This study seeks to analyze an investigative tool used in several countries and recently regulated in Brazil. The analysis begins with the description of the phenomenon of organized crime and its change throughout history, as well as the evolution of investigative techniques especially the infiltration of agents. Then we draw the legislative path that finally led to Law 12.850/13, that currently rules this institute. We then proceeded to examine each of the diploma elements to form the basis for the core of the work that are the procedural and evidentiary aspects of the tool. In the last chapter, we analyze the adequacy of the investigative technique to the principles of criminal procedure and those inherent to the police infiltration itself.
O presente trabalho busca analisar uma ferramenta investigativa conhecida como infiltra??o policial, usada em diversos pa?ses e recentemente regulamentada no Brasil. A ratiometric, luminescent Eu probe was sought for use in measuring intracellular pH, that can be excited in the range 355-405 nm. In order to achieve this, a series of 1- azathiaxanthone based chromophores has been synthesised and studied to find the best candidate to promote Eu emission. A series of 'D02A' derivatives was synthesised incorporating both the chosen chromophore (2-methyl-l-azathioxanthone) and a pH- dependent binding moiety (a tethered sulfonamide). Measurements were carried out in order to study and understand the properties of the complexes, especially the nature of the pH dependence. The protonation constants of the pendant pH 'switch' in each system were determined by luminescence titration. The influence on Eu emission of some endogenous anions and protein has also been examined by luminescence spectroscopy, along with calculations of their apparent binding constants in order to find the best candidate for measuring intracellular pH. A novel bicarbonate sensor has also been synthesised and studied incorporating a 7-(methylcarbamoylmethyl)-azathioxanthone sensitiser moiety. The luminescence properties have been thoroughly studied, and the changes in the photophysical properties and the sensitivity towards anion and protein binding rationalised. The complex displayed sensitivity towards endogenous anions and protein binding, however, a suitable calibration curve was obtained in the desired 5 - 30 mM HCO(_3) range in simulated endogenous anion mixture, using intensity ratio vs. pH plots. A series of ratiometric Eu(111)complexes has been synthesised incorporating an efficient sensitiser for measuring citrate concentrations in seminal and prostate fluid samples. Their luminescent properties were thoroughly studied using simulated prostate fluid as the background to find the best candidate for prostate adenocarcinoma detection. Preliminary studies have been undertaken to determine citrate levels in 'clinical' prostate and seminal fluid samples. Cellular uptake and localisation studies have also been undertaken with each complex revealing the complex uptake profile and the time-dependent localisation behaviour within the cell. Each complex showed no significant evidence for toxicity. Images were observed using unfixed cells, appropriate for live cell imaging applications.

The use of lanthanide complexes for the detection of biologically relevant species such as anions, pH and metal ions has grown significantly over the past decade. Such probes offer significant advantages over conventional probes; sharp narrow emission lines encode detailed spectral information and allow ratiometric analysis, and their luminescence is long-lived allowing selective spectral acquisition. Many lanthanide-based probes operate in aqueous media, but few have been applied to intracellular measurements. The introductory chapter considers the design of lanthanide based probes for cellular applications. The fundamentals of lanthanide emission are discussed, and how the ligand structure needs to be carefully constructed to maximise emission efficiency. Reported bicarbonate- and pH-responsive probes, both lanthanide based and non-lanthanide based, are reviewed, leading to a set of proposed novel probe structures. The synthesis of these probes with further reasoning behind their design is described in chapter two. The chapter concludes with an overview of the complexes in terms of their emission spectral form, hydration number, HPLC and mass spectrometry properties. Chapter three presents work on bicarbonate-responsive probes. Through a series of spectral titrations, affinity constants for a range of anions were assessed. A high selectivity for bicarbonate was observed over other oxy-anions, and in the presence of protein. The complexes exhibited a mitochondrial localisation profile, and the europium luminescence intensity was modulated reversibly in response to pCO2. The terbium analogues showed negligible change under such conditions, and thus a europium to terbium emission intensity ratio could be used to calibrate pCO2. This principle was also applied to the analysis of bicarbonate levels in simulated biological fluid, and in a serum sample. Several pH-responsive complexes are described in chapter four. Key aspects of pH-probe design were highlighted in early examples, leading to the final set of probes based on reversible sulphonamide ligation. Spectral titrations were performed to assess pKa values, and the use of europium to terbium emission intensity ratios was again found to offer a convenient method for assessing pH. Unprecedented changes in CPL were found in response to pH in these systems, and gem values could be used to report upon pH. The complexes were observed to exhibit a lysosomal distribution. Finally, chapter five contains experimental procedures for each compound synthesised, as well as general experimental procedures. Both sets of complexes described in this thesis show great promise for use as well-defined intracellular probes of bicarbonate or pH.

Limitations in the currently available technology for neural probes impede our progress towards a comprehensive brain activity map. The lack of understanding the brain function leads to limited options for the treatment of neurological disorders. In this thesis, I employed a two-step thermal drawing process (TDP), widely used in fabrication of optical fibers, to create arrays of metal microelectrodes embedded in a polymer cladding. The pitch and size of the electrodes were determined on the macroscale and preserved during the TDP. I have applied these fiber-inspired probes to record spontaneous and stimulated neural activity in vivo.

