The aim of this study was to investigate the stability of positional identity markers and phenotypic differences in isolated osteoblasts from distinct anatomic regions. In addition, the ability of heterotypic co-cultures to reprogramme site-specific Hoxa gene expression also tested. Rat osteoblastic cells from femurs and calvariae were harvested as matched pairs of cultures from 4 male rats. Cells were expanded extensively in medium supplemented with FGF-2, and were shown to maintain their osteoblastic phenotype as characterised by alkaline phosphatase (ALP) staining, osteopontin (OPN), osteocalcin (OCN) expression and osteoblast-associated gene expression in long term culture. Gene expression of cells was determined by quantitative RT-PCR. Differences in Hoxa gene expression as markers of positional identity were maintained for up to at least 10 passages, with calvarial cells remaining Hoxa-ve throughout. The transcription factors Msx2 and Irx5 were consistently more highly expressed in calvarial cells, whereas Tbx3 expression was elevated in femoral cells. Expression of the osteoblast-associated genes Bglap and Sppl were elevated in femoral cells, and also associated with increased osteopontin secretion and bone nodule formation. Runx2 was elevated in calvarial cells. Cells were also pre-labelled with fluorescent vital staining and co-cultured for 7 days prior to separating by fluorescence activated cell sorter to investigate the possibility of re-programming of Hoxa negative cells by direct contact with Hoxa +ve cells. However no evidence was seen of modulation of positional identity genes and phenotypes in these heterotypic cultures. In conclusion, the results demonstrate persistence of expression of positional identity gene markers and phenotypic differences between femoral and calvarial osteoblasts for prolonged periods in culture. These data suggest that these differences in regionally defined osteoblasts are inherently programmed in the cells as a result of their embryological position. The results may have considerable implications when considering the transplantation of autologous cells in tissue engineering.

This thesis comprises three phases with a combined aim which was to investigate the effect of position on the lumbar intervertebral disc (IVD). The effect of position on the lumbar IVD in asymptomatic subjects and subjects with discogenic low back pain (DLBP) was explored using positional Magnetic Resonance Imaging (pMRI). Convenience samples of 11 asymptomatic and 34 DLBP subjects were recruited to have sagittal and axial pMRI scans performed in sitting (Neutral, Flexed and Extended), standing and lying (Supine and Prone extension) positions. The sagittal plane migration of the nucleus pulposus (NP) of each lumbar IVD in each position was measured from the sagittal and axial pMRI scans. Within and between group inferential analysis was performed using nonparametric tests. Both the asymptomatic and DLBP subjects’ demonstrated that position had statistically significant effects on the sagittal plane NP migration. Both groups demonstrated significantly greater posterior sagittal plane NP migration in Neutral and Flexed sitting positions compared to the other positions. However, between group comparisons identified that the asymptomatic subjects also demonstrated significantly greater posterior sagittal plane NP migration than the DLBP subjects. This pattern was more common in the upper lumbar IVDs (L1/2 and L2/3) between positions and less common in the lower IVDs (L4/5 and L5/S1) between positions. New knowledge regarding the behaviour of the lumbar IVD emerged from this research. The differences detected between the asymptomatic and DLBP subjects suggest that some current theories regarding DLBP may be incorrect. The results also support imaging of DLBP subjects in sitting positions as opposed to current supine positions. Although the limitations of the study reduce generalisation of the results, the implications for clinical practice, imaging and suggestions for further research from this work are important to improve understanding and conservative management of DLBP.

Conjugated linoleic acid (CLA) is produced in the rumen by isomerization of linoleic acid and has been shown to be anticarcinogenic. The objective of this study was to determine the effects of feeding dietary CLA to pre-ruminant lambs or linoleic acid-rich safflower oil to weaned ruminating lambs. Fat content, CLA content and fatty acid (FA) composition of diaphragm, leg, rib, subcutaneous (SC) adipose and liver tissues were determined. The triacylglycerol (TAG) fractions of the total lipid (TL) from the diaphragm, rib and SC adipose tissues were analyzed for FA composition and FA positional distribution. The phospholipid (PL) fractions of the TL were analyzed for FA composition. The positional (PL) fractions of the TL were analyzed for FA composition. The positional distribution of FAs of liver PL was determined. Tissue fat content was affected by diet only in SC adipose tissue. Dietary supplementation with safflower oil increased the fat content of the subcutaneous adipose tissue whereas CLA supplementation resulted in decreased (P<0.05) fat content. Dietary supplementation with safflower oil increased (P<0.05) the CLA content of tissues by more than 200% whereas dietary supplementation with CLA did not affect CLA content. Positional distribution analysis indicated that CLA tends was localized to the sn-1/3 positions of TAG and the sn-2 position of the liver PL.

