The phosphorus (P) availability was tested on hydromorphic soils located in theSavavalley. 480 soil profiles covering the area of31 227 hawas analysed in our study. The plant available P was determined by the Ammonium-Lactate method. The P availability in the surface layer (0–30 cm) is very low (up to 5 mg P2O5/100 g of soil) in about 30% of the tested agricultural land (9 440 ha), next 32% (9 897 ha) is in the range of a low P availability (from 5.1 to 10 mg), while only 17% (5 445 ha) has a good or very good P availability (above 20 mg). Especially high frequency of low P availability was found in vertic gley, amphygley and hypogley soils (total8 680 haor 28% of tested agricultural land).

The objective of this work was to evaluate the effectiveness of ruzigrass (Urochloaruziziensis) in enhancing soil-P availability in areas fertilized with soluble or reactive rock phosphates. The area had been cropped for five years under no-till, in a system involving soybean, triticale/black-oat, and pearl millet. Previously to the five-year cultivation period, corrective phosphorus fertilization was applied once on soil surface, at 0.0 and 80 kg ha-1 P2O5, as triple superphosphate or Arad rock phosphate. After this five-year period, plots received the same corrective P fertilization as before and ruzigrass was introduced to the cropping system in the stead of the other cover crops. Soil samples were taken (0-10 cm) after ruzigrass cultivation and subjected to soil-P fractionation. Soybean was grown thereafter without P application to seed furrow. Phosphorus availability in plots with ruzigrass was compared to the ones with spontaneous vegetation for two years. Ruzigrass cultivation increased inorganic (resin-extracted) and organic (NaHCO3) soil P, as well as P concentration in soybean leaves, regardless of the P source. However, soybean yield did not increase significantly due to ruzigrass introduction to the cropping system. Soil-P availability did not differ between soluble and reactive P sources. Ruzigrass increases soil-P availability, especially where corrective P fertilization is performed.

