Photovoltaic (PV) system energy production is non-linear because it is influenced by the random nature of weather conditions. The use of machine learning techniques to model the PV system energy production is recommended since there is no known way to deal well with non-linear data. In order to detect PV system faults, the machine learning models should provide accurate outputs. The aim of this work is to accurately predict the DC energy of six PV strings of a utility-scale PV system and to accurately detect PV string faults by benchmarking the results of four machine learning methodologies known to improve the accuracy of the machine learning models, such as the data mining methodology, machine learning technique benchmarking methodology, hybrid methodology, and the ensemble methodology. A new hybrid methodology is proposed in this work which combines the use of a fuzzy system and the use of a machine learning system containing five different trained machine learning models, such as the regression tree, artificial neural networks, multi-gene genetic programming, Gaussian process, and support vector machines for regression. The results showed that the hybrid methodology provided the most accurate machine learning predictions of the PV string DC energy, and consequently the PV string fault detection is successful.

Power-Voltage (P-V) curve and Current-Voltage (I-V) curve determine the performance of the PV system. In this work, the arrangements of the PV module were reconstructed by adding the number of PV module in 3 strings configuration from 5 to 45. This method enhance the performance of the PV system as it able to show the characteristic of the P-V and I-V curve during partial shading and maximum irradiance despite higher number of PV panel. This study focuses on improving the PV array configuration and simulation speed of the PV panel. The simulation of small size PV array is possible, but the problem lies when the number of string and PV module used increases. New PV array configuration is flexible and easy to add string and increase the number of PV module. PV array configuration was modeled using MATLAB/SIMULINK software.

In PV (Photovoltaic) systems, the PV array is a structure in which many PV strings are connected in parallel. The voltage mismatch between PV strings, in which PV modules are connected in a series, occurs due to a voltage decrease in some modules. In this paper, research on the electrical characteristics of PV arrays due to a voltage mismatch was conducted. Considering the voltage mismatch, experiments on partial shading, the non-uniformity of irradiance, and the failure of bypass diodes were conducted on the PV module level. It was confirmed that the open-circuit voltage greatly decreased due to the failure of bypass diodes, which is among the causes of voltage mismatch. From the simulation results at the PV array level, it can be seen that a reverse current flowed into the low-potential string, which includes PV modules, causing the failure of the bypass diodes. Measuring the reverse current at one low-potential string, it was found that, in four parallel circuits, the reverse current was 12 A. For this reason, in large PV plants, an overcurrent can flow into the fuse due to the potential difference between strings, causing an output decrease of PV plants and the burnout of fuses.

The potential-induced degradation (PID) of photovoltaic (PV) modules is one of the most extreme types of degradation in PV modules, where PID-affected modules can result in an almost 25% power reduction. Understanding how module defects impact PID is key to reducing the issue. Therefore, this work investigates the impact of an anti-PID inverter on PV modules throughout three years of field operating conditions. We used electroluminescence (EL), ultraviolet fluorescence (UVF), and thermography imaging to explore the varieties of an anti-PID inverter connected to a PV string. It was discovered that a PV string with an anti-PID inverter could improve the output power of the modules by 5.8%. In addition, the performance ratio (PR) was equal to 91.2% and 87.8%, respectively, for PV strings with and without an anti-PID inverter.

<span lang="EN-US">As one of the most prominent renewable energy resources, Photovoltaic (PV) generation has been growing dramatically over the last years and its applications are classified into residential, commercial, and power plant class, based on its power capacity. Especially, a typical residential PV system has been configured with serially connected PV modules. For serially connected PV modules, shading is a critical factor to reduce the whole string PV output power. This paper presents study on performance analysis of a residential PV string under partial shading. This analysis contains how many maximum power points (MPPs) occur and how much power would be lost while a PV string is under partial shading with different shading positions and different shading intensities. PSIM simulation tool is used to verify the performance analysis. As a result, the number of MPPs is directly related with the number of shading intensities, regardless of the shading position. In this paper, it was verified that 8 maximum power points were generated for 8 solar intensities regardless of the location of the shadows under extreme conditions. These results can be utilized for fault diagnosis in the PV string.</span>

