Анотація:
Wälder stellen wichtige Ökosystemleistungen und -güter bereit und tragen damit maßgeblich zur Lebensgrundlage der Menschen bei. Sie unterliegen jedoch vielseitigen Bedrohungen, welche die volle Funktionsfähigkeit des Ökosystems gefährden und deren Entwicklung beeinträchtigen. Inwiefern sich diese auf die Funktionsweise und die Walddynamik in Kasachstan äußert, ist aufgrund der wenigen durchgeführten dendroklimatischen Untersuchungen im Detail unklar. Hinzu kommt, dass für die qualitative Bewertung der Klima-Wachstums-Beziehungen diese Studien in ihrer räumlichen und zeitlichen Abdeckung unzureichend sind. Zu den bereits verfügbaren Informationen fehlen zur Einschätzung gemeinsamer Muster und Unterschiede hinsichtlich der Reaktion des Baumwachstums auf das Klima qualitative Analysen. Hierfür wurden alle verfügbaren Publikationen zu den dendroklimatischen Untersuchungen in Kasachstan analysiert. Diese Analyse ermöglichte nicht nur die Zusammenfassung der verfügbaren Informationen zu den Beziehungen zwischen Klima und Wachstum, sondern lieferte zudem konkrete Informationen zu vorhandenen Forschungslücken. Einige dieser Lücken wurden durch die Untersuchungen der variierenden Klima-Wachstums-Beziehungen der Schrenks Fichte im Einzugsgebiet des Ile-Flusses geschlossen. Die durchgeführte Studie verdeutlicht unterschiedliche Reaktion von Früh-, Spätholz und Gesamtringbreite auf den Klimawandel. Der starke Einfluss des Klimawandels wurde durch die Abnahme des Temperatursignals um die 1970er-Jahre beobachtet. Eine erkennbare Periodizität bei Schwankungen der Baumringindizes lässt auf einen Einfluss bestimmter Klimaindizes, z. B. der Nordatlantischen Oszillation u. a. schließen. Die im Rahmen dieser Dissertation durchgeführten Untersuchungen tragen zur Entwicklung eines Baumring-Netzwerkes in Kasachstan bei. Die erhoben Daten sind für die Bewertung möglicher Auswirkungen des Klimawandels auf die Wälder der Schrenks Fichte in Kasachstan hilfreich.
Forests are very important for people’s livelihood. They provide important ecosystem services and goods. However there are many threats that interfere with the normal functioning and development of forest ecosystems. The influence of climate change processes on functioning and dynamics of forests in Kazakhstan is understudied, and is observed in the relatively small number of dendroclimatic studies. Additional limitations in understanding of climate-growth relationships arise from insufficient spatial and temporal coverage of those studies and the absence of a qualitative analysis of already available information for identification of common patterns and differences in the impacts of tree growth on climate. The main research goal of this thesis was accordingly to address these limitations. An analysis of all available publications on dendroclimatic studies in Kazakhstan was performed. This analysis not only summarized the available information on climate-growth relationships, but also provided certain information about existed research gaps. Some of those gaps were addressed by an investigation of variation of climate-growth relationships of Schrenk spruce in the Ile River basin. The study revealed certain differences in reaction of earlywood, latewood and total ring width on climate in four mountain ranges of the Ile River basin. The strong influence of climate change was observed in a decreasing temperature signal around the 1970s. Revealed periodicities in the fluctuation of tree-ring indices suggested an influence of certain climate indices such as North Atlantic Oscillation and the Tropospheric Biennial Oscillations. Collectively, the analysis in this dissertation contributes to development of the tree-ting network of Kazakhstan. Obtained information should help in assessment of possible impacts of climate change on Schrenk spruce forests in Kazakhstan.