Soil P fractionation was used to examine changes in soil inorganic and organic P under grazed irrigated pasture in a long-term field trial at Winchmore in Mid-Canterbury. The soil P fractionation scheme used involved sequential extractions of soil with O.5M NaHCO₃ @ pH 8.5 (NaHCO₃ P), 0.1M NaOH (NaOH I P), 1M HCl (HCl P) and 0.1M NaOH (NaOH II P). The Winchmore trial comprised 5 treatments: control (no P since 1952), 376R (376 kg superphosphate ha⁻¹ yr⁻¹ 1952-1957, none since), 564R (564 kg superphosphate ha⁻¹ yr⁻¹ 1952-1957, none since) 188PA (188 kg superphosphate ha⁻¹ yr⁻¹ since 1952) and 376PA (376 kg superphosphate ha⁻¹ yr⁻¹ since 1952: Topsoil (0-7.5cm) samples taken from the different treatments in 1958, 1961, 1965, 1968, 1971, 1974 and 1977 were used in this study. Changes in soil P with time showed that significant increases in soil inorganic P occurred in the annually fertilised treatments (l88PA, 376PA). As expected, the overall increase in total soil inorganic P between 1958 and 1977 was greater in the 376PA treatment (159 µg P g⁻¹) than that in the 188PA treatment (37 µg P g⁻¹). However, the chemical forms of inorganic P which accumulated in the annually fertilised treatments changed with time. Between 1958 and 1971 most of the increases in soil inorganic P in these treatments occurred in the NaHCO₃ and NaOH I P fractions. On the other hand, increases in soil inorganic P in the annually fertilised treatments between 1971 and 1977 were found mainly in the HCl and NaOH II P fractions. These changes in soil P forms were attributed to the combined effects of lime addition in 1972 and increased amounts of sparingly soluble apatite P and iron-aluminium P in the single superphosphate applied during the 1970's. In the residual fertiliser treatments (376R, 564R) significant decreases in all of the soil inorganic P fractions (i.e. NaHCO₃ P, NaOH I P, HCl P, NaOH II p) occurred between 1958 and 1977 following the cessation of P fertiliser inputs in 1957. This was attributed to continued plant uptake of P accumulated in the soil from earlier P fertiliser additions. However, levels of inorganic P in the different soil P fractions in the residual fertiliser treatments did not decline to those in the control which indicated that some of the inorganic P accumulated in the soil from P fertiliser applied between 1952 and 1957 was present in very stable forms. In all treatments, significant increases in soil organic P occurred between 1958 and 1971. The overall increases in total soil organic P were greater in the annually fertilised treatments (70-86 µg P g⁻¹) than those in the residual fertiliser (55-64 µg P g⁻¹) and control (34 µg P g⁻¹) treatments which reflected the respective levels of pasture production in the different treatments. These increases in soil organic P were attributed to the biological conversion of native and fertiliser inorganic P to organic P in the soil via plant, animal and microbial residues. The results also showed that annual rates of soil organic P accumulation between 1958 and 1971 decreased with time which indicated that steady-state conditions with regard to net 'organic P accumulation were being reached. In the residual fertiliser treatments, soil organic P continued to increase between 1958 and 1971 while levels of soil inorganic P and pasture production declined. This indicated that organic P which accumulated in soil from P fertiliser additions was more stable and less available to plants than inorganic forms of soil P. Between 1971 and 1974 small (10-38 µg P g⁻¹) but significant decreases in total soil organic P occurred in all treatments. This was attributed to increased mineralisation of soil organic P as a result of lime (4 t ha⁻¹) applied to the trial in 1972 and also to the observed cessation of further net soil organic P accumulation after 1971. Liming also appeared to affect the chemical nature of soil organic P as shown by the large decreases in NaOH I organic P(78-88 µg P g⁻¹) and concomitant smaller increases in NaOH II organic P (53-65 µg P g⁻¹) which occurred in all treatments between 1971 and 1974. The chemical nature of soil organic P in the Winchmore long-term trial was also investigated using 31p nuclear magnetic resonance (NMR) spectroscopy and gel filtration chromatography. This involved quantitative extraction of organic P from the soil by sequential extraction with 0.1M NaOH, 0.2M aqueous acetylacetone (pH 8.3) and 0.5M NaOH following which the extracts were concentrated by ultrafiltration. Soils (0-7.5cm) taken from the control and 376PA annually fertilised treatments in 1958, 1971 and 1983 were used in this study. 31p NMR analysis showed that most (88-94%) of the organic P in the Winchmore soils was present as orthophosphate monoester P while the remainder was found as orthophosphate diester and pyrophosphate P. Orthophosphate monoester P also made up almost all of the soil organic P which accumulated in the 376PA treatment between 1958 and 1971. This indicated that soil organic P in the 376PA and control treatments was very stable. The gel filtration studies using Sephadex G-100 showed that most (61-83%) of the soil organic P in the control and 376PA treatments was present in the low molecular weight forms (<100,000 MW), although the proportion of soil organic P in high molecular weight forms (>100,000 MW) increased from 17-19% in 1958 to 38-39% in 1983. The latter was attributed to the microbial humification of organic P and indicated a shift toward more complex and possibly more stable forms of organic P in the soil with time. Assuming that the difference in soil organic P between the control and 376PA soils sampled in 1971 and 1983 represented the organic P derived from P fertiliser additions, results showed that this soil organic P was evenly distributed between the high and low molecular weight fractions. An exhaustive pot trial was used to examine the relative availability to plants of different forms of soil inorganic and organic P in long-term fertilised pasture soils. This involved growing 3 successive crops of perennial ryegrass (Lolium perenne) in 3 Lismore silt loam (Udic Ustochrept) soils which had received different amounts of P fertiliser for many years. Two of the soils were taken from the annually fertilised treatments in the Winchmore long term trial (188PA, 376PA) and the third (Fairton) was taken from a pasture which had been irrigated with meatworks effluent for over 80 years (65 kg P ha⁻¹ yr⁻¹). Each soil was subjected to 3 treatments, namely control (no nutrients added), N100 and N200. The latter treatments involved adding complete nutrient solutions with different quantities of N at rates of 100kg N ha⁻¹ (N100) and 200kg N ha⁻¹ (N200) on an area basis. The soil P fractionation scheme used was the same as that used in the Winchmore long-term trial study (i.e. NaHCO₃ P, NaOH I P, HCl P, NaOH II p). Results obtained showed that the availability to plants of different extracted inorganic P fractions, as measured by decreases in P fractions before and after 3 successive crops, followed the order: NaHCO₃ P > NaOH I P > HCl P = NaOH II P. Overall decreases in the NaHCO₃ and NaOH I inorganic P fractions were 34% and 16% respectively, while corresponding decreases in the HCl and NaOH II inorganic P fractions were small «10%) and not significant. However, a significant decrease in HCl P (16%) was observed in one soil (Fairton-N200 treatment) which was attributed to the significant decrease in soil pH (from 6.2 to 5.1) which occurred after successive cropping. Successive cropping had little or no effect on the levels of P in the different soil organic P fractions. This indicated that net soil organic P mineralisation did not contribute significantly to plant P uptake over the short-term. A short-term field experiment was also conducted to examine the effects of different soil management practices on the availability of different forms of P to plants in the long-term fertilised pasture soils. The trial was sited on selected plots of the existing annually fertilised treatments in the Winchmore long-term trial (188PA, 376PA) and comprised 5 treatments: control, 2 rates of lime (2 and 4 t ha⁻¹ ) , urea fertiliser (400kg N ha⁻¹ ) and mechanical cultivation. The above ground herbage in the uncultivated treatments was harvested on 11 occasions over a 2 year period and at each harvest topsoil (0-7.5 cm) samples were taken from all of the treatments for P analysis. The soil P fractionation scheme used in this particular trial involved sequential extractions with 0.5M NaHCO₃ @ pH 8.5 (NaHCO₃ P), 0.1M NaOH (NaOH P), ultrasonification with 0.1M NaOH (sonicate-NaOH p) and 1M HCl (HCl P). In addition, amounts of microbial P in the soils were determined. The results showed that liming resulted in small (10-21 µg P g⁻¹) though significant decreases in the NaOH soil organic P fraction in the 188PA and 376PA plots. Levels of soil microbial P were also found to be greater in the limed treatments compared with those in the controls. These results indicated that liming increased the microbial mineralisation of soil organic P in the Winchmore soils. However, pasture dry matter yields and P uptake were not significantly affected. Although urea significantly increased dry matter yields and P uptake, it did not appear to significantly affect amounts of P in the different soil P fractions. Mechanical cultivation and the subsequent fallow period (18 months) resulted in significant increases in amounts of P in the NaHCO₃ and NaOH inorganic P fractions. This was attributed to P released from the microbial decomposition of plant residues, although the absence of plants significantly reduced levels of microbial P in the cultivated soils. Practical implications of the results obtained in the present study were presented and discussed.

The P-3 Orion maritime aircraft has been the U.S. Navy's primary maritime patrol aircraft since its fleet introduction in 1962. Naval Aviation Systems Command (NAVAIR) has determined that the P-3 fleet has sufficiently aged to warrant a replacement. The replacement aircraft is currently undergoing the conceptual phase of development and it is during this period that NAVAIR is interested in evaluating the trade-off between operational availability and the associated cost to achieve this operational availability. This thesis developed a simulation tool that was used to investigate relationships that affect cost and operational availability of the new (notional) aircraft on a deployment. The simulation tool was exercised for select scenarios in order to gain insights into the value of investing funds in additional aircraft versus the value of investing funds in increased component reliability. The simulation was developed to be very flexible and extensible, enhancing its value for future analyses. Required data inputs into the simulation tool are formatted utilizing a new technology called Extensible Markup Language (XML) which facilitates use of the data in nearly all computer software packages. The model is robust in nature and can be applied to a wide variety of aircraft.

