Abstract. The CALIPSO (Cloud-Aerosol Lidar and Infrared Pathfinder Satellite Observations) level 3 aerosol profile product reports globally gridded, quality-screened, monthly mean aerosol extinction profiles retrieved by CALIOP (the Cloud-Aerosol Lidar with Orthogonal Polarization). This paper describes the quality screening and averaging methods used to generate the version 3 product. The fundamental input data are CALIOP level 2 aerosol extinction profiles and layer classification information (aerosol, cloud, and clear-air). Prior to aggregation, the extinction profiles are quality-screened by a series of filters to reduce the impact of layer detection errors, layer classification errors, extinction retrieval errors, and biases due to an intermittent signal anomaly at the surface. The relative influence of these filters are compared in terms of sample rejection frequency, mean extinction, and mean aerosol optical depth (AOD). The “extinction QC flag” filter is the most influential in preventing high-biases in level 3 mean extinction, while the “misclassified cirrus fringe” filter is most aggressive at rejecting cirrus misclassified as aerosol. The impact of quality screening on monthly mean aerosol extinction is investigated globally and regionally. After applying quality filters, the level 3 algorithm calculates monthly mean AOD by vertically integrating the monthly mean quality-screened aerosol extinction profile. Calculating monthly mean AOD by integrating the monthly mean extinction profile prevents a low bias that would result from alternately integrating the set of extinction profiles first and then averaging the resultant AOD values together. Ultimately, the quality filters reduce level 3 mean AOD by −24 and −31 % for global ocean and global land, respectively, indicating the importance of quality screening.

This study discusses the rejection of the halal label certification on devil noodle products in the perspective of Islamic law and Law no. 33 of 2014. The results of the study concluded that the rejection of the halal label certification application based on SK46/Dir/LPPOM MUI/XII/14 and MUI Fatwa Number 4 of 2003 concerning Standardization of Halal Fatwas because the name of Satan is identic with the name of the enemy of Allah and it is feared that it can distance from remembering Allah and can be categorized as 'urf fasid (bad costome) that must be abandoned. Then, regarding the product name in Law no. 33 of 2014 is not an object that will be examined by halal auditors, but regarding the name and type of product, it is an initial file at the stage of submitting an application, then in article 33 of Law no. 33 of 2014 explains that the determination of product halalness is carried out by MUI in the MUI halal fatwa trial. In line with the conclusion above, the owner of the Surabaya devil noodle product must follow the procedure of LP POM MUI JATIM, namely changing the name of the devil with a good name, then for consumers who consume devil noodle, it is expected that the spicy level is adjusted to the body's ability and not excessive, and for LP POM MUI JATIM to be careful in accepting or rejecting the submission of MUI halal label certification, so that no party feels aggrieved or disappointed.

Recently, sensory threshold concepts have been expanded to include affective perceptions of foods and beverages, especially measures of product rejection; however, each threshold interpretation depends largely on the methodology employed. By substituting cricket powder for whole-wheat flour in snack crackers (0%, 5%, 10%, 15%, and 20%), we proposed modifications to two existing threshold methodologies: a modified consumer rejection threshold (M-CRT) using a 2-alternative choice (2-AC) preference test with “no preference” option; and a modified hedonic rejection threshold (M-HRT) based on less-than-neutral hedonic scores according to a one-sample t-test. We also proposed two new concepts: a rejection tolerance threshold (RTT) and an associated rejection range (RR), based on a binomial acceptability question and a probit regression model. These new methods provide more realistic interpretations of rejection-type thresholds based on user-defined allowable rejection levels, or “rejection tolerance” (for RTT), and a model-derived stimulus range (RR) to capture the rejection level of interest while providing flexibility for product formulation. All thresholds were investigated separately for color, flavor, texture and overall perception, elucidating differential sensorial effects of our novel ingredient on the food matrix. We suggest that conducting all demonstrated techniques in a single testing session gives a more expansive view of rejection thresholds while requiring minimal additional resources.

bstract This study aimed to determine the number of cases, causes, and main rejection factors of Indonesia's fishery products by the USA and European markets. Data were obtained from the websites of USFDA (USA) and RASFF (Europe) from 2010 to 2020. Pareto analysis and fishbone diagram were used for analyzing data that informants validated from selected exporters and experts. Within the observed period, there were 2,318 cases of rejection in the USA and 79 in Europe. The highest case was in 2011 in the USA and 2012 in European markets. Based on Pareto analysis of rejection cases, the main factors that accounted for more than 80% of rejections in the USA were filthy and Salmonella; meanwhile, in Europe, were mercury, poor temperature control, Salmonella, histamine, and cadmium. The fishbone diagram result with validation from fishery product exporter shows that human factor, such as the lack of coordination and communication between business actors, especially at the supplier level when selecting raw materials, was considered the cause of rejection (filthy). Establishing well-managed cooperation among business actors within an integrated fish supply chain management is essential to ensure the quality of fishery products. Reverse osmosis is efficiently used for producing drinking water from groundwater sources containing dissolved impurities, including fluoride, ammonia, lithium, strontium, boron, arsenic, etc. The principal problems of utilizing reverse osmosis include scaling on membrane surfaces, concentrate discharges, and low permeate TDS that often require conditioning. The main goal of this work was to demonstrate the viability of a newly developed methodology that relies on low-rejection nanofiltration membranes to improve product water quality by increasing its TDS and calcium content, and its economic efficiency compared to conventionally used reverse osmosis. Disadvantages of employing reverse osmosis for the production of drinking water are attributed to the fact that several pollutants (including lithium, ammonia, and boron) are monovalent ions and, as such, are poorly rejected by membranes as compared to calcium, sodium, sulfate, and chloride ions. Thus, in cases in which lithium or ammonia are present in high concentrations, high rejection membranes are usually used that result in low TDS of the product water. This article presents the results of research aimed at developing a new approach to changing the ratio of monovalent and divalent ions in product water. The new method described in this paper relies on low rejection membranes in a two-stage application that enables us to reduce monovalent impurities and increase the concentration of calcium and TDS values in product water while leaving lithium concentration unchanged. This is achieved by applying a two-stage scheme with low-rejection membranes instead of the reverse osmosis stage. The two-stage treatment using nanofiltration membranes results in the same rejection of lithium and product water quality as reverse osmosis. However, the ratio value of calcium and lithium concentrations in the concentrate of nanofiltration membranes appears to be significantly higher compared with the ratio measured in the feed groundwater. This can be attributed to different rejections of these ions by membranes. Therefore, concentration (reduction of volume) of the feed water with nanofiltration membranes and further dilution of the concentrate with deionized water produce the same concentration of lithium and are associated with an increase of 2–4 times the concentration of calcium. Treatment of this water in the second nanofiltration membrane stage produces drinking-quality water with the required lithium content and increased calcium concentration. We focus on the real-world example of groundwater treatment in Yakutia, Russia, an area where lithium concentration exceeds drinking standards by 24 times. The paper presents a technique of ion separation and demonstrates experimental results that provide lithium removal while increasing the calcium concentration and TDS value. The resulting concentrations are 2–5 times lower than those obtained via conventional use of reverse osmosis membranes. A series of experiments were conducted to remove lithium from groundwater and demonstrate the efficiency of the newly developed method of ion separation. Experimental results of the concentration of obtained values of lithium, calcium, and TDS in permeate and concentrate flows at each membrane stage demonstrate that they provide separation of monovalent and divalent ions and increase product water TDS without increasing lithium. This experimental approach increases calcium and TDS values in product water by 2–4 times compared with the use of reverse osmosis membranes. Calculations of operational costs for different options (the use of reverse osmosis, two-stage nanofiltration, and ion separation in a two-stage approach) are presented. These results confirm the economic advantage of nanofiltration membrane applications to remove lithium as compared to the use of high-rejection reverse osmosis membranes. The increase in product water TDS facilitates the further reduction of concentrate flow rate and operational costs. The economic comparison involved the calculation of the required membrane area and number of membrane elements at each stage, calcium carbonate scaling rates, reagent consumption to prevent scaling, and amounts of concentrate discharged into the sewer. Experimentally obtained results confirmed the feasibility of increasing the calcium concentration and TDS values in product water by 2–5 times while leaving the lithium concentration at the same level. Design characteristics to calculate operational costs for conventional and new options are calculated and demonstrate a sufficient (30–40%) reduction of operational costs compared to conventional use of reverse osmosis. The reduction in reagent consumption is attributed to the utilization of low-rejection nanofiltration membranes that have lower scaling propensities compared with reverse osmosis membranes and a smaller payment for concentrate discharge. The developed approach to using two-stage nanofiltration instead of single-stage reverse osmosis provides multiple advantages that include improved product water quality, lower concentrate consumption, and lower reagent consumption that are attributable to the use of low-rejection membranes. Different case studies are planned to demonstrate the efficiency of the proposed techniques to reduce ammonia, fluoride, and boron in drinking water.