This research both develops new methods and expands upon existing methodologies in order to improve our understanding of voting patterns and party competition in Britain. The thesis comprises five sections, each of which relates to a particular research focus. The first and principal section describes the process of determining a new method for decomposing electoral bias for three-party competition under simple plurality rules of voting. The study of electoral bias is important for voting systems that requires periodic boundary reviews intended to equalise electorate and to remove malapportionment. These papers describe both the process for developing the threeparty bias method and later its application to UK general elections from 1983 onwards. The second section uses aggregate data gathered for the elections to the Greater London Authority in order to understand the patterns of electoral support across the capital, particularly support for minor parties. A considerable amount of research effort has been expended upon providing reliable models for electoral forecasting both in the UK and elsewhere. The third section includes a paper that develops a forecast model that utilises aggregate local election data to estimate national vote shares for the three main parties in the UK. A fourth section brings together a series of papers that are linked by the themes of voter behaviour, either in terms of geographical or ballot context. A study of voter turnout in a London borough describes the relationship between proximity to polling station and electoral turnout at different types of election. A 8 number of papers included in this section also detail the effects of candidate ballot order on electoral support. The fifth and final section groups together two papers that using individual-level survey data to describe the pattern of candidate recruitment for local elections in Britain and, specifically, the under-recruitment of both women and Black, Asian and other minority ethnic candidates.

La localisation des objets dans l'espace est l'une des fonctions centrales du système visuel. Lorsqu'un observateur ou une cible se déplace, le mouvement de l’œil ou de l'objet peut être pris en compte pour calculer la position de l'objet à n'importe quel moment. Il a été démontré à plusieurs reprises que le mouvement visuel peut fortement influencer la position perçue d'un objet. Par exemple, la position d'un stimulus contenant une texture en mouvement (De Valois & De Valois, 1991; Ramachandran & Anstis, 1990), celle d'un flash présenté sur (Cavanagh & Anstis, 2013) ou à côté (Whitney & Cavanagh, 2000) d'une texture en mouvement, et même les positions d'apparition et de disparition d'objets en mouvement (Fröhlich, 1923) sont perçus comme étant déplacées dans le sens du mouvement. Dans le cadre de cette thèse, nous avons exploré la relation entre ces changements de position provoqués par le mouvement et différentes formes d'attention visuelle: 1) l'attention spatiale temporaire, 2) l'attention globale et locale, 3) l'attention spatiale maintenue, et 4) l'attention centrée sur les objets. Dans une première série d'expériences, nous avons étudié dans quelle mesure et de quelle manière l'attention module l'effet Fröhlich (i.e la position perçue du début du mouvement est déplacée dans le sens du mouvement). Dans les expériences 1 et 2, nous avons mesuré l'effet Fröhlich dans différentes conditions de d'indiçage et nous avons montré que les indices non valides ou tardifs entraînent des déplacements perceptifs plus importants. Dans l'expérience 3, nous avons comparé les déplacements provoqués par le mouvement lorsque les sujets portaient leur attention sur un ensemble d'objets en mouvement qui formaient un groupe à quand ils portaient leur attention à un seul stimulus parmi cet ensemble. Les résultats montrent que l'effet Fröhlich n'était présent que lorsque l'attention sélectionne un seul objet et qu'il disparaît lorsque le stimulus est perçu dans un contexte de groupe (global). Ainsi, ces résultats suggèrent que l'attention sélective est à la fois à l'origine de et module l'effet Fröhlich. Ayant établi que les délais temporels de l'attention augmentent les changements de la position provoqués par le mouvement, l'étude suivante était conçue pour explorer si la distribution spatiale de l'attention a le même effet. Dans cette étude, nous avons utilisé le « flash grab » - un changement de position illusoire aperçu quand une cible est brièvement présentée brièvement sur un fond en mouvement au moment de l'inversion du sens du mouvement (Cavanagh & Anstis, 2013). Les résultats indiquent que les attributs de la cible orthogonaux au mouvement du fond étaient déplacés, alors que les attributs parallèles au mouvement restaient intacts. Ceci suggère que le mouvement interagit avec la position des attributs de l'objet (et que l'attention focale sélectionne) avant que ces attributs ne soient regroupés en un objet. Localizing objects in space is one of the central functions of the visual system. When an observer or a target is moving, the motion of the eye or the object can be taken into account to compute the current object locations. It has been shown many times that visual motion can strongly influence the perceived position of an object. For example, a stationary patch containing moving texture (De Valois & De Valois, 199; Ramachandran & Anstis, 1990), a flash presented on (Cavanagh & Anstis, 2013) or next to (Whitney & Cavanagh, 2000) a moving texture, and even the onset and offset positions of the moving targets (Fröhlich, 1923) are perceived as shifted in the direction of motion. In this thesis we explore the relationship between these motion-induced position shifts and visual attention in the following forms: 1) transient spatial attention, 2) global and local attention, 3) sustained spatial attention, and 4) object-based attention. In the first series of experiments we looked at whether and how attention modulates the shift in localization of motion onset (Fröhlich effect). In Experiments 1 and 2 we measured Fröhlich effect under different cueing conditions and established that invalid or late cues produced larger perceptual shifts. In Experiment 3 we compare the motion-induced shifts when the subjects attended to a set of moving stimuli as a group and when they attended to an orientation singleton. We showed that the Fröhlich effect was only present when the target was individuated and disappeared when the stimulus was perceived globally. Thus, the Fröhlich effect appeared to be both produced and modulated by focal attention. Having established that temporal delays of attention increase motion-induced position shifts, the next study explored if spatial distribution of attention has a similar effect. In this study we used flash grab - an illusory position shift seen when a target is briefly flashed on top of a moving background that abruptly changes direction (Cavanagh & Anstis, 2013). Trials were presented in blocks and before each block a cue indicated a range of possible target locations. We found that the flash grab was reduced if the spatial distribution of targets was limited to a range of 90° or less. The final study asked whether motion shifts the perceived position of an object as a whole or if separate features of a single object are shifted independently. To test this we used the flash grab paradigm and briefly presented a shape on top of a moving background at the moment it changed direction. The results showed that the features of the target that were orthogonal to the background motion were shifted, whereas the features parallel to the motion were intact. This suggests that motion interacts with the position of the object's features (and focal attention selects them) before they are bound together into an object. In conclusion, we applied a variety of attentional manipulations to motion-induced position shifts, and examined the link between the strength of the illusion and the characteristics of attention used in a particular task. We found that 1) motion-induced position shifts require focused attention and the possibility to track an individual motion trajectory; 2) allowing attention to be allocated more efficiently in space reduces the illusion; and 3) motion-induced shifts operate on the feature-based and not object-based level