Fertilizer phosphorus (P) can become unavailable to crops due to immobilization of P in acidic soils through forming chemical bonds with iron (Fe) and aluminum (Al) amorphous oxides.  Organic chelating agents form strong bonds with metals in soil and may reduce P binding with Fe and Al.  Ethylenediamine tetraacetic acid (EDTA), hydroxyethyl ethylenediamine triacetic acid (HEEDTA), gluconic acid (GA), and citric acid (CA) were tested to determine their influence on water-soluble P (WSP), Mehlich-1 P and Mehlich-3 P in Loam and Sand soils fertilized with P and incubated for 49 days.  Soil P sorption capacity (PSC) was estimated from an oxalate extraction of Fe and Al, and chelates were applied at rates of 90 percent of the PSC.  The EDTA, HEEDTA, and CA significantly (P<0.05) reduced P sorption in the Loam and Sand when measured by WSP.  In soils without P fertilizer added, EDTA and HEEDTA significantly increased WSP, Mehlich-1, and Mehlich-3 P concentrations.  EDTA and HEEDTA were also applied at 0, 30, 60, 90, 120, and 150 percent PSC to produce a rate response curve for WSP in a second soil incubation.  With increasing chelating rate, there was a linear increase in WSP for both soils, thus indicating higher rates of chelating agents were most efficient at decreasing P sorption.
EDTA and HEEDTA were also tested in a 4-week greenhouse study for efficiency at increasing plant available P to corn (Zea mays L.) in two soils.  Phosphorus was added with and without the addition of chelating agents to the center of the pot, simulating a starter band of P.  After 4weeks, soils were analyzed for WSP, Mehlich-1, and Mehlich-3 P and corn above- and below-ground biomass was quantified and analyzed for total P concentration.  Without the presence of chelating agents, concentrations of WSP, Mehlich-1 P, Mehlich-3 P, above- and below-ground biomass, and TKP increased linearly as P fertilizer rates increased at 0, 9.6, 19.3, 28.9, and 38.5 kg P ha-1.  Decreased P sorption using chelating agents was not observed in this experiment.  However, with the results from the soil incubation, chelating agents do show potential for increasing plant available P, but the application and incorporation method needs to be further studied.

Legumes are unable to persist through post-fire succession in fynbos vegetation of the CFR unlike species in families such as Proteaceae and Restionaceae. The majority of fynbos legumes are seeders which tend to be shorter-lived than co-occurring resprouters. Seeders are likely to have a higher nutrient requirement than resprouters as they tend to invest more biomass- above ground, grow faster and produce more seed. In the oligo-trophic soils occupied by fynbos, symbiotic N2-fixation enables legumes to overcome low N availability but not low P availability. I hypothesized that: legumes are less effective at acquiring P from sparingly soluble sources compared with members of Proteaceae and Restionaceae ; legume seeders occupy soils with a higher nutrient status than resprouters. P-acquisition strategies of legumes and non-legumes were assessed.

La déficience des sols en éléments minéraux, particulièrement le phosphore (P) est une limitation majeure pour la croissance et le développement des légumineuses fixatrices d’azote. L’application des fertilisants phosphatés est une pratique traditionnelles pour satisfaire les besoins des plantes en P. Ainsi, pour tester l’efficacité d'utilisation du P pour la fixation symbiotique de l’azote (FSN) sous déficience en P, plusieurs lignées recombinantes (RILs) de haricot contrastantes dans leurs tolérance au déficit en P ont été utilisées. L’objectif principal de cette étude est d’évaluer l’aptitude de ces RILs pour l’amélioration de la fertilité phosphatée des sols déficients en P et sa relation avec la croissance et la nodulation de la légumineuse. Pour atteindre ces objectifs, des expériences ont été réalisées sous serre et en champs d'agriculteurs. Sous conditions contrôlées (serre) et non contrôlées (champs), les résultats ont montré une diminution du pH des sols associée à une augmentation du P assimilable. Une telle augmentation de la disponibilité du P a eu un effet positif sur la nodulation et la croissance de certaines des RILs testées. Aussi, l’amélioration de l’aptitude de ces RILs à fixer l’azote atmosphérique et l’élévation de la libération des protons H+ par les racines nodulées ont été quantifiés en milieu contrôlé. Ainsi la diminution du pH du sol a permis de réduire l’indisponibilité du P dans la solution du sol en le transformant en une forme directement biodisponible pour ces plantes. En outre, les résultats ont montré des différences significatives entre les différentes RILs en termes de la biomasse aérienne et nodulaire selon les sites d'observation. Nous concluons que, outre leur aptitude de fixation d’azote, l’utilisation efficace du phosphore pour sauver des engrais minéraux et de réduire les risques de pollution et pourrait améliorer la disponibilité des sols P
The deficiency of soil minerals, particularly phosphorus (P) is a major limitation for growth and development of nitrogen-fixing by legumes. The application of phosphate fertilizers is a traditional practice to meet the needs of plant P. Thus, to test the effectiveness of use of P for symbiotic nitrogen fixation (NSF) under P deficiency, several recombinant inbred lines (RILs) of beans in their contrasting tolerance to P deficiency have been used. The main objective of this study is to assess the ability of these RILs to improve the fertility of soils deficient in phosphorus (P) and its relationship with growth and nodulation of the legume. To achieve these objectives, some experiments were conducted in greenhouses and fields. Under controlled conditions (greenhouse) and uncontrolled conditions (field), the results showed decreasing in soil pH associated with Proton release H+ and P acquisition. The increasing in available P had a positive effect on nodulation and growth of some RILs tested. Also, improving the ability of these RILs to fix atmospheric nitrogen and release H+ by nodulated roots were quantified in a controlled environment. Thus the decreasing in soil pH has reduced the un-availability of P in soil solution by transforming it into a bio-available form to the plants. In addition, the results showed significant differences between RILs in biomass and nodulation as observation in sites. We conclude that, in addition to their ability to fix nitrogen, effective use of phosphorus to save mineral fertilizer and reduce the pollution risks and could improve the availability of soil P

