President Xi Jinping carried out a military reorganization carried out in 2015. The reorganization included forming the PLA Rocket Force as a new dimension tasked with handling missile weapons, both conventional and nuclear. This article uses offensive-defensive theory in analyzing the formation of PLARF as an effort to secure national interests. Furthermore, this article will explain why China formed PLARF and prefers to develop these missile weapons through offensive-defensive theory variables. Thus it can be said that the factor of weaponry technology plays a dominant role in the formation of PLARF. Keyword : PLARF, China, Offense-Defense Abstrak Presiden Xi Jinping melakukan reorganisasi militer pada tahun 2015. Reorganisasi tersebut diantaranya mengubah PLA Second Altilery Corps (PLASAC) menjadi PLA Rocket Force (PLARF) sebagai matra baru yang sejajar dengan matra liannya serta memiliki tugas untuk menangani persenjataan misil, konvensional maupun nuklir. PLARF dilengkapi persenjataan rudal dengan berbagai jelajah. Pengembangan teknologi persenjataan dilakukan untuk meningkatkan kemampuan PLARF. Secara geografis, PLARF diperlukan untuk melindungi wilayah territorial Tiongkok. Presiden Xi Jinping menggambarkan PLARF sebagai inti kekuatan deterrence strategis Tiongkok, yang diperlihatkan melalui parade militer secara besar. PLARF juga disiapkan untuk melakukan operasi gabungan, serta adanya kontrol CMC terhadap PLARF memberikan kewenangan yang unik .Artikel ini menggunakan teori ofensif-defensif dalam menganalisis reorganisasi militer yang dilakukan Tiongkok. Selanjutnya, artikel ini akan menjelaskan mengapa Tiongkok membentuk PLARF serta lebih memilih mengembangkan persenjataan misil tersebut melalui variabel teori ofensif-defensif. Background: Polyalthia longifolia which originates from India is rich with various useful phytochemicals which are valuable for human health. Accordingly, the current study was conducted to evaluate the combinational antimicrobial activity of P. longifolia Ethyl Acetate Fraction (PLEAF) with ampicillin, antioxidant and cytotoxicity activities. Methods: The evaluation of the synergistic activity of PLEAF fraction and ampicillin against MRSA local isolate was conducted with various antimicrobial assays. Results: The Minimum Inhibitory Concentration (MIC) values of PLEAF fraction (62.5 μg/mL) and ampicillin (5000 μg/mL) were found to decrease to 15.63 μg/mL for PLEAF and 2500 μg/mL for ampicillin respectively in the Fractional Inhibitory Concentration (FIC) assay against the MRSA bacteria. The 2,2-diphenyl1-picrylhydrazyl (DPPH) and nitric oxide free radical scavenging activities showed that PLEAF fraction possessed high antioxidant activity and the combinational of PLEAF fraction and ampicillin exhibited moderate antioxidant activity. The total phenolic content (TPC) of PLEAF was 168.22 ± 0.00407 μg GAE/g of PLEAF fraction. Discussion: henolic compounds might be responsible for the observed antioxidant and antimicrobial activity of PLEAF fraction. In addition, in 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) cytotoxicity test against Vero cells the PLEAF fraction was proven to be non-toxic (98.14% of cell viability) and the combination of PLEAF fraction and ampicillin treatment against the Vero cells showed an improved cell viability (52.44%) as compared with ampicillin alone in the treated group. Conclusions: The PLEAF fraction works well in combination with ampicillin to kill the MRSA local resistance strain. PLEAF fraction also showed favourable antioxidant activity and improved Vero cell viability in the presence of ampicillin which is an important attribute of PLEAF fraction to be used in the future combinational therapy.

AbstractPolylactic acid fibrils (PLAf) were employed as a fiber component in papermaking. The addition of 5 wt % of PLAf to bleached kraft birch pulp increased the tensile index of the resulting 100 g/m2 paper sheets by 20 % in comparison to sheets produced without PLAf. By heat-treating the paper sheets containing 5 wt % PLAf, a 35 % higher tensile index in comparison to sheets produced without PLAf was achieved. SEM imaging showed that the heat-treatment caused the PLAf to melt, which formed a film on the fiber web. The PLAf was ultrasonicated in an attempt to make its surface more hydrophilic and anionic and thus more compatible with cellulose. Chemical additives (cationic polyacrylamide, polyethylene imine and polyethylene glycol) were added to the PLAf/cellulose pulp mixture in order to increase the binding between the ultrasonicated PLAf and cellulose. Ultrasonication caused the PLAf length to decrease and the PLAf surface charge changed by 36 %, indicating that the PLAf became significantly more anionic. Neither ultrasonication of PLAf nor the chemical additives improved the paper sheets’ stretchability. Polyethyleneimine as an additive in an amount of 1 % increased the tensile index of heat-treated sheets made with 5 wt % of PLAf by 19 %.

