Visual Odometry is the process that estimates camera position and orientation based solely on images and in features (projections of visual landmarks present in the scene) extraced from them. With the increasing advance of Computer Vision algorithms and computer processing power, the subarea known as Structure from Motion (SFM) started to supply mathematical tools composing localization systems for robotics and Augmented Reality applications, in contrast with its initial purpose of being used in inherently offline solutions aiming 3D reconstruction and image based modelling. In that way, this work proposes a pipeline to obtain relative position featuring a previously calibrated camera as positional sensor and based entirely on models and algorithms from SFM. Techniques usually applied in camera localization systems such as Kalman filters and particle filters are not used, making unnecessary additional information like probabilistic models for camera state transition. Experiments assessing both 3D reconstruction quality and camera position estimated by the system were performed, in which image sequences captured in reallistic scenarios were processed and compared to localization data gathered from a mobile robotic platform

Com o avan?o nos algoritmos e no poder de processamento dos computadores, a sub?rea de Vis?o Computacional denominada de Structure from Motion (SFM) passou a fornecer ferramentas que comp?em sistemas de localiza??o visando aplica??es como rob?tica e Realidade Aumentada, em contraste com o seu prop?sito inicial de ser usada em aplica??es predominantemente offline como reconstru??o 3D e modelagem baseada em imagens. Sendo assim, este trabalho prop?e um pipeline de obten??o de posi??o relativa que tem como caracter?sticas fazer uso de uma ?nica c?mera calibrada como sensor posicional e ser baseado interamente nos modelos e algoritmos de SFM. Повний текст джерелаThis thesis takes place in the context of the European project WYSIWYD (What You Say is What You Did). The goal of this project is to provide transparency in Human-robot interactions, including by mean of language. The deployment of companion and service robots requires that humans and robots can understand each other and communicate. Humans have developed an advanced coding of their behavior that provides the basis of transparency of most of their actions and their communication. Until now, the robots do not share this code of behavior and are not able to explain their own actions to humans. We know that in spoken language, there is a direct mapping between languages and meaning allowing a listener to focus attention on a specific aspect of an event. This is particularly true in language production. Moreover, visual perception allows the extraction of the aspects of "who did what to whom" in the understanding of social events. However, in the context of human interaction, other important aspects cannot be determined only from the visual image. The exchange of an object can be interpreted from the perspective of the giver or taker. This introduces the notion of construal that is how a person interprets the world and perceive a particular situation. The events are related in time, but there are causal and intentional connexion that cannot be seen only from a visual standpoint. An agent performs an action because he knows that this action satisfies the need for another person. This may not be directly visible in the visual scene. The language allows specifying this characteristic: "He gave you the book because you like it." The first point that we demonstrate in this work is how the language can be used to represent these construals. In response, we have developed a system in which a mental model represents an action event. This model is determined by the correspondence between two abstract vectors: the force vector exerted by the action and the result vector corresponding to the effect of the applied force. The application of an attentional process selects one of the two vectors, thus generating the construal of the event. The second point that we consider in this work is how the construction of narrative discourse can be learned with a narrative discourse model. This model is based on both existing neural networks of production and comprehension of sentences that we enrich with additional structures to represent a context of discourse. We present also how this model can be integrated into an overall cognitive system for understanding and generate new constructions of narrative discourse based on similar structure, but different arguments. For each of the works mentioned above, we show how these theoretical models are integrated into the development platform of the iCub humanoid robot. This thesis will explore two main mechanisms to enrich the meaning of events through language. The work is situated between computational neuroscience, with development of neural network models of comprehension and production of narrative discourse, and cognitive linguistics where to understand and explain the meaning according to joint attention is crucial

Повний текст джерелаTumor comprise complex niche of the immune and non-immune components. The complex interaction between the tumor cells with its environment turns into either eradication or the growth and metastasis of the tumors. We have previously demonstrated the role of TLS (tertiary lymphoid structures) in lung tumors, in protective anti-tumor responses. Despite of this, tumors do develop via exploiting the regulatory mechanisms, particularly includes, infiltration of the Tregs (regulatory T cells). The aim of thesis was to study the putative role of Tregs in regulating the immune responses in lung cancer. This study strongly demonstrates the presence of FoxP3+ Tregs in the TLS as well as non-TLS areas of the lung tumors. Tregs mainly exhibit central and effector memory phenotype expressing vast repertoire of the activation and immune checkpoint molecules. The gene expression and flow cytometry data showed that Tregs express the co-stimulatory and inhibitory markers which are known to be involved in the their activation and immune suppression. The high density of the Ti-Tregs either in TLS or in nonTLS areas is associated with the poor survival of the NSCLC patients. When combined with the density of TLS mature DC or B cells or CD8+ T cells, a group of patients with the low DC, B cells and CD8+ T cells but high Tregs densities, had the worst clinical outcome. This allowed, to identify the NSCLC patients with highest risk of death. Thus, it be concluded that the Tregs create the immunosuppressive environment in the lung tumors by acting in both TLS and nonTLS areas of the tumors and thus could be possible reason for the reduced survival of the lung cancer patients