ENGLISH ABSTRACT: Flanged coaxial probes are widely used to conduct accurate, broadband permittivity measurements of various dielectric materials. A metrology study, discussed in [1], revealed that small perturbations in measured permittivity data, are due to escaping common-mode (CM) current that propagates onto exposed VNA feed cabling. This is not considered in published permittivity extraction algorithms, like the National Institute of Standards and Technology (NIST) full-wave code that assumes an infinite flange radius. To characterise this effect we validate a finite volume time domain (FVTD) CST simulation model of an SMA coaxial probe, by probing sensitive E-fields in a metallic shielding cylinder, placed around it. For this process, electro-optic (EO) E-field sensors are considered and a Mach-Zehnder type sensor is designed. Manufacturing difficulties discontinues this approach, but the revisited extended centre conductor E-field probing technique proves successful. The technique entails a high dynamic range, two-port VNA measurement. Through CST we gain knowledge of the physics behind the CM-problem and the behaviour of fields around the coaxial probe. Different shielding environments are simulated to establish their ability to impede CM-current coupling onto measurement cabling. To study the CM-effect on extracted permittivity results, the investigation is extended to Short-Open-Load (SOL) calibrated face-plane measurements of dielectric solids. A CST model, which considers escaping CM-energy, is used to generate open circuit (OC) calibration coefficients and to serve as an independent extraction method. We inspect the effect of different shielding environments and through CST, extract accurate permittivity results for e00, to a degree not previously achieved for such systems. This allows comment on the infinite-flange-radius assumption of the NIST method and proves the significance of the CM-effect.
AFRIKAANSE OPSOMMING: Geflensde koaksiale probes word algemeen in die praktyk gebruik om akkurate, wyeband permitiwiteitmetings van diëlektriese materiale te verrig. ’n Studie wat in [1] bespreek word, dui aan dat klein verskynsels in gemete resultate, verband hou met gemene-modus (GM) stroom, wat ontsnap en teenwoording is aan die buitekant van onbeskermde voerkabels. Hierdie verkynsel word nie deur huidige volgolf ekstraksie-algoaritmes, soos die van NIST wat ’n onëindige flensradius aanvaar, in ag geneem nie. Om die GM-effek te karakteriseer, verifieer ons ’n eindige-volume tyd-gebied CST simulasiemodel, deur sensitiewe metings binne ’n silidriese metaalskerm, wat om so ’n probe geplaas word. External nuclear medicine diagnostic imaging of early primary and metastatic lung cancer tumors is difficult due to the poor sensitivity and resolution of existing gamma cameras. Nonimaging counting detectors used for internal tumor detection give ambiguous results because distant background variations are difficult to discriminate from neighboring tumor sites. This suggests that an internal imaging nuclear medicine probe, particularly an esophageal probe, may be advantageously used to detect small tumors because of the ability to discriminate against background variations and the capability to get close to sites neighboring the esophagus. The design, theory of operation, preliminary bench tests, characterization of noise behavior and optimization of such an imaging probe is the central theme of this work. The central concept lies in the representation of the aperture shell by a sequence of binary digits. This, coupled with the mode of operation which is data encoding within an axial slice of space, leads to the fundamental imaging equation in which the coding operation is conveniently described by a circulant matrix operator. The coding/decoding process is a classic coded-aperture problem, and various estimators to achieve decoding are discussed. Some estimators require a priori information about the object (or object class) being imaged; the only unbiased estimator that does not impose this requirement is the simple inverse-matrix operator. The effects of noise on the estimate (or reconstruction) is discussed for general noise models and various codes/decoding operators. The choice of an optimal aperture for detector count times of clinical relevance is examined using a statistical class-separability formalism.

This Thesis is concerned with two cosmological probes of linear gravity. The first relates to Large Scale Structure (LSS) in the Universe, probed by galaxy surveys. The second to temperature anisotropics of the Cosmic Microwave Background (CMB), probed by the Wilkinson Microwave Anisotropy Map (WMAP). Both probe the matter and dark energy distributions in the Universe and can be used to test general relativity. The first part of this Thesis (Chapters 2 to 4) is concerned with the analysis of galaxy clustering in redshift space. The second part (Chapters 5 to 7) is concerned with the Integrated Sachs-Wolfe (ISW) effect using LSS-CMB cross-correlations. Chapter 1 introduces the cosmological theory and overviews the subsequent chapters. Chapter 2 gives a review of recent results from the 2 Micron All-Sky Survey (2MASS) and its Redshift Survey (2MRS). It includes work published in Erdogdu (a) et al. (2006) and Erdogdu (b) et al. (2006). Chapter 3 quantifies the clustering of 2MRS galaxies in redshift space. Chapter 4 uses results from Chapter 3 to constrain cosmological parameters. A selection of work from Chapters 3 and 4 will shortly become available in Rassat et al. (2008), entitled 'Redshift Space Analysis of 2MRS'. Chapter 5 overviews the late-time Integrated Sachs-Wolfe effect (ISW) and cross- correlations between the LSS and the CMB. Chapter 6 is also published in Rassat et al. (2007), entitled "Cross-correlation of 2MASS and WMAP3: Implications for the Integrated Sachs-Wolfe effect". It investigates a detection of the ISW effect and correlations which may affect statistical isotropy in the CMB ('Axis of Evil'). Chapter 7 uses the ISW effect to forecast constraints on dark energy parameters and general modifications of general relativity for the next generation of galaxy surveys, particularly the Dark UNiverse Explorer (DUNE) and the Dark Energy Survey (DES). Chapter 8 presents the overall conclusions of this Thesis. Chapter 9 discusses possible extensions to this work.

