β-thalassemia is one of the most frequently-occurring disorders due to altered hemoglobin protein synthesis (Modell & Darlison, 2008). In this disease, the β-globin gene is mutated, causing severe anemia and ineffective erythropoiesis. Patients can present with a number of life-threatening co-morbidities, including iron overload, extramedullary erythropoiesis, hypogonadism and osteoporosis (Origa, 2016). Current standard of care for patients with severe anemia involves blood transfusion and iron chelation. Allogeneic bone marrow transplant is the only curative option, but is constrained by the availability of matching donors and the occurrence of graft-versus-host disease. (Breda et al., 2016) As this is a monogenic disease, it makes it an attractive setting for genetic therapy at the DNA and RNA level. Currently, there are a few clinical trials and preclinical tests underway to evaluate the curative potential of lentiviral vectors to correct the mutated β-globin or add back a functional copy of β-globin (Jorge Mansilla-Soto, Isabelle Riviere, Farid Boulad, 2016, Negre et al., 2016) One of the processes that affects β-globin synthesis in β-thalassemia is occurrence of aberrant splicing. IVS-2-745 is a splicing mutation that occurs in intron 2 of the β-globin gene. It creates an aberrantly spliced form that incorporates an extra exon and leads to a premature stop codon. Here we report novel uniform 2’-O-methoxyethyl (2’-MOE) splice switching oligos (SSOs) to reverse this aberrant splicing. Lead 2’-MOE SSOs were generated to redirect splicing in the IVS-2-745 pre-mRNA. With these lead SSOs we have demonstrated aberrant to wild type splice switching. This switching led to an increase from 3-6% to up to 80% adult hemoglobin (HbA) in erythroid cells from thalassemic patients. Furthermore, we demonstrated a restoration of the balance between β-like- and α-globin chains, and up to an 87% reduction in toxic α-heme aggregates. While next examining the potential benefit of 2’MOE SSOs in a sickle-thalassemic phenotypic setting, we found reduced HbS synthesis and sickle cell formation due to HbA induction, as a result of 2’MOE SSO treatment. In summary, 2’MOE-SSOs are a promising therapy for splicing forms of β-thalassemia. Their ability to functionally modulate the thalassemia and sickle cell anemia phenotype by correcting the underlying defect offers a pharmacological treatment that is both direct and specific

Se realizó un ensayo con el objetivo de determinar el efecto del riego deficitario controlado (RDC) en variedades de olivo para aceite, Arbequina, Coratina, Frantoio y Coratina. Los tratamientos de déficit hídrico se iniciaron desde el momento de pinta hasta cosecha para cada una de las variedades. El tratamiento control tuvo un 100% de reposición de la evapotranspiración del cultivo (ETc) durante toda la temporada; un segundo tratamiento (T2) tuvo un 50% de reposición de la ETc desde el período de pinta hasta cosecha, duplicando el tiempo de espera entre los riegos y un tercer tratamiento (T3) con un 50% de reposición de la ETc desde el período de pinta hasta cosecha, disminuyendo a la mitad la precipitación de cada riego. El potencial hídrico xilemático se vió afectado en todas las variedades, excepto en Frantoio, y sólo antes de que comenzaran las precipitaciones, en donde, el tratamiento 2 presentó siempre los valores más bajos. Tanto en la var. Leccino como en la var. Frantoio, el contenido de agua en el fruto se vió reducido al momento de la cosecha, en donde los valores más bajos se obtuvieron en el tratamiento 2, alrededor de un 7% menos que el tratamiento control para el caso de Leccino. Esto trajo como consecuencia que el peso del fruto disminuyera y a su vez la concentración de aceite en éste aumentara. Al momento del procesamiento de los frutos en la almazara, se obtuvo mayores niveles de aceite en los tratamientos con RDC, esto debido básicamente a que los frutos al tener un menor contenido de agua, el proceso de extracción de aceite es más eficiente. Con respecto a las variedades, Arbequina y Coratina, en general, no presentaron diferencias en las distintas variables analizadas al momento de la cosecha, dado que los árboles no se encontraban en una situación de déficit hídrico, producto de las abundantes precipitaciones ocurridas ese otoño.
With the goal of determining the effect of deficit irrigation on olive trees, for oil production, var. Arbequina, Coratina, Frantoio and Leccino, treatments of hydric deficit between veraison and harvest were established. Treatments were; T1, control, with a water reposition of 100% of the crop evapotranspiration (Etc) during the whole season; T2, 50% of the Etc reached by doubling the delay between irrigations compared to the control frequency and; T3, 50% of Etc reposition reached by applying half the irrigation amount compared to the control, with the same frequency. The hidric potential xilematic met affected in all the varieties, except in Frantoio, and only before they began the rainfalls, where, the treatment 2 presented always the lowest values. The var. Leccino as well as var. Frantoio, reduced the fruit water content at harvest, as result of the controlled deficit irrigation. The lowest water content values were reached by treatment T2, with about 7% less water than T1, for var. Leccino, and about 4% less water content in the case of var. Frantoio, with no significant oil content reduction in both cases. Furthermore, as consequence of the controlled deficit irrigation, the weight of fruit decreased and the oil concentration increased. At the moment of the fruit processing in the oil mill, both cultivars showed improved efficiency in terms of oil extraction, and higher oil levels in the treatments with controlled deficit irrigation, basically due to minor fruit water content. With respect to var. Arbequina and var. Coratina, no significant differences for the analyzed variables was seen, do to the lack of hydric deficit close to harvest, given the effect of early rainfall events during autumn.

El plan de negocio consiste en una productora de eventos Familiares y Profesionales, que resuelva las necesidades de celebración en escenarios naturales y que ponga a disposición servicios adicionales. Esto permitirá generar una experiencia superior a los clientes y usuarios. El lugar elegido es la provincia de Talagante, por su oferta de entornos naturales y cercanía a las comunas del Gran Santiago y la provincia de Melipilla, que es el segmento geográfico definido. De esta forma, se firmará un contrato a cinco años con el centro de Eventos Olivos del Monte, ubicado en Talagante. Zona de alta demanda por sus entornos y servicios. Los entornos naturales, marcan una tendencia bien definida en el estudio de mercado y contar con un centro de eventos que cumpla con estas características es fundamental. Por ello, se seleccionó Los Olivos del Monte, con amplios y hermosos escenarios, donde se desarrollará una ventaja competitiva basada en actividades internas como marketing, adquisiciones, servicio de atención y seguimiento al cliente, hermoseamiento y mantención de las características distintivas del recurso elegido. Los Olivos Producciones, contará con un recurso único en la zona y que es el contar con un centro de eventos donde salón y carpa están inmersos en un bosque, esto hará sentir a los invitados que están experimentando de una celebración excepcional, además cuenta con hermosos escenarios como parques, parras, estacionamientos y piscina. El mercado de los matrimonios, como ejemplo de eventos familiares, asciende a $35.868 millones y el mercado de las capacitaciones en administración, eventos profesionales, a $27.704 millones. Por los montos involucrados y la alta atomización de empresas que compiten se estima una participación de mercado de 0,77% de los eventos familiares y un 0,19% de los eventos profesionales, al cierre del primer año. Para llegar a un 1,51% de participación en el mercado familiar y 0,38% del mercado de profesionales al quinto año, que es cuando se deberá tomar la decisión de continuidad o salida. Los socios aportaran capital y solicitarán financiamiento bancario, por un total de $74,8 millones. Así, el 100% de las utilidades obtenidas serán repartidas a los socios del proyecto, luego de cada ejercicio. Con esta inversión se espera obtener un VAN de $249 millones, una TIR de 72%, considerando un horizonte de evaluación de cinco años y una tasa de descuento de 13,13%. Con indicadores como EBITDA de un 4,4% al término del primer año y un 36% al quinto año, el proyecto tiene un Payback de 2,1 años desde el inicio de las operaciones. Resultando atractivo para los inversionistas.

