonnes, dure 37 min. La méthode de dosage a été validée pour les formes mono- et triphosphate et est applicable à des séries d'une vingtaine d'échantillons biologiques. D'autre part, dans une étude pré-analytique basée sur les plans d'expériences, nous avons comparé les conditions de préparation d'échantillons en vue du dosage de nucléotides intracellulaires dans 4 lignées cellulaires : 2 adhérentes (Messa et NCI-H292) et 2 en suspension (RL et L1210). Nous avons montré que les conditions pré-analytiques optimales dépendent de la lignée cellulaire soulignant ainsi l'importance de cette phase dans l'analyse des nucléotides. Enfin, l'expérience et les connaissances acquises lors du développement de la méthode de dosage des nucléotides ainsi que la large palette de molécules analysables avec cette méthode (nucléosides, nucléotides sucrés, autres métabolites), nous ont permis de développer des collaborations dans le domaine de la cancérologie avec différentes équipes de recherche. Par exemple, nous avons étudié l'implication des pools nucléotidiques dans le stress réplicatif induit par le stress oxydant et dans la reprogrammation cellulaire observée dans les cellules cancéreuses. Ainsi, les informations apportées par notre approche analytique, complémentaires des autres approches utilisées, ont montré l'implication des pools nucléotidiques dans la tumorigénèse. En conclusion, ce travail a permis de développer une technique analytique et de mettre en place une méthode de travail pour le dosage des nucléotides endogènes dans différents milieux biologiques Nucleotides, term including nucleoside mono-, di- and triphosphates, are endogenous compounds playing various roles in biology. They are components of nucleic acids, provide energy to metabolic reactions and act as carriers or second messenger. The study of endogenous nucleotides has become of great interest in physiological and pathological conditions. We developed a method for the quantification of endogenous nucleotides, using an on-line extraction on a WAX column coupled with LC-MS/MS. Analytical separation is performed on a Hypercarb column, without ion pairing agent in the mobile phase. The use of a triple quadrupole mass spectrometer following positive mode ionization allows the unambiguous identification of nucleotides presenting the same mass. Extraction and separation of nucleotides are achieved within 20 min and the method including re-equilibration of the two columns within 37 min. The method was validated for the quantification of nucleoside mono- and triphosphates, and could be applied to series of more than twenty biological samples. Secondly, in a study based on design of experiments, pre-analytical parameters influencing results of intracellular nucleotides were compared in four cell lines. We demonstrated that optimal pre-analytical parameters depend on cell lines. This clearly highlights the importance of pre- analytical conditions for the quantification of intracellular nucleotides to be as representative as possible of the real levels in cells. Then, thanks to experience acquired during the development and the validation of the analytical method, scientific collaborations have been established with several cancer research teams. For example, implication of nucleotide metabolism in replicative stress induced by oxidative stress or in the metabolic reprogramming in cancer cells was studied. Results obtained by our analytical approach were complementary to those obtained by other techniques. To conclude, our work consisted on the study of the entire workflow for the analysis of endogenous nucleotides in various biological samples

The synthesis of decalin systems possessing a quaternary carbon at C-9 was achieved via the tandem oxy-Cope/ene/Claisen rearrangement. Depending on the substitution pattern of the allyl ether 1, different reaction pathways may be followed to generate the decalin cores. In the case of 1,2-divinylchyclohexanol having a 1,2-disubstituted allyl ether, the tandem reaction afforded the decalin system in good yield. However, trisubstituted allyl ethers produced unique decalin products via a radical 1,3-shift. In addition, microwave irradiation of 1,2-divinylcyclohexanol propargyl ethers generated tetracyclic acetals via the tandem oxy-Cope/ene/Claisen/5-exo dig cyclization.