Data from a Coarse Acquisition (C/A) Global Positing System (GPS) map-grade receiver were evaluated to assess the accuracy of differentially corrected points. Many studies have focused on the accuracy of GPS units under ideal data collection conditions. Ideal conditions allow the collection of data with four satellites (3D mode), yet field data conditions are often less than ideal. Four satellites may not always be in view because of mountainous topography, heavy forest cover, or other obstructions which block satellite signals from the receiver. This study examines GPS accuracy when four satellites are not available, instead collecting data with only three satellites (2D mode).

3D GPS points compute four unknowns: x, y , z, and clock error. In comparison, 2D GPS points are less accurate as only three unknowns are calculated: x, y, and clock error. Elevation (or z) is not computed for 2D points, causing increased error in the horizontal (x, y) measurement. The effect of elevation was evaluated on 234 2D GPS data points. These points were collected and corrected at elevation intervals of true elevation, +-25 meters, +- 50 meters, and +-75 meters. These 2D points were then compared to surveyed points to measure the effect vertical error has on horizontal accuracy. In general, the more error in the vertical estimate during correction, the greater the horizontal error.

This thesis includes a study of a touch-free sensor systems and different principles for measuring a magnetic field. An in-depth study was conducted to describe two important principles in these contexts, i.e. Magnetoresistance principle and the Hall Effect principle. A market evaluation of different sensor chips developed on these principles was conducted. Furthermore, the study should be linked to existing principles to determine the position of a rotating axis. Function tests show that the performance requirement for each component was met. The results based on verification tests show that it is important to have a better mechanical connection between drive and device. Temperature tests show that the system meets the project specification at room temperature, but large angular deviations occur in temperature change, especially when driving with short steps. This thesis can be continued by reprogramming the source code and developing new software that can control more parameters and make it more accurate at positioning. Another recommendation would be to investigate and compare other sensor chips because there are several sensors that can apply to the system. Further improvements can be made by performing more tests on the system.
Examensarbetet innefattar en studie om beröringsfritt givarsystem och olika principer för att mäta ett magnetfält. En fördjupad litteraturstudie utfördes för att förklara två viktiga principer i dessa sammanhang d.v.s. magnetoresistiva principen och hallgivarprincipen. En marknadsutvärdering av olika sensor-chip som utvecklats enligt dessa principer genomfördes. Vidare ska studien kopplas till kända principer för att bestämma positionen för en roterande axel. Funktionstester visar att prestandakravet för varje komponent uppfylldes. Resultaten baserade på verifieringstester visar att det är viktigt att ha en bättre mekanisk koppling mellan drivdon och enhet. Temperaturtester visar att systemet uppfyller projektets kravspecifikation vid rumstemperaturen men det uppkommer stora vinkelavvikelser vid temperaturförändring framför allt vid körning med korta steg. Detta arbete kan fortsättas genom omprogrammering av källkoden och utveckling av ny programvara som kan styra fler parametrar och göra det mer exakt vid positionsbestämning. En annan rekommendation skulle vara att undersöka och jämföra andra sensorchip eftersom det finns flera sensorer som kan tillämpa i systemet. Ytterligare förbättringar kan göras genom att utföra fler tester på systemet.

