Brazil has presented tremendous progress in non-surgical embryo transfer (NSET) in sheep and goats. New instruments and techniques for non-surgical embryo recovery (NSER) and NSET in small ruminants were implemented. Recent improvements include refinement of the protocols for cervical relaxation combining oestradiol–oxytocin–cloprostenol treatment at specific times before NSER in sheep; recipient goats do not require any hormonal drugs to induce cervical dilation and direct embryo transfer by the cervical route yields excellent results. Transrectal ovarian ultrasonography (B-mode but especially colour Doppler) have proven to be accurate methods to localise and enumerate corpora lutea and luteinised unovulated follicles in recipient and donor does and ewes. An array of new criteria for selecting superior animals for NSER and NSET (e.g. cervical mapping) have been developed by Brazilian researchers. Extensive studies on both technologies were initially conducted in commercial breeds of goats and sheep but have been gradually extended to some native breeds of sheep (germplasm conservation) and dairy goat operations. It is speculated that, in future, NSER and NSET may become methods of choice for caprine and ovine embryo recovery and transfer in Brazil, and then globally. Due primarily to the efficiency of NSET in goats, a novel interspecies (e.g. bovine) IVP method may soon be developed on a large scale. The Brazilian experience is an invaluable source of information and know-how promoting the replacement of conventional surgical assisted reproductive technologies with non-surgical procedures and hence supporting the rapid development of the embryo transfer industry in small ruminants.

AbstractAs nanoscale science, engineering, and technology (NSET) becomes more integrated into precollege science curricula, it is crucial for teachers to develop coherent understandings of science principles (e.g., the structure of matter, size and scale, forces and interactions, and size-dependent properties) that allow them to coordinate these understandings from the macro- to the nanoscale. Furthermore, as teachers acquire new NSET content knowledge through professional learning opportunities, it is incumbent upon NSET educators to understand their developing content knowledge. To this end, we report results from a study in which we used a pre-/post-/delayed-posttest design to examine the change in 24 secondary (grades 7–12) science teachers’ NSET content knowledge as a result of their participation in a year-long professional development program that consisted of a 2-week intensive course and academic year follow-up activities. Participants showed significant gains from pretest to posttest and significant gains on the delayed test compared to the pretest. We also present trends that emerged in teachers’ open-ended responses that provided deeper insight into teachers’ NSET content knowledge. Finally, we discuss issues related to the assessment of teachers’ NSET content knowledge as well as the design of NSET professional development for teachers.

ContextEctopic ACTH syndrome (EAS) is principally associated with aggressive malignant tumors but also with neuroendocrine tumors of good prognosis. Recently, rare nonhepatocytic nested stromal and epithelial tumors (NSET) were characterized by their possible association with Cushing's syndrome of which biochemical and physiopathological features were still incompletely studied.ObjectiveTo describe the clinical and hormonal characteristics of an EAS originating from a liver NSET and further understand the mechanism of cortisol overproduction.Design and settingThis is a clinical case report from the Endocrinology Department of Caen University Hospital, France.Patient and interventionA 17-year-old female patient was found to have a large liver NSET with mild Cushingoid clinical features and intense biological hypercortisolism but moderate ACTH secretion. Resection of the tumor was curative with a 30-month follow-up.ResultsThe epithelial component of the tumor coexpressed ACTH mildly, corticotropin-releasing hormone (CRH) strongly, and 11β-hydroxysteroid dehydrogenase at a level comparable with normal human hepatocytes.ConclusionsLiver NSET is a new cause of EAS, which may evoke hypercortisolism by multiple biochemical pathways.