Considerando que existem informações controversas sobre os efeitos da interação P (fósforo) Zn (zinco), tem-se a hipótese que a interação P-Zn é uma resposta ao somatório de relações nutricionais que ocorrem na planta e no solo, implicando na modificação do desenvolvimento vegetal e em respostas adaptativas do algodoeiro (Gossypium hirsutum L.). Objetivou-se com o presente estudo avaliar os mecanismos de interação P-Zn no sistema solo-planta para o algodoeiro, realizando-se uma abordagem de química do solo (fracionamento de P e extração sequencial de Zn no solo), de dinâmica de absorção destes elementos (genômica - quanto à expressão de genes relacionados à absorção de P) e de fisiologia e nutrição de plantas (quanto à determinação das atividades da fosfatase ácida e da anidrase carbônica, metabolismo oxidativo, bem como as trocas gasosas e as avaliações fotoquímicas). Para alcançar os objetivos desta proposta foram realizados quatro experimentos, três em solução nutritiva e um em solo. No experimento I foi descrita a disponibilidade de P e Zn em solo de rizosfera em resposta a doses de P e Zn em duas classes de solo cultivados com algodoeiro. Já no experimento II, descreveu-se o efeito da interação P-Zn no acúmulo de nutrientes no tecido vegetal; nas alterações no processo fotossintético - determinadas pelas trocas gasosas - e consequentes alterações no crescimento das plantas, em resposta à nutrição de P e Zn. No experimento III foi verificada a disponibilidade fisiológica de P e Zn por meio da atividade enzimática da fosfatase ácida e da anidrase carbônica, bem como descrevito o efeito da interação nas respostas de trocas gasosas, fotoquímicas e do sistema antioxidante do algodoeiro. Considering that there is controversial information about the effects of P (phosphorus) Zn (zinc) interaction, it is hypothesized that the interaction P-Zn is a response to the sum of nutritional relations that occur in the level of plant and in the soil together, as a consequence of plant development and adaptive responses of cotton. The aim of the present study was to evaluate the mechanisms of P-Zn interaction in the soil-plant system for the cotton plant (Gossypium hirsutum L.), using a soil chemistry approach (P fractionation and sequential extraction of Zn in the soil), absorption dynamics of these elements (genomics - regarding the expression of genes related to P uptake) and plant physiology and nutrition (as regards the determination of acid phosphatase activities, oxidative stress, as well as gas exange and photochemical changes). To achieve the objectives of this proposal, four experiments were carried out, three in nutrient solution and one in soil. In the experiment I, we aimed to describe the availability of P and Zn in rhizosphere soil in response to P and Zn doses in two classes of soil cultivated with cotton (Gossypium hirsutum L.). In the experiment II, the objective was to investigate the effect of the interaction P-Zn on the accumulation of nutrients in the plant tissue; on changes in the photosynthetic process - determined by gas exchange; and consequent changes in cotton growth in response to P and Zn nutrition. In the experiment III, the objective was to verify the physiological availability of P and Zn by the enzymatic activity of acid phosphatase and carbonic anhydrase, as well as to describe the effect of the interaction in the gas exchanges, photochemical and antioxidant system responses of cotton plant. Finally, in the experiment IV it was characterized the PHO1 gene (phosphate transporters in roots to xylem) in cotton and evaluated the effect of the P-Zn interaction on PHO1 expression in Gossypium hirsutum

This book examines the prospects for infallibilism about knowledge, according to which one can know that p only if one has evidence which guarantees or entails that p. In particular, it focuses on the possibility of a non-sceptical infallibilism which rejects any kind of shifty view of knowledge, whether contextualist, relativist, or subject-sensitive invariantist. The availability of a non-shifty non-sceptical infallibilism seems to depend on whether such a view can defend a generous enough conception of evidence to allow us to have the knowledge we ordinarily take ourselves to have. In particular, such an infallibilist needs to allow that our evidence extends well beyond how things seem to us in our experience and includes claims about the external world. Thus, the infallibilism which is the focus of this book is committed to a generous conception of evidence. More precisely, I argue that infallibilism is committed to the following claims about evidence and evidential support: if p is evidence, then p is true; and if one knows that p, then p is part of one’s evidence, and p is evidence for p. However, I argue that these claims about evidence and evidential support are problematic. Furthermore, I argue that fallibilism can overcome the most serious objections levelled at it, which concern closure, concessive knowledge attributions, practical reasoning, and the threshold problem. So, I conclude that epistemologists who aim to avoid both scepticism and a shifty view of knowledge should be fallibilists.

AbstractThe starting point of this work is the intention of two farmers’ initiatives to disseminate locally developed and adapted cooking and sanitation technologies to smallholder households in Karagwe District, in northwest Tanzania. These technologies include improved cooking stoves (ICSs), such as microgasifiers, and a system combining biogas digesters and burners for cooking, as well as urine-diverting dry toilets, and thermal sterilisation/pasteurisation for ecological sanitation (EcoSan). Switching to the new alternatives could lead to a higher availability of domestic residues for soil fertility management. These residues include biogas slurry from anaerobic digestion, powdery biochar from microgasifiers and sanitised human excreta from EcoSan facilities. Such recycling-driven approaches address an existing problem for many smallholders in sub-Saharan Africa, namely, the lack of soil amenders to sufficiently replenish soil nutrients and soil organic matter (SOM) in soils used for agricultural activity. This example from Tanzania systematically examines the nexus of ‘energy-sanitation-agriculture’ in smallholder farming systems. The short-term experiments demonstrated that all soil amenders that were analysed could significantly enhance crop productivity. CaSa-compost – the product of co-composting biochar with sanitised human excreta – quadrupled grain yields. The observed stimulation of crop yield and also plant nutrition is attributed to improved nutrient availability caused by a direct increase of soil pH and of plant-available phosphorus (P) in the soil. The assessment of the lasting soil implications revealed that CaSa-compost and biogas slurry both show the long-term potential to roughly double yields of maize. Corresponding nutrient requirements can be adequately compensated through residue capturing and subsistence production of soil amenders. The potential of CaSa-compost for sustainable soil fertility management is superior to that of standard compost, especially with respect to liming, replenishing soil P and restoring SOM. Biogas slurry, however, yields inferior results in all aspects when compared to compost amendments.

Specific Objectives and Related Results: 1) Determine the effect of phosphorus availability on ethylene production by roots. Test the hypothesis that phosphorus availability regulates ethylene production Clear differences were found between the two plants that were studied. In beans ethylene production is affected by P nutrition, tissue type, and stage of development. There are genotypic differences in the rate of ethylene production by various root types and in the differential in ethylene production when P treatments are compared. The acceleration in ethylene production with P deficiency increases with time. These findings support the hypothesis that ethylene production may be enhanced by phosphorus deficiency, and that the degree of enhancement varies with genotype. In tomatoes the low-P level did not enhance significantly ethylene production by the roots. Wildtype cultivars and ethylene insensitive mutants behaved similarly in that respect. 2) Characterize the effects of phosphorus availability and ethylene on the architecture of whole root systems. Test the hypothesis that both ethylene and low phosphorus availability modify root architecture. In common bean, the basal roots give rise to a major fraction of the whole root system. Unlike other laterals these roots respond to gravitropic stimulation. Their growth angle determines the proportion of the root length in the shallow layers of the soil. A correlation between ethylene production and basal root angle was found in shallow rooted but not deep-rooted genotypes, indicating that acceleration of ethylene synthesis may account for the change in basal root angle in genotypes demonstrating a plastic response to P availability. Short-time gravitropic response of the tap roots of young bean seedlings was not affected by P level in the nutrient solution. Low phosphorus specifically increases root hair length and root hair density in Arabidopsis. We tested 7 different mutants in ethylene perception and response and in each case, the response to low P was lower than that of the wild-type. The extent of reduction in P response varied among the mutants, but every mutant retained some responsiveness to changes in P concentration. The increase in root hair density was due to the increase in the number of trichoblast cell files under low P and was not mediated by ethylene. Low P did not increase the number of root hairs forming from atrichoblasts. This is in contrast to ethylene treatment, which increased the number of root hairs partly by causing root hairs to form on atrichoblasts. 3) Assess the adaptive value of root architectural plasticity in response to phosphorus availability. A simulation study indicated that genetic variation for root architecture in common bean may be related to adaptation to diverse competitive environments. The fractal dimension of tomato root system was directly correlated with P level.

