In this paper, I investigate the use of SWOT analysis and the resource based view as tools to elaborate a business plan and in academic research by assessing the two methods’ strengths and weaknesses.   I found that SWOT analysis is useful to structure and prioritise information in a short and concise way. Even though, the information and the information collection methods as well as the concepts and theories used in combinaton with the model and their quality is of the most importance for a proper use of the model.   The resource based view is useful to reflect over which internal resources are the most valuable for the company, even though it can be difficult to assess the most important assets, i.e. the intangible assets. I also found that the fact that the resource based view, which is a explanantory theory, is used as a predicative theory causes uncertainaty and is problematic.

In order to achieve results that are reliable when using the finite element method one has to use an acceptable element mesh with respect to the shape and size of the elements. As a help to produce an acceptable mesh there are quality criteria that must be fulfilled in most pre-processors.

One objective with this thesis is to perform a sensitivity study that can be used as a basis for a Mesh guideline for chassis parts which is requested from engineers at Volvo 3P. The software used in the sensitivity study is ANSA as pre-processor, Nastran as solver and Metapost as post-processor.

In the first part of the sensitivity study three different models are used for studying quality criteria such as aspect ratio, skewness, mid point alignment, mid point deviation and element size. Solid elements of second order, which are used in the three models, can be generated in two ways, which constitutes another part of the sensitivity study. They may either be generated from the beginning or can be converted from first order elements. This means geometrically that if second order elements where generated from the beginning the element mesh would follow the shape of the component in a better way compared to the other method.

Recently a pre- and post-processing program called SimLab was introduced on the market. Since SimLab supports geometry import from several CAD-systems without loss of feature information, the automatic element mesh generation is supposed to be better as the mesh generator has access to more information concerning the geometry. An evaluation of SimLab is the second major objective of the thesis. More specifically, the evaluation concerns the possibility of using the software at Volvo 3P.

Results show a surprising insensitivity regarding the criteria and that the method of generating second order elements from the beginning is to be preferred. SimLab is a new program with big potential and the conclusion is that it is possible to use it at Volvo 3P.

Chapter One This chapter reviews the historical background of the fluorination using both elemental fluorine and N-F reagents. It relates the properties and preparations of these electrophilic reagents. And finally it discusses the use of these reagents in selective electrophilic fluorination (formation of C-F bond from C-H bond) of organic compounds. Chapter Two, Three and Four These chapters present our investigations into the fluorination of hydrocarbon systems containing an electron withdrawing group (ewg).Chapter Two shows the difference in reactivity of elemental fluorine and Selectfluor(^TM) towards aldehydes and ketones. Select fluor gives rise to a-fluorinated compound when reacting with ketones (acyclic and cyclic ones) whereas different results were observed using fluorine. In Chapter Three aldehydes are submitted to fluorination which occurred at the carbonyl and not on the chain. Finally, Chapter Four review the fluorination of nitriles using both fluorinating reagents. Chapter Five In this last Chapter we explore the fluorination of cyclic ethers (hydrocarbon systems containing an electron donating group) using both elemental fluorine and Selectfluor(^TM). Unexpected derivatives were identified. Chanter Six to Nine Experimental details relating to Chapter Two-Five.

The development of FIRST was a significant step in the field of silicon compilation. With FIRST, bit-serial signal processing systems could be rapidly implemented in silicon by high-level designers without requiring layout expertise. This thesis explores extensions to the compiler, but the methodology and techniques are not specific to FIRST and could be used in the more general VLSI arena. One major theme is the use of process independent layout, allowing the rapid update of a cell library to current state of the art process rules. After surveying other layout strategies, one particular layout style, gate matrix, was evaluated through the manual layout of a bit-serial, two's complement, multiplier utilising novel architectural features. The operation and architectural features of the multiplier are described, as these features were to be incorporated as options in newly generated cell libraries. SECOND, a full span silicon compiler; taking the high-level input description of FIRST but synthesizing layout to a process independent form (gate matrix) was developed using ideas gained from the manual assembly procedure. SECOND maintains and extends the hierarchy of FIRST using different assembly strategies for differing levels of hierarchy in the synthesis procedure. The hierarchy is described and the placement, routing and assembly procedures of the new elements of the hierarchy are covered. The automation tools used to generate the gate matrix layout of the lowest hierarchy level of SECOND are covered in a separate chapter. Using the same concepts of hierarchy, a tool ENGEN which transforms FIRST intermediate code to a gate level network description in HILO is also described as an alternative to SECOND in the search for process independence. The thesis ends with a suggestion of a bit-serial/bit-parallel frame for encouraging the acceptability of bit-serial systems.

