Повний текст джерелаMyocradial ischemia-reperfusion injuries require cardiprotective approaches to improve the repermeabilization of coronaries arteries. So in this work we looked at the mitochondrial permeability transition pore (mPTP) inhibition as this pore is implicated in ischemia-reperfusion injury. We used two pharmalogical strategies to inhibit it. (i) the first one targeted a mitonchondrial outer membrane receptor, the peripheral benzodiazepine (PBR), which is a putative component of mPTP. The cardioprotectio, mediated by PBR was evaluated by 4'-chlorodiazépam (CDZ) a specific PBR ligand. We used a model of global ischemia-reperfusion in isolated rat hearts and a model of regional myocardial ischemia-reperfusion in anaestheized rat. We showed that CDZ reduced infarct size afetr ischemia-reperfusion, improved mitochondrial funcions and decreased apotosis. We observed a reduction of cytochrome C and apoptosis inducing factors (AIF) release with a limitation of membrane permeability and with mPTP inhibition. We also demonstrated that this effect was related to a modification of the balance of the Bel-2 family proteins at the level of the mitochondrial outer member. Indeed, DCZ stabilized the association of bel-2 with the mitochondrial membrane and reduced the interaction of Bax. (ii) The second strategy aimed at stimulation the reperfusion injury salavage kinase (RISK), an endogenous protection cascade implied in pre and postconditioning, asthe cardioprotective effect mediated by RISK was suggested to be linked to mPTP inhibition. We used morphine a well-known cardioprotective opioid ligand which was shown to activated the RISK pathway and to inihibit glycogen synthase kinase -3beta (GSK-3beta). We showed that morphine reduced infarct size in a model of regional myocardial ischemia-reperfusion in rats improved mitochondrials functions. This cardioprotective effect was confirmed in isolated adult rat cardiomyocytes subjected to anoxia-reoxygenation as morphine delayed mPTP opening and increased cell survival. The protection afforded by morphine in vitro and in vivo was mimicked by the GSK-3Beta inhibitor SB16763, and was abolished by mortmannine a PI3K inhibitor. These results confirmed the involvement of the RISK pathway in the cardioprotective effect of both morphine and SB216763 and demonstrated a link between this pathway and mPTP inhibition. In a last part we showed that these two strategies had in common an antioxydant effect which appears necessary ti inhibit mPTP and to protect cardiomyocytes. Taken together these results demonstrate and pharmacological strategy limiting the mitochondrial permeability acting on mPTP is essential to protect the myocardium

Повний текст джерелаOne of the marked societal phenomena in recent decades is the aging of populations due to continually increasing lifespans and as a result, a considerable surge in the number and proportion of elderly, particularly in Western countries. In this demographic context, the rise of chronic diseases related to aging, including Alzheimer’s disease and other types of dementia, has become a major public health issue. The impact of modifiable environmental factors, evolution of the pathogenic mechanisms involved, and the lack of curative treatments illustrates the need for the development of interventions to prevent or delay the onset of these aging-related diseases. The present work demonstrates the importance of using age-adapted study models and experimental methods with the goal towards slowing or delaying age-related deleterious processes. Secondly, our results have identified membranes as an essential part for normal brain function. The composition and architectural changes in the neuronal membranes of elderly mice disrupt their functionality and reduce neuroprotective responsiveness such as those sought by anti-Alzheimer’s therapies. We also observed similar pro-aging-type changes in brain membranes of dyslipidemic mice fed a high-fat diet. Thus, disturbances of lipid homeostasis are correlated with an increased risk of developing aging-related cardiovascular and metabolic as well as neurodegenerative diseases. We finally demonstrated the preventive potential of dietary supplementation with docosahexaenoic acid, the most abundant long-chain polyunsaturated fatty acid in the brain, and observed its ability to restore a neuroprotective response that was impaired in older mice

Повний текст джерелаThe emergence and spread of bacteria that are multi-resistant to antibiotics has become a real public health concern. In particular, P. aeruginosa is one of the major human opportunistic pathogens for which it is urgent to develop new therapeutic strategies. P. aeruginosa is a microorganism that preferentially targets immuno-compromised individuals. Thus, patients suffering from cystic fibrosis, cancer, AIDS, or hospitalized for a long period are subjects at high risk of P. aeruginosa infection. In these patients, these infections are associated with high morbidity and mortality, due to the persistence and resistance of bacteria colonizing host tissues most often within biofilms.My thesis work is part of the objective of discovering new targets for the design of future innovative treatments. In this manuscript I describe the latest elements of characterization of the BAC system (for “Biofilm Associated Cluster”), which allow us to consider it as an original Psp system. This system is thus made up of 6 proteins encoded by the bacABCDEF operon. The expression of this operon would occur in response to stimuli and would be dependent on the sigma factor σ54 and the Gac/Rsm two-components system. The structure of BacA (protein presenting a DUF2170 domain), newly elucidated, is homologous to that of T3SS chaperone proteins suggesting that BacA is involved in the regulation of bacABCDEF expression. The BacB protein is a structural homolog of the PspA and Vipp1 proteins, suggesting that the BAC system is involved in membrane protection under the action of a yet unknown extra-cytoplasmic stress. The BacD protein appears to be a flotillin, a “scaffolding” protein known to reside in functional membrane microdomains, also called lipid rafts

Повний текст джерелаPseudomonas aeruginosa rhamnolipids are glycolipids known to trigger defense responses in grapevine and Arabidopsis thaliana but their mode of perception by plants is still unknown. As amphiphilic compounds, rhamnolipids have been proposed to interactdirectly with plasma membrane lipids. Here we show, by a biophysical approach involving solid state NMR and in silico molecular dynamic simulations, that the insertion of rhamnolipids does not disturb the dynamic of plant plasma membrane models. Inorder to characterize early gene expression modifications triggered by rhamnolipids a micro-array study on A. thaliana was realized, revealing a large transcriptional change. The potential of rhamnolipids to protect the agronomical plant Brassica napus was also investigated. Rhamnolipid triggering of chemical and physical defenses associated with efficient protection against the opportunistic pathogenic fungus Botrytis cinerea, used as a model, was shown. Those results highlight a real potential of RLs as biocontrol agents for Brassicaceae protection