* The scope and limitations of the hydroxy-directed Diels-Alder (HDDA) reaction were studied on 5-membered semicyclic dienes possessing secondary or tertiary allylic alcohols. A temporary magnesium tether provided by MgBr 2·OEt2 or vinylmagnesium bromide was utilized to regio- and stereoselectively control the Diels-Alder reaction. This methodology was applied towards the synthesis of the hydrindane moiety of isovelleral 2 in order to construct its analogues.* As well, the HDDA reaction was applied towards the total synthesis of havellockate 3. This thesis is divided into six parts, all centered around the development of dynamic (i.e., reversibly interacting) systems of molecules and their applications in dynamic combinatorial chemistry (DCC) and organic synthesis. Part one offers a general introduction, as well as a more detailed description of DCC, being the central concept of this thesis. Part two explores the potential of the nitroaldol reaction as a tool for constructing dynamic systems, employing benzaldehyde derivatives and nitroalkanes. This reaction is then applied in part three where a dynamic nitroaldol system is resolved by lipase-catalyzed transacylation, selecting two out of 16 components. In part four, reaction and crystallization driven DCC protocols are developed and demonstrated. The discovery of unexpected crystalline properties of certain pyridine β-nitroalcohols is used to resolve a dynamic system and further expanded into asynthetic procedure. Furthermore, a previously unexplored tandem nitroaldol-iminolactone rearrangement reaction between 2-cyanobenzaldehyde and primarynitroalkanes is used for the resolution of dynamic systems. It is also coupled with diastereoselective crystallization to demonstrate the possibility to combine several selection processes. The mechanism of this reaction is investigated and a synthetic protocol is developed for asymmetric synthesis of 3-substituted isoindolinones. Part five continues the exploration of tandem reactions by combining dynamic hemithioacetal or cyanohydrin formation with intramolecular cyclization to synthesize a wide range of 3-functionalized phthalides. Finally, part six deals with the construction of a laboratory experiment to facilitate the introduction of DCC in undergraduate chemistry education. De nos jours, l’augmentation du volume des données de séquençage est bien plus forte que celle de notre capacité à analyser ces données. En lien avec ce déluge de données et le besoin urgent de nouveaux outils bioinformatiques pour les analyser, notre travail consiste à développer de nouveaux algorithmes pour mieux comprendre les relations entre séquence, structure, et fonction des protéines. Les protéines contiennent de larges portions de séquences périodiques, qui forment des motifs d’acides aminés répétés les uns à la suite des autres que l’on appelle des répétitions en tandem. Elles se retrouvent dans 14% des protéines. De nombreuses études ont montré leur importance fonctionnelle ainsi que leur implication dans de nombreuses maladies humaines, notamment le cancer. Ici, nous montrons l’importance d’adopter une approche incluant plusieurs outils de détection de répétition en tandem afin de s’assurer d’obtenir le jeu de données le plus complet. Nous avons ainsi réalisé un pipeline approprié, et développé deux outils spécifiques : un filtre, pour gagner en rapidité, et un score, pour sélectionner les répétitions les plus pertinentes dans les régions structurées des protéines. Enfin, nous avons utilisé ce pipeline sur une sélection de 94 protéomes. In line with this data deluge and urgent needs in new bioinformatics tools our work deals with the development of new algorithms to better understand the sequence-structure-function relationship. Proteins contain a large portion of periodic sequences representing arrays of repeats that are directly adjacent to each other, so called tandem repeats (TRs). TRs occur at least in 14% of all proteins. Highly divergent, they range from a single amino acid repetition to domains of 100 or more repeated residues. Numerous studies demonstrated the fundamental functional importance of such TRs and their involvement in human diseases, especially cancers. Here we show the importance of integrating several TR detectors to get the most complete set of TRs in proteomes. We designed an appropriate pipeline and developed a filter to speed the process as well as a new scoring module to select relevant structured TRs. In addition, we undertook a large scale analysis of TRs in 94 proteomes. This large scale analysis allowed us to update previous census of TR showing that TRs occurs in 64% of all proteins and leads to a better understanding of TR in terms of their characteristics, composition and implication in human disease