Background: Polyalthia longifolia is a popular medicinal plant and has been widely used as a traditional remedy for centuries in curing of various ailments. The purpose of this study was conducted to determine the in situ antimicrobial synergistic effects between Polyalthia longifolia leaf ethyl acetate fraction (PLEAF) and ampicillin against MRSA local isolate by using modern microscopy technique. Methods: Hence, the evaluation of the synergistic activity of PLEAF and ampicillin against MRSA local isolate was conducted with scanning electron microscopy (SEM). Results: The combinational effect of PLEAF fraction and ampicillin exhibited significant antibacterial activity against MRSA. Bacterial cells observations showed invagination, impaired cell division, extensive wrinkles, cell shrinkage, the appearance of a rougher cell with fibrous matrix and clustered cells which confirmed the synergistic effect of PLEAF and ampicillin against MRSA local isolate by SEM. Conclusion: Conclusively, the in situ SEM observation proved the synergistic antimicrobial activity between PLEAF fraction and ampicillin to destroy the MRSA resistance bacteria which is an important aspect of PLEAF fraction to be used in the future combinational therapy.

Legionella pneumophila encodes an extracellular secreted phospholipase A named PlaA that is translocated by the type II secretion system. It plays an essential role in maintaining the integrity of Legionella-containing vacuoles in L. pneumophila pathogenesis. Here, it is shown that PlaA has a main lysophospholipase activity to hydrolyze fatty-acyl groups in lysophospholipids. Although it has a very low phospholipase A activity to catalyze the hydrolysis of fatty-acyl groups in phospholipids, PlaA can bind phospholipids such as 1,2-dipalmitoylphosphatidylcholine with a dissociation constant of 11.1 µM. Sequence-alignment analysis combined with activity assays revealed that PlaA contains a distinct substrate-binding site among the known structures of the phospholipase A family, implying that PlaA may present a novel mechanism for substrate recognition. Native PlaA and its selenomethionine (SeMet)-substituted form were purified and crystallized by vapour diffusion in hanging drops at 296 K. Diffraction data were collected to a resolution of 2.0 Å for native PlaA protein and to a resolution of 2.7 Å for SeMet-substituted PlaA protein. The crystals of native PlaA belonged to the monoclinic space group P21, while the crystals of SeMet-substituted PlaA belonged to the primitive orthorhombic space group P212121. Initial phases for PlaA were obtained from SeMet SAD data sets.

3541 Background: So far, no biomarkers of response to anti-angiogenic drugs are available in colorectal cancer (CRC) treatment. Liquid biopsy tracks dynamic mutational changes in CRC patients (pts). RAS mutant allele fraction in plasma (plMAF) is an independent prognostic marker in metastatic CRC (mCRC). We explored the predictive value of plMAF in RAS mutant pts treated in 1st line with chemotherapy +/- bevacizumab (bev). Methods: A multicentric prospective/retrospective analysis was conducted. We collected data from 226 mCRC pts and selected the subset not eligible for metastasis resection with basal plMAF sample evaluable for RAS mutant MAF quantification with digital PCR (BEAMing). Pts were stratified as high (≥ 5.8%) or low ( < 5.8%) plMAF. We investigated associations between clinicopathological variables and progression-free survival (PFS) stratified by plMAF RAS levels using Cox regression models and survival data were calculated by Kaplan-Meier method. Computational analysis of baseline CT scan data extracted 93 radiomics features of all the lesions per patient including 1) 1st class from density histogram distribution and texture analysis by 2) 2nd order and 3) higher order feature classes. The radiomic features distribution between pts with high and low pIMAF was assessed with Student’s t-test analysis. Results: From October 17 to May 19, 63 basal plasma samples were analysed with BEAMing. 42 pts (67.7%) were classified as high and 21 pts (32,3%) as low plMAF. In high plMAF subgroup, a statistically significant longer PFS favouring FOLFOX+bev was observed, compared to FOLFOX alone (10.7 vs 6.9 mts; HR: 0.30; p = 0.002). In low RAS plMAF subgroup, no differences in terms of PFS were observed in either arm (8.9 vs 8.7 mts; HR: 0.7; p = 0.6). Multivariate PFS model showed no association between RAS plMAF and clinicopathological variables, except for high RAS plMAF and treatment benefit with FOLFOX+bev. The CT-radiomics signature, that may translate tumor vascularization, differentiated patients with high vs low pIMAF (p = 0.002). 58 patients (92%) had similar radiomic score; 5 patients with high plMAF (8%) presented very heterogeneous radiomic score distribution. Conclusions: Tumor-borne RAS plMAFs may constitute a potential predictive biomarker of efficacy for anti-angiogenic drugs in mCRC. Next steps will include the identification of -histological, transcriptomic and radiomic- surrogate biomarkers of response that reflect tumor irrigational status.