In this paper, photovoltaic (PV) string failure analysis and health monitoring of PV modules based on a low-cost self-powered wireless sensor network (WSN) are presented. Simple and effective fault detection and diagnosis method based on the real-time operating voltage of PV modules is proposed. The proposed method is verified using the developed health monitoring system which is installed in a grid-connected PV system. Each of the PV modules is monitored via WSN to detect any individual faulty module. The analysis of PV string failure includes several electrical fault scenarios and their impact on the PV string characteristics. The results show that a degraded or faulty module exhibits low operating voltage as compared to the normal module. The developed health monitoring system also includes a graphical user interface (GUI) program which graphically displays the real-time operating voltage of each module with colors and thus helping users to identify the faulty modules easily. The faulty modules identification approach is further validated using the PV module electroluminescence (EL) imaging system.

Shingled photovoltaic (PV) modules with increased output have attracted growing interest compared to conventional PV modules. However, the area per unit solar cell of shingled PV modules is smaller because these modules are manufactured by dividing and bonding solar cells, which means that shingled PV modules can easily have inferior shading characteristics. Therefore, analysis of the extent to which the shadow affects the output loss is essential, and the circuit needs to be designed accordingly. In this study, the loss resulting from the shading of the shingled string used to manufacture the shingled module was analyzed using simulation. A divided cell was modeled using a double-diode model, and a shingled string was formed by connecting the cell in series. The shading pattern was simulated according to the shading ratio of the vertical and horizontal patterns, and in the case of the shingled string, greater losses occurred in the vertical direction than the horizontal direction. In addition, it was modularized and compared with a conventional PV module and a shingled PV module. The results confirmed that the shingled PV module delivered higher shading output than the conventional PV module in less shade, and the result of the shading characteristic simulation of the shingled PV module was confirmed to be accurate within an error of 1%.

Reduction of long-term degradation effects represents a long-time challenge in photovoltaic (PV) manufacturing industry. Modelling of long-term degradation types and their impact on maximum power of PV systems have been analysed in this article. Brief guidelines for PV cell-based modelling of PV systems have been illustrated. Special study case, PV string consisting of 12 PV modules, has been modelled in order to determine degradation and mismatch power losses. Modified methodology for prediction of annual energy production from PV string, based on horizontal irradiation and ambient temperature experimental measurements at the location of Belgrade, has been developed. Coefficient named ?degradation factor? has been introduced to include and validate degradation power losses. Economic considerations have indicated evident money income reduction, as a consequence of lower annual energy production related to long-term degradation.

The outdoor performance of n-type bifacial Si photovoltaic (PV) modules and string systems was evaluated for two different albedo (ground reflection) conditions, i.e., 21% and 79%. Both monofacial and bifacial silicon PV modules were prepared using n-type bifacial Si passivated emitter rear totally diffused cells with multi-wire busbar incorporated with a white and transparent back-sheet, respectively. In the first set of tests, the power production of the bifacial PV string system was compared with the monofacial PV string system installed on a grey concrete floor with an albedo of ~21% for approximately one year (June 2016–May 2017). In the second test, the gain of the bifacial PV string system installed on the white membrane floor with an albedo of ~79% was evaluated for approximately ten months (November 2016–August 2017). During the second test, the power production by an equivalent monofacial module installed on a horizontal solar tracker was also monitored. The gain was estimated by comparing the energy yield of the bifacial PV module with that of the monofacial module. For the 1.5 kW PV string systems with a 30° tilt angle to the south and 21% ground albedo, the year-wide average bifacial gain was determined to be 10.5%. An increase of the ground albedo to 79% improved the bifacial gain to 33.3%. During the same period, the horizontal single-axis tracker yielded an energy gain of 15.8%.