An extensive investigation into and charactaisation of factors influencing transgene expression following introduction of the transgoie into tobacco via Agrobacterium- mediated transformation was carried out. Characterisation of material supplied at the outset of this project revealed that this material was unacceptable for further analysis. It was thus deemed necessary to obtain large populations of transgenic tobacco heterogenous for levels of transgene expression. Characterisation of these populations (CaMV-lecA and ssRubisco-lecA plants) showed that all plants fell into one of four segregation classes based on segregation of the kanamycin-resistance selectable marker. Results showed that the majority of regenerants contained multiple nptII-containing inserts, while the presence of one or two such inserts was also found, albeit at a much lower frequency. Segregation analysis based on detection of the lecA transgene agreed, in the majority of cases, with these results. However, in a few cases it was found that data obtained from both segregation analyses did not agree, with the presence of a single lecA-containing transgene being detected in plants shown to contain two copies of the nptII-contaning transgene. This result indicates the occurrence of T-DNA rearrangement either within the tobacco genome or during T-DNA transfer and integration. Southern blot analyses allowed a detailed characterisation of T-DNA structure, copy number and number of integration sites to be undertaken. Results from these analyses revealed a higher frequency of T-DNA rearrangement within plants containing multiple inserts. However, such rearrangements did not correlate with a significant reduction in levels of transgene expression since all detected rearrangements were found to occur at or towards the left hand border of the T-DNA, that border distant to the lecA transgene. Plants containing more than one T-DNA were also frequently found to contain these T-DNAs arranged as an inverted repeat at a single locus although no significant relationship between copy number and the presence of such structures was found. Correlating transgene expression levels, as determined by radioimmunoassay-based quantitation of lectin protein in tissues of transgenic plants, with T-DNA copy number, organisation and structure revealed no significant relationship. It is thus feasible to conclude that the major contributory factor influencing levels of transgene expression is the location of T-DNA integration within the plant genome. Subsequent work concerned with investigating the nature of those integration site-specific factors i.e. 'position effect' indicated a possible role for methylation-induced modulation of gene expression. Results presented in this thesis provide an insight into the fate of transgenes following introduction into the plant genome and clearly demonstrate the importance of further exploring the molecular mechanisms underlying transgene expression variability.

Les transistors organiques à effet de champ (OFETs) dans lesquels le transport des charges se fait à travers un film mince de molécules organiques représentent une transformation de la technologie des transistors à effet de champ au regard de la technologie au silicium. Ils permettent notamment d’envisager le développement d’une électronique flexible à bas coût. Ce travail porte sur la synthèse, l’étude et l’utilisation en tant que couche active dans des OFETs de type n de couples d’isomères para- et méta-indacénodithiophènes (para- et méta-IDT) appauvris en électrons inédits. Une introduction aux OFETs de type n est tout d’abord présentée. Elle est suivie par la présentation de la synthèse des dérivés IDT et de l’analyse comparée de leurs propriétés physico-chimiques. La fabrication des OFETs, leur caractérisation et l’optimisation de leur architecture est enfin décrite, leurs performances montrant l’intérêt des IDTs pour les OFETs de type n. Différentes fonctionnalisations menant à des IDTs d’architecture 3π-2spiro sont également synthétisées afin d’étudier les propriétés intrinsèques de ces dérivés π-conjugués et d’envisager leur incorporation comme matrice hôte dans des diodes électrophosphorescentes organiques
Organic Field Effect Transistors (OFETs), in which the charge transport is carried through a thin film made of organic molecules represent a transformation of the FET technology regarding that based on Silicon. They offer in particular the possibility to manufacture low cost flexible electronics. This work is focused on the synthesis, the study and the use as active layer in n-type OFETs of novel, electron poor, couples of para- and meta-indacenodithiophenes isomers (para- and meta-IDT). First of all, an introduction to the field of n-type OFETs is presented, followed by the presentation of the synthesis of the IDT derivatives and the comparative analysis of their properties. Finally, the fabrication of the OFETs, their characterization and the optimization of their architecture is described. The performances recorded attest that these derivatives are of great interest for the n-type OFETs. Different 3π-2spiro IDT derivatives are also presented in order to study the IDTs intrinsic properties and to envisage their incorporation as host in phosphorescent organic light-emitting diodes

The purpose of the present study was to assess the pressure distribution responses to three hour seating bouts in a standard car seat condition compared to a Comfort Motion Technology (CMT) car seat condition. Twelve males participated in the study. Subjects came in for two separate sessions no more than one week apart. Pressure distribution on the backrest and seat pan was assessed in a three hour seating bout. Pressure variables associated with increased seating comfort were collected across six distinct 28 minute cycles and then averaged across these six cycles. The pressure variables were contact area, peak contact pressure, and force. The CMT car seat condition exhibited greater back contact area (p= .003) and less back peak pressure (p= .003) than the standard car seat condition. In addition, the CMT car seat condition had greater seat peak pressure (p< .001) and greater back force (p< .001) than the standard car seat condition. In conclusion, the CMT car seat condition is more comfortable than the standard car seat condition.

The purpose of this study was to investigate the relationship between strength and position control, with an emphasis on measurements at low, absolute levels of force, as well as to examine the effect of load on position control. 33 healthy and physically active male university students were subjected to maximum voluntary contraction (MVC) tests and a weighted positional knee extension task involving trials at force levels ranging from 0.5-20kg, with the goal of maintaining position. Among the principal findings was the fact that strength did not affect position variability at lower levels of force, while at higher force levels, the stronger subjects exhibited lower levels of position variability. Thus, it was concluded that general strength training up to a certain level is non-detrimental to force control at low levels of resistance, and that increases in strength may contribute positively to force control at moderate force levels. In accordance with previous research, variability increased as a function of load, while variability relative to load decreased with added weight. At the heaviest weights, strength became the main determinant of performance, while its importance at lower force levels was negligible. Mean and maximum frequency of velocity and acceleration exhibited a U-shaped relationship with weight, which at the lower end was attributed in part to an inability to perceive and correct for the small force oscillations, as well as low inertia.