This paper investigated the impact of the Nurturing talent in Science, Engineering and Technology (NSET) programme on teaching practices in rural schools within the Nkowankowa cluster in the Limpopo Province. Though NSET programme focused on Science, Mathematics and Technology, this study will only concentrate on Physical Sciences. Out of the population of 68 Science teachers, 30 participated in the completion of questionnaires and nine were interviewed. A mixed-methods approach with an exploratory design was used in this study to investigate the impact of the NSET programme on learner performance in Physical Sciences (focusing on teacher content and pedagogical delivery). For learners to consistently perform better, then one need to focus on teacher development in terms of content and pedagogy. The model that focuses on teacher development in order to address the performance of learners helps to sustain better results at school than focusing directly on learners since the teachers are likely to interact with more learners and stay at the same school longer. Questionnaires, interviews and document analysis were used as data collection methods. According to the research findings, the NSET programme had a positive impact on both the content and pedagogical delivery of teachers. This was revealed through the confidence educators gained and their ability to come up with new teaching strategies and improved teaching practices. The positive impact of the NSET programme is also noted through learners’ improved academic performance in Physical Science. The NSET programme conducted within the Nkowankowa cluster yielded positive results and the Limpopo Department of Basic Education (DBE) can adopt it as one of the best practices in the development of teachers and improvement of results in their schools.

The structural maintenance of chromosomes (SMC) family of proteins play key roles in the organization, packaging, and repair of chromosomes. Cohesin (Smc1+3) holds replicated sister chromatids together until mitosis, condensin (Smc2+4) acts in chromosome condensation, and Smc5+6 performs currently enigmatic roles in DNA repair and chromatin structure. The SMC heterodimers must associate with non-SMC subunits to perform their functions. Using both biochemical and genetic methods, we have isolated a novel subunit of the Smc5+6 complex, Nse3. Nse3 is an essential nuclear protein that is required for normal mitotic chromosome segregation and cellular resistance to a number of genotoxic agents. Epistasis with Rhp51 (Rad51) suggests that like Smc5+6, Nse3 functions in the homologous recombination based repair of DNA damage. We previously identified two non-SMC subunits of Smc5+6 called Nse1 and Nse2. Analysis of nse1-1, nse2-1, and nse3-1 mutants demonstrates that they are crucial for meiosis. The Nse1 mutant displays meiotic DNA segregation and homologous recombination defects. Spore viability is reduced by nse2-1 and nse3-1, without affecting interhomolog recombination. Finally, genetic interactions shared by the nse mutants suggest that the Smc5+6 complex is important for replication fork stability.

Nested stromal-epithelial tumor (NSET) of the liver is an extremely rare primary hepatic tumor with uncertain malignant potential. To date, only 11 cases have been described. We describe the case of a 2 1/2-year-old girl with an incidental liver mass. The mass was discovered on follow-up abdominal imaging for asymptomatic hydronephrosis diagnosed on antenatal ultrasound. Needle biopsy showed a mixed stromal and epithelial process in a nested pattern, with foci of ossification and no significant pleomorphism or necrosis. The nest cells stained with WT-1, cytokeratin 18, and CD56. Ossifying stromal epithelial tumor of the liver was strongly suspected. The findings were confirmed in the subsequent partial hepatectomy specimen. To our knowledge, this is the 12th case of NSET in the English-language literature and the 3rd case of NSET associated with genitourinary system abnormalities. Possible associations with dysregulated WT-1 expression are discussed.

ABSTRACT The Schizosaccharomyces pombe SMC proteins Rad18 (Smc6) and Spr18 (Smc5) exist in a high-M r complex which also contains the non-SMC proteins Nse1, Nse2, Nse3, and Rad62. The Smc5-6 complex, which is essential for viability, is required for several aspects of DNA metabolism, including recombinational repair and maintenance of the DNA damage checkpoint. We have characterized Nse2 and show here that it is a SUMO ligase. Smc6 (Rad18) and Nse3, but not Smc5 (Spr18) or Nse1, are sumoylated in vitro in an Nse2-dependent manner, and Nse2 is itself autosumoylated, predominantly on the C-terminal part of the protein. Mutations of C195 and H197 in the Nse2 RING-finger-like motif abolish Nse2-dependent sumoylation. nse2.SA mutant cells, in which nse2.C195S-H197A is integrated as the sole copy of nse2, are viable, whereas the deletion of nse2 is lethal. Smc6 (Rad18) is sumoylated in vivo: the sumoylation level is increased upon exposure to DNA damage and is drastically reduced in the nse2.SA strain. Since nse2.SA cells are sensitive to DNA-damaging agents and to exposure to hydroxyurea, this implicates the Nse2-dependent sumoylation activity in DNA damage responses but not in the essential function of the Smc5-6 complex.