El siguiente trabajo de investigación parte como respuesta a la falta de residencias universitarias en Lima, ciudad más importante del país, que a pesar de la gran oferta de estudiantes universitarios que se ha venido dando durante los últimos años, solo cuenta con una residencia universitaria propiamente dicha. Según cifras, del total de becas que se otorga todos los años en el Programa Nacional de Becas (PRONABEC), el 85% lo obtienen estudiantes del interior del país, que en su gran mayoría vienen a la capital a estudiar en universidades tanto públicas como privadas. Ante esta situación, los estudiantes optan por el alquiler de casas que no cuentan con espacios comunes donde puedan desarrollarse académica y socialmente. Es por ello que se plantea una residencia destinada exclusivamente para alumnos universitarios que se encuentran estudiando en alguna institución cercana. Por otro lado, el proyecto al estar ubicado estratégicamente en el distrito de Los Olivos, donde existen varios polos educativos y oportunidades de negocio, estará compuesto por tres tipologías: residencia, oficinas coworking y comercio local, ya que no solo se busca resolver el tema de estadía, sino que ayude a que los jóvenes tengan un mejor desarrollo profesional durante y después de su etapa universitaria, que tengan mayor conexión con su entorno directo y su comunidad, creando patios abiertos acompañados con áreas verdes de esparcimiento donde puedan intercambiar diferentes costumbres que cada uno tiene del lugar donde nacieron, logrando una interculturalidad entre los usuarios.
The research work below begins in response to the lack of student housing in Lima, the most important city in the country, which despite the great offer of university students that has been given in recent years, only has one university residence proper. According to statistics, of the total scholarships awarded every year in the National Scholarship Program (PRONABEC), 85% are obtained by students from country, who mostly come to the capital to study at both, public or private universities. Faced with this situation, students choose to rent houses that do not have common spaces where they can develop academically and socially. That is why a residence is proposed exclusively for young university students who are studying at a nearby institution. On the other hand, the project being strategically located in the district of Los Olivos, where there are several educational areas and business opportunities, because of that the project consists in three typologies: residence, coworking offices and local commerce, because not only do you seek to resolve the issue of stay, it also helps to young people have better professional development during and after their university stage , that have greater connection with their direct environment and their community, creating open plazas with green areas of recreation where they can exchange different customs that each has of the place where they were born, achieving an interculturality among users.

El Proyecto busca principalmente responder a la falta de una biblioteca pública en el distrito de los Olivos que abastezca de manera suficiente a las necesidades de los usuarios que habitan en él. Actualmente la biblioteca pública del los Olivos no cumple con abastecer la demanda de la población respecto a los estándares necesarios propuestos por la IFLA (Federación Internacional de Asociaciones de Bibliotecarios y Bibliotecas) de número de libros por habitante en el distrito. Además tiene como objetivo romper la brecha entre la biblioteca y la sociedad, cambiando la imagen de la biblioteca como un edificio elitista estancado en el tiempo a uno abierto a los usuarios, a la ciudad, al entorno natural que constantemente se actualice a los cambios tecnológicos. Siendo los Olivos un distrito joven y dinámico, marcado por su rápido crecimiento a partir de las migraciones ocurridas en el siglo XX, se caracteriza por tener una diversidad social y cultural donde coexisten habitantes de distintas culturas, costumbres y necesidades. Esta pluralidad tiene como consecuencia roces dentro de los mismos habitantes que desencadena mayormente en problemas como la marginación social, cultural y de genero. Con este proyecto se generará un punto de encuentro entre ciudadanos a modo de catalizador social jerarquizando la calidad espacial mediante espacios flexibles, dinámicos e integrados al entorno urbano y natural, promoviendo un espacio de descubrimiento, aprendizaje y integración.
The Project seeks mainly to respond to the lack of a public library in Los Olivo’s district in order to supply the needs of the users who live there. At the present time Los Olivo’s public library does not satisfy the population’s demand of needs in relationship to the standards proposed by the IFLA (International Federation of Associations of Librarians and Libraries) on the number of books per inhabitant in the district. It also aims to break the gap between the library and society, by changing the image of the library as an elitist building stuck in time to one open to users, to the city, and natural environment that is constantly updated to technological changes. Being Los Olivos a young and dynamic district, marked by its rapid growth from migrations in the twentieth century, it is characterized by having a social and cultural diversity where people of different cultures, customs and needs coexist. This plurality has a consequence: frictions within the same inhabitants that triggers problems such as social, cultural and gender exclusion. This project will generate a meeting point between citizens and act as a social catalyst, prioritizing spatial quality through flexible, dynamic spaces integrated into the urban and natural environment, promoting a space of discovery, learning and integration.

El objetivo de la presente tesis es evaluar la factibilidad de poner en marcha un Spa Urbano en el distrito de Los Olivos, denominado “Aqua Spa”, que brinde al visitante una experiencia vivencial única: “La cultura del Slow Life” a través de actividades de relajación como tratamientos corporales, circuitos de aguas, etc., que aprovechan los recursos naturales, como el agua y materia natural para los tratamientos del Spa. La propuesta de valor está basada principalmente en ofrecer un servicio de calidad, y una infraestructura Premium armónica evocando elementos naturales, como caídas de agua, espacios diseñados especialmente para desconectarse del día a día y mediante la arquitectura y el diseño pueda llevar al cliente a un nivel de relax y reflexión. El proyecto será un edificio de 3 niveles y estará ubicado en Los Olivos en zona comercial accesible, cabe destacar que su diseño contempla una preparación acústica para separar a las habitaciones de los tratamientos del mundo de afuera. “Aqua Spa” se posicionará como el primer spa dirigido a las familias de Lima Norte en particular a las que habitan en la zona de Los olivos como un servicio de lujo a precios competitivos con los centros existentes. La evaluación económica y financiera de los flujos de caja del proyecto demostró su factibilidad en este aspecto, sustentada en un VAN Económico de 65,981 dólares, lo que representa un retorno de inversión de cinco años. La inversión necesaria para llevar a cabo el proyecto es de 649,145 dólares; este importe será financiado con un aporte de capital del 30% por el promotor del proyecto y un 70% será financiado mediante un préstamo bancario. Finalmente, se concluye que el plan de negocio es viable, sustentado en una demanda existente de las personas para consumo de productos de cuidado personal, el gasto per cápita que está por debajo del gasto promedio en Latinoamérica, la creciente preocupación de las personas por el tema de salud (stress), la escasa oferta de establecimientos en la zona de Los Olivos, y la factibilidad operativa y económica del proyecto.