The Game Design Document is a well defined set of requirements for a game design and a methodology for game development. The most pertinent aspects of the GDD are described and the relative research collected, in order to better the reader's understanding of the concept. This is a work in two parts, theoretical and practical. Research was an ongoing effort while attempting toproduce a game design according to the content prescribed for the GDD. The resultant design was for a fictitious game dubbed Arcane Dominion.

Knowledge gained about the GDD is used to make a comparison and attempt to identify a possible evolution towards existing software engineeringmethodologies. Some differences are brought to light between the GDD and a popular contemporary methodology, Agile Development.

To compliment the research, one aspect of the designed game was implemented in a prototype and thereafter analyzed in order to obtain neededfeedback and be confronted with the complexities of game balancing pertaining to one chosen game strategy.

Computer science in general and software engineering in specific is changing very fast. Software engineers are constantly using more innovative and more efficient ways to develop new software than in the past. This continuous evolution of software development methodologies has a great impact on both the software developed and the environment that the developers work-in. Agile software development methodologies are used to overcome many issues in the software development processes. One of the issues which still exists and needs to be addressed is the preparation of proper documentation along with the software. The work presented in this dissertation focuses on software documentation. The work starts by a thorough literature review which focuses on different aspects of software documentation and different agile methodologies. The thesis focuses on finding out the challenges that the developers faces during their development process. Two major questions addressed in the thesis. First one is to find the motivation to document in agile envirionment, whih is based on the hypothesis that there do exist a motivation. The second question is that how should documentation be produced such that we could avoid maximum possible potential problems. These questions are addressed with the help of different perspectives of the stockholders (i.e. developers and users) and the existing methods for documentation. A questionnaire was developed based on the nine categories of documentation, like user documents and system documents etc.. It included different questions related to the types of documents created in software development processes, the software development stage at which the documents are created and the importance of the documents. Questions from this questionnaire are then posted on agile specific discussion forums. Where many experienced and fresh practitioners participated in the discussion. We had a detailed discussion on every component of documentation and problems were identified by the practitioners. The questionnaire was also sent to different companies practicing agile methodology. we received about 14 responses as it was detailed questionnaire with about 34 questions. The responses of the discussion forum and survey are then analyzed and conclusions were drawn. The conclusions include that all the participants consider software documentation very important to the success of a software development project. the question of motivation is answered from the literature and opinions we received from experienced practitioners. While seven factor are identified that affect your documentation, to help solve the question of how should documentation be done.

This thesis is divided into two sections. The first section covers techniques, strategies and mechanisms of oligosaccharide formation, illustrated with both literature and the author's results, highlighting the many subtleties involved in performing a glycosylation. These include attempts at trans-glycoside formation on a polymer support without the use of an acetyl directing group and the curious failure of an acetyl group to direct a glycosylation in the synthesis of a lactosamine derivative. The second section describes studies towards the development of stereocontrolled glycosylation protocol, based upon a intramolecular aglycon delivery strategy. Various tethers ranging from anomeric carbonates to anomeric phosphoranes were tried to realise this strategy but a solution remained elusive.

