Abstract : Recently, there is a great interest to study the flow characteristics of suspensions in different environmental and industrial applications, such as snow avalanches, debris flows, hydrotransport systems, and material casting processes. Regarding rheological aspects, the majority of these suspensions, such as fresh concrete, behave mostly as non-Newtonian fluids. Concrete is the most widely used construction material in the world. Due to the limitations that exist in terms of workability and formwork filling abilities of normal concrete, a new class of concrete that is able to flow under its own weight, especially through narrow gaps in the congested areas of the formwork was developed. Accordingly, self-consolidating concrete (SCC) is a novel construction material that is gaining market acceptance in various applications. Higher fluidity characteristics of SCC enable it to be used in a number of special applications, such as densely reinforced sections. However, higher flowability of SCC makes it more sensitive to segregation of coarse particles during flow (i.e., dynamic segregation) and thereafter at rest (i.e., static segregation). Dynamic segregation can increase when SCC flows over a long distance or in the presence of obstacles. Therefore, there is always a need to establish a trade-off between the flowability, passing ability, and stability properties of SCC suspensions. This should be taken into consideration to design the casting process and the mixture proportioning of SCC. This is called “workability design” of SCC. An efficient and non-expensive workability design approach consists of the prediction and optimization of the workability of the concrete mixtures for the selected construction processes, such as transportation, pumping, casting, compaction, and finishing. Indeed, the mixture proportioning of SCC should ensure the construction quality demands, such as demanded levels of flowability, passing ability, filling ability, and stability (dynamic and static). This is necessary to develop some theoretical tools to assess under what conditions the construction quality demands are satisfied. Accordingly, this thesis is dedicated to carry out analytical and numerical simulations to predict flow performance of SCC under different casting processes, such as pumping and tremie applications, or casting using buckets. The L-Box and T-Box set-ups can evaluate flow performance properties of SCC (e.g., flowability, passing ability, filling ability, shear-induced and gravitational dynamic segregation) in casting process of wall and beam elements. The specific objective of the study consists of relating numerical results of flow simulation of SCC in L-Box and T-Box test set-ups, reported in this thesis, to the flow performance properties of SCC during casting. Accordingly, the SCC is modeled as a heterogeneous material. Furthermore, an analytical model is proposed to predict flow performance of SCC in L-Box set-up using the Dam Break Theory. On the other hand, results of the numerical simulation of SCC casting in a reinforced beam are verified by experimental free surface profiles. The results of numerical simulations of SCC casting (modeled as a single homogeneous fluid), are used to determine the critical zones corresponding to the higher risks of segregation and blocking. The effects of rheological parameters, density, particle contents, distribution of reinforcing bars, and particle-bar interactions on flow performance of SCC are evaluated using CFD simulations of SCC flow in L-Box and T-box test set-ups (modeled as a heterogeneous material). Two new approaches are proposed to classify the SCC mixtures based on filling ability and performability properties, as a contribution of flowability, passing ability, and dynamic stability of SCC.<br>Résumé : Récemment, il y a un grand intérêt à étudier les caractéristiques d'écoulement des suspensions dans différentes applications environnementales et industrielles, telles que les avalanches des neiges, les coulées de débris, les systèmes de transport et les processus d’écoulement des matériaux. En ce qui concerne les aspects rhéologiques, la plupart des suspensions, comme le béton frais, se comportent comme un fluide non-Newtonien. Le béton est le matériau de construction le plus largement utilisé dans le monde. En raison de limites qui caractérisent le béton normal en termes de maniabilité et de capacité de remplissage de coffrage, il était nécessaire de développer une nouvelle classe de béton qui peut couler sous son propre poids, en particulier à travers les zones congestionnées du coffrage. Par conséquent, le béton autoplaçant (BAP) est un nouveau matériau de construction qui est de plus en plus utilisé dans les différentes applications. Étant donné sa fluidité élevée de BAP peut être utilisé dans certaines applications particulières, notamment dans la section densément renforcée. Cependant, la fluidité élevée rend le béton plus sensible à la ségrégation des gros granulats pendant l'écoulement (la ségrégation dynamique) et ensuite au repos (ségrégation statique). La ségrégation dynamique peut augmenter lorsque le BAP est coulé sur une longue distance ou en présence d'obstacles. Par conséquent, il est toujours nécessaire d'établir un compromis entre la fluidité, la capacité de passage, et la stabilité du BAP. Ceci doit être pris en considération afin de concevoir le processus de coulée et dosage des mélanges du BAP. Ceci est appelé la conception d'ouvrabilité du BAP. Une conception de maniabilité efficace et non coûteuse peut être achevée à travers la e prévision et l'optimisation de l'ouvrabilité des mélanges de béton pour les procédés de construction sélectionnés, notamment le transport, le pompage, la mise en place, le compactage, la finition, etc. En effet, les formulations de mélange doivent se confirmer à la qualité de la construction demandée, par exemple les niveaux exigés de fluidité, la capacité de passage, la capacité de remplissage, et la stabilité (statique et dynamique). Celui est nécessaire pour développer des outils théoriques afin d’évaluer dans quelles conditions les exigences de qualité de la construction sont satisfaites. Cette thèse est consacrée à la réaliser des simulations analytiques et numériques pour prédire la performance d'écoulement du BAP dans différents procédés de la mise en place du béton. L'objectif spécifique de cette étude consiste à simuler l'écoulement du BAP dans essais empiriques, notamment la boite en L et la boite en T pour évaluer la performance du BAP pendent la mise en place (la fluidité, la capacité de passage, la capacité de remplissage, et la ségrégation dynamique induite par cisaillement ou par gravité). Par conséquent, le BAP est modélisé comme matériau hétérogène. En outre, un modèle analytique est proposé pour prédire la performance à l'écoulement du BAP dans la boite en L en utilisant la théorie de Dam Break. D'autre part, les résultats des simulations numériques de l’écoulement du BAP dans une poutre renforcée sont comparés aux résultats expérimentaux par des profils de surface libres. Les résultats des simulations numériques de BAP coulée (modélisée comme un fluide homogène unique), sont utilisés pour déterminer les zones critiques correspondant à des risques plus élevés de ségrégation et de blocage. Les effets des paramètres rhéologiques, la masse volumique, le contenu des particules, la distribution de barres d'armature, et les interactions particule-barres sur les performances d'écoulement du BAP sont évaluées à l'aide de simulations MFN d’écoulement du BAP par les essais des L-Box et T-box (modélisée comme une matériau hétérogène). Deux nouvelles approches sont proposées pour classifier les mélanges du BAP sur la base de la capacité de remplissage, et les propriétés de performabilité, en fonction de la fluidité, la capacité de passage et de la stabilité dynamique du BAP.