La conversion photovoltaïque (PV) de l’énergie solaire repose sur la capacité qu’ont certains matériaux à convertir l’énergie des photons en courant électrique. Le développement des systèmes de conversion PV ces trente dernières années a permis des améliorations considérables en terme de coût et de performances dans le domaine des énergies renouvelables.Une cellule multi-jonctions (MJ), à base de matériaux semi-conducteurs III-V, est un empilement de sous-cellules aux gaps décroissants qui permet notamment une plus large utilisation du spectre solaire. Soumettre ces cellules PV à un flux solaire concentré permet d’augmenter significativement la puissance électrique créée par celles-ci, et ainsi d’abaisser substantiellement le coût de l’électricité produite.Le record du monde est actuellement détenu par le partenariat Soitec / Fraunhofer ISE avec un rendement de 46,0 % mesuré sur une cellule quadruple-jonctions en GaInP/GaAs//InGaAsP/InGaAs pour un taux de concentration de 508 X (où 1 X =1 soleil = 1 kW/m²).L’objectif du travail réalisé dans le cadre de cette thèse est de proposer une alternative aux cellules existantes plus simple à mettre en œuvre avec des cellules MJ monolithiques accordées sur substrat de GaSb pour des concentrations solaire de 1 000, soit une irradiance directe de 1 MW/m². Ce type de cellules, du fait de la très bonne complémentarité des gaps des matériaux et ses alignements de bandes favorables, constitue une alternative crédible et originale aux cellules existantes pour une utilisation sous flux solaire fortement concentré.Afin de mieux comprendre la cellule multijonctions III-Sb optimale, les travaux réalisés ont porté sur la fabrication et la caractérisation des trois sous-cellules fabriquées indépendamment. Ces trois échantillons épitaxiés sont l’Al0,9Ga0,1As0,07Sb0,93 (cellule Top), l’Al0,35Ga0,65As0,03Sb0,97 (cellule Middle) et le GaSb (cellule Bottom) ayant comme gaps respectifs 1,6 eV, 1,22 eV et 0,726 eV à 300 K.Le travail présenté dans cette thèse porte sur :- La réalisation et la mise au point de toutes les étapes technologiques nécessaires à la fabrication des cellules (dépôts métalliques, gravure humide et sèche par plasma …).- La caractérisation des métallisations par structure TLM (Transmission Line Method) dont le meilleur résultat obtenu concerne une métallisation tri-couche Cr/Pd/Au (30/30/30 nm) sur substrat GaSb type P.- La caractérisation sous obscurité courant-tension des paramètres électriques des cellules PV à température ambiante et en fonction de la température.- La caractérisation thermique par mesure de la conductivité thermique des matériaux et une cartographie de température de surface en fonction du flux solaire concentré en conditions réelles.- La caractérisation électro-optique par réponse spectrale, à partir de laquelle nous avons calculé le rendement quantique externe qui représente le rapport entre la quantité d’électrons créés et la quantité de photons incidente.- La caractérisation sous illumination à 1 soleil (1 000 W/m²) sous simulateur solaire et en conditions solaire dont nous avons comparé les paramètres électriques.- La caractérisation des cellules sous flux solaire (fortement) concentré au laboratoire PROMES. Les meilleurs rendements obtenus pour les cellules PV Bottom, Middle et Top respectifs de 4,6 % à 40 X (proche de l’état de l’art), 8,2 % à 96 X et 5,4 % à 185 X (première mondiale pour ces matériaux quaternaires).Ce travail a été cofinancé par le Ministère de l’Education et de la Recherche (Allocation ED) et le Labex SOLSTICE.Photovoltaic (PV) solar energy consists on the ability of certain materials to convert the photon energy into electric current. Exposing these PV cells to a concentrated solar flux can significantly increase the electrical power generated, and therefore substantially lower the cost of electricity yielded.The world record