This thesis aims at analyzing the psychodynamics of work related to public employees on probationary period from a Public Foundation using as methodology the psychodynamic of work. The probationary period is considered a sensitive, determinant and decisive stage in the professional life of the public employee, with individual, institutional and social repercussions. The theoretical framework used is the psychodynamics of work, advocated by Dejours (1992; 2004), having as its practical-methodological development the psychodynamic work clinic. The clinic represents the opportunity of an accessible and safe space for the expression and resignification of the working life of the subject. The study conducted eight clinic sessions with the participation of eleven public employees and five clinicians, with each session enduring for about sixty minutes. All the meetings were recorded and transcribed in full. The content analysis technique was used in order to organize the results and discussions in the following categories: a) work organization, b) meaning of work, c) subjective mobilization and pleasure at work, d) suffering, and e) Defensive strategies. It has been noted that the work organization is rigid, computerized and allows for rare opportunities for participation and discussion; however, on the other hand, it offers substantial institutional benefits such as remuneration, stability and the status of being a public employee. It has been found that it is the employee himself, in most cases, who gives meaning to his work, mobilizes himself, feels pleasure, suffers and defends himself, basing himself mainly on the choices available or on the mere glimpse of another work possibility or even another reality. The suffering appears in the relations with the leadership, in the discredit attributed to the evaluation of performance, as well as in the meeting between the prescribed and the real, considering that what is prescribed in the selection process is different from the activity performed, mostly characterized by monotony and repetition, with little room for modifications and contributions, and thus not sufficient for proper upward mobilization. There is cooperation, but one that is only related to technical aspects connected to work demands. It is noticed the absence of collective coping strategies to what bothers the employees. It was identified that they do not have the sense of belonging to the Foundation, understanding the probationary period as a passing professional stage. Finally, the study shows the development of the psychodynamic of work, within a specific envirment, to make visible the suffering at work and the subjective mobilization. Nevertheless, in the group studied, there was a limitation of the clinic regarding the possibility of producing greater interference and involvement of the workers in the context in which they work: there is a general perception, among the majority of the participants, that the current position is provisional and the change of public office something certain. This limited the number of sessions held, as well as demonstrated the non-engagement of workers in the quest towards changing the organization in which they work, since there is an individual quest to change jobs.
Esta pesquisa tem por objetivo analisar a psicodin??mica do trabalho de servidores p??blicos em est??gio probat??rio de uma Funda????o P??blica, utilizando como metodologia a cl??nica psicodin??mica do trabalho. Considera-se o est??gio probat??rio como uma etapa sens??vel, determinante e decisiva na vida profissional do servidor p??blico, repercutindo individual, institucional e socialmente. O referencial te??rico utilizado ?? a psicodin??mica do trabalho, preconizada por Dejours (1992; 2004), tendo como desdobramento pr??tico-metodol??gico a cl??nica psicodin??mica do trabalho. A cl??nica representa a oportuniza????o de um espa??o acess??vel e seguro para a express??o e ressignifica????o da vida laboral do sujeito. O estudo realizou oito sess??es da cl??nica com a participa????o de onze servidores e de cinco cl??nicos, com cerca de sessenta minutos de dura????o, cada sess??o. Todos os encontros foram gravados e transcritos na ??ntegra. Utilizou-se a t??cnica de an??lise de conte??do, a qual possibilitou organizar os resultados e a discuss??o nas seguintes categorias: a) organiza????o do trabalho, b) sentido do trabalho, c) mobiliza????o subjetiva e prazer no trabalho, d) sofrimento, e e) estrat??gias defensivas. Observouse que a organiza????o do trabalho ?? r??gida, informatizada e permite pouco espa??o para participa????o e discuss??o, mas, por outro lado, oferece expressivos benef??cios institucionais como a remunera????o, a estabilidade e o status em ser servidor p??blico. Constatou-se que ?? o pr??prio servidor, na maioria dos casos, que d?? sentido ao seu trabalho, se mobiliza, sente prazer, sofre e se defende, baseando-se principalmente nas escolhas e no vislumbre de outro trabalho ou mesmo de outra realidade. O sofrimento aparece nas rela????es com as chefias, no descr??dito atribu??do ?? avalia????o de desempenho, bem como no encontro entre o prescrito e o real, tendo em vista que o prescrito em edital ?? diferente da atividade realizada, sendo essa caracterizada pela monotonia e repeti????o, com pouca margem para modifica????es e contribui????es, sendo assim, n??o suficiente para a devida mobiliza????o. Existe coopera????o, sendo est?? ligada somente a aspectos t??cnicos referentes a demandas de trabalho. Percebe-se a aus??ncia de estrat??gias coletivas de enfrentamento do que incomoda os servidores. Identificou-se que eles n??o possuem a sensa????o de pertencimento ?? Funda????o, entendendo a fase de est??gio como um per??odo profissional passageiro. Por fim, o estudo mostra o desenvolvimento da psicodin??mica do trabalho, no ambiente especifico, para tornar vis??vel o sofrimento no trabalho e a mobiliza????o subjetiva. No entanto, no grupo estudado, percebeu-se uma limita????o da cl??nica no que diz respeito a produzir maior interfer??ncia e envolvimento dos trabalhadores no contexto em que atuam: h?? uma ideia, na maioria dos participantes, de que o cargo atual ?? provis??rio e a mudan??a de cargo p??blico algo certo Isso limitou o n??mero de sess??es realizadas, bem como demonstrou o n??o engajamento dos trabalhadores na busca por mudar a organiza????o em que trabalham, uma vez que h?? uma busca individual por mudar de cargo p??blico.

