The influence of driver's rage on driving safety is one of the centers of traffic accident research. Road rage is a common potential factor affecting driving safety. Real-time monitoring of driver's emotional state and timely intervention measures can effectively reduce the incidence of traffic accidents. At present, the reliability of driver emotion recognition based on single consistent factor for road rage needs to be further improved. Therefore, based on the multi-feature fusion method, this paper proposes a recognition method of driver's road rage emotional state that integrates facial expression, driving control, voice text and physiological characteristics, and proposes the analysis of fusion strategy. In order to improve the reliability and implementability of road rage symptom judgment, this paper studies the state identification methods of road rage and integrates the characteristics of road rage. The focus of this study is to solve the key problem of insufficient reliability of road rage recognition based on facial expressions through the combination of multiple features and multiple data, aiming at the defects of the study on the single feature of road rage, and effectively improve the accuracy of road rage state emotion detection.

AbstractThe receptor for advanced glycation end-products (RAGE) is a multi-ligand receptor which belongs to the pattern recognition receptor family and can bind to various ligands such as advanced glycation end-products (AGEs), members of the S100 protein family, glycosaminoglycans, amyloid β peptides, high-mobility group box-1 (HMGB1) and nucleic acids through its extracellular domain. The RAGE-ligand interaction leads to the activation of MAP kinase and NF-kB signaling pathways. Further ligand-induced up-regulation of RAGE is involved in various patho-physiological situations including late diabetic complications, Alzheimer disease and several other neurodegenerative diseases. A secreted soluble isoform of RAGE (sRAGE), corresponding to the extracellular domain only, has the ability to block RAGE-associated cellular activation and signaling. Further application of recombinant sRAGE has been shown to block RAGE-mediated pathophysiological conditions in various models of cancer or multiple sclerosis. These finding demonstrates sRAGE as a therapeutic tool to block RAGE-associated inflammatory signaling. In this manuscript, we describe a two-step simple, novel and convenient method for expressing and purifying scalable quantities of biologically active murine form of sRAGE by using E.coli as an expression host. The method we propose has several advantages over the current available methods particularly in terms of yield and quality of preparation. The sRAGE produced by this expression system retains all the secondary structural properties as analyzed by the ligand binding affinities. The produced protein also retains all the DNA-RAGE binding functional properties and thus can be a valuable tool for studying dynamics of this novel RAGE ligand. Moreover this method can be utilized by researchers to generate biologically active endotoxin-free sRAGE for in vivo applications to study and treat RAGE-associated pathologies.

AbstractExamining two sets of archived materials that include a corpus of narratives that reflect on the period of apartheid in South Africa and posters used by anti-apartheid activists, the paper teases out the operations of racism and the manifestations of rage on the Black body. Critical discourse analysis and affect as theory and method are applied to trace the work of racism and its affective consequences and resistances. Here affect is deployed to read the terrain of the corporeal and the discursive. Black rage is seen as a response to White supremacy and it has the following outcomes: it can have destructive consequences, can enable psychological release of pent up anger, and can simultaneously be an expression of self-love.

In mammals, glycated serum albumin (gSA) contributes to the pathogenesis of many metabolic diseases by activating the receptors (RAGE) for advanced glycation end products (AGEs). Many aspects of the gSA–RAGE interaction remain unknown. The purpose of the present paper was to study the interaction of glycated human albumin (gHSA) with RAGE using molecular modeling methods. Ten models of gHSA modified with different lysine residues to carboxymethyl-lysines were prepared. Complexes of gHSA–RAGE were obtained by the macromolecular docking method with subsequent molecular dynamics simulation (MD). According to the MD, the RAGE complexes with gHSA glycated at Lys233, Lys64, Lys525, Lys262 and Lys378 are the strongest. Three-dimensional models of the RAGE dimers with gHSA were proposed. Additional computational experiments showed that the binding of fatty acids (FAs) to HSA does not affect the ability of Lys525 (the most reactive lysine) to be glycated. In contrast, modification of Lys525 reduces the affinity of albumin for FA. The interspecies differences in the molecular structure of albumin that may affect the mechanism of the gSA–RAGE interaction were discussed. The obtained results will help us to learn more about the molecular basis for the involvement of serum albumin in the AGE/RAGE axis and improve the methodology for studying cellular signaling pathways involving RAGE.

Objective. Fatty liver is a rising global health concern, significantly increasing the burden of health care cost. Nonalcoholic fatty liver disease (NAFLD) has a correlation with metabolic syndrome and its complications. Method. We reviewed the literature regarding the mechanisms of developing NAFLD through AGE-RAGE signaling. Results. NAFLD, metabolic syndrome, and production of advanced glycation end-products (AGEs) share many common risk factors and appear to be connected. AGE induces production of the receptor for AGE (RAGE). AGE-RAGE interaction contributes to fat accumulation in the liver leading to inflammation, fibrosis, insulin resistance, and other complications of the fatty liver disease. The immune system, especially macrophages, has an important defense mechanism against RAGE pathway activities. Conclusion. Soluble form of RAGE (sRAGE) has the capability to reduce inflammation by blocking the interaction of AGE with RAGE. However, sRAGE has some limitations, and the best method of usage is probably autotransplantation of transfected stem cells or monocytes, as a precursor of macrophages and Kupffer cells, with a virus that carries sRAGE to alleviate the harmful effects of AGE-RAGE signaling in the settings of fatty liver disease.