Sustainability is a vague concept specifically in the context of a corporate world. There are numerous definitions for corporate sustainability and just as many ways of evaluating it. This work attempts to define, structure and assess corporate sustainability in a standardized robust manner through development of a comprehensive framework. The framework is developed based on Global Reporting Initiative (GRI) guidelines to serve as a common measure system allowing for meaningful assessment of current state and comparison between companies in a variety of industries. Based on this framework and earlier developed Sustainability Assessment by Fuzzy Evaluation (SAFE) model a quantitative method is developed in MATLAB code. The new method is demonstrated on five companies within software industry through evaluation of publicly available data. The outcome of the evaluation is a relative ranking of companies with respect to economic, social and environmental aspects as well as intermediate components of each. Additionally high impact components, which have the potential to improve the ranking outcome the most, are identified for one of the companies as an example of practical application of such assessment. Evaluation of these components could serve as a base for recommendations development of further management action on improving of company's sustainability.

Thesis (M.S.)--Massachusetts Institute of Technology, Dept. of Architecture, 1989.

The act of designing in architecture is a complex process. Many designers, when probed for reasons to explain their actions, are either unable to answer questions, or provide explanations that are not true descriptions of their actions. Frequently the designer will answer that his or her reason for making a particular design decision is based on 'feeling' or 'intuition.' Under this model the design process assumes a 'mystical' aura. Architectural designers can create, yet are unable to say how they do so. Often that which can be explicitly discussed by the designer is the least significant part of his or her design process. It is unlikely that designers are 'channeling' information from cosmic sources. Rather, they are working with knowledge that is largely tacit. This thesis attempts to de-mystify the process of architectural design. Through a close scrutiny of existing literature, incorporation of personal experience as an architect, and testing of theories with lay, novice, and expert designers a theory of design methodology is proposed.

The tracking modulation index (Km) is a key performance parameter for any autotracking antenna and should especially be considered for classically difficult targets such as missiles and/or fast moving aircraft. Antenna subsystems are typically characterized by their gain to temperature ratio (G/T) to optimize receive data bit error rates (BER) for distant targets. One important parameter often overlooked for telemetry autotracking antennas is a graded value for the available tracking modulation index (Km) that is common in radar autotracking applications. Tracking modulation performance is a major contributor for minimizing the antenna pointing error during an autotrack mission. Autotracking radar antenna specifications typically include tracking modulation as a major design parameter, many receive-only autotrack antennas used for telemetry applications do not consider this important parameter for the intended tracking mission which may result in poor autotracking performance. This paper investigates the effects of tracking modulation levels on system pointing errors for various classes of feed topologies.

Master’s Degree Dissertation
The present dissertation covers academic concerns in disruptive change that causes value displacements in today’s competitive economic environment. To enhance survival capabilities organizations are increasing efforts in more untraditional business value assets such intellectual capital and competitive intelligence. Dynamic capabilities, a recent strategy theory states that companies have to develop adaptive capabilities to survive disruptive change and increase competitive advantage in incremental change phases. Taking advantage of the large amount of information in the World Wide Web it is propose a methodology to develop applications to gather, filter and analyze web data and turn it into usable intelligence (WeCIM). In order to enhance information search and management quality it is proposed the use of ontologies that allow computers to “understand” particular knowledge domains. Two case studies were conducted with satisfactory results. Two software prototypes were developed according to the proposed methodology. It is suggested that even a bigger step can be made. Not only the success of the methodology was proved but also common software architecture elements are present which suggests that a solid base can be design for different field applications based on web competitive intelligence tools.