ABSTRACT We previously showed that Legionella pneumophila secretes, via its type II secretion system, phospholipase A activities that are distinguished by their specificity for certain phospholipids. In this study, we identified and characterized plaA, a gene encoding a phospholipase A that cleaves fatty acids from lysophospholipids. The plaA gene encoded a 309-amino-acid protein (PlaA) which had homology to a group of lipolytic enzymes containing the catalytic signature GDSL. In Escherichia coli, the cloned gene conferred trypsin-resistant hydrolysis of lysophosphatidylcholine and lysophosphatidylglycerol. An L. pneumophila plaA mutant was generated by allelic exchange. Although the mutant grew normally in standard buffered yeast extract broth, its culture supernatants lost greater than 80% of their ability to release fatty acids from lysophosphatidylcholine and lysophosphatidylglycerol, implying that PlaA is the major secreted lysophospholipase A of L. pneumophila. The mutant's reduced lipolytic activity was confirmed by growth on egg yolk agar and thin layer chromatography and was complemented by reintroduction of an intact copy of plaA. Overexpression of plaA completely protected L. pneumophila from the toxic effects of lysophosphatidylcholine, suggesting a role for PlaA in bacterial detoxification of lysophospholipids. The plaA mutant grew like the wild type in U937 cell macrophages and Hartmannella vermiformis amoebae, indicating that PlaA is not essential for intracellular infection of L. pneumophila. In the course of characterizing plaA, we discovered that wild-type legionellae secrete a phospholipid cholesterol acyltransferase activity, highlighting the spectrum of lipolytic enzymes produced by L. pneumophila.

Background After incision keratinocytes in the epidermis become activated to produce a range of pain-related mediators. microRNA 203 (miR-203) is known to be involved in keratinocyte growth, differentiation, and skin inflammation. We hypothesized that one or more of these mediators might be under the control of miR-203. Methods The expression of miR-203 and its target gene, phospholipase A2 activating protein (PLAA), were examined after hind paw incision in mice. We investigated the local effect of intraplantar PLAA peptide injection in normal mice and the effects of a selective secretory phospholipase A2 inhibitor (HK064) on PLAA or incision-induced mechanical allodynia. Last, we investigated the role of substance P signaling in regulating miR-203 and PLAA expression in vitro and in vivo. Results Levels of miR-203 were strongly down-regulated in keratinocytes after incision. Informatics-based approaches identified PLAA as a likely candidate for regulation by miR-203. PLAA caused mechanical allodynia and conditioned place aversion but not thermal sensitization. HK064 reduced mechanical allodynia after incision and after intraplantar injection of PLAA. Using preprotachykinin gene knockout mice or with neurokinin-1 selective antagonist LY303870 treatment, we observed that substance P-mediated signaling was also required for miR-203 and PLAA regulation after incision. Finally, using the rat epidermal keratinocyte cell line, we observed that a miR-203 mimic molecule could block the substance P-induced increase in PLAA expression observed under control conditions. Conclusions miR-203 may regulate expression of the novel nociceptive mediator PLAA after incision. Furthermore, the regulation of miR-203 and PLAA levels is reliant upon intact substance P signaling.

Background. During minimally invasive surgery, efficient and nontoxic hemostats are important for difficult to access bleeding areas. Polylactic acid is an ecofriendly hemostatic agent and we aimed to evaluate the efficacy of a polylactic acid nonwoven fabric (PLAF) developed by Toray Industries, Inc, on liver hemostasis in a preclinical study. Materials and Methods. PLAF consists of both 1-µm diameter fibers and 100-µm diameter beaded fibers. Four rats were used, and 2 trough-shaped resections of the liver parenchyma were performed (n = 8 lobes). Immediately after the resection, PLAF (PLAF group: n = 4 lobes) or rayon gauze (Rayon group: n = 4 lobes) were applied on the resected plane and compressed manually. We compared the mean time to hemostasis and blood loss per lobe, as well as histological findings between the groups. Results. The PLAF group had a significantly shorter bleeding time ( P = .006), and showed lower blood loss compared with the Rayon group ( P = .076). Histopathological evaluation showed a large amount of beads on the liver surface in the PLAF group. Aggregated red blood cells evident by electron microscopy and von Willebrand factor immunofluorescence were seen surrounding the beads. The PLAF group showed significantly greater von Willebrand factor expression than the Rayon group ( P = .004). Discussion. This new PLAF showed superior outcomes thanks to its unique characteristic of forming beaded nanofibers, and it has the potential to be an efficient hemostat in minimally invasive surgery in the human body.