BackgroundAnimal studies suggest that advanced glycation end-products (AGEs) and their interaction with receptor for AGEs (RAGE) are involved in sarcopenia, but their relationship in human skeletal muscles has yet to be elucidated.ObjectivesWe aimed to determine whether RAGE expression in human skeletal muscle is associated with serum AGE levels and sarcopenia-related changes.MethodsWe reviewed 33 consecutive women (mean age, 65 years) with distal radius fracture who had consented to donate a sample of forearm flexor muscle for research purposes, which was taken during surgical fracture repair. The muscle RAGE expression was measured with immunohistochemistry staining and serum AGE levels using ELISA method. We compared RAGE expression and AGE levels in patients with and without sarcopenia. We also correlated RAGE expression with such clinical parameters as age, body mass index, bone mineral density (BMD), as well as sarcopenia-related changes, including grip strength, appendicular skeletal muscle mass, and muscle cross-sectional area (CSA) ratios.ResultsTwelve patients (36%) were diagnosed with sarcopenia according to the Asian Working Group for Sarcopenia. They had a significantly higher RAGE expression (p = 0.044) and AGE level (p < 0.001) than those without sarcopenia. The RAGE expression correlated significantly with a high serum AGE level (r = 0.510, p = 0.011) and correlated inversely with a muscle CSA ratio (r = -0.696, p < 0.001).ConclusionThis study shows that RAGE expression increases in sarcopenic patient skeletal muscles. This expression also correlates positively with serum AGE levels and inversely with muscle CSA ratios. Further studies are necessary to determine whether targeting RAGEs can be a therapeutic option for sarcopenia.References[1]Singh R, Barden A, Mori T, Beilin L. In diabetes mellitus (DM) patients, the accumulation of advanced glycation end products (AGE) leads to inflammation and oxidative stress through the activation of specific receptors for AGE (RAGE). Glycated albumin (gHSA) makes a significant contribution to the overall level of AGE in human body and, as a result, to the pathogenesis of DM and concomitant diseases. The mechanism of interaction of gHSA with RAGE is practically not studied. The purpose of the present paper is to study the binding of gHSA to RAGE using molecular modeling methods, to find the main sites of interaction and structural features of glycation sites that determine the efficiency of complex formation with RAGE. Ten gHSA models were constructed using molecular docking and molecular dynamics (MD) methods; each model corresponded to one modified lysine residue (carboxymethyl-lysine): Lys64, Lys73, Lys137, Lys233, Lys262, Lys317, Lys378, Lys525, Lys573, Lys574. Complexes of gHSA with the V-domain of RAGE were constructed using the macromolecular docking method, and their stability was studied using MD simulation. In the constructed gHSA models, the carboxyl groups of glycated Lys317 and Lys525 form intramolecular salt bridges with surrounding amino acids; in other cases, the carboxyl groups of the modified lysines are free to interact with positively charged amino acid residues on the RAGE surface. According to the macromolecular docking data and subsequent MD simulation, the complex of RAGE with gHSA glycated at Lys233 is most effective in terms of strength and specificity. Specific RAGE complexes with gHSA glycated at Lys317 and Lys574 are not formed. The obtained data on the interaction of gHSA with RAGE will help to understand the role of albumin in the pathophysiology of DM and advance towards the prevention and development of effective therapy for this disease.

Based on the methodology of artificial neural networks, models describing the dependence of the level of RAGE inhibitory activity on the affinity of compounds for target proteins of the RAGE-NF-kB signal pathway have been costructed. A validated database of the structures and activity levels of 183 known compounds, which were tested for RAGE inhibitory activity was formed. The analysis of the AGE-RAGE signaling pathways was carried out, 14 key RAGE-NF-kB signal pathway nodes were found, for which 34 relevant target proteins were identified. A database of 66 valid 3D models of 22 target proteins of the RAGE-NF-kB signal chain was compiled. Ensemble molecular docking of 3D models of 183 known RAGE inhibitors into sites of 66 valid 3D models of 22 relevant RAGE target proteins was performed and minimum docking energies for each compound were determined for each target. According to the method of artificial multilayer perceptron neural networks, classification models were constructed to predict level of RAGE inhibitory activity based on the calculated affinity of compounds for significant target proteins of the RAGE-NF-kB signaling chain. The prognostic ability of these models of RAGE-inhibitory activity was evaluated, the maximum accuracy according to ROC-analysis was 90% for a high level of activity. The sensitivity analysis of the developed multitarget models were carried out, the most significant targets of the RAGE-NF-kB signal transmission chain were determined. It was found that for high level of RAGE inhibitory activity, the most significant biotargets are not AGE receptors, but eight signaling kinases of the RAGE-NF-kB pathway and transcription factor NF-kB1. Thus, it is suggested that known compounds with high RAGE-inhibitory activity are preferential inhibitors of signal kinases.

Objective T1-weighted MRI images are commonly used for volumetric assessment of brain structures. Magnetization prepared 2 rapid gradient echo (MP2RAGE) sequence offers superior gray (GM) and white matter (WM) contrast. This study aimed to quantitatively assess the agreement of whole brain tissue and deep GM (DGM) volumes obtained from MP2RAGE compared to the widely used MP-RAGE sequence. Methods Twenty-nine healthy participants were included in this study. All subjects underwent a 3T MRI scan acquiring high-resolution 3D MP-RAGE and MP2RAGE images. Twelve participants were re-scanned after one year. The whole brain, as well as DGM segmentation, was performed using CAT12, volBrain, and FSL-FAST automatic segmentation tools based on the acquired images. Finally, contrast-to-noise ratio between WM and GM (CNRWG), the agreement between the obtained tissue volumes, as well as scan-rescan variability of both sequences were explored. Results Significantly higher CNRWG was detected in MP2RAGE vs. MP-RAGE (Mean ± SD = 0.97 ± 0.04 vs. 0.8 ± 0.1 respectively; p<0.0001). Significantly higher total brain GM, and lower cerebrospinal fluid‏ volumes were obtained from MP2RAGE vs. MP-RAGE based on all segmentation methods (p<0.05 in all cases). Whole-brain voxel-wise comparisons revealed higher GM tissue probability in the thalamus, putamen, caudate, lingual gyrus, and precentral gyrus based on MP2RAGE compared with MP-RAGE. Moreover, significantly higher WM probability was observed in the cerebellum, corpus callosum, and frontal-and-temporal regions in MP2RAGE vs. MP-RAGE. Finally, MP2RAGE showed a higher mean percentage of change in total brain GM compared to MP-RAGE. On the other hand, MP-RAGE demonstrated a higher overtime percentage of change in WM and DGM volumes compared to MP2RAGE. Conclusions Due to its higher CNR, MP2RAGE resulted in reproducible brain tissue segmentation, and thus is a recommended method for volumetric imaging biomarkers for the monitoring of neurological diseases.

