Повний текст джерелаB-cell malignancies are a heterogeneous group of clinical conditions including indolent diseases such as Chronic Lymphocytic Leukemia (CLL) and highly aggressive lymphoproliferative disorders such as Burkitt’s lymphoma. B-cell malignancies treatments take advantage of dose-intensive chemotherapeutic regimens and immunotherapy via monoclonal antibodies. Unfortunately, they may lead to insufficient tumor distribution of therapeutic agents and cause several adverse effects. Thus, we propose a novel therapeutic approach for the treatment of CLL and Burkitt’s lymphoma in which high-doses of the association of hydroxychloroquine and chlorambucil (HCQ/CLB) or fludarabine were loaded inside biodegradable nanoparticles (BNPs) coated with an anti-CD20 antibody. First of all, a Burkitt’s lymphoma cell line (BJAB), two CLL cell lines (MEC1 and EHEB) and cells purified from patients’ blood samples were used to confirm CD20 expression and to assess BNPs binding and internalization. These studies demonstrated BNPs ability to bind malignant B cells and to enter inside cells in a process different from endocytosis. Then, BNPs therapeutic effect was evaluated by MTT test, AnnV/PI assay and western blot to put in evidence apoptosis induction and autophagy inhibition. These experiments demonstrated drugs-loaded BNPs ability to kill malignant B cells with comparable effects than those obtained with free drugs whereas empty BNPs were practically ineffective. In vivo BNPs characterization included the evaluation of their toxicity, biodistribution and therapeutic effect. C57/BL mice were used to evaluate BNPs toxicity which was studied considering survival, loss of body weight and several tissue markers in the blood. Mice receiving 8 injections of free HCQ+CLB died in this experiment whereas animals challenged with the same amount of drugs encapsulated inside BNPs did not show toxic effects suggesting BNPs safety. The importance of antiCD20 antibody in the homing of BNPs was confirmed by in vivo Time-Domain Optical Imaging performed in localized B-cell malignancy-bearing mice. This analysis suggested the ability of antiCD20-conjugated BNPs to specifically target tumor B-cells, with a pick after 24-48 hours. On the contrary, untargeted BNPs localization inside tumor was significantly decreased. In this analysis it was also evident that the liver is the main site of BNPs’ elimination while in the other organs the presence of fluorescent BNPs was very low. Finally, BNPs ability to treat a new xenograft human/SCID leukemia and Burkitt’s lymphoma mouse model was studied. Drugs-loaded BNPs were able to improve HCQ/CLB efficacy in vivo allowing the cure of treated all Burkitt’s lymphoma-bearing mice and 3 out of 7 leukemia-bearing animals. All these data together put the basis for the potential use of BNPs in the treatment of B-cell malignancies.
I tumori a cellule B sono un gruppo eterogeneo di patologie che comprendono sia malattie indolenti, come la leucemia linfatica cronica (LLC), sia aggressive, come il linfoma di Burkitt. Il trattamento delle patologie a cellule B prevede sia l’utilizzo di chemioterapici (agenti alchilanti e analoghi delle purine) che di anticorpi monoclonali. Nonostante la varietà di terapie esistenti, l’efficacia di questi farmaci è limitata dalla mancata specificità per le cellule tumorali e dall’induzione di gravi effetti collaterali. Per ovviare ai limiti delle terapie attuali, è stato quindi proposto l’utilizzo di nanoparticelle coniugate con un anticorpo antiCD20, specifico per le cellule B, e contenenti alte concentrazioni di chemioterapici (idrossiclorochina e clorambucile o fludarabina). Le nanoparticelle sono state caratterizzate in vitro e in vivo durante questo progetto di dottorato. Inizialmente sono stati effettuati studi in vitro al fine di valutare l’espressione del CD20 sulla superficie di una linea cellulare di linforma di Burkitt (BJAB), due linee di LLC (MEC1 e EHEB) e cellule purificate da campioni di sangue di pazienti affetti da LLC. In seguito, il legame e l’internalizzazione delle nanoparticelle a queste cellule sono stati dimostrati suggerendo anche come le nanoparticelle vengano internalizzate attraverso un meccanismo diverso dall’endocitosi. L’effetto terapeutico in vitro delle nanoparticelle è stato valutato con test MTT, AnnessinaV/PI e tramite western blot al fine di evidenziare l’induzione di apoptosi e l’inibizione dell’autofagia, meccanismi attraverso cui i farmaci utilizzati sono noti agire. Questi esperimenti hanno dimostrato che le nanoparticelle cariche di chemioterapici sono in grado di uccidere le cellule B tumorali con effetti paragonabili a quelli ottenuti da pari concentrazioni di farmaci liberi dimostrando come il processo di produzione delle nanoparticelle non influisca sull’efficacia dei chemioterapici. Al contrario, nanoparticelle vuote non sono in grado di uccidere le cellule dimostrando la mancata tossicità dei polimeri da cui sono costituite. Dopo aver confermato il legame e l’internalizzazione delle nanoparticelle che inducono la morte delle cellule B tumorali, sono stati effettuati esperimenti in vivo tra cui studi di tossicità al fine di valutare eventuali effetti collaterali indotti dal trattamento, studi di biodistribuzione e la valutazione degli effetti terapeutici. Gli studi di tossicità sono stati effettuati in topi sani valutando parametri quali la perdita di peso, la sopravvivenza e la tossicità sistemica. Nanoparticelle cariche di farmaci presentano un profilo tossicologico sicuro mentre pari dosi di farmaci liberi inducono la morte di tutti gli animali trattati. Questi esperimenti dimostrano quindi come l’inserimento di farmaci all’interno di nanoparticelle prevenga gli effetti collaterali normalmente indotti dai chemioterapici. Secondariamente, sono stati effettuati studi di biodistribuzione di nanoparticelle coniugate o meno con un anticorpo antiCD20. Questi studi effettuati tramite Optical Imaging dimostrano come nanoparticelle coniugate con l’anticorpo antiCD20 si localizzino preferenzialmente nella massa tumorale in 24-48 ore in quantità maggiore rispetto a nanoparticelle non coniugate con l’anticorpo. Inoltre, da queste analisi risulta evidente come il fegato sia il maggiore sito di eliminazione delle nanoparticelle mentre in altri organi la presenza di nanoparticelle è molto bassa. Infine, un modello disseminato di linfoma di Burkitt e un modello di LLC sono stati sviluppati in topi SCID (Severe Combined ImmunoDeficiency) iniettando rispettivamente cellule BJAB intraperitoneo e cellule MEC1 endovena. I modelli sono stati caratterizzati e utilizzati per valutare la potenziale applicazione delle nanoparticelle nel trattamento di queste patologie. Questi studi hanno dimostrato come le nanoparticelle siano in grado di aumentare l’efficacia dei chemioterapici e di curare tutti i topi affetti da linfoma di Burkitt e 3/7 topi affetti da leucemia. Concludendo, questi risultati suggeriscono la potenziale applicazione delle nanoparticelle cariche di chemioterapici nel trattamento di LLC e linfoma di Burkitt.