In order to advance our understanding towards potential biotechnology improvement of plant performance, we studied root responses to limited P in two different plants, Arabidopsis and tomato. Arabidopsis is among the most studied model plants that allows rapid application of molecular and developmental experiments while tomato is an important crop, with application in agriculture. Using Arabidopsis we found that steroid hormones modulate the extent of root elongation in response to limited P, by controlling the accumulation of iron in the root. We also found that the availability of P and iron control the activity of the steroid hormone in the root. Finally, we revealed the genes involved in this nutrient-hormone interaction. Hence, the ferroxidase LPR1 that promotes iron accumulation in response to low P is repressed by the transcription factor BES1/BZR1. Low P inhibits the steroid hormone pathway by enhancing the accumulation of BKI1. High levels of BKI1 inhibit the activity of the steroid hormone receptor at the cell surface and iron accumulation increases inside the root, resulting in a slow growth. Together, the extent of root elongation depends on interactions between an internal cue (steroid hormone) and cues derived from the availability of P and iron in the environment. Using tomato, we found that the response of two cultivated tomato varieties (M82 and New Yorker) to limited P is distinct from that of the wild species, Solanumpennellii. This is implicated at both the levels of root development and whole plant physiology. Specifically, while the root system architecture of cultivated tomato is modulated by limited P availability, that of the wild type species remained unaffected. The wild species appears to be always behaving as if it is always in phosphate deprived conditions, despite sufficient levels of phosphate. Hyper-accumulation of metals appears to mediate this response. Together, this knowledge will be used to isolate new genes controlling plant adaptation to limited P environment.