RAGE signal transduction via the RAGE-NF-κB signaling pathway is one of the mechanisms of inflammatory reactions that cause severe complications in diabetes mellitus. RAGE inhibitors are promising pharmacological compounds that require the development of new predictive models. Based on the methodology of artificial neural networks, consensus ensemble neural network multitarget model has been constructed. This model describes the dependence of the level of the RAGE inhibitory activity on the affinity of compounds for 34 target proteins of the RAGE-NF-κB signal pathway. For this purpose an expanded database of valid three-dimensional models of target proteins of the RAGE-NF-κB signal chain was created on the basis of a previously created database of three-dimensional models of relevant biotargets. Ensemble molecular docking of known RAGE inhibitors from a verified database into the sites of added models of target proteins was performed, and the minimum docking energies for each compound in relation to each target were determined. An extended training set for neural network modeling was formed. Using seven variants of sampling by the method of artificial multilayer perceptron neural networks, three ensembles of classification decision rules were constructed to predict three level of the RAGE-inhibitory activity based on the calculated affinity of compounds for significant target proteins of the RAGE-NF-κB signaling pathway. Using a simple consensus of the second level, the predictive ability of the created model was assessed and its high accuracy and statistical significance were shown. The resultant consensus ensemble neural network multitarget model has been used for virtual screening of new derivatives of different chemical classes. The most promising substances have been synthesized and sent for experimental studies.

Abstract Aim NIH3T3 cell line with expression of human receptor for advanced glycation end-products (hRAGE) transduced with lentivirus vectors was used to analyze affinity, biological activity, and/or molecular mechanisms of molecules targeting the hRAGE pathway. Method The DNA fragment coding for hRAGE gene was integrated into the genome of NIH3T3 cells using lentivirus transduction. Cells expressing hRAGE were selected with puromycin, and the level of hRAGE expression was analyzed by Western blot. To establish a stable cell line, colonies of hRAGE-expressing cells were generated, and the level of RAGE expression in each engineered cell line was analyzed within 20 generations. Flow cytometry assay was used to verify affinity of anti-hRAGE antibody binding to hRAGE on the surface of engineered cells. The engineered NIH3T3 cell line was applied to assess effects of anti-hRAGE blocking antibody on amyloid β-induced cells apoptosis by CCK-8 assay. Results The engineered NIH3T3 cell line (hRAGE-NIH3T3) could stably express human RAGE. Commercial anti-RAGE polyclonal antibody could recognize and bind to human RAGE on the surface of hRAGE-NIH3T3 but not original NIH3T3 cells. In addition, hRAGE-NIH3T3 was more sensitive to RAGE pathway-dependent stimulation. Our data show that the hRAGE-NIH3T3 cell line established is an excellent tool in the study of RAGE-targeting molecules based on the cellular level, biological function, and RAGE-mediated molecular mechanisms.

Background/Aims: Advanced glycation end products (AGEs) induce epithelial mesenchymal transition (EMT) in renal proximal tubular epithelial cells (PTECs) by promoting the two EMT regulators, transforming growth factor beta (TGF-β) and connective tissue growth factor (CTGF). However, the exact signaling mechanism remains largely unclear. Methods: We investigated the promotion to high mobility group box 1 (HMGB1) in renal tubular epithelial HK-2 cells by AGE-BSA with quantitative PCR and western blot assay, and then determined the regulatory role of HMGB1 in the AGE-BSA-induced CTGF and TGF-β. In addition, the dependence of the receptor of advanced glycation end products (RAGE) was also examined in the CTGF and TGF-β promotion by AGEs and HMGB1 in HK-2 cells using the RNAi method. Results: It was demonstrated that AGEs induced translocation and release of HMGB1 from tubular epithelial HK-2 cells, and the released HMGB1 enhanced the promotion to CTGF and TGF-β by AGEs in HK-2 cells. On the other side, the HMGB1 knockdown by siRNA attenuated the AGE-BSA-induced expression of TGF-β. Moreover, the CTGF and TGF-β promotion in HK-2 cells by AGEs and HMGB1 was RAGE-dependent. Conclusion: Our results indicated that AGEs induced HMGB-1 and promoted the CTGF and TGF-β in renal epithelial HK-2 cells RAGE-dependently. And there was a synergism between AGEs and HMGB1 in the RAGE signaling activation. The in vitro data suggested that the AGE-RAGE and HMGB-1-RAGE signaling might play an important role in the promotion of CTGF and TGF-β in the renal fibrosis process of diabetic nephropathy.

One of the human-caused factors that decreases traffic safety is road rage behaviors. Many of the behaviors thought to be indicative of road rage are clearly intended to be aggressive. The purpose of this study was to examine the frequency of road rage incidents experienced by road users both as victims and perpetrators. Furthermore, the goal was to investigate the impact of various and other factors on road rage behaviors in the past year. The questionnaire was completed by 633 individuals who agreed to participate in the study and reside in Istanbul, Izmir, and Ankara. All of the participants were Turkish, aged 19–76 years (M = 37.84 years, SD = 10.10); 57.3% were females, and 42.7% were males. The survey application was started with ethical approval. Using the snowball sampling method, a survey link or paper pencil forms were distributed to participants. Participants were requested to fill out the questionnaire form including information and various forms of road rage. Data were analyzed using the chi-square test and binary logistic regression analysis. Using the SPSS 28 program, all data were statistically analyzed to a significance level of p < 0.05. According to our findings, low-level road rage is most common among road users. For perpetration, this rate is 43.6%, while for victimization it is 63.2%. Overall, the results of the analysis suggest that low or high-levels of road rage perpetration and victimization may be influenced by driving a vehicle, carrying a weapon and certain demographic factors like gender, age, education level, and marital status. Numerous factors interact to facilitate road rage's emergence. In our study, various forms of road rage are significantly influenced by factors such as age, gender, education, marital status, driving a vehicle, carrying a weapon. We believe it is crucial to examine this issue from multiple perspectives. Additionally, programs for prevention and intervention that effectively reduce the prevalence of road rage incidents are critical.