Partial Least Squares (PLS) is an important statistical technique with multipleand diverse applications, used as an effective regression method for correlated orcollinear datasets or for datasets that are not full rank for other reasons. A shorthistory of PLS is followed by a review of the publications where the issues with theapplication PLS that have been discussed. The theoretical basis of PLS is developedfrom the single value decomposition of the covariance, so that the strong links between principal components analysis and within the various PLS algorithms appear as a natural consequence. Latent variable selection by crossvalidation, permutation and information criteriaare examined. A method for plotting crossvalidation results is proposed that makeslatent variable selection less ambiguous than conventional plots. Novel and practicalmethods are proposed to extend published methods for latent variable selection byboth permutation and information criteria from univariate PLS1 models to PLS2 multivariate cases. The numerical method proposed for information criteria is also more general than the algebraic methods for PLS1 that have been recently published as it does not assume any particular form for the PLS regression coefficients. All of these methods have been critically assessed using a number of datasets, selected specifically to represent a diverse set of dimensions and covariance structures. Methods for simulating multivariate datasets were developed that allow controlof correlation and collinearity in both regressors and responses independently. Thisdevelopment also allows control over the variate distributions. Statistical design ofexperiments was used to generate plans for the simulation that allowed the factorsthat infuence PLS model fit and latent variable selection. It was found that all thelatent variable selection methods in the simulation tend to overfit and the feature inthe simulation that causes overfitting has been identified.

This dissertation provides three novel methodologies to the field of political science. In the first chapter, I describe how to make causal inferences in the face of dynamic strategies. Traditional causal inference methods assume that these dynamic decisions are made all at once, an assumption that forces a choice between omitted variable bias and post-treatment bias. I resolve this dilemma by adapting methods from biostatistics and use these methods to estimate the effectiveness of an inherently dynamic process: a candidate's decision to "go negative." Drawing on U.S. statewide elections (2000-2006), I find, in contrast to the previous literature, that negative advertising is an effective strategy for non-incumbents. In the second chapter, I develop a method for handling measurement error. Social scientists devote considerable effort to mitigating measurement error during data collection but then ignore the issue during analysis. Although many statistical methods have been proposed for reducing measurement error-induced biases, few have been widely used because implausible assumptions, high levels of model dependence, difficult computation, or inapplicability with multiple mismeasured variables. This chapter develops an easy-to-use alternative without these problems as a special case of extreme measurement error and corrects for both. In the final chapter, I introduce a model for detecting changepoints in the distribution of contributions data because it allows for overdispersion, a key feature of contributions data. While many extant changepoint models force researchers to choose the number of changepoint ex ante, the game-changers model incorporates a Dirichlet process prior in order to estimate the number of changepoints along with their location. I demonstrate the usefulness of the model in data from the 2012 Republican primary and the 2008 U.S. Senate elections.

As a field, Management Consulting is mostly unknown to students of public universities. This unfortunate circumstance is a product of favoritism of elite, private colleges in the hiring processes of most consulting firms. Regardless of the presence or lack of intention of pursuing management consulting as a career path, undergraduates can benefit from the application of management consulting principles, especially in terms of business problem solving. Undergraduate consulting field projects help to hone analytical skills, apply course material, and serve the community. In order to more easily facilitate undergraduates participating in such projects, this thesis first outlines best practices by top firms in the management consulting industry. It then adapts these best practices to provide a broad methodology for undergraduate use in pursuing consulting projects, as well as a specific methodology for one of the more common undergraduate--‐level consulting tasks: marketing analysis and design. The thesis then applies these methodologies through a case study, a consulting engagement performed by Eller Consulting Club for a local entrepreneur. Finally, the thesis provides guidelines for securing employment in the management consulting industry, focusing on the interview process. True to consulting form, the thesis is presented in a straightforward, easily accessible slide deck.

The report describes a prestudy project for Epiroc Rock Drills AB at the department PLM Solutions. The company consists of a number of divisions thatall work with or are affected by CAD models. PLM Solutions has long seen a need for a common methodologybut due to the priorities of other projects this has been delayed. The purpose of the prestudy was to provide a mapping of the CAD methodology at the company today and also investigate the need for methodology. Based on the mapping, a recommendation was given ofhow PLM Solutions shouldproceed with a major methodology project. The recommendation wastocreate a common overall CAD methodology and take advantage of the already existing methodsin the organization. As the problem was treated as a prestudy, the method of a prestudy model is also the case. The model is general and adapted to fit this particular project. The approach deals with the phases: start-up, background analysis and solutions. The background analysis examines the current situation analysis using models for interviewing techniques. A larger part of the basis for the prestudy rests on interviews with stakeholders. The result is presented based on the same phases that the method consists of. The result is a mapping of the methodology and needs of the divisions, this is also visualized using a SWOT analysis. The results arealso the recommendation and requirements based on the interviews and business case. Continued work should as a suggestion include several more interviews, collecting more material from around the divisions, more detailed calculations and a focus on the departments that are affected by the models.

