Thin-ply laminates exhibit a higher degree of freedom in design and altered failure behaviour, and therefore, an increased strength for unnotched laminates in comparison to thick-ply laminates. For notched laminates, the static strength is strongly decreased; this is caused by a lack of stress relaxation through damage, which leads to a higher stress concentration and premature, brittle failure. To overcome this behaviour and to use the advantage of thin-ply laminates in areas with high stress concentrations, we have investigated thin-ply hybrid laminates with different metal volume fractions. Open hole tensile (OHT) and open hole compression (OHC) tests were performed with quasi-isotropic carbon fibre reinforced plastic (CFRP) specimens. In the area of stress concentration, 90° layers were locally substituted by stainless steel layers of differing volume fractions, from 12.5% to 25%. The strain field on the specimen surface was evaluated in-situ using a digital image correlation (DIC) system. The embedding of stainless steel foils in thin-ply samples increases the OHT strength up to 60.44% compared to unmodified thin-ply laminates. The density specific OHT strength is increased by 33%. Thick-ply specimens achieve an OHC strength increase up to 45.7%, which corresponds to an increase in density specific strength of 32.4%.

PurposeOCT-angiography (OCT-A) offers a non-invasive method to visualize retinochoroidal microvasculature. As glaucoma disease affects retinal ganglion cells in the macula, macular microcirculation is of interest. The purpose of the study was to investigate regional macular vascular characteristics in patients with ocular hypertension (OHT), pre-perimetric primary open-angle glaucoma (pre-POAG) and controls by OCT-A in three microvascular layers.Material and methods180 subjects were recruited from the Erlangen Glaucoma Registry, the Department of Ophthalmology, University of Erlangen and residents: 38 OHT, 20 pre-POAG, 122 controls. All subjects received an ophthalmological examination including measurements of retinal nerve fibre layer (RNFL), retinal ganglion cell layer (RGC), inner nuclear layer (INL), and Bruch’s Membrane Opening-Minimum Rim Width (BMO-MRW). Macular vascular characteristics (vessel density, VD, foveal avascular zone, FAZ) were measured by OCT-A (Spectralis OCT II) in superficial vascular plexus (SVP), intermediate capillary plexus (ICP), and deep capillary plexus (DCP).ResultsWith age correction of VD data, type 3 tests on fixed effects showed a significant interaction between diagnosis and sectorial VD in SVP (p = 0.0004), ICP (p = 0.0073), and DCP (p = 0.0003). Moreover, a significance in sectorial VD was observed within each layer (p<0.0001) and for the covariate age (p<0.0001). FAZ differed significantly between patients’ groups only in ICP (p = 0.03), not in SVP and DCP. For VD the AUC values of SVP, ICP, and DCP were highest among diagnostic modalities (AUC: 0.88, 95%-CI: 0.75–1.0, p<0.001).ConclusionRegional reduced macula VD was observed in all three retinal vascular layers of eyes with OHT and pre-POAG compared to controls, indicating localized microvascular changes as early marker in glaucoma pathogenesis.

Orthotopic heart transplantation (OHT) represents an effective alternative for individuals with end-stage heart disease. The current literature reports only the responses of OHT patients to greater than or equal to 4 mo of exercise training (ET) and frequently lacks adequate controls. Most programs currently treating OHT patients usually provide 6-12 wk of ET. This study describes the effects of a 10-wk supervised ET program in 12 male OHT patients and 5 other male OHT patients who served as a comparison group. Graded exercise tests were performed before and after ET. After ET, maximal O2 consumption was significantly greater for the ET group than the comparison group (P less than 0.05) and the mean increase in peak heart rate was 18 +/- 4 and 6 +/- 4 (SE) min-1 for ET and comparison groups, respectively (P less than 0.05). Maximal ventilation was also significantly greater for the ET group at after ET, while resting heart rate and blood pressure and peak blood pressure, O2 pulse, respiratory rate, and ventilatory equivalents for O2 and CO2 were not significantly changed. We conclude that after OHT a 10-wk ET program improves maximal O2 consumption and, by improving peak heart rate, improves O2 delivery.

Purpose. To evaluate the changes of flash electroretinography (fERG) and pattern visual evoked potentials (pVEP) in ocular hypertension (OHT) patients.Methods. Twenty-five OHT patients and 30 healthy volunteers were enrolled for this cross-sectional study. Opthalmologic examinations, visual field tests, pVEP and fERG were performed. The main outcome measures were the differences between pVEP and fERG parameters.Results. The mean age of OHT patients and volunteers were 57 ± 12.25 years (range 30–65 years), and 53.25 ± 12.0 years (range 30–65 years), respectively. The mean amplitude of the pVEP was statistically lower in the OHT group (P< 0.05). Latency of the two groups was different; however, the difference was not statistically significant (P> 0.05). In fERG of OHT group, there was a significant decrease in the amplitude of the oscillatory potentials (Ops), and a significant delay in latency of rod and cone waves (allP< 0.05). There was no significant change in the flicker fERG waves between the two groups (P> 0.05).Conclusions. Although we found a decrease in Ops amplitude and a prolonged latency in flicker fERG, only the decrease in Ops amplitude was statistically significant between the two groups. The amplitude of Ops wave and amplitude of pVEP may reflect early glaucomatous damage in OHT patients.