Повний текст джерелаGalectins are bridging glycosylated molecules by their β-galactoside moieties, forming a network involved in many physiological functions. Extracellularly, galectins function has been mainly described through carbohydrate binding activity. The first example of a carbohydrate-independent interaction outside the cell concerns galectin-1 (Gal1) and its ligand, the pre-BCR. This interaction has a crucial role in B-cell development through the formation of a Gal1-dependent lattice between pre-BII and stromal cells. Our previous study revealed that Gal1 interacts with a motif of the pre-BCR (λ5-UR) on a surface adjacent to the Gal1 CBS (Carbohydrate Binding Site) and allows Gal1 to undergo selective affinity changes in its carbohydrate-binding activity in vitro.Using solution state on cell NMR spectroscopy, we investigated the effect of λ5-UR interactions on Gal1 binding activity for its physiological ligands expressed on pre-BII and stromal cell surfaces. We show that λ5-UR can specifically induce a ligand binding shift of Gal1 when bound to its cell surface ligands. We also identified one glycan epitope for which Gal1 increases its affinity upon λ5-UR interaction and deciphered the intermolecular contact changes induced by λ5-UR. In addition, glycan arrays screening combined to NMR relaxation data showed that λ5-UR changes the internal dynamics of specific Gal1 CBS residues resulting in the targeting of specific glycan epitopes. This cellular and structural NMR study ever conducted at atomic resolution for a member of the galectins demonstrates that Gal/unglycosylated protein interactions can act as physiological regulators of Gal1 lattice interactions at the cell surface

Повний текст джерелаCell therapy for regeneration, has received extensive attention and the accumulated evidence from both pre-clinical and clinical studies suggests that it has the potential to restore heart function. However, the results from first clinical trials are mixed, with benefits ranging from absent to transient and, at most, marginal. These studies indicate that adult stem cells, whether muscular or bone marrow-derived, fail to generate new cardiomyocytes, capable to improve cardiac function. Emerging evidence suggests that several subpopulations of resident cardiac stem cells (CSCs) have the ability to differentiate into cardiac myocytes, vascular smooth muscle and endothelial cells. CSCs represent a logical source to exploit in cardiac regeneration therapy bacause, unlike other adult stem cells, they are likely to be intrinsecally programmed to generate cardiac tissue in vitro and to increase cardiac tissue viability in vivo. In addition, autologous CSCs can be employed avoiding ethical and immunological problems associated with the use of embryonic stem cells. Recently, a group of our network has successfully isolated CPCs/CSCs from small biopsies of human myocardium and expanded them ex vivo trough several generations without loosing differentiation potential into cardiomyocytes and vascular cells, bringing autologous transplantation of cardiac stem cells closer to clinical translation. However cell therapy in general suffers limitatations related to variable cell retention, survival and significant cell death or apoptosis, early after implantation in the diseased myocardium. Furthermore, cell transplantation may not always be suitable for catastrophic events like large myocardial damage. For this reason, hybrid therapies that incorporate tissue engineering are being developed as potentially new therapeutic approaches for repair of myocardial tissue. Tissue engineering (TE) involves seeding a biodegradable scaffold with cells that grow into morphologically recognizable tissue both in vitro and in vivo. Recent advances in cell culture and TE have facilitated the development of suitable cell-engineered biodegradable grafts. The optimal biomaterials and cell types, however, have not been identified. Our hypothesis is that autologous cardiac stem cells could represent the most efficient and reliable cell type to be used for an hybrid therapy (tissue engineering/stem cells) to restore myocardial function in ischemic myocardial desease. TE joints the physical replacement of the diseased structure with new cardiac tissue built from a biodegradable scaffold, with the regenerating activity of the optimal cell types. A bioengineered tissue graft (biocomplex) would be the ideal treatment to repair cardiac ischemic diseases. The possibility to compare the biological activity of the CSCs with other adult SCs, should definitely individuate and characterize the advantages and disadvantages of the best clinical applicable biocomplex. Moreover, the creation of the appropriate animal model and the realization of diagnostic protocols aimed to monitor the in vivo cell fate, will be used as pre-clinical background for large animals and phase I-II clinical studies.

CCND1 encodes two mRNA isoforms, « a » and « b » by alternative splicing. The respective role of each protein in the B-cell transformation process remains unknown. By using various cellular models and gain- and loss-of-function strategies we report that : downregulation of CCND1 expression is not sufficient for cell proliferation arrest and cell death promotion ; ectopic expression of cyclin D1a inhibits mitochondrial activity through the recruitment of VDAC channel ; ectopic expression of cyclin D1b allows in vivo engraftment through the activation of neoangiogenesis ; cyclins D1a/b and K regulate cell metabolism