Photovoltaic (PV) systems have recieved a lot of attention in recent decades due to their accessibility and advancements in PV technology. The protection of PV systems from faults such as String to String (SS), String to Ground (SG), Open circuit (OC), and partial shading are the key challenges to the realization of cost-effective and environmentally friendly PV systems. Such unusual circumstances reduce the maximum available PV power. Partial shading and breakdowns in a PV array must therefore be noticed quickly for enhanced system efficiency and reliability. The significant fault current in PV systems can be detected using the existing safety devices in PV systems, such as fuses and residual current detectors. The flowing fault current being of low order is not significant enough for current protection devices to detect if the solar and/or fault mismatch is modest and the fault resistance is high. As a result, under cloudy and low irradiance conditions, the traditional protection devices fail to identify problems, resulting in reliability concerns and photovoltaic fire threats. In this context, a fault diagnosis scheme for PV systems is presented in this paper, which includes feature extraction using the Discrete wavelet transform, and classification of various defects on the PV system using Decision tree.

Xanthomonas oryzae pv. oryae (Xoo) est l'agent causal de la bactériose vasculaire du riz (BLB), maladie entraînant des pertes de rendement importantes dans la plupart des régions rizicoles, notamment en Afrique. La virulence des souches Asiatiques de Xoo dépend des effecteurs de la famille AvrBs3/PthA ou TAL (pour Transcription Activator-Like). Des études appro fondies sur le mode d'action des effecteurs TAL ont montré que l'activité de virulence ou d'avirulence que les TAL confèrent à la bactérie dépend essentiellement de leur interaction avec les gènes de sensibilité et/ou de résistance correspondants chez le riz. Les souches de Xoo originaires d'Afrique isolées récemment ne présentent que huit effecteurs de type TAL dans leur génome et leur rôle dans la virulence est jusqu'à présent inconnu. Les travaux de cette thèse ont porté sur la caractérisation des effecteurs de type TAL dans la souche africaine de Xoo BAI3. Une mutagenèse systématique par recombinaison homologue sur l'ensemble des huit gènes tal de la souche BAI3 a été effectuée et a conduit à l'identification et à la caractérisation du gène talC. TalC se caractérise par 21.5 répétitions et est phylogénétiquement proche des TAL de Xanthomonas oryzae pv. oryzicola (Xoc). Le mutant BAI3ΔtalC est incapable de développer les symptômes de la maladie sur plante sensible. Toutefois, les bactéries se multiplient très bien au niveau de l'apex foliaire proche du site d'inoculation, laissant supposer que TalC est requis pour la colonisation du système vasculaire. Parmi les cibles directes potentielles identifiées via l'analyse du transcriptome de feuilles de riz sensible (BAI3 versus BAI3ΔtalC), Os11N3 code une protéine de la famille des noduline-3 MtN3 et est fortement induit durant l'infection par la souche sauvage BAI3. Nous avons identifié dans la région promotrice de Os11N3 une séquence nucléotidique ciblée par TalC et démontré qu'elle était fonctionnelle via des expériences d'expression en trans dans N. benthamiana. Les travaux de cette thèse montrent pour la première fois que les effecteurs de type TAL contribuent de manière importante à la virulence de la souche africaine Xoo BAI3. Ces travaux contribueront à l'amélioration génétique des lignées de riz pour la résistance à la bactériose vasculaire du riz en Afrique
Xanthomonas oryzae pv. oryzae (Xoo) is the causal agent of Bacterial Leaf Blight (BLB) on rice, a serious disease causing important yield losses in the main rice growing regions including Africa. The virulence of Asian Xoo strains mainly depends on the type III effectors of avrBs3/pthA gene family, namely TAL (for Transcription Activator Like) effectors. In depth studies on the function of TAL effectors revealed that the virulence and/or the avirulence activities conferred by these effectors requires the binding and the induction of the corresponding S and/or R genes. African Xoo strains was shown to harbor 8 TAL effectors in their genomes. However, the contribution of these TAL effectors to Xoo virulence is still unknown. This work reports on the identification and characterization of TAL effectors in the African Xoo strain BAI3R. A random mutagenesis based on homologous recombination in the genes encoding TAL effector was conducted in Xoo str ain BAI3R and led to the identification of talC. TalC harbors 21.5 repeats in its central domain and is phylogenetically more related to TAL effectors of Xanthomonas oryzae pv. oryzicola (Xoc). The BAI3RΔtalC mutant is seriously impaired in its virulence on susceptible rice varieties. Interestingly, bacteria are still able to grow at wild-type levels in the apex of the leaf, suggesting a requirement of talc for vascular colonization. Potential direct host targets were identified by conducting a transcriptomic analysis of rice leaves challenged with Xoo strain BAI3R vs. BAI3RΔtalC. Among the identified targets, the rice gene Os11N3 was found to be highly induced upon infection by the wild type strain but not the mutant one. A DNA target box for TalC was located in the Os11N3 upstream region and proved to be functional using GUS assays. We also show that the Os11N3 341-bp upstream region is transcriptionnally activated by TalC. Our results demonstrated for the first time that TAL effectors play an important role in the virulence of Xoo strain BAI3R. Our work will contribute to better improve rice for resistance to bacterial leaf blight