Preflight, in-flight, and postflight IOP measurements were collected aboard two U.S. Space Shuttle Program missions in normotensive control, OHT, and glaucomatous crewmembers. METHODS: Five subjects (three controls, one glaucomatous, one OHT) were studied aboard 2-wk Space Shuttle missions. Baseline IOP (triplicate; handheld tonometry) was recorded during training 12 mo preflight, in flight (114 d), and postflight (329 d). Subjective symptoms were recorded via questionnaires. Data were analyzed using a spreadsheet with two-sample t-tests. P-value < 0.05 determined significance. RESULTS: IOP increased for all in-flight vs. preflight measurements for controls (N 3, 48.9, 16.9, 5.85), OHT (N 1, 20.3), and glaucomatous (N 1, 32.2) groups. IOP eventually returned to baseline postflight [Return (R)35 d], except for the astronaut with OHT (R917). Subjective symptoms, likely multifactorial, included blurredvision, decreased visual acuity, and headaches. DISCUSSION: IOP increased during spaceflight and normalized upon return. Astronauts and commercial spaceflight participants may need screening for elevated IOP to potentially prevent sequelae related to glaucoma and OHT, the former which requires treatment in flight and the latter which may need prophylaxis. Previous studies have shown elevated IOP upon entry into microgravity with various normalization timeframes in flight and postflight. It is unclear how increased IOP relates to spaceflight-associated neuro-ocular syndrome (SANS); however, several hypotheses exist. Treatment strategies should be available for acute and chronic ocular pathology during spaceflight despite the unique challenges of eye-drop application in microgravity. Purpose To correlate functional damage over time detected by standard automated perimetry (SAP) and frequency doubling technology (FDT) with central corneal thickness (CCT) in patients with ocular hypertension (OHT). Methods Seventy-eight OHT patients underwent CCT measurements, SAP, and FDT (the latter two also after 12 and 18 months). Patients were divided into three equally sized groups of 26 patients each: thin (<540 üm), normal (540–580 üm), and thick cornea (>580 üm). The frequency of abnormal FDT and SAP results was analyzed over time (Pearson χ2 test). Results Six of 26 patients with thin corneas (23.1%) presented an abnormal FDT test at baseline, compared to 1 of 26 (3.8%) in the normal thickness cornea group and 1 of 26 (3.8%) in the thick cornea group. After 12 months, the abnormal FDT tests were as follows, respectively: 9 of 26 (34.6%), 2 of 26 (7.7%), and 2 of 26 (7.7%). For SAP the abnormal results were as follows, respectively: 8 (30.1%), 5 (19.2%), and 2 (7.7%). After 18 months, the abnormal FDT tests were as follows, respectively: 16 (61.5%), 5 (19.2%), and 5 (19.2%). For SAP, the abnormal results were as follows, respectively: 10 (38.5%), 5 (19.2%), and 2 (7.7%). Conclusions OHT patients with thinner corneas have a greater risk of developing functional damage over time.

BackgroundThere are more than one million National Health Service visits in England and Wales each year for patients with glaucoma or ocular hypertension (OHT). With the ageing population and an increase in optometric testing, the economic burden of glaucoma-related visits is predicted to increase. We examined the conversion rates of OHT to primary open-angle glaucoma (POAG) in England and assessed factors associated with risk of conversion.MethodsElectronic medical records of 45 309 patients from five regionally different glaucoma clinics in England were retrospectively examined. Conversion to POAG from OHT was defined by deterioration in visual field (two consecutive tests classified as stage 1 or worse as per the glaucoma staging system 2). Cox proportional hazards models were used to examine factors (age, sex, treatment status and baseline intraocular pressure (IOP)) associated with conversion.ResultsThe cumulative risk of conversion to POAG was 17.5% (95% CI 15.4% to 19.6%) at 5 years. Older age (HR 1.35 per decade, 95% CI 1.22 to 1.50, p<0.001) was associated with a higher risk of conversion. IOP-lowering therapy (HR 0.45, 95% CI 0.35 to 0.57, p<0.001) was associated with a lower risk of conversion. Predicted 5-year conversion rates for treated and untreated groups were 14.0% and 26.9%, respectively.ConclusionLess than one-fifth of OHT patients managed in glaucoma clinics in the UK converted to POAG over a 5-year period, suggesting many patients may require less intensive follow-up. Our study provides real-world evidence for the efficacy of current management (including IOP-lowering treatment) at reducing risk of conversion.