Повний текст джерелаIn the recent years the increased presence of human made compounds that mimic the action of estrogens termed endocrine disrupters (ED) in environment and in food and the exposure to these compounds during fetal and neonatal period has been hypotized to be the cause of the raise of disorders of male reproductive function, such a decrease of sperm count, increase in the incidence of testicular cancer and cryptorchidism and hypospadias termed Testicular Dysgenesis Syndrome (TDS). For these reason, it is important to know how the estrogens and xenoestrogens, a class of ED, act during the fetal development and to know the mechanism by which these compounds exert their effects. AIMS: To study the expression of estrogen receptors (ERs) in the embryonic precursors of the adult gametes termed PGCs and to analyze the existence in such cells of intracellular molecular pathways modulable by estrogens and xenoestrogen lindane. To verify the presence of functional ER-beta in embryonic testicular somatic cells using an ERE-luc and AP1-Luc assay and to evaluate estrogenic activity of putative EDs on mammalian embryonic testis. RESULTS: The data described in this thesis highlights the existence of functional estrogen-dependent pathways in embryonic mouse gonads in particular in testis, both in germ and somatic cells. We found that E2 is able to activate via ER-beta multiple intracellular signalling in PGCs and that the xenoestrogens, lindane affect the survival in such cells through a direct pro-apoptotic action likely resulting from its adverse effect on AKT activity. Othermore, we described for the first time the existence of a functional ERα pathway in putative Leydig cells from early stage of testis development and describe an in vitro assay that can be used to evaluate estrogenic activity of compounds on mammalian embryonic testis. CONCLUSIONS: These results support the notion of the TDS origin during early stages of testis development. While data are accumulating showing direct effect of estrogens and EDs on gene expression and specific functions of somatic cells of the embryonic testes, in particular Leydig cells, such results on germ cells are lacking and further studies are needed to investigate the effects of these compounds on embryonic germ cell function including epigenetic regulation.

Повний текст джерелаAfter antigen recognition, B cells are activated and interact with other cells within secondarylymphoid organs (dendritic cells, T lymphocytes …) to form a germinal center. In the GC, the IgH locusis reorganized in order to increase the affinity of immunoglobulins for antigen through somatichypermutation (SHM) of V(D)J regions and to configure them into several forms harboring diversifiedmodes of action after “class switc recombination” (CSR). Both mechanisms are initiated by ActivationInduced Deaminase (AID) which targets DNA cytosines to convert them into uracil, then causing singleor double strand breaks in DNA when the mismatchs are located close to each other. It has been shownthat AID can target the IgH locus 3’ regulatory region on specific regions called LS, then leading to thetotal deletion of IgH locus C genes, loss of BCR expression and cell death by locus suicide recombination(LSR). In our study, we created a human Cμ knock-in model distal to the hs4 element of the 3’RR, in anattempt to rescue cells after the LSR event. Our model showed that this insertion indeed succeededinto replacing LSR by “class switching to humanized IgM” and also qualitatively modulated someaspects of the humoral response. This new LSR reporter model additionally supports the hypothesisthat LSR is regulated and increases with B cell activation. Studies of ex vivo B cells from the modelsuggest that LSR can occur in T dependent and independent manners, but is induced by triggering TLR4but not TLR9. Studies of the human IgM repertoire showed a biased use of VH families, and notably themouse VH5 family was used more frequently than in the control group. The BCR repertoire bias stronglysuggests that LSR is at least in part a matter of affinity of the BCR variable regions for antigens andligands that remain to be characterized

Повний текст джерелаLa leucemia linfatica cronica di tipo B (LLC-B) è la forma più comune di leucemia nell’adulto ed è caratterizzata dall'accumulo clonale di piccoli linfociti B CD5+ nel sangue periferico, nel midollo osseo e negli organi linfatici, dovuto sia ad un aumento della proliferazione che ad un difetto dei meccanismi di morte cellulare programmata. La resistenza all’apoptosi in questi linfociti favorisce la progressione della malattia attraverso un’aumentata sopravvivenza del clone neoplastico e l’induzione della resistenza ai farmaci citostatici. Tali alterazioni sono imputabili sia a fattori intrinseci che a fattori estrinseci derivanti dal microambiente. Poiché l’aumentata sopravvivenza ed il progressivo accumulo del clone linfocitario risultano selettivamente favoriti dall'interazione con cellule accessorie non tumorali presenti nel microambiente in cui esso risiede, in questa tesi abbiamo focalizzato l’attenzione sulle cellule mesenchimali staminali (MSC), allo scopo di valutare il loro ruolo nella sopravvivenza e nella compartimentalizzazione del clone B leucemico. Le MSC costituiscono una frazione esigua (inferiore allo 0,01%) della popolazione di cellule midollari, sono cellule staminali multipotenti in grado di differenziare in diversi tessuti di origine mesenchimale; sono inoltre dotate di proprietà immunomodulatorie verso linfociti B, T, Natural Killer e cellule dendritiche. In questa tesi le MSC sono state isolate dal sangue midollare di 47 pazienti affetti da LLC-B e sono state caratterizzate fenotipicamente e funzionalmente mediante analisi citofluorimetrica (positività per CD73, CD90 e CD105, negatività per CD31, CD34 e CD45) e colture differenziative (adipociti ed osteociti) confrontandole con MSC di donatori sani. Successivamente si sono allestite co-colture di linfociti B di pazienti affetti da LLC e MSC allo scopo di valutare l’effetto delle MSC sul clone neoplastico di LLC. Le MSC ottenute dai pazienti non hanno presentato alterazioni dal punto di vista fenotipico né funzionale rispetto alle MSC di donatori sani, tuttavia esse hanno sviluppato interazioni capaci di favorire la sopravvivenza del clone leucemico. Gli esperimenti di co-coltura hanno dimostrato infatti che le MSC esercitano un effetto anti-apoptotico sui linfociti B neoplastici, documentato da un aumento significativo della sopravvivenza delle cellule B di LLC dopo 7 giorni di coltura, effetto verificatosi anche con MSC di donatori sani e invece molto meno marcato nei linfociti B normali. Tale attività anti-apoptotica si è osservata, seppur di minore intensità, anche nei linfociti B di pazienti sottoposti a trattamento chemioterapico in vivo con Fludabarina e Ciclofosfamide, suggerendo che le MSC possano essere implicate anche nei meccanismi di chemio resistenza del clone di LLC. Infine l’analisi della migrazione cellulare dei linfociti B patologici in presenza di terreno condizionato derivante dalle colture di MSC ha dimostrato la capacità delle MSC di produrre stimoli chemiotattici in grado di richiamare in sede midollare il clone maligno, ma non i linfociti B normali. I dati riportati suggeriscono che le MSC nei pazienti affetti da LLC-B, sebbene non mostrino alterazioni dal punto di vista fenotipico e funzionale, svolgono un ruolo attivo nel favorire la sopravvivenza e la compartimentalizzazione delle cellule B neoplastiche a livello midollare.