O presente estudo teve por finalidade avaliar os efeitos da contagem de células somáticas (CCS) no leite cru (nos níveis de 100-200.000, 400-500.000 e >800.000 céls./mL) sobre as características físicas, químicas, microbiológicas, sensoriais e funcionais dos queijos Minas Frescal e tipo Mussarela. Utilizou-se um delineamento experimental em parcelas subdivididas em blocos, considerando-se a contagem de células somáticas como efeito principal, os dias de análise como subparcelas e os processamentos como blocos. Cada tipo de leite foi obtido da ordenha de animais previamente selecionados de acordo com o nível de células somáticas. As etapas de elaboração dos queijos incluíram a pasteurização do leite (65º C, 30 minutos), adição de cloreto de cálcio, fermento (para a Mussarela) e coalho, coagulação e obtenção do coágulo, dessoragem, salga na massa, filagem, moldagem, salga na salmoura (para a Mussarela), e embalagem dos produtos. Os queijos foram mantidos em B.O.D. a 4 ºC e avaliados nos dias 2, 9, 16, 23 e 30 após a fabricação. A seqüência de elaboração dos queijos Minas e tipo Mussarela foi repetida 5 e 3 vezes, respectivamente, para cada tratamento. As análises realizadas nos queijos foram: pH, acidez, percentuais de gordura, matéria seca (MS), cinzas, nitrogênio total (NT), nitrogênio não protéico (NNP), nitrogênio não caseinoso (NNC), índice de proteólise, ácidos graxos livres (AGL), textura, avaliação sensorial, propriedades funcionais (capacidade de derretimento a 107 ºC e percentual de óleo livre - somente no queijo tipo Mussarela), contagem de mesófilos, psicrotróficos e número mais provável a 35 e 45 ºC. No queijo Minas Frescal, não houve interação entre CCS e dias de armazenamentos nas avaliações físico-químicas, microbiológicas, funcionais e índice de lipólise, porém houve efeito significativo (P<0,05) para índices de proteólise e profundidade da proteólise. Já para mesófilos, psicrotróficos, acidez, matéria seca, firmeza e ácidos graxos livres houve efeito significativo (P<0,05) ao avaliar dias de armazenamento. A avaliação sensorial, no primeiro dia de análise, não apresentou diferença significativa nos atributos, porém o queijo de alta CCS (> 800.000 céls./mL) diferiu dos demais no dia 30, em que recebeu menor nota em todas as características. No queijo tipo Mussarela, houve interação entre CCS e dias de armazenamento apenas para a capacidade de derretimento, resultando em aumento do derretimento no decorrer do tempo. Para as características de pH, ácidos graxos livres, índices de proteólise, extensão e profundidade da proteólise, mastigabilidade e elasticidade houve efeito significativo (P<0,05) para dias de armazenamento. Entre os atributos avaliados na sensorial, apenas a aparência apresentou diferença entre tratamentos. O rendimento dos queijos, Minas frescal e tipo Mussarela, não foram influenciados pela quantidade de células somáticas dos leites. Os resultados do trabalho indicaram que o leite destinado à fabricação dos queijos Minas e tipo Mussarela deve apresentar CCS até 400-500.000 céls./mL, de maneira a evitar alterações na qualidade dos produtos ao longo do período de armazenagem.<br>The aim of the present study was to evaluate the effect of somatic cells counts (SCC) in raw milk (at levels of 100-200,000, 400-500,000 and 800,000 cells./mL) on physical, chemical, microbiological, sensorial and functional characteristics of Minas frescal and Mozzarella type cheeses. A completely randomized block design was used, considering SCC as the main effect, the days of analysis as sub parcels and the processing batches as the blocks. Each type of milk was obtained from cows previously selected according to its individual SCC. The manufacture of cheeses included: pasteurization of milk (65ºC, 30 minutes), addition of calcium chloride, starter culture (for Mozzarella) and rennet, coagulation and separation of the curd, whey drainage, salting (for Minas cheese), stretching of the curd, kneading and salting in brine (for Mozzarella), and packing the products. The cheeses were stored in a B.O.D. oven at 4ºC and evaluated on days 2, 9, 16 23 and 30 after the manufacture. The sequence of elaboration of the Minas frescal and Mozzarella cheeses was repeated 5 and 3 times, respectively, for each treatment. The analyses carried out in the cheeses were: pH, acidity, percentages of fat, dry matter (DM), ashes, total nitrogen (NT), non-protein-nitrogen (NPN), non-casein nitrogen (NCN), proteolysis, free fatty acids (FFA), texture, sensorial evaluation, functional properties (melting capacity the 107 ºC and percentage of free oil - only in the cheese Mozzarella), mesophile and psychrotrophic counts and the most probable number at 30 and 45ºC. For Minas frescal cheese, no interaction was found between SCC and days of storage when considering the data on chemical, physical, microbiological, functional and lipolysis index. However, a significant effect (P<0.05) was observed for proteolysis and depth of proteolysis. For mesophiles, psychrotrophics, acidity, dry matter, firmness and free fatty acids, there was a significant effect (P<0.05) for days of storage. The sensorial evaluation made on the first day of analysis did not present significant differences for all attributes. However, the Minas cheese made with high SCC (800,000 cells/mL) differed on day 30, when it received worse grades for all the attributes. For the Mozzarella cheese, an interaction between SCC and days of storage was observed only for the melting capacity, which resulted in an increment of the melting along the time of storage. The parameters of pH, free fatty acid, proteolysis, extension and depth of proteolysis, springiness and elasticity had significant effect (P<0.05) for days of storage. Amongst the attributes evaluated in the sensorial, only the appearance presented difference between treatments. The yield of Minas frescal and Mozzarella cheese was not influenced by the amount of somatic cells in the original milk. Results indicated that milk used for the manufacture of Minas frescal and Mozzarella cheeses should present SCC up to 400-500,000 cells/mL, in order to avoid quality changes in those products during storage.

Bei dem Prozess einer Wasserstoffinduzierten Spannungsrisskorrosion (H-SpRK) handelt es sich um einen zeitabhängigen Vorgang, der zu einer Reduzierung der Duktilität und Widerstandskraft des Spannstahls führt und daraus resultierend ein sprödes und schlagartiges Versagen eines Spannbetonbauwerkes zur Folge haben kann. Der Prozess selbst und insbesondere auch die ihn beeinflussenden Parameter sind für die im Bauwesen verwendeten hochfesten Spannstähle älterer Produktion weitestgehend unerforscht. Die Relevanz für bestehende Bauwerke ist jedoch nicht zuletzt durch einzelne dokumentierte und untersuchte Schadensfälle nachgewiesen.
Ziel der vorliegenden Arbeit war es, die zur Verfügung stehenden Bauwerksuntersuchungen aus den zurückliegenden ca. 10 Jahren statistisch zu analysieren und auszuwerten. Auf dieser Basis war ein Berechnungsmodell wahrscheinlichkeitstheoretischer Basis zu entwickeln, mit dem die Gefahr eines spröden Bauwerksversagens für das Gesamttragwerk beurteilt und hinsichtlich der Auswirkungen auf das einzuhaltende Sicherheitsniveau bewertet werden kann.
Es wurden insgesamt 31 Bauwerksuntersuchungen statistisch ausgewertet und beurteilt. Die zur Verfügung stehenden Daten wurden analysiert und hinsichtlich der Prüfqualität sowie der Quantität der Proben bewertet. Dabei war festzustellen, dass aufgrund fehlender konkreter Vorgaben im Regelwerk eine sehr heterogene Datenbasis vorliegt. Nicht alle Untersuchungsergebnisse konnten in die weitere Auswertung einbezogen werden. Die in ausreichender Datenqualität und Datenumfang geeigneten Untersuchungen wurden hinsichtlich ihrer Relevanz für den Prozess einer H-SpRK analysiert und die Auswirkungen einzelner Parameter bewertet.
Im Ergebnis der materialtechnischen Untersuchungen und statistischen Auswertung der Bauwerksuntersuchungen wurde ein Berechnungsmodell vorgestellt, das den gleichzeitigen Ausfall von Spannstahl in allen Bereichen des Bauwerkes berücksichtigt. Zusätzlich zum Standardverfahren des beschriebenen Vorgehens wird das Modell um den Ansatz eines korrelierten Spannstahlausfalls erweitert. Außerdem wird für Bauwerke mit einem statisch unbestimmten Anteil der Vorspannung sowie für Konstruktionen mit gestaffelter Spannstahlbewehrung die Anwendung des Verfahrens konkretisiert.
Neben der Erstellung des Berechnungsmodells wurden Vorschläge zu Vorgaben für die Bauwerksprüfung vorgestellt. Dazu zählen insbesondere die Festlegung von Prüfintervallen sowie einheitliche Vorgaben zu den verwendeten Prüfmethoden und -verfahren. Bezüglich der Festlegung von Mindestumfängen von Proben wird zwischen bestehenden und auch weiter zu nutzenden Bauwerken einerseits sowie Probennahmen im Rahmen von Rückbaumaßnahmen unterschieden.