The granite processing industry generates large amounts of bottom granite dust waste every day. After the drying and heating process of concrete mixture production, the granite dust is blown and collected in the filtering nozzle. This very fine particle granite dry fly dust, with a particle size maximum distribution of 500 μm, can easily be blown away by wind and cause serious environmental impacts. The use of this waste material would be an effective way to reduce such impacts. Therefore, this paper presents an experimental study on the potential of granite dust as a filler in enhancing the mechanical performance of a hybrid basalt/glass (WB/GCSM) composite. The unhole and open hole tensile (UHT and OHT) properties, low velocity impact (LVI) properties, quasi-static indentations (QSI) properties, flexural properties, interlaminar shear stress (ILSS) properties, and morphology of the developed WB/GCSM composites were evaluated. To meet the objective of this study, composite specimens were produced using 1.5–60 μm granite fly dust at three (3) different loadings (1, 3 and 5 wt%). This granite fly dust was incorporated into polyurethane resin using a mechanical stirring technique. The production of FRP laminates then completed using a hand lay-up and vacuum bagging technique. Four types of the WB/GCSM composites systems, i.e., [WB/GCSM], [WB/GCSM/1GD], [WB/GCSM/3GD] and [WB/GCSM/5GD] were fabricated and compared. The analysis results for the mechanical tests revealed that the incorporation of granite dust of up to 3 wt% had increased the UHT, OHT, LVI, QSI, flexural and ILSS properties of all WB/GCSM composites systems. Higher levels of damage tolerance in UHT and OHT tests, and increased ductility index in the LVI test were obtained when granite dust was added up to 5 wt%. However, a remarkable improvement in all mechanical properties was noticed for [WB/GCSM/1GD], which recorded the highest mechanical performance among all WB/GCSM composite systems.

The current drive for sustainability demands recyclable matrices for composite materials. Vitrimers combine thermoset properties with reprocessability, but their mechanical performance in highly loaded applications, for instance, composites for aeronautics, is still to be demonstrated. This work presents the complete mechanical characterization of a new vitrimer reinforced with carbon fiber. This vitrimer formulation consists of functional epoxy groups and a new dynamic disulfide crosslinks-based hardener. The testing campaign for the vitrimer composites encompassed tension, compression, interlaminar shear strength (ILSS), in-plane shear (IPS), open-hole tension (OHT) and compression (OHC), filled-hole compression (FHC) and interlaminar fracture toughness tests under mode I and II. Test conditions included room temperature and high temperature of 70 °C and 120 °C, respectively, after moisture saturation. Tension and flexural tests also were applied on the neat vitrimer resin. The results compared well with those obtained for current aeronautic materials manufactured by Resin Transfer Molding (RTM). The lower values observed in compression and ILSS derived from the thermoplastic veils included as a toughening material. This work demonstrates that the vitrimer formulation presented meets the requirements of current matrices for aeronautic-grade carbon-reinforced composites.

Purpose To evaluate the efficacy, safety, and tolerability of Timogel® preservative-free once daily compared to timolol 0.5% ophthalmic solution bid in patients with ocular hypertension (OHT) and patients with primary open-angle glaucoma (POAG). Methods A total of 75 patients with OHT and patients with POAG treated with timolol 0.5% bid with intraocular pressure (IOP) ≤21 mmHg were enrolled. They underwent complete ophthalmologic examination, IOP measurements (at trough and daytime curve), evaluation of side effects, Schirmer test, break-up time [BUT], blood pressure, heart rate, ocular diastolic perfusion pressure measurements, and acceptance (Comparison of Ophthalmic Medications for Tolerability). Patients switched to Timogel® and were re-evaluated 3 months later. The analysis of variance and the Pearson χ2 tests were used to test differences between the treatments. Results Intraocular pressure reduction at trough was 23.6% with timolol 0.5% and 22.3% with Timogel®. No statistical differences were observed in IOP values at trough and in the daytime curve between the 2 treatments. Local and systemic side effects were less frequent with Timogel® (hazard ratio: p<0.05). Patients demonstrated a significant improvement of Schirmer test and BUT (p<0.05) and a reduction of dryness and foreign body sensation (42.6% vs 15.4%; p<0.01) after switching to Timogel®. Mild and short-lasting blurred vision after Timogel® instillation occurred in about 18.5% of patients. A total of 82% of patients were satisfied or very satisfied with Timogel® vs 61% with previous treatment (p<0.01). Conclusions Timogel® preservative-free dosed once every morning has a 24-hour hypotensive effect with a better safety profile than timolol 0.5% bid and it is well-accepted by patients. The once-daily dosing improved acceptance and compliance.