is currently held by the partnership Soitec / Fraunhofer ISE with an efficiency of 46.0 % measured on a four-junction cell GaInP/GaAs//InGaAsP/InGaAs for a concentration ratio of 508 X (where 1 X = 1 sun = 1 kW/m²).The objective of the work in this thesis is to propose an alternative to existing cells, easier to implement with monolithic MJ cells grown on a GaSb substrate for solar concentrations of 1 000, which corresponds to a direct irradiance of 1 MW/m². This type of cell, due to the good complementary of the material gaps and its favorable band alignments, is a realistic and original alternative to existing cells for use under highly concentrated solar flux.To better understand the optimal multijunction III-Sb cell, the work carried out consisted on the manufacturing and characterization of the three sub-cells independently.These three epitaxial samples are Al0,9Ga0,1As0,07Sb0,93 (Top cell), the Al0,35Ga0,65As0,03Sb0,97 (Middle cell) and GaSb (Bottom cell) having as respective gaps 1.6 eV, 1.22 eV and 0.726 eV at 300 K.The work presented in this thesis is:- The establishment of all the technological steps required to manufacture the cells (metal deposition, wet and dry plasma etching ...).- The characterization of metallization by TLM structure (Transmission Line Method) with the best result being a three-layer metallization Cr/Pd/Au (30/30/30 nm) on a GaSb P-type substrate.- The characterization under dark of current-voltage electrical parameters of PV cells at room temperature and in function of the temperature.- The thermal characterization by measuring the thermal conductivity of the materials and a surface temperature mapping in function of the concentrated solar flux in realistic conditions.- The electro-optical characterization by spectral response, from which we calculated the external quantum efficiency which is the ratio between the amount of electrons created and the amount of incident photons.- The characterization under 1 sun illumination (1 000 W/m²) in a solar simulator and in realistic conditions of which we compared the electrical parameters.- The characterization of solar cells under (highly) concentrated solar flux in the PROMES laboratory.The best efficiencies for Bottom, Middle and Top PV cells respectively are 4.6 % for 40 X (close to the state of the art), 8.2 % for 96 X and 5.4 % for 185 X (world first for these quaternary materials).This work was cofounded by the Ministry of Education and Research (ED Research grant) and Labex SOLSTICE

A novel tandem time-of-flight (TOF) mass spectrometer has been developed for studying the photo-induced dissociation of large molecules and elemental clusters. It consists of a linear first stage TOF analyser for primary mass separation and precursor ion selection, and a second orthogonal reflecting field TOF analyser for product ion analysis. The instrument is equipped with a large volume throughput molecular beam source chamber allowing the production of jet-cooled molecules and molecular clusters, as well as elemental clusters, using either a pulsed laser vaporisation source (LVS) or a pulsed arc cluster ion source (PACIS). A second differentially pumped chamber can be used with effusive sources, or for infrared laser desorption of large molecules, followed by laser ionisation. These primary ions can then be irradiated with a second, high energy laser to induce photodissociation. Detailed information about the fragmentation mechanisms can be deduced from the product ion mass spectra. A theoretical overview of the technique of tandem time-of-flight mass spectrometry is presented, together with a detailed description of the experimental procedures and equipment used. In order to assist with the design and optimisation of the instrument a "virtual" mass spectrometer was drawn to scale using the SIMION software program, in order to simulate ion trajectories for differing voltage, geometry's and dimensions of the ion optics. An ion gate was designed and manufactured to provide primary mass selection following the first time-of-flight mass analyser. The device consisted of four layers of interleaved wires; primary ions could be selectively transmitted by application of a fast rising high voltage pulse to the middle set of wires. The mass gate was measured to have a mass resolving power, m/Dm = 30.