An RND (resistance-nodulation-cell division) transporter, called the PseC protein, was identified at the left border of the syr-syp genomic island of Pseudomonas syringae pv. syringae strain B301D. The PseC protein exhibited amino acid homology to a putative RND transporter of Ralstonia solanacearum with identities of 61% (i.e., PseC). The pseC mutant strain showed a larger reduction in syringopeptin secretion (67%) than syringomycin secretion (41%). A β-glucuronidase assay with a pseA::uidA reporter construct indicated that the GacS/A two-component system controls expression of the pseA gene. Expression of the sypA gene by mutant strain B301D-HK4 corresponded to approximately 13% of that by parental strain B301D, whereas the syrB1 gene expression by mutant strain B301D-HK4 was nearly 61%. Mutant strain B301D-HK4 was reduced in virulence by about 58% as compared to parental strain B301D. A drug-supersensitive acrB mutant of E. coli showed increased resistance to acriflavine and tetracycline upon heterologous expression of the pseA, pseB, and pseC genes. Thus, the PseC protein, an RND transporter, has an important role in secretion of syringomycin and syringopeptin. An ATP-binding cassette (ABC) transporter, called the PseF protein, was identified at the left border of the syr-syp genomic island. The PseF protein exhibited amino acid homology to a putative ABC transporter of E. coli W3104 with identities of 57.6% (i.e., PseF to MacB). The pseF mutant strain showed significant reduction in secretion of syringomycin (74%) and syringopeptin (71%). Expression of the sypA gene by mutant strain B301D-HK7 was approximately 6.9% as compared to that of parental strain B301D, while the syrB1 gene expression by mutant strain B301D-HK7 was nearly 14.6%. Mutant strain B301D-HK7 was less virulent by approximately 67% than parental strain B301D. Expression of the pseF gene was induced approximately six times by strain B301D grown on SRMAF, as compared to that of strain B301D grown on SRM. During infection of bean plants by P. syringae pv. syringae strain B728a, expression of the pseF gene increased 3 days after inoculation. Thus, the PseF protein, an ABC transporter, responsible for secretion of syringomycin and syringopeptin is required for full virulence.