Повний текст джерелаLa leucemia linfatica cronica a cellule B (LLC-B) è la forma più comune di leucemia nell’adulto ed è caratterizzata dall’accumulo di piccoli linfociti B CD5+ nel sangue periferico, nel midollo osseo e negli organi linfatici, dovuto sia a un aumento della proliferazione, che a un difetto dei meccanismi di morte cellulare programmata. Tra i fattori che contribuiscono alla sopravvivenza del linfocita neoplastico un ruolo fondamentale svolge l’attività anomala di diversi enzimi con attività fosfotransferasica che modulano le principali vie di segnalazione intracellulare, tra cui la lipide chinasi PI3K, la serina/treonina chinasi PKC, e le tirosin-chinasi Syk e Lyn. L’attività di ricerca svolta durante il mio dottorato si è concentrata sulla Src chinasi Lyn, la quale in questa patologia risulta sovra-espressa, iperattiva e delocalizzata nel citosol come componente di un complesso multiproteico aberrante. All’interno di questo complesso, Lyn è associata al chaperone molecolare Hsp90, il quale la preserva in conformazione attiva, così contribuendo all’elevata tirosin-fosforilazione basale tipica della LLC-B. Lo scopo della mia indagine è stato identificare le molecole substrato dell’anomala attività di Lyn, potenzialmente coinvolte nei fenomeni di resistenza all’apoptosi dei cloni leucemici. Grazie all’applicazione di metodi biochimici e biomolecolari, nonché a un approccio bioinformatico, abbiamo identificato due substrati proteici di Lyn, la proteasi caspasi 8 e la proteina fosfatasi 2A (PP2A). In primo luogo, abbiamo dimostrato che nelle cellule B di LLC, la procaspasi 8, zimogeno della caspasi 8, è fosforilata in Tyr380 ad opera di Lyn citosolico; tale fosforilazione comporta non solo l’inibizione dell’attività caspasica ma anche il riarrangiamento strutturale della proteasi stessa, che nei cloni leucemici localizza nel citosol in forma dimerica. Tale inibizione viene rimossa usando sia inibitori di Lyn, come il dasatinib, sia inibitori di Hsp90, come geldanamicina, quest’ultimo provocando la rottura del complesso citosolico e quindi il calo dell’attività di Lyn e in ultima analisi attivando le vie apoptotiche estrinseca ed intrinseca. In secondo luogo, abbiamo verificato che Lyn esercita un controllo negativo su uno dei fattori chiave della modulazione dei segnali che regolano la sopravvivenza cellulare, la serina/treonina fosfatasi PP2A. I nostri dati indicano l’esistenza di un meccanismo d’inibizione dell’attività fosfatasica di PP2A, che dipende sia dalla fosforilazione come tale in Tyr307 della subunità catalitica, ma anche dalla stabilizzazione dell’interazione della fosfatasi con il suo inibitore proteico cellulare SET, il quale è anch’esso sovra-espresso nelle cellule di LLC-B. La formazione di questo complesso stabile mantiene la fosfatasi in uno stato inattivo contribuendo quindi alla persistenza dei segnali di sopravvivenza mediati in particolar modo dalla serina/treonina chinasi Akt. Inoltre, allo scopo di individuare nuove molecole in grado di riattivare PP2A e quindi utili a contrastare i segnali di sopravvivenza delle cellule leucemiche, abbiamo verificato l’utilità di un noto attivatore di PP2A, il fingolimod (FTY720), già impiegato nella sclerosi multipla e ultimamente rivelatosi efficace nel causare la morte cellulare in vari tipi di modelli di neoplasia. Il meccanismo di azione risiede nella capacità di rimuovere l’inibitore SET dalla subunità catalitica di PP2A. Questo composto, tuttavia, possedendo attività immunosoppressiva a causa dell’induzione della degradazione del recettore della sfingosina ma anche effetti collaterali a carico del sistema cardio-vascolare, può rivelarsi inadeguato nell’ottica di una più complessa strategia terapeutica in campo oncologico. Quindi sono stati sviluppati degli analoghi strutturali che non interferissero con il recettore della sfingosina ma destabilizzassero il complesso della PP2A fosforilata e SET. In questo modo è stato identificato un composto (MP07-66), con un’efficacia sovrapponibile al fingolimod sia nel disgregare il complesso che nell’inibire la protein chinasi Akt, fondamentale nel preservare i segnali di sopravvivenza. Inoltre tale composto era in grado di indurre apoptosi su cellule leucemiche, il quale effetto veniva incentivato dalla presenza di dasatinib con meccanismo sinergico, ulteriormente confermando il ruolo della fosforilazione nella stabilità del complesso PP2A/SET. In conclusione, questo lavoro identifica PP2A e caspasi 8 quali substrati dell’attività citosolica aberrante di Lyn, confermando il ruolo chiave di questa chinasi nel supportare i molteplici segnali anti-apoptotici presenti nella LLC-B. I meccanismi molecolari di resistenza all’apoptosi qui identificati possono costituire dei potenziali bersagli per lo sviluppo di nuove terapie ad uso clinico.

Furthermore, the characterization of the major regulatory proteins and effectors, upstream and downstream of Akt, has led to the conclusion that the cascade of reactions of this signaling pathway in hMSC follows a canonical pathway. Pharmacological inhibitors were used to determine the potential mechanism responsible for cell migration and invasion. Blocking PI3K/Akt pathway resulted in decreased hMSC migration. The use of pharmacological inhibitors specific for individual Akt isoforms allowed us to discriminate the different role of Akt1 and Akt2 in the migration of the hMSC. Through our analysis, we demonstrated that pharmacological inactivation of Akt2, but not that of Akt1, significantly decreased cell migration and invasion. Although these results are not fully comprehensive for the understanding of the phenomenon, in the complex indicate that the activation of Akt2 plays a critical role in allowing the migration of the hMSC. The demonstration that the Akt2 isoform is required for the chemotaxis of direct hMSC, makes this kinase a potential pharmacological target to modulate the migration of such cells.