The Utah Department of Transportation (UDOT) Traffic and Safety Division continues to advance the safety of the state roadway network through network screening and decision making tools. In an effort to aid UDOT in meeting this goal, the Department of Civil and Environmental Engineering at Brigham Young University (BYU) has worked with the Statistics Department in developing analysis tools for highway safety, specifically the Utah Crash Prediction Model (UCPM) and the Utah Crash Severity Model (UCSM). Additional tools and methodologies, such as the "Hot Spot Identification and Analysis," have been created to summarize the roadway characteristics, crash data, and possible countermeasures of roadway segments with safety problems. This research focuses on the creation of a three part "Roadway Safety Analysis" methodology, which applies and automates the cumulative work of recently completed highway safety research conducted for UDOT. The first part is to prepare the roadway data and crash data for the statistical analysis. The second part is to perform the network screening statistical analysis; rank the segments by state, UDOT Region, and county; and select segments of interest. The third part is to compile and publish the Roadway Safety Analysis reports for the selected segments of interest. These parts are accomplished using the automation tools and graphical user interfaces (GUIs), which are documented in three respective volumes of user manuals. The automation tools and GUIs were developed with checks and processes to allow the Roadway Safety Analysis methodology to be completed with new, updated roadway and crash datasets. The Roadway Safety Analysis methodology allows future iterations of the UCPM and UCSM analysis and compilation of the Roadway Safety Analysis reports to be conducted in a user friendly environment. A series of critical data columns were identified to communicate the need for data consistency for future iterations of this safety research. An example of the entire process of the Roadway Safety Analysis methodology is given to illustrate how the three parts tie together. The overall process has automated data processing tasks, which saves time and resources for the analyst to investigate possible safety measures for segments of interest. The goal of this document is to outline the trajectory in which I have been working the past several years. I would like to comment and detail the production of several projects, including Site Nine: Indefensible Structures, Text 11: Site Translations, Study For Casablanca: Maps For Access And Improvised Housing, and Lavon, Texas - Levittown, New York. The reach of the projects, and it is perpetually adaptive to new concerns, is not to assign a space between art, architecture, and planning. Even though there is certainly space which the works will invoke between these areas. This document and the projects would question why there should be such a space and what it could achieve. It may be possible to see at such a point that assigning nomenclature can be considered a quick task and the value is the content of a word, not by the name by which it is called. This is not to say meaning and context cannot be derived from the title of disciplines or fields, they in fact provide considerable insight. The aforementioned projects, and those to follow, would look into the social climates of a location and only after considerations of the political, economic, and communicative indicators, courses of intervention would be developed. I would like to note, ever more in the continuation of these works, the implementation of intervention becomes less so. The projects, in chronology, quietly move from methods of production to methods of research. This was due to an increasing appeal to the cultural sensitivity of any and all methods of production, in both domestic and foreign capacities.