Tais mudan?as perpassam as sociedades contempor?neas de um mundo globalizado. No ?mbito da educa??o, muitas expectativas s?o postas no sistema educacional. Nesse cen?rio, a forma??o e o exerc?cio da doc?ncia se torna uma tarefa cada vez mais complexa e necess?ria para redefinir o papel da educa??o numa sociedade chamada do conhecimento. Aprender a ser professor e tornar-se professor, nesse mil?nio, requer aprofundar a compreens?o a respeito dessa ?rea, t?o estrat?gica, que envolve os sistemas de ensino, a escola e seus professores. O per?odo de inser??o dos professores na carreira docente, em especial na rede p?blica, tem se revelado como uma etapa complexa por esta quest?o estar na g?nese do processo de algu?m aprender a ser professor. Envolveu professores que estavam estreando na doc?ncia e outros que, tendo experi?ncias de sala de aula de outros contextos, eram iniciantes na rede estadual de ensino. Para tanto, buscamos tra?ar o perfil s?cio demogr?fico desses professores; identificar as percep??es e viv?ncias que marcaram o ingresso deles na rede, as principais a??es desenvolvidas, a repercuss?o das mesmas e aspectos que foram conformando o desenvolvimento profissional. Ap?s a defini??o de uma amostra estat?stica, a pesquisa selecionou 84 professores aprovados no concurso realizado em 2011 para o cargo de professor das s?ries iniciais, convocados e nomeados em 2012 e lotados na 1? Dired (Natal). ? luz de procedimentos explorat?rios e descritivos, analisamos um conjunto de dados apoiados nas abordagens qualitativa e quantitativa da pesquisa em educa??o. Para recolha das informa??es utilizamos question?rio com perguntas fechadas e abertas. Os dados coletados foram sistematizados com o aux?lio do Software Modalisa (vers?o 6.0) e para a an?lise destes recorremos ? An?lise de Conte?do (BARDIN, 1977) e aos recursos da estat?stica descritiva. Os resultados evidenciam que, diferentemente do que ? revelado em outros estudos, o ingresso dos docentes nas escolas da rede estadual n?o foi t?o traum?tico. Foi marcado por sentimentos muito mais positivos que negativos, tendo proporcionado a eles descobertas e aprendizagens, al?m de importante protagonismo dos docentes nas atividades que deveriam estar sendo assumidas, formalmente, pela SEEC e compartilhada com as escolas e respectivas inst?ncias de gest?o.
We are more than a decade (2016) living in a new millennium brings us rapid social, political, technological changes, among others. Such changes pervade contemporary societies in a globalized world. In education, high expectations are placed on the educational system. In this scenario, the teaching exercise becomes an increasingly complex and necessary task to redefine the role of education in a society called knowledge. Learning to be a teacher and become a teacher, in this millennium, it requires a deeper understanding about this as strategic area that involves the education systems, the school and its teachers. The insertion period of teachers in the teaching profession, particularly in public has been revealed as a complex step to be concerned the process of learning to be a teacher. These early years are considered profitable for the development of teachers. As can be seen in the arguments of researchers who are supported in the framework of the professionalization of teachers having as representative: Marcelo Garcia (1999), Gauthier (1998); Ramalho, Nu?ez and Gauthier (2004); Cortez (2003), Novoa (1997), Garcia (1999), Day (2001), Hubermam (1995), Veeman (1984) Canary (2001) among others. The present study aims to investigate the integration and development of teachers in the education system of the newborn, whose focus on tr?sanos the probationary period, regulated by Complementary Law No. 322 of 11/01/2006. Involved teachers who were debuting in teaching and others, and classroom experiences from other contexts were beginners in state schools. Therefore, we seek to profile demographic partner of these teachers; identify the perceptions and experiences that have marked their entry into the network, the main actions developed, the impact thereof and aspects that were shaping professional development. After the definition of a statistical sample, the survey selected 84 teachers passed the competition held in 2011 for the post of professor of early grades, called and named in 2012, crowded the 1st Dired. In light of exploratory and descriptive procedures , we analyze a set of data supported the qualitative and quantitative approaches to research in education . To collect the information used a questionnaire with closed and open questions. The data collected were organized with the help of Software Modalisa (6.0 ) and for the analysis of these supported the resort to content analysis ( Bardin, 1977) and descriptive statistics . The results show that , contrary to what is revealed in other studies , the entry of teachers in the state public schools was not as traumatic. It was marked by much more positive feelings than negative , having provided the findings and learning teachers , and their important role in the activities that should be being shared with instances of SEEC and school management.

