Повний текст джерелаAculeatins and FR235222 are two families of natural products that are highly effective against apicomplexan parasites, responsible for malaria and toxoplasmosis. In the first part of this work, we developed a new reaction involving a “one pot” phenolic oxidation cascade sequence using a hypervalent iodine reagent in catalytic condition. This flexible synthetic strategy will increase accessibility to new aculeatin derivatives to achieve bioactive compounds. In the second part, we were interested in FR235222, an HDACi (histone deacetylase inhibitor) which targets the class I human HDAC and parasite T. gondii HDAC3 responsible for toxoplasmosis. HDACs are proteins that play an important role in the control of epigenetic mechanisms. In this study, a FR235222 fluorescent derivative was synthesized to confirm the FR235222 target in human cells through cellular localization studies. The synthesis and assessment of the activity of new HDACi analogues, combined with molecular modelisation studies, allowed to demonstrate for the first time that the keto-hydroxyl zinc binding group and the flexibility of the linker would be responsible for this selectivity on human class I HDACs. Finally, with these results in hand and the identification of structural differences between human and parasitic HDAC3, prediction studies (docking) have revealed structural determinants to design inhibitors selective for apicomplexan parasites HDACi

Повний текст джерелаThis work is developed in 3 chapters. The first chapter describes an ethopharmacological work in two Mestizos communities from Peruvian Amazonia. Results include an inventory of 46 plants grouped according to their uses and traditional preparations. In vitro activities on three parasites (Plasmodium falciparum, Leishmania donovani, Trypanosoma brucei gambiense) and their cytotoxicity are reported. Among all these plants Grias neuberthii (Lecythidaceae) and Costus curvibracteatus (Costaceae) showed a strong antiparasitic activity, associated with a strong cytotoxicity for C. curvibracteatus.The second chapter describes the exploration by mass spectrometry (MS) in a biomimetic environment mimicking the digestive vacuole of Plasmodium (DVP), the intermolecular bond between heme and ligands. Results for methoxyflavones suggested that there is no positive relation between the stability of the heme adduct and the biological activity on two P. falciparum strains (3D7 and W2). A correlation was suggested between the presence of a methoxy substitution in R5 of the flavone, heme binding and hydrophobicity (cLogP). This relation could be partially explained by the influence of the carbonyl group on hydrogen bounding. Docking analyses were performed to understand the electrostatic forces involved in the binding. The same kind of study was applied on original fluorescent probes based on artemisinin skeleton (ART). Stability of the heme adduct with the probes, evaluated by CID, showed similarities with ART. Stability of three probes in different conditions mimicking Plasmodium biology were evaluated.The third chapter presents the development of a crude extract biodereplication method, using heme-binding methodology by MS. The plant Piper coruscans (Piperaceae) was selected for this application. MSMS adducts visualization was performed by molecular networking. Targeted products were isolated by centrifugal partition chromatography or preparative liquid chromatography, in one or two steps. Thirteen molecules were isolated, including ten already known products: six flavanones, three chalcones, one alkylamide, one indanone isolated for the first time like a natural product and two new coumpounds: one kavalactone and one cinnamic acid derivative. Among all these molecules, a chalcone validated the biological activity of the plant and showed an intermolecular bound with heme