This review article explores how Ronelda S. Kamfer’s novel Kompoun (2021) deconstructs and diversifies the white patriarchal space of the plaas (farm) by reinscribing it with a highly situated ‘plaasfeminism’ emerging from the female characters in the novel. This critical reinscription through the lives of the McKinney women from the Overberg is necessary, but certainly not triumphant. For Nadia, the protagonist, the idyll of the plaas consists of her admiration of and longing for her maternal forebears and thus provides a source of strength and personhood, but the plaas is also quite literally the scene of a crime from which her family fails to protect her. Kompoun complicates mainstream notions of feminist resistance by charting the internal contradictions of female subjectivity and highlighting the vulnerable position of the McKinney children, who grow up in a community where both adult men and women pose a threat of emotional and physical abandonment and abuse. Yet, in times of need, Nadia manages to mobilise her personal image of the plaas’ beauty as motherly and the women who live there as tough as coping strategies that suspend her imprisonment in the harmful dynamics around her.

Synergizing integrin and cell-membrane heparan sulfate proteoglycan signaling on biomaterials through peptidic sequences is known to have beneficial effects in the attachment and behavior of osteoblasts; however, controlling the exact amount and ratio of peptides tethered on a surface is challenging. Here, we present a dual molecular-based biointerface combining integrin (RGD) and heparin (KRSR)-binding peptides in a chemically controlled fashion. To this end, a tailor-made synthetic platform (PLATF) was designed and synthesized by solid-phase methodologies. The PLATF and the control linear peptides (RGD or KRSR) were covalently bound to titanium via silanization. Physicochemical characterization by means of contact angle, Raman spectroscopy and XPS proved the successful and stable grafting of the molecules. The biological potential of the biointerfaces was measured with osteoblastic (Saos-2) cells both at short and long incubation periods. Biomolecule grafting (either the PLATF, RGD or KRSR) statistically improved (p < 0.05) cell attachment, spreading, proliferation and mineralization, compared to control titanium. Moreover, the molecular PLATF biointerface synergistically enhanced mineralization (p < 0.05) of Saos-2 cells compared to RGD or KRSR alone. These results indicate that dual-function coatings may serve to improve the bioactivity of medical implants by mimicking synergistic receptor binding.

Resumo Introdução Usualmente, as luxações sacroilíacas são tratadas com parafusos iliossacrais ou com placas anteriores à articulação sacroilíaca (ASI). Este estudo compara a rigidez e carga máxima suportada pelos dois tipos de fixações acima citados, utilizando pelves sintéticas. Método Dez pelves sintéticas foram divididas em dois grupos (n = 5). No grupo denominado PlaCF, a ASI foi fixada com duas placas anteriores. No grupo ParCF, a ASI foi fixada com dois parafusos iliossacrais no corpo da primeira vertebra sacral (S1). A rigidez e carga máxima suportada por cada montagem realizada, foi mensurada. A análise estatística foi realizada através do teste U de Mann-Whitney (p < 0.05 foi considerado estatisticamente significativo para todas as análises). Resultados A carga máxima suportada até a falha da fixação pelos grupos PlaCF e ParCF foram respectivamente 940 ± 75 N e 902 ± 56 N, não havendo diferença estatística entre eles. A rigidez obtida pelo grupo ParCF foi maior e com diferença estatística em relação ao grupo PlaCF (68.6 ± 11.1 N/mm e 50 ± 4.0 N/mm respectivamente). Conclusão Apesar da menor rigidez obtida no grupo PlaCF, as placas anteriores à ASI podem ser uma ótima opção no tratamento da luxação sacroilíaca quando os parafusos iliossacrais não puderem ser utilizados. Outros estudos são necessários para detectar possíveis diferenças entre os dois procedimentos do ponto vista cirúrgico e clínico.

Accurate dynamic modelling of the Tandem Wing Quadcopter Hybrid (TWQH) platform is becoming increasingly important, as strict requirements are progressively being imposed on the UAV (unmanned aerial vehicles) control system in the transition phase from vertical to level flight and vice versa. This work aims to present preliminary design performances in the steady and manoeuvring flight of the platform thrusted by only four rotors directed upward. The aerodynamic effects of tandem wings make a significant contribution to the evaluation of performance, controllability and manoeuvrability in hovering and low speed phase. More specifically, steady, level, symmetrical pull-up and coordinated turns approaches of the quad model are considered and the results obtained in the simulation study are presented and discussed.