This work describes the synthesis of new chemiluminescent ester compounds and their applications in immunoassay. The new compounds are 2,6-dibromo-4-(2-succinimidyl-oxycarbonylethyl)phenyl 10-methylacridinium-9-carboxylate trifluoromethanesulfonate (II), 2,6-dimethyl-4-(2-succinimidyloxycarbonylethyl)phenyl 10-methylacridinium-9-carboxylate trifluoromethanesulfonate (III), 2-(2-succinimidyloxycarbonylethyl)phenyl 10-methylacridinium-9-carboxylate trifluoromethanesulfonate (IV), 4,6-dibromo-2-(2-succinimidyloxycarbonylethyl)phenyl 10-methylacridinium-9-carboxylate trifluoromethane-sulfonate (V), 4,6-dimethyl-2-(2-succinimidyloxycarbonylethyl)phenyl 10-methyl-acridinium-9-carboxylate trifluoromethanesulfonate (VI) and 4-bromo-6-methyl-2-(2-succinimidyloxycarbonylethyl)phenyl 10-methylacridinium-9-carboxylate trifluoromethane-sulfonate (VII). All these compounds contain an acridinium ester unit, which provides the chemiluminescent moiety, a phenolate unit as a leaving group and a succinimidyl ester unit as a means of linking to biological molecules. The thesis is divided into two parts. The introduction gives a broad background and understanding of chemiluminescent biological probes. Initially the process of light emission is explained in brief. This is important because it is light emission from a chemical reaction that is chemiluminescence. This is followed by a description of the major chemiluminescence reagents, especially acridinium ester species as examples, and of how these can be used as labels in chemiluminescence immunoassay. The second part describes the synthesis of compounds (II), (III),(IV), (V), (VI), and (VII), together with som basic studies of theirchemiluminescent properties and attachment to Biblogical materials.

One of the primary targets of current and especially future cosmological observations are light thermal relics of the hot big bang. Within the Standard Model of particle physics, an important thermal relic are cosmic neutrinos, while many interesting extensions of the Standard Model predict new light particles which are even more weakly coupled to ordinary matter and therefore hard to detect in terrestrial experiments. On the other hand, these elusive particles may be produced efficiently in the early universe and their gravitational influence could be detectable in cosmological observables. In this thesis, we describe how measurements of the cosmic microwave background (CMB) and the large-scale structure (LSS) of the universe can shed new light on the properties of neutrinos and on the possible existence of other light relics. These cosmological observations are remarkably sensitive to the amount of radiation in the early universe, partly because free-streaming species such as neutrinos imprint a small phase shift in the baryon acoustic oscillations (BAO) which we study in detail in the CMB and LSS power spectra. Building on this analytic understanding, we provide further evidence for the cosmic neutrino background by independently confirming its free-streaming nature in different, currently available datasets. In particular, we propose and establish a new analysis of the BAO spectrum beyond its use as a standard ruler, resulting in the first measurement of this imprint of neutrinos in the clustering of galaxies. Future cosmological surveys, such as the next generation of CMB experiments (CMB-S4), have the potential to measure the energy density of relativistic species at the sub-percent level and will therefore be capable of probing physics beyond the Standard Model. We demonstrate how this improvement in sensitivity can indeed be achieved and present an observational target which would allow the detection of any extra light particle that has ever been in thermal equilibrium. Interestingly, even the absence of a detection would result in new insights by providing constraints on the couplings to the Standard Model. As an example, we show that existing bounds on additional scalar particles, such as axions, may be surpassed by orders of magnitude.

Chitin is a glycopolymer consisting of +-(1, 4)-linked N-acetyl-D-glucosamine residues that occurs widely in nature and is a constituent of many organisms that are pathogenic to humans, including insects, fungi, and parasitic nematodes. As these organisms depend on the ability to break down chitin at key points of their life-cycle, inhibitors of the enzymes termed chitinases that catalyse the hydrolysis of chitin, are of considerable interest as potential drugs and insecticides. Although chitin is absent from mammalian physiology, two human chitinases along with several chitin-binding proteins (chi-lectins) have been associated with the onset or transmission of several major human diseases such as asthma, Legionnaire’s disease and osteoarthritis. Therefore, selective inhibitors of chitinases are now of considerable interest as new drug leads and biochemical probes. Until recently, few broad spectrum chitinase inhibitors had been identified. The natural cyclopentapeptide, argifin, has been shown to be a potent inhibitor of several bacterial-type family 18 chitinases including Aspergillus fumigatus chitinase B1 (AfChiB1). With the aid of high resolution X-ray structures we have designed and prepared linear fragments of the natural product cyclopentapeptide argifin using a combination of SPPS and solution phase synthesis. This has allowed us to determine that the Nmethyl guanylurea motif serves as a starting point for the generation of novel, drug-like, peptidomimetic inhibitors family 18 chitinases. We have also demonstrated that the cis configuration about the Arg(MC)-N-MePhe peptide bond is essential to retain any significant biological activity. This dipeptide motif is also found in another naturally occurring cyclopentapeptide, banyasin A, extracted from the cyanobacteruim Nostoc sp. Banyasin A also contains a rare +-amino acid, 3-amino-2-methyl-5E-octenoic acid (Amoa), in which the stereochemistry at the C3 and C5 of Amoa has not been resolved. The diastereoselective synthesis of Amoa for the preparation of banyasin A has also been established using chiral oxazolidinone-based aldol chemistry, which has allowed us to successfully prepare a single diastereoisomer of the natural product cyclopentapeptide incorporating this +-amino acid. New methodology for the preparation of argifin has also been developed to reduce the propensity towards the formation of undesired side products and to prepare the natural product on a larger scale.