This thesis presents a unification of kinematic and force-based methods for the design and analysis of planetary gear trains along with a discussion of potential applications in tandem biking. Specifically, this thesis will provide a simple solution technique for the general case of a two-degree of freedom (2DOF) planetary gear train along with new graphical design aids. It will also address the use of epicyclic gear trains as a power coupling in a tandem bike. In the current literature, planetary gear trains are given a clear treatment with regard to the pure kinematics of the system, but little or no literature exists that includes the torques present in the system. By treating both the kinematics and torque balance of the most general case, this thesis attempts to fill a void in the current literature. After developing the solution to the general two-degree of freedom case using the Willis formula, a force analysis will be performed using the conservation of energy principle assuming zero losses. Once the total solution is known, nomographs will be presented as a simple design tool. These graphical aids enable the designer to simultaneously approximate both speeds and torques for the mechanism. After fully developing a satisfactory solution technique and design tools, these will be applied to the problem of coupling the power provided by the riders of a tandem bicycle. Master of Science

Les séquences répétées en tandem sont constituées de motifs adjacents. Elles constituent une classe de séquences génétiques dont font partie microsatellites et minisatellites. Dans cette thèse, nous traitons le problème de la comparaison de séquences répétées en tandem sous un modèle évolutif particulier. Plus précisément, nous nous intéressons au problème de leur alignement dans lequel, en plus des trois opérations classiques, mutation, insertion et délétion, nous considérons l'amplification en tandem et la contraction en tandem. L'amplification copie un facteur de la séquence, c'est-à-dire un ou plusieurs caractère(s), et met le ou les exemplaire(s) du facteur copié à côté du facteur original, la contraction est l'événement inverse. L'amplification (resp. la contraction) est dite « n-aire d'ordre m », si elle copie (resp. retire) m motif(s) n fois. Nous proposons une méthode donnant un score d'alignement, qui est une métrique, entre deux séquences répétées en tandem, sous un modèle comprenant les cinq opérations précédemment citées où l'amplification et la contraction sont unaires d'ordre 1. Le problème est difficile car les opérations ne sont pas commutatives. Notre solution fait appel à de l'algorithmique de graphe. Nous avons réalisé un programme nommé MS_Align qui implémente cette méthode. Il s'agit du premier programme capable d'aligner des cartes de minisatellites. À l'aide de ce programme, nous avons étudié des données biologiques provenant du minisatellite humain MSY1. Comme nous le montrons, notre modèle évolutif s'applique bien à ce type de séquences d'ADN. Nous avons construit à partir de nos résultats des arbres phylogénétiques semblables à ceux obtenus grâce à d'autres marqueurs du chromosome Y indépendants de MSY1, nos arbres offrent une meilleure résolution. Une partie de cette thèse est consacrée au problème général où nous relaxons les contraintes sur les amplifications et contractions.

Une répétition en tandem avec évolution consiste en une suite de copies plus ou moins contigue͏̈s où chaque copie est fortement similaire à celle qui la précède et à celle qui lui succède. Il n'existe pas de notion de modèle comme pour les répétitions en tandem "classiques" et la première et la dernière copie peuvent être complètement différentes. Ce "nouveau" type de répétition a été mis en évidence durant l'assemblage du génome humain. Dans ce manuscrit, je commence par définir, de façon formelle, les répétitions en tandem avec évolution à partir des observations effectuées dans les séquences biologiques. Après avoir effectué des tests sur des logiciels de recherche de répétitions, je conclus qu'il n'existe pas de logiciel capable de les détecter efficacement. J'ai alors conçu des algorithmes capables de détecter ce type de répétitions : un premier algorithme, quadratique en la longueur de la séquence, puis un algorithme linéaire. Ces algorithmes ont été implantés afin de mettre à disposition des biologistes des outils qui détectent efficacement ces répétitions dans de très grandes séquences génomiques, telles les chromosomes humains. Plusieurs méthodes de parallélisation sont également présentées. Les résultats des tests réalisés sur des chromosomes montrent la présence de ces répétitions dans plusieurs génomes An evolutive tandem repeat consists in a series of almost contiguous copies, such that each copy is strongly similar to its predecessor and its successor. There is no model and the first and the last copy can be completely different. It differs from a ``classical'' tandem repeat that considers a model and a radius around this model. This ``new'' type of repeat has been pointed out during the assembly of the human genome. In this manuscript, I first define, in a formal way, the evolutive tandem repeats from observations achieved in biological sequences. After having carried out tests using well known existing softwares, I conclude that no one was able to detect it correctly and that they are not part of the sequences that have been submitted to databanks. I therefore designed algorithms that are able to locate this specific type of repeats: a first algorithm, quadratric in the length of the sequence, then a linear one. These algorithms have been implemented in order to provide biologists with tools that detect efficiently these repeats in real large sequences, such as human chromosomes. Several parallelisation methods are presented too. The results of the carried out tests on chromosomes show the existence of such repeats in genomes