This study investigated the involvement of RNA folding in the synthesis of a fusion protein with beta-galactosidase activity. The coding gap region of the Prevotella loescheii adhesin gene plaA was fused in-frame with the Escherichia coli lacZ gene on plasmid pSK105. N-Terminal sequencing of the expressed plaA-lacZ protein indicated that it resulted from translational initiation at a fortuitous ribosomal-binding site within the plaA sequence at nt 570. Specific mutations were introduced in the stem-loop region that precedes the gap sequence. Analysis of stem-loop mutants, together with the introduction of compensatory mutations that restored activity, supports a requirement for stem-loop formation within the plaA sequence preceding the translational initiation site. A mutation reducing the predicted size of the loop, but preserving the stem structure, inactivated fusion protein synthesis. A suppressor mutation predicted to restore the size of the loop restored efficient fusion protein synthesis. In addition, the sequence preceding the translational start site of the plaA-lacZ fusion has several similarities to sequences that function as translational enhancers in prokaryotes. These include a stem-loop structure, an A-U rich region preceding the initiation codon, and a region of homology to 16S rRNA.Key words: site-directed mutagenesis, stem-loop formation, fusion protein, translational initiation, translational enhancer.

The surface of polylactic acid fiber (PLAF) was treated with self - manufactured starch nanocrystals (SNCs) to improve its interfacial adhesion with the starch matrix. Determination of optimum dispersion conditions of SNCs dispersion by transmittance. The effects of direct oscillatory deposition, chemical grafting SNCs modification, and SNCs dispersion concentration on the surface treatment effect of PLAF and its interfacial bonding with starch matrix were investigated. The results showed that the SNCs were successfully introduced into the PLAF surface, moreover, the modifying effect of the chemical grafted SNCs was better than the direct oscillatory deposition, the distribution of SNCs particles was more uniform on the fiber surface, strong chemical bonding to the fiber surface, make up for the surface damage of the fibers caused by aminolysis. At the SNCs dispersion concentration of 10 g/L, the PLAF tensile strength could be maintained at 582 MPa, a large amount of starch was evenly attached to the surface after pulling out. It showed that a strong interface bond with the starch matrix, the highest IFSS value was 2.57 MPa and the increase was by about 63 %. Therefore, the introduction of SNCs for interfacial modification of PLAF is of great significance and deserves further exploration.

ABSTRACT Cdc48 (p97/VCP) is an AAA-ATPase molecular chaperone whose cellular functions are facilitated by its interaction with ubiquitin binding cofactors (e.g., Npl4-Ufd1 and Shp1). Several studies have shown that Saccharomyces cerevisiae Doa1 (Ufd3/Zzz4) and its mammalian homologue, PLAA, interact with Cdc48. However, the function of this interaction has not been determined, nor has a physiological link between these proteins been demonstrated. Herein, we demonstrate that Cdc48 interacts directly with the C-terminal PUL domain of Doa1. We find that Doa1 possesses a novel ubiquitin binding domain (we propose the name PFU domain, for PLAA family ubiquitin binding domain), which appears to be necessary for Doa1 function. Our data suggest that the PUL and PFU domains of Doa1 promote the formation of a Doa1-Cdc48-ubiquitin ternary complex, potentially allowing for the recruitment of ubiquitinated proteins to Cdc48. DOA1 and CDC48 mutations are epistatic, suggesting that their interaction is physiologically relevant. Lastly, we provide evidence of functional conservation within the PLAA family by showing that a human-yeast chimera binds to ubiquitin and complements doa1Δ phenotypes in yeast. Combined, our data suggest that Doa1 plays a physiological role as a ubiquitin binding cofactor of Cdc48 and that human PLAA may play an analogous role via its interaction with p97/VCP.

ABSTRACT The gene encoding a poly(dl-lactic acid) (PLA) depolymerase from Paenibacillus amylolyticus strain TB-13 was cloned and overexpressed in Escherichia coli. The purified recombinant PLA depolymerase, PlaA, exhibited degradation activities toward various biodegradable polyesters, such as poly(butylene succinate), poly(butylene succinate-co-adipate), poly(ethylene succinate), and poly(ε-caprolactone), as well as PLA. The monomeric lactic acid was detected as the degradation product of PLA. The substrate specificity toward triglycerides and p-nitrophenyl esters indicated that PlaA is a type of lipase. The gene encoded 201 amino acid residues, including the conserved pentapeptide Ala-His-Ser-Met-Gly, present in the lipases of mesophilic Bacillus species. The identity of the amino acid sequence of PlaA with Bacillus lipases was no more than 45 to 50%, and some of its properties were different from those of these lipases.