Prolonged application may result in P accumulation up to levels that impair plant nutrition, increase P loss, and promote eutrophication in downstream waters. This study aims to shed light on the RW- and BS-P forms in soils and to follow the processes that determine P reactivity, solubility, availability, and loss in RW and BS treated soils. The Technion group used sequential P extraction combined with measuring stable oxygen isotopic composition in phosphate (δ18OP) and with 31P-NMR studies to probe P speciation and transformations in soils irrigated with RW or fresh water (FW). The application of the δ18OP method to probe inorganic P (Pi) speciation and transformations in soils was developed through collaboration between the Technion and the UCSC groups. The method was used to trace Pi in water-, NaHCO3-, NaOH-, and HCl- P fractions in a calcareous clay soil (Acre, Israel) irrigated with RW or FW. The δ18OP signature changes during a month of incubation indicated biogeochemical processes. The water soluble Pi (WSPi) was affected by enzymatic activity yielding isotopic equilibrium with the water molecules in the soil solution. Further it interacted rapidly with the NaHCO3-Pi. The more stable Pi pools also exhibited isotopic alterations in the first two weeks after P application, likely related to microbial activity. Isotopic depletion which could result from organic P (PO) mineralization was followed by enrichment which may result from biologic discrimination in the uptake. Similar transformations were observed in both soils although transformations related to biological activity were more pronounced in the soil treated with RW. Specific P compounds were identified by the Technion group, using solution-state 31P-NMR in wastewater and in soil P extracts from Acre soils irrigated by RW and FW. Few identified PO compounds (e.g., D-glucose-6-phosphate) indicated coupled transformations of P and C in the wastewater. The RW soil retained higher P content, mainly in the labile fractions, but lower labile PO, than the FW soil; this and the fact that P species in the various soil extracts of the RW soil appear independent of P species in the RW are attributed to enhanced biological activity and P recycling in the RW soil. Consistent with that, both soils retained very similar P species in the soil pools. The HUJ group tested P stabilization to maximize the environmental safe application rates and the agronomic beneficial use of BS. Sequential P extraction indicated that the most reactive BS-P forms: WSP, membrane-P, and NaHCO3-P, were effectively stabilized by ferrous sulfate (FeSul), calcium oxide (CaO), or aluminum sulfate (alum). After applying the stabilized BS, or fresh BS (FBS), FBS compost (BSC), or P fertilizer (KH2PO4) to an alluvial soil, P availability was probed during 100 days of incubation. A plant-based bioassay indicated that P availability followed the order KH2PO4 >> alum-BS > BSC ≥ FBS > CaO-BS >> FeSul-BS. The WSPi concentration in soil increased following FBS or BSC application, and P mineralization further increased it during incubation. In contrast, the chemically stabilized BS reduced WSPi concentrations relative to the untreated soil. It was concluded that the chemically stabilized BS effectively controlled WSPi in the soil while still supplying P to support plant growth. Using the sequential extraction procedure the persistence of P availability in BS treated soils was shown to be of a long-term nature. 15 years after the last BS application to MN soils that were annually amended for 20 years by heavy rates of BS, about 25% of the added BS-P was found in the labile fractions. The UMN group further probed soil-P speciation in these soils by bulk and micro X-ray absorption near edge structure (XANES). This newly developed method was shown to be a powerful tool for P speciation in soils. In a control soil (no BS added), 54% of the total P was PO and it was mostly identified as phytic acid; 15% was identified as brushite and 26% as strengite. A corn crop BS amended soil included mostly P-Fe-peat complex, variscite and Al-P-peat complex but no Ca-P while in a BS-grass soil octacalcium phosphate was identified and o-phosphorylethanolamine or phytic acid was shown to dominate the PO fraction that impair plant nutrition, increase P loss, and promote eutrophication in downstream waters. This study aims to shed light on the RW- and BS-P forms in soils and to follow the processes that determine P reactivity, solubility, availability, and loss in RW and BS treated soils. The Technion group used sequential P extraction combined with measuring stable oxygen isotopic composition in phosphate (δ18OP) and with 31P-NMR studies to probe P speciation and transformations in soils irrigated with RW or fresh water (FW). The application of the δ18OP method to probe inorganic P (Pi) speciation and transformations in soils was developed through collaboration between the Technion and the UCSC groups. The method was used to trace Pi in water-, NaHCO3-, NaOH-, and HCl- P fractions in a calcareous clay soil (Acre, Israel) irrigated with RW or FW. The δ18OP signature changes during a month of incubation indicated biogeochemical processes. The water soluble Pi (WSPi) was affected by enzymatic activity yielding isotopic equilibrium with the water molecules in the soil solution. Further it interacted rapidly with the NaHCO3-Pi. The more stable Pi pools also exhibited isotopic alterations in the first two weeks after P application, likely related to microbial activity. Isotopic depletion which could result from organic P (PO) mineralization was followed by enrichment which may result from biologic discrimination in the uptake. Similar transformations were observed in both soils although transformations related to biological activity were more pronounced in the soil treated with RW. Background: In recent years a major expansion of electricity production by coal combustion has taken place in Israel, the United States and the rest of the world. As a result, a large amount of CCPs are created that include bottom ash, fly ash, flue gas desulfurization (FGD) gypsum and other combustion products. In Israel 100,000 tons of fly ash (10% of total CCPs) are produced each year and in the US a total of 123 million tons of CCPs are produced each year with 71 million tons of fly ash, 18 million tons of bottom ash and 12 million tons of FGD gypsum. Many new scrubbers are being installed and will come on-line in the next 2 to 10 years and this will greatly expand the amount of FGD gypsum. One of the main substrates used in Israel for growth media is volcanic ash (scoria; tuff). The resemblance of bottom coal ash to tuff led us to the assumption that it is possible to substitute tuff with bottom ash. Similarly, bottom ash and FGD gypsum were considered excellent materials for creating growth mixes for agricultural and nursery production uses. In the experiments conducted, bottom ash was studied in Israel and bottom ash, fly ash and FGD gypsum was studied in the US. Major Achievements: In the US, mixes were tested that combine bottom ash, organic amendments (i.e. composts) and FGD gypsum and the best mixes supported growth of tomato, wheat and marigolds that were equal to or better than two commercial mixes used as a positive control. Plants grown on bottom ash in Israel also performed very well and microelements and radionuclides analyses conducted on plants grown on bottom coal ash proved it is safe to ingest the edible organs of these plants. According to these findings, approval to use bottom coal ash for growing vegetables and fruits was issued by the Israeli Ministry of Health. Implications: Bottom coal ash is a suitable substitute for volcanic ash (scoria; tuff) obtained from the Golan Heights as a growth medium in Israel. Recycling of bottom coal ash is more environmentally sustainable than mining a nonrenewable resource. The use of mixes containing CCPs was shown feasible for growing plants in the United States and is now being evaluated at a commercial nursery where red sunset maple trees are being grown in a pot-in-pot production system. New potential for engineering chloroplasts to express novel traits has stimulated research into relevant techniques and genetic processes, including plastid transformation and gene regulation. This proposal continued our long time BARD-funded collaboration research into mechanisms that influence chloroplast RNA accumulation, and thus gene expression. Previous work on cpRNA catabolism has elucidated a pathway initiated by endonucleolytic cleavage, followed by polyadenylation and exonucleolytic degradation. A major player in this process is the nucleus-encoded exoribonuclease/polymerasepolynucleotidephoshorylase (PNPase). Biochemical characterization of PNPase has revealed a modular structure that controls its RNA synthesis and degradation activities, which in turn are responsive to the phosphate (P) concentration. However, the in vivo roles and regulation of these opposing activities are poorly understood. The objectives of this project were to define how PNPase is controlled by P and nucleotides, using in vitro assays; To make use of both null and site-directed mutations in the PNPgene to study why PNPase appears to be required for photosynthesis; and to analyze plants defective in P sensing for effects on chloroplast gene expression, to address one aspect of how adaptation is integrated throughout the organism. Our new data show that P deprivation reduces cpRNA decay rates in vivo in a PNPasedependent manner, suggesting that PNPase is part of an organismal P limitation response chain that includes the chloroplast. As an essential component of macromolecules, P availability often limits plant growth, and particularly impacts photosynthesis. Although plants have evolved sophisticated scavenging mechanisms these have yet to be exploited, hence P is the most important fertilizer input for crop plants. cpRNA metabolism was found to be regulated by P concentrations through a global sensing pathway in which PNPase is a central player. In addition several additional discoveries were revealed during the course of this research program. The human mitochondria PNPase was explored and a possible role in maintaining mitochondria homeostasis was outlined. As polyadenylation was found to be a common mechanism that is present in almost all organisms, the few examples of organisms that metabolize RNA with no polyadenylation were analyzed and described. Our experiment shaded new insights into how nutrient stress signals affect yield by influencing photosynthesis and other chloroplast processes, suggesting strategies for improving agriculturally-important plants or plants with novel introduced traits. Our studies illuminated the poorly understood linkage of chloroplast gene expression to environmental influences other than light quality and quantity. Finely, our finding significantly advanced the knowledge about polyadenylation of RNA, the evolution of this process and its function in different organisms including bacteria, archaea, chloroplasts, mitochondria and the eukaryotic cell. These new insights into chloroplast gene regulation will ultimately support plant improvement for agriculture