Chez les eucaryotes, la voie de protéolyse Ub/ protéasome 26S est responsable de la dégradation sélective de la plupart des protéines intracellulaires. Cette dégradation par le protéasome 26S est initiée par une polyubiquitination de la protéine réalisée grâce à l’action d’une cascade enzymatique impliquant 3 types d'enzymes nommées « ubiquitin-activating enzyme » (E1), « ubiquitin-conjugating enzyme » (E2) et « ubiquitin-protein ligase » (E3). Il existe différentes classes d’ubiquitines ligases (E3), parmi lesquelles la plus connue est le complexe SCF (Skp1-Cullin-F-box). La protéine SKP1 fixe à la fois la Culline et la F-box qui va reconnaitre spécifiquement la protéine cible. Contrairement aux protistes, les champignons et certains vertébrés qui possèdent un unique gène SKP1 fonctionnel, de nombreux animaux et espèces de plantes présentent plusieurs SKP1 homologues. Vingt et un et trente deux gènes SKP1 ont été décrits respectivement chez Arabidopsis thaliana et Oryza sativa. En dépit de l’importance du complexe SCF, chez le riz, peu de travaux décrivent les interactions entre les dizaines de protéines « SKP1-like » et les centaines de protéines F-box. Dans un premier temps, nous avons collecté et analysé les séquences de 288 gènes « SKP1-like » appartenant à 17 espèces, dont la mousse Physcomitrella patens, cinq monocotylédones et 11 eudicotylédones. Les analyses structurales et phylogénétiques de ces gènes indiquent qu’ils peuvent être divisés en différentes sous-familles. Nos analyses ont montré qu’OSK1 et OSK20 chez le riz constituent une classe de gènes SKP1 à intron unique conservé. Dans un deuxième temps, nous avons étudié le profil d’expression des gènes « SKP1-like » chez le riz. Notre investigation sur le nombre d’EST a montré que les gènes OSK1 et OSK20 sont les plus largement représentés dans les bases de données EST publiques. La méta-analyse de l’expression des gènes « SKP1-like » chez le riz, indique que les gènes OSK présentent des profils d'expression hétérogènes selon les tissus et les conditions physiologiques. Les résultats des intearctions protéine-protéine en double hybride ont révélé que les protéines OSK présentent différentes capacités d’interactions avec les protéines F-box. Cependant, OSK1 et OSK20 semblent interagir avec la plupart des protéines F-box testées. Les études de localisation subcellulaire ont indiqué que OSK1 et OSK20 sont des protéines nucléaires et cytosoliques. En se basant sur les divers résultats obtenus dans ce travail, nous pouvons suggérer que chez le riz, les gènes OSK1 et OSK20 sont fonctionnellement équivalents aux gènes ASK1 et ASK2 chez Arabidopsis thaliana. Nous pouvons également proposer les équivalents de ces gènes chez les autres espèces végétales dont le génome a été séquencé<br>In eukaryotes, the ubiquitin Ub/26S proteasome pathway is responsible for the selective degradation of most intracellular proteins. This cellular process is initiated by protein polyubiquitination mediated by a three-step cascade involving: an ubiquitin-activating enzyme (E1), an ubiquitin-conjugating enzyme (E2) and an ubiquitin-protein ligase (E3). The E3 ubiquitin ligases contain several classes, among which the best-known are Skp1-Cullin-F-box (SCF) complexes. The SKP1 protein binds both Cullin and F-box which recognizes specifically the target proteins. Whereas protists, fungi and some vertebrates have a single functional SKP1 gene, many animal and plant species possess multiple SKP1 homologues. Twenty one and thirty-two SKP1-related genes have been described respectively in the Arabidopsis and Oryza sativa genome. Despite the importance of the SCF complex, there have been a few reports of systematic surveys of interactions between the dozens of SKP1-like proteins and the hundreds of F-box proteins in rice. In a first step, we retrieved and analyzed 288 SKP1-like genes belonging to 17 species including the moss Physcomitrella patens, five monocots and 11 eudicots. Structural and phylogenetic analysis of rice OSK genes and other plant SKP1-like genes have indicated that the different members of the plant SKP1 can be split into different subfamily. Our analyses indicated that OSK1 and OSK20 belong to a class of SKP1 genes that contain one intron at a conserved position. In a second step, we studied expression profiles of the rice Skp1-like genes. Our EST survey indicated that OSK1 and OSK20 are the most widely represented genes in public EST databases. Meta-analysis of the expression of rice SKP1-like genes indicated that OSK genes exhibit an expression profile that was heterogeneous in terms of tissues, conditions and overall intensity. Yeast two-hybrid results revealed that OSK proteins display a differing ability to interact with F-box proteins. However, OSK1 and OSK20 seemed to interact with most F-box proteins tested. Subcellular localization studies indicated that OSK1 and OSK20 are nuclear and cytosolic proteins. Based on the results obtained in this study, we can suggest that rice OSK1 and OSK20 are likely to have similar functions as do the Arabidopsis ASK1 and ASK2 genes. Similarly, we suggest a list of functional equivalent in the other sequenced plant genomes

The Society for Conservation Biology held their 21st annual conference in Port Elizabeth during July 2007. This was the first SCB conference hosted on the African continent. Over 1500 conservation professionals and students from throughout the world were brought together by the SCB annual meeting. The local organizing committee was interested in the economic impacts associated with the conference and approached the NMMU Economics Department to conduct an economic impact assessment of the conference. The Economics Department invited the researcher to undertake this study towards obtaining an MCom degree in Economics. The economic impact of conferences stems from two sources, namely: expenditure by delegates (the demand-side) and the expenditure by conference organizers (the supply-side). The study focused on the economic impacts of the conference for the Eastern Cape. The conference expenditure produced an increase in demand in the Eastern Cape. Only new expenditure originating from outside the Eastern Cape were considered. An increase in the demand for one industry’s output will create additional demand for the outputs of its supplying industries, because industries are connected through forward and backward linkages. These inter-industry linkages produce a multiplier effect. The initial direct conference expenditure created secondary impacts. The latter were indirect and induced expenditures. In addition to secondary impacts, the SCB conference produced spill-over impacts. The spill-over impacts of the conference were noted (but not quantified). The expenditure by delegates was determined by means of a delegate expenditure questionnaire conducted during the conference. The expenditure by the conference organizers was determined in consultation with the organizers, using their financial statements. The multiplier impacts were estimated by means of an input-output (IO) analysis, using a Social Accounting Matrix (SAM) of the Eastern Cape as the underlying database. These multipliers (Type 1 and 2 multipliers) were estimated using open and closed multiplier models. It was deduced that the 2007 SCB conference created a significant and positive net economic impact in the Eastern Cape. The total direct cash injection of the conference was R12.141 million. Using a Type 1 multiplier this direct stimulus is estimated to have caused an extra R16.502 million increase in gross output. Using a Type 2 multiplier this direct stimulus was estimated to have caused a R19.884 million increase in gross output. The total cash injection of the conference contributed R6.093 million to GVA in terms of a Type 1 multiplier and a R7.344 million increase in GVA in terms of a Type 2 multiplier. Household incomes in the Eastern Cape were increased by R3.384 million. These results confirm that the hosting of major events and conferences is indeed a tool for promoting regional economic development.