ANNs are only accurate for the scope of the given training data which is not suitable for real life impact localisation due to the large range of possible impact variation. Impact data was collected for a variation of impact cases (angle, mass and energy) on a sensorized curved composite panel. From observation of the obtained data, a robust signal Time of Arrival (TOA) extraction method is proposed using a Normalised Smooth Envelope Threshold (NSET) which is a modification of the currently known Normalised Threshold (NT) method. Two ANNs were trained using TOA extracted with the NT and NSET method from a baseline case and tested with TOA extracted from cases having added variation of impact condition. The results show that the proposed NSET method results in more accurate results for impact cases different to the training case and thus allows for only a single impact training case to accurately predict cases with multiple variation. This enhances the applicability of ANNs for impact localisation in real life conditions.

ABSTRACT The rad18 gene of Schizosaccharomyces pombe is an essential gene that is involved in several different DNA repair processes. Rad18 (Smc6) is a member of the structural maintenance of chromosomes (SMC) family and, together with its SMC partner Spr18 (Smc5), forms the core of a high-molecular-weight complex. We show here that both S. pombe and human Smc5 and -6 interact through their hinge domains and that four independent temperature-sensitive mutants of Rad18 (Smc6) are all mutated at the same glycine residue in the hinge region. This mutation abolishes the interactions between the hinge regions of Rad18 (Smc6) and Spr18 (Smc5), as does mutation of a conserved glycine in the hinge region of Spr18 (Smc5). We purified the Smc5-6 complex from S. pombe and identified four non-SMC components, Nse1, Nse2, Nse3, and Rad62. Nse3 is a novel protein which is related to the mammalian MAGE protein family, many members of which are specifically expressed in cancer tissue. In initial steps to understand the architecture of the complex, we identified two subcomplexes containing Rad18-Spr18-Nse2 and Nse1-Nse3-Rad62. The subcomplexes are probably bridged by a weaker interaction between Nse2 and Nse3.

Structural Maintenance of Chromosomes (SMC) complexes are important for many aspects of the chromosomal organization. Unlike cohesin and condensin, the SMC5/6 complex contains a variant RING domain carried by its Nse1 subunit. RING domains are characteristic for ubiquitin ligases, and human NSE1 has been shown to possess ubiquitin-ligase activity in vitro. However, other studies were unable to show such activity. Here, we confirm Nse1 ubiquitin-ligase activity using purified Schizosaccharomyces pombe proteins. We demonstrate that the Nse1 ligase activity is stimulated by Nse3 and Nse4. We show that Nse1 specifically utilizes Ubc13/Mms2 E2 enzyme and interacts directly with ubiquitin. We identify the Nse1 mutation (R188E) that specifically disrupts its E3 activity and demonstrate that the Nse1-dependent ubiquitination is particularly important under replication stress. Moreover, we determine Nse4 (lysine K181) as the first known SMC5/6-associated Nse1 substrate. Interestingly, abolition of Nse4 modification at K181 leads to suppression of DNA-damage sensitivity of other SMC5/6 mutants. Altogether, this study brings new evidence for Nse1 ubiquitin ligase activity, significantly advancing our understanding of this enigmatic SMC5/6 function.

Structural maintenance of chromosomes (SMC) complexes are critical chromatin modulators. In eukaryotes, the cohesin and condensin SMC complexes organize chromatin, while the Smc5/6 complex directly regulates DNA replication and repair. The molecular basis for the distinct functions of Smc5/6 is poorly understood. Here, we report an integrative structural study of the budding yeast Smc5/6 holo-complex using electron microscopy, cross-linking mass spectrometry, and computational modeling. We show that the Smc5/6 complex possesses several unique features, while sharing some architectural characteristics with other SMC complexes. In contrast to arm-folded structures of cohesin and condensin, Smc5 and Smc6 arm regions do not fold back on themselves. Instead, these long filamentous regions interact with subunits uniquely acquired by the Smc5/6 complex, namely the Nse2 SUMO ligase and the Nse5/Nse6 subcomplex, with the latter also serving as a linchpin connecting distal parts of the complex. Our 3.0-Å resolution cryoelectron microscopy structure of the Nse5/Nse6 core further reveals a clasped-hand topology and a dimeric interface important for cell growth. Finally, we provide evidence that Nse5/Nse6 uses its SUMO-binding motifs to contribute to Nse2-mediated sumoylation. Collectively, our integrative study identifies distinct structural features of the Smc5/6 complex and functional cooperation among its coevolved unique subunits.