Les polymères dynamiques ont la particularité d’être composés de monomères liés entre eux par des liaisons covalentes ou non et dont la formation est réversible dans certaines conditions. Leur structure et leurs propriétés sont donc susceptibles de s’adapter avec les conditions dans lesquelles ils sont placés, et cela même après la polymérisation. Les polysaccharides sont formés par des séquences de saccharides connectés entre eux par des liaisons glycosidiques. Si on remplace cette liaison glycosidique par un lien covalent réversible, comme la fonction imine, on obtient alors un analogue dynamique d’un polysaccharide. Notre but a été de préparer des analogues dynamiques de polysaccharides naturels basés sur la formation réversible d’imines (hydrazones, oximes). Différents types de monomères ont été synthétisés par fonctionnalisations de saccharides ou d’oligosaccharides naturels (glycosylations, oxydations, protections et déprotections sélectives,…)
Reversible polymers, or dynamic polymers, result from the reversible association of monomers using covalent (dynamic covalent polymers) or non covalent (supramolecular polymers) chemical bonds. As the polymerisation is reversible, the structure, the molecular weight distribution, and the composition of the polymer are susceptible to evolve even after polymerisation. According to their dynamic character this polymers are adaptative and can respond to an external stimuli (temperature, pH, presence of a molecular target) by a constitutional change, and so they can be considered as smart materials. Our goal was to prepare dynamic covalent polymers, based on reversible imine bonds formation (hydrazones or oximes), that are analogs of natural polysaccharides. This dynamic glycopolymers could be interesting as materials, and have biorecognition properties combined with the possibility to adapt their sequence and composition in presence of a biological target

En la producción de aceite de oliva se debe enfrentar el problema de añerismo de esta especie, siendo particularmente dañino por la intensidad en que se manifiesta, afectando la disponibilidad de materia prima. Con el objeto de regular la carga frutal, en olivos bajo un sistema orgánico, se estableció un ensayo mediante diferentes regímenes de poda, utilizando árboles de 8 años de edad, de la variedad aceitera Coratina, en su año alta producción. El estudio fue realizado en el fundo “El Oliveto” (33º48 latitud sur y 71º05 longitud oeste, altitud 205 m.s.n.m.), localidad de Cholqui, comuna de Melipilla, provincia de Melipilla, Región Metropolitana, Chile. Se realizaron tres tratamientos, los cuales se efectuaron a salidas de invierno, el 12 de septiembre del 2006, cuando los árboles presentaban el estado fenológico de ramillete expuesto. Se contó con un tratamiento control, sin poda; un segundo tratamiento con un 33% de remoción de la madera del año y un tercer tratamiento con un 50% de remoción de la madera del año. Cada uno de los tratamientos contó de cinco repeticiones; siendo la unidad experimental el árbol. En los tratamientos de poda, se logró un aumento en el crecimiento de brotes durante la temporada, respecto del tratamiento testigo. Además, se redujo el porcentaje de frutos retenidos, lo que repercutió tanto en un mayor tamaño y peso de fruto, como en el porcentaje de aceite al momento de la cosecha. A medida que la intensidad de poda fue mayor, menor fue la producción de los olivos y la carga frutal obtenida. Finalmente con ambos tipos de poda, se obtuvo similares producciones de aceite por árbol (kg aceite/árbol).
Alternate bearing is particularly damaging in the olive trees because of the intensity with which it manifests itself, affecting the availability of raw material for oil production. With the objective of regulating fruit load in olive trees under an organic system, a trial with different pruning systems was established using eight-year-old trees from of the oil Coratina cultivar in their full-bearing year. The study was carried out on the “El Oliveto” farm (33º 48´ South lat. and 71º 05´ West long., at 205 m.a.s.l.) at the Cholqui location, Melipilla commune, Melipilla province, Metropolitan Region of Chile. Three treatments were performed in late winter (Sept. 12, 2006), when the olive trees were in the phenological stage of “exposed bouquet”. Treatments consisted of: (1) control without pruning; (2) 33% removal of one-year-old wood; and (3) 50% one-year-old wood. Five replications of each treatment were done, with a single olive tree being the experiment unit. An increase in shoot growth was achieved with the pruning treatments during the season compared to the control treatment. There was a decrease in the percentage of retained fruits with the pruning treatments, resulting in a greater fruit size and weight as well as in a greater oil content at harvest. As pruning was intensified, olive production decreased. As a consequence, similar oil yields per tree (kg oil/tree) were obtained with both types pruning.

El distrito de Los Olivos tiene las condiciones necesarias para convertirse en el distrito más competitivo de Lima Norte. A pesar de ello, sus líderes y gobernantes no han logrado formular una visión de futuro que los ubique en una posición más competitiva. La inseguridad ciudadana, los índices de pobreza, el acceso a la educación de calidad y a la asistencia sanitaria, la informalidad y la falta de oportunidades son algunos de los problemas cotidianos que agobian a su población. A ello se suma el contexto político, económico y social por el que atraviesa el país. Para salir de esta situación se propone el presente plan estratégico del distrito. El cual ha sido desarrollado siguiendo la metodología del Modelo Secuencial del Proceso Estratégico, propuesto por el Dr. Fernando A. D’Allesio Ipinza en su libro El Proceso Estratégico: Un enfoque de gerencia. Este proceso estratégico contempla elementos de análisis, definición de objetivos y estrategias que involucra a diversos actores para la consecución de una visión de futuro retadora. Que se complementa con herramientas de evaluación y control que deben ser ejecutadas permanentemente dada la naturaleza de un entorno cambiante y susceptible a la coyuntura externa. La aplicación del presente trabajo busca como resultado el desarrollo del distrito de Los Olivos y la felicidad de su población. The district of “Los Olivos” has the necessary conditions to become the most competitive distric of Lima Norte. But spite of that , its leaders and governors have not been able to formulate a vision of the future that put there in most competitive position. The insecurity of the citizens, the indices of poverty, the access to quality education and the health assistance, the informality and lack of opportunities are some of the everyday problems that overwhelm its population. In addition to it, including the political, economical and social context by which the country is going through. To get out from this situation the present strategical plan is proposed, this plan has been developed following the methodology of the sequential model from the strategical process porposed by Dr. Fernando A. D’Allesio Ipinza in his book “The strategical process: a focus of management”. This strategical process contemplates elements of analysis, definitions of objetives and strategies that involve diverse actors for the achievement of a vision of challenging future. This is completed with tools of evaluation and control that must be carried out permanently given the nature of a changing evironment and susceptible of the external joint. The application of this work looks for the result of the development of the district of “Los Olivos” and the happiness of its inhabitants. The plan contains the strategical guidelines so the district takes advantages of its factors of production in an effective way and to develop competitive advantage to attract investments, generate sources of employment, reduce inequality gaps, improve security, raise the income per capita of its population and better their wellness. After that, “Los Olivos” will be a cultural, modern and safe district, transformed into a better place to live

Este proyecto consiste en el diseño de un Centro de Salud del Primer Nivel de Atención especializado en la rehabilitación integral de niños y adolescentes. Se basa en una arquitectura terapéutica con la que se diseña un establecimiento de salud generando espacios amplios y abiertos hacia jardines terapéuticos, integrando los espacios naturales y recreativos con las áreas de tratamiento. Se busca resolver la problemática de nuestra ciudad en la cual tenemos un déficit de centros de salud especializados en la rehabilitación pediátrica. El proyecto se ubica estratégicamente en el distrito de Los Olivos por la demanda en este sector de la población, y la falta de centros de salud de la zona. La población a servir son menores de 18 años con ciertas limitaciones, para que puedan desarrollarse y reintegrarse en la sociedad de manera adecuada.
The project consists of the design of a Pediatric Rehabilitation Center. It is based on a therapeutic architecture that is used in designing a healthcare building by creating wide and open spaces to the gardens thus contributing to the user’s rehabilitation.  It is intended to integrate the therapeutic gardens and natural areas of the project alongside the treatment areas with the objective to provide care for children and young people from an early age in order for them to develop and reintegrate themselves into society adequately. The project's location is in Los Olivos's district in the northern part of Lima due to the high demand of users and the lack of infrastructure in the area to help them.