This work aimed to study the flux behavior, rejection and concentration factors, and energy demand to establish the optimal conditions during ultrafiltration of sweet buttermilk to produce ice cream. The experiments were conducted with a UF25-PAN membrane at a transmembrane pressure of 0.2, 0.35 and 0.5 MPa, and a volume reduction ratio (VRR) of 2, 3, 4, and 5. Total protein, fat, ash, and dry matter contents, phospholipid composition, and acidity of retentates and permeate were determined. The increase in the VRR led to a decrease in the permeate flux and an increase in the energy demand, rejection, and concentration factors of the main components of sweet buttermilk. The highest values of rejection and concentration factors established for fat were 98.65% and 4.93, respectively. The permeate flux and energy demand increased with the rise in the transmembrane pressure. The total phospholipids increased 2.8 times at VRR 5 compared to the initial buttermilk. The use of VRR 3 gave the best ratio between the permeate flux, the energy demand, and relatively high values of concentration and rejection factors. The use of VRR 5 will enrich the ice cream to the greatest extent to obtain a product with the highest level of biologically active substances (proteins, phospholipids, minerals).

Metal casting is a process of metallurgical forming by liquidize of material up to certain high temperature to achieve perfect chemical reaction and high level quality products. Auto pour means pouring the metal liquid into the mould not by manual operation but more than automatically done by a set of machine without any human intervention. This paper aims to describe the process of automation for brake drums where several parameters are compare with manual process. The standard time, cycle time, rejection rate, dimension, chemical composition and product quality have been analyzed. The results shows that the standard time reduce to 40%, rejection rate less 5%, tensile strength 3% higher because of better pouring process. The other parameters are the same for both manual or automatic pouring process. The conclusion of this research is that the auto pour provides better results in term of productivity and quality.

In the present condition, customer demand has increased gradually due to technological development and globalisation in this world. So this made the industrialist to deliver the products at right quantity at right place at right time with shorter lead time. But due to high rejection rate the industries fails to meet the customer demand which in turn reduces the customer satisfaction. Hence, this research an attempt was made to reduce the defects of casting components in an original equipment manufacturer. The study was carried out in a foundry of brake drum manufacturing industry. The problem was inside that the industry could not meet the customer demand due to high rejection rate and the major contributor for high rejection is found to be sand drop. Hence, proposed solutions have been implemented to achieve the objectives. After implementation of proper lean tools it was observed from the results that, the rejection rate was reduced by about 4% and the quality level was increased by about 4.4%.

One of the most attracting production systems that has recently been vastly explored by practitioners and academicians is hybrid make-to-stock/make-to-order. Having a hierarchical production planning structure considered, this paper develops a multi-stage model to cope with the operational decisions, including order acceptance/rejection, product lot sizing, overtime capacity planning, outsourcing, and due date setting. Moreover, the proposed framework also comprises providing alternative products for the coming orders in order to enhance service level of the firm to the customers. In order to validate the presented framework, it is applied in a real industrial case study and the obtained results approve validity of the proposed framework.

The effects of Hibiscus sabdariffa (roselle; Hs) by-product (2–5%) and Pleurotus djamor (pink oyster; Pd) powder (5–7.5%) as meat replacers on the physicochemical and sensorial properties of beef patties were analyzed. The addition of these non-meat ingredients significantly decreased moisture and increased fiber content, and did not affect the protein level of the product. The antioxidant effect of the roselle by-product was limited, while Pleurotus djamor favored the oxidation processes. The samples supplemented with roselle by-product and mushroom powder presented significantly lower microbial counts (total viable counts, enterobacteria, and Pseudomonas) than control, but texture and sensorial parameters were significantly affected. The patties darkened due to the presence of the Hibiscus by-product, while the color of the samples containing 5% Pleurotus djamor was hardly modified. These samples, together with the control samples, were the most sensorially appreciated. The addition of these ingredients provoked a decrease in texture parameters, being less pronounced in the samples with only 2% of roselle by-product. In spite of the good antimicrobial and antioxidant properties of Hibiscus by-products, its inclusion in meat should be moderate (2–2.5%) to avoid consumer rejection when Pleurotus djamor is also included in the formulation.

The countries and regions which suffer from the shortage of fresh water resources have developed sea and brackish water desalination plants to supply drinking water. The desalination process has usually been designed from the rejection ratio of salt to meet the level of soluble residue, chloride concentration for drinking purposes. The Japanese government revised the drinking water quality standards to enhance an appropriate drinking water quality management for many hazardous micro-pollutants in drinking water. Since there has not been sufficient studyies on the performance of R.O. desalination processes on micro hazardous constituents, the authors carried out a pilot plant study and field study of several desalination plants for the public water supply. Most of the constituents of R.O. filtrate meets the drinking water quality standard under the design and operational condition that to attain 99% of the salt rejection. However, Langlier's index, boron and bromoform produced different figures from other inorganic constituents and organic micro pollutants. The rejection ratio of boron ranged only from 43 to 78%. Although the rejectio ratio of boron can be improved by increasing the driving pressure and by adjusting pH, the filtrate cannot meet the drinking water quality standard. Chlorination in the R.O. desalination process produces bromoform that can be removed by the R.O. membrane. However, bromide ion in the filtrate can enhance the formation of disinfection byproducts, if the filtrate is mixed with fresh water, in the distribution system of the public water supply.

Continuous improvement is the prime requirement for all industries to sustain and grow in the competitive global market. This paper is a case study of a manufacturing industry in Taiwan, facing the problem of rejection in the brushless motor product. The DMAIC (Define, Measure, Analyze, Improve, and Control) cycle was used to improve processes to reduce the rejection rate. In the Define phase, anything related to processes, products, suppliers, customers, and customer needs related to good quality products were determined. SIPOC diagrams (Supplier, Input, Process, Output, and Customer) and Critical to Quality (CTQ) were identified. The Measure phase focused on data collection by determining the baseline of the process and determining the root causes of the process. DPMO and the Control Chart were applied in this phase. In the Analyze phase, the causes of production process failures that result in defective products were identified. Tools like Pareto Diagram, Fishbone Diagram, and FMEA were used in this phase. In the Improve phase, the improvement solutions in overcoming priority problems were determined by using the 5W + 1H tool. Several improvement solutions were implemented, such as improvement in inspection methods, re-selection for the supplier, increasing the number of workers, providing training to workers, and others. In the control phase, the Six Sigma values were improved. In January–May 2022, the Six Sigma level increased from 5.11 to 5.44.