Purpose: Drugs with cardiovascular protective properties (statins, acetylsalicylic acid (ASS)) were assumed to have positive effects on patients suffering from glaucoma disease. The present retrospective study aimed to investigate the influence of statins, ASS or a combination of both on the glaucoma conversion and progression rate in glaucoma suspects and glaucoma patients with a 20-year follow-up period. Methods: A retrospective analysis of 199 eyes of 120 patients (63 male, 57 female) of the Erlanger Glaucoma Registry (EGR; ClinicalTrials.gov Identifier: NCT00494923; ISSN 2191-5008, CS-2011) was performed considering systemic therapy with statins, ASS or a combination of both: 107 eyes with ocular hypertension (OHT) and 92 eyes with pre-perimetric primary open-angle glaucoma (pre-POAG). All patients received an ophthalmological examination including morphometric and functional glaucoma diagnostics. Glaucoma conversion was defined as the conversion of OHT to pre-POAG. Glaucoma progression was defined as confirmed visual field loss. Data were shown as percentages. Statistical analysis was performed by Chi-Quadrat tests. Results: 1. Glaucoma conversion/progression was observed in 46.7% of the subjects, additionally in combination with hypercholesterinemia in 76.8%. 2. Statins: 27.3% of eyes under systemic statin therapy showed a conversion/progression. Patients taking statins ≥ 10 years yielded a reduced conversion/progression rate (p = 0.028, non-significant after Bonferroni–Holm). 3. ASS: 34.7% of eyes under systemic ASS therapy showed a conversion/progression. A significantly lower conversion/progression rate was observed after ASS therapy ≥ 12 years (p = 0.017, significant after Bonferroni–Holm). 4. ASS and statins: 25.0% of eyes under combined therapy showed a conversion/progression. A significantly reduced conversion/progression rate was reached after 8 years of combined therapy (p = 0.049, non-significant after Bonferroni–Holm). Conclusions: Patients with ocular hypertension and early glaucoma seem to benefit from adjuvant cardiovascular protective therapy. 2588 Background: The aims were to characterize the genetic profiles of UGT1A4, UGT2B7 and UGT2B15in Asian healthy populations (Chinese, Malay, Indians, N=80 each) and investigate the effects of these SNPs on the disposition of TAM in Asian breast cancer patients (N=202). Methods: Healthy Asian subjects and Asian breast cancer patients were genotyped for UGT1A4, UGT2B7 and UGT2B15 SNPs. Plasma levels of tamoxifen and its metabolites in Asian patients were determined via LC-MS/MS analysis. Genotypic-phenotypic differences were examined using non-parametric tests. Haplotypic effects were examined using haplotype-specific generalized linear model. Results: Screening of the 5′ upstream, exonic, exonic-intronic junctions and 3′ downstream regions identified 61, 77 and 47 SNPs in UGT1A4, UGT2B7 and UGT2B15respectively. A total of 16, 14 and 20 tag-SNPs were identified in these genes respectively and genotyped in patients. Haplotypic analysis revealed associations between LD block 1 (-1548A>G, -1531C>T, -419G>A, -219C>T, -163G>A, 142T>G, 448T>C, 804G>A, IVS1+196T>C, IVS1+346T>G, IVS1+414A>G, IVS2+307A>G) and higher plasma Tam-N-Gluc level and MRTAM-N-Gluc/TAM after adjustment for significant covariates. The median (range) MRTAM-N-Gluc/TAM was 2.1- and 1.9-fold higher among patients carrying one and two copies of H2 (GCATAGCACTGG), respectively, compared to patients who carried two copies of H1 haplotype (ACGCGTTGTTAA) [H1/H1 vs H1/H2 vs H2/H2: 0.35 (0.11 - 2.09) vs 0.74 (0.19 - 3.60) vs 0.66 (0.52 - 1.66), P < 0.0001]. Similar association was observed between H2 and Tam-N-Gluc level [H1/H1 vs H1/H2 vs H2/H2: 0.75 (0.15 - 4.45) vs 1.29 (0.19 - 9.20) vs 1.66 (0.62 - 2.83) ng/ml, P= 0.004]. UGT2B15 LD block 3 (1568A>C, *168C>T, *186A>T, *+630C>G and *+888A>G) was associated with modest decrease and increase in (E)-END and MRZ-NDM-4-O-GLUC/E-END, respectively. UGT2B7haplotypes were not associated with O-glucuronidations of 4-OHT and END. Conclusions: This study highlights the involvement of UGT1A4 haplotypes in the variability in TAM N-glucuronidation while UGT2B7 and UGT2B15 variants played limited role in the variabilities of O-glucuronidations of 4-OHT and END in Asian breast cancer patients. The Fontan operation is the current surgical procedure to treat single-ventricle congenital heart disease, by splitting the systemic and pulmonary circulations and thus permitting lifespan to adulthood for the majority of newborns. However, emerging data are showing that Fontan-associated liver disease (FALD) is an increasing related cause of morbidity and mortality in patients with the Fontan circuit. We described the clinical, laboratory, and transient elastography (TE) findings in a case series of adults with the Fontan circuit, and also correlated data with post-mortem histological features, aimed to define the prognostic value of TE in the staging of FALD. All patients presented signs of a long-standing Fontan failure, characterized by reoperation need, systemic ventricle dysfunction, and FALD stigmata (liver and spleen enlargement, portal vein and inferior vena cava dilation, and abnormal liver function tests). Liver and spleen stiffness (LS and SS) values were indicative of significant liver fibrosis/cirrhosis and the presence of suggestive portal hypertension (LS mean 35.9; range 27.3–44.7 kPa; SS mean 42.1, range 32.2–54.5 kPa). Post-mortem evaluations confirmed a gross hepatic architecture distortion in all cases. All patients died from severe complications related to liver dysfunction and bleeding. TE correlated well with pathological findings and FALD severity. We propose this validated and harmless technique to monitor liver fibrosis extension and portal hypertension over time in Fontan patients, and to identify the optimal timing for surgical reoperations or orthotopic-heart transplantation (OHT), avoiding a higher risk of morbidity and mortality in cases with severe FALD.