We mapped 8 QTL implicated in testis, development, prostate growth, sperm vitality and morphology. We performed fine mapping analysis of a QTL of reduced testis weight associated with teratozoospermia, localised on MMU11. We proposed a candidate locus encompassing four testis expressed gene. Moreover, in order to understand gene expression regulation in interspecific mosaic genome, we analysed testis transcriptome of three IRC strains compared with parental strains testis transcriptome. In this study, we describe how spretus genes are regulated when introgressed in musculus background. This study gives some insight concerning gene flow tolerance across the specie barrier during emergence of mosaic genome

Повний текст джерелаHepatitis B virus (HBV) infection is a major health problem worldwide as (1) despite an effective preventive vaccine over 240 million individuals are chronically infected and (2) the actual viral suppressive treatments available do not eliminate viral DNA from cells. Thus, infected individuals are at a high risk of developing hepatocellular carcinoma (HCC) and understanding viral replication mechanisms and how it impacts on hepatocarcinogenesis is a major challenge.The role of the HBx protein, notably in viral replication and oncogenic processes, is the subject of many publications. However, many studies have often used non-physiological infection conditions. My thesis project has addressed these limitations by using cellular models, including primary human hepatocytes which can be infected by HBV, to investigate HBx’s role in these processes. I have shown that HBx is indispensable for HBV replication and that HBx associates with the infected cell’s DDB1/ E3 ubiquitin complex to target its Smc5/6 complex for degradation via the proteasome. These studies have identified that the Smc5/6 complex is a novel viral restriction factor that acts at an epigenetic level to block viral replication. This unexpected role of SMC5/6 has led to new research into the evolutionary conservation of restriction factors for episomal DNA viruses. As SMC5/6 is implicated in DNA Damage Repair (DDR), the last section of my thesis reports how SMC5/6 degradation in infected cells can sensitise cells to the cell killing effects of DNA damaging agents such as ionizing radiation and hydroxyurea. These results open-up possibilities for HCC treatment where HBx expression may be of therapeutic benefit

Повний текст джерелаInvasive fungal diseases (IFDs) represent an important cause of morbidity and mortality in patients affected by lymphoproliferative diseases. In the recent years, the introduction of new drugs, in particular those targeting Bruton tyrosine kinase (BTK), has allowed dramatic improvement in the prognosis of patients with chronic lymphocytic leukemia (CLL) and other B-cell clonal disorders. Although these small molecules were initially considered less immunosuppressive than chemo-immunotherapy, an increasing number of reports have described the occurrence of unexpected opportunistic fungal infections, in particular invasive aspergillosis, in the first 6 months of treatment. BTK represents a crucial molecule in the transmission of signaling cascade from several immune receptors, such as pathogen-associated molecular patterns (PAMPs, i.e. β-glucans, chitin and mannans), CD11b/CD18, triggering receptor expressed on myeloid cells 1 (TREM-1) and Toll-like receptors (TLRs), that allow the recognition of fungi by the innate arm of the immunity. However, the immunopathogenetic mechanisms underlying the development of IFDs in patients treated with BTK inhibitors are not fully elucidated, and multiple pathways might be involved. Our in vitro study, based on different immunological functional assays, aimed at characterize the specific off-target effects of BTK inhibitors on anti-mold innate immune response mediated by neutrophils, monocytes, macrophages, as well as platelets, obtained from patients with B-cell neoplasms. Pharmacological inhibition of BTK reduced signaling pathways activated by Aspergillus fumigatus, determining an exacerbation of an immunosuppressive signature and a significant defect in the secretion of inflammatory cytokines, either in neutrophils, macrophages and monocytes isolated from patients and healthy donors. In neutrophils, the inhibition of BTK significantly hampered the phagocytic activity (mean reduction of 51,6% and 37,6%, with two different BTK-inhibitors). Similarly, we found a remarkable reduction in the phagocytic function of CLL-associated macrophages (nurse-like cells, NLC), as well as of CD14+ circulating monocytes (either in CLL patients and healthy volunteers). Concordantly, secretion of TNF-α by NLC, assessed by cytokine secretion assay (CSA), was strongly induced by pulsing the cells with Aspergillus fumigatus conidia and was markedly reduced by two different drugs targeting BTK. Furthermore, we demonstrated, for the first time, that the exposure to BTK-inhibitors impairs several immune functions of platelets, i.e. activation (as indicated by the low levels of P-selectin expression), direct killing activity and ability to adhere to conidia. Hyphal damage test, performed on neutrophils, monocytes, macrophages and platelets in the presence of pharmacological BTK-inhibition, documented a striking reduction of the anti-hyphal lytic activity in all aforementioned cell types, either in patients and healthy volunteers. Overall, these effects concur to a failure in completely counteracting conidia germination. Our results revealed specific modifications in the innate response in patients with B-cell neoplasms induced by the inhibition of the BTK pathway, underlying the importance of this targetable kinase as a “guardian” of the innate immunity.

Повний текст джерелаWhilst adult neurogenesis is a limited process in mammals, it is, in contrast, intense in teleost fish and occurs in the whole brain. Fish exhibit specific features concerning the aromatase, the enzyme that synthesises estradiol (E2). Indeed, brain aromatase (aroB) is up-regulated by E2 in radial glial cells (RGC). These RGC are known progenitor cells in mammals, during embryonic neurogenesis. In contrast, their role is unknown in fish. The aim of this work was to study the potential link between E2 and the strong neurogenic activity of fish. We show that RGC are progenitor cells in the adult brain of zebrafish (Danio rerio). Moreover, we characterized and validated a transgenic zebrafish line aroB/GFP. We also found that aromatizable and non aromatizable androgens can induce aroB through estrogen receptors. We also show that the onset of the estrogen regulation of aroB appears between 24 and 48h