Abstract Introduction. Blood is a vital resource, and its use is growing worldwide. Although the demand for blood is high, there are cases in which blood products are rejected. The number of rejected blood components in hospitals is reported to be between 0.1% and 6.7%. (1–5) Objectives. Quality management includes good practices in transfusion activity, an important part of which is limiting the rejection of labile blood products. The objective of this study is to analyze the use and rejection of blood products in the Constanza County Emergency Clinical Hospital between 2012–2021, including 2 years of the Covid-19 pandemic. Material and method. The work presents a retrospective study of transfusion activity over a period of 10 years, with the analysis of units of labile blood products rejected compared to the number of units transfused to hospitalized patients. Results and discussions. In our institution, with a large number of transfused patients and a very large volume of administered blood products, the rejection was on average 1.82% (3,887 rejected products out of the 213,869 received products). Most rejected products were erythrocyte concentrates (1491 units). The good practice measures implemented have led to a decrease of the number of rejected products, with a reduction of the number of rejected units in the last 3 years by an average of 142 units per year. Conclusions. Quality assurance is a legal obligation in the transfusion sector. The implementation of the norms of good practice according to the Order M.S. no.329 / 2018 led to the decrease of the number of rejected products from 2.81% in 2018 to 1.16% in 2021. An increasingly restrictive blood test is used.

Regular exercise has many health benefits, however a major problem in the United States is that Americans do not exercise enough to reap these advantages. Although there are many ways that one can be motivated to exercise, the use of wearable technologies such as fitness tracking devices show great promise as an individual, and cost effective solution. On the other hand, many people who try out these devices return them leading to lower than idea acceptance rates for these devices. We examined online product reviews for wearable fitness devices in order to discover which factors led to product acceptance or rejection. We performed a qualitative analysis of user reviews across many websites and devices followed by a quantitative exploratory analysis using stepwise multiple regression predicting users’ experience. Overall, our results support that four high-level themes: usability, trust, motivation, and wearability determined a user’s experience.

The consumer acceptance of novel enhanced-quality products and their willingness to buy such products may be a crucial topic in the field of marketing. The aim of this study was to analyze the association between consumers’ perceptions of food quality and their acceptance of enhanced meat products and novel packaging. The study was conducted using the Computer-Assisted Personal Interview (CAPI) method in a random group of 1009 respondents, who were recruited as a representative sample based on data from the Polish National Identification Number database. The participants were asked about the most important quality determinants of food products of animal origin and about quality improvement methods and their acceptance of those methods. The quality determinants of animal-based food products were indicated as follows: origin, production technology, manufacturer, components and nutritional value, visual and sensory characteristics, expiry date, and cost. The quality improvement methods were clustered into groups that were associated with product enhancement and application of novel packaging, and the acceptance of those methods was also verified. Indicating specific quality determinants of animal-derived food products affects the consumer acceptance of product enhancement (p = 0.0264) and novel packaging as quality improvement methods (p = 0.0314). The understanding that enhancement is applied for the purpose of quality improvement did not influence the acceptance of products (p = 0.1582), whereas the knowledge that novel packaging is applied influenced the acceptance (p = 0.0044). The obtained results suggested that in the case of application of novel packaging, a higher level of knowledge may be a reason for consumer’s rejection of the resulting products, but the appearance and taste of products may contribute to the higher acceptance of novel packaging. Educating consumers may improve their acceptance of product enhancement, as concerns about the addition of food preservatives may lead them to reject enhanced products.

The ‘insurance ova’ hypothesis (Anderson, 1990) views dizygotic twinning as a by-product of selection for multiple ovulation which sometimes – in error – results in the birth of twins. From this viewpoint, polyovulation is a mechanism which reduces the risks of fertilization failure or embryo defect/mortality. If DZ twin births are a ‘side-effect’ of a mechanism which compensates for defective embryos one might predict that embryo defect rates and twinning rates will covary. This prediction is tested using national-level data on twinning rates and rates of trisomy-21 (Down's syndrome), and a strong positive correlation is found, even when controlling for maternal age. One suggestion that follows from this finding is that intra- and interpopulation variation in both twinning rates and Down's syndrome rates may result, in part, from individual variation in pre-implantation rejection of embryos in the very early stages of pregnancy. In this paper it is proposed that the ‘insurance ova’ explanation for twinning in humans could be expanded to incorporate a model of rejection of anomalous embryos, be they anomalies of number or type. Variation in the efficiency of an embryo rejection mechanism, combined with variation in frequency of polyovulation, would have consequences for individual reproductive success.

The principle of recognition based on the use of the properties of a thin gas layer and the supporting surface of equipment, which provided the creation of a number of universal devices capable of solving the problem of sorting and culling piece parts by weight. A promising method of recognition by the generalized criterion (specific load) is presented on the basis of which two new orientation criteria were obtained (the area of the support surface of the product and its mass) are obtained on the basis of this method. It is proved that the gas layer has a high sensitivity to changes in the characteristic parameters of products, i. e. is able to recognize them. There is a need to develop automatic sorting and rejection devices for a class of small piece products with implicit design features and having understated physical and mechanical properties, as they can not apply to traditional means of automation, loading and orientation operations with mechanical grips, weighing in the stream. There are significant difficulties in the culling of piece products with bevels, shells, etc., which led to the creation of devices that provide manipulation of a wide variety of sizes of specific products with a decrease in dry mechanical friction between the product and the bearing parts of the equipment. Classification of specific product types and configurations of circuits pneumatic devices that use the effects in the thin gas bearing strata (the working surfaces and air chambers, view their movements, technological capabilities). In the manufacture of liquid dyes, almost complete desalting, which helps to improve the stability of the product, enhances the solubility of the dye. Diafiltration is used to allow a high level of desalting to be achieved. The process of desalination of aqueous dye-salt solutions by polymeric nanofiltration membranes using commercially available modules was studied. The influence of dye and salt concentration on the salt rejection and pressure applied on the flux as well as comparison of individual NF membranes for desalting purposes is presented. The great interest is also devoted to the mathematical modelling of nanofiltration and description of discontinuous diafiltration by periodically adding solvent at constant pressure difference.

We model complexity in social programs as a by-product of the screening process. While a more rigorous screening process may improve targeting efficiency, the associated complexity is costly to applicants and induces incomplete take-up. We integrate the study of take-up with the study of rejection (Type I) and award (Type II) errors, and characterize optimal programs when policy makers choose screening intensity (and complexity), an eligibility rule, and a benefit level. Consistent with many real-world programs, optimal programs feature high complexity, incomplete take-up, classification errors of both Type I and II and, in some cases, “excessive” benefits. Currently, reconfigurable hardware devices feature a high density of heterogeneous resources to enable multitasking and offer flexibility in application needs. These concepts raise the need for efficient management of hardware tasks and hardware resources. The scheduling of hardware tasks is highly dependent on placement. Placement focuses on allocation of hardware resources required by the scheduled hardware tasks. In this paper, we propose novel three-level resource management that investigates enhancement of placement quality by reducing task rejection, configuration overheads, and by optimizing resource utilization. Improving placement quality will produce significant enhancement of performance for scheduling and overall execution time of the application in FPGA. Hence, the placement problem is formulated into a constrained optimization problem and resolved with powerful solvers using the Branch and Bound method. The obtained results of an application of heterogeneous hardware tasks show an average resource utilization of 36% of the available resources on the reconfigurable region and an overall overhead of 11% of total application running time, and we have eliminated the issue of task rejection. Compared to static implementation, the gain in resource utilization within the reconfigurable region achieves up to 43%.