The estimation of effort is at the heart of project tasks, since it is used for many purposes such as cost estimation, budgeting, monitoring, project planning, control and software investments. Researchers analyze problems of the estimation, propose new models and use new techniques to improve accuracy. However up to now, there is no comprehensive estimation methodology to guide companies in their effort estimation tasks. Effort estimation problem is not only a computational but also a managerial problem. It requires estimation goals, execution steps, applied measurement methods and updating mechanisms to be properly defined. Besides project teams should have motivation and responsibilities to build a reliable database. If such methodology is not defined, common interpretation will not be constituted among software teams of the company, and variances in measurements and divergences in collected information prevents to collect sufficient historical information for building accurate models. This thesis proposes a methodology for organizations to manage and execute effort estimation processes. The approach is based on the reported best practices, v empirical results of previous studies and solutions to problems &
conflicts described in literature. Five integrated processes: Data Collection, Size Measurement, Data Analysis, Calibration, Effort Estimation processes are developed with their artifacts, procedures, checklists and templates. The validation and applicability of the methodology is checked in a middle-size software company. During the validation of methodology we also evaluated some concepts such as Functional Similarity (FS) and usage of Base Functional Components (BFC) in effort model on a reliable dataset. By this way we evaluated whether these subjects should be a part of methodology or not. Besides in this study it is the first time that the COSMIC has been used for Artificial Neural Network models.

My aIm In this thesis is to analyse the 'evolutionary analogy', a particular form. of evolutionary epistemology which claims that scientific change is governed by the same mechanisms, or by mechanisms analogous to those at work in organic evolution, mainly natural selection. In tenns of research questions, the overall aim of this thesis is to answer 'Is the process of scientific change analogous to or even the same as organic change?' Many philosophers proposed evolutionary theories of scientific change - evolutionary analogies. By drawing analogies or even equating the mechanisms of organic and scientific evolution they described the process underlying the latter but also justified its value, implicitly or explicitly, by a simple analogy: better theories are those which survive old ones, as better species are those which survive previous ones. The results of these philosophers, however, have not been satisfactory and a novel approach is needed. In this thesis I am interested in a purely descriptive philosophy of science and my position will be based on the critique of the most recent and comprehensive attempt to defend the evolutionary analogy, made by David Hull. In order to answer the research question I have formulated above, the following issues will be addressed: what is organic evolution;. what is meant by 'evolutionary analogy'; how analogy/identity can be evaluated; how evolutionary analogy/identity has been defended by philosophers; how these defences perform; what is the best defence available; whether it passes the evaluation and, if not, whether it can be improved; if all the positions fail, whether it is possible to conceive alternative analogies between other relevant processes which do not incur the same problems. By addressing these issues, I will be able to conclude that the process of scientific change is different from organic change and that only loose analogies can be defended.

Pedagogical methodology has rarely been considered as a driving force in educational facilities planning and design. Because of constantly changing learning modalities, state standardization of learning and incredibly diverse educational demographic, designing learning spaces that support various pedagogies is paramount if our students are to succeed in the knowledge-based economy. From its beginnings as the rural one-room school house to the contemporary single volume, teaching and learning methodologies are continually changing and adapting to contemporary necessity. Little emphasis has been placed over the qualitative aspects of facilities planning, and how children may benefit from varying spatial environments. No practical and consistent concept of open-space learning has ever been established when considering open-concept education. This is the fundamental tenet to this master's thesis. This thesis will analyze contemporary learning methodologies with particular emphasis on physical learning and the transition to digital learning. Through the analysis of varying pedagogies specific qualitative themes, modalities, and hierarchical mechanisms will be made known and the architectural design strategies will draw upon these themes. The thesis presents information necessary to designing within a neighborhood sensitive to history and place. The rich significance of place in the Mississippi/Yazoo Delta will be a driving force in the design of a K-6 Elementary school to replace the existing school. The thesis will additionally draw upon the analysis of various precedents, demographic studies, site analyses, and code analyses to establish an architecture rich in character and performative qualities necessary for growing minds.