Im Mittelpunkt der Arbeit stand die Entwicklung, Darstellung, Charakterisierung sowie Applikation maßgeschneiderter, self-assembly-fähiger Moleküle, die gezielt auf TiO2- und nicht auf SiO2-Oberflächen anbinden. Die resultierenden Monoschichten (SAMs) wiesen eine Biokompatibilität sowie Biofunktionalität auf. Dazu wurden neue bisher noch nicht beschriebene Moleküle entwickelt, die auf einer Kombination von funktionalisierten Oligo(ethylenglykol)-Einheiten mit Monoalkylphosphorsäure- und Alkylphosphonsäurederivaten basieren. Diese Verbindungen konnten durch die Anwendung der Self-Assembly-Technik erfolgreich aus wässriger Lösung auf TiO2-Substrate adsorbiert werden. The aim of this work was the design, preparation, characterization and application of tailor-made, self-assembly molecules, which are able to adsorbed selectively on TiO2 surfaces but not on SiO2. The resulting self-assembled monolayers (SAMs) had a biocompatibility and bio functionality. For this purpose new molecules have been developed, which are not described in the literature. These compounds are derivatives of monoalkyl phosphoric acids or alkyl phosphonic acids and contain a terminal functional oligo (ethylene glycol) unit. The compounds were assembled on the TiO2-surface by self-assembly technique from aqueous solution. The adsorbed layers were characterized by different analytical tools, like angle resolved XPS, spectroscopic ellipsometry and SPR-spectroscopy. The selective adsorption of SAMs on TiO2-surfaces and the formation of stable SAMs make it possible to prevent or minimize non specific protein adsorption and also to bind selected proteins via specific surface reactions

Esto debido a que es la única zona en la ciudad que no posee de un centro de esta tipología de manera próxima, dificultando el tratamiento de un considerable número de pacientes. Los cuales, deben transportarse a pesar de sus limitaciones hacia la zona sur y este de la capital; Instituto Nacional de Rehabilitación Dra. Adriana Rebaza Flores en el distrito de Chorrillos y Hogar Clínica San Juan de Dios en el distrito de San Luis, respectivamente. Se trata de un centro de categoría II-E, con funciones de promoción, prevención, recuperación y rehabilitación. El cual propone como énfasis el recorrido arquitectónico con elementos que permitan el bienestar anímico como circulación accesible, ingreso de luz, uso de color y vegetación, lo cual permite una sanación más pronta sin excesivo uso de medicación.
This project shows us the social need to implement a rehabilitation center in the north zone of Lima. This place was choosed because it is the only area in the city that does not have a center of this typology, making it difficult to treat a huge number of patients. They must travel despite their limitations to the south and east zone of the capital: The national rehabilitation institute Dra. Adriana Rebaza in the district of Chorrillos and the rehab center of “San Juan de Dios” in the district of San Luis, respectively. The category of this project is II-E, they have the function of promotion, prevention, recovery and rehabilitation. This project, proposes an emphasis on the “architectural path with elements that allow the animic welfare as accessible circulation, light income, use of color and vegetation, which facilitates a faster healing without excessive use of medication.

In this thesis newly generated high-resolution Oligo-Miocene climate proxy records from Walvis Ridge ODP Site 1264 (south-eastern Atlantic Ocean) are presented (Chapters 2 and 3). The records are tuned to an eccentricity solution (Chapter 3) and they are compared to published Atlantic and Pacific palaeoclimate chronologies (Chapters 2 and 4). The main research objectives are 1) to identify astronomical pacemakers of global significance and test earlier pacing theories, 2) to describe global climate and oceanographic change on astronomical and tectonic time scales and 3) to test the strong hysteresis in ice sheet models that suggest a very stable Antarctic ice sheet once formed. Chapter 1 gives a general introduction on the “mid”-to-late Oligocene climatic, oceanographic, geographic and cryospheric settings. Climate evolution and dynamics, together with the major underlying processes are introduced. In Chapter 2, high-resolution early Miocene stable oxygen and carbon isotope chronologies from Walvis Ridge Site 1264 are presented. The data are analysed on an untuned age model to identify the principal astronomical pacemakers, without introducing power on orbital frequencies. A dominance of variance in all datasets on 100-kyr timescales is found. The δ18O data are used to parameterize a suite of 1D ice sheet models and show that between 20 – 80% (avg. ~50%) of the δ18O signal can be explained by changes in Antarctic ice volume. (This chapter has been published as: D. Liebrand, L. J. Lourens, D. A. Hodell, B. de Boer, R. S. W. van de Wal and H. Pälike. Antarctic ice sheet and oceanographic response to eccentricity forcing during the early Miocene. Climate of the Past, 7, 869–880, 2011) In Chapter 3, extended stable-isotope records together with X-ray fluorescence core scanning data from Walvis Ridge Site 1264 are presented. The records span an 11-Myr mid Oligocene through early Miocene time interval. Ages are calibrated to eccentricity, are in good agreement with the GTS2012 and independently confirm the Oligo-Miocene time scale to the ~100-kyr level. The ~2.4-Myr long-period eccentricity cycle is identified as the main pacemaker of Oligo-Miocene climate events, as identified in the benthic isotope records, at shorter astronomical (eccentricity) periodicities. In Chapter 4, the high-resolution Oligo-Miocene benthic stable-isotope chronology from Site 1264 is compared to published records from the Atlantic and Pacific to further identify and explore possible global climate pacemakers. In addition, an investigation of long-term trends and inter-/intra-basin isotopic gradients and their implications for ice volume reconstruction and palaeoceanographic studies are discussed. Methods are explored to quantify the apparent change in geometry of ~100-kyr cycles in our benthic δ18O data and the analyses indicate an increased cycle asymmetry (i.e. sawtooth patterns) throughout the Oligo-Miocene. This change in cycle geometry is interpreted as a measure of changing boundary conditions and used to track the evolution of a threshold response mechanism in Earth’s climate system. In Chapter 5 the main results of this thesis are summarised, the implications for our understanding of the Oligo-Miocene are discussed and perspectives are given on future work.