Повний текст джерелаActivation of B lymphocytes is triggered by the specific binding of antigens (Ag), tethered in vivo at the surface of Ag-presenting cells, to the B Cell Receptor (BcR). Engagement of the BCR with Ag leads to the formation of an immunological synapse, which triggers B cell activation and promotes the acquisition of membrane-bound Ag for processing it into endo-lysosomes, loading onto MHCII molecules and presentation to CD4+ T cells. Interaction with T cells is required for B cell activation and differentiation into high-affinity antibody producing cells. In this thesis we have identified and highlighted the roles of Cdc42 and the PAR polarity complex as key regulators of B lymphocyte polarization. This work is the first demonstration that B lymphocytes polarize their machinery toward particulate antigens. Furthermore, polarization of the MT cytoskeleton and MHCII+ containing lysosomes at the immune synapse is fundamental for efficient Ag extraction, processing, and loading onto MHCII molecules and for presenting Ag to CD4+ T cells. This thesis shows that the Rho GTPase Cdc42 is activated upon BCR engagement and controls MTOC polarization toward the IS. Disruption of Cdc42 leads to impairments in both cytoskeleton and vesicle polarity and in Ag presentation. We further show that the PAR polarity complex is a key effector of Cdc42 in this process. First, the kinase aPKCÇ is activated under BCR engagement; it localizes on lysosomes and relies on Cdc42 activation. Second, aPKCÇ silencing results in the loss of MTOC and lysosome polarization. Finally, aPKCÇ silencing in B lymphocytes impairs Ag processing and Ag presentation to CD4+ T cells. The last results of my work show that Par3 concentrates at the immune synapse, which indicates a local polarity cue where the centrosome is efficiently docked. Par3 acts together with dynein to induce MTOC polarization at the immune synapse and is essential for efficient Ag internalization and presentation to CD4+ helper T cells. We conclude that upon B cell activation Cdc42 controls together with PAR the dynamic re-organization of the microtubule cytoskeleton, which is essential to direct the trafficking events required for Ag processing and presentation

Повний текст джерелаTerminal B cell differentiation through the germinal center leads to the production of antibody-secreting cells: plasma cells (PC) with a long lifespan and high affinity for the antigen. This reaction involves the formation of an anatomical microstructure that includes a dark zone: site of intense proliferation and affinity maturation of the BCR of the centroblasts, and a light zone in which the class switch recombination of the BCR and centrocyte (CC) selection take place. This differentiation is finely controlled by follicular helper T cells (Tfh) that produce molecules such as IL-4, CD40L and IL-21. This phD work focus on the CD23 expression on the surface of CC during their metamorphosis into PC. Firstly, I showed that the expression of the low affinity IgE receptor is regulated by Tfh derived IL-4 and CD40L. Moreover, I showed that CC exposed to Tfh signals and which do not express the CD23 were the ones that have the ability to differentiate into PC. In this context, I identified at the single cell level a transcriptional signature expressed specifically by these cells which contains a gene never described in PC, encoding the transcription factor DEC2 whose function remains to be determined. In addition, I studied CD23 expression in follicular lymphoma and showed the existence of differences between these two populations, suggesting a preferential survival and differentiation of CD23neg cells compared to CD23pos cells

Повний текст джерелаRecruitment of histone deacetylases (HDACs) by transcription factor c-Myc to promoter regions of regulatory protein- and microRNA-coding genes is essential for its proto-oncogenic functions. Considering the widespread involvement of c-Myc in lymphomagenesis, we studied the antitumor effects of the new-generation pan-HDAC inhibitor ITF2357 (Givinostat®) in c- Myc-overexpressing human B-cell non-Hodgkin lymphoma (B-NHL) models. ITF2357 significantly delayed the in vitro growth of B-NHL cell lines by inducing G1 cell-cycle arrest, eventually followed by cell death. These events correlated with the extent of c-Myc protein down-regulation and were associated with the induction of the c-Myc targeting microRNAs let-7a and miR-26a. Since microRNA modulation was not as persistent as c-Myc repression, we investigated whether cap-dependent translation was also affected and found, in effect, that 4E-BP1, eIF4E, and eIF4G, as well as its major positive regulators, Akt and PIM kinases, were inhibited in function of the cell sensitivity to ITF2357 antitumor activity. In vivo, ITF2357 significantly hampered the growth of 2 human Burkitt’s lymphoma models and, in combination with low-dose cyclophosphamide, achieved complete remissions in most animals, equaling or even exceeding the activity of standard cyclophosphamide. Collectively, these findings provide the rationale for testing the clinical advantages of adding ITF2357 to conventional treatments for c-Myc overexpressing lymphomas.