La bactériose vasculaire du riz (BLB) et à stries foliaires (BLS) causées respectivement par Xanthomonas oryzae pv. oryzae (Xoo) et X. oryzae pv. oryzicola (Xoc) sont deux maladies émergentes en Afrique de l'Ouest, suite à l'expansion de la culture du riz et à l'introduction de variétés à haut rendement au cours de ces dernières décennies. Trois nouvelles races de Xoo ont été caractérisées en Afrique dont on a montré, sur la base d'une analyse génétique, leur spécificité africaine. En revanche, une étude réalisée sur une dizaine de souches de Xoc isolées au Mali en 2003, démontre qu'elles sont apparentées à des souches de Xoc asiatiques. En Asie, plusieurs gènes de résistance à Xoo ont été identifiés et déployés dans les programmes de lutte contre BLB. Cependant aucun gène de résistance à Xoc n'a été encore identifié chez le riz. Les objectifs de notre étude étaient (i): d'implémenter les collections de souches de Xoo et Xoc Africaines disponibles mais incomplètes, à l'aide de nouvelles campagnes d'échantillonage réalisées de 2009 à 2012 dans différentes zones agroécologiques du Burkina Faso et du Mali, (ii) de déterminer la diversité génétique de ces souches, (iii) d'identifier et caractériser de nouvelles sources de résistance contre BLB et BLS au sein d'accessions de riz cultivées au Burkina Faso. Nos résultats ont montré que les souches africaines de Xoc sont hautement variables tant d'un point de vue génétique que du pouvoir pathogène. L'analyse par PCR de deux effecteurs de types III conservés (xopAJ et xopW) permet de différencier les souches de Xoc en deux groupes, xopAJ étant absent dans la majorité des souches et une insertion de 1050 bp étant détectée dans la séquence codante de xopW de certaines souches. Néanmoins, il apparait que la forte diversité génétique des Xoc n'est pas corrélée à leur origine géographique, ni à la période de collecte, ou à la nature de l'hôte. Les souches de Xoo caractérisées appartiennent toutes à la race A1 qui n'avait pas encore été signalée au Mali. Au regard de la diversité des souches et de leur évolution, il est important d'envisager un plan de surveillance épidémiologique à plus large échelle des populations de Xo dans les régions concernées en Afrique de l'Ouest. Enfin, nous avons montré que certaines variétés de riz cultivées au Burkina Faso présentent un phénotype de résistance spécifique des souches africaines de Xoo et ce, à tous les stades de développement de la plante. Ces données originales contrastent par rapport au phénotype des lignées de riz résistantes de référence (Xa4, xa5 et Xa7 efficaces uniquement au stade de tallage maximum). Eu égard à l'absence de gènes de résistance dans le riz efficaces contre Xoc, ces variétés qui constituent également une source de résistance efficaces contre la diversité des souches de Xoc africaines, offrent potentiellement un nouveau moyen pour assurer le contrôle du BLS au Burkina Faso et éventuellement dans d'autres pays Africains
Bacterial Leaf Blight (BLB) and Bacterial Leaf Streak (BLS) diseases respectively caused by Xanthomonas oryzae pv. oryzae (Xoo) and X. oryzae. pv oryzicola (Xoc) are two emerging diseases of rice in West Africa, due to the recent expansion of rice cultivation and introduction of improved rice varieties over the last decade. Three news Xoo races were characterized based on genetic analysis, demonstrating their african specificity. In contrast, a study achevied on about ten Xoc strains isolated in Mali in 2003, show that they are related to asian Xoc strains. In Asia, several R genes against Xoo have been identified and deployed in breeding program to control BLB. In contrast, no R gene against Xoc has been identified in rice. The objectives of this PhD thesis are to (i) complete the Xo collections of African isolates upon annual sampling operated from 2009 to 2012 in various agroecological areas of Burkina Faso and Mali, (ii) determine the genetic diversity of these strains , (iii) identify and characterize news sources of resistance genes to BLB and BLS within rice accessions cultivated in Burkina Faso.Our results showed that african Xoc are highly diverse genetically and phenotypically. PCR-based analyse of two conserved type III effector gene (xopAJ and xopW) differentiated two groups of Xoc strains, with xopAJ not detected in a majority of African Xoc strains and 1050 bp insertion detected in xopW gene for few strains. However, the high genetic diversity observed among the Xoc strains is not correlated to geographical origin, sampling data or host plant species. Xoo strains characterized belong all to race A1 previously reported by Gonzalez et al. (2007) in Burkina Faso. Given the diversity of X. oryzae strains and their evolution, it is essential to establish a large scale epidemiological monitoring of Xo populations in concerned regions in west Africa.At last, some accessions cultivated in Burkina Faso showed specific resistance to african Xoo strains at all plant development stages. These original data contrast with rice lines carring Xa4, xa5 and Xa7 resistance genes against BLB, which are only effective at maximun tillering stage.Given no sources of effective resistance genes against BLS is available in rice, these accessions which were also efficient against a set of Xoc strains representative of the diversity in Africa, represent a huge potential source for the control of BLS in Burkina Faso, and eventually in others african countries

