Problematiken för anskaffning av standardsystem inom en verksamhet kan bli omfattande,

särskilt ifall företaget tar förhastade beslut, och därmed blir anskaffningen en belastning

istället för tvärtom. Metodutvecklingen för anskaffningen av standardsystem är ett område

som inte är utforskat i stor utsträckning. Idag finns det en generell metod som används vid

anskaffning av standardsystem. Metoden kallas för SIV-metod (Standardsystem i

verksamheter) och har utvecklats av bl.a. Anders G. Nilsson i samband med ett projekt som

drevs inom institutet V i Stockholm i början av 1990-talet. SIV-metoden har inte utvecklats

färdigt och behöver därför prövas i fler praktiska fall. Syftet med denna uppsats är ta fram

metoden och pröva den i praktiken för att se hur pass användbar den är. Vi har arbetat med

företaget Koneo i Växjö som är ett IT - relaterat företag och har varit i behov av att

anskaffa ett standardsystem som i deras fall handlade om ett kundvårdssystem, CRM

(Customer Relationship Management).

I vår studie har vi tillämpat SIV-metoden för att se hur användbar den är i praktiken. Efter

genomgånget projekt kan vi konstatera att metoden är användbar i den mening att den ger

vägledning och tar vara på de grund delar som ingår i processen vid anskaffning av

standardsystem.


The problems that occur when obtaining or realising Standardsytem within an organisation,

can be exhaustive if the enterprise rushes to a desicion. Furthermore, this practical

application could be a burden instead of an opportunity. The method for obtaining realising

a standardsystem is an area that has not been studied to any greater extents. In the present,

the SIV-method (SIV: Standardsystem in business) is a general method used in the

relization of a standardsystem. The SIV-method has been developed by amongst others,

Anders G. Nilsson, in a project assigned by the V institute in Stockholm, Sweden. The

reason for this thesis is that the SIV-method is not considered complete and therefore we

will attempt to investigate this in a practical case. The purpose of this thesis is to test the

Siv-method in an organisations process for acquiring a standardsystem.

This research is co-operated in with the enterprise of Koneo. That is in need of a

standardsytem that concerns and operates the intergration of a Customer Relation

Managment system, CRM- system. After accomplishing our project we can establish that

SIV-method is useful, by means of that it takes into consideration main parts of the process

when obtaining a standardsystem.

The complexity of System-on-a-Chip (SoC) is continuing to increase due to the shrinking die size, increase in the number of sub-modules, power efficiency, performance, higher functionality and used protocols. This has an impact on the verification process related to the overall design process. For the verification process, there are commercial products that can be applied in order to verify and test certain Intellectual Properties (IP) but also platforms that lack these tools. This thesis focuses on the issue where a system has to be constructed that helps the verification and testing process of a data bus protocol used by the Device Under Testing (DUT). The study of the Serial Peripheral Interface (SPI) gives the examples of some issues that can be faced during applying this data bus protocol to any given system. Different kinds of testing and verifying methods are addressed in order to show what the tools can be when applying new DUT to a system or examining the data bus protocol it uses. The flow of a design process is studied by showing the iterations of a particular system that was to be created to meet the need that were introduced while examining the issues relating this subject. This flow can be said to start from ground level and end to the final iteration where the system could be created from. The functionality and structure of a Multipurpose Verification System that was created during this thesis are explained. The proofing process of this system is showed by examining the simulation and synthesis reports. The outcomes and future development ideas are discussed as well. This thesis showed that the study in hand has benefits to Nokia as the applying company and the system could be added to the company tool library after modifying it to be used as a stand-alone IP
Järjestelmäpiirien (SOC) kompleksisuus on jatkuvassa kasvussa johtuen johdinpiirien pienenemisestä, alijärjestelmien määrän kasvusta, vaatimuksista tehonkulutuksessa, suorituskyvyn kasvusta, toiminnallisuuden kasvusta ja käytettävistä protokollista. Näillä attribuuteilla on vaikutusta kokonaissuunnitteluprosessin verifiointiprosessiin. Verifiointiprosessiin on tällä hetkellä saatavilla kaupallisia sovelluksia, joita voidaan hyödyntää kolmannen osapuolen suunnitteleman systeemin testaukseen ja verifioitiin. Samalla on myös olemassa testausalustoja, joista nämä sovellukset puuttuvat. Tämä diplomityö keskittyy tilanteeseen, jossa täytyy rakentaa systeemi joka helpottaa testattavassa laitteessa (DUT) käytettävän tietoväyläprotokollan verifiointi- ja testausprosessia. Serial Peripheral Interface (SPI)-väyläprotokollan tutkimus tuo esiin esimerkkejä, joihin voidaan törmätä, kun kyseistä väyläprotokollaa käytetään missä tahansa sitä hyödyntävässä systeemissä. Diplomityössä tutkitaan erilaisia testaus- ja verifiointimetodeja, jotta voidaan osoittaa mitä erilaisia työkaluja voidaan hyödyntää, kun uusi testattava laite lisätään olemassa olevaan systeemiin tai tutkitaan tämän käyttämää väyläprotokollaa. Kokonaissuunnitteluprosessia on tutkittu esittelemällä tietyn järjestelmän iteraatiovaiheita, joka kehitettiin ratkaisemaan aiemmin tarkasteltuja, tähän aiheeseen liittyviä ongelmia. Suunnitteluprosessin voidaan katsoa alkaneen tilanteesta, jossa mitään konkreettista ei ollut vielä valmiina ja päättyvän tilanteeseen jossa järjestelmän viimeinen iteraatio voitiin alkaa konkretisoimaan. Monikäyttöisen syntetisoituvan verifiointijärjestelmän funktionaalisuus ja rakenne esitellään. Tutkimalla simulointi- ja synteesiraportteja näytetään tämän järjestelmän varmennusprosessi. Diplomityön toteutumista ja tulevaisuuden jatkokehitysideoista keskustellaan. Tämä diplomityö osoittaa, että Nokia soveltajayrityksenä pystyy hyödyntämään tämän tutkielman lopputulemia. De flesta metoder, verktyg och arbetssätt som används vid traditionell projektledning har använts under en lång tid trots att de yttre förutsätt-ningarna har förändrats. Systemutvecklingsbranschen upplevde de traditionella metoderna att driva projekt som oerhört statiska och tröga, därför utvecklades mer lättrörliga metoder som kom att kallas agila systemutvecklingsmetoder. Fram till år 2007 fanns inte en strukturerad process för hur valet och implementeringen av en agil systemutveckl-ingsmetod bör gå till. Det finns idag några ramverk som beskriver hur val och implementering av en agil metod kan bedrivas. Inget av dessa ramverk tar dock i beaktande eller utgår ifrån en organisations strategi eller vikten av strategisk passform. Syftet med denna rapport har varit att undersöka hur en agil systemutvecklingsmetod kan väljas utifrån strategisk passform. Undersökningen har även syftat till att studerade förändringar som kan krävas i en organisations projektstyrningsmodell för att införa en agil systemutvecklingsmetod. För att besvara syftet har en fallstudie utförts på Statens tjänstepensionsverk. På SPV har en ut-värdering med TS-metoden genomförts för att bedöma olika agila systemutvecklingsmetoders strategiska passform. Med hjälp av TS-metoden har agila systemutvecklingsmetoder bedömts mot en organi-sations strategiska mål. Resultatet från utvärderingen var att de agila systemutvecklingsmetoderna DSDM och Scrum ansågs ha den bästa strategiska passformen. Då DSDM till stor del är ett ramverk är en kom-bination av dessa båda systemutvecklingsmetoder möjlig. TS-metoden har resulterat i en relativ bedömning mellan de agila systemutveckl-ingsmetoderna. Utifrån denna bedömning har de agila systemutveckl-ingsmetoderna kunnat rangordnas utifrån ett strategiskt perspektiv. Införandet av en kombination av metoderna har inneburit att arbetet kommer att utföras iterativt istället för sekventiellt och detta har inneburit att beslutspunkterna i projektstyrningsmodellen har förändrats. Även styrning efter TKQ har försvunnit.
Most of the methods, tools and approaches used in traditional project management come from the Cold War days. System development sector experienced the traditional methods of managing projects as extremely static and slow and therefore developed more agile methods, agile sys-tem development methods. By the year 2007 there was not a structured process for the selection and implementation of an agile method. There is currently some framework that describes how the selection and im-plementation of an agile method can be conducted. None of these frameworks will be considered or assume an organization strategy and the importance of strategic fit. The purpose of this report was to investi-gate how an agile software development method can be chosen based on strategic fit. The survey also aimed to study the changes that would be required in an organization's project management model in order to introduce an agile system development methodology. To answer the question, an evaluation with TS-method was performed. In the TS-method has agile software development methodologies assessed against an organization's strategic goals. The result from the evaluation was that the agile software development methods DSDM and Scrum were con-sidered to have the best strategic fit. When the DSDM largely is a framework a combination of the two agile software methods is possible. TS-method gave a relative assessment between the agile software devel-opment methods. Based on this assessment, the agile software devel-opment methods could be ranked from a strategic perspective. The in-troduction of a combination of methods has meant that the work will be performed iteratively rather than sequentially, and this has meant that the decision points in the project management model have changed. Although control with TKQ has disappeared.