Diabetes mellitus (DM) is a multifactorial disease associated with hyperglycemia and an increased risk of complications in the cardiovascular system. During hyperglycemia, there is a significant decrease in GLP-1 and GLP- 1R expression, which have been known to have protective effects on the cardiovascular system. Increased RAGE expression due to hyperglycemia can also lead to inflammation, contributing to the pathogenesis of diabetes complications, specifically cardiovascular diseases. Administration of Hibiscus sabdariffa Linn. (HSL) can improve cardiovascular function, partly by increasing GLP-1 expression and suppressing RAGE expression. The aim of this study is to investigate the effect of HSL administration on RAGE, GLP-1, and GLP-1R expression in the heart. This study is an experimental study using 9-to-11-week-old DDY mice divided into four groups: control (K), DM control (KDM), positive control DM with quercetin (KQ), and DM with HSL at a dose of 400 mg/kgBW (DM-HSL). The experimental animals received treatment for four weeks. The analysis of GLP-1R and RAGE expression was conducted using RT-PCR, while GLP-1 expression were obtained using the ELISA method. The results of this study indicate a decrease in RAGE expression and an increase in GLP-1 and GLP-1R expression in the hearts of diabetic mice. Quercetin administration was also found to decrease RAGE expression, increase GLP-1 expression, and raise GLP- 1R expression in the hearts of diabetic mice. These findings suggest that HSL administration has the potential to protect the cardiovascular system in diabetes, which is related to the activity of GLP-1 and GLP-1R and RAGE.

Abstract Originating on New York’s Wall Street, the Occupy movement was “an international network of protests against social and economic inequality that began in [September] 2011 in response to the downturn of 2008” (Thorson et al. 2013, 427). Whilst there has been research on online activity in relation to Occupy, the scope of linguistic analysis to date has been somewhat narrow. Furthermore, the focus on new media has indirectly led to an absence of analysis of institutionally-endorsed traditional media texts. We adopt a mixed-method approach of corpus analysis and discourse analysis of national newspaper articles to answer questions such as ‘Is Occupy associated with a semantic field of violence and aggression?’ and ‘Who is represented as having agency?’ Our results indicate that, in our small corpus of media texts, Occupy and its supporters were predominantly portrayed negatively at the movement’s height; even though protesters are reported to have been peaceful in their majority, the English-speaking media we analysed still aligns them with language suggestive of aggression, conflict and even violence.

Experiments conducted in recent years on animals and research works worldwide show a linkage between calprotectin and occurrence and development of the abdominal aortic aneurysm (AAA). Additionally, a correlation between the level of the receptor for advanced glycation end products (RAGE) and the diameter of the abdominal aorta was found. The purpose of this study was to investigate whether calprotectin and the RAGE plasma level may be a biomarker of human AAA occurrence. We determined two groups of research participants: a group of 32 patients aged 53–88 undergoing primary endovascular aneurysm repair and a control group of 43 volunteers aged 59–82 without the AAA. All the patients from the study group had their blood samples drawn in order to determine the level of calprotectin and RAGE in plasma. The second follow-up examination was carried out after three months. The concentration of calprotectin and RAGE in plasma was determined with the use of the immunoenzymatic method (ELISA). The study showed that patients with the AAA had significantly higher mean calprotectin and RAGE plasma levels (p = 0.0001 and p = 0.0002, respectively) as compared to the control group. After the AAA repair operations, the level of concentration of the calprotectin decreased significantly (p = 0.0002). So far, no studies on the connection between the increase of the calprotectin and RAGE in the patient’s plasma with the AAA have been published. Calprotectin may be a promising biomarker related to the occurrence of AAA. Larger studies are needed to fully elucidate and confirm the role of calprotectin in the development and progression of the aneurysm.

Rage against the Machine is known for their politics as well as their music, which the later helped creating an aggressive-heavy rock-rap genre. Article presents a song by Rage against the Machine band related to ideological movement in America, titled Sleep Now in the Fire. This effort brings an understanding ofideology that is embraced by the band. The method is through literature study. Presentation begins with a short biography of the band, theoretical concepts, and analysis of the song as well its music video. It is concluded that the song represents ideological criticism toward capitalism using Marxism. Both of these lyric and music video represents Marxism as it shares the same movement to fight capitalism—which in this case, Rage against the Machine is using their music, lyric, and video to fight the crime against humanity and cultural imperialism.

Objective. The interaction between advanced glycation end products and their cellular receptor (RAGE) has an important role in the pathogenesis of diabetic microvascular complications. The aim of this study was to investigate the relationship between the 2184A/G polymorphism in the RAGE gene and diabetic nephropathy in Chinese Han patients with type 2 diabetes mellitus. Methods. A total of 868 patients with type 2 diabetes mellitus (486 without and 382 with diabetic nephropathy) were enrolled in this study. The genotype and allele frequencies of the 2184A/G polymorphism were detected using the polymerase chain reaction-restriction fragment-length polymorphism method. Results. The G allele and AG + GG genotype frequencies in patients with diabetic nephropathy were significantly lower than those in patients without diabetic nephropathy (P=0.001 and P=0.005, resp.). After adjustments for possible confounders, multivariate logistic regression analyses showed that the 2184A/G polymorphism was independently associated with diabetic nephropathy (OR = 0.46, 95% CI: 0.22–0.92, P=0.028). Conclusions. Our study indicated that the 2184A/G polymorphism in the RAGE gene was significantly associated with diabetic nephropathy in Chinese Han patients with type 2 diabetes.