In this study we compare the effectiveness of the Normalised Smoothed Envelope (NSET) method for Artificial Neural Network (ANN) based impact localisation under simulated environmental and operational conditions with respect to other ANN based localisation methods developed by other studies. It is shown that when the testing and training impact case is the same, most studies give comparably good accuracy of localisation irrespective of feature extraction method or structure geometry. However, when the testing and training impact cases are not the same, only the NSET method is able to negate the variations caused by various impact cases and provide good localisation accuracy for an ANN trained using only a single impact case thus allowing for smaller training data set size requirements and increasing feasibility for real life application.

The Smc5-Smc6 holocomplex plays essential but largely enigmatic roles in chromosome segregation, and facilitates DNA repair. The Smc5-Smc6 complex contains six conserved non-SMC subunits. One of these, Nse1, contains a RING-like motif that often confers ubiquitin E3 ligase activity. We have functionally characterized the Nse1 RING-like motif, to determine its contribution to the chromosome segregation and DNA repair roles of Smc5-Smc6. Strikingly, whereas a full deletion of nse1 is lethal, the Nse1 RING-like motif is not essential for cellular viability. However, Nse1 RING mutant cells are hypersensitive to a broad spectrum of genotoxic stresses, indicating that the Nse1 RING motif promotes DNA repair functions of Smc5-Smc6. We tested the ability of both human and yeast Nse1 to mediate ubiquitin E3 ligase activity in vitro and found no detectable activity associated with full-length Nse1 or the isolated RING domains. Interestingly, however, the Nse1 RING-like domain is required for normal Nse1-Nse3-Nse4 trimer formation in vitro and for damage-induced recruitment of Nse4 and Smc5 to subnuclear foci in vivo. Thus, we propose that the Nse1 RING-like motif is a protein–protein interaction domain required for Smc5-Smc6 holocomplex integrity and recruitment to, or retention at, DNA lesions.

Abstract Cu nanoparticles and 2D-MoS2 nanosheets have been synthesised and the morphology of the prepared samples has been characterised using high-resolution transmission electron microscopy and field emission scanning electron microscopy. Steady-state fluorescence studies exhibit quenching of fluorescence intensity upon the addition of quencher in varying amounts. Time-resolved fluorescence studies show a reduction of fluorescence lifetime in the presence of quencher. A thorough investigation of energy transfer between MoS2 and copper nanoparticles has been accomplished using steady-state and time-resolved fluorescence studies. The results are more consistent with the nanometal surface energy transfer (NSET) theory, which follows a 1/d 4 distance dependence than the Förster resonance energy transfer theory exhibiting 1/d 6 distance dependence. For the MoS2–Cu pair, the NSET efficiency, the proximal distance between donor–acceptor pairs and the rate of energy transfer has been explored. Energy transfer-based fluorescent biosensors have been regarded as potential candidates in biosensing applications. Glutathione, an endogenous tripeptide compound, plays a vital role as an antioxidant in the human body. Irregular glutathione levels have been linked to several adverse illnesses. Henceforth, detection of glutathione levels is indispensable. The Cu–MoS2 pair was used for glutathione sensing in nanomolar concentration (nM), which demonstrated a turn-off sensing behaviour.

Disaster education allows community members to initiate pre-disaster measures at the individual level. The National Society for Earthquake Technology (NSET) has initiated a school safety program in selected government schools in Nepal, one of the most disaster prone countries. This study outlines the current condition of disaster education in Nepal, identifies teachers’ levels of awareness and evaluates the effect of NSET's work. A survey of 130 teachers was conducted in over 40 schools in which it was found that while a level of disaster education in Nepal is widespread, it has not been implemented systematically but depends on the awareness of individual teachers. More teachers in NSET project schools provided disaster education than in the typical government and private schools that were observed. However, the quality of education provided was found to be the same. The teaching focused particularly on the effects of disasters in which the teachers had personal experience. These topics were often covered even if they were not included in the textbooks. The majority of teachers surveyed reported a need for curriculum development in this area. However, considering the results listed above, it can be considered that teacher training is the most important step to improve disaster education in Nepal.

