In the current context of the growth of healthcare expenditures, health economics can help decision making in the process of resources allocation in the field of health. Neurodegenerative diseases (ND) are huge and costly diseases for which no cure is available to date. Prevention can be an axis of care of theses pathologies. It could help delay the disease progression or complications linked to these ones or to its treatments. In the frame of NDs, health economics can help to make choices in order to provide quality of healthcare to patients in the context of constraint resources. This thesis aimed to describe and analyze methodological characteristics of studies which assessed NDs costs, paying particular intention to informal costs. Our works revealed significant variations of methodological characteristics of a study to another. An additional effort must be performed by the scientific community in the field of methodological validation. This thesis also aimed to assess medical and economic consequences of prevention strategies in Alzheimer disease (AD) and Parkinson disease (PD). The observed results concerned the implementation of a directed prevention program in AD (I.e. multidomain intervention) and of a targeted prevention program in PD (i.e. therapeutic education). They confirmed the need to implement studies on prevention program on substantial time horizons

In Drosophila, glutamate is the neurotransmitter at neuromuscular junctions (NMJ) and in the central nervous system (CNS). We studied Drosophila homologues of genes coding for re-uptake glutamate transporters (EAAT) and vesicular glutamate transporters (VGLUT). DEAAT1, the only EAAT Drosophila glutamate transporter, is expressed in glial extensions that project in the neuropil. Inactivation of dEAAT1 by RNA interference led to increased duration of muscle potential, behavior deficits, shortened life span, increased sensitivity to oxidative stress and neuropil degeneration. These results demonstrate that dEAAT1 participates to the removal of glutamate at synapses, and prevent glutamate-induced neurotoxicity. We isolated DVGLUT a VGLUT gene. DVGLUT represented the first available marker of central glutamatergic synapses. Surprisingly, DVGLUT positive glutamatergic synapses were found in high abundance in the CNS. This suggests that glutamate is a major neurotransmitter in the Drosophila

Alzheimer's disease is a neurodegenerative aging-related dementia that is characterized by loss of memory and cognitive disorders. Toxicity of soluble oligomers of [beta]-amyloid peptide (sOA[beta]) is a key element in early stages of the disease. Absence of curative therapies, chronic aspects of the pathogenic mechanisms implicated and influence of common risk factors shared with the cardiovascular diseases, including dietary parameters and lipid metabolism, should widely encourage considering the interest of preventive interventions allowing slowing the progression and delaying the clinical onset of Alzheimer's-related troubles. Therefore, nutritional approaches could appear as a strategy able to reduce the prevalence of this disease. Early Alzheimer's mouse model allowed us to assess the preventive potential of diets supplemented in docosahexaenoic acid (DHA, C22:6 n-3). Our results show that adequate dietary intake of DHA lead to increased levels in different brain structures. Consequently, hippocampal and cortical synaptic functions were preserved, even upon acute exposure to A[beta] oligomers, maintaining or improving the cognitive capacities of A[beta]-exposed mice. These improvements were positively correlated with DHA-enrichment associated in hippocampus and with preserved synaptic integrity. We also designed nutritional strategies in order to evaluate the beneficial effects of DHA on diet-induced dyslipidemia as well as on cognitive impairment and neurodegenerative processes associated with normal or pathologic aging. Our results show that dietary DHA can prevent high-fat diet-induced dyslipidemia and delay cognitive decline related with normal or pathological aging

Alzheimer's disease (AD) is the most common cause of neurodegenerative disease and represents a major public health issue worldwide. Many therapeutic strategies have been explored for several decades, however, there is still no curative treatment and the priority remains prevention. In this thesis work, we focused on the preventive approach based on lipids, in particular omega-3 polyunsaturated fatty acids (n-3 PUFAs). N-3 PUFAs play an important role in the development, maintenance and function of the brain, and they have been the subject of particular interest in the prevention of cognitive deficits associated with neurodegenerative diseases. The objective of this work is to study the functionality and bioavailability of nanoliposomes (NL) rich in n-3 PUFAs, in order to assess their neuroprotective potential to develop new preventive strategies in aging-related diseases such as AD. To do this, a green extraction technique was used to prepare NL, from natural resources from salmon lecithin rich in n-3 PUFA. This thesis is based on three parts. The first part is devoted to a bibliographical review of new techniques to facilitate the access of molecules to the brain. NL shows a promising role, being able to improve the transport of molecules across the blood-brain barrier and reach relevant brain regions. In the second part of this work, the bioavailability of NL rich in n-3 PUFA was studied in a primary culture of cortical neuronal cells from rat embryos and in a mouse model. This study shows for the first time the brain bioavailability of NL rich in n-3 PUFA in in vitro and in vivo models. Finally, in the third part of this thesis, the physicochemical properties and transfer mechanisms of NL were studied in a primary culture of cortical neurons. These results provide new information on the interaction between NL and neurons and are promising with regards to the use of NL rich in n-3 PUFA, opening up new possibilities in the development of preventive and neuroprotective therapeutic strategies for neurodegenerative diseases such as AD