Diabetes mellitus (DM) often accompanies liver dysfunction. Astragali Radix is a traditional Chinese herbal medicine that is widely administrated to ameliorate the symptoms of diabetes as well as liver dysfunction, but its acting mechanism is still not yet fully recognized. Advanced glycation end products (AGEs) play a key role in promoting diabetic organ dysfunction. Both hyperglycemia and AGEs can induce insulin resistance, hepatocyte damage and liver dysfunction. We designed this study to explore the effects of the phytoestrogen Calycosin, a major active component of Astragali Radix, on AGEs-induced glucose uptake dysfunction in the hepatocyte cell line and relevant mechanisms. MTT and BrdU methods were applied to evaluate cell viability. 2-NBDG was used to observe glucose uptake by a live cell imaging system. Immunofluorescence method was carried out to investigate GLUT1, GLUT4, and RAGE protein expressions on cell membrane. cAMP content was determined by an EIA method. We found Calycosin concentration-dependently ameliorated AGEs-induced hepatocyte viability damage. AGEs dramatically reduced basal glucose uptake in hepatocytes, and this reduction could be reversed by Calycosin administration. By immunofluorescence detection, we observed that Calycosin could inhibit AGEs-induced GLUT1 expression down-regulation via estrogen receptor (ER). Furthermore, Calycosin decreased AGEs-promoted RAGE and cAMP elevation in hepatocytes. These findings strongly suggest that Calycosin can ameliorate AGEs-promoted glucose uptake dysfunction in hepatocytes; the protection of cell viability and ER-RAGE and GLUT1 pathways play a significant role in this modulation.

Introduction. In the study of Alzheimer’s disease (AD), the cause-and-effect relationship between neurodegenerative changes and the accompanying amyloid angiopathy is becoming increasingly important. The accumulated clinical data indicates that an important contribution to the pathogenesis of AD is made by neurovascular unit dysfunction, including disruption in permeability of the blood-brain barrier (BBB), microcirculation, and metabolic coupling of cells.Aim. To study the molecular mechanisms of disturbed brain microcirculation and the structural and functional integrity of the BBB in experimental models of AD in vitro under the modulation of CD147 and RAGE.Materials and methods. The study was carried out on C57BL/6 mice. First, we formed an AD model in animals of the experimental group. Then, we isolated and cultured primary cells of the brain, modulated the activity of CD147 and RAGE in endothelial cells using siRNA CD147, siRNA RAGE, cyclophilin A and Aβ1-42, and formed a BBB model in vitro. Further, we assessed transendothelial electrical resistance in the BBB model in vitro, registered the marker molecules of angiogenesis and analyzed the expression of APP in endothelial cells. Statistical processing of the obtained data was carried out using the methods of nonparametric statistics: the Mann – Whitney U test for comparing independent samples and the Wilcoxon test for comparing dependent samples. The level of statistical significance of differences was p ≤ 0.05.Results and discussion. Knockdown of RAGE led to a statistically significant increase in TEER, an intensification of neoangiogenesis, and a decrease in the level of APP expression. At the same time, although CD147 knockdown led to an increase in TEER, it also led to controversial effects on angiogenesis and an increase in APP expression.Conclusion. Analyzing the data obtained, it can be concluded that RAGE and CD147 silencing in the cells of cerebral microvessels can become a promising method for reducing their pathological permeability.

The pathogenesis of IgAV, the most common systemic vasculitis in childhood, appears to be complex and requires further elucidation. We aimed to investigate the potential role of galactose-deficient immunoglobulin A1 (Gd-IgA1), high-mobility group box 1 (HMGB1), receptor for advanced glycation end products (RAGE) and protocadherin 1 (PCDH1) in the pathogenesis of IgAV. Our prospective study enrolled 86 patients with IgAV and 70 controls. HMGB1, RAGE, Gd-IgA1 and PCDH1 in serum and urine were determined by the enzyme-linked immunosorbent assay (ELISA) method at the onset of the disease and after a six-month interval in patients and once in the control group. Serum concentrations of HMGB1, RAGE and PCDH1 and urinary concentrations of HMGB1, RAGE, Gd-IgA1 and PCDH1 were significantly higher in patients with IgAV than in the control group (p < 0.001). Concentrations of HMGB1 (5573 pg/mL vs. 3477 pg/mL vs. 1088 pg/mL, p < 0.001) and RAGE (309 pg/mL vs. 302.4 pg/mL vs. 201.3 pg/mL, p = 0.012) in the serum of patients remained significantly elevated when the disease onset was compared with the six-month follow-up interval, and thus could be a potential marker of disease activity. Urinary concentration of HMGB1 measured in the follow-up period was higher in patients with nephritis compared to IgAV without nephritis (270.9 (146.7–542.7) ng/mmol vs. 133.2 (85.9–318.6) ng/mmol, p = 0.049) and significantly positively correlated with the urine albumine to creatinine ratio (τ = 0.184, p < 0.05), the number of erythrocytes in urine samples (τ = 0.193, p < 0.05) and with the outcome of nephritis (τ = 0.287, p < 0.05); therefore, HMGB1 could be a potential tool for monitoring patients with IgAV who develop nephritis. Taken together, our results imply a possible interplay of Gd-IgA1, HMGB1, RAGE and PCDH1 in the development of IgAV. The identification of sensitive biomarkers in IgAV may provide disease prevention and future therapeutics.

Background: Aggressive behavior among geriatric psychiatry inpatients spontaneously declines during serial measurements over time with rating scales. Method: Evidence for decline in aggressive behavior, over a 6-week period, on a continuing-care geriatric psychiatry ward was examined with the Staff Observation Aggression Scale (SOAS) and the Rating Scale for Aggressive Behavior in the Elderly (RAGE). Results: The RAGE and all its subitems did not spontaneously decline. The total number of aggressive incidents on the SOAS, the SOAS total score, and the subitem scores, except for the SOAS results subitem, spontaneously declined after Week 4 and became established by Week 5. Conclusion: A “run in” period of at least 4 weeks may be needed in intervention studies using the SOAS to reduce contamination by spontaneous decline.

Background: This study aims to investigate potential markers of psoriasis and aging, and to elucidate possible connections between these two processes. Methods: The serum samples of 60 psoriatic patients and 100 controls were analysed, and the levels of four selected parameters (AGEs, RAGE, NAD, and elastin) were determined using commercial ELISA kits. Serum C-reactive protein was assayed using an immune-nephelometry method. Findings: Among the patients, the levels of CRP, AGEs, and RAGE were all increased, while the levels of NAD were reduced when compared to the control group. A negative correlation between the levels of AGEs and NAD was found. A negative correlation between age and the NAD levels among the control group was observed, however among the patients the relationship was diminished. While there was no difference in the levels of native elastin between the patients and the controls, a positive correlation between the levels of native elastin and age and a negative correlation between the levels of native elastin and the severity of psoriasis were found. Conclusions: The results of our study support the notion of psoriasis and possibly other immune-mediated diseases accelerating the aging process through sustained systemic damage. The serum levels of CRP, NAD, AGEs, and RAGE appear to be promising potential biomarkers of psoriasis. The decrease in the serum levels of NAD is associated with (pro)inflammatory states. Our analysis indicates that the levels of native elastin might strongly reflect both the severity of psoriasis and the aging process.