Анотація:
This diploma thesis addresses the problem of liquid and gas simulation, it particularly deals with computer simulation of flow of viscous newtonian liquids with a free surface. A main goal of this work is to create an efficient simulation model, utilizing the benefits of current GPU parallel architecture for general-purpose computing. I chose to implement Smoothed Particle Hydrodynamics, a lagrangian particle-based method. A significant portion of this thesis consists of speed analysis of the implemented algorithm, comparison with other authors' achievements in the field and a demonstration of benefits brought by GPU involvement in the computation. As an output of the thesis I present an interactive computer program that allows for real-time simulation (and visualization) of water-like fluids.

Neste trabalho foi feito um estudo investigativo sobre animação de fluidos utilizando sistemas de partículas. Baseado nas propostas apresentadas por Muller et al., esta dissertação objetiva investigar e compreender o uso do método Lagrangeano baseado em partículas, conhecido como Smoothed Particle Hydrodynamics (SPH), para simulação de fluidos. A validação do método foi feita através da implementação de uma biblioteca capaz de animar fluidos a taxas interativas. Para testar a eficácia e eficiência do método, a biblioteca desenvolvida permite a instanciação de diferentes configurações, incluindo o tratamento de colisões do fluido com obstáculos, o tratamento da interação entre dois fluidos distintos e o tratamento de forças externas exercidas pelo usuário via um mecanismo de interação.
This work investigates the use of particle-based system for fluid animation. Based on proposals presented by Müller et al., the goal of this dissertation is to investigate and fully understand the use of a Lagrangian method known as Smoothed Particle Hydrodynamics (SPH) for fluid simulations. A library has been implemented in order to validate the method for fluid animation at interactive rate. To demonstrate the method effectiveness and efficiency, the resulting library allows the instantiation of different configurations, including the treatment of fluid-obstacle collisions, interaction between two distinct fluids, and fluid-user interaction.