Повний текст джерелаIl carcinoma della cervice uterina rappresenta la seconda causa di morte per tumore tra le giovani donne fra i 15 e i 44 anni, dopo il tumore al seno. Si stimano infatti a livello mondiale 530.000 nuovi casi di tumore cervicale all’anno e circa 275.000 decessi. Lo sviluppo del tumore alla cervice uterina è imputabile all’infezione persistente, sessualmente trasmessa, di alcuni genotipi ad alto rischio di Papillomavirus (HPV); in particolare più del 70% di tutti i tumori cervicali è correlato con la presenza di HPV16 e 18 mentre il rimanente 25% è legato all’infezione causata da altri genotipi di HPV come 31, 33, 45 e 58. Le donne immunocompetenti sono in grado di eliminare spontaneamente le infezioni dei genotipi ad alto rischio in 12-18 mesi. La seroconversione che deriva dall’infezione non avviene in tutte le donne e, laddove succede, i titoli degli anticorpi neutralizzanti sono molto bassi e non proteggono da successive reinfezioni. Inoltre, il 10% delle donne non è in grado di eliminare il virus e l’infezione persistente a livello della mucosa cervicale, è il punto di partenza di una serie di eventi molecolari che portano allo sviluppo di lesioni neoplastiche. Nel 2006-2007 sono stati approvati e commercializzati due vaccini profilattici anti-HPV costituiti dalle proteine L1 del capside virale, assemblate a formare degli pseudovirioni (VLP) tridimensionalmente identici alle particelle virali native ma non infettivi: Cervarix (GSK) contenente le VLP di HPV16 e 18 e Gardasil (Merck) contenente le VLP di HPV6, 11, 16 e 18, quindi protettivo anche nei confronti di infezioni genitali di tipo benigno come i condilomi. Gli studi clinici hanno dimostrato che entrambi i vaccini sono sicuri, capaci di indurre elevati livelli di anticorpi neutralizzanti contro la proteina L1 ed efficaci nel proteggere dall’infezione da parte dei genotipi di HPV presenti nel vaccino. Tuttavia, i vaccini sono in commercio solo da pochi anni e sono necessari ulteriori studi per correlare l’efficacia di protezione con l’entità delle risposte immunitarie indotte. Per studiare l’immunità a lungo termine, la determinazione dei titoli anticorpali (neutralizzanti e non) è uno strumento utile ma non sufficiente, ed è necessario quindi quantificare i linfociti B memoria antigene-specifici. Inoltre, sono ancora pochi i dati disponibili in letteratura sull’induzione delle risposte immunitarie in funzione dell’età di somministrazione del vaccino e soprattutto la maggior parte dei dati di efficacia e di risposte immunitarie indotte da Gardasil e Cervarix provengono dalle aziende che commercializzano i vaccini. In questo contesto, lo studio indipendente da noi eseguito aveva tre obiettivi: 1) sviluppare e standardizzare una metodica di B-cell Elispot per la quantificazione dei linfociti B memoria HPV genotipo-specifici; 2) quantificare i linfociti B memoria e i titoli anticorpali, specifici per ciascun antigene vaccinale, in una popolazione di adolescenti (12 anni) e di donne (20-45 anni) vaccinate con Gardasil o Cervarix, arruolate 1-6 mesi o 4 anni dopo la vaccinazione; 3) comparare l’immunogenicità dei due vaccini anti-HPV. In particolare, ad oggi sono state arruolate 283 volontarie di cui per il vaccino Gardasil n=121 per il gruppo 12 anni, n=112 per il gruppo 20-45 anni e per il vaccino Cervarix n=60 per il gruppo 12 anni. L’arruolamento dei soggetti vaccinati con Cervarix è ancora in corso e lo studio verrà completato nei prossimi 6 mesi. I risultati dello studio hanno dimostrato che le frequenze dei linfociti B memoria HPV genotipo-specifiche indotte dal Gardasil sono elevate 1-6 mesi dopo la vaccinazione in entrambe le coorti di età e, nonostante diminuiscano nel corso del tempo, dopo 4 anni continuano ad essere significativamente elevate indipendentemente dall’età di somministrazione del vaccino. Al contrario, i titoli anticorpali sono influenzati dall’età di somministrazione del vaccino. Infatti, i titoli anticorpali misurati nella coorte di adolescenti sono sempre significativamente superiori rispetto a quelli misurati nella corte delle donne, sia 1-6 mesi sia 4 anni dopo la vaccinazione. In aggiunta, la percentuale di individui i cui titoli anticorpali non sono più misurabili, dopo 4 anni dalla vaccinazione, è significativamente superiore nelle donne rispetto alle adolescenti. Relativamente alla comparazione delle risposte immunitarie indotte dai due vaccini anti-HPV, i risultati preliminari ottenuti in un gruppo di adolescenti vaccinate con Cervarix dimostrano che 1-6 mesi dopo la vaccinazione il Cervarix induce titoli anticorpali e frequenze di linfociti B memoria anti-HPV16 e 18 significativamente superiori rispetto ad adolescenti vaccinate con Gardasil. I risultati ottenuti potranno essere un utile ausilio per il Ministero della Salute per tracciare le linee guida di prevenzione primaria del tumore alla cervice uterina.

Повний текст джерелаDans les conditions physiologiques normales, la glycémie est maintenue dans d'étroites limites. La sécrétion d'insuline par les cellules B pancréatiques joue un rôle majeur dans ce contrôle. En effet, l'insuline est la seule hormone capable d'empêcher une élévation excessive de la glycémie. La cellule B pancréatique constitue un exemple d'oscillateur cellulaire dont la dynamique non linéaire permet l'apparition d'une activité électrique "en salves" ("bursting"). Lors de la sécrétion, les cellules B présentent des oscillations du potentiel membranaire, alternances de phase actives, où la membrane est dépolarisée et durant laquelle des potentiels d'action sont produits, et de phases silencieuses, où le potentiel membranaire est stable et hyperpolarisé. La durée relative de la phase active est directement carrelée au taux de sécrétion d'insuline. Cette activité électrique est produite par une modification des flux ioniques transmembranaires. Il est maintenant établi que l'élévation de la concentration intracellulaire de Ca2+ ([Ca2+]i) qui se produit lors de la phase active est impliquée dans le déclenchement de l'exocytose. Néanmoins, les mécanismes impliqués dans la régulation de l'activité électrique restent mal compris.
Dans ce travail, nous examinons le rôle d'une protéine présente dans la membrane de la cellule B, l'échangeur Na+/Ca2+ ,dans la modulation des oscillations du potentiel membranaire et de la [Ca2+]i .Depuis sa découverte au niveau des cellules cardiaques et de l'axone de calmar, l'échange Na+/Ca2+ a été identifié dans de nombreux autres types cellulaires dont la cellule B pancréatique. Ce transporteur utilise le gradient électrochimique du Na+ comme source d'énergie pour l'expulsion des ions Ca2+ du cytosol. Dans les cellules cardiaques, il représente un mécanisme important d'expulsion du Ca 2+ intracellulaire. De plus, étant donné la stoechiométrie de la réaction de transport, son activité induit un courant (I Na/Ca) qui est impliqué dans la prolongation des potentiels d'action cardiaques. Au niveau de la cellule B pancréatique, le rôle de l'échange Na+/Ca2+ reste mal compris. L'absence d'inhibiteur spécifique de la protéine handicape sérieusement l'approche expérimentale.
Dès lors, nous avons utilisé une approche basée sur la modélisation mathématique afin de clarifier l'impact de l'activité de l'échange Na+/Ca2+ sur l'activité électrique et la régulation de [Ca2+]i de la cellule B pancréatique. La modélisation de l'activité électrique de la cellule B pancréatique repose sur le formalisme développé par Hodgkin et Huxley pour la cellule nerveuse. Dans ce contexte, l'application de la conservation de la charge au circuit équivalent de la membrane cellulaire fournit un système d'équations différentielles ordinaires, non linéaires.
Lors de la première partie de notre travail, nous avons mis au point un dispositif expérimental permettant de mesurer le courant électrique lié aux phénomènes de transport ioniques dans les membranes cellulaires. Nous avons pu détecter I Na/Ca et étudier son activation par la [Ca2+]i et le potentiel membranaire. Ces données expérimentales nous ont permis de proposer un modèle minimal pour I Na/Ca. D'autre part, afin d'évaluer expérimentalement l'effet de l'échange Na+/Ca2+ sur l'activité électrique de la cellule B pancréatique, nous avons étudié l'effet d'une réduction de la concentration extracellulaire de Na+ sur les oscillations du potentiel membranaire.
D'un point de vue théorique, à partir de notre modèle minimal pour I Na/Ca ,nous avons élaboré différents modèles mathématiques de l'activité électrique des cellules B. Ces modèles fournissent une prédiction correcte, qualitativement et quantitativement, de l'effet d'une réduction de la concentration extracellulaire de Na+ sur l'activité électrique périodique de la cellule B pancréatique. D'autre part, nos simulations numériques nous ont permis de démontrer la capacité de l'échange Na+/Ca2+ à moduler la durée relative de la phase active des oscillations du potentiel membranaire. De plus, nous avons pu mettre en évidence un mécanisme physiologique original liant la concentration extracellulaire de glucose et l'activité du transporteur. Enfin, nous nous sommes intéressés aux effets induits par la présence de l'échange Na+/Ca2+ sur l'activité électrique périodique et la régulation de [Ca 2+]i de cellules B couplées électriquement et hétérogènes en leurs paramètres. En effet, dans les conditions physiologiques, les cellules B constituent une population de cellules hétérogènes, électriquement couplées au sein des îlots pancréatiques. Il est établi que ce couplage joue un rôle essentiel dans l'apparition et la régulation de l'activité électrique périodique. Nous avons étudié différents modèles mathématiques correspondant à un réseau de cellules électriquement couplées. Nos simulations numériques nous ont permis de démontrer que l'échange Na+/Ca 2+ joue un rôle clef dans la régulation de la [Ca2+]i au sein d'un réseau de cellules B couplées électriquement. Il prévient, localement, l'apparition de niveaux de [ca2+]i trop élevés, potentiellement dangereux pour le métabolisme cellulaire, causés par l'hétérogénéité des paramètres cellulaires.