ENGLISH ABSTRACT: Bacterial canker of stone fruit, caused by Pseudomonas syringae pv. syringae, is one of the most destructive crop diseases in South Africa. Chemical control has failed completely and effective long-term management strategies will have to rely on the breeding of resistant host trees. To assist in such breeding programmes, investigations into the molecular basis of the interaction between P. s. pv. syringae and stone fruit trees have been undertaken in collaboration with the ARC-Fruit, Wine and Vine Research Institute in Stellenbosch. The aim of this dissertation was to clone and identify genes that are involved in interaction between the bacterial canker pathogen and stone fruit trees. In the first part of the study, the harpin encoding gene of a local strain of the pathogen, P. s. pv. syringae NV, was amplified in a polymerase chain reaction (PCR) strategy with primers based on the hrpAZB sequences of the bean pathogen, P. s. pv. syringae 61. Sequencing of this hrpZpssNvgene revealed a high degree of homology (96%) between the harpin encoding genes and harpin proteins of the two strains. The hrpZpssNvgene was subsequently cloned into the pMAL-c2 expression vector and expressed in Escherichia co/i. This system was used for the production of purified, biologically active, recombinant HrpZpSSNV protein. In the second part of the study, differential display (DD) technology was used to identify genes that are induced in stone fruit trees in response to P. s. pv. syringae and/or its harpin elicitor. For this purpose, actively growing shoots of two Prunus sa/icina cultivars, the moderately resistant cv. 'Laetitia' and the highly susceptible cv. 'Songold' were treated with recombinant harpinpssNvprotein or live P. s. pv. syringae NV bacteria. An untreated control and wounding control was included in the experiment. Total RNA was isolated for comparative mRNA analysis 24 hours after treatment. DD profiles were generated with fifteen primer combinations. Eight candidate bands were re-amplified, cloned and sequenced. Reverse transcription PCR was employed to verify the expression patterns of the cloned bands in the original RNA sample set. Two bands, DDc and DD4 were shown to be differentially expressed between treatments and/or cultivars, while no differences in the expression levels of the remaining six bands (DDa, DDe, DD3, DD5, DD6 and DD7) were observed. BLAST similarity searches yielded significant matches for DDe, DD4 and DD7 with plant defense-related genes.

Trutranskriptase-PKR is gebruik om die ekspressiepatrone van die gekloneerde bande in die oorspronklike RNA monsters na te gaan. Daar is vasgestel dat twee van die bande, DDc en DD4, differensieel tussen kultivars en/of behandelings uitgedruk is, terwyl geen verskille in die ekspressievlakke van die oorblywende ses bande (DDa, DOe, 003, DOS, 006 en DO7) waargeneem is nie. BLAST-soektogte het betekenisvolle ooreenkomste vir DDe, DD4 en DD7 met plant weerstandsgeassosieerde gene opgelewer.