The research presented in this thesis addressed knowledge gaps for three medicinal plant taxa, Souroubea spp. (Marcgraviaceae) and Platanus (Platanaceae) as well as Echinacea spp. (Asteraceae). The primary pharmacological mechanism of Souroubea sympetala and Platanus occidentalis were well established, with pentacyclic triterpenes identified as major active principles. My results indicate that major triterpenoids, and crude plant extracts, selectively inhibited monoacyglycerol lipase (MAGL) activity but not fatty acid amide hydrolase (FAAH) activity. These data suggest a possible secondary anxiolytic mechanism of action through the endocannabinoid system (ECS). My study of herb-drug interactions of Souroubea and Plantanus products showed some potential risk when combined with a classic benzodiazepine class drug, diazepam, and I proposed a mechanism through in vitro CYP450 enzyme inhibition. The pharmacokinetic study revealed the difficulty of detecting betulinic acid in animal blood. To support the development a commercial botanical composed of these medicinal plants, an extraction method and a highly sensitive and selectivity HPLC-APCI-MS based quantification method was successfully developed and validated. Part II of this thesis focused on the impact of phytochemical variation and hepatic metabolism on the ECS activity of Echinacea spp. and explored the potential for new applications of Echinacea spp. as a natural health product. My research indicated that considerable variability in the content of phenolic and alkylamide (AKA) compounds reflected similar variability in in vitro bioactivity at ECS-related pharmacological targets. Following biochemometric analysis, several phenolic compounds and AKAs in Echinacea spp. were found to be significant independent variables determining FAAH inhibition and CB receptor activation. Hepatic metabolism was also found to affect the FAAH inhibition of AKA, as increased FAAH inhibitory effects were observed after CYP450-mediated metabolism of both individual AKAs and crude extracts of E angustifolia and E. purpurea, suggesting a “pro-drug” mechanism. Dose dependent activities were observed with oral administration of both E angustifolia and E. purpurea root extract in rat paw model of inflammation and pain. Further tests indicated these activities can be partially blocked by co-administration of CB1 and CB2 receptor antagonists AM251 and AM630, respectively. This evidence suggests activity for peripheral pain was at least partially mediated through the ECS.

It is a great research interest to integrate the Cell/B.E. processor into an AMD Opteron system. The result is a system benefiting from the advantages of both processors: the high computational power of the Cell/B.E. and the high I/O throughput of the Opteron. The task of this diploma thesis is to accomplish, that Cell-SPU code initially residing on the Opteron could be executed on the Cell under the GNU/Linux operating system. However, the SPUFS (Synergistic Processing Unit File System), provided from STI (Sony, Toshiba, IBM), does exactly the same thing on the Cell. The Cell is a combination of a PowerPC core and Synergistic Processing elements (SPE). The main work is to analyze the SPUFS and migrate it to the Opteron System. The result of the migration is a project called RSPUFS (Remote Synergistic Processing Unit File System), which provides nearly the same interface as SPUFS on the Cell side. The differences are caused by the TCP/IP link between Opteron and Cell, where no Remote Direct Memory Access (RDMA) is available. So it is not possible to write synchronously to the local store of the SPEs. The synchronization occurs implicitly before executing the Cell-SPU code. But not only semantics have changed: to access the XDR memory RSPUFS extends SPUFS with a special XDR interface, where the application can map the XDR into the local address space. The application must be aware of synchronization with an explicit call of the provided ''xdr\_sync'' routine. Another difference is, that RSPUFS does not support the gang principle of SPUFS, which is necessary to set the affinity between the SPEs. This thesis deals not only with the operating system part, but also with a library called ''libspe''. Libspe provides a wrapper around the SPUFS system calls. It is essential to port this library to the Opteron, because most of the Cell applications use it. Libspe is not only a wrapper, it saves a lot of work for the developer as well, like loading the Cell-SPU code or managing the context and system calls initiated by the SPE. Thus it has to be ported, too. The result of the work is, that an application can link against the modified libspe on the Opteron gaining direct access to the Synergistic Processor Elements.

This study has   focused on system development approaches and system development   methodologies. The purpose of the study has been to present a view on the   current situation of different development approaches (agile, traditional,   iterative and hybrid) and methodologies by highlighting the situation from a   number of perspectives. Questions that are answered are: which system   development approaches and methodologies that can be said to be most   frequently used based on the respondents, if the mentioned approaches can be   given certain characteristics based on a selection of factors and the   respondents' attitude towards the approach they practice. The study has been   conducted as a survey where the answers have been obtained via social media.   The result shows that agile methodologies dominate and that they are often   included in various forms of hybrid and agile approaches. Furthermore, it has   not been possible to create an in-depth knowledge of the approaches based on   the selected factors. Finally, it is noted that project members who practice   an agile approach have a great confidence in the approach and have a strong   conviction that it contributes to increase the quality of the software.

The core of this thesis is to provide an exhaustive identi cation of the vibrational properties of the cluster assembled sp-sp2 pure carbon material. This system consists in an amorphous sp2 matrix where spCCs are embedded and stabilized by sp2 fragments and it is synthesized by depositing, through the Supersonic Cluster Beam Deposition (SCBD) technique, sp-sp2 pure carbon clusters produced in a Pulsed Microplasma Cluster Source (PMCS). Although this material has been continuously studied for long time mainly by Raman spectroscopy, its vibrational features are not clear yet. In order obtain a better comprehension on the presence, structure and stability of spCCs in the sp-sp2 pure carbon, new experimental and theoretical eff orts are required. In particular it is necessary to clearly and unambiguously identify chains with di erent lengths and terminations in order to obtain a clear description of the physical and chemical properties characterizing the sp-sp2 material. Here I propose the synthesis, characterization and modeling of spCCs terminated by the simplest sp2 termination, i.e. Naphthalene, which well approximates an sp2 end-group present in the sp-sp2 system (graphene nano-ribbons). The combined experimental-theoretical study of the solution containing dinaphtylpolyynes with di erent length, performed through Infrared and Multi-Wavelength Resonant Raman spectroscopy combined with structural and vibrational ab-initio density-functional theory simulations, leads to the deep comprehension of the vibrational properties of the dinaphtylpolyynes. The model provided for dinaphtylpolyynes can be then used for the interpretation of the intricate results obtained by performing Infrared and Multi-Wavelength Resonant Raman experiment on the complex sp-sp2 pure carbon material.

