Повний текст джерелаL-fucose and oligosaccharides containing of L-fucose present interesting biological properties notably in the prevention of the metastases, in the reaction of inflammation, in the treatment of rheumatoid arthritics, in the vaccination (antigens) and in the cosmetic domain against the dehydration of the skin. A production of L-fucose as of oligosaccharides containing of L-fucose with the aim of their use in therapies, pushed numerous laboratories to a screening of organisms synthetizing polysaccharides containing of L-fucose. Our work, concerned first of all the production of the polysaccharides by Sinorhizobium M5N1 cPRK290 noted N and Enterobacter noted Sc strains, later these polysaccharides were characterized. And secondly we produced oligosaccharides containing of L-fucose from polysaccharides. This production of oligosaccharides required the use of several methods (acid hydrolysis, thermal hydrolysis, in the medium of culture and enzymatic degradation). The enzymatic method allows to obtain oligosaccharides in a reproducible way

Повний текст джерелаA lot of progress had been done in recent years to understand the regulation of synthesis and secretion of cellulases in the fungus Trichoderma reesei. However, the production cost of cellulases still remains one of the most important limiting steps in the production of bioethanol from lignocellulose. It is in this context that my PhD project has been developed: to genetic engineering T. reesei strains to increase its cellulase production and its adaptation to industrial conditions. First, we conducted a transcriptome analysis to an improved lineage of industrial strains during cellulase production following conditions close to the industrial process (lactose as inducer and fed-batch culture in bioreactor). We found specifically regulated genes for the most performant strain, possibly involved in its high cellulase production capacity. Then, we identified which regulated genes from transcriptome were involved in cellulase production. For this purpose mutated strains for highly regulated genes were constructed and their phenotype assessed. Three mutated genes showed to impact cellulase production and their function gave us an insight into new mechanisms being regulated. Finally, we explored the expression differences when we used a lignocellulose hydrolysate as cellulases inducer instead of lactose. A transcriptomic study of cellulases production on lignocellulose hydrolysates and lactose, allowed us to characterize the differences in regulated genes between these inducers. In addition, a group of genes probably related to detoxification was identified and could be used in the future to develop an inhibitor resistant strain

Повний текст джерелаHuntington's Disease (HD) is a fatal and progressive neurodegenerative disease. There is no cure for this genetic disease caused by an abnormal CAG-repeats expansion within the coding sequence of the Huntingtin (HTT) gene. The mutated isoform of the protein (mut-HTT) is associated with massive neuronal loss in the striatum and the cortex. Several studies have demonstrated that reduction of mut-HTT levels decreases symptoms associated with HD and increases life span in mouse model of the disease. Both pre-clinical and clinical programs aimed at HTT-lowering are currently under development. In this project, we perfomed a high throughput screening to identify small molecules with mut-HTT lowering activities in neural stem cells or striatal neurons derived from patient-specific human induced pluripotent stem cells. We miniaturized and automated an assay to detect mut-HTT level in human cells that was used for the screening of drug repurposing libraries. We identified several small molecules with mut-HTT-lowering cellular activities. Additional biochemical analyses confirmed those activities and for some revealed cell type specificity. Overall, these results open up new therapeutic perspectives for patients with Huntington's Disease