T cells are activated by recognition of foreign peptides displayed on the surface of antigen presenting cells (APCs), an event that triggers assembly of a complex microscale structure at the T cell-APC interface known as the immunological synapse (IS). It remains unresolved whether the unique physical structure of the synapse itself impacts the functional response of T cells, independent of the quantity and quality of ligands encountered by the T cell. As a first step toward addressing this question, we fabricated multicomponent protein surfaces that surrogate the role of APCs and studied T cell responses as a function of synapse structure. To pattern multiple proteins on surfaces, we synthesized and characterized a new polymer, poly(o-ntrobenzyl methacrylate-r-methyl methacrylate-poly(ethylene glycol) methacrylate (PNMP), a photoresist that can be processed under mild aqueous conditions. Based on the pH- and temperature-sensitive solubility of UV-exposed PNMP random terpolymers in aqueous buffers, two-component protein patterning was achieved under conditions that avoid exposing proteins to conditions outside the narrow range of physiological pH, ionic strength, and temperature where their stability is greatest.

Using a photolithographic strategy we developed employing this novel PNMP photoresist polymer, we created multicomponent protein surfaces presenting micron-scale arrays of tethered T cell receptor (TCR) ligands (anti-CD3 'activation sites') surrounded by a field of tethered intercellular adhesion molecule-I (ICAM-1), as a model substrate on which T cells could be seeded to mimic T cell-APC interactions. CD4+ T cells seeded on these surfaces polarized and migrated; on contact with activation sites, T cells assembled an IS with a structure modulated by the physical pattern of ligand encountered. On surfaces patterned with focal spots of TCR ligand, T cells stably interacted with activation sites, proliferated, and secreted cytokines. In contrast, T cells interacting with activation sites patterned to preclude centralized clustering of TCR ligand failed to form stable contacts with activation sites, exhibited aberrant PKC-[Theta] clustering in a fraction of cells, and had significantly reduced production of interferon-[gamma]. These results suggest that focal clustering of TCR ligand characteristic of the 'mature' IS may be required under some conditions for full T cell activation.

Повний текст джерелаSpin crossover (SCO) compounds with a large hysteresis centered around room temperature (RT) are being constantly pursued although such behavior is very rare and most often noticed in coordination networks. In this context, a new molecular discrete compound, the complex [Fe(PM-PeA)2(NCSe)2], showing a large SCO hysteresis spanning RT has been synthesized in both singlecrystal and powder forms. Then, two research lines emerged. The first one concerns the multi-scales study of the structure-properties relationships, combining single-crystal and powder X-ray diffraction at variable temperature. A particular focus has been made on the microstructural scale, almostunexplored until now. For the first time, the coherent domain sizes and the micro-deformation rate has been quantified for a molecular discrete SCO compound. The second part of this work investigates in detail the pressure-induced SCO. The in situ single-crystal X-ray diffraction studies has been carried out to perform a complete characterization of the crystal structure under pressure. Thereafter, an accurate high-pressure X-ray diffraction measurement on the powder, with a synchrotron radiation,provides a fine track of the pressure-induced SCO and showed a piezo-hysteresis. Experiments coupling pressure and temperature brought crucial information pertaining to variation of bulk moduli withtemperature, the piezo-hysteresis width, the temperature dependence of the pressure transition and pressure-induced SCO abruptness, are provided in detail for the first time

Повний текст джерелаThe objective of this study was to evaluate the relationship between the structure (at physical and/or molecular level) of wheat bran and aleurone and their health effects. Keeping the composition of bran and aleurone constant, their structure was modified by mechanical and/or enzymatic treatments in order to disrupt their complex matrix structure and increase the bioaccessibility of their dietary fibre and phenolic compounds. The antioxidant capacity of the bran and aleurone fractions was evaluated by a standard in vitro test and by an in vitro gastric digestion model. The grinding disrupted the physical structure of bran and aleurone increasing their specific surface leading to a greater exposition of ferulic acid moiety (FA) which increased their antioxidant capacity. Enzymatic treatment performed on aleurone acted on its molecular organisation releasing its FA from arabinoxylans (AX). This destructuration improved the antioxidant capacity of aleurone even more than the one allowed by grinding. As aleurone is rich in highly fermentable AX, the effects of its better accessibility and depolymerisation were evaluated using an in vitro model mimicking the human colon. The physical destructuration of aleurone by grinding did not improve its fermentability, i.e. the colonic metabolism of phenolic compounds and the formation of short-chain fatty acids were similar compared to the native aleurone. Nevertheless, the enzymatic degradation of aleurone produced higher amount of soluble AX and bioavailable FA improving the metabolism of colonic FA in metabolites with anti-inflammatory properties. An in vivo mouse study with a high-fat diet was used to evaluate the potential of physically and molecularly destructured aleurone to counteract the metabolic disorders, such as obesity, oxidative stress and inflammation. The physical destructuration of aleurone did not have any positive effect on the metabolic disorders. The aleurone fraction, which presented the highest level of soluble AX and bioavailable FA, reduced the obesity (body weight gain, adiposity, and leptin secretion) and insulin resistance in mice. But no significant differences were observed in the oxidative stress and inflammation status of mice fed with any of the aleurone-based diets. In conclusion, we demonstrated that the aleurone health effects were clearly linked to the integrity of its structure. Its health effects have been increased by modification of its structure mainly at molecular level by degrading its cell wall to increase the bioaccessibility and bioavailability of nutritionally interesting compounds, such as AX and FA