A raiva é uma antropozoonose e causa a morte de 55 mil pessoas anualmente no mundo. Neste trabalho foi utilizado um sistema genético derivado do Vírus da Floresta de Semliki (SFV) carregando RNA para expressão gênica da glicoproteína do vírus da raiva (RVGP) e da proteína verde fluorescente (GFP), a título de controle do experimento. Lotes de partículas virais de SFV-RVGP e SFV-GFP foram obtidos em células BHK-21 e quantificados através de qRT-PCR. Foram realizados ensaios de infecção tanto com vírus ativado e não ativado em duas linhagens de macrófagos murinos IC-21 e J774A-1. A expressão da proteína RVGP foi avaliada pelo teste de Imunofluorescência Indireta (IFI) e a expressão da proteína GFP analisada por Microscopia de Fluorescência e Citometria de Fluxo em ambas as linhagens. A entrada do vírus SFV em células de mamíferos ocorre após 2 horas da infeção e de acordo com o experimento no qual analisamos a titulação viral do sobrenadante celular, indicaram que até as 72 horas pós infecção a titulação viral se manteve com o mesmo valor inicial, sugerindo que não houve entrada dos vírus na célula por transdução ou por fagocitose.
Rabies is a anthropozoonosis and causes the death of 55,000 people worldwide each year. In this work we used a genetic system derived from the Semliki forest virus (SFV) carrying RNA for gene expression of the rabies virus glycoprotein (RVGP) and green fluorescent protein (GFP), by way of experiment control. Plots of viral particles of SFV-RVGP and SFV-GFP were obtained in BHK-21 cells and quantified by qRT-PCR. Infection assays were performed both with activated and not activated in two virus strains of murine macrophages IC-21 and J774A-1. The expression of RVGP protein was assessed by indirect immunofluorescence test (IFI) and the expression of GFP protein analyzed by fluorescence microscopy and flow cytometry in both strains. The SFV entry of viruses into mammalian cells occurs after 2 hours of infection and according to the experiment in which we analyzed the viral titer of the cell supernatant showed that by 72 hours post-infection viral titer remained at the same initial value, suggesting no entry of virus into the cell by transduction or by phagocytosis.

The study’s objective is to analyze the role that Hong Kong has in the global system; with the aim of putting light on how Hong Kong has evolved, the analysis starts with an historical review of Hong Kong, from its early period till the Handover to China in 1997 under the “One country, two systems” formula and the establishment of the HKSAR. Moving to the core of the analysis, the study examines those that are considered, from a foreign investor’s viewpoint, the points of strength of Hong Kong: the focus will be on the main aspects that make it important and attractive for the international business community and on the reasons why it is convenient to invest in or through it. For this purpose, a triple perspective is adopted: the analysis of Hong Kong’s advantages is structured into the three main aspects in which Hong Kong, in light of the literature and the data examined, excels; first, the role of Hong Kong as a business hub is discussed, then its position as an international financial center and as a trade and connectivity hub is examined. Furthermore, a second perspective is used to conduct the analysis: all of the Hong Kong’s advantages belonging to these three macro areas are explained in relation to a dichotomy, that is Hong Kong’s independence from and proximity to China. Finally, the analysis is summed up in a brief conclusive part.

Streptomyces are Gram-positive bacteria belonging to the Actinobacteria. They are predominantly soil-dwelling saprophytic organisms. S. coelicolor is often used as a model organism for the study of the mechanisms underlying the processes of development and antibiotic production. S. scabies is one of only a few members of the genus that is pathogenic, being the causal agent of common scab of potato. Bioinformatic analysis of the genomes of both S. coelicolor and S. scabies revealed the presence of genes encoding components of putative Type VII protein secretion systems (T7SSs). To investigate the role of these putative T7SSs in S. coelicolor and S. scabies, marked deletions were constructed in genes encoding core components of the machinery. Phenotypic analysis revealed that deletion of genes encoding the T7SS FtsK/SpoIIIE ATPases (FSD) resulted in delayed actinorhodin production in S. coelicolor and slightly accelerated aerial development in S. scabies. In S. scabies, deletion of the genes encoding the WXG100 proteins, canonical substrates of T7SSs, led to a marked reduction in aerial hyphae formation and sporulation, in a media-dependent manner when compared to the wild-type strain. Closer inspection by scanning electron microscopy revealed that the spore chains of these mutant strains were under-developed and spores were irregularly sized. Strains of S. scabies lacking the WXG proteins (but not the FSD protein) were also resistant to lytic infection by the bacteriophage FC31. Taken together, these observations indicate that the major role of the WXG100 proteins in Streptomyces is intracellular.Plant infection assays indicate that the T7SS of S. scabies does not significantly contribute to virulence in several laboratory models of plant infection.

Neuropathogenesis of HIV-associated neurocognitive disorders (HAND) is likely instigated by chronic immune activation in response to residual infection in the central nervous system (CNS), where combined antiretroviral therapy (cART) has limited access. Monocyte/macrophages (Mo/Mϕ) constitute the predominant population of infected cells in the CNS and play a major role in HIV-induced neuropathogenesis. Emergence of compartmentalized HIV subpopulations in the brain corresponds with accumulation of HIV-infected Mo/Mϕ and is consistent with acquired immune deficiency syndrome (AIDS)-related neuropathology. We used a rhesus macaque model of neuroAIDS to elucidate the role of Mo/Mϕ in establishing CNS infection and the emergence of compartmentalized virus in the brain. To do this, we: 1) performed phylogenetic analysis of viral sequences from peripheral and CNS compartments and determined the incidence of Mo/Mϕ infection in CNS tissues to identify sources of CNS viral subpopulations that emerge with AIDS-related neuropathology; 2) optimized a method for obtaining single genome viral sequences from Mo/Mϕ populations extracted from tissues and 3) performed phylogenetic analysis of viral sequences from bone marrow (BM) and CNS Mo/Mϕ and determined the incidence of Mo/Mϕ infection in the BM to assess whether BM Mo/Mϕ are sources of infected Mo/Mϕ that accumulate in the CNS with AIDS-related neuropathology. We found that animals with AIDS-related neuropathology had a higher incidence of Mo/Mϕ infection and compartmentalized SIV subpopulations in CNS tissues compared to animals without neuropathology. Additionally, CSF virus, which is used to assess the presence of CNS virus compartmentalization in living patients, was not compartmentalized even with significant compartmentalization in the brain and severe AIDS-related neuropathology (Chapter 2). Relative to animals without CNS pathology, animals with AIDS-related neuropathology had a higher incidence of Mo/Mϕ infection in the BM and viral sequences from BM and CNS perivascular Mo/Mϕ clustered with sequences from trafficking monocytes and CNS tissues (Chapter 4). The results suggest that infected Mo/MΦ in CNS tissues are sources of compartmentalized virus and that infected Mo/Mϕ in the BM are sources of infected Mo/Mϕ that accumulate in the CNS with AIDS-related neuropathology. In summary, the data in this dissertation suggest that targeting Mo/Mϕ may prevent CNS infection and inflammation associated with HIV-induced neuropathogenesis