In today’s industry, the competitive market, the short life time of the products and rapid change in customer demand forms a big trend of appearance of new manufacturing system. Reconfigurable Machine Tool (RMT) is a kind of solution for future machining systems, thus it can not only provide customized solutions to the operation requirements but also is cost-effective. Thethesis aims to create and implement methodology of RMT design for manufacturing industry. The methodology is introduced and extended according to the five principals of modular machine tool design. The methodology will be applied step by step herein so as to make the methodology more clear. First of all is to get the concept or process requirements from the customer. The customer requires three reconfigurable part families for a high speed milling application including tables, spindles, and cutters. In this case, to fulfill customer’s requirements and concept, the dual spindle (Multi-tool) RMT is selected as an example to interpret the RMT design methodology. Secondly, the most important point in methodology of RMT design is to analyze the valuable proposal of the four principles of modular design based on extensive experience. This work turns the four principles, separation, and unification (standardization), connection, and adaptation, to practical design methodology. Based on the classical four principles, the principle of reusability is an addition principal in consider of zero waste concept. In order to perform the RMT design methodology effectively, it is necessary to complement the advantages of the reconfiguration in dual spindle RMT design and the reconfiguration for other part families in according to the RMT design methodology. A final step utilizes computer software to model the configuration in 3 dimensions. And evaluations can be discussed in Degree of Freedom (DOF), stiffness analysis and number of modules.

The HOF.MeCN complex, formed from the reaction of elementary fluorine with aqueous acetonitrile, was discovered by Shlomo Rozen in 1987 and, in a series of publications, he demonstrated that the complex is a uniquely effective electrophilic oxygen transfer agent. However, it is estimated that the HOF.MeCN complex has a half life of a few hours at 0ºC and thus, must be produced and used immediately when required. In addition to this, highly exothermic, rapid oxidation processes can be problematic with reaction control and safety when reagents are added into an excess of a highly oxidizing medium. Consequently, scale-up of oxidations to a larger industrial level using the HOF.MeCN complex in batch processes would not be possible. In recent years, continuous flow reactors using microchannels have been viewed as a viable method for avoiding many of the problems encountered when a laboratory process is scaled-up. The low chemical inventory of such reactors means that even highly reactive reactions can be performed safely and, with the application of multiple reaction channels in parallel, large quantities of product can be easily obtained. In this thesis we present new continuous flow methodology for the in situ generation of HOF.MeCN and, without isolation, immediate substrate oxidation in a two-step process. The continuous process, therefore, provides a genuine method for oxidizing large quantities of material, without the problems associated with batch oxidations. Scale-up oxidations of various amines, alkenes, and anilines are also presented along with a safe and accurate method for calibrating HOF.MeCN amounts.

Test theories are mathematical constructions with a methodological purpose. They enable experimental or other observational evidence, definitively to select a physical theory from out of a class of alternatives-a formal deductive method of testing which is (as I will show) distinct from the traditional hypothetico-deductive one. I believe that the study here of test-theory methodology is important for philosophers of science if they are properly to understand (i) methods of theory appraisal in physics, (ii) certain key conceptual issues in the foundations of physics, and (iii) more generally, the growth and structure of physical theory (history). I believe that this study is important also for physicists who desire a clearer understanding of the power, limitations, and conceptual presuppositions of the test-theory method. (i) Methods. I show how test theories establish relations between physical theory and phenomena which are at once stronger and more systematic than relations established by the hypothetico-deductive method. I use this result to defend some positivist methodological doctrines which philosophers today generally reject. I also use the result to criticise certain post-positivist doctrines which are meant to replace the very positivist doctrines I aim to defend. I compare some philosophical accounts of empirical confirmation with test-theory methodology in order to illuminate some features of the test-theory method and criticise standard philosophical accounts of confirmation. In this discussion I explore limitations of the test-theory method and identify general conditions which both knowledge and the world must satisfy if the method is to work. (ii) Foundations. I address some key issues in the foundations of physics issues concerning the nature of space, time and causation. I discuss how test-theory methodology reveals the extent to which the very foundations of physics are empirically determinable. This extent is limited I argue: my discussion points to the furthest application but also to some notable limitations of the test-theory approach. (iii) History. I argue that Newton pioneered the use of test-theory constructions in theoretical physics, and I show how twentieth century physicists have brought the test-theory method to a high level of sophistication. I argue both that the test-theory idea makes possible a coherent view of the history of physics, and that the history of physics deepens our appreciation of the power of the test-theory idea. There is unity, I argue, at the level of test-theory methodology across the seemingly disparate historical phases of physical inquiry (classical versus modern). There is power, I maintain, in a method which can fruitfully be applied in seemingly diverse conceptual contexts (Newtonian versus Einsteinian).