Повний текст джерелаGuanine-rich DNA or RNA sequences can adopt non-canonical structures composed of four strands called guanines quadruplexes or G-quadruplexes. These structures are made by the formation and stacking of guanine quartets; they can be found in various region of the genome. G-quadruplexes motifs are frequently found during the selection of aptamers by the so-called SELEX method: a bias exists in the proportion of guanines in comparison with other nucleotides in a database gathering together known aptamers sequences. We did an in silico then in vitro systematic analysis of aptameric motifs described in the literature. We used a prediction algorithm currently developed in the laboratory to determine which sequences car form G-quadruplexes in silico. In order to check those predictions, a biophysical test allowing to quickly assay a lot of sequences was set up. Our results demonstrate that a lot of published aptamers are able to fold into G-quadruplexes. Another part of this work is related to the use of the SELEX with two different goals. First of all we did an in vitro selection against a G-quadruplexes ligand, in order to tell exactly which nucleic motifs can interact with this molecule. We then carried out SELEX experiments against several DNA or RNA biologically relevant G-quadruplexes (human telomeric motifs, minisatellite repetitions and sequences from UTR of mRNA), with the aim of getting specific probes for these conformations

Повний текст джерелаAntigenic lipids are presented to T-cells by CD1 proteins (CD1a to CD1d) expressed at the antigen presenting cell surface. In tuberculosis context, which the etiological agent is Mycobacterium tuberculosis, several lipid antigens have been described and it is important to note that most of them are presented by the isoforme CD1b. Thus, phosphatidyl-myo-inositol mannosides (PIM), and di-acylated sulfoglycolipids (Ac2SGL) are two glycolipid families of the mycobacterial cell-wall able to stimulate T-cell clones in vitro. The first line of my thesis work has consisted in the structural characterization of the antigenic species belonging to these two glycolipid families and, in an other way, to decipher intracellular processing mechanisms that manage PIM presentation in which CD1e, the fifth isoform of CD1 proteins, plays a key and non-presenting role not defined. Secondly, my research has consisted in looking for new antigens to expand the repertoire of immunogenic lipids involved in the anti-mycobacterial immunity. PIM are present in several glycosylated forms, PIM2 to PIM6 (possessing 2 to 6 mannosyl units), but also in different acylated forms, mono- to tetra-acylated. To be presented and recognized by T-cells, PIM6 have to be processed in PIM2 by a lysosomal mannosidase expressed in presenting cells. This degradation requires the CD1e protein. In this study, we have shown that CD1e has properties of lipid transfer protein, in vitro, specifically towards some PIM acyl-forms: di-acylated PIM. However, only tetra-acylated forms are antigenic and are able to stimulate T cells. Actually, we have demonstrated that a degradation of the lipidic part exist also in antigen presenting cells. All the data show that, in vivo, tetra-acylated PIM6 would be firstly processed in their lipidic part in order to generate non-conventional di-acylated forms that CD1e could be able to extract them from membranes. With this extraction, CD1e could allow PIM6 digestion by an a-mannosidase in an antigenic di-acylated PIM2 that CD1e could also transfer to the presenting isoform, CD1b. Sulfoglycolipids (SGL), contrary to PIM, are specific to M. Tuberculosis. Among SGL, only Ac2SGL have been described as immunogenic antigens able to, in vitro, activate CD8+ T-cells that have bactericide properties. Ac2SGL are constituted by a sulfated trehalose esterified by a classical fatty acid (palmitic acid or stearic acid) and a hydroxyphthioceranoic acid with a variable structure with up to 50 carbon atoms. In order to characterize which species can activate Ac2SGL specific T-cells, we improved the purification method and we analyzed with accuracy the structure of each molecular species constituting this glycolipid family by mass spectrometry and NMR spectroscopy. Thus, it has been highlighted new minority populations of Ac2SGL. The characterization of each population contribution in the anti-mycobacterial T response is an aim of this work. We also initiated the purification and the characterization of a new antigen produced by M. Tuberculosis. In these models, only the invidual species responses to environment are accounted for.Testing the importance of complementary processes showed a significant effect of colonization dynamics, but only a slight effect of biotic relationships. These processes led us to considerate a more complex assemblage structure than the one usually assumed by the biomonitoring methodologies.Therefore we implemented both neural networks models and fuzzy logic methodologies, in order to refine French ecoregions and to describe relationships between species traits and environment