Sepsis is defined as a life-threatening organ dysfunction caused by a dysregulated host response system and can occur when the immune system over- or under- reacts to an infection. Klebsiella spp. has been found to be one of the leading causes of sepsis, and the increasing occurrence of antibiotic resistance observed has become a major concern in clinical care. To study the genome and increase knowledge of the biodiversity of K. pneumoniae, K. variicola, and K. oxytoca, bacterial isolates were collected from blood, urine, nasopharynx, and wounds of patients with suspected sepsis. Next-generation sequencing was performed, and the presence of antibiotic resistance genes and plasmids were studied. Furthermore, a prediction of traits for each phylogroup was performed and the results from whole-genome sequencing were compared to phenotypic results. Among the K. pneumoniae isolates obtained, almost half had been misidentified by standard phenotypic methods and were found to be K. variicola, K. quasipneumoniae, and K. quasivariicola. A significant difference in the number of antibiotic resistance genes were observed between K. pneumoniae and K. variicola compared to K. oxytoca, however no significant difference was observed between K. pneumoniae and K. variicola, suggesting the underestimated pathogenicity of K. variicola. A genetic agreement was observed between the type of beta-lactamase harboured and presence or absence of nitrogen-fixation genes to the phylogroup, providing a way of species identification. Further studies should be conducted on the pathogenicity and virulence of K. variicola and K. quasipneumoniae to avoid misidentification, find organism-specific treatments, and narrow down the antibiotic prescription.

Syftet med denna rapport är att kartlägga Scania Production System (SPS), hur det motiverades och implementerades inom Scanias organisation samt dra kopplingar mellan tidigare reformer och SPS. Vi vill ta reda på hur Scania lyckats skapa ett eget skräddarsytt produktionssystem som visat sig vara så framgångsrikt och hur detta infördes inom hela organisationen. SPS är ett resultat av Toyota och deras Toyota Production System (TPS) efter ett nära samarbete mellan Scania och Toyota sedan början av nittiotalet. Scanias värderingar, principer och prioriteringar beskrivs av SPS och har visat sig vara en av företagets konkurrensfördelar genom ökad effektivitet samt produktivitet inom produktionen. TPS fick mycket av sin inspiration av de tidigare reformerna Taylorismen och Fordismen ur rationaliseringsrörelsen. Detta genom Taylors idéer om standardisering och Fords löpandebands-princip. Toyota var även bland dem första att utforma och tillämpa de logistiska reformerna Total Quality Management, Lean Production och Just-In-Time i sitt produktionssystem. Reformer som används och är välkända inom många företag inte minst Scania och SPS. Rapporten uppfyllde sitt syfte genom intervjuer och analys av konceptlitteratur för att kunna konstatera att det finns en röd tråd inom företaget och att det finns en tydlig koppling mellan SPS och tidigare reformer. Avslutningsvis lyckades vi även analysera och dra slutsatser om hur SPS utformades, motiverades och implementerades. Utformningen av SPS gick till genom ett nära samarbete mellan ledningar på Scania och Toyota vilken Scania på så sätt kunnat ta inspiration ifrån och kunnat anpassa systemet efter Scanias kultur. Hur sedan SPS motiverades och implanterades föregick i en ledningsprocess som Scania tillämpade inom organisationen. Lösningen kom att bli avgörande för framtiden, ledarskapet. Man tillämpade ett nytt tankesätt genom hela verksamheten vilket har visat sig vara framgångsrikt.
The purpose of this report is to map the Scania Production System (SPS), how it was motivated and implemented within Scania's organization, as well as linking previous reforms with SPS. We want to find out how Scania managed to create its own customized production system that proved being successful and how it was introduced throughout the organization. SPS is a result of Toyota and their Toyota Production System (TPS) following a close collaboration between Scania and Toyota since the early 1990s. Scania's values, principles and priorities are described by SPS and have proven to be one of the company's competitive advantages through increased efficiency and productivity in production. TPS received much inspiration from the earlier reforms of Taylorism and Fordism from the rationalization movement. This through Taylor's ideas about standardization and Ford's running band principle. Toyota was also one of the first to design and apply the logistics theory’s Total Quality Management, Lean Production and Just-In-Time in its production system. Reforms used and well known in many companies, not least Scania and SPS. The report fulfilled its purpose through interviews and analysis of conceptual literature to find that there is a red thread within the company and that there is a clear link between SPS and previous reforms. In conclusion, we also managed to analyse and draw conclusions about how the SPS was designed, motivated and implemented. The design of SPS was achieved through close collaboration between Scania and Toyota wires, which enabled Scania to inspire and adapt the system to Scania's culture. The way in which SPS was motivated and implanted preceded a management process that Scania applied to within the organization. The solution came to be decisive for the future, leadership. A new way of thinking was applied throughout the business, which has proved to be successful.