La matière organique particulaire (POM) des aérosols atmosphériques présente une composition chimique particulièrement complexe. Au mieux, seulement 20 % en masse des particules parviennent à être identifiés par les techniques de spéciation moléculaire. D'autre part, les méthodes d'analyse globale conduisent à une simplification importante de la matrice et par conséquent à une perte d'informations. Les travaux menés au cours de cette thèse s'inscrivent dans un objectif de développement de méthodes d'analyse chimique innovantes et complémentaires, permettant une meilleure connaissance et une meilleure compréhension de la fraction organique des aérosols atmosphériques. Trois méthodes d'analyse fonctionnelle par spectrométrie de masse tandem (MS/MS) ont été mises au point, pour la détermination quantitative des fonctions chimiques carboxyliques (RCOOH), carbonyles (R-CO-R') et nitros (R-NO2). Dans les trois cas, la précision des mesures a été estimée par le calcul de la variabilité obtenue pour l'analyse de 25 mélanges références constitués de 16 à 31 composés selon la fonction étudiée, dans des proportions différentes d'un mélange à un autre. Le résultat de cette étude statistique montre une erreur analytique inférieure à 20 % pour chacune des trois fonctions. Les limites de détection (de l'ordre de 0,005 mM) et la gamme de linéarité (0,01 - 0,5 mM) ont permis d'appliquer les méthodes développées sur des échantillons d'aérosols organiques secondaires produits en chambre de simulation, d'aérosols à l'émission, et sur des aérosols atmosphériques. Les résultats obtenus sont cohérents avec la littérature et notre connaissance actuelle des aérosols. Plusieurs pistes de réflexion sur l'utilisation des méthodes développées sont enfin présentées et montrent l'intérêt de cette nouvelle approche en termes d'études des sources et de vieillissement de l'aérosol.

This thesis explores the development of the tandem oxy-Cope/ene rearrangement and its application toward the synthesis of natural antibiotic tetrodecamycin. These studies have been carried out to better understand the chemical synthesis of decalin natural products, and provide a springboard from which further studies can be launched. The first part of the study focueses on the tandem oxy-Cope/ene rearrangement and the unravelling of the intricate aspects governing the selectivity, stereochemistry, and transition state conformers at play. As a result of this study, we have developed a general methodology for rapidly constructing decalins bearing a ring junction alcohol via cascade oxy-Cope/ene rearrangement of 1,2-divinyl cyclohexanols. The second part of this study involved the application of this methodology to the decalin skeleton present in tetrodecamycin. Described in this section is the development of the synthetic chemistry en route to the core of the natural product, along with the serendipitous discovery of a tandem oxy-Cope/Claisen/ene/Claisen rearrangement. Finally, the decalin core provided by the tandem rearrangement was fully functionalised to resemble that of the natural product (including all 6 contiguous stereocentres), and the synthetic efforts toward the completion of tetrodecamycin are described.

Although molybdenum blue solutions have been known for more than twocenturies, an understanding of their chemical nature is only beginning to emerge.This dissertation aimed at elucidating the structural nature of the polydisperse,nanoscopic components in the solution phases and the solid states of partiallyreduced polyoxomolybdate (Mo-POM). The study offered at least fourcontributions to the area: (1) a rational protocol for the molecular recognition ofMo-POM with de novo organic hosts. (2) demonstration of kinetic precipitation ofa dynamic mixture of polyoxomolybdates and application of the technique to thestudy of the dynamic mixture by TEM (3) characterization of the Mo-POMnanostructures by an unusual combination of complementary analyticaltechniques. (4) a general approach for the synthesis of crown-ethers-containingtripodal molecules.The molecular recognition of Mo-POM with designer tripodal hexaminetris-crown ethers opened a window to the solution phase structures of Mo-POMnanoscopic components. Studies with a series of structurally analogous hostsprobed the relationship between the structure of the molecular host and theformation of nanostructures.An unusual combination of complementary analytical protocols: flow fieldflowfractionation, electron microscopy (transmission and scanning), andinductively coupled plasma – emission spectroscopy, was used to monitor thesolution-phase evolution of Mo-POM nanostructures. The crystallization – drivenformation of keplerate Mo-POM and solution-phase evolution of structurallyrelated nanoscopic species were apparent in the self-assembling process ofpartially reduced Mo-POM.