Modern solid state chemistry is inconceivable without theoretical treatment of solids thanks to the availability of efficient and accurate computational methods. Being developed mainly by physicist's community and deeply rooted in the formalism of reciprocal space, they often lack connections to familiar chemical concepts, indispensable for the chemical understanding of matter. Quantum chemical topology approach is a powerful theory able to efficiently recover chemical entities from the abstract description of a system given by its density matrices. It can be used to partition any many-electron system into the atoms, using the topology of electron density or for instance into atomic shells, using the topology of ELI-D field. Various characteristics of interactions between these chemical building blocks can be obtained applying bonding indicators, e.g. from the analysis of domain-averaged properties. Quantum chemical topology methods have been extended in the current work for the applications on the diversity of theoretical methods widely used for the description of solids nowadays – from the mean field Kohn-Sham density functional theory to the reduced one-electron density matrices functional theory or from the scalar-relativistic methods to the many-component formalisms employing spinor wavefunctions. It has been shown, that they provide chemically meaningful description of the bonding which is universally applicable to any class of extended systems, be it ionic insulator, covalent solid or metal. It has been shown, that the relativistic effects on the chemical bonding can be easily revealed using extensions of bonding indicators developed in the current work. Classical chemical concepts like Zintl-Klemm concept can be easily recovered with these descriptions. Intimate connection between the class of the material and the degree of chemical bonding delocalization has been also established. All these methods have been successfully applied to the various classes of solids and delivered novel insights on their crystal structure, properties, solid state transitions and reactivity.

Highly maneuverable ocean going vehicles require quick response, control, and guidance to maintain accurate track keeping characteristics. Ocean vehicles, however, may experience significant lags in their inertial positional information that limit their reaction times. This thesis investigates the effects of these lags on guidance and control. The relationship of critical visibility versus the controller time constant and its effect on the stability of the guidance/control scheme is analyzed. Results are presented based on time domain and frequency response techniques using a dynamic model of the Naval Postgraduate School Autonomous Underwater Vehicle II (NPS AUV II), for which a complete set of hydrodynamic coefficients and geometric properties is available.

As is generally known, pressure-resistant solenoids for valves are used in hydraulics and pneumatics. These solenoids, for example, consist of a so-called pole tube with an integrated armature and a separate solenoid coil. Suitable displacement measuring systems are used in order to measure the armature´s as well as valve spool´s position constnatly; these ar directly mounted to the solenoid. Exeamples here are Hall Effect sensors or LVDT transducers. In case of lesser requirements, it is also possible to measure the valve spool´s position directly and in simplified manner. Here, partially standard proximity sensors are employed, which use the valve spool directly as measuring object. In these cases, however, only discrete position measurement is possible. A constant displacement measurement is not necessary. By means of the sensorless position-control developed by MSM, the armature´s position can be executed directly by analyzing the solenoid coil, partly replacing the usual sensors, under adapted basic conditions.

The mosiac, cell-autonomous expression of genes resulting from chromosomal rearrangement and relocation next to broken heterochromatin is termed position effect variegation (PEV). Since the gene is inactivated due to chromatin changes, this system allows the genetic study of chromatin structure and function using mutations which rescue the mosaic phenotype. These mutations called suppressors of variegation, Su(var)s, must influence chromatin structure. The genetic characterization of several groups of Su(var)s has been undertaken in this study using Drosophila melanogaster. Variegation of the light gene, located in heterochromatin, is enhanced by several Su(var) mutations on chromosome two. This opposite effect suggests that products of these Su(var)s are essential for functioning heterochromatin and deleterious for euchromatic environments. Other Su(var)s have slight or no effects on the same variegating rearrangements, demonstrating functional differences, among the Su(var)s tested. A group of Su(var)s located within 4 map units near the centromere of chromosome three was characterized using deficiency mapping, new compound autosome formation and inter se complementation based on newly established homozygous phenotypes. Two Su(var)s mapped to 87B on 3R, while one Su(var) maps to 3L according to compound mapping. Inter se complementation, in combination with mapping data, suggests that four seperate loci make up this group of Su(var)s. Eight of nine Su(var)s are extremely sensitive to heterochromatic deletions as shown by their responses to loss of 2R heterochromatin, as well as the Y chromosome. In contrast, Su(var)A130 is insensitive to both forms of heterochromatic deficiencies. Su(var)s show complicated reactions to maternal verses paternal source effects. Six of nine Su(var)s show a female-specific temperature sensitive maternal effect. Some maternal and paternal effects are observed at 22 C. Su(var)A57 is maternal semi-lethal and suppressed at 29 C. This characterization has better defined these mutants, making them ammenable to molecular study.

The purpose of this study was to analyze specific biomechanical and physiological variables in disabled swimmers performing a functional backcrawl. The effect of selectively placed flotation devices on the body position, exercise heart rate, and perceived exertion of the swimmer was compared to swimming with no flotation. The participants in the study included eight able-bodied males 21.5 (+2.51) yrs. and four disabled males 29.25 (+2.22) yrs. Each subject performed the backcrawl for 3 consecutive lengths of a 25 meter pool under four separate trial conditions: free swimming, swimming with the hip flotation device, the knee flotation device, and the ankle flotation device. Immediately following each trial a post-exercise heart rate was taken and the subject was asked to rate perceived exertion. Full recovery was allowed after each trial. Trials were video taped and digitized to determine the mean angles at the neck, trunk, hip, and knee during one complete stroke cycle. The investigator recognizes that this generalization presents the possibility for a bias in the data and that further study would warrant the utilization of the different phases of the stroke for determination of body angles. The results of the study demonstrated that a significant difference did exist between the disabled and able-bodied hip angle during free swimming. Within the disabled group, no significant differences were evident when comparing the body angles during free swimming to the same angles while swimming with each of the flotation devices. Correlations between heart rate and ratings of perceived exertion for all subjects (n=12) were significant in each of the four trial conditions.