Complex multidimensional capturing setups such as plenoptic cameras (PC) introduce a trade-off between various system properties. Consequently, established capturing properties, like image resolution, need to be described thoroughly for these systems. Therefore models and metrics that assist exploring and formulating this trade-off are highly beneficial for studying as well as designing of complex capturing systems. This work demonstrates the capability of our previously proposed sampling pattern cube (SPC) model to extract the lateral resolution for plenoptic capturing systems. The SPC carries both ray information as well as focal properties of the capturing system it models. The proposed operator extracts the lateral resolution from the SPC model throughout an arbitrary number of depth planes giving a depth-resolution profile. This operator utilizes focal properties of the capturing system as well as the geometrical distribution of the light containers which are the elements in the SPC model. We have validated the lateral resolution operator for different capturing setups by comparing the results with those from Monte Carlo numerical simulations based on the wave optics model. The lateral resolution predicted by the SPC model agrees with the results from the more complex wave optics model better than both the ray based model and our previously proposed lateral resolution operator. This agreement strengthens the conclusion that the SPC fills the gap between ray-based models and the real system performance, by including the focal information of the system as a model parameter. The SPC is proven a simple yet efficient model for extracting the lateral resolution as a high-level property of complex plenoptic capturing systems.

Recently the EPA placed Aeromonas hydrophila on the Candidate Contaminant List (CCL). It has long been known to be a pathogen of cold blooded animals and now is a suspected human opportunistic pathogen as well. Among the various virulence factors produced by A. hydrophila, the cytolytic enterotoxin (AHCYTOEN) is by far one of the most important contributors to the pathogenicity of the organism. This factor is also produced by other pathogenic Aeromonas spp. In this study, PCR technology was used to detect AHCYTOEN gene from a simulated water distribution system. A set of primers was selected to amplify the unique sequence of a pathogenic island, AHCYTOEN gene. To examine the sensitivity of the PCR, serial dilutions of pure A. hydrophila culture were tested. The PCR technique used was sensitive enough to detect samples containing less than 10.0 cells/ml. Source water, bulk water, and simulated distribution biofilm samples were examined for the gene. Biofilm and bulk water samples exposed to raw source water were collected on 4 occasions during a 24-day period. PCR technology detected the AHCYTOEN gene from 100 % of the bulk water samples and 85% of tightly bound biofilm (TB) samples from a simulated water distribution system while no positive results were observed in loosely bound biofilm samples (LB). After the inlet line of the system was changed to normally treated distribution water, 11 biofilm samples were collected on 3 occasions during 15 day sampling period along with bulk water samples. No positive results were observed from the bulk water and LB samples while 91% of TB samples tested for the presence of the gene. No significant difference was observed in detection by PCR from biofilm samples before and after the switch to chloraminated water.

Salmonella enterica serovar Typhimurium (S. Typhimurium), an intracellular pathogen, causes gastroenteritis in humans and a systemic disease in mice. The ability of Salmonella to replicate inside host cells requires translocation of effector proteins across the vacuolar membrane, mediated by the Salmonella pathogenicity island‐2 (SPI‐2) type three secretion system (T3SS). However, the repertoire of effectors involved in this process has not been defined. The first part of this PhD work focused on SrfJ, a putative effector of the SPI‐2 T3SS with similarity to human lysosomal glucosylceramidase. Expression of its gene was dependent on SsrA/B, a two‐component regulatory system required for expression of most SPI‐2 effector genes. Expression of srfJ was also shown to occur under SPI‐2 T3SS activation conditions. However, there was no detectable secretion or translocation of the protein, although a srfJ mutant strain had an intracellular replication defect in primary bone marrow‐derived macrophages. Using a dual‐fluorescence reporter system that allows direct measurement of intracellular replication, the contribution to replication of individual SPI‐2 T3SS effectors was investigated. The replication kinetics of S. Typhimurium deletion mutants for all known SPI‐2 effectors were measured and compared in mouse bone marrow‐derived macrophages. Several mutant strains with replication defects were identified, thereby revealing that intracellular replication is the result of the contribution of numerous effectors. Two S. Typhimurium polymutant strains were generated whose replication defects closely resemble that of a SPI‐2 T3SS null mutant and are severely attenuated in virulence in vivo. These strains retained an intact T3SS and delivered a CD8+ T cell epitope via the SPI‐2 T3SS into the cytoplasm of infected cells. Since an S. Typhi mutant strain lacking the SPI‐2 T3SS has been shown to be safe and immunogenic in humans, these polymutant strains could have applications in vaccine design, as carrier strains for the delivery of heterologous antigens.