Background:Chronic kidney disease (CKD) is a condition associated with progressive loss of kidney function and kidney damage. The two common causes of CKD are diabetes mellitus and hypertension. Other causes of CKD also include polycystic kidney disease, obstructive uropathy and primary glomerulonephritis. The receptor for advanced glycation end-products (RAGE) is a multi-ligand cell surface receptor of the immunoglobulin superfamily and it has been associated with kidney disease in both non-diabetic and diabetic patients. Presently, data on the association between RAGE polymorphisms and CKD in the Malaysian population is limited, while numerous studies have reported associations of RAGE polymorphisms with diabetic complications in other populations. The present study aims to explore the possibility of using RAGE polymorphisms as candidate markers of CKD in Malaysian population by using association analysis. Methods:A total of 102 non-diabetic CKD patients, 204 diabetic CKD patients and 345 healthy controls were enrolled in the study. DNA isolated from blood samples were subjected to genotyping of RAGE G82S, −374T/A, −429T/C, 1704G/T and 2184A/G polymorphisms using real-time polymerase chain reaction (PCR). The 63-bp deletion, a polymorphism in the RAGE gene promoter, was genotyped using conventional PCR method and visualized using agarose gel electrophoresis. The collective frequencies of genotypes with at least one copy of the minor alleles of the four polymorphisms were compared between the non-diabetic CKD patients, diabetic CKD patients and healthy controls. Results:After adjustment of age, gender and ethnic groups in binary logistic regression analysis, the G82S CT + TT genotypes were associated with non-diabetic CKD patients when compared with diabetic CKD patients (p= 0.015, OR = 1.896, 95% CI = 1.132–3.176). After further adjustment of CKD comorbidities, the G82S CT + TT genotypes were still associated with non-diabetic CKD patients when compared with diabetic CKD patients (p= 0.011, OR = 2.024, 95% CI = 1.178–3.476). However, it cannot be suggested that G82S polymorphism was associated with CKD in non-diabetic patients in this study. This is because there were no significant differences in the frequencies of G82S CT + TT genotypes between non-diabetic CKD patients and healthy controls. In addition, the RAGE −374T/A, −429T/C, 1704G/T, 2184A/G and 63-bp deletion polymorphisms were also not associated with non-diabetic CKD patients and diabetic CKD patients in this study. Conclusion:The G82S, −374T/A, −429T/C, 1704G/T, 2184A/G and 63-bp deletion polymorphisms examined in this study were not associated with chronic kidney disease in the Malaysian patients.

C-reactive protein (CRP) is a commonly used serum biomarker for detecting sepsis in neonates. After the onset of sepsis, serial measurements are necessary to monitor disease progression; therefore, a non-invasive detection method is beneficial for neonatal well-being. While some studies have shown a correlation between serum and salivary CRP levels in septic neonates, the causal link behind this correlation remains unclear. To investigate this relationship, CRP was examined in serum and saliva samples from 18 septic neonates and compared with saliva samples from 22 healthy neonates. While the measured blood and saliva concentrations of the septic neonates varied individually, a correlation of CRP levels between serum and saliva samples was observed over time. To clarify the presence of active transport of CRP across the blood–salivary barrier (BSB), transport studies were performed with CRP using in vitro models of oral mucosa and submandibular salivary gland epithelium. The results showed enhanced transport toward saliva in both models, supporting the clinical relevance for salivary CRP as a biomarker. Furthermore, CRP regulated the expression of the receptor for advanced glycation end products (RAGE) and the addition of soluble RAGE during the transport studies indicated a RAGE-dependent transport process for CRP from blood to saliva.

This paper reported the fabrication of cellular Al foams without thickening agent addition via melt foaming method with the porosity rage of 62.4%-83.3% and the pore size of approximate 2.0mm. CaCO3 was selected as the blowing agent. 1.0wt.%-3.0wt.% Mg was added in the matrix. The gas releasing reaction mechanism was revealed by TG-DTA (Thermo Gravimetric Analysis-Differential Thermal Analysis) experiments. The influence of the adding methods of Mg and CaCO3 on the foaming process was studied and the results showed that, compared to adding powder mixture, the method of adding Mg bulk into the melt before the addition of CaCO3 produces Al foams with better pore structure. Finally, the compressive behaviors of Al foams were investigated.

Abstract Introduction: In forensic practice, it is well known that the mechanism and dating of traumatic injuries is one of the primary responsibilities of this specialty. Currently, it is a subject still debated by researchers, and so far, an infallible marker that would objectively support their intravitam/postmortem occurrence has not yet been identified. However, studies have shown that the HMGB1-RAGE axis is rapidly activated after trauma and might be an essential element to help solve the forensic problem of wound dating. Purpose: To compare the values of HMGB1-RAGE expression occurring in wounds produced intravitally shortly before death and in wounds produced postmortem and to quantify the differences arising between them. Material and method: For this prospective study, skin fragments were collected from the site of wounds in autopsied cadavers at the County Clinical Service of Forensic Medicine Constanta (SCJML Constanta), wounds produced intravitally and with a maximum survival time of 60 minutes. Postmortem wounds and control fragments from volunteers undergoing surgery for skin tumours were also collected. The main conditions were: chronological documentation of the lesion and absence of neoplastic or inflammatory conditions. Ninety-six autopsy cases between 2021–2022 met the criteria for inclusion in the study. A control fragment accompanied each fragment from the wound. Routine Hematoxylin-Eosin (HE), Perls and Van Gieson Werhoeffstaining, as well as immunohistochemistry with HMGB1 and RAGE markers were performed on each fragment and a score based on staining intensity was determined. Results: Routine staining was not useful in assessing vitality in segments with survival time up to 30 min. Immunohistochemically, both markers showed increased values compared to control values (p<0.0001) and to lesions produced postmortem. An interesting aspect is the lack of reactivity in the lesion’s margins for both markers. Conclusions: Although further research is needed, the results of our study support the hypothesis that the HMGB1-RAGE axis is useful in assessing the vital reaction in skin wounds.