Multi-string configuration is commonly seen in practical photovoltaic (PV) applications, to secure the Maximum Power Point (MPP) for each string, individual string is equipped with its own DC-DC converter for voltage regulation. This study presents an integrated framework that achieves dual purposes simultaneously: maximum power point tracking (MPPT) control and detection of the change in internal resistance. An multi-channel extremum seeking control (ESC) strategy is developed to maximize the power output of multi-string PV system by regulating the voltage input to the DC-DC converter, overall MPPT can be obtained by delivering the power from strings to the DC bus. Simulation results show that the ESC can achieve multi-string overall MPPT with periodic dither signals such as sinusoidal and square wave signals. As change in internal resistance is coupled with many degradation factors for the PV cell, e.g. increase in the series resistance and decrease in the shunt resistance, the capability of integrated detection of internal resistance can be used as a diagnostic probe for PV online diagnostics. Square Wave ESC dither signal is a useful tool to detect the change of internal resistance as the rise time or settling time on a square wave rising edge is more amplified with internal change of a PV system. Therefore, the voltage output transient under square-wave dithering can be changed remarkably with degradation induced change of internal resistance. Simulation results have also confirmed such correlation. The ESC with square wave dithering can thus provides the dual benefits of MPPT and internal resistance change simultaneously.

This paper presents a Maximum Power Point Tracking (MPPT) strategy for multi-string photovoltaic (PV) systems using the Simultaneous Perturbation Stochastic Approximation (SPSA) algorithm. The multi-string PV system considered is a decentralized control configuration, controlling the voltage reference to each PV module but based on the feedback of the total power at the DC bus. This requires only one pair of voltage and current measurements. The MPPT control problem for such topology of multi-string PV systems features a high input dimension, which can dramatically slow down the searching process for the real-time optimization process involved. The SPSA algorithm is considered in this study due to its remarkable capability of fast convergence for high dimensional search problems endorsed by various applications recently. Simulation study is performed for an 8-string PV system, and experimental study is performed for a 4-string PV system. Good performances are observed for both simulation and experimental results.

Induced Systemic Resistance (ISR) is a highly variable property that can be induced by compost amendment of potting media and soils. For example, previous studies showed that only 1 of 79 potting mixes prepared with different batches of mature composts produced from several different types of solid wastes were able to suppress the severity of bacterial leaf spot of radish caused by Xanthomonas campestris pv. armoraciae compared with disease on plants produced in a nonamended sphagnum peat mix. In this project, microbial consortia in the rhizosphere of plants grown in ISR-active compost-amended substrates were characterized. The plants used included primarily cucumber but also tomato and radish. Rhizosphere microbial consortia were characterized using multiple molecular tools including DGGE (Israel) and T -RFLP (Ohio) in both ISR-active field plots and potting media. Universal as well as population-specific bacterial and fungal PCR primers were utilized. T -RFLP analyses using universal bacterial primers showed few significant differences in overall bacterial community composition in ISR-active and inactive substrates (Ohio). In addition, the community members which were significantly different varied when different ISR-activecomposts were used (Ohio). To better characterize the shifts in microbial community structure during the development of ISR, population specific molecular tools were developed (Israel, Ohio).-PCR primers were designed to detect and quantify bacterial groups including Pyrenomycetes, Bacillus, Pan toea, Pseudomonas, Xanthomonas and Streptomyces as well as Trichoderma and Fusarium; two groups of fungi that harbor isolates which are ISR active (Isreal and Ohio). Bacterial consortia associated with cucumber plants grown in compost-amended potting mixtures were shown to be dominated by the phylogenetic taxon Bacteroidetes, including members of the genus Chryseobacterium, which in some cases have been shown to be involved in biocontrol (Israel). Nested-PCR-DGGE analyses coupled with long l6S rDNA sequencing, demonstrated that the Chryseobacteriumspp. detected on seed and the root in compost-amended treatments were derived from the compost itself. The most effective ISR inducing rhizobacterial strains were identified as Bacillus sp. based on partial sequencing of l6S rDNA. However, these strains were significantly less effective in reducing the severity of disease than Trichoderma hamatum382 (T382). A procedure was developed for inoculation of a compost-amended substrate with T -382 which consistently induced ISR in cucumber against Phytophthora blight caused by Phytophthora capsiciand in radish against bacterial spot (Ohio). Inoculation of compost-amended potting mixes with biocontrol agents such as T -382 and other microbes that induce systemic resistance in plants significantly increased the frequency of systemic disease control obtained with natural compost amendments.