We analyzed clinical experience with percutaneous closure of instances of left atrial appendage with thrombus (LAAT) irresponsive to antithrombotic therapy in patients treated in three high-volume cardiology centers. Clinical and procedural data regarding consecutive patients who underwent percutaneous left atrial appendage closure (PLAAC) due to LAAT were retrospectively analyzed. The study population consisted of 17 patients (11 men; 68 ± 14 years; CHA2DS2VASC 4.7 ± 1.9; HASBLED 3 (0–5)) with LAAT confirmed by transesophageal echocardiography, and included 5 patients with mechanical heart valves. Most of the patients (94.1%) received anticoagulation therapy before PLAAC. All LAATs were located in distal portions of the appendage and occupied less than 30% of its volume. Occluding-device implantation was successful in 17 patients; in one, a residual leak was disclosed. Appropriate positioning of occluders required more than 1 attempt in 6 individuals (35.3%); in 3 others (17.6%), the subjects’ devices had contact with thrombi. No procedural complications were noted. Midterm follow-up (median: 10 months) revealed no procedure-related complications or clinically diagnosed thromboembolism. Transesophageal echocardiography (TEE) performed after six months revealed device-related thrombus in one patient. We concluded that LAAT irresponsive to antithrombotic therapy might be effectively treated with PLAAC, even in patients with mechanical-valve prostheses.

Abstract After a hint from Lang et al.'s (1997) defence cascade, researchers considered cognitive process experienced when encountering mental stress to be composed of four elements: (serially) first attention (Attent), second unpleasant affect (UnplAff) and sometimes pleasant affect (PlAff), and third cognitive coping (CogCop). The present study investigates the effect of each cognitive element on the provocation of the well-known vascular-dominant reaction pattern during mirror tracing: elevation of mean blood pressure mainly because of increases in total peripheral resistance. Twenty-four male students first underwent four computer-simulated mirror-tracing practices of 3 min each, then a 7 min adaptation followed by a 3 min baseline, and further four kinds of actual mirror tracing trials (Attent, UnplAff, PlAff, and CogCop) of 3 min each. Results on the cardiovascular measures indicated that every mirror-tracing trial indisputably provoked the vascular-dominant reaction pattern. An alpha-adrenergic vascular sympathetic activation was heightened. Self-report measures on the four cognitive elements suggested that heightened Attent seemed to contribute to provoking the reaction pattern. Although the UnplAff and PlAff trials had an active coping feature in a narrow sense, they could not provoke the cardiac-dominant reaction pattern. Differences in task difficulty among the mirror tracings could not explain the results. The implications of these results are discussed in order to better understand cardiovascular hemodynamics during mental stress.

Bacterial phospholipase A1 (PLA1) is used in various industrial fields because it can catalyze the hydrolysis, esterification, and transesterification of phospholipids to their functional derivatives. It also has a role in the degumming process of crude plant oils. However, bacterial expression of the foreign PLA1-encoding gene was generally hampered because intracellularly expressed PLA1 is inherently toxic and damages the phospholipid membrane. In this study, we report that secretion-based production of recombinant PlaA, a bacterial PLA1 gene, or co-expression of PlaS, an accessory gene, minimizes this harmful effect. We were able to achieve high-level PlaA production via secretion-based protein production. Here, TliD/TliE/TliF, an ABC transporter complex of Pseudomonas fluorescens SIK-W1, was used to secrete recombinant proteins to the extracellular medium. In order to control the protein expression with induction, a new strain of P. fluorescens, which had the lac operon repressor gene lacI, was constructed and named ZYAI strain. The bacteriotoxic PlaA protein was successfully produced in a bacterial host, with help from ABC transporter-mediated secretion, induction-controlled protein expression, and fermentation. The final protein product is capable of degumming oil efficiently, signifying its application potential.

Age-dependent changes in leaf photosynthetic characteristics (i.e. parameters of the light response curve (maximum photosynthetic rate (Pmax), quantum yield (Φ) and the convexity parameter (θ)), stomatal conductance (gs) and dark respiration rate (Rd)) of an exotic perennial legume, Medicago sativa L. (lucerne), and two potential pasture legumes native to Australia, Cullen australasicum (Schltdl.) J.W. Grime and Cullen pallidum A. Lee, grown in a glasshouse for 5 months at two phosphorus (P) levels (3 (P3) and 30 (P30) mg P kg–1 dry soil) were tested. Leaf appearance rate and leaf area were lower at P3 than at P30 in all species, with M. sativa being the most sensitive to P3. At any leaf age, photosynthetic characteristics did not differ between P treatments. However, Pmax and gs for all the species and Φ for Cullen species increased until full leaf expansion and then decreased. The convexity parameter, θ, did not change with leaf age, whereas Rd decreased. The estimates of leaf net photosynthetic rate (Pleaf) obtained through simulations at variable Pmax and Φ were lower during early and late leaf developmental stages and at lower light intensities than those obtained when Φ was assumed to be constant (e.g. for a horizontally placed leaf, during the 1500°C days developmental period, 3 and 19% reduction of Pleaf at light intensities of 1500 and 500 µmol m–2 s–1, respectively). Therefore, developmental changes in leaf photosynthetic characteristics should be considered when estimating and simulating Pleaf of these pasture species.