In this thesis we discuss recent results on the medium/probe interaction in the strongly coupled plasma produced in heavy-ion collisions. Such processes involve physics at multiple energy scales making a consistent theoretical description challenging. We show how insights from various regimes can be combined to extend our understanding of the underlying physics. As a first example, we start with the novel contributions to the drag force acting on a heavy quark moving through the strongly coupled holographic plasma. The new terms are proportional to the coefficient of the axial anomaly and to the chiral asymmetry in the medium. These chiral contributions to the drag force act either parallel to or antiparallel to an external magnetic field or to the vorticity of the fluid. We show that the chiral drag force vanishes for heavy quarks that are at rest in a suitably chosen frame in the weak field limit. In this frame, the heavy quark at rest sees counter propagating momentum and charge currents, both proportional to the axial anomaly coefficient, but feels no drag force. This provides strong concrete evidence for the absence of dissipation in chiral transport. Then, we extend this result introducing a concept of an "anomalous wind" felt by probes at rest. This phenomenon modifies previous results on the medium/probe interaction. We demonstrate that by deriving the anomalous correction to the screening length of the heavy quarkonium color potential. Finally, we discuss the strong magnetic field limit to the leading order in the weak medium/probe coupling. It is shown that the drag force is suppressed along the magnetic field and in the exact chiral limit tends to zero. This anisotropy in the drag force is present in the system even at zero chiral asymmetry and may lead to strong observable effects. Then, we turn to the main available probes of the plasma produced in experiments - jets. We supplement the holographic description corresponding to the infinite medium/probe coupling limit with an initial parameter distribution for an ensemble of jets motivated by the weakly coupled dynamics and similar to the case of proton-proton collisions. The model constructed in this way, combining insights from both weakly and strongly coupled regimes, is used to analyze the evolution of an ensemble, as it propagates through an expanding cooling droplet of strongly coupled plasma as in heavy ion collisions. Each jet in the ensemble is represented holographically by a string in the dual 4 + 1-dimensional gravitational theory. Firstly, the full string dynamics is approximated by assuming that the string moves at the speed of light. We study the evolution of the jet opening angle distribution upon propagating the droplet and study the medium effect on the mean opening angle within a simple two parametric phenomenological model. Then, we extend this result analyzing the full string dynamics for a range of possible initial conditions. That gives access to the dynamics of holographic jets just after their creation. We analyze the full jet shape modification and find the result that the jet shape scales with the opening angle at any particular energy. Further, we construct an ensemble of dijets with energies and energy asymmetry distributions taken from events in proton-proton collisions and jet shape taken from proton-proton collisions and scaled according to our result. We study how these observables are modified after the ensemble of dijets is propagated through the strongly-coupled plasma. The results of this simple model is in qualitative agreement with the experimental data.

In this thesis, I investigate several ways to probe gravity in the strong-field regime. These investigations focus on observables from the gravitational dynamics, i.e. when time derivatives are large: thus I focus on sources of gravitational waves. Extreme mass-ratio inspirals (EMRIs) can be very sensitive probes of strong-field physics. Predicting observables from EMRIs must be done numerically, so accurate numerical methods are required to ensure that any comparison with measurement is not spoiled by numerical artefacts. The first investigation of this thesis is a spectral (in the angular sector), pseudospectral (in the radial sector) time-domain PDE solver for perturbations of a Kerr black hole (i.e. solving the Teukolsky equation). The method exhibits good convergence and prompts much future investigation. A second approach to probing strong gravity is to consider theories which are general relativity (GR) with a few small corrections and investigate the effect of these corrections on observables. Since gravitational waves are the prime observable and they control the long-term evolution of dynamical systems, I investigate their properties in almost-GR theories. The second investigation of this thesis is a study of the propagation and energy content of gravitational waves in these theories. I find that in a large class of theories, approaching the asymptotically at part of spacetime, gravitational waves propagate in the same fashion as in GR and have the same effective stress-energy tensor as in GR. Next, I study the strong-field correction to the structure of a Schwarzschild black hole in a class of theories. Finally, with these ingredients, I investigate the leading corrections to the dynamics and observables of a comparable mass-ratio inspiral using post-Newtonian techniques. The main result is the appearance of dipolar scalar radiation in this class of theories. The dipolar radiation has a frequency dependence which does not arise in GR and is a distinct signature of corrections. Such signatures should be testable using gravitational wave detection and pulsar timing.