The antibotulinal baceriocins, propionicin PLG-1 and jenseniin G., were the first to be identified, purified and characterized for the dairy propionibaceria and are produced by Propionibacterium thoenii P127 and P. thoenii/jensenii P126, respectively. Objectives of this project were to (a) produce polyclonal antibodies for detection, comparison and monitoring of propionicin PLG-1; (b) identify, clone and characterize the propionicin PLG-1 (plg-1) and jenseniin G (jnG) genes; and (3) develop gene transfer systems for dairy propionibacteria using them as models. Polyclonal antibodies for detection, comparison and monitoring of propionicin PLG-1 were produced in rabbits. Anti-PLG-1 antiserum had high titers (256,000 to 512,000), neutralized PLG-1 activity, and detected purified PLG-1 at 0.10 mg/ml (indirect ELISA) and 0.033 mg/ml (competitive indirect ELISA). Thirty-nine of 158 strains (most P. thoenii or P. jensenii) yielded cross-reacting material; four strains of P. thoenii, including two previously unidentified bacteriocin producers, showed biological activity. Eight propionicin-negative P127 mutants produced neither ELISA response nor biological activity. Western blot analyses of supernates detected a PLG-1 band at 9.1 kDa and two additional protein bands with apparent molecular weights of 16.2 and 27.5 kDa. PLG-1 polyclonal antibodies were used for detection of jenseniin G. PLG-1 antibodies neutralized jenseniin G activity and detected a jenseniin G-sized, 3.5 kDa peptide. Preliminary immunoprecipitation of crude preparations with PLG-1 antibodies yielded three proteins including an active 3-4 kDa band. Propionicin PLG-1 antibodies were used to screen a P. jensenii/thoenii P126 genomic expression library. Complete sequencing of a cloned insert identified by PLG-1 antibodies revealed a putative response regulator, transport protein, transmembrane protein and an open reading frame (ORF) potentially encoding jenseniin G. PCR cloning of the putative plg-1 gene yielded a 1,100 bp fragment with a 355 bp ORF encoding 118 amino acids; the deduced N-terminus was similar to the known PLG-1 N-terminus. The 118 amino acid sequence deduced from the putative plg-1 gene was larger than PLG-1 possibly due to post-translational processing. The product of the putative plg-1 gene had a calculated molecular weight of 12.8 kDa, a pI of 11.7, 14 negatively charged residues (Asp+Glu) and 24 positively charged residues (Arg+Lys). The putative plg-1 gene was expressed as an inducible fusion protein with a six-histidine residue tag. Metal affinity chromatography of the fused protein yielded a homogeneous product. The fused purified protein sequence matched the deduced putative plg-1 gene sequence. The data preliminarily suggest that both the plg-1 and jnG genes have been identified and cloned. Demonstrating that antibodies can be produced for propionicin PLG-1 and that those antibodies can be used to detect, monitor and compare activity throughout growth and purification was an important step towards monitoring PLG-1 concentrations in food systems. The unexpected but fortunate cross-reactivity of PLG-1 antibodies with jenseniin G led to selective recovery of jenseniin G by immunoprecipitation. Further refinement of this separation technique could lead to powerful affinity methods for rapid, specific separation of the two bacteriocins and thus facilitate their availability for industrial or pharmaceutical uses. Preliminary identification of genes encoding the two dairy propionibacteria bacteriocins must be confirmed; further analysis will provide means for understanding how they work, for increasing their production and for manipulating the peptides to increase their target species. Further development of these systems would contribute to basic knowledge about dairy propionibacteria and has potential for improving other industrially significant characteristics.

Research objectives were: 1) To study the effect of diet energy density, level of exercise, thermal conditions and reproductive state on cardiovascular function as it relates to oxygen (O2) mobilization. 2) To validate the use of heart rate (HR) to predict energy expenditure (EE) of ruminants, by measuring and calculating the energy balance components at different productive and reproductive states. 3) To validate the use of HR to identify changes in the metabolizable energy (ME) and ME intake (MEI) of grazing ruminants. Background: The development of an effective method for the measurement of EE is essential for understanding the management of both grazing and confined feedlot animals. The use of HR as a method of estimating EE in free-ranging large ruminants has been limited by the availability of suitable field monitoring equipment and by the absence of empirical understanding of the relationship between cardiac function and metabolic rate. Recent developments in microelectronics provide a good opportunity to use small HR devices to monitor free-range animals. The estimation of O2 uptake (VO2) of animals from their HR has to be based upon a consistent relationship between HR and VO2. The question as to whether, or to what extent, feeding level, environmental conditions and reproductive state affect such a relationship is still unanswered. Studies on the basic physiology of O2 mobilization (in USA) and field and feedlot-based investigations (in Israel) covered a , variety of conditions in order to investigate the possibilities of using HR to estimate EE. In USA the physiological studies conducted using animals with implanted flow probes, show that: I) although stroke volume decreases during intense exercise, VO2 per one heart beat per kgBW0.75 (O2 Pulse, O2P) actually increases and measurement of EE by HR and constant O2P may underestimate VO2unless the slope of the regression relating to heart rate and VO2 is also determined, 2) alterations in VO2 associated with the level of feeding and the effects of feeding itself have no effect on O2P, 3) both pregnancy and lactation may increase blood volume, especially lactation; but they have no effect on O2P, 4) ambient temperature in the range of 15 to 25°C in the resting animal has no effect on O2P, and 5) severe heat stress, induced by exercise, elevates body temperature to a sufficient extent that 14% of cardiac output may be required to dissipate the heat generated by exercise rather than for O2 transport. However, this is an unusual situation and its affect on EE estimation in a freely grazing animal, especially when heart rate is monitored over several days, is minor. In Israel three experiments were carried out in the hot summer to define changes in O2P attributable to changes in the time of day or In the heat load. The animals used were lambs and young calves in the growing phase and highly yielding dairy cows. In the growing animals the time of day, or the heat load, affected HR and VO2, but had no effect on O2P. On the other hand, the O2P measured in lactating cows was affected by the heat load; this is similar to the finding in the USA study of sheep. Energy balance trials were conducted to compare MEI recovery by the retained energy (RE) and by EE as measured by HR and O2P. The trial hypothesis was that if HR reliably estimated EE, the MEI proportion to (EE+RE) would not be significantly different from 1.0. Beef cows along a year of their reproductive cycle and growing lambs were used. The MEI recoveries of both trials were not significantly different from 1.0, 1.062+0.026 and 0.957+0.024 respectively. The cows' reproductive state did not affect the O2P, which is similar to the finding in the USA study. Pasture ME content and animal variables such as HR, VO2, O2P and EE of cows on grazing and in confinement were measured throughout three years under twenty-nine combinations of herbage quality and cows' reproductive state. In twelve grazing states, individual faecal output (FO) was measured and MEI was calculated. Regression analyses of the EE and RE dependent on MEI were highly significant (P<0.001). The predicted values of EE at zero intake (78 kcal/kgBW0.75), were similar to those estimated by NRC (1984). The EE at maintenance condition of the grazing cows (EE=MEI, 125 kcal/kgBW0.75) which are in the range of 96.1 to 125.5 as presented by NRC (1996 pp 6-7) for beef cows. Average daily HR and EE were significantly increased by lactation, P<0.001 and P<0.02 respectively. Grazing ME significantly increased HR and EE, P<0.001 and P<0.00l respectively. In contradiction to the finding in confined ewes and cows, the O2P of the grazing cows was significantly affected by the combined treatments (P<0.00l ); this effect was significantly related to the diet ME (P<0.00l ) and consequently to the MEI (P<0.03). Grazing significantly increased O2P compared to confinement. So, when EE of grazing animals during a certain season of the year is estimated using the HR method, the O2P must be re measured whenever grazing ME changes. A high correlation (R2>0.96) of group average EE and of HR dependency on MEI was also found in confined cows, which were fed six different diets and in growing lambs on three diets. In conclusion, the studies conducted in USA and in Israel investigated in depth the physiological mechanisms of cardiovascular and O2 mobilization, and went on to investigate a wide variety of ruminant species, ages, reproductive states, diets ME, time of intake and time of day, and compared these variables under grazing and confinement conditions. From these combined studies we can conclude that EE can be determined from HR measurements during several days, multiplied by O2P measured over a short period of time (10-15 min). The study showed that RE could be determined during the growing phase without slaughtering. In the near future the development microelectronic devices will enable wide use of the HR method to determine EE and energy balance. It will open new scopes of physiological and agricultural research with minimizes strain on animals. The method also has a high potential as a tool for herd management.

