We have investigated the role for diacylglycerol (DAG) in membrane bud formation in the Golgi apparatus. Addition of propranolol to specifically inhibit phosphatidate phosphohydrolase (PAP), an enzyme responsible for converting phosphatidic acid into DAG, effectively prevents formation of membrane buds. The effect of PAP inhibition on Golgi membranes is rapid and occurs within 3 min. Removal of the PAP inhibitor then results in a rapid burst of buds, vesicles, and tubules that peaks within 2 min. The inability to form buds in the presence of propranolol does not appear to be correlated with a loss of ARFGAP1 from Golgi membranes, as knockdown of ARFGAP1 by RNA interference has little or no effect on actual bud formation. Rather, knockdown of ARFGAP1 results in an increase in membrane buds and a decrease of vesicles and tubules suggesting it functions in the late stages of scission. How DAG promotes bud formation is discussed.

Background: Previously, we reported on an opioid receptor quantitative systems pharmacology (QSP) model to evaluate naloxone Methods: In this study, we extended our model to include higher systemic levels of fentanyl (up to 100 ng/ml) and the newly approved 8mg IN naloxone dose (equivalent to 4 mg) Results: As expected, at the lower peak fentanyl concentrations (25 ng/ml and 50 ng/ml), the simulations predicted that 2 mg, 4 mg, 5 mg, and 10 mg IM doses of naloxone displaced fentanyl and reached below the 50% receptor occupancy within 10 minutes. However, at the concentration of 75 ng/ml, the simulation predicted that the 2 mg dose of naloxone failed to reach below the 50% occupancy within 10 minutes. Interestingly, at the highest peak concentration of fentanyl studied (100 ng/ml), the model predicted that the 4 mg of naloxone IM (equivalent to 8 mg IN) failed to reach below the threshold of 50 % occupancy within 10 minutes or even within 15 minutes (data not shown). In contrast, the model predicted successful reversals when 5 and 10 mg IM doses were utilized. Conclusion: These results support the notion that acutely administered higher doses of naloxone are needed for rapid and adequate clinical reversal, particularly when higher systemic exposure of the potent synthetic opioids occurs.

A defining feature of adaptive behavior is our ability to change the way we interpret sensory stimuli depending on context. Rapid adaptation in behavior has been attributed to frontal cortical circuits, but it is not clear if sensory cortexes also play an essential role in such tasks. In this study we tested whether the auditory cortex was necessary for rapid adaptation in the interpretation of sounds. We used a two-alternative choice sound-categorization task for rats in which the boundary that separated two acoustic categories changed several times within a behavioral session. These shifts in the boundary resulted in changes in the rewarded action for a subset of stimuli. We found that extensive lesions of the auditory cortex did not impair the ability of rats to switch between categorization contingencies and sound discrimination performance was minimally impaired. Similar results were obtained after reversible inactivation of the auditory cortex with muscimol. In contrast, lesions of the auditory thalamus largely impaired discrimination performance and, as a result, the ability to modify behavior across contingencies. Thalamic lesions did not impair performance of a visual discrimination task, indicating that the effects were specific to audition and not to motor preparation or execution. These results suggest that subcortical outputs of the auditory thalamus can mediate rapid adaptation in the interpretation of sounds.

After clathrin-mediated endocytosis, clathrin removal yields an uncoated vesicle population primed for fusion with the early endosome. Here we present the first characterization of uncoated vesicles and show that myo6, an unconventional myosin, functions to move these vesicles out of actin-rich regions found in epithelial cells. Time-lapse microscopy revealed that myo6-associated uncoated vesicles were motile and exhibited fusion and stretching events before endosome delivery, processes that were dependent on myo6 motor activity. In the absence of myo6 motor activity, uncoated vesicles remained trapped in the actin mesh, where they exhibited Brownian-like motion. Exit from the actin mesh occurred by a slow diffusion-based mechanism, delaying transferrin trafficking to the early endosome. Expression of a myo6 mutant that bound tightly to F-actin produced immobilized vesicles and blocked trafficking. Depolymerization of the actin cytoskeleton rescued this block and specifically accelerated transferrin delivery to the early endosome without affecting earlier steps in endocytosis. Therefore actin is a physical barrier impeding uncoated vesicle trafficking, and myo6 is recruited to move the vesicles through this barrier for fusion with the early endosome.