The article identifies some trends in the process of transformation of advertising discourse. There is a rejection of traditional advertising practices and a shift in advertising communication towards Internet technologies (banners, pop-up notifications). One of the trends in modern advertising communication is the appeal to new methods and tools that allow achieving the goal of an advertising campaign by varying the impact on the advertising consumer. New forms of advertising communication due to the diversification of information carriers are described. Specific features of the reconfiguration of advertising discourse are established: interactivity, shifting the focus from products as such to brand promotion, various forms of advertising mimicry, «blurring» through meta-discursive practices and referring to the game register, «tightening» as a result of referring to shock advertising strategies. There is a radicalization of institutional advertising, manifested in the re-creation of pathos or dysphoria in the field of anti-smoking advertising campaigns, road safety, etc. It is emphasized that significant changes occur both at the level of creating an advertising product (at the stages of determining the target audience, formulating an advertising message, giving it the appropriate form) and at the level of advertising distribution (in particular, choosing the appropriate media for advertising information).

Every company tries to find ways to maintain its market position in an increasingly competitive business environment. In order to be successful, companies need to achieve their objectives and goals to make their vision a reality while adhering to their mission. Company claims report in the few years still hasn’t solved yet. Such complaints are significant because customersatisfaction can only be improved if the root causes of the problem is clearly identified. In the plastic industry that produces parts for the automotive industry, a systematic method named 8D is used to analyze product conformity to specifications. The 8D method consists of nine disciplines problem-solving. The original purpose of 8D method was to eliminate the defect causing the problem, thus restoring customer satisfaction and level up the quality level of the company. This research established the suitability of the 8D for complaint settlement. 8D involves teamwork to solve the problem and using a 9-step structural approach. This case study resulted that the 8D is effective. The total customer claims at the end of last year until this year in Dec 2020 totaled zero (0) and process rejection for incorrect dimensions were reduced from 0.07 % to 0.01%.

The main objective of this research is to reduce the blowholes by analyzing the factors which are affected during the casting process. The process parameters are optimized and change is made in the design part to reduce the blowhole and to increase the efficiency of the high pressure die casting machines. Product manufactured from every manufacturing process shows some defects. For supplying quality product to the customer these defects must be reduced. In this work, an attempt is made to reduce the rejection due to the blowhole defect is found out through why-why analysis technique. Process capability of current high pressure die casting manufacturing process is checked. Manufacturing process found capable to manufacture the components. Current problem of blowhole defect is solved making an improvement in design of die which we insert. In gate directions are changed so as to obtain modified improved flow pattern. Using magma flow simulationsoftware existing and modified design has then been compared. It is found that, modified design shows superior results and using this, the defect of blowholes is minimized up to satisfactory level.

Linked to potential health problems and toxicity to crops, boron is present in seawater at concentrations of ranging from 4 to 7 mg/L, and not readily removed by reverse osmosis technology. Commercially available seawater reverse osmosis (SWRO) membranes possess a wide range of rejection characteristics for boron in seawater under ambient temperature and pH, ranging from approximately 50% for low-energy membranes to greater than 90% for the newest high rejection membranes. This level of rejection is typically insufficient to reduce boron concentrations in natural seawater to less than recommended levels. Current World Health Organization (WHO) drinking water concentrations for boron are limited to 0.5-mg/L. Two techniques utilized to mitigate boron concentrations are (1) increasing the dissociation of boric acid by increasing pH prior to SWRO; and, (2) utilizing a second pass reverse osmosis system, potentially coupled with pH adjustment. Utilizing these techniques, the authors tested commercially available SWRO membranes from three different manufacturers utilizing feed water alkalization, coupled with a second pass system. Utilizing feed water alkalization alone, the authors found that all three SWRO membranes were able to produce permeate complying with WHO regulations. Using second pass RO, a boron concentration of less than 0.5 mg/L was achieved for feed pH greater than 6, and less than 0.1-mg/L for pH of 10.

Polyamide (PA) membranes possess properties that allow for selective water permeation and salt rejection, and these are widely used for reverse osmotic (RO) desalination of sea water to produce drinking water. In order to design high-performance RO membranes with high levels of water permeability and salt rejection, an understanding of microscopic PA membrane structures is indispensable, and this includes water transport and ion rejection mechanisms on a molecular scale. In this study, two types of virtual PA membranes with different structures and densities were constructed on a computer, and water molecular transport properties through PA membranes were examined on a molecular level via direct reverse/forward osmosis (RO/FO) filtration molecular dynamics (MD) simulations. A quasi-non-equilibrium MD simulation technique that uses applied (RO mode) or osmotic (FO mode) pressure differences of several MPa was conducted to estimate water permeability through PA membranes. A simple NVT (Number, Volume, and Temperature constant ensemble)-RO MD simulation method was presented and verified. The simulations of RO and FO water permeability for a dense PA membrane model without a support layer agreed with the experimental value in the RO mode. This PA membrane completely rejected Na+ and Cl− ions during a simulation time of several nano-seconds. The naturally dense PA structure showed excellent ion rejection. The effect that the void size of PA structure exerted on water permeability was also examined.

Economic data are frequently generated by stochastic processes that can be modeled as realizations of random functions (functional data). This paper adapts the specification test for functional data developed by Bugni, Hall, Horowitz, and Neumann (2009, Econometrics Journal12, S1–S18) to the presence of missing observations. By using a worst case scenario approach, our method is able to extract the information available in the observed portion of the data while being agnostic about the nature of the missing observations. The presence of missing data implies that our test will not only result in the rejection or lack of rejection of the null hypothesis, but it may also be inconclusive.Under the null hypothesis, our specification test will reject the null hypothesis with a probability that, in the limit, does not exceed the significance level of the test. Moreover, the power of the test converges to one whenever the distribution of the observations conveys that the null hypothesis is false.Monte Carlo evidence shows that the test may produce informative results (either rejection or lack of rejection of the null hypothesis) even under the presence of significant amounts of missing data. The procedure is illustrated by testing whether the Burdett–Mortensen labor market model is the correct framework for wage paths constructed from the National Longitudinal Survery of Youth, 1979 survey.