<p>One source of input data for software evolution research is data stored inside a software configuration management repository. The data includes different versions of a software system’s source code as well as version history metadata, such as check-in dates or log messages. Inherently, extracting this data manually is a time- and labor intensive task. The subsequent preprocessing step and the appropriate storage of the results, necessary to utilize the data for further analysis, is an additional effort for the researcher.</p><p>The goal of this thesis is to design and implement a front-end plug-in for an existing software comprehension tool, the VizzAnalyzer, providing the capability to extract and analyze multiple versions and evolutional information of software systems from SCM repositories and to store the results. Thereby, the implemented solution provides the infrastructure for software evolution research.</p>

The <i>SCL</i> gene is a critical haematopoietic transcription factor, with a pattern of expression that is highly conserved from mammals to teleost fish. Its central role in haematopoiesis and its complex expression pattern make it a model gene for studying transcriptional networks. The full set of <i>cis</i>-regulatory elements active in the <i>SCL</i> locus is probably still to be discovered, and will be required for a more complete understanding of its transcriptional regulation. <i>SCL</i> has also been a model gene for studying comparative sequence analysis. This has been exploited in this thesis to approach two questions. Firstly: how can different species be utilized and combined to improve the accuracy of comparative sequence analysis? Secondly: what previously undiscovered <i>cis</i>-regulatory elements are active in the <i>SCL</i> locus, and how do they act? BAC clones containing the <i>SCL</i> loci of dog and platypus as well as the paralogous <i>LYL1</i> locus of a marsupial, the stripe-faced dunnart, were isolated and sequenced and the <i>SCL</i> locus in rat was annotated. Human/dunnart sequence comparisons appeared to be more selective than human/mouse comparisons in the <i>LYL1</i> locus, but still permitted the identification of all putative promoters within the clone. Phylogenetic footprinting determine five transcription factor binding sites within the <i>LYL1</i> promoter, with a pattern very reminiscent of those seen in critical <i>SCL</i> regulatory elements. Functional analyses confirmed <i>LYL1</i> promoter activity and nuclear binding. Comparisons of two-, three-, and four-way eutherian alignments at the <i>SCL</i> locus revealed that the four-way alignment was the most sensitive and specific for identifying both regulatory regions and transcription factor binding sites. Adding platypus sequence vastly improved specificity, but at the expense of sensitivity. Four candidate regulatory regions was identified, three of which were then shown to act as lineage-restricted regulatory elements, probably affecting transcription of the co-ordinately expressed downstream gene, <i>MAP17</i>. It is concluded that to maximize sensitivity and specificity in the identification of eutherian regulatory regions, sequences from multiple eutherian species should be combined for comparative analysis.

Under the unifying theme of supply chain learning, this three essay dissertation extends scholarship by investigating these multi-tier relationships. Theory is emerged, grounded in data, gathered from functioning supply chains in an effort to provide scholars and practitioners with an increased understanding of the SCL phenomena. Essay 1, entitled "Supply Chain Learning: An Exploratory Literature Review" examines the current literature in an attempt to address the shortcomings and emerge areas that have been less explored and less understood. By exposing these areas of research opportunities using a grounded theory methodology, a framework was emerged allowing identification of the limitations of extant literature and providing a springboard for future research. This framework also allowed further investigation into the SCL processes and expansion of the current understanding by providing academia with a comprehensive review of the literature and revealing the shortcomings that exist related to SCL.
Using the framework emerged in Essay 1, Essay 2 entitled "Toward Supply Chain Learning: A Focus on the Customers of Logistics Service Providers" explores the rationalization and cognitive processes of senior level executives of firms utilizing national or global supply chains. These respondents are directly engaged in creating, establishing and operating relationships with third party logistics (3PL) providers within a functioning supply chain. By examining the relationships and processes from the point of view of customers of third party logistics providers, a unique perspective provides insight into these relationships. Using semi-structured interviews with these executives, grounded theory was once again used to emerge theory explaining the phenomena of SCL. In particular, this research examines the elements studied from the perspective of customers of third party logistics providers as they seek to develop new processes and solutions in hopes of obtaining a competitive advantage by adaptive learning with the help of their providers and trading partners. In addition, this research increases our understanding of SCL by examining a relationship between customers and 3PL providers, their experiences and outcomes.
Essay 3, entitled "Practical Application of Supply Chain Learning" focuses on the implications of the learning relationship and its impact on the practitioner. By providing the findings of the research in a context relatable to practitioners, this culmination of findings allows practitioners to relate the findings directly to their existing supply chains. Realizing that many supply chains are relationship driven, this research focuses on the findings of previous research to provide a more holistic view of the learning relationship process as it exists in multiple tiers of their existing supply chain. Providing a step-by-step explanation of the SCL process as emerged from previous research, executives are provided a tool to better identify, analyze and understand these processes as relatable in their existing environment.
As a methodical analysis of the IOL process, these essays provide the foundation for understanding the relationship process that exists between learning partners in a supply chain. Essay 1 provides basis for theory development by examining current literature and exposing the shortcomings while also emerging a preliminary framework on which to build future research. Essay 2 follows up on these deficiencies and attempts to saturate understanding of the IOL process, particularly from the point of view of 3PL customers in an existing supply chain. Essay 3 delivers these findings to executives in a relatable format, providing a holistic understanding of the phenomena. In summation, this dissertation provides theory emerged from data, explaining the learning relationship from the point of view of the customers of 3PL services, the cognitive dimensions and outcomes of these decisions as they relate to learning in the supply chain.

On December 11, 2008, the financial world was in a panic as the United States Securities and Exchange Commission (SEC) announced the arrest of Bernard L. Madoff of Bernard L. Madoff Investment Securities, LLC, for orchestrating a $65 billion Ponzi scheme. An investigation took place into Madoff's practices, and as a result, it was revealed the SEC failed to catch Madoff years earlier as a result of its business practices. After this became known, the SEC faced reputational harm. This qualitative analysis seeks to discover through identification and analysis of themes and sub-themes of response strategies, the extent to which the SEC applied Situational Crisis Communication Theory (SCCT) in its crisis response, in order to examine SCCT's merit as a theory in government crisis communication research. This study also offers additional response strategies imposed by the SEC to suggest further expansion of SCCT in a government context.<br>Master of Arts<br>On December 11, 2008, Bernard L. This thesis describes the results of two process system engineering studies: (1) hydrogen pinch analysis; and (2) application of COSMO-SAC (conductor-like screening model â segment activity coefficient) to electrolytes. Part (1) presents an automated spreadsheet method that can quickly help minimize fresh hydrogen consumption and maximize hydrogen recovery and reuse in petroleum refineries and petrochemical complexes. Part (1) has appeared as a featured article on engineering practice in the Chemical Engineering Magazine, volume 115, pp. 56-61, June 2008. We present an automated spreadsheet on our research group website (www.design.che.vt.edu) and describe procedures for using the spreadsheet in this thesis. Part (2) discusses the application of the conductor-like screening model â segment activity coefficient (COSMO-SAC), a liquid-phase activity-coefficient model, to electrolytes. We offer detailed procedure for obtaining sigma profiles for electrolytes. A sigma profile is a molecular-specific probability distribution of the surface-charge density, which enables the application of solvation-thermodynamic models to predict vapor-liquid and solid-liquid equilibria, and other properties. We propose to add an additional term to the exchange energy to account for ion-ion attractive and repulsive forces. We also look at the resulting exchange energy behavior. Although accurate numerical results are not achieved, we are able to produce results that match literature data by adding an adjustment factor.<br>Master of Science