Bacteraemic sepsis has a high mortality that can be reduced by early diagnosis and initiation of appropriate antimicrobial therapy 1. Rapid confirmation of the diagnosis and identification of the causal agent provide guidance on the adequacy and duration of antimicrobial therapy and on the need for source investigation. Clinical microbiology laboratories have rightly placed great emphasis on this aspect of their practice. As causal organisms are usually present in low titre, direct microscopy is impractical and laboratories have generally relied on culture of blood in broth, which is relatively insensitive and too slow to influence initial management. Phenotypic methods for the identification and antimicrobial susceptibility testing (AST) of isolates have progressively improved over the last two decades, but still require significant periods of incubation. Similarly, commercial blood culture systems have been refined with better systems for automated detection of growth in broth, but still require incubation for up to five days and subculture for the isolation of pathogens. Constantly monitored blood culture systems and automated identification/AST are now the norm in most clinical laboratories. Although there will undoubtedly be further development of phenotypic methods, with incremental improvements in sensitivity and time-to-detection, research and development now concentrated on molecular detection methods has the potential to result in a paradigm shift in our approach to the microbiological diagnosis of this condition.

Yeast that naturally exhaust the glucose from their environment differentiate into three distinct cell types distinguishable by flow cytometry. Among these is a quiescent (Q) population, which is so named because of its uniform but readily reversed G1 arrest, its fortified cell walls, heat tolerance, and longevity. Daughter cells predominate in Q-cell populations and are the longest lived. The events that differentiate Q cells from nonquiescent (nonQ) cells are initiated within hours of the diauxic shift, when cells have scavenged all the glucose from the media. These include highly asymmetric cell divisions, which give rise to very small daughter cells. These daughters modify their cell walls by Sed1- and Ecm33-dependent and dithiothreitol-sensitive mechanisms that enhance Q-cell thermotolerance. Ssd1 speeds Q-cell wall assembly and enables mother cells to enter this state. Ssd1 and the related mRNA-binding protein Mpt5 play critical overlapping roles in Q-cell formation and longevity. These proteins deliver mRNAs to P-bodies, and at least one P-body component, Lsm1, also plays a unique role in Q-cell longevity. Cells lacking Lsm1 and Ssd1 or Mpt5 lose viability under these conditions and fail to enter the quiescent state. We conclude that posttranscriptional regulation of mRNAs plays a crucial role in the transition in and out of quiescence.

AbstractClimate in the Arctic has warmed at a more rapid pace than the global average over the past few decades leading to weather, snow, and ice situations previously unencountered. Reindeer herding is one of the primary livelihoods for Indigenous peoples throughout the Arctic. To understand how the new climate state forces societal adaptation, including new management strategies and needs for preserved, interconnected, undisturbed grazing areas, we coupled changes in temperature, precipitation, and snow depth recorded by automatic weather stations to herder observations of reindeer behaviour in grazing areas of the Laevas Sámi reindeer herding community, northern Sweden. Results show that weather and snow conditions strongly determine grazing opportunities and therefore reindeer response. We conclude that together with the cumulative effects of increased pressures from alternative land use activities, the non-predictable environmental conditions that are uniquely part of the warming climate seriously challenge future reindeer herding in northern Sweden.

Traditionally, biosensor development has focused on molecules with a defined metabolic role that can be exploited by enzyme-based systems. Antibodies have the ability to move beyond this range of analytes, and are particularly useful in detecting small, hapten molecules. Electrochemically based biosensor developments have been less fruitful in this regard, as enzyme labelling is required, and such assays require the separation from bound and unbound species. These separations and the removal of background signals result in the increased complexity of the assay format, making it unsuitable for rapid sensor analysis. We have developed an electrochemical sensor based on anti-bodies that does not require the separation of bound and unbound molecules in a competition immunoassay format. This removes the need for several washing and separation steps as is normally employed in this type of assay. This allows single-step immunoassays to be performed using this system, and also allows for the real-time monitoring of antibody—antigen interactions. We have shown that such assays are possible in both batch and flow-injection formats and we are currently developing an assay for the pesticide atrazine. Tentative results show that analysis with this system is possible in the p.p.m. to p.p.b. range.

Background: While the global maternal mortality ratio (MMR) shows a decreasing trend, there is room for improvement. Midwifery education has been under scrutiny to ensure that graduates acquire knowledge and skills relevant to the local context. Objective: To review the basic professional midwifery qualification and pre-practice requirements in countries with lower MMR compared with Malaysia. Methods: A rapid review of country-specific Ministry of Health and Midwifery Association websites and Advanced Google using standardised key words. English-language documents reporting the qualifications of midwives or other requirements to practise midwifery from countries with a lower MMR than Malaysia were included. Results: Sixty-three documents from 35 countries were included. The minimum qualification required to become a midwife was a bachelor’s degree. Most countries require registration or licensing to practise, and 35.5% have implemented preregistration national midwifery examinations. In addition, 13 countries require midwives to have nursing backgrounds. Conclusion: In countries achieving better maternal outcomes than Malaysia, midwifes often have a degree or higher qualification. As such, there is a need to reinvestigate and revise the midwifery qualification requirements in Malaysia.