Recent statistical analyses of the clinical outcome have revealed that the presence of donor-specific anti-HLA antibodies (DSAs) in pre-transplant recipient is correlated with increased risk for graft rejection after allogeneic hematopoietic stem cell (HSC) transplantation. However, while cytotoxic T lymphocytes (CTLs) recognizing mismatched HLA molecule of the donor have been shown to be involved in graft rejection, there has been no biological evidence in humans that graft rejection is mediated by DSAs. In the present study, we demonstrate a case of cord blood allograft rejection in which DSAs acted to mediate graft rejection. Interestingly, CTL clones specific for donor HLA molecule were also isolated, suggesting that humoral and cellular immune responses were together responsible for allograft rejection. A patient with graft rejection after HLA-mismatched cord blood transplantation was studied. The white blood cell count increased transiently, subsequently decreased to undetectable level, and finally graft rejection was diagnosed on day 34. Screening for pre-transplant anti-HLA antibodies was not routinely performed at that time. We initially presumed that DSA would be responsible for graft rejection. The patient serum on day 36 were screened for HLA antibodies and further evaluated to determine HLA specificities using a LABScreen Single Antigen Kit. An antibody against HLA-B* 54:01, which was expressed in donor cells but not in patient cells, was detected. To test the effect of the DSA against HLA-B* 54:01 on HSC engraftment, antibody-dependent cellular cytotoxicity (ADCC) activity was assessed using unrelated HLA-B* 54:01-positive bone marrow mononuclear cells (BMMNCs) pre-cultured with patient serum and pre-transplant NK cells. The patient serum clearly showed an inhibitory effect on colony formation. Complement-dependent cytotoxicity activity of the DSA could not be detected. These data suggest that the DSA impaired the cord blood engraftment through ADCC activities. We next determined if cellular immunity was also responsible for allograft rejection. Two independent CTL clones, CD4-CD8+ and CD4+CD8-, were isolated from the patient blood at the time of graft rejection by limiting dilution. Both CTL clones were of patient origin by using short tandem repeat analysis and showed cytotoxicity against donor cells but not patient cells in Cr release assay. A CTL stimulation assay using COS cells transfected with various mismatched donor HLA cDNA constructs demonstrated that CD8+ and CD4+CTL clones recognized HLA-B* 54:01 and -DRB1* 15:02 molecules, respectively, both of which were expressed in donor cells but not in patient cells. To test the effect of the CTL clones on engraftment, a colony forming assay using either HLA-B* 54:01 or -DRB1* 15:02-positive unrelated BMMNCs pre-cultured with the corresponding CTL clones was performed. Each of these CTL clones inhibited colony formation from unrelated BMMNCs sharing target HLA in a dose-dependent manner. These data suggest that the CTLs also impaired the cord blood engraftment. Furthermore, anti-HLA-B* 54:01 antibody was detected in cryopreserved pre-transplant patient serum, and in nested PCR assays specific for the HLA-B* 54:01-specific CTL clone’s uniquely rearranged T cell receptor V beta chain, PCR product was detected by amplification of cDNA from pre-transplant patient peripheral blood mononuclear cells. Thus, both HLA-B* 54:01-specific antibody and CTL clone developed in the patient prior to transplantation. The present data provide the first direct evidence showing that humoral and cellular immune responses were involved in the graft rejection. Of note is the recognition of the same mismatched HLA-B* 54:01 molecule by the DSA and CTL, suggesting that humoral and cellular immune processes acted together to mediate allograft rejection. Given the difficulty in detecting HLA-specific CTLs in pre-transplant patient blood in contrast to the easiness in screening for DSAs, the presence of DSA not only means a direct deleterious effect on donor cells but it may also reflect the presence of CTLs that causes allograft rejection. Further studies are warranted to clarify whether the present observation can be duplicated in other patients with DSA. We are conducting the study to clarify whether patients with anti-HLA antibodies have CTLs that recognize the same HLA molecules before transplantation. Disclosures: No relevant conflicts of interest to declare.

Promoting quality within the context of agile software development, it is extremely important as well as useful to improve not only the knowledge and decision-making of project managers, product owners, and quality assurance leaders but also to support the communication between teams. In this context, quality needs to be visible in a synthetic and intuitive way in order to facilitate the decision of accepting or rejecting each iteration within the software life cycle. This article introduces a novel solution called Product Quality Evaluation Method (PQEM) which can be used to evaluate a set of quality characteristics for each iteration within a software product life cycle. PQEM is based on the Goal-Question-Metric approach, the standard ISO/IEC 25010, and the extension made of testing coverage in order to obtain the quality coverage of each quality characteristic. The outcome of PQEM is a unique multidimensional value, that represents the quality level reached by each iteration of a product, as an aggregated measure. Even though a value it is not the regular idea of measuring quality, we believe that it can be useful to use this value to easily understand the quality level of each iteration. An illustrative example of the PQEM method was carried out with two iterations from a web and mobile application, within the healthcare environment. A single measure makes it possible to observe the evolution of the level of quality reached in the evolution of the product through the iterations.

To compete effectively in the global marketplace of the twenty-first century, manufacturing companies are trying to maintain a high level of flexibility and responsiveness to achieve agility and to remain competitive. Manufacturers are under tremendous pressure to improve productivity and quality while reducing costs. The new competition is in terms of reduced cost, improved quality products with higher performance, a wider range of products and better services all delivered simultaneously to enhance value to customers. In such environment providing good quality product at low cost for a medium scale industry has become very tough. To provide good quality product at low cost, small industries need a formulation of some manufacturing approaches like agile manufacturing to manufacture defect free products within their materials cost limit. Medium scale steel manufacturing industries like Magnum steel limited (MSL), banmore are facing problems of higher rejections in form of wastes so as to increase their cost. This paper aims to analyze the application of agile manufacturing in magnum steel limited in order to reduce wastages through implementing lean tools and techniques. An agility audit questionnaire is used for assessing the agility level of the company to identify the current level of performance within the company with respect to the key elements of agility. MSL’s agile experience is reported including a list of recommendation for improving its competitiveness to offer solution alternatives not only to the current problems but also to the ones that may be encountered in the future.

The consumption of cookies is widely extended throughout the world, although their formulas contain ingredients such as saturated fats or refined flours that are considered harmful to health. In addition, cookies are generally made from wheat flour, and nowadays there is a growing concern about gluten intolerance, thus the demand for gluten-free products is increasing. In this regard, the aim of the present study is to reformulate traditional cookies by replacing wheat flour and butter by flours and oils from nuts and seeds. Within these seeds, poppy or chia are not commonly consumed ingredients as they can cause rejection by consumers. Thus, a study was performed to evaluate the neophobia level of consumers and the consumer acceptance for the inclusion of these novel ingredients in cookies. The results have been obtained by measuring physical parameters, proximate composition and consumer evaluation of five batches of cookies. By replacing butter and wheat flour with maize flour, almond, walnut, chia or poppy seed flours and oils, an increase of protein, fat and fiber has been observed as well as a decrease in carbohydrate content; thus, the resultant cookies would be a good source of vegetal protein as well as a source of oleic and linoleic acid with potential benefits on health. The cookies in general have similar physical properties and a positive consumer acceptance in texture, taste and external aspect. The Food Neophobia Scale results suggest that non-neophobic consumers gave higher scores than neophobic consumers in all the parameters. The resultant product would be a functional product able to substitute traditional ones not only directed to celiac people but all type of consumers because of their beneficial composition.

In 1999, Lola und Bilidikid emerged as the creative product of Turkish-born director, Kutluğ Ataman, and provided a rare example and highly nuanced exploration of homosexuality in a Turkish-German context. This article explores the film’s representation of a group of characters marginalised not only on the level of their cultural background, but also on the level of sexual identity. Through a close analysis of the film, various demonstrations of and responses to homosexuality as a sub-culture within a patriarchal, migrant (and therefore already marginalised) group will also be explored, including particularly the notions of drag and performance. These responses will be analysed from the perspective of the protagonists, who are themselves not only both gay and of Turkish origin, but also demonstrate almost fundamentally conflicting approaches to homosexuality. This is particularly evident in the varying degrees to which they either accept or reject their own homosexuality. The article will also explore the consequences of the acceptance and rejection of their respective homosexualities and the extent to which this informs the construction and/or perpetuation of a particular performance of masculinity.