Electromechanical probing applications continuously require smaller pitches, faster manufacturing and lower electrical resistance. Conventional techniques, such as MEMS based cantilever probes have their shortcomings in terms of the lowest pitch that can be achieved, cost and yield. Given their promising mechanical and electrical properties, carbon nanotubes (CNTs) are strong candidates for future probing applications. A new class of metal-CNT hybrid electromechanical probes is presented where vertically aligned carbon nanotube structures, grown with a chemical vapor deposition (CVD) technique, act as elastic springs, and a metal coating on the probes is used for increased electrical conduction. This design and architecture presents a scalable approach where thousands of probes can be fabricated in very short production times. 1.5 Ohm resistance and reliable performance for 6000 cycles at 50 [mu]m over-travel was achieved for a column of 200 [mu]m x 200[mu]m cross-section and 1plm of Au deposition. In-situ scanning electron microscope mechanical compression tests revealed a unique deformation mechanism of the CNT structures where continued compression results in successive buckle formation which later can serve as micro-bellows and elastic springs.
(cont.) A novel stiffness tuning method is presented to control the elastic properties of a given CNT probe by controlling the initial compressing amount. Further stiffness tuning is achieved by changing gas composition during CVD growth where CNT diameter and density is modified. Lateral compression and densification tests show that these CNT structures are highly anisotropic and have very different deformation mechanisms in vertical and lateral directions. Mechanical properties resulting from two main CVD growth techniques, namely fixed catalyst where a thin film of catalyst layer is deposited onto the growth substrate, and floating catalyst where the catalyst particles are introduced in the gas phase, are compared. It is found that floating catalyst CVD growth yields much stiffer structures due to the relatively larger CNT diameters. As the adhesion of CNT structures to the growth substrate is very weak and the support layer is typically an insulator, a versatile transfer printing technique is developed which enables simultaneous placement and reinforcement of the probes on a wide range of substrates, including metals and printed circuit boards. Electromechanical performance and failure mechanisms of fully functional metal-CNT hybrid probes are presented.

Initially, complexes containing a trifluoromethyl group were examined for 19F MRI/S, due to the large chemical shift range, favourable NMR properties, and the absence of a background signal in vivo. The use of paramagnetic lanthanide ions increases relaxation rates, allowing faster acquisition times and increased signal intensity per unit time. In addition, chemical shift non-equivalence is enhanced, which is important for the development of responsive probes. Examples are presented that report selectively on changes to citrate concentration levels in the presence of other anions or signal enzyme hydrolysis. To improve signal intensity for in vivo applications, conjugate systems have been developed containing a number of fluorinated lanthanide complexes covalently bound to high molecular weight adducts (glycol chitosan, PAMAM dendrimer). These systems resulted in one major species being observed in solution, despite the broad molecular weight ranges exhibited by these polymeric entities. Preliminary in vivo MRI studies in animal models were undertaken with the glycol chitosan conjugate. In order to enhance the signal further, a set of fluorinated complexes was synthesised containing a pyridyl moiety designed to increase the rigidity of the overall system, thereby reducing dynamic exchange broadening. Additionally, the 19F NMR spectra of these complexes displayed much larger lanthanide induced shifts than observed for the previous systems. By substituting the CF3 group for a tert-butyl moiety, the corresponding 1H NMR signal was shifted well beyond the standard diamagnetic range, leading to the development of a new series of complexes for use as 1H MRI and MRS probes. These complexes have been applied to preliminary in vivo MRI studies. A further system has been developed that displays pH-dependent behaviour, observed by both 1H NMR spectroscopy and in phantom imaging studies.

The time in the history of the Universe when the first stars and galaxies formed and ionized the gas in the intergalactic medium is known as the Epoch of Reionization. This transformative time period, which took place within the first billion years after the Big Bang, is still relatively unexplored due to the significant difficulties associated with observing so far back in time. The theme of this thesis is tying together existing (mostly indirect) and upcoming observations with simulations. Papers I and II deal with Lyα emitting galaxies. The Lyα emission line is very sensitive to neutral hydrogen, which was plentiful during the Epoch of Reionization. Therefore, observations of distant Lyα galaxies may be used to indirectly tell us something about when and how reionization took place. Properly interpreting the observations is, however, far from straightforward. In Paper I we develop a new method for combining large, low-resolution simulations of the intergalactic medium with small, high-resolution simulations of individual galaxies. We use this method to reproduce existing observations. In Paper II we use the same method to make predictions for future observations. Another observational probe of the EoR is the 21-cm emission line from neutral hydrogen. This line is the most promising probe for directly studying the neutral gas in the early Universe, and several radio telescopes are currently gathering data to observe it. The 21-cm signal is affected by a multitude of cosmological and astrophysical effects, all of which need to be understood in order to interpret the upcoming observations. One such effect is the non-random shifts in redshifts caused by the peculiar velocity of matter flowing towards higher-density regions. This effect, known as redshift space distortions, is the topic of papers III and IV, while paper V deals with another observational effect called the lightcone effect.