The major earthquake in April 25, 2015 of Mw 7.8 and aftershock of intensity Mw 7.3 on May 12, 2015 has caused not only a substantial death toll and huge economic losses, but also heavy damage to many buildings. This paper outlines the common observed damage patterns of different types of buildings in Kathmandu valley induced by the earthquake and their constructional deficiencies..We visited Department Of Urban Development and Building Construction (DUDBC) of Nepal Government, and Nepal Society for Earthquake Technology (NSET) and got various information regarding structural damages caused by Gorkha earthquake. After acquiring knowledge on this topic through internet and from NSET and DUDBC, the structural failure analysis of buildings affected during the earthquake in Kathmandu Valley was done by photo observation. Both unreinforced masonry buildings and reinforced masonry structures suffered low to heavy destruction. The construction and structural deficiencies were identified to be the major cause of failure, however local soil amplification, foundation problems, liquefaction associated damages and local settlement related damages were also significantly observed during this earthquake and reported in the paper. The Gorkha earthquake sequences delivered unprecedented opportunity to augment the understanding on seismic performance of the buildings. This paper is also motivated to point out the faintness in the past to current constructional practice of buildings, provide preventive measure and convey awareness to stake holders for future safer building construction practice.

The April 2015 Gorkha earthquake in Nepal revealed the relative effectiveness of the Nepal Standard or the national building code (NBC), and irregular compliance with it in different parts of Nepal. Much of the damage to more than half a million residential structures in Nepal may be attributed to the prevalence of owner-built or owner-supervised construction and the lack of owner and builder responsiveness to seismic risk and training in the appropriate means of complying with the NBC. To explain these circumstances, we review the protracted implementation of the NBC and the role played by one organization, the National Society for Earthquake Technology—Nepal (NSET), in the implementation of the NBC. We also share observations on building code compliance made by individuals in Nepal participating in workshops led by the Earthquake Engineering Research Institute's 2014 class of Housner Fellows.

The primary purpose was to examine changes in balance, lower extremity (LE) power and flexibility following 10 weeks of taekwondo (TKD) training and to determine if this was different in children classified as healthy weight (HW) and overweight (OW)/obese. Participants included 17 children (HW: n = 11, OW/obese: n = 6). Data were collected on balance, LE power and flexibility at baseline and 10 weeks. Balance was assessed with eyes open in normal (NSEO), tandem (TSEO), single (SLEO) stances and with eyes closed for normal (NSEC) and tandem (TSEC) stances. Center of pressure displacements in mediolateral (Xavg) and anteroposterior (Yavg) directions; and average velocity (Vavg) were calculated. Analyses included two-way ANOVA and Mann Whitney U tests (P < 0.05). Balance data indicated significant interaction effects for Xavg in NSEO, Yavg in TSEO; time effects for Yavg in NSEO, NSEC and SLEO and Vavg in SLEO conditions. A significant group effect was shown for Vavg in the NSEO, NSEC and TSEO and for Yavg in TSEC conditions. Flexibility decreased significantly with TKD. Findings suggest that 10 weeks of TKD training may improve balance in children, and OW/obese group may have greater improvements in balance with eyes open compared to their peers.

Through their recommendation, we visited National Society of Earthquake Technology (NSET) and Earthquake Safety Solution (ESS) which gave us chance to meet with Daniell King and Harry Toward who were researching in Nepal about base isolation method. We visited a village named Sankhu ward No.4 near Nepal Engineering College. We asked people how feasible this project can be. Many people showed positive responses towards it. Then we went to Sankhu Municipality for the collection of secondary data. So especially in developing countries like Nepal, this project will give a lot of benefit. Further it is expected to give better results than other resistive methods.