Background: The main cause of complications in diabetes mellitus is a condition of oxidative stress due to increased reactive oxygen species (ROS). The increase in ROS can be triggered by the increase in advanced glycation end-products (AGE), which binds to its receptor (RAGE). The result of increased ROS will lead to lipid peroxidation in cell membranes and induce particular increase in malondialdehyde (MDA). Objectives: The objective of this study was to evaluate the benefits of physical exercise in preventing complications of the kidneys caused by hyperglycemia. Method: This study was a pure experimental research with post-test only group design. Subjects were 10 male Sprague Dawley rats induced with 35 mg/kgdose of streptozotocin (STZ). The subjects were divided into two groups, i.e. the STZ group and STZ group with physical exercise. Physical exercise was given in 9 weeks using a treadmill, in 5 times/week frequency, moderate intensity, 0 degree of tilt, gradually increased speed and duration. Result: Regular and measurable physical exercise can decrease the blood glucose level(0,048) and reduce the RAGE expression in glomerulus(0,003) but increase MDA level in kidney (0,767). Conclusion: Regular and measurable physical exercise can decrease the blood glucose level and reduce the RAGE expression in glomerulus. Bangladesh Journal of Medical Science Vol.18(3) 2019 p.484-490

Neutrophil-mediated innate host defense mechanisms include pathogen elimination through bacterial phagocytosis, which activates the 5-lipoxygenase (5-LOX) product synthesis. Here, we studied the effect of synthetic oligodeoxyribonucleotides (ODNs), which mimic the receptor-recognized sites of bacterial (CpG-ODNs) and genomic (G-rich ODNs) DNAs released from the inflammatory area, on the neutrophil functions after cell stimulation with Salmonella typhimurium. A possible mechanism for ODN recognition by Toll-like receptor 9 (TLR9) and RAGE receptor has been proposed. We found for the first time that the combination of the magic peptide LRR11 from the leucine-rich repeat (LRR) of TLR9 with the CpG-ODNs modulates the uptake and signaling from ODNs, in particular, dramatically stimulates 5-LOX pathway. Using thickness shear mode acoustic method, we confirmed the specific binding of CpG-ODNs, but not G-rich ODN, to LRR11. The RAGE receptor has been shown to play an important role in promoting ODN uptake. Thus, FPS-ZM1, a high-affinity RAGE inhibitor, suppresses the synthesis of 5-LOX products and reduces the uptake of ODNs by neutrophils; the inhibitor effect being abolished by the addition of LRR11. The results obtained revealed that the studied peptide-ODN complexes possess high biological activity and can be promising for the development of effective vaccine adjuvants and antimicrobial therapeutics.

Background and Aims. Radix Rehmanniae and Corni Fructus (RC) have been widely applied to treat diabetic nephropathy (DN) for centuries. But the mechanism of how RC plays the therapeutic role against DN is unclear as yet. Methods. The information about RC was obtained from a public database. The active compounds of RC were screened by oral bioavailability (OB) and drug-likeness (DL). Gene ontology (GO) analysis was performed to realize the key targets of RC, and an active compound-potential target network was created. The therapeutic effects of RC active compounds and their key signal pathways were preliminarily probed via network pharmacology analysis and animal experiments. Results. In this study, 29 active compounds from RC and 64 key targets related to DN were collected using the network pharmacology method. The pathway enrichment analysis showed that RC regulated advanced glycosylation end product (AGE-) RAGE and IL-17 signaling pathways to treat DN. The animal experiments revealed that RC significantly improved metabolic parameters, inflammation renal structure, and function to protect the kidney against DN. Conclusions. The results revealed the relationship between multicomponents and multitargets of RC. The administratiom of RC might remit the DM-induced renal damage through the AGE-RAGE signaling pathway to improve metabolic parameters and protect renal structure and function.

We study the singular limit of the compressible Navier–Stokes system in the whole space ℝ3, where the Mach number and Froude number are proportional to a small parameter ε → 0. The central issue is the local decay of the acoustic energy proved by means of the RAGE theorem. The result is quite general and the proposed approach can be applied to a large variety of problems that concern propagation of acoustic waves in compressible fluids. In particular, the method can be used for showing stability of various numerical schemes based on the so-called hybrid methods.

Modern agriculture is developing towards the direction of intelligent. We can realize the nondestructive measurement of plant leaf area by using the digital camera. In this paper, we make the same ratio between the camera’s screen and the background plate to overcome the problem of geometric distortion. Then we use two perpendicular digital cameras from the front and side to collect images respectively for curved leaf. Because there are characteristics of the image grey value mutation on the rage of the vane, we can extract the leaf by image segmentation. The leaf area can be calculated by the statistic of the pixels number according to the projection principle. Experiments show that, the error of leaf area measurement reduces from 13.51% to 5.93% by binocular vision. So this method not only can get the measurement of leaf area data, but also can effectively avoid the two-dimensional image distortion and improve the accuracy of leaf area calculation.

This study was aimed to identify and analyze the meaning and message in several sentences found in the novel Rage of Angels by Sidney Sheldon published in 1980. The selected sentences in the novel, which are the data of this study, were collected by applying the documentation method and note-taking technique which then were analyzed using the descriptive qualitative method. The data were presented informally in descriptive form. This study analyzes the types of meaning based on the theory proposed by Leech (1974) and the messages of the novel based on the biographical approach theory by Wellek and Warren (1962). As a result, all the seven types of meaning, namely connotative meaning, conceptual meaning, affective meaning, stylistic meaning, reflected meaning, collocative meaning, and thematic meaning, were found in the novel. In relation to the message of the novel, based on the biographical approach, there were five messages that the author delivered.