This thesis describes the synthesis and analysis of organometallic complexes that feature oligo-phenylene ethynylene based ligands. Chapter 1 introduces the general topic of molecular electronics and provides a general overview of the interest in phenylene ethynylene systems as foundation architectures for molecular wires. The role of metal complexes in molecular electronics and the application of the cluster - surface analogy to the study of model systems is also described. Chapter 2 describes the synthesis of oligo-phenylene ethynylene pro-ligands of the general form Мe(_3)SіС≡СС(_6)H(_4)C≡СС(_6)H(_4) (1-5), in which R is either an electron donor (Me, OMe) or acceptor (СО(_2)Ме, NO(_2), CN). The compounds were synthesised either via Sonogashira Pd/Cu cross-coupling reactions or via the nucleophilic attack of benzoquinones by lithiated acetylide anions and subsequent reduction. The desilylation of these compounds afforded the terminal alkynes НС≡СС(_6)Н(_4)С≡СС(_6)Н(_4)R (6-10). Furthermore, the extended "three-ring" 1,4- bis(phenyl ethynylbenzene) derivatives Мe(_3)SіС≡СС(_6)Н(_4)С≡СС(_6)Н(_4) ≡C(_6)H(_4)С≡СSiMe(_3), 11 and Мe(_3)SіС≡СС(_6)Н(_4)С≡СС(ОМе)(_2)(Н)(_2)С≡С(_6)Н(_4)С≡CSiMe(_3), 13 and the terminal alkyne НС≡СС(_6)Н(_4)С≡СС(OMe)(_2)(H)(_2)C≡ C(_6)(h)(_4)C≡CH (14) have also been prepared. These compounds were fully spectroscopically characterised and in the case of 14 the molecular structure analysis is discussed. Chapter 3 discusses the synthesis of the gold(I) oligo-phenylene ethynylene complexes The complexes were prepared by treating the ligands precursors 1-5 with АuСl(Рl(_3)) (L = Ph or Су) in the presence of NaOMe to afford complexes Аu(С≡СС(_6)Н(_4)С≡СС(_6)Н(_4)R)PPh(_3) [R = Me (15), OMe (16), СО(_2)Me (17), NO(_2) (18) and CN (19) and Аu(С≡СС(_6)Н(_4)С≡СС(_6)Н(_4)R)PCy(_3) [R = Me (20), OMe (21) and NO(_2) (22)]. The "three-ring" complexes {Аu(РРh(_3))}2(μ-С≡СС(_6)H(_4)С≡СС(_6)Н(_4)С=С(_6)Н(_4)С≡С), 23 and {Аu(РРh(_3)}(_2)( μ-C≡CC(_6)H(_4)C≡CC(_6)(OMe)(_2)H(_2)C≡C(_6)H(_4)C≡C 24 were also prepared. These complexes were spectroscopically characterised and molecular structural analyses reveal intermolecular interactions between the phenylene ethynylene portion of the molecules in the solid state, but not aurophilic interactions. Chapter 4 examines the synthesis of half-sandwich Ru(L(_2))Cp' [L = РРh(_3), Cp' = Cp ; L(_2) = dppe, Cp' = Cp*) acetylide complexes derived from simple phenyl, tolan and oligo(phenylene ethynylene) based acetylenes. The electrochemical properties of these complexes have been explored, as have some of the molecular structural details. Chapter 5 describes the synthesis of some cluster complexes. The gold acetylide complexes Аu(С≡СС(_6)Н(_4)С≡СС(_6)Н(_4)R)(РРh(_3)) react readily with Ru(_3)(CO)(_10)(μ-dppm) to afford phenylene ethynylene derivatives Ru(_3)(μ-АuРРh(_3))(μ-С(_2)С(_6)Н(_4)С≡СС(_6)Н(_4)-R)(μ-dppm)(C0)(_7) (38-42) in which the conjugated organic moiety is "end-capped" by the cluster and an R group that is either electron donating or withdrawing (R = Me, OMe, СО(_2) Мe, NO(_2), CN). The clusters 38-42 are linked to the hydrido clusters Ru(_3)(μ-Н)(μ-С(_2)С(_6)Н(_4)С≡СС(_6)Н(_4)R)(СО)(_7) (36 and 37) through the well-known isolobal relationship between H and Аu(РR(_3)). In addition, the bis-cluster {Ru(_3)(μ-dppm)(CO)(_7)}(_2){(μ-ΑuΡΡh(_3))}(_2)(μ-C(_2)C(_6)H(_4)C≡CC(_6)H(_4)C≡CC(_6)H(_4)C(_2)) (43) has also been prepared. All the clusters reported in this chapter were crystallographically determined. Structural, spectroscopic, photophysical and electrochemical studies were conducted and have revealed little electronic interaction between the remote substituent and the organometallic end-caps. Chapter 6 explores the novel, preparative scale stoichiometric transmetallation reactions involving the simple Аu(С≡СR)(РРh(_3)) (R = Ph or С(_6)Н(_4)Мe) complexes. These gold(I) complexes have been treated with several inorganic and organometallic compounds MXL(_n) [M = metal, L(_n) = supporting ligands, X = halide], to afforded the corresponding metal-acetylide complexes M(C≡CR)L(_n), with representative examples featuring metals from Groups 8-11. The acetylide products were fully characterised by usual spectroscopic methods including the molecular structural analysis. Chapter 7 concludes the general summary of the thesis and discusses briefly the findings achieved in each chapter and the vital role of oligo-phenyelene ethynylenes ligands in the construction of numerous organometallic complexes which show interesting and promising properties for the molecular wires development. In addition, further future work is also proposed on other systems that feature this ligands.

Halothermothrix orenii, a gram negative strictly anaerobic thermohalophilic bacterium has been isolated from sediments of a Tunisian salt lake (Chott El Guettar). H. orenii grows optimally at 60˚C with 10% NaCl. Phylogenetically, H. orenii is placed at the base of phylum Firmicutes, in the family Haloanaerobiaceae, order Haloanaerobiales, and is the only member of this order whose genome has been completely sequenced. The availability of the genome sequence will not only enhance our understanding of survival and adaptation strategies of microbes living under dual extremities of high salt and temperature, but will also provide information on novel biocatalysts which could have potential application in research, industrial and bioremediation processes. To date, our laboratory has studied a large number of genes that encode Glycosyl Hydrolases and Transferases from H. orenii, including AmyA and AmyB of the α-amylase family 13, a sucrose phosphate synthase (SPS) and a Fructokinase (Frk). Biochemical analysis demonstrated that all these recombinant enzymes are optimally active between 55-65˚C and in addition, the extracellular amylases AmyA and AmyB are most active with NaCl concentration between 5-25%. All the recombinant enzymes have been crystallized and their structures solved, but their molecular basis of thermohalophilicity has yet to be determined. In the present study, data mining of the genome of H. orenii further identified six potential α-amylase coding genes (designated Amy1, Amy2, Amy5, Amy6, Amy7 and Amy8). Primers were designed for cloning and overexpression of these genes into two vectors, namely flexi pFN6A and pET22b (+). Clones of Amy1, Amy6, Amy7 and Amy8 were discarded from the freezer by a member of the plant laboratory in my school, and therefore only clones Amy2 and Amy5 were available for studies. Amy5 clone has been studied in detail and forms the basis of this thesis. The Amy5 gene, which consists of 1701 nucleotides, was determined by BLAST analysis to code for an oligo-1,6-glucosidase, was cloned in pET22b (+) vector and the recombinant enzyme successfully over-expressed in the E. coli B strain (BL21 (DE3)). The recombinant enzyme (designated rAmy5) was purified to homogeneity using immobilized metal affinity chromatography (IMAC). The molecular weight of rAmy5 was determined by SDS to be 66kDa, which is close to the expected theoretical calculated value of 66.766kDa. The rAmy5 hydrolysed p-nitrophenyl-α-D-glucopyranoside (PNPG). The temperature optimum for enzyme activity was 60˚C (temperature activity range between 45-65˚C) and the pH optimum was between pH 6 and 6.5 (pH activity range between pH 5.3-7). rAmy5 activity was inhibited by metal ions in the order Cd 2+ > Cu2+ > Zn2+ > Fe3+ > Fe2+ > Ni2+ > NH4+, but enhanced by Ca2+ and Mn2+ with Mg2+ and Co2+ having no effect. The recombinant enzyme was tolerant to proteolysis when tested in presence of various concentrations of Trypsin. DTT (Dithiothreitol) had no effect on enzyme activity suggesting that cysteine residues did not play any role in providing thermostability to the enzyme molecule. Activity of the enzyme against PNPG was completely inhibited by 100mM D-glucose or 0.2mM PNP (p-nitrophenol). EDTA was found to have no effect on enzyme activity. The enzyme activity was enhanced in the presence of salts such as 0.5M NaCl and 1M KCl by 22.5% and 45% respectively. Denaturants inhibited enzyme activity in the order SDS > Tris > Urea. Sugars inhibited the enzyme activity in the order D-glucose > D-fructose > Mannitol > Xylose > Cellobiose > Lactose > Cellobiose > Lactose > Mannose > Raffinose > Galactose. The enzyme displayed no activity when tested against the following PNP-based glycosides: 2-Nitrophenyl-β-D-fucopyranoside, p-Nitrophenyl-α-L-arabinopyranoside, p-Nitrophenyl-α-D-xylopyranoside, p-Nitrophenyl-β-D-xylopyranoside, 2-Nitrophenyl-β-D-glucopyranoside, p-Nitrophenyl-β-D-fucopyranoside, 2-Nitrophenyl-1-thio-β-D-galactopyranoside, and p-Nitrophenyl-β-D-glucuronide. The activation energy of enzyme was found to be 81.3kJ/mol. The enzyme was thermostabilized at 60˚C in the presence of salts and combination of metal ions in the following order: 0.5MNaCl > 0.5M NaCl + 25mM CaCl2 > 1M KCl > 1M KCl + 25mM CaCl2. Presence of 25mM CaCl2 alone was found to destabilize the enzyme at 60˚C. The half-life of the enzyme increased four times in presence of 0.5M (10%) NaCl at 60˚C. The enzyme followed Michaelis Menten kinetics for PNPG and was found to have a KM and VMAX of 1.3mM and 333.3mM PNPG/min per mg of enzyme respectively. The enzyme had a high specific activity and was found to be active against a variety of substrates. The probable order of its specificity was PNPG > dextrin > dextran > isomaltotriose > amylose > pullulan > palatinose > soluble starch > trehalose > maltotetraose > maltose > isomaltose > maltooligosaccharide > panose. The enzyme was more active against substrates containing (1→6)-α-D-glucosidic linkages. In silico analysis revealed that the Amy5 putative protein sequence consisted of a highly conserved ‘QpDln’ signature for oligo-1,6-glucosidase sub-family. The high thermostability of the rAmy5 enzyme could be correlated to an increased presence of prolines and preference for glycine over alanine in the Amy5 sequence. A preference for charged residues over uncharged residues indicated the possible reason for the increased thermostability with NaCl of the rAmy5 enzyme. The fact that the Amy5 gene sequence had a significant identity (63.7%) to the sequence of solved crystal structure of oligo-1,6-glucosidase (1UOK) from Bacillus cereus formed the basis for a comparative homology modelling of Amy5 protein and as result a 3-Dimensional model of the Amy5 protein could be predicted. Comparative analysis of the predicted Amy5 model with 1UOK revealed that the residues of deep catalytic cleft of the latter were found at the same positions (Asp199, His 328, and Asp329), except that the Glu255 residue of 1UOK aligned with position Glu256 of Amy5. These catalytic residues are considered necessary for enzyme function rather than enzyme substrate specificity. Study of crystal structures of various enzymes has revealed that there are various subsites (amino acid residues) which associate with catalytic residues to impart the character to the enzyme to act on specific substrates. In addition to the present study of comparative modelling and biochemical characterization of the rAmy5 enzyme, protein crystallography studies could add to our understanding of structure-function relationship. In our present study, an attempt to crystallize the rAmy5 protein was made using a hanging drop method by screening it against 240 different buffer conditions (using JBScreen Mixed Crystallography kit, Jena Biosciences, Germany), but crystals were not observed during the three-month incubation period.