The chirality of biological systems can be probed using highly emissive lanthanide complexes with the aid of circularly polarised luminescence and emission spectroscopy. Such chirality probes can be synthesised through the incorporation of a remote chiral centre within the ligand framework, which can preferentially stabilise a particular stereoisomer giving an enantiopure complex of well-defined helicity. Alternatively, lanthanide chirality probes can be derived from achiral or dynamically racemic ligands, where the selective induction of a CPL signal can be monitored as a function of the nature and concentration of a selected chiral analyte. A series of chiral lanthanide complexes has been synthesised. Each complex is based on an amide substituted 1,4,7-triazacyclononane system derived from either R-(+) or S-(-)-α-methylbenzyl amine. The stereochemistry of the amide moiety controls the helicity of the complex, and one major diastereoisomer is formed for each lanthanide metal. The absolute stereochemistry of the major diastereoisomer was determined by X-ray crystallography (S-Δ-λλλ and R-Λ-δδδ). Inclusion of an aryl-alkynyl chromophore generated complexes that exhibited large extinction coefficients (up to 55,000 M-1 cm-1) and high quantum yields (up to 37%) in water. A second set of bright Eu (III) complexes has been prepared based on an achiral heptadentate ligand system, which vary in the nature of the pyridyl donor (phosphinate, carboxylate and amide). The binding of a number of chiral acids including lactate, mandelate and cyclohexylhydroxyacetate was monitored by a change in the emission spectrum and the induction of strong CPL. Empirical analysis of the ΔJ = 4 region of each of the Eu (III) complexes allows an assignment of the complex-anion adducts as R-Δ and S-Λ. Furthermore, variations in the sign and magnitude of CPL allow the enantiomeric purity of samples with unknown enantiomeric composition to be assessed. Finally, several dynamically racemic lanthanide chirality probes have been synthesised and characterised. Induced CPL has been assessed, which arises as a result of the change in complex constitution upon binding to important chiral biomolecules such as, sialic acid, O-phosphono-amino acids and peptides and oleoyl-L-lysophosphatidic acid (LPA). This work presents the first example of induced CPL in the detection of cancer biomarkers, sialic acid and LPA, and demonstrates the utility of this class of dynamically racemic Eu (III) complexes as chirality probes.

Using three complementary approaches, this work sought to tackle the widespread problem of antibiotic resistance. To circumvent the resistance mechanisms developed by bacteria, it is necessary to establish drug candidates that act on novel therapeutic targets, such as the ion channels used by bacteria to modulate homeostasis. Examples include the potassium efflux channel, Kef, and the mechanosensitive channel of small conductance, MscS, which are not found in humans. How these targets function must be well understood before drug candidates can be developed, as such, their identification and investigation is often accompanied by the evolution of the analytical techniques used to study them. Membrane protein mass spectrometry is one technique showing potential in the study of ion channels. However, spectra can be clouded by the detergents used to solubilise ion channels from their native membranes. Undertaken herein was the synthesis of some fluorescent glycolipid detergents, which it was hypothesised could be encouraged to dissociate from ion channels via laser-induced excitation within the gas phase of a mass spectrometer, thereby improving the clarity with which spectra can be obtained. For Kef, an unconfirmed mechanism of action had previously been proposed. To explore the suggestion that sterically-demanding central residues are important for channel activation, solid phase peptide synthesis was used to isolate three tripeptide analogues of N-ethylsuccinimido glutathione, a known activator with a high affinity for Kef. A competition fluorescence assay suggested these tripeptides bound to Kef with an affinity lower than predicted, allowing the conclusion that a more detailed assessment of the steric bulk required for activation was necessary before a mechanism of action could be confirmed. Lysophosphatidylcholine has been shown to activate MscS, although it is not known how. Affinity chromatography between MscS and lysophosphatidylcholine was proposed as a means by which specific binding interactions could be investigated. For this technique an amino-derivative of lysophosphatidylcholine was necessary and its challenging synthesis is also detailed herein.

Probation and mediation service is an institution which has been gaining increasingly vital and important position in the field of criminal law. The service focuses on a very wide range of subjects and its activities aim not only to monitor but also to help. It realizes the execution of alternative sentences and also shows activity in pre-trial proceedings and in proceedings before the court decision. It cooperates with many different organizations, it is involved in many projects. For criminal justice it has become an essential and equal partner in dealing with the consequences of crimes committed by both adult offenders and juveniles. It contributes to the favourable development of the re-education of accused and convicted people, supervises proper execution of sentenced alternative punishments and the achievement of their purpose. It makes sure that the collective process was fulfilled to deal with all the adverse consequences of illegal acts of the offenders and that the victims utilize their legal rights and entitlements. Probation and mediation service provides a comprehensive view of both criminal activities and the personalities of offenders.

O presente trabalho teve por objetivo identificar os fatores que concorrem para que alguns indivíduos na terceira idade se encontrem em situação de pobreza. Mais concretamente, pretendeu-se identificar o perfil socioeconómico de pessoas em situação de pobreza através de um modelo explicativo probit, que estimasse o peso de vários fatores para a referida situação. Porque em Portugal, mais concretamente em zonas rurais, existe uma forte componente de autoconsumo, entrámos em linha de conta com as linhas de pobreza monetária e não-monetária. Esta caracterização é feita a partir do Inquérito às Despesas das Famílias IDEF 2010/2011. O modelo revelou o impacto na probabilidade de se ser pobre (em termos monetários e totais) de variaveis como o nível de ensino do idoso, a sua idade, tipo de ADP onde se encontra inserido, localização rural/ urbana e fonte mais frequente de rendimento. Foram ainda discutidas as implicações do modelo para políticas sociais de apoio.
The present study had the aim to indentify the factors associated to poverty in the elderly. More precisely, we intended to indentify the socioeconomic profile of people in poverty situation through a probit model, capable of estimating the weight of various factors for the referred situation. Because in Portugal, more specifically, in rural areas, there is a strong component o autoconsumption, we took into account the monetray and non-monetary poverty lines. This characterization was made using the Housing Buget Survey 2010/2011. The model revealed an impact on the probability of being poor (in terms of monetary and total income) of variables such as education, age, type of household, and main type of income. We also discuss the implications of the model for social policies.