Abstract Chromatin insulators protect erythroid genes from being silenced during erythropoiesis, and the disruption of barrier insulator function in erythroid membrane gene loci results in mild or severe anemia. We showed previously that the USF1/2-bound 5′HS4 insulator mediates chromatin barrier activity in the erythroid-specific chicken β-globin locus. It is currently not known how insulators establish such a barrier. To understand the function of USF1, we purified USF1-associated protein complexes and found that USF1 forms a multiprotein complex with hSET1 and NURF, thus exhibiting histone H3K4 methyltransferase- and ATP-dependent nucleosome remodeling activities, respectively. Both SET1 and NURF are recruited to the 5′HS4 insulator by USF1 to retain the active chromatin structure in erythrocytes. Knock-down of NURF resulted in a rapid loss of barrier activity accompanied by an alteration of nucleosome positioning, increased occupancy of the nucleosome-free linker region at the insulator site, and increased repressive H3K27me3 levels in the vicinity of the HS4 insulator. Furthermore, suppression of SET1 reduced barrier activity, decreased H3K4me2 and acH3K9/K14, and diminished the recruitment of BPTF at several erythroid-specific barrier insulator sites. Therefore, our data reveal a synergistic role of hSET1 and NURF in regulating the USF-bound barrier insulator to prevent erythroid genes from encroachment of heterochromatin.

Adversarial Optimization provides a reliable, practical way to match two implicitly defined distributions, one of which is typically represented by a sample of real data, and the other is represented by a parameterized generator. Matching of the distributions is achieved by minimizing a divergence between these distribution, and estimation of the divergence involves a secondary optimization task, which, typically, requires training a model to discriminate between these distributions. The choice of the model has its trade-off: high-capacity models provide good estimations of the divergence, but, generally, require large sample sizes to be properly trained. In contrast, low-capacity models tend to require fewer samples for training; however, they might provide biased estimations. Computational costs of Adversarial Optimization becomes significant when sampling from the generator is expensive. One of the practical examples of such settings is fine-tuning parameters of complex computer simulations. In this work, we introduce a novel family of divergences that enables faster optimization convergence measured by the number of samples drawn from the generator. The variation of the underlying discriminator model capacity during optimization leads to a significant speed-up. The proposed divergence family suggests using low-capacity models to compare distant distributions (typically, at early optimization steps), and the capacity gradually grows as the distributions become closer to each other. Thus, it allows for a significant acceleration of the initial stages of optimization. This acceleration was demonstrated on two fine-tuning problems involving Pythia event generator and two of the most popular black-box optimization algorithms: Bayesian Optimization and Variational Optimization. Experiments show that, given the same budget, adaptive divergences yield results up to an order of magnitude closer to the optimum than Jensen-Shannon divergence. While we consider physics-related simulations, adaptive divergences can be applied to any stochastic simulation.

Bacterial colonies infection is one of the causes of bloodstream disease, and it can be a fatality. Therefore, medical diagnoses require fast identification and classification of organisms. Artificial Intelligence with deep learning (DL) can now be developed as a rapid bacterial classification. The research aims to combine deep learning and support vector machines (SVM). The ResNet-101 model of the DL algorithm extracted the image’s features using transfer learning then classified by the SVM classifier. According to the experimental results, this model had 99.61% accuracy, 99.58% recall, 99.58% precision, and 99.97% specificity. The technique presented might enhance clinical decision-making.

Manual scanning electron microscope (SEM) analysis is historically considered to be slow and tedious resulting in a low volume of data. This is due in large part to the mechanics of moving stage locations and recording image and spectral data. Conversely, high volume data acquired using automated SEM analysis has been associated with the need for complex systems for data management and analysis. In addition, the proliferation of high volume digital microscopy and its attendant “ tonnage” of paper images has lead to the desire for a “green” (filmless and hardcopy-reduced) operation.There are some classes of projects which are amenable to automated feature analysis - discrete features that are distinct from a background material. However, many projects require operator intervention in order to identify the region or points of interest. Yet, these projects may also require that large data sets be acquired and analyzed for statistical rigor.

Detection of bacteria in implants can be time consuming and equivocal. Routine histology of biopsy specimens from implant sites requires approximately 24 hours and is frequently indecisive for the demonstration of bacteria. Culture requires a minimum of 24-48 hours and may require repetitive cultures or subcultures which result in inhibition of the growth of commensals as well as conditions which favor the growth of suspected pathogens. Direct examination of implant specimens with the scanning electron microscope can show bacteria but the time, special equipment, and hazards associated with preparative techniques such as critical point drying can be prohibitive. Another limitation is the small size of the drying chamber which restricts the specimen size. Nation showed that a rapid procedure for drying insect soft tissues utilizes stepwise dehydration with graded ethanols followed by very brief treatment with hexamethyldisilazane (HMDS). It did not require specialized equipment.