El proyecto que se está desarrollando en un Centro Oncológico de categoría I-3 según el MINSA, que cumpla con la arquitectura sensorial para darle mayor confort a los usuarios durante tu tratamiento. El proyecto encuentra en el distrito de Los Olivos, Lima. Lo que motiva a esta investigación es la falta infraestructura especializada para el tratamiento del cáncer. El propósito de proyecto es realizar un centro ambulatorio que cubra esta necesidad ya que actualmente es un problema de salud pública. El centro de salud propuesto tiene como objetivo ser un apoyo en los tratamientos que da el INEN. El INEN, la atención a los pacientes de aproximadamente de 17 min, por lo que hay déficit de atención, puesto que la atención adecuada para los pacientes con cáncer en un instituto debe de ser de 20 a 30 minutos. El centro se ubicará en el distrito de Los Olivos, ya que tiene como objetivo descentralizar los servicios oncológicos. Actualmente se están tiene estrategias para mejorar estos servicios, sin embargo, en estas no cuentan con la mención de un centro especializado que descentralice el INEN. El centro oncológico estará diseñado tomando en cuenta referencias a nivel internacional, un estudio de los procedimientos dados en el INEN y la normativa peruana brindada por el MINSA.
This thesis project develops and Oncology Center of category I-3, according to MINSA which complies with the sensorial architecture in order to give comfort to the patients and users. This project will be located in Los Olivos district, Lima. What motivates me about this research is the lack of specialized infrastructure for cancer treatment. The purpose of this project is to design an outpatient center that meets this current public health problem. The health center has the intent to support the treatments that INEN provide. In this institution (INEN), the attention last at least 17 minutes, this means there is a deficit, because the optimal time should be around 20 to 30 minutes. This new center would be located in Los Olivos district towards to decentralize the oncology services. This Oncology Center will be design taking notice of the international references, the study of procedures done by INEN and the Peruvian regulations set by MINSA.

La presente investigación tiene como propósito principal calcular el Índice de Progreso Social (IPS) para el distrito de Los Olivos. La metodología de la investigación se en basó en el Social Progress Imperative (SPI), la cual permite determinar el Índice de Progreso Social mediante la integración de cuatro componentes en tres dimensiones: (a) Necesidades humanas básicas, (b) Fundamentos de bienestar, y (c) Oportunidades. El tipo de investigación es de tipo cuantitativa, no experimental y con alcance descriptivo, en donde la información base, se obtuvo de una recolección de datos primaria a través de encuestas realizadas a una muestra de la población del distrito, por medio de un cuestionario de preguntas proporcionados por CENTRUM PUCP Graduate Business School, además se recolecto la información a través de fuentes secundarias. El tamaño de la muestra con los cuales se obtuvieron los resultados fue de 400 jefes del hogar, a los cuales se les entrevisto teniendo en cuenta las cuatro zonas del distrito. El cálculo del IPS del distrito de Los Olivos es el primer precedente con el que cuenta el distrito y podrá ser utilizado para la toma estratégicas de decisiones en el ámbito social y ambiental. El resultado obtenido del IPS del distrito de Los Olivos es de 67.48, clasificado en el nivel “Medio Alto”, cabe destacar que se encuentra por encima del IPS de Lima Metropolitana. Los resultados a nivel de dimensiones posicionan a Necesidades humanas básicas en primer lugar con un puntaje de 75.13 clasificado como “Alto”, seguido de Oportunidades con un puntaje de 64.97 clasificado como “Medio Bajo”, y Fundamentos de bienestar con un puntaje de 62.35 clasificado como “Medio Bajo”. La investigación muestra objetivamente que los resultados a nivel de indicadores que componen el IPS en el distrito de Los Olivos necesitan atención y sirve como punto de partida para que las autoridades municipales consideren este trabajo de investigación como fuente para su gestión.
The purpose of this research paper is to calculate the Social Progress Index (IPS) of the district of Los Olivos in order to ascertain the results in the dimensions that the model poses. To achieve this, we have followed the methodology proposed by the Social Progress Imperative (SPI) and the information was gathered through surveys provided by CENTRUM PUCP which were taken to the family heads of each home of the district. Additionally, a quantitative research was included containing a descriptive analysis in the district. In the same way, it contains information proceeding from secondary sources. This study is the first made without precedents in the district of Los Olivos to measure the IPS and may be used to take strategic decisions. The IPS is centered in offering detailed information of the population through the following measurements: (a) basic human needs (b) welfare foundations, (c) opportunities. The results obtained from the IPS of the district of Los Olivos is 67.48, which represents a “Medium High” classification level which is higher than the IPS of Lima. This indicator was obtained from the average of the results of the three measurements: (a) basic human needs with a score of 75.13 classified as “High”, (b) welfare foundations with a score of 62.35 classified as “Medium Low” and, (c) opportunities with a score of 64.97 classified as “Medium Low”. This shows that the components of welfare focused in the basic human needs are well established in comparison with the other measurements. This research shows in an objective manner the indicators that the district of Los Olivos needs to improve: it likewise serves as a starting point that will enable the municipal authorities to take decisions and actions in order to improve their social and environmental indicators in such a way that they can likewise improve the quality of life and opportunities of the population that lives in the district of Los Olivos.