The project presented in this report is set out to develop a portable electronic system to be used as a medicine pill container. With the functionality to configure up to twelve daily repeated alarms, the purpose of the medicine dosage system is first and foremost to remind the user when they should take their medicine. Secondly, LED lights and user-recorded voice notifications are to be implemented to further aid the user in taking the right medicine each time. The device is also to have a memory log, recording up to one hundred missed dosages, enabling an authorized medicine professional to verify the medicine adherence of the user.Prior to the start of the project, an outline for the functionality and physical appearance of the device was set by the project owner, Victrix AB. This project covers the hardware and software development, as well as the design choices within. The aim is to follow the proposed functionality specification as close as possible, while making justified hardware and software choices considering simplicity, efficiency, power consumption, and availability. By following the specification, the goal is ultimately to increase the medicine adherence for users of the device developed with this thesis.Using qualitative research methods, a valid background study was created, preceding the development of the medicine dosage system. Hardware for a first prototype of the device was then chosen based on the gathered information about existing technologies and related work. With thorough testing and recurrent information exchange with the client, a prototype of the medicine dosage system, based on an Arduino microcontroller, was constructed. The prototype was evaluated to fulfill 92% of the requirements considered as high priority by Victrix.
Projektet som presenteras i denna rapport är tänkt att utveckla ett portabelt elektroniskt system för användning som en medicinsk pillerbehållare. Med funktionaliteten att konfigurera upp till tolv dagligen upprepande alarm är syftet med medicindoseringssystemet först och främst att påminna användaren när de ska ta sin medicin. Lysdioder och användarens egna inspelade röst som notifikationer ska implementeras för att vidare hjälpa användaren att ta rätt medicin vid varje tillfälle. Enheten ska också ha en minneslogg som sparar upp till etthundra missade doseringar, vilket gör det möjligt för auktoriserad sjukvårdspersonal att verifiera användarens följsamhet till medicineringen.En översiktlig beskrivning av funktionaliteten samt det fysiska utseendet av enheten skrevs av projektägaren Victrix AB innan projektet startades. Det som detta projekt täcker är hårdvaruoch mjukvaruutvecklingen, så väl som där tillhörande designval. Projektet siktar på att följa den föreslagna funktionalitetsspecifikationen så nära som möjligt, och samtidigt göra välgrundade val för hårdoch mjukvara med enkelhet, effektivitet, energiförbrukning och tillgänglighet i åtanke. Genom att följa specifikationen är det slutliga målet att frambringa ökad medicinföljsamhet för användare av den med det här projektet utvecklade enheten.Utvecklingen av medicindoseringssystemet föregicks av en befogad bakgrundsstudie utformad genom användningen av kvalitativa forskningsmetoder. Hårdvara att användas för en första prototyp av enheten valdes sedan baserat på den insamlade informationen om existerande teknologier och relaterat arbete. Genom grundliga tester och regelbundet informationsutbyte med kunden konstruerades en prototyp av medicindoseringssystemet baserat på en Arduinomikrokontroller. Prototypen utvärderades att uppfylla 92% av kraven som Victrix ansåg vara av hög prioritet.

GSHP systems are common in Sweden but there are few evaluations quantifying the performance of the systems and highlighting problem that occurs during operations. The research project Annex 52 Long-term performance measurement of GSHP systems serving commercial, institutional and multifamily building part of IEA HPT TCP proves the need to systematically be able to evaluate GSHP systems. This thesis aims to expand the knowledge of how to evaluate GSHP systems and provide case studies for Annex 52. Three residential ground source heating systems used for heating has been evaluated and analyzed in this study. The evaluation has consisted of three parts. The first part analyzes the operation and stability of the GSHP systems. The second part consist of a detailed study of the performance of the GSHP systems. The seasonal performance factor has been calculated for different system boundaries based on the work done by SEPEMO. In addition, a method on how to evaluate the efficiency of the heat pumps based on the two temperature levels, source side temperature and the heat sink temperature, that the heat pump is operating at throughout a year has been developed within this thesis. This has included a method on how to normalize the temperatures based on the operation of the heat pump in order to quantify one temperature for each the two temperature levels. The third part consist of a comparison of the mean secondary fluid temperature between the calculated temperature using the software EED and the measured temperatures. This includes a comparison evaluation and sensitivity analysis on input parameters during the design of the borehole heat exchanger fields. This study has expanded the available reference cases of GSHP systems in Sweden. It also can be used as a guideline for those who will evaluate future GSHP systems. Designers of GSHP system will also benefit from the recommendations listed in this thesis regarding instrumentation and possible problems that may occur. The results show that the evaluation successfully managed to quantify the performance and operational issues that have occurred for each system. The method developed in this study was able to quantify the operation of the different systems based on the temperature levels and can be used for future GSHP evaluations of similar system type.
Bergvärmesystem är vanligt förekommande i Sverige men trots detta finns det få studier där prestandan har utvärderats och de vanligt förekommande problemen under drift har belysts. Forskningsprojektet Annex 52 Annex 52 Long-term performance measurement of GSHP systems serving commercial, institutional and multi-family building som är en del av IEA HPT TCP visar på behovet av att systematisk utvärdera bergvärmesystem. Detta examensarbete syftar till att utveckla och bidra till kunskap om hur bergvärmesystem kan utvärderas och att bidra med exempelstudier till Annex 52. Inom detta examensarbete har tre bergvärmesystem som betjänar flerbostadshus utvärderats och analyserats. Utvärderingen bestod av tre analyser. I den första analyserades driften av bergvärmesystemen och hur stabilt systemet har varit historiskt. Detta följdes av en detaljerad analys av olika nyckeltal för bergvärmesystemen. Årsverkningsgraden har beräknats för olika gränsdragningar vilka baseras på det tidigare arbetet utfört av SEPEMO. Inom detta examensarbete har även en metod tagits fram för att utvärdera verkningsgraderna för en värmepump baserat på de två temperaturnivåerna, köldbärarsidan och värmebärarsidan, som värmepumpen arbetar med under ett år. Till detta har en metod tagits fram om hur temperaturen kan normaliserats baserat på driften av värmepumparna för att kvantifiera en temperatur vardera för de två temperaturnivåerna. I den tredje utvärderingen jämfördes den beräknade medelfluidtemperaturen av köldbäraren i borrhålen med den uppmätta temperaturen. Till detta utfördes en känslighetsanalys av hur indata av dessa beräkningar påverkar resultaten.