This thesis was aimed to evaluate if sentiment related to stocks expressed on the subforum “Wallstreetbets” also reflects the traded volume in the stock market. For this purpose, a collection of comment data from posts filtered under the “Hot” section was issued between the 6th of April 2021 and the 20th of April 2021 on daily basis at 22.00 (GMT+2). The comments were preprocessed to filter out noise, and thereafter comments that contained mentions of stocks were analyzed using VADER, an algorithm for grading sentiment. In total sentiment regarding 13 different stocks were fitted into a mixed effect model with random slopes and intercepts. The results showed a positive correlation between sentiment expressed and the traded volume. This indicates that by studying the forum we can better understand how people invested in stocks make investment decisions, which potentially could lead to a competitive advantage over time.

Position Effect Variegation (PEV) is the mosaic expression of a gene that has been moved out of its optimal environment and into a different area on the chromosome. Changing a gene’s environment may have profound effects on its eligibility for proper expression, which is a complicated process regulated by many factors. The PEV phenomenon is used as an assay to study gene expression as regulated by chromatin structure. In this study, the Drosophila melanogaster white gene was used as a reporter to study the various effects of CHD1, a chromatin regulating factor, on PEV gene expression. Inspired by preliminary data generated by the Armstrong Lab where overexpression of CHD1 resulted in suppression of gene silencing of the brown gene and loss of CHD1 resulted in enhancement of gene silencing, this study uses PEV as an assay to examine whether loss of function chd1 mutant alleles function dominantly to enhance silencing of the white gene when it is placed in a repressive chromatin environment. Surprisingly, I found that a chd1 loss of function mutant allele dominantly suppressed gene silencing (meaning I saw an increase in gene expression), suggesting that the CHD1 protein is normally required for effective silencing. The results demonstrated that CHD1 is a dominant modifier of PEV gene expression. CHD1 significantly modifies gene expression by suppressing silencing of the white gene inserted into pericentric heterochromatin on the second and fourth chromosomes and an insertion into the medial region of the fourth chromosome, while it shows no significant modification of the white gene inserted into telomeric heterochromatin of the fourth chromosome. Together, these intriguing results regarding varying gene expression at different chromosomal sites show that PEV is a dynamic phenomenon meriting further research and studying the effects of CHD1 as a modifier of PEV may be influential to understanding the mechanism and characteristics of gene expression.

This study investigated the effect of shoulder position on grip strength in 30 female students from Florida International University. A Jamar dynamometer was used to measure the grip strength in three testing positions (0,90 and 135 degrees of shoulder flexion with full elbow extension). The highest mean grip strength measurement was found at 135 degrees of shoulder flexion, followed by 0 degrees and then 90 degrees. An ANOVA indicated that there was a significant difference between at least two of the three positions. A Fisher's LSD post hoc test indicated that mean grip strength at 135 degrees of flexion was significantly higher than at 0 and 90 degrees of flexion.

A study was undertaken to identify the effect of head flexion/extension on singing voice quality. The amplitude of the fundamental frequency (F0), and the singing power ratio (SPR), an indirect measure of singer’s formant activity, were measured. F0 and SPR scores at four experimental head positions were compared with the subjects’ scores at their habitual positions. Three vowels and three pitch levels were tested. F0 amplitudes and low frequency partials in general were greater with more extended head positions, while SPR increased with neck flexion. No effect of pitch or vowel was found. Gains in SPR appear to be the result of damping low frequency partials rather than amplifying those in the singer’s formant region. Raising the amplitude of F0 is an important resonance tool for female voices in the high range, and may be of benefit to other voice types in resonance, loudness, and laryngeal function.

The purpose of this study was to investigate the influence an increase in strand strength and the effect the as-cast vertical location had on transfer length, development length, and flexural strength and to resolve the discrepancies regarding the definition of the top-bar/strand effect. Two types of test specimens were fabricated and tested investigating each respective item. The increase in strand strength was found to influence transfer length, development length, and flexural strength, while the as-cast vertical location was only found to influence transfer length, and in turn development length. Contrary to the historical definition, the top-bar/strand effect was found to be more dependent on the amount of concrete cast above the strand than the amount below it, with transfer lengths showing a steady increase with a decrease in the amount of concrete cast above the strand. As a result of the findings of this study, a new transfer length equation was proposed and a previously proposed flexural bond length equation was recommended for use in lieu of the current code provisions. The current equations for flexural strength were found to give adequate estimates for flexural strength, although a decrease in ductility was noted.