e17016 Background: Occult Hepatitis B (OHB)and Hepatitis C (OHC) in patients with hematological malignancies (HM) are understudied, mainly because biopsies to identify these viruses in the liver are often not performed due to an associated risk of complications (e.g., bleeding). We describe our retrospective experience of patients with HM tested for OHB and OHC. Methods: Between 09/09 and 11/10, 18 unselected consecutive patients with an abnormal liver panel were tested for both OHB and OHC which was defined as detectable viremia in the absence of HBsAg and anti-HCV, respectively. Severe liver dysfunction was defined as ALT >10 times the upper limit of normal (>560 IU/L) and/or a total bilirubin >6 mg/dL. Plasma samples were tested for HBV DNA and HCV RNA by using real-time PCR (Cobas Taqman HBV and HCV Tests-Roche Molecular Systems). Results: Most patients (12 or 67%) had leukemia, followed by lymphoma (5 patients or 28%). Two patients (11%) had OHB, and none had OHC.The 2 patients with OHB were born in areas with an intermediate to high prevalence of HBV; both had only low grade viremia (<29 IU/mL). At the time HBV DNA testing was performed, peak ALT levels in these 2 patients were 843 IU/L and 6,047 IU/L compared to a median peak ALT of 302 IU/L (range, 84-1,320 IU/L) among those without OHB (P=0.03). Severe liver dysfunction occurred in the 2 (100%) patients with OHB compared to 6 (38%) of the non-OHB cases (P=0.1). Durations of ALT elevation were 12 and 17 days in patients with OHB, compared to a median duration of 13 days (range, 5-73 days) in non-OHB patients. Co-infections were identified in 1 patient with OHB and in 10 (63%) without OHB. The 2 patients with OHB and 8 (50%) patients without OHB died within 6 months after HBV DNA testing. Conclusions: Although our data are limited, in a group of cancer patients in whom a liver biopsy is not available, detection of HBV DNA or HCV RNA in serum in the absence of HBsAg or anti-HCV identifies a subset of patients with occult viral hepatitis, a phenomenon of unclear clinical significance.

The effects of sex steroids on spermatogenesis and testicular androgen secretion were studied in juvenile (spermatogonia present in testes) African catfish. Fish were implanted with Silastic pellets containing 11-ketotestosterone (KT), 11β-hydroxyandrostenedione (OHA), androstenetrione (OA), androstenedione (A), testosterone (T), 5α-dihydrotestosterone (DHT), or estradiol-17β (E2). Control groups received steroid-free pellets. Two weeks later, testis tissue fragments were incubated with African catfish luteinizing hormone (LH) and the amount of OHA secreted in vitro (the main androgen produced by African catfish testes) was quantified. Tissue fragments were then fixed for histological analysis of spermatogenesis. Treatment with KT, OHA, and OA stimulated testicular growth and spermatogenesis (spermatocytes and spermatids were found), whereas T, DHT, A, or E2 had no such effects. All steroids, except for DHT and E2, reduced OHA secretion in the absence and presence of LH to ∼10% of the control values. Previous studies have shown that KT, OHA, and OA have little effect on circulating LH levels in juvenile male African catfish, so that these androgens probably had direct effects on the testis. Inasmuch as OHA, OA, and KT have largely similar effects and because OHA and OA are converted to KT in vivo, we suggest that KT is physiologically the most relevant androgen for the initiation of spermatogenesis in African catfish.

Purpose To evaluate the performance of various tests with automated and subjective evaluation for primary diagnostics of glaucoma in a normal clinical setting. Methods Subjects referred because of suspicion of glaucoma were recruited. All subjects had full ophthalmologic evaluation with stereophotography of the optic nerve head (ONH), red-free retinal nerve fiber layer (RNFL) photography, scanning laser polarimetry (SLP), optical coherence tomography (OCT), and standard automated perimetry (SAP). Analysis of all results was made jointly by 5 glaucoma specialists, with a consensus for each eye as healthy, glaucomatous, or glaucoma suspect. Results from each method were evaluated against this consensus, followed by a subjective evaluation of each method by a glaucoma specialist, comparing this with automated classification by the devices. Results Of the 101 subjects and 202 eyes examined, 23 eyes were by consensus glaucomatous, and 23 were glaucoma suspect. Sensitivity was best with ONH photos and worst with SAP, while SLP had a better sensitivity but poorer specificity than did RNFL photographs and OCT. Subjective evaluation of SLP, OCT, and SAP data gave better sensitivity and specificity than did classification by numeric values from the devices only, with OCT performing better in automatic classification than did SLP or SAP. Conclusions None of the current methods is superior to others in diagnosing glaucoma, and the accuracy of automated tests was better when subjectively evaluated by an experienced ophthalmologist. Diagnosis of glaucoma should be based on a combination of test results interpreted by a clinician.