The suprachiasmatic nucleus (SCN) of the hypothalamus is the master mammalian pacemaker, co-ordinating the multitude of cell-autonomous circadian oscillators across the body to ensure internal synchrony, as well as maintaining an adaptive phase relationship with the light-dark cycle via projections from the retina. Intercellular communication between SCN clock neurons synchronises their oscillations, resulting in coherent output signals to the periphery. Vasoactive intestinal peptide (VIP), a neuropeptide expressed in the retinorecipient ventrolateral region of the SCN, is vital to this circuit-level co-ordination by signalling to its cognate VPAC2 receptor. In addition, VIP is important for the integration of light input into the SCN oscillation. The aims of the work presented in this thesis were to determine the roles of the VIP and VPAC2 cells in controlling circadian rhythmicity, and to elucidate the mechanisms of VIP signalling that underpin these roles. The first two experimental chapters utilise intersectional genetics and viral transduction to address separable roles for the VIP and VPAC2 cell populations. By diphtheria toxin-mediated cell ablation, or by adjusting cell-autonomous periodicity or rhythmicity specifically in these cell populations, I have identified that the VPAC2 cells are important for period setting and rhythmicity of both the SCN ex vivo and mouse behaviour in vivo, while the VIP cells play a vital role in behavioural rhythmicity and phase coherence across the SCN. The next two chapters use application of VIP to SCN slices to address mechanisms of phase-resetting through pharmacological manipulation and microarray analysis. I find that VIP has long lasting effects on all major circadian parameters of the SCN slice oscillation at both the cellular and circuit levels, and that it achieves this through a diversity of molecular pathways, in particular through cAMP/Ca2+ response elements within gene promoters. The final chapter focuses primarily on DUSP4, a negative regulator of the MAP kinase pathway that I have demonstrated to be upregulated by VIP. Here I demonstrate that DUSP4 affects the steady-state period of SCN slices, as well as influences phase shifting characteristics of both slices and mice. To conclude, the work presented here furthers our knowledge of neuropeptidergic communication in mammalian pacemaking. I have undertaken extensive characterisation of the molecular mechanisms through which the VIP neuropeptide influences SCN oscillators, and I have determined differential roles for the VIP and VPAC2 neurons in circadian timekeeping.

Finite element analysis is versatile and used widely in a range of engineering andscientific disciplines. As time passes, the problems that engineers and designers areexpected to solve are becoming more computationally demanding. Often theproblems involve the interplay of two or more processes which are physically andtherefore mathematically coupled. Although parallel computers have been availablefor about twenty years to satisfy this demand, finite element analysis is still largelyexecuted on serial machines. Parallelisation appears to be difficult, even for thespecialist. Parallel machines, programming languages, libraries and tools are used toparallelise old serial programs with mixed success. In some cases the serialalgorithm is not naturally suitable for parallel computing. Some argue that rewritingthe programs from scratch, using an entirely different solution strategy is a betterapproach. Taking this point of view, using MPI for portability, a mesh free elementby element method for simple data distribution and the appropriate iterative solvers,a general parallel strategy for finite element analysis is developed and assessed.

Ma thèse a pour objet le rôle de la déesse-fleuve Sarasvatī dans le Ṛgveda, le plus ancien texte de l’Inde en sanskrit védique (2ème millénaire av. N. è). Sarasvatī est l’une des rares divinités védiques dont le culte persiste jusqu’aux temps modernes, et cela malgré son statut mineur dans le Ṛgveda où son nom apparaît dans 71 strophes et trois hymnes qui lui sont adressés. La thèse cherche à résoudre la dichotomie entre la nature physique et mythologique de la déesse, notamment que les tribus védiques vénéraient un fleuve du nom de Sarasvatī qui se desséchait progressivement en même temps qu’elles la célébraient comme déesse de la poésie. Une analyse des attributs, des actions et des requêtes adressés à la divinité montre qu’elle a cinq fonctions distinctes : fleuve, elle est aussi une déesse de la guerre, des joutes verbales, de la poésie et de la fertilité. Ces diverses fonctions ne sont pas en contradiction les unes avec les autres : elles sont liées entre elles par une série de connexions. Il apparaît que le fleuve formait jadis une frontière territoriale séparant les tribus indo-aryennes de la population indigène du sous-continent indien, ce qui a fait de Sarasvatī une déesse guerrière. La confrontation guerrière se répercute dans la sphère du sacrifice où Sarasvatī a une fonction en tant que déesse des joutes poétiques. Cette interprétation de Sarasvatī est tout à fait inédite et jette une nouvelle lumière sur son personnage dans la religion védique<br>This thesis examines the role of the river goddess Sarasvatī in the Ṛgveda, the most ancient text of the Indian corpus from the second millennium B. C. E. Sarasvatī is one of the few Vedic deities to have endured through to modern-day Hinduism despite her minor status in the Ṛgveda where her name appears just 71 times with only three hymns dedicated to her. The thesis seeks to resolve the dichotomy between the physical and mythical reality of the goddess by examining why the Vedic people worshipped a river that was seemingly in a state of desiccation and simultaneously revered it as a goddess of poetry. An analysis of all her attributes, actions, and requests reveals that her personality comprises five distinct aspects, which determine her unique role in the Vedic religion: she is a river but also a goddess of war, verbal contests, poetry, and fertility. These aspects are not at variance with one another, but are rather linked via a series of connections in such a way that her function as a goddess of poetry is tied to her physical nature. It seems that the river at one time formed a territorial boundary dividing the migrating Indo-Aryans tribes from the indigenous population of the Indian subcontinent, which in turn led to Sarasvatī becoming a warrior goddess. This military competition is then reflected in the sacrificial domain where she holds the function of a goddess of poetic contests. The frequent requests for Sarasvatī to assist the warrior-patrons in battle and the poets in the verbal contests show that her most central roles are as a goddess of competition and victory. This wholly-revised perspective thus throws new light on her function in the Vedic religion