Notre laboratoire s’intéresse depuis de nombreuses années à l’implication des télomères en pathologie humaine. Les télomères sont des structures nucléoprotéiques présentes aux extrémités des chromosomes. Ils jouent un rôle important dans le bon fonctionnement et la durée de vie des cellules, et ils nécessitent, de ce fait, une régulation et une protection stricte. De plus, dans différents organismes, les télomères sont associés à la répression des gènes proximaux. Ce phénomène épigénétique appelé effet de position télomérique (TPE) implique la propagation d’une structure chromatinienne répressive. Très étudié chez S. Cerevisiae, cet effet a aussi été démontré chez l’homme et dépend de protéines télomériques et de remodelages chromatiniens. Ainsi, le traitement des cellules humaines par des agents modifiant l’architecture chromatinienne tels que l’inhibiteur des histones déacetylases, la Trichostatine A, permet de lever le TPE et d’induire l’expression d’un gène en position subtélomérique. En revanche, la 5-aza-2’-deoxycytidine, un inhibiteur de la méthylation de l’ADN, n’abroge pas ce type de silencing suggérant l’implication de modifications spécifiques de ces régions dont l’organisation nucléosomale est particulière. Afin d’identifier de nouveaux composant chimiques capables d’inhiber les effets de position télomériques dans les cellules humaines, nous avons effectué le criblage d’une banque de molécules bioactives «Prestwick Chemical Library » sur la plateforme du Centre pour les Molécules Bioactives (CMBA) au CEA de Grenoble. Les criblages primaires et secondaires de cette chimiothèque ont permis à nos collaborateurs de nous livrer une liste de 15 touches confirmées, inhibitrices de la répression transcriptionnelle dépendante des télomères et observée expérimentalement grâce au gène rapporteur de la luciférase. Après avoir testé l’ensemble de ces molécules dans notre laboratoire, deux polyphénols (Acacetine et Chrysine) étaient capables de lever spécifiquement le TPE sans modifier l’expression du gène rapporteur placé hors contrôle télomérique. Nous nous sommes dès lors demandé à quel niveau ces deux molécules agissent sur le TPE (raccourcissement ou prolongement des tailles des télomères, perte ou gain de protéines associées aux télomères et/ou création ou pas d’un dysfonctionnement télomérique). L’ensemble de ces analyses a montré que le traitement des cellules humaines avec ces deux molécules est accompagné par un raccourcissement de la taille des télomères, un dysfonctionnement télomérique et la création de fusion entre chromatides sœurs. La découverte de ces molécules ouvre de nouvelles perspectives sur le traitement des maladies associées à des mécanismes télomériques, le cancer mais aussi des maladies rares comme la dystrophie facio-scapulo-Humérale, dont nous avons parallèlement étudié les mécanismes moléculaires
Telomeres are protective structures present at the ends of linear chromosomes and consist of simple repeated DNA sequences and specialized proteins. They are essential for the stable maintenance of eukaryotic chromosomes and they can regulate the lifespan of cells. In addition to their essential functions, telomeres are, in diverse organisms, specialized sites with regard to gene expression. Indeed, the transcription of genes located next to telomeres is repressed: this phenomenon was termed telomere position effect (TPE). As observed in the yeast, S. Cerevisiae -where TPE is best characterized- TPE in human cells depends of both telomere length and architecture. The treatment of cells with Trichostatin A, an inhibitor of class I and II histone deacetylases antagonizes TPE while treating the cells with 5-aza-2’-deoxycytidin, a demethylating agent, has no apparent effect on telomeric repression. Overall, position effects at human chromosome ends are dependent on a specific higher-order organization of the telomeric chromatin. In order to identify new molecules able to modulate TPE in human cells, we carried out the screening of the «Prestwick Chemical Library », a bank of bioactive molecules, on the CMBA-CEA platform in Grenoble. A total of 15 molecules were identified as anti-TPE factors after primary and secondary screening based on the increase in fluorescence measurement. After testing all of these molecules in our laboratory, two polyphenols, Acacetin and Chrysin, were retained as potent inhibitors of TPE and further characterized as drugs targeting telomeres specifically. We then asked whether TPE reversal was dependent on telomere length. The incubation in the presence of Acacetin and Chrysin affects telomere length suggesting that increased expression of the subtelomeric reporters is associated with a shortening of telomeres. To check whether these molecules revert telomeric silencing, by affecting telomere integrity, we analyzed chromosome stability in cells treated with Acacetin or Chrysin. The two molecules exhibit a rapid increase in the frequency of telomere induced DNA damage foci, suggesting that these drugs trigger telomeric dysfunction. Drug treatment also significantly increases the frequency of telomeric abnormalities observed in metaphase with a significant increase of end-to-end fusions between sisters chromatid. The discovery of these new compounds targeting specifically chromosome ends opens new avenue for the targeted treatment of diseases linked to telomeric pathways such as cancer but also rare diseases such as Facio-Scapulo-Humeral dystrophy for which we investigated the molecular mechanisms

In this modern period, societal and religious groups are strongly divided regarding the acceptability of abortion. Despite so many attempts by various groups to find a middle ground, the debate on abortion still remains largely polarized, at its most dramatic point with the extreme conservatives claiming abortion to be the moral equivalent of murder and the extreme liberals see it as devoid of moral import. And this polarization is due to the legal battle that continues to shadow moral discussions. An acceptance of an ethical nuance will here play as a concession on the deeply contested question of whether abortion should be a legally protected option for a woman, and to an extent blame for the continued crudeness which can be laid at the doorstep of a moral theory itself. Apparently, the ethical literature on abortion has focused almost exclusively on the tiniest moral assessment on whether and when abortion is morally permissible. This question is a crucial one indeed and its answer is desperately sought in this thesis by exposing the position of the Catholic Church.