Carbohydrates are a complex group of biomolecules with a high structural diversity. Their almost omnipresent occurrence has generated a broad field of research in both biology and chemistry. This thesis focuses on three different aspects of carbohydrate chemistry, synthesis, structure elucidation and the conformational analysis of carbohydrates.

The first paper describes the synthesis of a penta- and a tetrasaccharide related to the highly branched capsular polysaccharide from Streptococcus pneumoniae type 37. In the second paper, the structure of the O-antigenic repeating unit from the lipopolysaccharide of E. coli 396/C1 was determined along with indications of the structure of the biological repeating unit. In addition, its structural and immunological relationship with E. coli O126 is discussed. In the third paper, partially protected galactopyranosides were examined to clarify the origin of an intriguing 4JHO,H coupling, where a W-mediated coupling pathway was found to operate. In the fourth paper, the conformation of methyl a-cellobioside is studied with a combination of molecular dynamics simulations and NMR spectroscopy. In addition to the expected syn-conformation, detection and quantification of anti-ø and anti-ψ conformers was also possible.

The focus of this thesis is to examine the structural properties of polysaccharides produced by bacteria, as well as the dynamic and conformational behavior of a synthetically derived oligosaccharide. The primary structures of the O-polysaccharide repeating units of four different Escherichia coli (E. coli) strains, namely O175, O177, O103 and TD2158, as well as the first report of a capsular polysaccharide produced by lactic acid bacteria Leuconostoc mesenteroides ssp. cremoris PIA2 are reported in paper I–V. Structural analyses have been performed using a combination of nuclear magnetic resonance spectroscopy and chemical component analysis. The elucidated structures in paper I–III, as well as paper V, are composed of linear repeating units of varying composition and length. In paper IV, the structure of the O-polysaccharide repeating unit of E. coli TD2158 is determined to be a branched hexasaccharide structure with a heterogeneous substitution pattern, with either a β-GlcpNAc or β-Glcp residue branching to the backbone chain. Incubation with bacteriophage HK620 tailspike protein shows that the polysaccharide is selectively cleaved at the α-GlcpNAc-(1→2)-α-Rhap-linkage of the backbone chain, yielding a 9:1 ratio of β-GlcpNAc/β-Glcp containing hexasaccharides after digestion. In paper VI the conformational properties of a trisaccharide, which constitutes an internal epitope of the LeaLex hexasaccharide over-expressed on the surface of squamous lung cancer cells, have been analyzed using NMR spectroscopy and molecular dynamics simulations. The β-(1→3)-linkage of the trisaccharide was shown to be highly flexible.

The performance of devices based on small molecules depends on their molecular architecture and their two- and three-dimensional organisation into thin films. In this thesis scanning probe microscopy and density functional theory calculations are used to elucidate packing of poly(aniline) oligomers, oligo(aniline)s, into films formed by vacuum thermal evaporation and solution-processing. Thermal deposition of oligo(aniline)s under ultra-high vacuum conditions and subsequent study by scanning tunneling microscopy at various temperatures highlights two key issues with the preparation of oligo(aniline) thin films by this method. Firstly, the underlying copper substrate was shown to catalyse the fragmentation of oligo( aniline)s even at temperatures as low as 30 K. Secondly, upon an interfacial layer of fragmented molecules further disordered layers form, these layers are disordered due to presence of multiple isomeric forms of oligo(anilines). These two issues will prevent the preparation of high-quality oligo(aniline) thin films, and therefore limit the successful application of these molecules into devices. Computational studies of the isomerism of oligo(aniline)s undertaken by ab initio quantum calculations revealed a wealth of information about the gas-phase behaviour of these molecules. The energy barriers to cis/trans, syn/anti and ring rotation are extracted, as well as the kinetics of all these processes. Relatively low energetic barriers coupled with the small energy differences between isomers suggest that thin films of these molecules will be subject to conformational disorder than will inhibit their performance in applications. This led to the calculation of the molecular structures of ladder oligo( aniline )s. These were shown to be planar and rigid, and therefore offer attractive potential synthetic targets if the unique properties of oligo( aniline)s are to be exploited. Self-assembly of an amphiphilic oligo( aniline) into thin films by solution processing was investigated by tapping-mode atomic force microscopy and scanning tunneling microscopy. The driving forces for the formation of self-assembled thin films of oligo ( aniline)s are a delicate balance of non-covalent interactions. By taking a methodical approach, thin films of vertically or horizontally oriented oligo( aniline) molecules in their semi-conducting or conducting state could be formed by simple drop-casting. Important thin film properties are highly anisotropic, and the ability to control the orientation of molecules within a film is crucial for maximising device performances. Scanning tunneling microscopy of self-assembled monolayers of oligo(aniline)s reveals their surface structures, and offers the potential to manipulate them on a single molecule basis.

Whether or not a causal relationship exists between climate and mammal body size is one of the longest-standing and most intractable questions in ecology. The classic model of body size evolution (Bergmann's Rule) holds that body size is driven by temperature, but more recent hypotheses have suggested that other climatic variables or biotic interactions may play a more important role. The use of paleoecological data to address this question allows variables that are tightly correlated in modern ecosystems to be teased apart and allows body size patterns to be observed through time, adding an extra dimension to analyses. This dissertation details the findings of two paleoecological tests of Bergmann's Rule in the Oligo-Miocene (30-5 Ma), one tracking body size and climate through time in the northwestern United States and another tracking geographic body size trends through time along the west coast of North America. In both cases, body size was analyzed in three representative families of mammals: equids, canids, and sciurids. Such large-scale analyses are dependent on fossils that can be placed in a reliable taxonomic, geologic, and temporal context, and this dissertation also focuses on a reevaluation of the canid fauna of Oregon's Juntura Formation that places a critically important Late Miocene carnivore fauna in just such a context. Two genera of canids - Epicyon and Carpocyon - are described from the fauna for the first time, with important implications for regional biostratigraphy. The body size analyses show no consistent relationship between body size and any climatic variable. Further, body size patterns vary widely between taxa at several levels, suggesting that one universal driver of body size evolution does not exist. Not only is there no evidence for Bergmann's Rule in Oligo-Miocene mammals, but comparative analyses of geographic body size patterns in the modern genera Odocoileus, Canis, and Spermophilus fail to show the latitudinal gradients upon which Bergmann's Rule is predicated. The apparent existence of such trends in some taxa may be the result of anthropogenic extirpation at low latitudes, further underscoring the importance of including paleontological data when formulating models predicting the response of biotic variables to environmental change.

Thesis presents synthesis and study of thiophene monomers, oligomers and polymers. The new series of thieno[3,4-b]pyrazine copolymers based on 3-dodecylthiophene and pyrazine monomers were prepared by oxidative polymerization with FeCl3. The effective synthetic method for preparation of 3-alkylthiophenes and thiophene oligomers was developed by optimizing of Kumada cross-couplig. The mentioned method could be realized for multikilos scale with possibility of transfer to pilot plant production. The second part of thesis focuses on synthesis and study of new thiophene compounds, which consist of both 2,3-diazo-1,3-butadiene bridge with two terminal chromophores and two thiophene units linked together via -position by pyrazine or hydrazine bridge. Finally, the new type of regular alternating copolymer consists of 2,2´:5´,2´´-terthiophene-5,5´-dicarboxylic acid (TEDA) and polyethyleneoxide (PEO) was prepared. Formation of polymer nano-subunits as separated phases in solid state was confirmed by TEM.