Rare disease variants are receiving increasing importance in the past few years as the potential cause for many complex diseases, after the common disease variants failed to explain a large part of the missing heritability. With the advancement in sequencing techniques as well as computational capabilities, statistical methodology for analyzing rare variants is now a hot topic, especially in case-control association studies. In this thesis, we initially present two related statistical methodologies designed for case-control studies to predict the number of common and rare variants in a particular genomic region underlying the complex disease. Genome-wide association studies are nowadays routinely performed to identify a few putative marker loci or a candidate region for further analysis. These methods are designed to work with SNP data on such a genomic region highlighted by GWAS studies for potential disease variants. The fundamental idea is to use Bayesian methodology to obtain bivariate posterior distributions on counts of common and rare variants. While the first method uses randomly generated (minimal) ancestral recombination graphs, the second method uses ensemble clustering method to explore the space of genealogical trees that represent the inherent structure in the test subjects. In contrast to the aforesaid methods which work with SNP data, the third chapter deals with next-generation sequencing data to detect the presence of rare variants in a genomic region. We present a non-parametric statistical methodology for rare variant association testing, using the well-known Kolmogorov-Smirnov framework adapted for genetic data. it is a fast, model-free robust statistic, designed for situations where both deleterious and protective variants are present. It is also unique in utilizing the variant locations in the test statistic.

Flowgraph models are directed graph models for describing the dynamic changes in a stochastic process. They are one class of multistate models that are applied to analyse time-to-event data. The main motivation of the flowgraph models is to determine the distribution of the total waiting times until an event of interest occurs in a stochastic process that progresses through various states. This thesis applies the methodology of flowgraph models to the study of Markov and SemiMarkov processes. The underlying approach of the thesis is that the access to the moment generating function (MGF) and cumulant generating function (CGF), provided by Mason’s rule enables us to use the Method of Moments (MM) which depends on moments and cumulant. We give a new derivation of the Mason’s rule to compute the total waiting MGF based on the internode transition matrix of a flowgraph. Next, we demonstrate methods to determine and approximate the distribution of total waiting time based on the inversion of the MGF, including an alternative approach using the Pad´e approximation of the MGF, which always yields a closed form density. For parameter estimation, we extend the Expectation-Maximization (EM) algorithm to estimate parameters in the mixture of negative weight exponential density. Our second contribution is to develop a bias correction method in the Method of Moments (BCMM). By investigating methods for tail area approximation, we propose a new way to estimate the total waiting time density function and survival

This work describes the development of three methods for the fluorination of electrophilic substrates:1.) The reaction of caesium fluoride with perfluoro(2-methylpent-2-ene) leads to the formation of perfluoro(2-methylpentan-2-yl)caesium. This perfluoroalkyl carbanion has been shown to undergo significant fluoride ion exchange at temperatures above 60 C. Thus, reaction of a solution of the carbanion with a suitable electrophile resulted in the selective formation of a carbon-fluorine bond or perfluoroalkylation of the electrophile.2.) Caesium fluoride has been developed as a moderately effective nucleophilicfluorinating agent in the Room Temperature Ionic Liquid (RTIL) solvent [BMIM][PF(_6)]. The fluorination of a range of volatile substrates was studied, and fluorination in the absence of a conventional organic solvent was demonstrated. Recycling of the solvent has been investigated, as has the decomposition of [BMLM] [PF(_6)] in the presence of caesium or potassium fluoride at elevated temperatures.3.) Reaction of a highly fluorinated azaheterocycle with DMAP leads to the formation of a fluoride salt. This salt, formed in situ, was used as a source of nucleophilic fluoride ion for the fluorination of a range of electrophiles. Several of the fluoride salts were converted to their more stable tetrafluoroborate or triflate analogues and characterised.