In the past two decades, there have been significant advances in the development of self-centering (SC) seismic force resisting systems. However, examples of SC systems used in practice are limited due to unusual field construction practices, high initial cost premiums and deformation incompatibility with the gravity framing. A self-centering beam moment frame (SCB-MF) has been developed that virtually eliminates residual drifts and concentrates the majority of structural damage in replaceable fuse elements. The SCB consists of a I-shaped steel beam augmented with a restoring force mechanism attached to the bottom flange and can be shop fabricated. Additionally, the SCB has been designed to eliminate the deformation incompatibility associated with the self-centering mechanism.
The SCB-MF system is investigated and developed through analytical, computational, and experimental means. The first phase of the work involves the development of the SCB concepts and the experimental program on five two-thirds scale SCB specimens. Key parameters were varied to investigate their effect on global system hysteretic response and their effect on system components. These large-scale experiments validated the performance of the system, allowed the investigation of detailing and construction methods, provided information on the behavior of the individual components of the system. The experimental results also provided data to confirm and calibrate computational models that can capable of capturing the salient features of the SCB-MF response on global and component level.
As a part of the second phase, a set of archetype buildings was designed using the self-centering beam moment frame (SCB-MF) to conduct a non-linear response history study. The study was conducted on a set of 9 archetype buildings. Four, twelve and twenty story frames, each with three levels of self-centering ratios representing partial and fully self-centering systems, were subjected to 44 ground motions scaled to two hazard levels. This study evaluated the performance of SCB-MFs in multi-story structures and investigated the probabilities of reaching limit states for earthquake events with varying recurrence period.
The experimental and computational studies described in this dissertation demonstrate that the SCB-MF for steel-framed buildings can satisfy the performance goals of virtually eliminating residual drift and concentrating structural damage in replaceable fuses even during large earthquakes.<br>Ph. The quest for high-throughput proteomics in recent years has revealed a number of critical issues. Whilst improved 2-D gel electrophoresis (2-DE) sample preparation, staining and imaging issues are being actively pursued by industry, reliable high-throughput spot matching and quantification remains a significant bottleneck in the bioinformatics pipeline. The flow of data to mass spectrometry through robotic spot excision and protein digestion so far has been restricted. The purpose of this thesis is to describe a computational framework that is suitable for large-scale image mining and statistical cross-validation of 2-DE. This is in response to the development of a statistical 2-DE ontology that underpins the emerging Human Proteome Organisation’s Proteomics Standards Initiative General Proteomics Format (HUPO PSI GPS).The thesis begins with a comprehensive review of the role of bioinformatics in 2-DE. A platform for Statistical Expression Analysis (SEA) is then proposed, which performs the analysis statistically and simultaneously between sets of gel replicates. The method circumvents the drawback of traditional approaches in using symbolic representation of spots at the very early stages of the analysis, which despite greatly simplifying data handling and management, introduces persistent errors due to inaccuracies in spot modelling and matching. With the proposed SEA, the method is fully automated and small insignificant expression changes over one gel pair can be revealed when reinforced by the same consistent changes in others. To achieve this, we present an integrated image registration and bias field correction technique to normalise sets of gels for direct comparison. With the Robust Advanced Image Normalisation (RAIN) algorithm, image intensity distribution is used, rather than selected features. A new method of volume-invariant warping is proposed which ensures the volume of protein expression under transformation is preserved. In this thesis, the relative bias field between reference and sample gel is modelled by multiplicative and additive piecewise B-spline surfaces. To overcome the massive computational burden SEA entails, a cluster computing framework has been developed. It utilises Condor middleware, JPEG-LS lossless image compression and probabilistic task replication for novel distributed image processing. Also, the image analysis has been designed and implemented with GPGPU (general purpose computation on graphics processors), which brings a significant increase in computational performance. The validation of the proposed techniques was performed with 2-DE image data produced by University College Dublin, along with large datasets formed from the HUPO Human Brain Proteome Project (HBPP). The HBPP is a worldwide academic and industrial collaboration of neuroproteomic centres, with the aim of characterising the human and mouse brain proteomes for studying human neurodegenerative diseases. Compared with existing methods, results from the proposed analysis framework show substantial improvements in computational throughput and expression analysis sensitivity.

Background: The term Open Innovation describes a phenomenon where organizations are no longer solely relying on internal R&D to innovate technologies that they then would bring to market. Instead, firms are looking outside for ideas or working with other organizations to develop technologies in order to capitalize on efficiencies of existing knowledge, reduce investment risks and increase speed to market. We identified a need to analyze the existing open innovation adoption frameworks in order to track similarities and differences so that we could create a new framework which is more comprehensive. Purpose: The purpose of this thesis is to create a framework for the analysis of the open innovation adoption process which is based on organizational change and other relevant streams of research. In order to do this we were to find answers to the following research questions: 1) How can organizational change theory be used to analyze the movement from a closed to open innovation strategy? 2) In addition to organizational change theory, which research streams should be considered when analyzing the open innovation adoption process? 3) To what extent does our new framework provide an adequate foundation for analyzing the open innovation adoption process at SCA Hygiene Products? Results: Our new framework is meant for the analysis of open innovation adoption process. The framework helps researchers to analyze how different organizations have adopted open innovation based on the four research streams that we have analyzed. Spinal Cord Stimulation (SCS) is a treatment for lumbar back pain and despitethe proven effcacy of the technology, there is a lack of knowledge in how the treatment outcome varies between different patients groups. Furthermore, since the method is costly, in the sense of material, surgery and follow-up time, a more accurate patient targeting would decrease healthcare costs. Within recent years, Real World Data (RWD) has become a vital source of information to describe the effects of medical treatments. The complexity, however, calls for new, innovative methods using a larger set of useful features to explain the outcome of SCS treatments. This study has employed machine learning algorithms, e.g., Random Forest Classier (RFC) boosting algorithms to finally compare the result with the baseline; Logistic regression (LR). The results retrieved was that RFC tend to classify successful and unsuccessful patients better while logistic regression was unstable regarding unbalanced data. In order to interpret the insights of the models, we also proposed a Soft Accuracy Measurement (SAM) method to explain how RFC and LR differ. Some factors have shown to impact the success of SCS. These factors were age, income, pain experience time and educational level. Many of these variables could also be supported by earlier studies on factors of success from lumbar spine surgery.

This dissertation focuses on the use of subcarrier multiplexing (SCM) in multimode fibers, utilizing carrier frequencies above what is generally utilized for multimode fiber transmission, to achieve high bit rates. In the high frequency region (i.e., frequencies larger than the intermodal bandwidth), the magnitude response of multimode fiber does not decrease monotonically as a function of the frequency but is shown to become relatively flat (but with several deep nulls) with an amplitude below that at DC. The statistical properties of this frequency response at high frequencies are analyzed. The probability density function of the magnitude response at high frequencies is found to be a Rayleigh density function. The average amplitude in this high frequency region does not depend on the frequency but depends on the number of modes supported by the fiber. To transmit a high bit rate signal over the multimode fiber, subcarrier multiplexing is adopted. The performance of the SCM multimode fiber system is presented. The performance of the SCM system is significantly degraded if there are some subcarriers located at the deep nulls of the fiber. Equalization and spread spectrum techniques are investigated but are shown to be not effective in combating the effects of these nulls. To cancel the effects of these deep nulls, training process and diversity coding are considered. The basic theory of diversity coding is given. It is found that the performances of the system with training process and the system with diversity coding are almost identical. However, diversity coding is more appropriate since it requires less system complexity. Finally, the practical limits and capacity of the SCM multimode fiber system are investigated. It is shown that a signal with a bit rate of 1.45 Gbps can be transmitted over a distance up to 5 km.<br>Ph. D.