ABSTRACT Stabilization and processing of stalled replication forks is critical for cell survival and genomic integrity. We characterize a novel DNA repair heterodimer of Nse5 and Nse6, which are nonessential nuclear proteins critical for chromosome segregation in fission yeast. The Nse5/6 dimer facilitates DNA repair as part of the Smc5-Smc6 holocomplex (Smc5/6), the basic architecture of which we define. Nse5-Nes6 (Nse5/6) mutants display a high level of spontaneous DNA damage and mitotic catastrophe in the absence of the master checkpoint regulator Rad3 (hATR). Nse5/6 mutants are required for the response to genotoxic agents that block the progression of replication forks, acting in a pathway that allows the tolerance of irreparable UV lesions. Interestingly, the UV sensitivity of Nse5/6 mutants is suppressed by concomitant deletion of the homologous recombination repair factor, Rhp51 (Rad51). Further, the viability of Nse5/6 mutants depends on Mus81 and Rqh1, factors that resolve or prevent the formation of Holliday junctions. Consistently, the UV sensitivity of cells lacking Nse5/6 can be partially suppressed by overexpressing the bacterial resolvase RusA. We propose a role for Nse5/6 mutants in suppressing recombination that results in Holliday junction formation or in Holliday junction resolution.

The Smc5/6 complex plays essential roles in chromosome segregation and repair, by promoting disjunction of sister chromatids. The core of the complex is constituted by an heterodimer of Structural Maintenance of Chromosomes (SMC) proteins that use ATP hydrolysis to dynamically associate with and organize chromosomes. In addition, the Smc5/6 complex contains six non-SMC subunits. Remarkably, and differently to other SMC complexes, the Nse1 and Nse2 subunits contain RING-type domains typically found in E3 ligases, pointing to the capacity to regulate other proteins and complexes through ubiquitin-like modifiers. Nse2 codes for a C-terminal SP-RING domain with SUMO ligase activity, assisting Smc5/6 functions in chromosome segregation through sumoylation of several chromosome-associated proteins. Nse1 codes for a C-terminal NH-RING domain and, although it has been proposed to have ubiquitin ligase activity, no Smc5/6-dependent ubiquitylation target has been described to date. Here, we review the function of the two RING domains of the Smc5/6 complex in the broader context of SMC complexes as global chromosome organizers of the genome.

The Neutrosophic Set (Nset) represents the uncertainty in data with fuzzy attributes beyond true and false values independently. The problem arises when the summation of true (Tr), false (Fa), and indeterminacy In values crosses the membership value of one, that is, Tr+In+Fa<1. It becomes more crucial during decision-making processes like medical diagnoses or any data sets where Tr+In+Fa<1. To achieve this goal, the FNset is recently introduced. This study employs the Interval-Valued Fermatean Neutrosophic Set (IVFNset) as its chosen framework to address instances of partial ignorance within the domains of truth, falsehood, or uncertainty. This selection stands out due to its unique approach to managing such complexities within multi-decision processes when compared to alternative methodologies. Furthermore, the proposed method reduces the propensity for information loss often encountered in other techniques. IVFNS excels at preserving intricate relationships between variables even when dealing with incomplete or vague information. In the present work, we introduce the IVFNset, which deals with partial ignorance in true, false, or uncertain regions independently for multi-decision processes. The IVFNset contains the interval-valued Trmembership value, Inmembership value, and Famembership for knowledge representation. The algebraic properties and set theory between the interval-valued FNset have also been presented with an illustrative example.

A parametric investigation of the effect of impactor stiffness as well as environmental and operational conditions on impact contact behaviour and the subsequently generated lamb waves in composite structures is presented. It is shown that differing impactor stiffness generates the most significant changes in contact area and lamb wave characteristics (waveform, frequency, and amplitude). A novel impact localisation method was developed based on the above observations that allows for variations due to differences in impactor stiffness based on modifications of the reference database method and the Akaike Information Criterion (AIC) time of arrival (ToA) picker. The proposed method was compared against a benchmark method based on artificial neural networks (ANNS) and the normalised smoothed envelope threshold (NSET) ToA extraction method. The results indicate that the proposed method had comparable accuracy to the benchmark method for hard impacts under various environmental and operational conditions when trained only using a single hard impact case. However, when tested with soft impacts, the benchmark method had very low accuracy, whilst the proposed method was able to maintain its accuracy at an acceptable level. Thus, the proposed method is capable of detecting the location of impacts of varying stiffness under various environmental and operational conditions using data from only a single impact case, which brings it closer to the application of data driven impact detection systems in real life structures.