The authors report a case of a dysgenetic male pseudohermaphroditism with a 45,X/46,XY karyotype in a mosaic form, which was diagnosed in an infant. The one-week-old infant was evaluated because of proximal hypospadias and retention of the right testis. The results of hormonal tests were the followings: serum FSH 5.2 mU/ml; LH: 2.0 mU/ml; testosterone: 144.3 ng/dl; androstendione: 0.42 µg/l; 17-hydroxyprogesterone: 1.12 ng/ml. Chromosomal analysis revealed 45,X/46,XY karyotype. Fluorescent in vitro hybridization showed that 51% of the lymphocytes had the Y chromosome and the SRY gene. Analysis of the SRY showed no deletion in the AZF a,b,c regions. Pelvic magnetic resonance imaging indicated the presence of vagina between the bladder and the rectum, and it showed a mass measuring 15×8 mm in the right inguinal canal as well as an oval gonadal mass with a size of 13×7 mm in the left scrotum. During surgical intervention, performed at the age of one, the right gonad was removed and biopsy of the scrotal testis was performed. Histological examination revealed dysgenetic testis in both sides. The authors emphasize the necessity of cytogenetic and endocrinological investigations of newborns with perineoscrotal hypospadia and bilateral or unilateral maldescent testes immediately after birth. Surgical removal of the dysgenetic testicular tissue located in the abdominal cavity and its histological evaluation provides separation of mixed gonadal dysgenesis, dysgenetic male pseudohermaphroditism, bilateral gonadal dysgenesis and ovotestis in the 45,X/46,XY mosaic cases. Avaliamos as concentrações de GH, IGF-1 e IGFBP-3 em 10 pacientes com acromegalia em atividade, antes e após tratamento com octreotida subcutânea (OCT-sc) e LAR (OCT-LAR). Verificamos o valor preditivo dos testes agudo e de curto prazo (após 21 dias) na resposta do GH e IGF-1. As avaliações de médio prazo foram realizadas após 6 meses de tratamento com cada formulação. Os valores de GH (µg/l; IFMA), nadir de GH no oGTT e IGF-1 (µg/l; IRMA) pré-tratamento foram 13,9±6,3; 11,4±6,3; 717±107, respectivamente. Os tratamentos de 21 dias com OCT-sc ou OCT-LAR reduziram os níveis de GH (2,9±1,1 e 4,4±1,2) e IGF-1 (491±80 e 512±80). Redução após 6 meses de tratamento foi similar com as duas formulações: GH basal (2,8±0,9 e 1,9±0,5), nadir GH-GTT (1,6±0,4 e 1,6±0,5) e IGF-1 (583±107 e 515±83). A IGFBP-3 não foi bom parâmetro para avaliação destes pacientes. O teste agudo não foi indicador da ocorrência de efeitos colaterais. Os testes, agudo e de curto prazo, foram capazes de predizer a resposta do GH ao tratamento crônico com a OCT-sc e OCT-LAR.

To analyze in mild Alzheimer’s disease (MAD) patients, GDS-4 (Reisberg Scale), whether or not some psychophysical tests (PTs) support OCT macular findings in the same group of MAD patients reported previously.Methods. Twenty-three MAD patients and 28 age-matched control subjects with mean Mini Mental State Examination of 23.3 and 28.2, respectively, with no ocular disease or systemic disorders affecting vision were included. Best-corrected visual acuity (VA), contrast sensitivity (CS) (3, 6, 12, and 18 cpds), color perception (CP), and perception digital test (PDT) were tested in one eye of each patient.Results. In comparison with the controls, MAD patients presented (i) a significant decrease in VA, PDT, and CS for all spatial frequencies analyzed, especially the higher ones, and (ii) a significant increase in unspecific errors on the blue axis (P<0.05in all instances). In MAD patients, a wide aROC curve was plotted in all PTs.Conclusions. In MAD, CS, VA, and the tritan axis in CP were impaired. The PTs with the greatest predictive value are the higher spatial frequencies in CS and tritan unspecific errors in CP. PT abnormalities are consistent with the structural findings reported in the same MAD patients using OCT.

Laboratory animals often develop abnormal repetitive (stereotypic) behaviour that can influence both physiology and behavioural test results. Such abnormal behaviours usually develop in suboptimal environment and increase over time. To explore the development of stereotypic forms of activity night home-cage behaviour of laboratory mice was analysed and collated with the behaviour in open field (OFT), hole-board (HBT) and tail suspension (TST) tests twice within 16 weeks. Mice expressed few stereotypies and their duration lessened over time from median 8.2 to 1.0 %. In contrast, grooming behaviour increased significantly from 29.5 to 49.6 %. Home-cage grooming correlated with the latency of locomotion start in OFT and with the immobility time in TST. Intensity and stability of stereotypic activity and grooming inf­luenced the duration of grooming in OFT: the mice with unstable stereotypies groomed more than others, and mice with the lowes home-cage grooming level also groomed in OFT the most. Intensity and stability of night grooming influenced the behaviour in TST: the mice with unstable level of grooming were the most mobile in this test. Abnormal home-cage activity may indicate impaired welfare, and that, in turn, may affect test activity, so researchers need to keep it in mind when planning animal behaviour experiments.

Eight oat (Avena sativa L.) cultivars grown at 27 locations during 1980, 1981, 1982, 1983 and 1984 were used to study the effect of genotype-by-environment interactions and their implications on oat cultivar testing and recommendation for Alberta. The study showed that accurate regional performance results would be obtained only if Alberta was divided into at least six rain-fed and two irrigated areas. The study also showed that the number of locations, years and replications that are currently used in some of the regional testing areas of Alberta are insufficient to detect (at P = 0.05) a 5% yield difference between the standard check (Cascade) and a test cultivar. The effects of increasing or decreasing the number of locations, years and replications on the precision of future regional oat tests in Alberta are discussed.Key words: Avena sativa L., oats, cultivar-environment interactions, regional tests