Soybean is one of the most important grain legumes grown in US and worldwide, and is a major component of human and animal protein diets. Despite improvements in management practices, and the introduction of improved soybean cultivars, soil borne pathogens continue to cause tremendous yield loss in soybean production each year. Among soil borne pathogens; Soybean Cyst Nematode (SCN) or Heterodera glycines together with Sudden Death Syndrome (SDS) induced by Fusarium virguliforme are responsible for the most damages in soybean fields. The most effective way to control these two pathogens is to develop resistant cultivars. Resistance to any population (HgType) of H. glycines, requires a functional allele at rhg1/Rfs2 locus. The rhg1/Rfs2 gene encodes a receptor-like kinase (RLK) protein. By analysing near isogenic lines (NIL) segregating for rhg1/Rfs2, rhg1-like loci were found at other locations most conservedly on LG B1. While the nature of rhg1 allele was thought to be recessive, heterozygous NIL segregating at the rhg1 locus showed that the resistant allele was dominant. Rhg1 was also inferred to be multigeneic due to absence of recombination between the RLK and other 2 genes. Functional and structural analyses were conducted on the leucine rich repeat (LRR) from RLK protein encoded by GmRLK18-1 within the Rhg1/Rfs2 locus. The LRR of GmRLK18-1 showed a high binding affinity to CLE-like peptides found in both nematode secretions and plant developmental control. Crosslinking assays and native gel analysis of GmRLK18-1-LRR validated its model as a crystal homo-dimer. Larger proteins were also shown to bind the LRR domain, in far-Western analyses both methionine synthase and cyclophilin bound strongly to the LRR domain. Homology and ab-initio modeling of the LRR domain of the GmRLK18-1 was predicted as both a monomer and a homodimer containing intrinsically unstructured regions. Amino acid substitutions found among GmRLK18-1 allotypes A87V, Q115K and H274N were predicted to play crucial roles in protein function and stability. The receptor like kinase (RLK) GmRLK18-1 within the Rhg1/Rfs2 locus underlies a pleiotropic resistance to both SCN and SDS. The resistance allele was shown to be dominant in both heterozygous NILs at Rhg1/Rfs2 and transgenics (hetero- or hemi-zygous). The RLK was found to provide a partial resistance to SCN and importantly a nearly complete resistance to both root and leaf symptoms of SDS. In the presence of Rhg4, the RLK-transgenic plants developed nearly full resistance to SCN. Therefore the RLK was proven to underlie a major portion of the Rhg1/Rfs2 locus.

It has recently been discovered that some, if not all, classical novae emit GeV gamma-rays during outburst, but the mechanisms involved in the production ofgamma-rays are still not well understood. We present here a comprehensive multiwavelength data set-from radio to X-rays-for the most gamma-ray-luminous classical nova to date, V1324 Sco. Using this data set, we show that V1324 Sco is a canonical dusty Fe II-type nova, with a maximum ejecta velocity of 2600 km s(-1) and an ejecta mass of a few x 10(-5) M-circle dot. There is also evidence for complex shock interactions, including a double-peaked radio light curve which shows high brightness temperatures at early times. To explore why V1324. Sco was so gamma-ray luminous, we present a model of the nova ejecta featuring strong internal shocks and find that higher gamma-ray luminosities result from higher ejecta velocities and/or mass-loss rates. Comparison of V1324. Sco with other gamma-ray-detected novae does not show clear signatures of either, and we conclude that a larger sample of similarly well-observed novae is needed to understand the origin and variation of gamma-rays in novae.

The polymers are analyzed physically and chemically using atomic force microscopy, profilometry, scanning electron microscopy, optical microscopy, contact angle measurement, FT infra-red spectroscopy and energy dispersive X-ray spectrometry. PEEK is found to be the most suitable polymer and FT Infra-red spectroscopy an informative analytic tool.

This thesis report presents a method to analyze the communication architecture for the Intelligent Transportation Systems (ITS). The report also includes a case study on ASN.1 notation and analysis of its encoding rules. Included in the report is also: (i) accompanying instruction on how to use ASN.1 compilers to produce the C/C++ message encoder/decoder, and (ii) analysis of Non-IP communications of Communication Access for Land Mobiles (CALM-FAST) protocol stack in ITS. The thesis is a part of the research project entitled “SCOOP”, a joint project between SCANIA CV AB and KTH. The purpose of this thesis is to contribute to the ultimate goal, which is to equip a vehicle with necessary hardware and software technology to provide a platooning behavior in the GCDC 2011 competition. This goal is achieved by the means of wireless communication system for both vehicle to vehicle and vehicle to road side units communications in the platoon. Overall, this thesis introduces the important usage of ASN.1 in implementation of cut-edge telecommunication systems especially in V2V and V2I communication; and clarifies the CALM-FAST protocol stack in mobile nodes.<br>Kartlägga CALM-FAST protokollet och hur det användes tillsammans med den i tävlingen GCDC 2011 fastslanga kommunikationsprotokollet. Studio inerente al tema della manutenzione su condizione, applicata ad un impianto per la produzione di capsule in plastica, in collaborazione con il gruppo SACMI S.C., a Imola, all’interno della divisione Closures & Cointainers. Presentata la tematica della manutenzione degli impianti di produzione e la sua evoluzione negli ultimi decenni, si pone l’attenzione sulle attività aziendali e l’ambito in cui si è incentrato lo studio. Segue una trattazione teorica del tema della Big Data Analysis e di come questa può essere sfruttata in termini di manutenzione predittiva e stima della RUL di un un particolare componente, concentrandosi sulle tecniche più recenti applicate in merito.
Successivamente viene spiegato nel dettaglio il funzionamento della linea produttiva esaminata ed è affrontata la gestione del database di dati raw raccolti dai sensori presenti sull’impianto esaminato. Dopo averli presentati, per i diversi gruppi funzionali della linea presa in esame sono stati generati grafici su un periodo di nove mesi di produzione presso un cliente, andando a valutare: condizioni di funzionamento, possibili anomalie, trend che giustificassero usura o malfunzionamento di uno o più componenti. I risultati sono stati, quando possibile, confrontati con i report manutentivi reali, per avere conferme della robustezza dell’informazione generata dai sensori, in un’ottica che ha come possibile obiettivo quello di porre le basi per un futuro sistema di manutenzione su condizione.

A major step in the biosynthesis of many membrane proteins is their insertion into the membrane of the endoplasmic reticulum (ER). The insertion of a multi-spanning membrane protein is a complex process since several transmembrane (TM) domains have to be correctly integrated in order to enable its correct assembly. At present it is unclear how the integration of multiple TM domains is co-ordinated by the ER translocon. The aim of this study was to analyse the molecular environment of the TM domains of a model seven TM domain protein, opsin, so as to better understand the mechanism by which integration occurs. For this purpose, stable 'integration intermediates' of defined lengths representing distinct stages of opsin biosynthesis were generated by in vitro translation of truncated mRNA in the presence of semi-permeabilised cells. Cysteine-mediated, site-specific cross-linking and immunoprecipitation were employed to examine the environment of these integration intermediates. In addition, cysteine-specific modification reagents with different physical properties were used to investigate the environment of opsin TM3 during its insertion at the ER membrane. Opsin TM domains exhibit unique patterns of adduct formation with the ER translocon components, Sec61α and Sec61β. TM1 associates with the Sec61 complex at two distinct stages during nascent chain extension, and this behaviour is dependent on the presence of subsequent TM domains. The re-association of TM1 with the transloconmay well facilitate the co-ordinated integration of TMs 1-3 into the lipid bilayer. Opsin TM4 exits the Sec61 complex as soon as the subsequent TM domain is synthesised, while TM5, TM6 and TM7 remain associated with the ER translocon throughout protein synthesis, suggesting their concerted release upon chain termination. Evidence is provided that opsin is integrated via a single Sec61 heterotrimer, despite the fact that the ER translocon appears to consist of multiple copies of the Sec61 complex. On the basis of this work, a model is presented describing the complete integration of opsin at the ER membrane.