Die Khumalo-saak handel oor ’n bejaarde man (K) se aansoek om ’n spoliasiebevel teen L (die eienaar van ’n plaas waaroor K jare lank vir hom ’n pad na sy woning toegeëien het) wat eensklaps sy grond toegespan het en geweier het om die heksleutels aan K – ’n okkupeerder ingevolge artikel 1 van die Wet op die Uitbreiding van Sekerheid van Verblyfreg 62 van 1997, woonagtig op ’n buurplaas – te oorhandig ten einde hom in staat te stel om steeds die pad te gebruik. Ofskoon daar ’n ander pad na K se woning was, was dit uiters swak en slegs geskik vir gebruik deur 4×4 voertuie. Aangesien K net ’n ou bakkie besit het, het hy geweldig ongerief verduur as gevolg van L se optrede. L het aangevoer dat hy goeie redes gehad het vir sy optrede, onder andere bodembewaring en om wilddiewe hok te slaan, asook dat K boonop nie ingevolge ’n serwituut van via toegang oor sy plaas gehad het nie. Die grondeisehof het op grond van ’n bedenklike toepassing van artikel 22(2)(c) van die Wet op Herstel van Grondregte 22 van 1994 vir hom jurisdiksie toegeëien. Verder het die hof, strydig met die robuuste en spoedeisende aard van die mandament van spolie, die beswaar dat die applikant sy aansoek eers na twee jaar en agt maande geloods het, summier verwerp en daarnaas, ook strydig met bestaande beginsel en praktyk, in besonderhede op die meriete van K se saak ingegaan om tot die beslissing te geraak het dat daar ’n stilswyende precarium tussen K en L gesluit is, omdat L se werknemers wat sienderoë toegelaat het dat K die pad oor L se grond gebruik, as L se verteenwoordigers vir doeleindes van sluiting van die precarium-ooreenkoms bevind is. Ingevolge die normale reëls moes die prekaris, K, dus volgens die hof redelike kennis van die precarium dans, L, ontvang het voordat laasgenoemde sy plaas kon afsper. Die hof het in wese “geregtelik kennis geneem” van hierdie precarium en hom geensins gesteur aan die bestaande reëls van die kontraktereg en, in die besonder, die verteenwoordigingsreg nie. Veral oppervlakkig was die hof se behandeling van die vereiste van skending van quasi-besit vir die mandament van spolie in gevalle waar die applikant nie in daadwerklike beheer van ’n onroerende saak was nie. Die hof het hier in werklikheid deur middel van die toepassing van spoliasiebeginsels spesifieke nakoming van die precarium-kontrak gelas. Daar word betoog dat die hof uit sy pad gegaan het om K uit sy verknorsing te help, ofskoon daar in die lig van geldende regsreëls geen regverdiging vir sodanige regshulp was nie. Hoogstens het die feit dat L jarelank vir K toegelaat het om sy pad te gebruik op toestemming – ’n eensydige regshandeling wat te eniger tyd herroep kan word – neergekom en L was dus ten volle daarop geregtig om sy plaas af te sper. Daar kan verder geensins betoog word dat daar ’n grondwetlike verpligting op L gerus het om K se gebruik van die pad oor sy plaas te gedoog nie. Deur hierdie uitspraak het die hof ’n gevaarlike presedent geskep.

The present work aimed to search for a released peptide from proteolytic action on a silver barb fish muscular protein that confers health benefit through antioxidation activity. Changes in the physicochemical, microbiological, and protein characteristics of plaa-som samples during eight days of both spontaneous traditional fermentation (Batch 1; B1) and spontaneous fermentation with the addition of pineapple (Batch 2; B2) were determined. Results showed a correlation between an increase in the total acidity and bacterial counts with the length of fermentation duration, where the pH gradually decreased at the end of fermentation. Protein hydrolysis during fermentation was indicated by an increase in the amount of TCA-soluble peptide contents that peaked on day 5 (D5) in both batches (B1D5 and B2D5), which displayed their highest DPPH radical-scavenging inhibition of plaa-som protein hydrolysates (PSPHs). Twelve peptide fractions of the best PSPH were separated by ultrafiltration using molecular weight cut off (MWCO) at 3 and 10 kDa, and they were also purified by size exclusion chromatography. Results demonstrated that stronger peptides B2D5 - 3 kDa - F1 and B1D5 - 10 kDa - F1 were arranged in 12 peptides, which exhibited the highest reducing power, more than their radical-scavenging inhibition (p < 0.05). Therefore, it was concluded that both peptides obtained from PSPH have released antioxidative peptides that could be beneficial towards consumer's health, particularly the spontaneous plaa-som fermented with the addition of pineapple.