Повний текст джерелаNell’ambito del processo di attivazione dovuto all’interazione con l’antigene (Ag), le cellule B proliferano e iniziano un processo di maturazione terminale attraverso la formazione dei centri germinativi (GC). All’interno dei GC, a seguito dell’ipermutazione somatica delle regioni variabili del recettore delle cellule B (BCR) ed il cambiamento di isotipo delle immunoglobuline, i cloni che hanno raggiunto alta affinità per l’Ag possono andare incontro a due possibili destini: differenziamento in plasmablasti (PB) che secernono anticorpi (Ab) o in cellule B della memoria quiescenti (MBC). Il meccanismo molecolare che determina il destino delle cellule B umane durante il differenziamento tardivo in PB o MBC è poco conosciuto. Studi recenti hanno rivelato nuovi aspetti del programma trascrizionale responsabile della maturazione di cellule B in topo o in popolazioni di cellule umane, ma la disponibilità limitata di campioni e la difficoltà nell'isolamento di MBC Ag-specifiche da sangue periferico hanno reso l’analisi di questi tipi cellulari particolarmente complicata. Per questo sono stati raccolti campioni di sangue da donatori sottoposti a vaccinazione stagionale contro l’influenza. Questi campioni sono stati processati immediatamente dopo il prelievo, effettuato in corrispondenza di due particolari momenti: 8 e 22 giorni dopo la vaccinazione, rispettivamente picchi della risposta mediata da PB e da MBC . I campioni di sangue periferico sono stati usati per l’isolamento di PB, NAIVE e MBC Ag-specifiche sfruttando marcatori di superficie. Per l’analisi del profilo di espressione genica è stato ottimizzato un metodo che permette di effettuare qPCR di numerosi geni in cellule B umane isolate come singola cellula. Tale approccio è stato usato inizialmente per uno studio pilota dell’espressione di 21 geni di interesse su cellule isolate da un primo soggetto, permettendoci di discriminare cellule appartenenti alle tre diverse popolazioni. Successivamente questo protocollo è stato ottimizzato sfruttando la tecnologia del 96.96 Dynamic Array prodotto da Fluidigm, sistema che permette di effettuare RT-qPCR su 96 singole cellule per 96 geni in una singola reazione. Con questo metodo ad alta resa abbiamo analizzato 240 singole cellule appartenenti alle popolazioni di MBC Ag-specifiche, PB e NAIVE (80 cellule ciascuna) di un secondo soggetto, permettendoci di analizzare il profilo di espressione di 96 geni coinvolti nelle vie di differenziamento delle cellule B. Attraverso un’analisi statistica di raggruppamento gerarchico dei dati di espressione appartenenti a tutti i campioni processati, abbiamo riunito sotto tre gruppi diversi per espressione genica le cellule appartenenti alle tre diverse popolazioni e abbiamo identificato i geni che le caratterizzano. La Linear Discriminant Analysis, una tecnica di riduzione dimensionale supervisionata, sottolinea come i PB siano particolarmente differenti da MBC Ag-specifiche e NAIVE, che invece condividono un profilo più simile. Sfruttando diversi metodi di analisi statistica, sono stati identificati i geni significativamente espressi in maniera diversa tra le tre popolazioni. Così facendo sono stati individuati sia geni il cui ruolo nella maturazione delle cellule B è noto, sia geni conosciuti principalmente per la loro funzione in altri processi o altre fasi dello sviluppo di queste cellule (FOXP1, POU2AF1, IRF2). Inoltre abbiamo confrontato i profili di espressione delle MBC Ag-specifiche con MBC isolate da un donatore sano non vaccinato, per identificare possibili differenze nei profili di espressione di MBC recentemente attivate e MBC circolanti, lontane dall'attivazione Ag-specifica. Tale analisi ha identificato 16 geni espressi differentemente in maniera significativa, evidenziando un profilo di espressione che denota uno stato di attivazione per le MBC recentemente contattate dall'Ag. Per studiare ulteriormente l’eterogeneità delle MBC Ag-specifiche, tramite PCR abbiamo amplificato e sequenziato le regioni variabili delle catene pesanti (VH) delle immunoglobuline espresse dalle stesse cellule, ma gli studi di correlazione mostrano solo deboli associazioni tra maturazione del BCR (in termini di tasso di mutazione delle VH) e dati di espressione genica. Al contrario, è stata individuata associazione significativa tra la selezione dell'isotipo del BCR e l’espressione di due geni, in particolare l’espressione di RORα è associata alla classe IgA, mentre TBX21 all’IgG, in accordo con studi precedenti effettuati in popolazioni di cellule B murine. In conclusione, i geni identificati da questo studio come discriminanti delle MBC recentemente attivate dall'Ag potrebbero essere ulteriormente studiati in qualità di potenziali marker della risposta B alla vaccinazione in uomo.