The thesis adapts Bourdieu’s theory of hexis as a method for approaching the Baillie (Hegel/Baillie, 1910/1931) and Pinkard (Hegel/Pinkard, 2008) translations of Hegel’s Die Phänomenologie des Geistes (Hegel, 1807/1970) as embodiments of a translatorial practice informed by social and philosophical contextual factors. The theoretical concept of a translatorial hexis is analogous to Bourdieu’s habitus but differs in that the translatorial hexis embodies a specifically dominant, honour-seeking stance of the translator with regard to the micro-dynamics of the surrounding sub-fields; the translatorial hexis is also embodied primarily in the detail of the text and in the peritexts to the translations. Chapters 3 and 4, which focus on the Baillie and Pinkard translations, are each divided into three sections: an analysis of the historical background to the translation in terms of interrelated Bourdieusian fields defined by rival positions vying for academic reputation; an analysis of lexical patterning identified in TT corpora with reference to the translations of two ‘dialectically ambiguous’ terms Geist [mind/spirit] and aufheben [cancel/preserve/sublate]; an analysis of peritexts to the two translations. Starting with a discussion of Hegel’s ‘dialectical ambiguity’ in chapter 1 and an elaboration of the Bourdieusian theoretical framework in chapter 2, the thesis attempts to explain the lexical findings with reference to the concept of translatorial hexis in a manner which takes philosophical and sociological factors into consideration as determinants of the translators’ strategies. The analysis focuses on the positioning of Sir James Black Baillie with regard to Absolutist and Personalist versions of British Idealism and Terry Pinkard with regard to the non-metaphysical readings of Hegel and the development of communitarian ideologies. The publication of new translations of Hegel’s works and new critical works on German Idealism suggest that a Hegel revival is in full progress. Given the centrality of translation to this phenomenon, it is appropriate that translation studies should contribute to the discussion, especially to demonstrate the value of a self-reflexive, multi-disciplinary approach which brings linguistic analysis and sociological contextualisation to bear on some of the philosophical issues at stake.

Two metakaolins were evaluated for use as supplementary cementitious materials in cement-based systems. The metakaolins varied in their surface area (11.1 v. 25.4 m2/g), but were quite similar in mineralogical composition. Performance of metakaolin mixtures was compared to control mixtures and to mixtures incorporating silica fume as partial replacement for cement at water-to-cementitious materials ratios of 0.40, 0.50, and 0.60. In this study, the early age properties of fresh concrete and the mechanical and durability properties of hardened concrete were examined. Early age evaluations aimed to determine the reactivity of metakaolin (heat of hydration) and its effect on mixture workability (slump, setting time, unit weight). In addition, three types of shrinkage were monitored in metakaolin-cement systems: chemical, autogenous, and free. Compressive, tensile, and flexural strength and elastic modulus were measured at various concrete ages. The influence of metakaolin addition on durability was assessed through accelerated testing for sulfate resistance, expansion due to alkali-silica reaction, and through rapid chloride permeability measurements.
To further quantify the underlying mechanisms of metakaolin's action, the microstructure of pastes was examined. Calcium hydroxide (CH) content was determined using thermogravimetric analysis and verified using differential thermal analysis. Surface area and pore size distribution were evaluated via nitrogen adsorption. These analyses yielded information about the pozzolanic reactivity of metakaolin, associated CH consumption and pore structure refinement, and resulting improvements in mechanical performance and durability of metakaolin-concretes.

The Arabidopsis SON1 F-box protein was implicated in regulating a pathogen defense pathway, but its exact function in wild-type plants is unknown. As an F-box protein it was predicted that SON1 would assembles into a SON1-SCF ubiquitin ligase complex that recruits specific plant defense-related proteins for proteolysis. The yeast 2-hybrid assay was used to screen for potential substrates for a putative SON1-SCF ligase, leading to the identification of Arabidopsis LNK2 as a SON1-binding factor.
Comprehensive protein-protein interaction analysis has shown that the binding of SON1 to LNK2 protein is specific, because closely related, full-length Arabidopsis F-box proteins do not interact with LNK2. However, when tested in isolation, some fragments derived from the paralog proteins did bind SON1, suggesting that higher order structure or inter-domain interference affects the ability of SON1 to recruit substrate. When analyzed for interaction domains, three regions of SON1 were identified that bind to LNK2 and a LNK2 binding region spans a conserved amino acid region. Phylogenetic analysis revealed that there is a paralogous gene called LNK1 in Arabidopsis, and both LNK1 and LNK2 are restricted to the plant kingdom. LNK2 and LNK1 are seen to be widely distributed in embryophyte seed and spore plants. Genetic analysis and complementation tests showed that LNK1 and LNK2 regulate flowering and photo-morphogenesis redundantly.
Though lnk1 and lnk2 mutants look similar to wild-type plants, lnk1 lnk2 double mutant plants possess a long hypocotyls and flower late compared to wild-type plants. Because SON1 is implicated in plant defense, lnk mutants were assessed for susceptibility to a virulent oomycete pathogen, Hyaloperonospora arabidopsidis (Hpa). Interestingly, lnk mutants supported less disease development, suggesting a role of the wild-type LNK proteins in the enabling of pathogen colonization or in repressing host defenses. To confirm that each of the phenotypes described were a consequence of lnk1 and lnk2 mutations, wild type LNK1 and LNK2 were introduced into lnk1 lnk2 plants and examined. For most phenotypes, genetic complementation and thus restoration of a WT phenotype was observed. However, differences were uncovered in the ability of LNK genes to rescue the phenotypes, indicating specialization of function.
The interaction of LNK2 with SON1 suggests that SON1-dependent ubiquitination and proteasomal degradation may control LNK2 abundance. I show by a cell free protein degradation assays that proteasome-based degradation of LNK2 as well as LNK1 is possible. Data showed that SON1 binds to ASK1 in-planta suggest the existence an SCFSON1 complex that targets LNK2 for polyubiquitination and its subsequent degradation by the proteasome. The data presented in this dissertation indicates a role for LNKs in flowering and plant defense and also suggest that proteasome-base regulation of LNK turnover may be utilized to regulate LNK protein abundance and proper maintenance of the circadian clock.

This project examines the ontoepistemological underpinnings of diversity education in the field of communication by focusing on the points where diversity, pedagogy and communication intersect. In this study I seek to understand how we come to know what we know about diversity, or the social construction of differentness, and how we share this information with others. I analyzed three popular interpersonal communication textbooks, examining the patterns revealed in the text, in order to address these questions.
This study uses three complimentary methods to reveal the number of occurrences that center on diversity in the text (content analysis), to interpret themes reflected by the patterns discovered in the text (thematic analysis) and a creative twist on the coding process that opens the analysis process to the coders and includes their input as participants to this study (reflexive content analysis). The results of this study revealed three-hundred ten occurrences of the social construction of difference across all three textbooks but only a portion of those, seventy-six, suggested social constructionist underpinnings of these constructs. This study shows how we have missed an opportunity at the introductory level to expand our student's knowledge of issues in diversity.