Various body positions have been used in the scientific literature when performing the isometric midthigh pull resulting in divergent results. We evaluated force production in the isometric midthigh pull in bent (125° knee and 125° hip angles) and upright (125° knee, 145° hip angle) positions in subjects with (>6 months) and without (<6 months) substantial experience using weightlifting derivatives. A mixed-design ANOVA was used to evaluate the effect of pull position and weightlifting experience on peak force, force at 50, 90, 200, and 250 ms. There were statistically significant main effects for weightlifting experience and pull position for all variables tested, and statistically significant interaction effects for peak force, allometrically scaled peak force, force at 200 ms, and force at 250 ms. Calculated effect sizes were small to large for all variables in subjects with weightlifting experience, and were small to moderate between positions for all variables in subjects without weightlifting experience. A central finding of the study is that the upright body position (125° knee and 145° hip) should be used given that forces generated are highest in that position. Actual joint angles during maximum effort pulling should be measured to ensure body position is close to the position intended.

Transgenic plant technology has greatly facilitated the study of plant gene structure and function. It has, however, become apparent that the position of integration into the genome can influence transgene expression both quantitatively and qualitatively. One aim of the work presented in this thesis was to investigate the effects of a chicken B-globin gene matrix attachment region to insulate against these position effects in transgenic tobacco (Nicotiana tabacum). It was found that, while this sequence mediates position-independent transgene expression in mice, it does not do so in transgenic tobacco. Concurrently, a second aim of this thesis was to identify plant genes that are expressed either constitutively or in the majority of cell types. Such gene loci may contain native plant DNA elements that could confer position-independent transgene expression and would, furthermore, provide additional promoters for genetic engineering. The approach was to introduce a promoterless gusA transgene, situated adjacent to the left border repeat of a T-DNA, to generate plant gene-reporter gene fusions in vivo following transformation of potato (Solanum tuberosum). Expression of the transgene is only expected if the gene has become integrated downstream from an active endogenous plant promoter, thereby tagging it and facilitating its subsequent isolation. A transgenic line was identified exhibiting transgene expression in leaf, stem, root and tuber tissue. Isolation of genomic flanking DNA by inverse PCR identified the site of T-DNA integration as being within a locus containing patatin genes, and analysis of transgene expression reconciled the expression level and pattern to that expected from a class I patatin gene promoter.

In this thesis, we utilized 1,3,5-tri(thiophen-2-yl)benzene (TTB) core, which is reported previously, as the core moiety of C3-symmetry compounds. By modifying amide group onto thiophene group, we expect the non-covalent interactions of amide group such as hydrogen bond interactions and Van der Waals force would induce molecules to assemble into nanostructure. We use scanning electron microscopic and tunneling electron microscopic to investigate the nanostructure. We have modified different alkyl amide moiety onto C3 position of thiophene and obtained two compounds, TTB-3EH and TTB-3DA. In organic solvents such as N, N-dimethylformamide and acetonitrile, TTB-3EH forms fibril and with a concentration of 5 wt.% TTB-3EH results in the formation of gel. At the same time, TTB-3DA forms nanospheres in organic solvents that mentioned above. Besides, we exchange the linking of the alkyl amide group onto C5 position of thiophene and obtain two compounds, TTB-5EH and TTB-5DA, which form nanospheres in the mixture of acetonitrile and hexane solvents.

Leader-member exchange (LMX) have been found as significant and positive impacts in organizational leadership studies. Thus, it’s important to conduct further study of the antecedents of LMX. Based on the fact that past research has less concerned about the subordinates’ intentions in dealing with interpersonal emotions, this study used subordinates’ attachment styles as the antecedent of LMX, in which the theory of mind was used to explain the interactions between leaders and their members. First, the questionnaires of pre-test were collected and conducted with the item analysis, exploratory factor analysis, reliability analysis, and validity analysis for the development of the official scale. Official data was collected in three technological universities located in Mid-Taiwan. Samples are students engaging in full-time jobs and study in the Department of Continuing Education of these three universities. 227 valid questionnaires were collected. The confirmatory factor analysis, reliability analysis, and validity analysis were conducted to see the goodness-of-fit between the collected data and the measurement model. Furthermore, the test of structural model was processed to testify the relationships between the exogenous latent variables and the endogenous latent variables. Finally, using SEM (structural equation modeling, LISREL8.80) to testify the four proposed hypotheses. The results of this study indicated that: (1) the different attachment styles of subordinates had impacts upon the development of LMX. (2) The subordinates with different attachment styles perceived different positional resources. (3) The subordinates’ perceptions on positional resources would inference the development of LMX. (4) Subordinates’ attachment styles had partial direct inference on LMX, and partial indirect influence on LMX through perceived positional resources.