To investigate the effect of competitive incentives under peer review, we designed a novel experimental setup called the Art Exhibition Game. We present experimental evidence of how competition introduces both positive and negative effects when creative artifacts are evaluated and selected by peer review. Competition proved to be a double-edged sword: on the one hand, it fosters innovation and product diversity, but on the other hand, it also leads to more unfair reviews and to a lower level of agreement between reviewers. Moreover, an external validation of the quality of peer reviews during the laboratory experiment, based on 23,627 online evaluations on Amazon Mechanical Turk, shows that competition does not significantly increase the level of creativity. Furthermore, the higher rejection rate under competitive conditions does not improve the average quality of published contributions, because more high-quality work is also rejected. Overall, our results could explain why many ground-breaking studies in science end up in lower-tier journals. Differences and similarities between the Art Exhibition Game and scholarly peer review are discussed and the implications for the design of new incentive systems for scientists are explained.

Sirolimus is an immunosuppressive drug used to prevent graft rejection. Therapeutic drug monitoring is required as with other immunosuppressive drugs. Previous studies have shown the interactions between sirolimus and drugs that affect the activity of cytochrome P450 3A4 and P-glycoprotein. There is an increasing tendency for the use of herbal remedies in many countries. Medicinal herbs are rich sources of natural bioactive compounds that could interact with drugs. Parsley, Petroselinum crispum, is a food, spice, and also a medicinal herb. We report a case of a renal transplant recipient who had a supratherapeutic blood level of sirolimus due to consuming excessive parsley to highlight a possible herb–drug interaction. This is the first case report describing sirolimus–parsley interaction. Herb–drug interactions are especially important for drugs with a narrow therapeutic window. For this reason, healthcare professionals should question all patients, especially transplant patients, about the use of herbs or herbal products and report interactions. Plain Language Summary Parsley, a commonly consumed food, affects the level of an important drug in a renal transplant recipient: A case report Sirolimus is a drug that suppresses the immune response used to prevent organ rejection in people who have had kidney transplants. In order to reach the optimum balance between therapeutic efficacy and adverse effects, sirolimus blood levels should be closely monitored. AimThe aim of this audit was to establish whether immunosuppression was being prescribed correctly and whether target levels were being reached during the first six weeks post-transplant.1 2MethodThe standards were discussed and agreed, due to an absence of standardised local or national written protocols, with the lead paediatric cardiothoracic transplant consultant and a specialist transplant liaison nurse. The paediatric transplant database provided a list of patients between October 2016 and July 2018, from which paediatric cardiac transplant patients were included in this audit. All data were collected retrospectively, for the first six weeks post-transplant, from patient’s electronic records.ResultsTwenty-three patients were included in the audit; fifteen males and eight females and the mean age was 6 years old. The standards for the timing and dosing of the first ciclosporin dose were met for 87% and 78% of patients respectively. Six patients (26%) had a ciclosporin level within the target range by day 4 post-transplant, for the remaining seventeen patients the average was day 9 post-transplant. The mean levels remained within this range or slightly above after day 9. Azathioprine or mycophenolate was started within 7 days of transplant in 6 patients (23%). Four patients (17%) had documented episodes of rejection; in one patient all other standards were met and in the other three only one additional standard was not met. Nineteen patients (83%) did not have a documented episode of rejection.ConclusionsPost-transplant management is individualised based on multiple factors such as clinical conditions e.g. renal/liver impairment and whether other agents such as ATG or steroids are being used.1 The lack of documentation around the treatment decisions made it difficult to explain deviations from standards in this audit. Ciclosporin standards were not met completely but were most likely unfeasible due to a narrow target range and the time between first dose and level monitoring. There did not appear to be a clear association between standards not being met and episodes of rejection. It would be beneficial to repeat this as a larger, prospective audit using revised standards.ReferencesCostanzo MR, Costanzo MR, Dipchand A, et al. Xenotransplantation of pig organs receives substantial attention for being comparable to human’s. However, compatibility constraints involving hyper-acute rejection (HAR) still block clinical applications. Transgenesis of human complement regulatory proteins has been proposed to overcome xenorejection. Pigs expressing human-CD55 have been widely tested in experimental surgery. Still, no standardized method has been developed to determine tissue expression of human decay-accelerating factor (DAF), hCD55’s product, or to predict the ability to overpass HAR. Here we describe objective procedures addressing this need. Organs and tissues from five hCD55 transgenic pigs were collected and classified according to their xenotransplantation value. The ability to overcome HAR was assessed by classical complement pathway hemolysis assays. Quantitative PCR mRNA expression and Western blot protein level studies were performed. Real-time cytotoxicity assays (RTCA) on fibroblast cultures exposed to baboon and human sera informed on longer-term rejection dynamics. While greater hCD55/DAF expression correlated with better performance, the results obtained varied among specimens. Interestingly, the individual with highest mRNA and protein levels showed positive feedback for hCD55 transcript after challenge with human and baboon sera. Moreover, hCD55 expression correlated to DAF levels in the liver, lung and intestine, but not in the heart. Moreover, we found significant correlations among valuable and non-valuable tissues. In sum, the methodology proposed allows us to characterize the hCD55 transgene functioning and performance. Moreover, the correlations found could allow us to predict hCD55/DAF expression in surrogate tissues, thus eliminating the need for direct biopsies, resulting in preservation of organ integrity before xenotransplantation.

The aim of this research is to review the relationship between university students’ resilience and levels of social exclusion and forgiveness. Study group of the research includes 355–206 (58%) female and 149 (42%) male–students who marked at least one item in Risk Factors Determination List. This study is a correlational survey model. The Resilience Scale, The Risk Factors Determination List, The Social Exclusion Scale for Adolescents and Forgiveness Scale are used as data collection tools. Pearson Product-Moment Correlation coefficient and Multiple Linear Regression Analysis are used in data analysis. In the wake of correlation analysis, a significant relationship cannot be found between resilience and forgiveness level. A negative and significant relationship is found between resilience and exclusion and negligence sub-dimensions of social exclusion. In the wake of regression analysis, sub-dimensions of social exclusion predict resilience. In order to increase the resilience of university students, rejection by their friends should be minimized, and in order to prevent individuals from being exposed to social exclusion, communication skills can be improved. Social support, which is among the protective factors of resilience, has an important place in life of university students. Therefore, social activities that every student can participate in can be hold.

The world’s growing population is highly dependent on animal agriculture. Animal products provide nutrient-packed meals that help to sustain individuals of all ages in communities across the globe. As the human demand for animal proteins grows, the agricultural industry must continue to advance its efficiency and quality of production. One of the most commonly farmed livestock is poultry and their significance is felt on a global scale. Current poultry farming practices result in the premature death and rejection of billions of chickens on an annual basis before they are processed for meat. This loss of life is concerning regarding animal welfare, agricultural efficiency, and economic impacts. The best way to prevent these losses is through the individualistic and/or group level assessment of animals on a continuous basis. On large-scale farms, such attention to detail was generally considered to be inaccurate and inefficient, but with the integration of artificial intelligence (AI)-assisted technology individualised, and per-herd assessments of livestock became possible and accurate. Various studies have shown that cameras linked with specialised systems of AI can properly analyse flocks for health concerns, thus improving the survival rate and product quality of farmed poultry. Building on recent advancements, this review explores the aspects of AI in the detection, counting, and tracking of poultry in commercial and research-based applications.