The “Novel streaming potential (SP) and thermal sensor techniques for monitoring water and nutrient fluxes in the vadose zone” project ended Oct. 30, 2015, after an extension to complete travel and intellectual exchange of ideas and sensors. A significant component of this project was the development and testing of the Penta-needle Heat Pulse Probe (PHPP) in addition to testing of the streaming potential concept, both aimed at soil water flux determination. The PHPP was successfully completed and shown to provide soil water flux estimates down to 1 cm day⁻¹ with altered heat input and timing as well as use of larger heater needles. The PHPP was developed by Scott B. Jones at Utah State University with a plan to share sensors with Shmulik P. Friedman, the ARO collaborator. Delays in completion of the PHPP resulted in limited testing at USU and a late delivery of sensors (Sept. 2015) to Dr. Friedman. Two key aspects of the subsurface water flux sensor development that delayed the availability of the PHPP sensors were the addition of integrated electrical conductivity measurements (available in February 2015) and resolution of bugs in the microcontroller firmware (problems resolved in April 2015). Furthermore, testing of the streaming potential method with a wide variety of non-polarizable electrodes at both institutions was not successful as a practical measurement tool for water flux due to numerous sources of interference and the M.S. student in Israel terminated his program prematurely for personal reasons. In spite of these challenges, the project funded several undergraduate students building sensors and several master’s students and postdocs participating in theory and sensor development and testing. Four peer-reviewed journal articles have been published or submitted to date and six oral/poster presentations were also delivered by various authors associated with this project. We intend to continue testing the "new generation" PHPP probes at both USU and at the ARO resulting in several additional publications coming from this follow-on research. Furthermore, Jones is presently awaiting word on an internal grant application for commercialization of the PHPP at USU.

Improving plant cell wall (fiber) degradation remains one of the highest priority research goals for all ruminant enterprises dependent on forages, hay, silage, or other fibrous byproducts as energy sources, because it governs the provision of energy-yielding nutrients to the host animal. Although the predominant species of microbes responsible for ruminal fiber degradation are culturable, the enzymology and genetics underpinning the process are poorly defined. In that context, there were two broad objectives for this proposal. The first objective was to identify the key cellulosomal components in Ruminococcus albus and to characterize their structural features as well as regulation of their expression, in response to polysaccharides and (or) P AA/PPA. The second objective was to evaluate the similarities in the structure and architecture of cellulosomal components between R. albus and other ruminal and non-ruminal cellulolytic bacteria. The cooperation among the investigators resulted in the identification of two glycoside hydrolases rate-limiting to cellulose degradation by Ruminococcus albus (Cel48A and CeI9B) and our demonstration that these enzymes possess a novel modular architecture specific to this bacterium (Devillard et al. 2004). We have now shown that the novel X-domains in Cel48A and Cel9B represent a new type of carbohydrate binding module, and the enzymes are not part of a ceiluiosome-like complex (CBM37, Xu et al. 2004). Both Cel48A and Cel9B are conditionally expressed in response to P AA/PPA, explaining why cellulose degradation in this bacterium is affected by the availability of these compounds, but additional studies have shown for the first time that neither PAA nor PPA influence xylan degradation by R. albus (Reveneau et al. 2003). Additionally, the R. albus genome sequencing project, led by the PI. Morrison, has supported our identification of many dockerin containing proteins. However, the identification of gene(s) encoding a scaffoldin has been more elusive, and recombinant proteins encoding candidate cohesin modules are now being used in Israel to verify the existence of dockerin-cohesin interactions and cellulosome production by R. albus. The Israeli partners have also conducted virtually all of the studies specific to the second Objective of the proposal. Comparative blotting studies have been conducted using specific antibodies prepare against purified recombinant cohesins and X-domains, derived from cellulosomal scaffoldins of R. flavefaciens 17, a Clostridium thermocellum mutant-preabsorbed antibody preparation, or against CbpC (fimbrial protein) of R. albus 8. The data also suggest that additional cellulolytic bacteria including Fibrobacter succinogenes S85, F. intestinalis DR7 and Butyrivibrio fibrisolvens Dl may also employ cellulosomal modules similar to those of R. flavefaciens 17. Collectively, our work during the grant period has shown that R. albus and other ruminal bacteria employ several novel mechanisms for their adhesion to plant surfaces, and produce both cellulosomal and non-cellulosomal forms of glycoside hydrolases underpinning plant fiber degradation. These improvements in our mechanistic understanding of bacterial adhesion and enzyme regulation now offers the potential to: i) optimize ruminal and hindgut conditions by dietary additives to maximize fiber degradation (e.g. by the addition of select enzymes or PAA/PPA); ii) identify plant-borne influences on adhesion and fiber-degradation, which might be overcome (or improved) by conventional breeding or transgenic plant technologies and; iii) engineer or select microbes with improved adhesion capabilities, cellulosome assembly and fiber degradation. The potential benefits associated with this research proposal are likely to be realized in the medium term (5-10 years).