FundaÃÃo Cearense de Apoio ao Desenvolvimento Cientifico e TecnolÃgico
Cowpea is an important legume of the Northeast of Brazil and its yield is affected by Colletotrichum fungi. These fungi are known to occur on and cause disease anthracnose on the broad range of plants. In addition, the rapid and transient accumulation of reactive oxygen species, termed âoxidative burstâ, is one of the earliest observable reaction of plant cells to microbial infection. As plant H2O2 is believed to have direct antimicrobial effects on pathogens it was raised the hypothesis that alteration of H2O2 concentrations by pharmacological compounds could confer differences in susceptibility of the cowpea genotype BR-3-Tracuateua to C. gloeosporioides attack. Thus, seeds were germinated on humid Germitest paper and after four days seedlings were transferred to hydroponic solution. Eight days later, the primary leaves were excised, infiltrated with glucose oxidase (GO+G), salicylic acid (AS), catalase (CAT) and diphenyleneiodonium chloride (DPI) transferred to Petri dishes and inoculated with 2.0 x 105 spores mL-1 fungal suspension on the adaxial surface. After, leaves were placed in darkness and collected at 12, 24, 48 and 72 hours. By GO+G and SA leaf treatments showed 144.96 and 186.05 nmol H2O2 g-1 fresh mass (FM), respectively, and the fungus presented a subcuticular, intramural necrotrophic strategy, forming secondary hyphae associated with a quick spread and a rapid killing of the host cells. On the leaves treated with SA, it was observed lipid peroxidation and the absence of melanized appresoria. However, CAT and DPI treatment leaves presented 55.50 nmol H2O2 g-1 FM and the fungus showed hemibiotrophic infection-type, with globular vesicles and primary and secondary hyphae formation. All results suggest that despite H2O2 influenced directly the fungal infection process-type it does not confer resistance of cowpea to C. gloeosporioides.
FeijÃo-de-corda à um importante legume presente na regiÃo Nordeste do Brasil e sua produÃÃo à afetada por fungos do gÃnero Colletotrichum. Estes fungos sÃo conhecidos por causar antracnose em uma grande variedade de plantas. AlÃm disso, o acÃmulo rÃpido e transiente de espÃcies reativas de oxigÃnio, nomeado de âoxidative burstâ, à uma das primeiras reaÃÃes observÃveis em cÃlulas de plantas diante infecÃÃo microbiana. Como H2O2 produzido por plantas à considerado por apresentar efeitos antimicrobianos diretos, foi levantada a hipÃtese que alteraÃÃes nas concentraÃÃes de H2O2 por substÃncias farmacolÃgicas poderiam conferir diferenÃas na susceptibilidade do genÃtipo de feijÃo-de-corda, BR-3-Tracuateua, ao ataque do fungo C. gloeosporioides. Deste modo, sementes foram germinadas em papÃis Germistest umedecidos e, depois de 4 dias, plÃntulas foram transferidas para soluÃÃo hidropÃnica. Passados oito dias, as folhas primÃrias foram cortadas, infiltradas com glucose oxidase (GO+G), Ãcido salicÃlico (AS), catalase (CAT) e cloreto de difenilenoiodÃnio (DPI), transferidas para placas de Petri e inoculadas com suspensÃo do fungo ajustada para concentraÃÃo de 2,0 x 105 esporos mL-1. Em seguida, as folhas foram mantidas no escuro e foram coletadas em 12, 24, 48 e 72 horas. Folhas tratadas com GO+G e AS mostraram 144,96 e 186,05 nmol H2O2 g-1 de massa fresca (MF), respectivamente, e o fungo apresentou estratÃgia subcuticular, intramural necrotrÃfica, formando hifas secundÃrias associadas com Ãgil crescimento e rÃpida morte das cÃlulas hospedeiras. Nas folhas tratadas com AS, foi observada peroxidaÃÃo de lipÃdios e ausÃncia de apressÃrios melanizados. Contudo, folhas tratadas com CAT e DPI apresentaram 55,50 nmol H2O2 g-1 MF e, o fungo, mostrou modo de infecÃÃo hemibiotrÃfico, com vesÃculas globulares e formaÃÃo de hifas primÃrias e secundÃrias. Todos os resultados sugerem que, apesar do H2O2 tenha influenciado diretamente o modo de infecÃÃo do fungo, este nÃo conferiu resistÃncia no feijÃo-de-corda ao C. gloeosporioides.

Feijão-de-corda é um importante legume presente na região Nordeste do Brasil e sua produção é afetada por fungos do gênero Colletotrichum. Estes fungos são conhecidos por causar antracnose em uma grande variedade de plantas. Além disso, o acúmulo rápido e transiente de espécies reativas de oxigênio, nomeado de ‘oxidative burst’, é uma das primeiras reações observáveis em células de plantas diante infecção microbiana. Como H2O2 produzido por plantas é considerado por apresentar efeitos antimicrobianos diretos, foi levantada a hipótese que alterações nas concentrações de H2O2 por substâncias farmacológicas poderiam conferir diferenças na susceptibilidade do genótipo de feijão-de-corda, BR-3-Tracuateua, ao ataque do fungo C. gloeosporioides. Todos os resultados sugerem que, apesar do H2O2 tenha influenciado diretamente o modo de infecção do fungo, este não conferiu resistência no feijão-de-corda ao C. gloeosporioides.

The thesis deals with electronic communication through data mailboxes. It provides the legal framework that regulates this kind of communication and how to work with data mailboxes as a business entity. The theoretical part is about issues and defining basics concepts, legal framework and ways how to work with the data mailboxes. The practical part is devoted to the company, which uses the data mailboxes to comunicate with authorities and also with clients. Based on consultation with users is evaluated what pros and cons brought them this news and also is quantified their cost savings, which are associated with communication through data mail boxes.

The main topic of the diploma thesis is the development of data boxes in the Czech Republic from the very beginning to its current valid legislation. It focuses on the law No. 300/2008 on electronic operations and authorized conversion of documents. This law has integrated the data boxes into the Czech rule of law. Besides, attention was given to the main terms related to the electronic communication via data boxes (data message, data box, establishment of a data box, types of data boxes). Furthermore, in the thesis is also mentioned the project of data boxes at the town hall for the region of Prague 13. The possibility of submission a request on establishment of a data box for individuals has also been discussed. In the conclusion were given remarks and recommendations related to the issues.

The thesis deals with project data boxes, which was launched in 2009. The aim of this project is electronic communication between public administration, local government and natural or legal persons. Data exchange system is one of the basic elements of e-government is used to send and receive data messages. Thanks to the legal framework of the data report elevated to the same level as registered mail with legally essential effects. The main part of the thesis, excluding the current legislation, the analysis of the recoverability of data boxes at Moore Stephens Ltd. and the proposal of appropriate software solutions for optimal utilization of the data mailbox and archive data messages. We were also examined possible system errors and any suggestions for improvement.

This master's thesis deals with the use of text classification for sorting of incoming emails. First, there is described the Knowledge Discovery in Databases and there is also analyzed in detail the text classification with selected methods. Further, this thesis describes the email communication and SMTP, POP3 and IMAP protocols. The next part contains design of the system that classifies incoming emails and there are also described realated technologie ie Apache James Server, PostgreSQL and RapidMiner. Further, there is described the implementation of all necessary components. The last part contains an experiments with email server using Enron Dataset.