Students with complex support needs require individualized education programs (IEPs) to describe their present levels of academic achievement and functional performance (PLAAFP), and the annual goals and supplementary aids and services (SAS) that will be provided to enable them to make progress in the least restrictive environment. Previous research has found that IEPs do not reflect recommended practices and that IEP quality varies by educational placement in inclusive and separate class settings. The purpose of this study was to evaluate a national sample of IEPs of elementary-aged students with complex support needs to determine whether placement predicts IEP quality. We used multilevel regression to measure the extent to which placement predicts overall IEP quality as well as the quality of IEP components, including PLAAFP, goals, and SAS. We did not detect statistically significant differences in IEP quality by placement for any of these IEP components or for overall quality; instead, we found the IEPs consistently failed to meet quality indicators across all four placement types. Given these findings, we suggest implications for future research aimed at improving IEP quality for students with complex support needs.

The tensile properties of PLA/PBAT blends, PLA fibre reinforced PBAT composite (PLAF) at room temperature and -18°C were investigated. The concentrations of PLA in the blends were 10%, 20%, 30% and 40% (by volume). There was an improvement of elastic modulus (E) for PLA/PBAT blends when PLA was 40%. There was no significant difference of ultimate tensile strength (UTS) among the blends. For the same concentration of PLA (40%) in PLA-PBAT mixture, PLAF exhibited higher values of E and UTS than that of PLA/PBAT blends. Elongation of PLA/PBAT blends rapidly decreased upon the addition of PLA to the blends. The values of E and UTS for PLA/PBAT blends and composite, neat PLA, and PP increased with the decreasing of temperature from room temperature to -18°C. The effect of decreasing temperature was not observed on elongation. It was appeared from the results obtained for FTIR and DSC measurements that PLA and PBAT were immiscible, separating into two phases.

São apresentados nove táxons de Euglenophyta incolores como resultado do estudo taxonômico em ambientes lênticos (lagoas, açude, canal e banhados) de duas áreas próximas a Laguna dos Patos: a Lagoa do Casamento e ecossistemas associados (30º03'- 30º34'S e 50º25'- 50º47'W) e região do Butiazal de Tapes (30º23'30º38'S e 51º16'- 51º29'W) na Planície Costeira do Rio Grande do Sul. As coletas de rede e espremido de macrófitas aquáticas abrangeram as estações de outono e primavera de 2003. Quanto à distribuição todos os nove táxons de Euglenophyta incolores são novas citações para a Planície Costeira do Estado, sendo primeiros registros para o Brasil: Cyclidiopsis acus Korsch., Entosiphon polyaulax Skuja, Gyropaigne kosmos Skuja, Menoidium tortuosum (Stokes) Lemm. var. playfairii Bour. e Rhabdomonas mirabilis (Playf.) Schroeckh, Lee & Patterson, e ainda para o Rio Grande do Sul: Menoidium gracile Playf., Menoidium pellucidum Perty e Menoidium obtusum Pringheim. O ecossistema banhado foi o que apresentou maior riqueza destacando-se o banhado entre a Lagoa do Capivari e Lagoa do Casamento por ter sido encontrado sete táxons dentre os nove identificados. A área da Lagoa do Casamento apresentou maior riqueza de Euglenophyta incolores do que os ambientes próximos ao Butiazal de Tapes, possivelmente por esta área possuir maior contribuição antrópica.

Thai traditional fermented fish products (Plaa-som) from four provinces (Ubon Ratchathani, Surin, Sisaket, and Khon Kaen) in the northeast part of Thailand were collected and analyzed to determine their salt content, total acidity, and pH. Yeasts in all samples were isolated and identified to the genus and species level based on sequence analysis of the D1/D2 of the large subunit (LSU) rRNA gene and the internal transcribed spacer (ITS) region. The results revealed that the salt content, total acidity, and pH values are in the range of 2.01–6.9%, 0.62–1.9%, and 4.4–6.57%, respectively. Moreover, 35 strains of yeast were isolated and identified as eight genera, namely Candida, Diutina, Filobasidium, Kazachstania, Pichia, Saccharomyces, Torulaspora, and Yarrowia with 17 species. The ascosporogenous yeast, Kazachstania, was the most dominant genus found and was widely distributed among the fermented food samples. In addition, a new strain of yeast, Kazachstania surinensis, was also discovered in Plaa-som samples. Thus, this study is the first to report the presence and wide distribution of these yeasts in fish fermentation products.

The article discusses aspects of the organization of distance learning using educational platforms, the capabilities of Google Tables and distance communication systems. The experience of organizing distance learning also in the mode of full-time education is given. The transition to a distance learning format was the only way to continue training and effective feedback with students in a pandemic, but, as practice has shown, the skills of organizing training in a distance format have been closely integrated into the educational process, especially for an open school. The use of a Russian educational platform with ready-made tools for teaching, monitoring and analyzing the data obtained made it possible in the shortest possible time to restructure the learning process with minimal labor costs for both school teachers and students and their parents.