The research problem: Bacterial spot and bacterial speck diseases of tomato are causedby strains of Xanthomonas campestris pv. vesicatoria (Xcv) and Pseudomonas syringae pv.tomato (Pst), respectively. These bacteria colonize aerial parts of the plant and causesignificant losses in tomato production worldwide. Protection against Xcv and Pst bycultural practices or chemical control has been unsuccessful and there are only limitedsources of genetic resistance to these pathogens. In previous research supported in part byBARD IS-3237-01, we extensively characterized changes in tomato gene expression uponthe onset of spot and speck disease resistance. A remarkable finding of these studies wasthe inducibility in tomato leaves by both Xcv and Pst strains of genes encodingtranscriptional activator of the GRAS family, which has not been previously linked todisease resistance. Goals: Central goals of this research were to investigate the role of GRAS genes in tomatoinnate immunity and to assess their potential use for disease control.Specific objectives were to: 1. Identify GRAS genes that are induced in tomato during thedefense response and analyze their role in disease resistance by loss-of-function experiments.2. Overexpress GRAS genes in tomato and characterize plants for possible broad-spectrumresistance. 3. Identify genes whose transcription is regulated by GRAS family. Our main achievements during this research program are in three major areas:1. Identification of tomato GRAS family members induced in defense responses andanalysis of their role in disease resistance. Genes encoding tomato GRAS family memberswere retrieved from databases and analyzed for their inducibility by Pst avirulent bacteria.Real-time RT-PCR analysis revealed that six SlGRAS transcripts are induced during theonset of disease resistance to Pst. Further expression analysis of two selected GRAS genesshowed that they accumulate in tomato plants in response to different avirulent bacteria orto the fungal elicitor EIX. In addition, eight SlGRAS genes, including the Pst-induciblefamily members, were induced by mechanical stress in part in a jasmonic acid-dependentmanner. Remarkably, SlGRAS6 gene was found to be required for tomato resistance to Pstin virus-induced gene silencing (VIGS) experiments.2. Molecular analysis of pathogen-induced GRAS transcriptional activators. In aheterologous yeast system, Pst-inducible GRAS genes were shown to have the ability toactivate transcription in agreement with their putative function of transcription factors. Inaddition, deletion analysis demonstrated that short sequences at the amino-terminus ofSlGRAS2, SlGRAS4 and SlGRAS6 are sufficient for transcriptional activation. Finally,defense-related SlGRAS proteins were found to localize to the cell nucleus. 3. Disease resistance and expression profiles of transgenic plants overexpressing SlGRASgenes. Transgenic plants overexpressing SlGRAS3 or SlGRAS6 were generated. Diseasesusceptibility tests revealed that these plants are not more resistant to Pst than wild-typeplants. Gene expression profiles of the overexpressing plants identified putative direct orindirect target genes regulated by SlGRAS3 and SlGRAS6. Scientific and agricultural significance: Our research activities established a novel linkbetween the GRAS family of transcription factors, plant disease resistance and mechanicalstress response. SlGRAS6 was found to be required for disease resistance to Pstsuggesting that this and possibly other GRAS family members are involved in thetranscriptional reprogramming that takes place during the onset of disease resistance.Their nuclear localization and transcriptional activation ability support their proposed roleas transcription factors or co-activators. However, the potential of utilizing GRAS familymembers for the improvement of plant disease resistance in agriculture has yet to bedemonstrated.