Injection in the LHC is a delicate moment, since the LHC collimation system cannot offer adequate protection during beam transfer. For this reason, a complex chain of injection protection devices has been put in place. Among them, the SPS scrapers are the multi-turn cleaning system installed in the SPS aimed at halo removal immediately before injection in the LHC. The upgrade in luminosity of the LHC foresees beams brighter than those currently available in machine, posing serious problems to the performance of the existing injection protection systems. In particular, the integrity of beam-intercepting devices is challenged by unprecedented beam parameters, leading to interactions potentially destructive. In this context, a new design of scrapers has been proposed, aimed at improved robustness and performance. This thesis compares the two scraping systems, i.e. the existing one and the one proposed for upgrade. Unlike any other collimation system for regular halo cleaning, both are "fast" systems, characterised by the variation of the relative distance between the beam and the absorbing medium during cleaning, which enhances the challenge on energy deposition values. Assets/liabilities of the two systems are highlighted by means of numerical simulations and discussed, with particular emphasis on energy deposition in the absorbing medium, time evolution of the beam current during scraping and losses in the machine. Advantages of the system proposed for upgrade over the existing one are highlighted. The analysis of the existing system takes into account present operational conditions and addresses the sensitivity to settings previously not considered, updating and extending past studies. The work carried out on the upgraded system represents the first extensive characterisation of a multi-turn cleaning system based on a magnetic bump. Results have been obtained with the Fluka-SixTrack coupling, developed during this PhD activity from its initial version to being a state-of-art tracking tool for cleaning studies in circular machines. Relevant contributions to the development involve the handling of time-varying impact conditions. An extensive benchmark against a test of the scraper blades with beam has been carried out, to verify the reliability of results. Effcts induced in the tested blades confirm the high values of energy deposition predicted by the simulation. Moreover, the comparison with the time profile of the beam intensity measured during scraping allowed the reconstruction of the actual settings of the blades during the test. Finally, the good agreement of the quantitative benchmark against readouts of beam loss monitors finally proves the quality of the analyses and the maturity of the coupling.

The effect of competing food sources on termite consumption of hexaflumuron bait and subsequent mortality was examined. Also, the effect of hexaflumuron on the termite gut fauna was evaluated to determine if hexaflumuron could kill termites via a secondary mode of action. Hexaflumuron consumption in no-choice and choice tests was evaluated at 2d and 5d. Total diet consumption was not different between the treatment groups. Hexaflumuron consumption was reduced by a factor of 3 in the presence of a competing control diet and reduced by a factor of 57 in the presence of an inulin diet. In the choice test, termites preferred inulin over hexaflumuron. Termite mortality after hexaflumuron consumption was quantified at 5d, 15d, 20d, 25d, and 35d in three treatment groups. Mean mortality for termites fed only hexaflumuron diet was significantly higher than termites fed only control diet. Mean mortality for termites given a choice was no different than mortality for termites fed only hexaflumuron or control diet. LT50s for termites fed control diet, hexaflumuron diet, or both diets in the choice test, were 24.4d, 18.7d, and 20.6d, respectively. The choice test LT50 did not differ from the LT50 of either no-choice test. Termites fed only hexaflumuron diet had an earlier LT50 than termites fed only control diet. The effect of hexaflumuron on gut fauna survival was evaluated at 5d and 15d. No significant differences were found in total numbers of protozoa in termites fed either hexaflumuron or control diet. Pyrsonympha was the only protozoa significantly reduced by hexaflumuron consumption.

Escherichia coli possède une grande plasticité génomique comme en témoigne la diversité des souches à l’intérieur de cette espèce bactérienne. Bien que la majorité des souches soient inoffensives ou à tout le moins opportunistes, plusieurs ont acquis des facteurs de virulence spécifiques leur procurant un pouvoir pathogénique. Les souches pathogènes comme E. coli O157 :H7 sont responsables de cas de morbidité, mortalité et pertes économiques importantes dans l’industrie agro-alimentaire dans le monde entier. L’évolution bactérienne est un mécanisme continuel qui se fait via l’échange d’éléments génétiques mobiles, de mutations ponctuelles et autres réarrangements génétiques. Ces changements génétiques peuvent procurer des avantages sélectifs permettant une adaptation bactérienne rapide face aux stress et changements environnementaux et favorisant le développement de pathogènes émergents. Dans la première partie de ce projet, nous avons étudié la région intergénique mutS-rpoS, qui est une des plus grandes sources de polymorphisme chromosomique chez les entérobactéries. Notre analyse génomique comparative a permis de confirmer le polymorphisme à l’intérieur même d’un ensemble de souches Escherichia spp., Salmonella spp. et Shigella spp. De plus, nous avons pu confirmer que certains types de polymorphismes dans la région mutS-rpoS étaient fortement associés à certains types de pathogènes chez E. coli. Dans notre analyse, nous avons ressorti un groupe de gènes à l’intérieur de la région mutS-rpoS qui pourraient sevir comme marqueur chromosomique intéressant pour les E. coli extra-intestinaux (ExPEC), un groupe comprennant des souches hautement pathogènes et difficiles à définir par les tests actuelllement disponibles. Dans notre analyse bio-informatique, nous avons isolé ce groupe de gènes associé aux ExPEC et nous l’avons caractérisé in sillico. Nous avons également inclus dans l’analyse deux souches hypermutables du genre Escherichia spp. de notre collection, isolées d’animaux de ferme. L’hypermutabilité ou la capacité d’acquérir des mutations plus rapidement que la normale accélère le processus d’évolution et la capacité d’adaptation de ces souches. La région mutS-rpoS est reliée au système de réparation de l’ADN bactérien (MMRS) et pourrait être impliquée dans l’apparition du phénotype d’hypermutabilité. Durant les dernières années, de plus en plus d’espèces du genre Escherichia ont été isolées de cas cliniques d’animaux et d’humains. Ces souches atypiques ont un potentiel de virulence très élevé, des combinaisons de gènes de virulence et des variants génétiques différents des souches typiques, et certaines souches ont même évolué en tant que pathogènes. Les souches de l’espèce E. albertii ont été isolées récemment et ont un grand potentiel de virulence autant chez les humains que chez les oiseaux. Ces souches sont souvent confondues avec d’autres organismes pathogènes comme E. coli dans les tests biochimiques, et le manque de connaissances sur E. albertii rend son identification difficile. Dans la deuxième partie de ce projet, nous avons identifié des gènes spécifiques aux souches d’E. albertii ainsi que des gènes de virulence présents chez E. albertii par comparaisons génomiques, ce qui a permis de développer et optimiser un test PCR (réaction en chaîne par polymérase) visant l’identification génomique rapide et fiable d’E. albertii.