Повний текст джерелаTropical diseases, including parasitosis, are major public health problems in developing countries. The few existing treatments are becoming less effective due to the emergence of pharmacoresistances, justifying the search for new therapeutic agents with innovative mechanisms. Electrophilic compounds displaying in their structure a Michael acceptor moiety (MA) are able to inhibit some of the parasites enzymatic systems, essential for their survival, and are potent modulators of the parasited host immune response. This confers them antiparasitic properties. In this context, this work presents the isolation of sesquiterpene lactones from the species Pseudelephantopus spiralis (Less.) Cronquist. These secondary metabolites display MA moieties in their structure and are potent biological active compounds. The species Pseudelephantopus spiralis (Less.) Cronquist was chosen on the basis of chimiotaxonomical and ethnopharmacological criteria and due to the very good antileishmanial activity of the ethanolic extract of the aerial parts on Leishmania amazonensis (IC50=0.9 µg/mL). The phytochemical study of P. spiralis led to the isolation of 7 sesquiterpene lactones exhibiting a hirsutinolide-type core in their structure, isolated from petroleum ether, ethanolic and aqueous extracts of plant's aerial parts. The isolated compounds were identified by spectroscopic techniques (NMR, MS) and tested in vitro for their anti-parasitic potential against two models: L. infantum and Plasmodium falciparum. In parallel their cytotoxicity on two healthy cell lines was evaluated in order to determine their selectivity indexes. Two of the isolated compounds, diacetylpiptocarphol and piptocarphin A, displayed a good activity on both parasite models but lacked of selectivity (SI<1). These two compounds were also tested for their effects on the macrophages/monocytes stimulation. Especially the promotion of the expression of the scavenger receptor CD36 on macrophages surface and the cytoprotective Heme Oxygenase enzyme HO-1 was evaluated. CD36 receptor is involved in the recognition and the nonopsonic phagocytosis of Plasmodia-parasitized erythrocytes and HO-1 induction reduces the severity of trypanosomiasis and malaria in murine models. An analytical study using high resolution mass spectrometry and sequential fragmentation was led at the Louvain Drug Research Institute (Brussels) in order to study the fragmentation of isolated hirsutinolides and to develop a method for their detection by UHPLC-DAD-MS/MS in complex extracts. Повний текст джерелаAbstract: Within the ARISTO project, this thesis addresses the need to advance the Environmental Risk Assessment (ERA) of pesticides for soil microbes. Given the magnitude of pesticide applications annually and the possible accumulation of active ingredients and their main residues in soil, there is an urgent need to develop an accurate and effective risk assessment strategy. Particular attention is directed towards ammonia oxidizers, crucial contributors to the soil nitrogen cycling. While soil microbial responses to pesticides are relatively well studied, there is a significant knowledge gap regarding the effects on free-living nematodes, organisms closely interacting with the microbial community. This thesis aims also to explore this gap by investigating the responses of both soil microbes and free-living nematodes communities to pesticide mixtures, through a lab-to-field experimental approach.Molecular techniques such as qPCR and next generation sequencing were employed to study changes in microbial community due to pesticide mixture application, while impacts on free-living nematodes were recorded with the traditional taxonomical identification. Our findings suggest that pesticide mixtures do not have a stronger impact on the microbial and nematode communities than that produced by each of the pesticide studied. Overall, molecular techniques targeting microbes and taxonomical identification of nematode provide comprehensive information on the impact of pesticide mixture on the key players of the soil food web, that could be of interest to be implemented in a further revision of the current pesticides EU regulation

Повний текст джерелаRed microalgae from Rhodella violacea species product a soluble exopolysaccharide release in the media. In this study, optimal culture conditions for exopolysaccharide production were investigated, following photosynthesis activity and culture conditions. This study allowed us to determinate R. violacea optimal media for growth and exopolysaccharide production, which is f/2 media supplemented in nitrogen and phosphorus. Optimal physicochemical parameters are an irradiance of 420 μmol photons.m-2.s-1, a temperature of 24°C, and a pH of 8.3. Photobioreactor of 5 L used to cultivate R. violacea in optimal conditions, gave 0.5 g.L-1 of EPS. Structural analysis of the EPS revealed the production of a proteoglycan, principally composed by xylose, sulfated and with a high molecular mass (1.2 106 g.mol-1). The polymer is complex, as it contains different monosaccharide: rhamnose, galactose, arabinose and glucuronic acid. The antiparasitic effect of polymers from microalgae, and macroalgae were investigated on microsporidia, in vitro against Encephalitozoon cuniculi using Human foreskin fibroblasts, and in vivo against Nosema ceranae using bees. Exopolysaccharide from R. violacea decreases microsporidia growth in vivo and in vitro. In addition, in vivo the polymer allows decrease in bees’ mortality. Polysaccharide from others origins also have antiparasitic effet, such as exopolymer from Porphyridium purpureum and marinum, and sulphated carragheenans which reduce microsporidia growth, and decrease bees’ mortality