Abstract Targeted intravenous (IV) Busulfan (TBu) in combination with cyclophosphamide (Cy) is widely used as a preparative regimen in children with thalassemia major undergoing matched allogeneic haematopoietic stem cell transplantation (HSCT) to reduce toxicity. At our centre, we have been using TBu/Cy regimen since 2011 for very young children (Pesaro class I/II risk status) and with matched related donors. We have compared the outcome of HSCT in this group of patients with a retrospective cohort (from 1995-2009) of age and risk status matched patients receiving fixed dose oral Bu without PK monitoring. All patients with beta thalassaemia major undergoing HSCT with matched related donor between January, 2011 - May, 2018, receiving IV Bu (0.8mg/kg Q6H days x 4 days) / Cy (50mg/kg x 4 days) with anti-thymocyte globulin based conditioning regimen were included in this study. Busulfan levels were monitored after the 1st dose of busulfan and further doses adjusted to achieve a target range of 900-1300um*min. The Bu plasma levels achieved on day 1 and on day 3 were compared with HSCT outcome endpoints including chimerism status on D28, overall and event-free survival (OS, EFS), and graft rejection. There were 52 children, median age of 3 years (range:1-6); 44 class I/II and 8 class III low risk. Different proprietary Bu products were used: 26 received Bucelon™ (Celon Labs, Hyderabad, India), 23 patients received Buslera™ (Biem Pharma, Ankara, Turkey), and 3 received Bufatas™ (Intas Pharma, Ahmedabad, India). The 3rd dose of Bu was increased in 35, decreased in 2 and unchanged in 15 patients. The median 1st dose Bu AUC was 625 um*min (range: 115- 2466) while the 9th dose AUC was 1105 um*min (range: 543-2656). Target Bu AUC was achieved in 40 patients (77%) as assessed by Bu AUC on Day 3 while the AUC in the remaining 12 patients (23%) was lower than 900 um*min (range: 543-872). Twenty-three of the 50 evaluable patients showed mixed chimerism (MC) or rejection on day 28 (46%; MC level 1 - 14; MC level 2 - 3; MC level 3 - 2 and >90 recipient chimerism - 4, as per our previous data- Fouzia et al, BMT, 2017). 11/23 (47%) had graft rejection; The OS and EFS were 96% and 79% respectively. Two patients died - 1 class III, of diffuse alveolar hemorrhage/ idiopathic pulmonary syndrome and the other of complications related to primary graft failure. Correlation of PK with all demographic variables by univariate analysis did not reveal any significant associations. However, while 11 out of 39 patients (28%) with 9th dose Bu AUC in the lower three quartiles rejected the graft (Q1: <913 (543-906)-3rej; Q2: 936-1100 -2rej; Q3: 1110-1271- 6rej), none of the 13 patients (0%) in the highest quartile (>1292 (1299-2656) um*min) rejected the graft (p=0.034). While none of the 13 patients in Q4 died, 7 had hepatotoxicity (grade 2 and above) and mucositis (grade 2 and above). The incidence of hepatotoxicity in the first three quartiles were as follows: Q1: 11 had grade 2-3 hepatotoxicity and 2 had grade 2-3 mucositis; in Q2: 5 had grade 2-3 hepatotoxicity while 7 had grade 2-3 mucositis; Q3: 3 had grade 4 hepatotoxicity while 4 had grade 2-3 mucositis (p=ns with respect to toxicity in Q4 vs. the other 3 quartiles). There was no significant association between the busulfan formulation used and the incidence of graft rejection. We then compared the HSCT outcome parameters in these patients with age and Pesaro class matched retrospective cohort of thalassemia patients (n=79) who had received a similar conditioning regimen but with oral Bu without PK guided dose adjustment. There was a significantly better OS (Fig A) in the TBu cohort compared to the oral Bu (p=0.03) but this did not translate to better EFS (Fig B) due to increased incidence of graft rejection (Figure C). In conclusion, our data suggests that targeting higher Bu AUCs within the therapeutic window could reduce the risk of graft rejection and improve OS without increasing toxicity. Strategies for rapid dose adjustments after the first dose PK are needed to better achieve these target values to reduce rejections and improve outcome. Figure. Figure. Chimeric Antigen Receptor (CAR) T cell therapy has generated unprecedented efficacy in the treatment of multiple hematologic malignancies. For relapsed/refractory Multiple Myeloma (MM), autologous CAR-T products directed against the B cell maturation antigen (BCMA), such as Poseida's P-BCMA-101, have demonstrated significant efficacy. P-BCMA-101 is comprised of a high-percentage of stem cell memory T cells (TSCM), resulting in a product that is much safer and potentially more durable than other anti-BCMA autologous product candidates. However, as individualized products, all autologous CAR-T products are expensive to manufacture and dependent upon patient T-cells of variable quality. We are developing P-BCMA-ALLO1, an off-the-shelf allogeneic (allo) BCMA-specific CAR-T product candidate derived from healthy donor material, which provides numerous advantages over autologous products, increasing patient access by being immediately available and greatly reducing manufacturing cost and variability. P-BCMA-ALLO1 is produced using two key platform technologies: the nonviral piggyBac® (PB) DNA Modification System and the high-fidelity Cas-CLOVER™ (CC) Site-Specific Gene Editing System. The mRNA coding for hyperactive, or "Super PB" transposase (SPB), and CC enzymes are codelivered with the P-BCMA-ALLO1 PB-based DNA transgene via electroporation to healthy donor T cells to stably integrate the transgene, as well as to knockout (KO) several mediators of allo graft-versus-host and host-versus-graft responses to maximize patient safety and durability of response. The P-BCMA-ALLO1 transgene encodes three genes, a BCMA-specific single-domain variable heavy chain (VH)-CAR (VCAR) gene, a drug selection gene to generate a ~100% CAR+ product, as well as a caspase-based safety switch gene to reduce or eliminate the product in vivo, if desired. The CC System is used to KO the endogenous T Cell Receptor (TCR) and beta-2 microglobulin, thereby decreasing Major Histocompatibility Complex (MHC) class I expression. KO of these key targets is aimed to prevent graft-versus-host disease, as well as reduce host-versus-graft rejection of the product. The CC System can efficiently edit resting T cells, thereby maintaining a high-percentage of TSCM cells, and does not create unwanted off-target mutations, another important consideration when creating an allo product candidate. To maximize the number of doses produced from a single manufacturing run, we have developed a proprietary "booster molecule" that allows for significant expansion of TCR-KO CAR-TSCM cells to potentially produce hundreds of doses. To date, large-scale manufacturing of significant doses of potent allo CAR-T products has been challenging for the field. P-BCMA-ALLO1 manufacturing uses a potentially unlimited number of individual serial donors. We have currently produced P-BCMA-ALLO1 at both research and near-commercial scale from >35 donors with >97% manufacturing success. While a range of TCR-KO efficiencies was observed (~50-90%), the final product was always >99% homozygous TCR-KO after a purification step. Overall expansion of TCR-KO cells ranged from ~2-20 fold, and after removal of unedited TCR+ cells ~0.42-7.04x10e9 TCR-KO cells were recovered from 0.75x10e9 starting cells. However, working at clinical production scale (starting with ~3x10e9 cells), up to 250 doses of P-BCMA-ALLO1 could be manufactured per run, at a dose of 150x10e6 cells/patient. Importantly, with this level of donor and manufacturing robustness, no significant prior screening of donor material, other than to meet standard FDA requirements, would be needed. P-BCMA-ALLO1 made from multiple donors were comprised of an exceptionally high-percentage of the desirable TSCM cells (CD45RA+CD62L+CD45RO-) and had minimal to no expression of exhaustion markers, such as PD-1 or Lag3. Furthermore, P-BCMA-ALLO1 demonstrated potent efficacy in the RPMI-8226 xenograft model in NSG mice across multiple products generated from separate individual healthy donors. Altogether, these data demonstrate a robust, reproducible and highly scalable manufacturing process. Moreover, this manufacturing process can easily be expanded for use with additional CAR targets for treatment of other hematologic or solid tumor malignancies. This study aims to know whether there is a correlation between students’ Quranic memorization and their English vocabulary retention. This study chose 28 of seventh graders at Islamic Junior High School Az-Zahra Lampung in 2018/2019 academic year as the sample by means of cluster random sampling technique. This study applied correlational research design. The documented data of Quranic memorization of learners and the English vocabulary preservation were collected from the test and then analyzed for hypothetical testing employing Pearson's Product Moment equation. The outcome of the hypothetical test shows that Sig. (value) was generated by the value of significance = 0.000 < α = 0.05. It means the acceptance of Ha and the rejection of H0. Furthermore, based on the r value interpretation table, it is known that the observed r is a high correlation as the value of observed r is 0.622 in the 0.600 – 0.800 level. It can therefore be inferred that there is a significant correlation between the Quranic memorization of learners and the acquisition of English vocabulary.