Ce travail de thèse décrit la préparation et l'étude des propriétés d'auto-assemblage de nouveaux amphiphiles fonctionnalisés par des sucres. Des glycosides propargyliques du lactose et de la N-acétyl-glucosamine ont été conjugués par chimie click (cycloaddition de Huisgen catalysée par des sels de cuivre) à des dérivés de poly(ethyleneglycol) dont une des extrémités a au préalable été modifiée par une fonction azide et l'autre par un bloc hydrophobe de type polyphénylène ou bien aliphatique. Après une caractérisation par résonance magnétique nucléaire et spectrométrie de masse, les propriétés d'auto-assemblage de ces amphiphiles ont été étudiées par diffusion dynamique de la lumière (DLS), diffraction des rayons-X aux petits angles (SAXS) et microscopie électronique. Il a été montré qu'en phase aqueuse, les systèmes amphiphiles dérivés du PEG 900 s'auto-assemblent pour former de micelles de taille extrêmement régulière dont le diamètre moyen est de l'ordre de 10 nm. La présence et la biodisponibilité des sucres à la surface de ces nanoparticules ont également pu être démontrées par diffusion dynamique de la lumière avec les lectines PNA et WGA. Les interactions spécifiques observées entre les lectines et micelles associées aux propriétés d'encapsulation de ce type de nanoparticules permettent d'imaginer de futures applications pour la délivrance de médicaments ou encore l'imagerie médicale
This thesis reports the preparation and characterization of new rod-coil amphiphiles functionalized with oligo- and polysaccharides through Huisgen 1,3-dipolar cycloaddition reactions between species functionalized by an azide group on one side and an terminal alkyne on the other catalyzed by copper. The amphiphiles were synthesized and characterized based on different hydrophobic parts conjugated with the polymer poly(ethylene oxide) PEO with a hydrophilic spacer arm and the oligo- and polyssaccharides 2-propargyl-2-acetamido-2-deoxy-β-D-glucopyranose (GlcNAc) and propargyl β-D-galactopyranosyl-(14)-β-D-glucopyranose (Lac). The amphiphiles synthesized were characterized in terms of their chemical structure and composition through nuclear magnetic resonance (NMR), Fourier transform infrared (FTIR) spectroscopy, mass spectroscopy (MALDI-TOF-MS and ESI-MS) and high resolution mass spectroscopy (HRMS). After the dissolution in water, the amphiphiles self-associate in highly regular micelles with an average diameter of 2RH ~ 10 nm. Dynamic light scattering (DLS), transmission electron microscopy (TEM) and small angle x-ray scattering (SAXS) were used in order to investigate the structure and dynamics of these saccharide amphiphiles. The presence of carbohydrate epitopes on the surface of the micelles and their bioavailability for the segmentation of lectin were also demonstrated by DLS. Specific interactions of GlcNAc and Lac residues with lectins from wheat germ agglutinin (WGA) and peanut agglutinin (PNA), respectively, reveal the potential applications of such amphiphilic derivatives of carbohydrates as vectorizing systems, both simple and specific to a drug delivery site

La discriminación en el Perú es un fenómeno social de larga data. Desde la llegada de los españoles al continente americano, o incluso desde mucho antes, los primeros pobladores experimentaron la sensación de sentirse subyugados física y psicológicamente por un abanico de motivos que lo único que acarreó fue una condición categórica de desigualdad y prejuicio colectivo que afianzó una serie de conflictos sociales que todavía pululan en nuestro país. El presente trabajo busca mostrar, sin sesgo alguno, uno de los tantos fenómenos de discriminación que transita sin rostro ni identidad propia entre nosotros. Este fenómeno es denominado discriminación lingüística, y se ha elegido un colegio del distrito de Los Olivos, con el fin de mostrar la existencia de actos de exclusión por la manera de hablar de estudiantes migrantes. Por tales consideraciones, mediante el uso de la metodología sugerida en la investigación, sustentaremos las causas y modos en los que este fenómeno acciona en una realidad específica: la escuela. La existencia de la discriminación lingüística nos demuestra que todavía en la actualidad hay que hablar con cautela, pronunciar con atino y escribir con «finura», para no ser víctimas de inquisidoras críticas o comentarios que todavía siguen siendo el rezago de pensamientos acientíficos. Por tanto, la reflexión y el juicio flemático de esta realidad palmaria es la columna vertebrar de este trabajo de investigación, que, sin lugar a dudas, pretende ser el inicio de otras investigaciones que busquen el punto de quiebre entre lo moralmente censurable y lo realmente analizable.

Xyloglucans are polysaccharides found as storage polymers in seeds and tubers, and as cross-linking glycans in the cell wall of plants. Their structure is complex with intricate branching patterns, which contribute to the physical properties of the polysaccharide including its binding to and interaction with other glycans such as cellulose.

Xyloglucan is widely used in bulk quantities in the food, textile and paper making industries. With an increasing interest in technically more advanced applications of xyloglucan, such as novel biocomposites, there is a need to understand and control the properties and interactions of xyloglucan with other compounds, to decipher the relationship between xyloglucan structure and function, and in particular the effect of different branching patterns. However, due to the structural heterogeneity of the polysaccharide as obtained from natural sources, relevant studies have not been possible to perform in practise. This fact has stimulated an interest in synthetic methods to obtain xyloglucan mimics and analogs with well-defined structure and decoration patterns.

Glycosynthases are hydrolytically inactive mutant glycosidases that catalyse the formation of glycosidic linkages between glycosyl fluoride donors and glycoside acceptors. Since its first conception in 1998, the technology is emerging as a useful tool in the synthesis of large, complex polysaccharides. This thesis presents the generation and characterisation of glycosynthases based on xyloglucanase scaffolds for the synthesis of well-defined homogenous xyloglucan oligo- and polysaccharides with regular substitution patterns.

Oligo(ethylene glycol) groups have been used as substituents in poly(p-phenyleneethynylene)s (PPEs) to provide solubility, and to boost quantum yield. Properties such as water-solubility and increased quantum yield in aqueous solution make these conjugated systems promising for biosensory applications. In this thesis, a PPE containing a branched ethylene glycol side chain is synthesized as part of a polymer array for glycan biosensing. I also report that the same side chain can be put to use in a red-emissive polymer to lend water solubility. Another monomeric unit, containing ethylene glycol chains, is incorporated into a PPE to create an ampiphilic polymer. The versatility of these polymers allows them to be used for a variety of purposes, some of which will be described herein.

Electronic conductive polymers such as polyacetylenes, polyanilines and polythiophenes, are not easy to process due to lack of tractability and solubility. Chemical modifications of the polythiophenes backbone have been utilized to improve the processibility. Our goal is to synthesize and characterize 3-[oligo(dimethylsiloxane)]- thiophene macromonomers as precursors for potentially processible, conductive derivatives of polythiophene. The precursor will be used in the preparation of various poly {[3- oligo(dimethylsiloxane)]thiophene-co-3-methylthiophene} random copolymers. By increasing the chain length of the low glass transition (Tg) side chain, we hope to produce processible elastomeric material. w-(Si-H)-DMS, 3-propenylthiophene (i.e. precursors for the macromonomers), and the macromonomer were characterized (as appropriate) by 'HNMR, SiNMR, C NMR, DSC, IR, UV-VIS, GPC, and GC-MS.