Objective Retinal structural changes which were investigated by optical coherence tomography (OCT) have been reported in schizophrenia. Since cognitive dysfunction is a core feature of schizophrenia, the correlations between retinal findings and cognitive functions of patients and their healthy siblings may provide insight into the pathophysiological processes of the disorder. We aimed to investigate the relationship between neuropsychiatric tests and retinal changes in schizophrenia patients and their healthy siblings.Methods We measured OCT parameters and cognitive performance (via Trail Making Tests, verbal fluency tests, and The Digit Span Tests) of 72 participants (36 patients with schizophrenia and 36 healthy siblings) and disease severity (with Positive and Negative Syndrome Scale, Global Assessment of Functioning, and Clinical Global Impression scales) in patients with schizophrenia and evaluated the relationship between retinal findings and clinical parameters, especially neurocognitive tests.Results We found decreased ganglion cell layer-inner plexiform layer thickness and macular volume in the patient group. There were strong correlations between neurocognitive tests and OCT findings in both groups. On the other hand, there was not any correlation between retinal findings and disease parameters.Conclusion The cognitive symptoms of schizophrenia may be more closely related to structural changes in the retina.

Abstract ‘Vates’ collards were direct seeded into double-row beds on 9 Sep and later thinned to 4 inches apart within rows. Each plot was 17 ft long and consisted of 3 36-inch beds with 2 empty beds between plots. The treatments were arranged in a complete randomized block design with 4 replicates. Spray treatments were applied at a rate of 25 gal/acre using a CO2-powered backpack sprayer equipped with a 20-inch boom and 2 Teejet size 18 nozzles angled inward. Eight sprays were applied on weekly starting 4 Oct. Efficacy was determined 6 or 7 d after each application by counting the larvae and pupae of CL and the larvae of SCW on 15 plants/plot.

Purpose. To evaluate the diagnostic accuracy of retinal ganglion cell (RGC) counts as estimated by combining data from standard automated perimetry (SAP) and spectral domain optical coherence tomography (SD-OCT).Methods. Healthy individuals and glaucoma patients were included in this cross-sectional study. All eyes underwent 24-2 SITA SAP and structural imaging tests. RGC count estimates were obtained using a previously described algorithm, which combines estimates of RGC numbers from SAP sensitivity thresholds and SD-OCT retinal nerve fiber layer (RNFL) average thickness.Results. A total of 119 eyes were evaluated, including 75 eyes of 48 healthy individuals and 44 eyes of 29 glaucoma patients. RGC count estimates performed better than data derived from SD-OCT RNFL average thickness or SAP mean deviation alone (area under ROC curves: 0.98, 0.92, and 0.79;P<0.001) for discriminating healthy from glaucomatous eyes, even in a subgroup of eyes with mild disease (0.97, 0.88, and 0.75;P<0.001). There was a strong and significant correlation between estimates of RGC numbers derived from SAP and SD-OCT (R2=0.74;P<0.001).Conclusion. RGC count estimates obtained by combined structural and functional data showed excellent diagnostic accuracy for discriminating the healthy from the glaucomatous eyes and performed better than isolated structural and functional parameters.

"ABSTRACT. Introduction: Coaches should be able to estimate successfully different physical attributes of junior tennis player’s performance such as power, in order to monitor players’ progress and to design the most appropriate training program. However, this process requires reliable and valid field tests. Objective: Aim of this study was to examine absolute and relative reliability of overhand ball throw (OBT) test within testing session and to investigate its validity. Methods: 257 Hungarian junior boy and girl tennis players (aged 11- 17) separated into four groups, performed OBT and serve speed (SS) tests of standardised protocol. Results: Dependent sample t-test revealed no significant (p= 0.31-57>0.05) difference between test and retest sample means within testing session and magnitude of effect size (dz=0.1-0.5) were trivial for all groups. Furthermore, all groups had low typical percentage error (CV= 3-4 %), and standard error of measurement values was consistently low (SEM= 0.12- 0.18). Within test-retest consistency illustrated strong relative reliability (ICC= 0.98-0.99). Moreover, significantly large to very large positive correlations were found between OBT and SS (r= 0.57-0.81; p˂0.01) tests. The coefficient of determination indicated that OBT explained 32-65% of the SS for groups. Conclusions: These findings suggest that absolute and relative reliability of OBT test is high within testing session and validity of OBT test is acceptable for measuring power ability of flat serve execution in junior tennis players."

Summary The suppressive effect of a black oat, Avena strigosa, breeding line KH1a on Meloidogyne spp. was examined in pot tests and on Meloidogyne incognita in pot and field tests. In pot tests, roots of black oats were examined 42-46 days after inoculation of 500 second-stage juveniles. There were significantly fewer egg masses on the roots of KH1a than on 12 black oat cultivars examined. KH1a was a poor host for four isolates of M. incognita, two isolates of M. arenaria and one isolate of M. javanica, and a non-host for one isolate of M. hapla. The effect of autumn and spring cropping of KH1a on soil nematode density was examined in M. incognita-infested fields. Nematode density after autumn cropping of KH1a was significantly lower than that after susceptible black oat, resulting in significantly lower Pf/Pi in KH1a (0.10 and 0.13) than in susceptible black oat (0.42 and 0.74). Damage indices of the succeeding crop, sweet potato, were significantly lower in KH1a plots than in susceptible black oat plots. In spring cropping, there were significantly fewer soil nematodes in KH1a plots than in susceptible black oat plots 3 weeks after cultivation. In both autumn and spring cropping, there was no significant difference in soil nematode density between KH1a and bare fallow. These results suggested that KH1a is a good alternative to current susceptible cultivars for the management of M. incognita.