This study investigated the influence of social status on academic performance of secondary school students with disabilities in Kwara state. The descriptive survey of a correlational type was adopted for this study. The population for the study comprised all secondary school students with disabilities in Kwara State, while 200 students with disabilities constituted a sample for the study. Social Peer Rejection Scale developed by Asher et al.’s (2001) was adapted for this study. The adapted scale was validated and has a reliability coefficient of 0.89. The data collected were analysed using percentage for demographic data and research questions while the two null hypotheses formulated were tested at 0.05 level of significance using Pearson’s Product Moment Correlation (PPMC). The results of the findings revealed that 87.0% (n=147) of the secondary school students with disabilities were social rejected and as a result, it has negative influence of the academic performance. The findings also showed that social acceptance influences academic performance of students with disabilities in Kwara State. Implications for counselling practice and recommendations were also made.

PT X is a manufacturer of cardboard box whose products are indispensable for various fields. The problem identified in NH1X36B pre-printed box, which is a shoes box, is the high defect rate that exceed company target of 2%. This study aims to reduce the defect rate of the product. The Quality-Management (QM) and Cross Industry Standard Process for Data Mining (CRISP-DM) approach was conducted by integrating Six Sigma and data mining. The Business Understanding phase was intended to define business and data mining objectives, SIPOC (Supplier-Input-Process-Output-Customer) diagram, and Critical-to-Quality (CTQ). In Data Understanding phase, it is known that the Defects Per Million Opportunities (DPMO) value is 1210.12. Data preparation phase was carried out with data cleaning, reduction, and discretization. Based on the Modeling result using Decision Tree C4.5 and FP-Growth algorithm, it is known that the dominant attributes causing high rejection are smeared ink, white spots, uneven varnish, and delamination. Decision Tree model accuracy of 90.24% indicates that the model is performing well. Analysis using FMEA yielded priority correction to the causes of smeared ink, uneven varnish, and delamination. Process improvement in Deployment phase was the application of plate cleaning and mounting form, printing process checklist, and SOP for sheet inspection. The improvement plans managed to improve the quality by rising sigma level from 4.533 to 4.648 sigma and decrease defect rate to 1.559%.

Dimethyl ether (DME), derived from methanol, has been issued as an alternative to diesel and LPG. This study will continue the research of an optimized process control design for DME purification plants, specifically the methanol purification process. This way, water can be separated, and the main product, methanol, can be recycled for DME synthesis. A setpoint-based linear model of a methanol–water purification system has been developed with four controlled variables (CV) with objectives to; maintain a separated liquid water stream in the bottom stage (Stage 30) of the distillation column for methanol–water separation at 11.22%, keep the purified liquid methanol at a condenser at 58.81 °C and 49.96% of level, and the last CV is the cooler in the distillate stream, to keep the purified methanol’s top product at 40.75 °C. To complete the model, a first order plus dead time (FOPDT) disturbance model is created against the inlet temperature and flow rate of the feed, the major cause of disturbances in the industry. Using a traditional proportional integral controller connected to each controlled variable, a multi-loop control system is formed with optimization and compared to the disturbance rejection of multivariable model predictive control (MMPC). The final improvement against the feed temperature and flow for CV1, CV2, CV3, and CV4 is shown by, respectively, Integral Absolute Error (IAE) values of 79.49%, 99.90%, 100%, and 99.99% and Integral Square Error (ISE) values of 97.1%, 100%, 99.99%, and 100%.

Nanofiltration, used in the Méry-sur-Oise water treatment plant (90 MGD) provides a softened water with a low organic matter content from which micropollutants and microorganisms have been removed. The experience acquired on the site since 1992 has revealed that the use of a crystal formation inhibitor is essential to slow down the clogging of the nanofiltration membranes, essentially on the third stage. Given the complexity of the action of the antiscalants (threshold effect, metal ion sequestration capacity, particle dispersion capacity), it is difficult to theoretically define the type of product to be used and its optimal dosing level. In addition to laboratories or modelling studies, pilot experiments are needed to assess antiscalants and optimise their doses. Nanofiltration cycles being long (1-2 months), it is nevertheless difficult to compare results of cycles, and the objective of the project was to develop a methodology for sequestering agent testing. To carry out strictly comparative studies of commercially available products as well as studies using the same product but at several different dosage levels, a pilot unit comprising three parallel nanofiltration trains was built. The pilot unit is fed with 2nd stage concentrate, produced without sequestering agent, and each trains has its own separate antiscalant injection point. The first cycle aimed to provide answers to the following: (i) choice of the antiscalant best adapted to the treatment of water from the river Oise, (ii) identification of the clogging agents, (iii) the efficiency of the chemical regeneration procedure. A production cycle using the same antiscalant at three different doses (0, 1 and 3 g.m-3) demonstrated that this product acted through a threshold effect. Analysing the cleaning waters provided further information on the type of clogging agents deposited on the membrane. Although for all the cycles the overall nature of the clogging is identical, essentially composed of organic matter and mineral elements (calcium, phosphorus, aluminium, etc.). The similarity of the water permeability and salt rejection at the beginning of two consecutive cycles tends to demonstrate that the chemical regeneration procedure (caustic soda/citric acid) is efficient.

In the manufacturing firms, hospitals, banks, schools and research institutes etc., total customer satisfaction with products and services have very much importance and also necessary for their survival. Quality services and reducing wastages of all types are the primary need of every organisation. Quality Circle is (QC) is one of the techniques that can be utilised to solve industrial problems and can reduce the rejection level. In present paper, QC concept is implemented to solve wastage problem by taking a real case study of an automobile industry. The observed data was analysed through various problem solving techniques such as 80/20 rule and fishbone diagram etc. Finally, Why-Why analysis was carried out to identify the root cause of the problem and suggested actions to solve the problem. The following results were obtained: Gradually reduction in materials wastage minimises the rejection and hence reduces the overhead expenses of the firm. Further, QC helps in motivating employees to do their best for the organisation, which not only improves the productivity of the organisation, but also helps in discovering hidden talent and creative skill of the workforce. Finally, QC helps in developing the sense of belongingness towards organisations.