High competition requires new methods, tools, and technologies for implementing marketing efforts. Mind maps represent marketing information in a compact way, which facilitates classification and visualization of the marketing phenomenon under analysis. The method of text analysis can be applied to marketing research because mind maps that represent marketing information are a structured text; such an approach makes it possible to subject mind maps to linguistic processing. This research combined the conventional semantic analysis with comparing mind maps as a method of semantic-graph analysis. Initial marketing data were converted into a structured graph of lexical units, which were them compared pairwise to calculate similarity, taking into account the structural-multiple approach. The methods of semantic text analysis proved applicable to marketing data and effective in extracting additional information from a set of heterogeneous sources or different experts. Text analysis tools demonstrated a good potential for broadening the rage of marketing research methods.

Purpose The purpose of this study is to distinguish those emotions which customers express verbally during a failed remote service encounter from those which they do not. The study further attempts to investigate the post-consumption customer behaviour of verbally expressed and unexpressed negative customer emotions. Design/methodology/approach The authors used a survey-based research design. The hypotheses were tested through the “partial least squared structural equation modelling” method. Findings This study shows that in a failed remote service encounter, customers verbally express retaliatory rage emotions, such as anger and rage. At the same time, they are able to suppress rancorous rage emotions, such as disgust and contempt and do not express them verbally. The authors demonstrate that after emotions are verbally expressed during a failed remote service encounter, they are followed by the post-consumption behaviours of negative word of mouth and revenge; when emotions are not expressed verbally during a failed service encounter, they are followed up by exit behaviour. Research limitations/implications The effects of variables, such as switching costs and individual and situational factors, can be investigated in the model. Future studies can also explore the role of organizational interventions, such as explanation and apology, on negative customer emotions during failed remote service encounters. Their moderating impact on customer behaviour during and after the encounters can be investigated. Practical implications This study has much practical relevance in the post-COVID-19 world, where remote service delivery is becoming the new normal in many sectors. In remote service delivery situations, verbally unexpressed negative emotions can remain undetected; however, they have negative consequences for firms. This study underscores the need to train frontline employees to notice these unexpressed emotions so that service recoveries can be initiated. Originality/value This paper contributes to the area of dysfunctional customer behaviour and service recovery. The existing literature has not explored whether some negative emotions are expressed during a failed service encounter and then acted upon later, and some emotions are not expressed but acted upon later. This study addresses the problem of firms getting caught unawares when they find customers resorting to undesirable post-consumption behaviour without demonstrating any verbal expressions during the preceding failed service encounters.

Objective. Hyperuricemia (HUA) is a common metabolic disease caused by disordered purine metabolism. We aim to reveal the mechanisms underlying the anti-HUA function of Simiao pill and provide therapeutic targets. Methods. Simiao pill-related targets were obtained using Herbal Ingredients’ Targets (HIT), Traditional Chinese Medicine Systems Pharmacology (TCMSP), and Traditional Chinese Medicine Integrated Database (TCMID). HUA-associated targets were retrieved from GeneCards, DisGeNET, and Therapeutic Targets Database (TTD). Protein-protein interaction (PPI) network was constructed using the Search Tool for the Retrieval of Interacting Genes/Proteins (STRING) database, ggraph and igraph R packages. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were performed using ClusterProfiler. The top 10 core targets were identified through cytoHubba. Molecular docking was conducted using PyMOL and AutoDock high-performance liquid chromatograph (HPLC) analysis was performed to identify effective compounds of Simiao pill. Results. Simiao pill-HUA target network contained 80 targets. The key targets were mainly involved in inflammatory responses. Insulin (INS), tumor necrosis factor (TNF), interleukin-6 (IL6), interleukin 1 beta (IL1B), vascular endothelial growth factor A (VEGFA), leptin (LEP), signal transducer and activator of transcription 3 (STAT3), C-C motif chemokine ligand 2 (CCL2), interleukin-10 (IL10), and toll like receptor 4 (TLR4) were the top 10 targets in the PPI network. GO analysis demonstrated the main implication of the targets in molecular responses, production, and metabolism. KEGG analysis revealed that Simiao pill might mitigate HUA through advanced glycation end-product- (AGE-) receptor for AGE- (RAGE-) and hypoxia-inducible factor-1- (HIF-1-) associated pathways. IL1B, IL6, IL10, TLR4, and TNF were finally determined as the promising targets of Simiao pill treating HUA. Through molecular docking and HPLC analysis, luteolin, quercetin, rutaecarpine, baicalin, and atractylenolide I were the main active compounds. Conclusions. Simiao pill can mitigate HUA by restraining inflammation, mediating AGE-RAGE- and HIF-1-related pathways, and targeting IL1B, IL6, IL10, TLR4, and TNF.

Introduction This study aimed to investigate amniotic fluid (AF) proteins that were differentially expressed between patients with cervical insufficiency (CI) and asymptomatic short cervix (SCX, ≤ 25 mm), and whether these proteins could be predictive of spontaneous preterm birth (SPTB) in these patients. Method This was a retrospective cohort study of 129 singleton pregnant women with CI (n = 80) or SCX (n = 49) at 17 to 26 weeks who underwent amniocentesis. An antibody microarray was used to perform comparative proteomic profiling of AF from matched CI (n = 20) and SCX (n = 20) pregnancies. In the total cohort, an ELISA validation study was performed for 15 candidate proteins of interest. Subgroup analyses of patients with CI and SCX were conducted to evaluate the association between the 15 proteins and SPTB at < 32 weeks of gestation. Results Eighty-six proteins showed intergroup differences. ELISA validation confirmed significantly higher levels of AF EN-RAGE, IL-8, lipocalin-2, MMP-9, S100A8/A9, thrombospondin-2, and TNFR2 in patients with CI than in those with SCX. Multivariable analysis showed that increased AF levels of EN-RAGE, S100A8/A9, and uPA were independently associated with SPTB at < 32 weeks in patients with CI; whereas in patients with SCX, high AF levels of APRIL, EN-RAGE, LBP, and TNFR2 were independently associated with SPTB at < 32 weeks. Conclusions Multiple AF proteins show altered expression in patients with CI compared with SCX controls. Moreover, several novel mediators involved in inflammation were identified as potential biomarkers for predicting SPTB after the diagnosis of CI and SCX. These results provide new insights into target-specific molecules for targeted therapies to prevent SPTB in patients with CI/SCX.