The clinical progression of small-cell lung cancer (SCLC) remains pessimistic. The aim of the present study was to promote the understanding of the clinical significance and mechanism of O-linked N-acetylglucosamine (GlcNAc) transferase (OGT) in SCLC. Wilcoxon tests, standardized mean difference (SMD), and Kruskal–Wallis tests were utilized to compare OGT level differences among the experimental and control groups. The univariate Cox regression analysis, Kaplan–Meier curves, and receiver operating characteristic curves were applied to determine OGT’s clinical relevance in cancers. The Spearman correlation analysis and enrichment analysis were utilized to explore the underlying mechanisms of OGT in cancers. For the first time in the field, we provide an overview of OGT in 32 cancers using a large number of samples (n = 21,196), determining distinct OGT expression in 25 cancers and its prognosis effects in 12 cancers. Furthermore, using 950 samples from multiple sources, upregulated OGT was found in both mRNA and protein levels in SCLC (SMD = 0.93, 95% CI [0.24, 1.63]). Higher OGT levels represented a more unfavorable disease-free interval for SCLC patients ( p < 0.001 ). The research also identified OGT expression as a potential marker for SCLC prediction (sensitivity = 0.79, specificity = 0.86, and AUC = 0.88). The high expression of OGT in SCLC may result from the positive regulation of two transcription factors—DEK and XRN2. We primarily investigated the underlying mechanisms of OGT in SCLC. Herein, based on the analyses from pan-cancer to SCLC, OGT demonstrated conspicuous clinical significance. OGT may be an underlying biomarker for the treatment and identification of some cancers, including SCLC.

Abstract The in-plane shear strengths (IPSS) of copy paper, filter paper, and sack paper were obtained from an asymmetric four-point bending (AFPB) test. Rectangular tabs of medium-density fibreboard (MDF) were bonded to the paper specimen. The length of the clearance between the tabs was varied, and the influence of the clearance on the IPSS value was investigated. The IPSS obtained from the AFPB test was compared with that obtained from a 35° off-axis tension (OAT) test, which was proposed in a previous study. The IPSS values obtained from the AFPB tests on the copy paper and sack paper were significantly lower than those obtained from the 35° OAT tests because of the localised buckling caused by the negative principal stress. In contrast, the IPSS values obtained from the AFPB and OAT tests on the filter paper were in agreement when the clearance length was >5 mm because the buckling effect was excluded.

An isolate of Magnaporthe grisea was collected from a blast lesion on oat in Brazil. Sequence analysis of the rDNA-ITS-2 region and DNA fingerprinting with repetitive elements revealed that the Avena isolate belongs to the "crop isolate group" and is similar to Triticum isolates. At high temperature (28°C), the Avena isolate caused severe disease symptoms on primary leaves of oat and wheat. When the temperature was decreased to 20°C, wheat leaves expressed resistance to the Avena isolate. Cytologically, this temperature-dependent resistance was associated with an increase in the incidences of papilla formation and a hypersensitive reaction. Pathogenicity tests with various plant species at 20°C revealed that the Avena isolate is exclusively parasitic on oat. To elucidate genetic mechanisms of this species-specific parasitism, the Avena isolate was crossed with a Triticum isolate and resulting F1 progenies were subjected to pathogenicity tests on oat seedlings. In the F1 population, avirulent and virulent cultures segregated in a 1:1 ratio, suggesting that the specific parasitism on oat is controlled by a single gene. Aim: to assess correlations between ocular ultrasound (US) biometry and optical coherence tomography (OCT) and hemoglobin A1c (HbA1c) in children with type 1 diabetes (T1D). Patients and Methods: 79 children were enrolled. The control group included 52 age-matched healthy children. Axial length (AL, mm) and lens thickness (LT, mm) were measured by US. Central corneal thickness (CCT) and central macular thickness (CMT) were measured by OCT (Cirrus 2000, Carl Zeiss Meditec, Germany). Correlation analysis to assess the relationship between HbA1c, US, and OCT was performed. Results: 55.7% of the participants were boys (n=44) and 44.3% were girls (n=35). Children were assigned into six groups depending on HbA1c levels. Group 1 (HbA1c <6.0%) included 8 children, group 2 (HbA1c 6.1–7.0%) 29 children, group 3 (HbA1c 7.1–8.0%) 21 children, group 4 (HbA1c 8.1–9.0%) 8 children, group 5 (HbA1c 9.1–10%) 5 children, and group 6 (HbA1c >10%) 8 children. The mean AL was 22.7±1.1 mm. The greatest AL was reported in groups 2 and 4, while the least AL was reported in groups 3 and 6. The greatest LT was reported in groups 4 and 6. In group 1, LT was similar to those in the control group. In all groups but group 2, CCT was greater than in controls. CMT demonstrated a negative correlation with HbA1c levels in the study groups. CMT was the greatest in children with T1D of 5 years’ durati Conclusion: in children with T1D, US and OCT findings correlate with HbA1c levels and T1D duration. Ocular biometry, CCT, and CMT measurements are useful tests illustrating the level of compensation of systemic disease.