Plant cell infection is tightly controlled by cell surface receptor-like kinases (RLKs). Like other RLKs, the Medicago truncatula entry receptor LYK3 laterally segregates into membrane nanodomains in a stimulus-dependent manner. Although nanodomain localization arises as a generic feature of plant membrane proteins, the molecular mechanisms underlying such dynamic transitions and their functional relevance have remained poorly understood. Here we demonstrate that actin and the flotillin protein FLOT4 form the primary and indispensable core of a specific nanodomain. Infection-dependent induction of the remorin protein and secondary molecular scaffold SYMREM1 results in subsequent recruitment of ligand-activated LYK3 and its stabilization within these membrane subcompartments. Reciprocally, the majority of this LYK3 receptor pool is destabilized at the plasma membrane and undergoes rapid endocytosis in symrem1 mutants on rhizobial inoculation, resulting in premature abortion of host cell infections. These data reveal that receptor recruitment into nanodomains is indispensable for their function during host cell infection.

Abstract Abstract 646 Chromatin modification and remodeling activities play a central role in organizing nuclear functions in eukaryotic genome. Chromatin domains with characteristic epigenetic marks are organized by chromatin insulator. The chicken b-globin insulator, 5′HS4, is an excellent model system to study how insulator maintains gene function and prevents the encroachment of repressive heterochromatin. We showed previously that USF1/2 bound 5′HS4 insulator mediates chromatin barrier activity by recruiting and organizing active histone modifications in the chicken b-globin locus. However, it is not clear how insulator establishes such a barrier. To further understand the physical function of USF1, we purified the USF1-associated protein complexes. We found that USF1 forms a multiprotein complex with hSET1 and NURF exhibiting histone H3K4 methyltransferase and ATP-dependent chromatin remodeling activities, respectively. Both hSET1 and NURF complexes are specifically recruited to the 5′HS4 insulator by USF1 to retain the active chromatin structure. Knock-down of BPTF, a component of NURF complex, resulted in a rapid loss of barrier activity that protects a transgene from chromatin silencing. The loss of barrier by the BPTF knock-down is accompanied by an alternation of nucleosome positioning that expands the nucleosome to the nucleosome free linker region in the HS4 insulator site and increases repressive H3K9 dimethylation. Furthermore, the suppression of hSET1, a H3K4 methyltransferase, leads to not only a loss of barrier activity, but also a loss of H3K4 dimethylation and H3K9/K14 acetylation, as well as the recruitment of BPTF at chromatin insulator sites. Thus, our data reveal a molecular mechanism in which histone modifying enzyme hSET1 and chromatin remodeling complex NURF collaborate together to maintain the 5′HS4 chromatin barrier activity and to prevent the encroachment of adjacent heterochromatin. Tooth and bone specimen require extensive demineralization for careful analysis of cell morphology, as well as gene and protein expression levels. The LacZ gene, which encodes the ß-galactosidase enzyme, is often used as a reporter gene to study gene-structure function, tissue-specific expression by a promoter, cell lineage and fate. This reporter gene is particularly useful for analyzing the spatial and temporal gene expression pattern, by expressing the LacZ gene under the control of a promoter of interest. To analyze LacZ activity, and the expression of other genes and their protein products in teeth and bones, it is necessary to carry out a complete demineralization of the specimen before cutting sections. However, strong acids, such as formic acid used for tooth demineralization, destroy the activities of enzymes including those of ß-galactosidase. Therefore, most protocols currently use mild acids such as 0.1 M ethylene diamine tetra-acetic acid (EDTA) for demineralization of tooth and bone specimen, which require a longer period of treatment for complete demineralization. A method by which hard tissue specimens such as teeth and bones can be rapidly, but gently, decalcified is necessary to save time and effort. Here, we report a suitable method for rapid demineralization of mouse teeth in 0.1M EDTA at 42˚C without any loss of ß-galactosidase activity.

School refusal is a complex phenomenon that has been subject to definitional and diagnostic confusion. The aetiology of school refusal remains incompletely understood; heterogeneity rather than homogeneity prevails. In the behaviour management of school refusal, the clinician may plan either a gradual or rapid school return. Although a graduated return to school may be necessary in certain cases, secondary complications are minimized by an immediate return to school. Both graduated and rapid treatment require a flexible and integrative approach drawing on classical, operant and vicarious conditioning principles. Little research has been conducted on the effectiveness of the behavioural management of school refusal, as well as psychological adjustment to school.
